    هدف مطالعه

    سرطان پروستات دومین سرطان شایع و پنجمین عامل مرگ ناشی از سرطان مردان در سال 2012 در جهان می باشد. مسیر PI3K/AKT نقش اساسی در بیماری زایی سرطان پروستات دارد و به جهت نقش کلیدی این مسیر در پیشروی سرطان آن را به هدف جذابی برای استراتژی های جدید درمانی تبدیل می کند. یکی از روندهای تنظیمی ژنها، میکرو RNAها می باشند که توانایی ویژه ای برای کنترل چندین ژن و مسیر به طور همزمان، آنها را کاندید قابل توجهی برای اهداف درمانی می کند. در این بررسی بر آن بودیم که میکروRNA موثر بر مسیر PI3K/AKT را با روش های بیوانفورماتیکی به دست بیاوریم و نتایج حاصله را در سلول های رده پروستات بررسی نماییم.

    مواد و روش ها

    برای پیدا کردن میکروRNA موثر بر مسیر PI3K/AKT، شش ژن این مسیر که به عنوان هدف های دارویی مطرح هستند را در ده الگوریتم متفاوت پیش بینی بررسی کردیم. سپس به صورت بیوانفورماتیکی میکروRNA حاصله را از نظر میزان بیان در بافت پروستات سالم و رده های سلولی سرطان پروستات وهمچنین از لحاظ هدف گیری در نرم افزارهای آنالیز چرخه مورد ارزیابی قرار دادیم. به صورت عملی بیان میکروRNA های کاندید در نمونه کنترل و رده های سلولی سرطان پروستات ارزیابی گردید.

    یافته ها

    میرهای خانواده 29 بیشترین شناسایی را از مجموعه 6 ژنی داشتند و سایر بررسی های بیوانفورماتیک نیز موید نتیجه حاصله بودند. بررسی ها نشان دهنده کاهش بیان معنی دار میرهای خانواده 29 در رده های سلولی سرطان پروستاتDU-145،PC3 و LNCAP نسبت به نمونه کنترل سالم می باشد.

    نتیجه گیری

    و ی ار و ر ن میرهای خانواده 29، نشان دهنده امکان استفاده از میرهای خانواده 29 برای مهار مسیر PI3K/AKT در سرطان پروستات می باشد.

    هدف
    دندریمرها نانوساختارهای سه بعدی هستند که کاربردهای زیادی در پزشکی از جمله دارورسانی و تصویر برداری پیدا کرده اند. دندریمر آنیونی پلی اتیلن گلیکول-سیترات، پتانسیل بالایی برای دارو رسانی و افزایش حلالیت داروهای نامحلول در آب دارد ولی روش سنتز آن زمانبر و چند مرحله ای است و از مواد سمی مانند دی کلرو متان در سنتز آن استفاده شده است. در این تحقیق روشی ساده تر و یک مرحله ای و با استفاده از شیمی سبز برای سنتز آن ابداع شد.
    مواد و روش ها
    چهار روش مختلف جهت بهینه سازی سنتز این دندریمر مورد بررسی قرار گرفت. تایید و تعیین ساختمان محصولات توسط DLS، FTIR و LC-MS انجام شد و میزان سمیت سلولی با استفاده از تست XTT بررسی شد.
    نتایج
    در این تحقیق روش سنتز دندریمر G2 در یک مرحله و بدون تخلیص G1 ابداع شد که در مقایسه با روش های قبلی ارائه شده برای سنتز این دندریمر نه تنها به زمان بسیار کمتری نیاز دارد بلکه از مواد غیر سمی و شیمی سبز نیز در آن استفاده شده است. همچنین با توجه به نتایج تست XTT سمیت قابل توجهی تا غلظت 800 میکرومولار در رده های سلولی Hela و Vero مشاهده نشد.
    نتیجه گیری
    نتایج نشان می دهد که سنتز یک مرحله ای دندریمر آنیونی پلی اتیلن گلیکول-سیترات نسل دو (G2) روش ساده و مناسب برای ساخت این دندریمر می باشد. همچنین این دندریمر زیست سازگار بوده و می تواند به عنوان حامل مناسبی جهت اهداف دارو رسانی مورد استفاده قرار گیرد.

    زمینه و هدف

    جهت بهبود حلالیت و فراهمی زیستی کورکومین در درمان سرطان، یک پلی یورتان کاتیونی نوین و آب پایه سنتز شد و به عنوان یک نانو حامل به منظور بارگیری کورکومین، مورد استفاده قرار گرفت. ما اثر نانوداروی آماده شده را بر روی ملانوما (F10B16) و فیبروبلاست (L929) مورد بررسی قرار دادیم.

    مواد و روش ها

    مورفولوژی، سایز و توانایی ورود به سلول نانوذرات سنتز شده، به ترتیب، با استفاده از زتا سایزر، میکروسکوپ الکترونی روبشی انتشار میدان، میکروسکوپ نیروی اتمی و میکروسکوپ فلورسانس بررسی شد. به این ترتیب پیش بینی شد که کورکومین در هسته آبگریز حامل های پلی یورتان بارگیری شود. بعد از تعیین غلظت مناسب و انجام تست ورود به سلول، میزان تاثیر گذاری بر سلول های سرطانی و نرمال با تست MTT و Real time PCR مورد بررسی قرار گرفت.

    نتایج

    قطر نانوذره بارگیری شده و پراکنش آن، در دمای 25 درجه سانتی گراد به ترتیب 1/7 ± 62/37 نانومتر 2/1 ± 0/080 بود. میزان بازده کپسوله سازی کورکومین در پلی یورتان 0/2 % ± 87 می باشد. مطالعات مورفولوژیکی نیز، کروی شکل بودن و پراکنش مناسب نانوذرات، بدون تجمع قابل توجه را تایید کردند. نتایج حاصل از تست MTT نشان می دهد که IC50 محاسبه شده برای زمان 24 و 48 ساعت به ترتیب 36/2 و 25/4 مایکرومولار می باشد. نتایج Real time PCR نشان می دهد که نانوسامانه به مقدار قابل توجهی میزان بیان mRNA ژنهای VEGF، Bcl-2، MMP-9 و COX-2 را کاهش و BAX را افزایش می دهد.

    نتیجه گیری

    نتایج ما، شواهد قابل قبولی در مورد اثر بازدارنده تکثیر سلولی و القای مرگ برنامه ریزی شده نانوسامانه دارویی کورکومین، بر روی سلول های سرطان پوست نشان داد در حالی که اثر قابل توجهی بر روی سلول های نرمال مشاهده نشد.

    هدف

    اندومتریوز بیماری شایعی است که طی آن غدد و استرومای اندومتر در بیرون از حفره رحمی به صورت نابجا رشد می کند. ایجاد مدل های حیوانی جهت ارزیابی این بیماری کمک کننده خواهد بود. بنابراین در این مطالعه سعی شد دو مدل حیوانی اندومتریوز با استفاده از قطعات بافت و سلول های جداشده آندومتر انسان ایجاد شود و با هم مقایسه گردد.

    مواد و روش ها

    ابتدا بافت اندومتر انسان در فاز تکثیری یا ترشحی از زنان بستری در بیمارستان امام خمینی تهران که تحت هیسترسکوپی به علل خوش خیم قرار گرفته بودند تهیه شد و پس از تایید نرمال بودن بافت، قسمتی از بافت اندومتر انسان به تکه هایی به ابعاد 2 میلی متر مکعب بریده شد و بقیه بافت برای جداسازی سلول ها و کشت تا چهار مرحله مورد استفاده قرار گرفت. در مدل اول تکه های اندومتر به ابعاد 2 میلی متر مکعب و در مدل دوم 6 10×2 سلول اندومتر حاصل از پاساژ چهارم به صورت زیر جلدی به موش های تحت تیمار با اشعه گاما منتقل شد و به مدت 20 روز موش ها در شرایط کنترل شده و استریل نگهداری شدند. خونگیری از موش ها قبل و پس از پیوند جهت تعیین سطح استروژن انجام گرفت و بافت اکتوپیک ایجاد شده در هر دو مدل جمع آوری شدند و جهت ارزیابی مرفولوژیک با بکارگیری رنگ هماتوکسیلین و ائوزین و بررسی ترشحات غدد از واکنش اسید پریودیک شیف (PAS) استفاده شد همچنین تعداد مقاطع غدد در واحد سطح محاسبه و شمارش شد.

    نتایج

    20 روز پس از پیوند بافت غددی شبیه بافت اندومتر در زیر جلد موش در هر دو مدل دیده شد و تعداد مقاطع غدد در واحد سطح 2mm در مدل اول 17/18 ± 57/55 و در مدل دوم 77/98 ± 271/57 بود. وسعت غدد ایجاد شده در مدل دوم بیشتر از مدل اول بود (0/05≥P) و ترشحات غدد با رنگ آمیزی PAS واکنش مثبت داشتند. همچنین میزان استروژن تولید شده در هر دو مدل نسبت به گروه کنترل نرمال بیشتر و در مدل دوم نیز بیشتر از مدل اول بود (0/05≥P).

    نتیجه گیری

    در مجموع مدل آندومتریوزیس با بکارگیری سلول های آندومتر کشت شده، کارایی بیشتری را از نظر مرفولوژی و سطح غدد شکل گرفته و فعالیت استروژنی بالا نشان داد و می تواند در مطالعات اندومتریوزی از این مدل استفاده شود.

    سابقه و هدف

    CtxB باتحریک تولیدآنتی بادی علیهCtxB منجر به اثر بخشی واکسن شده است تلاشهای مختلفی در جهت افزایش تولید این آنتی بادی صورت پذیرفته شده است.کایتوسان یک پلی ساکاریدی است که قادر است به مخاط بچسبد این ماده با پتانسیل بسیار زیاد برای تحویل واکسن خوراکی با توجه به ویژگی های منحصر به فرد از جمله زیست سازگاری، زیست تخریب پذیری،چگالی بار بالاوغیرسمی می باشد.

    مواد و روش ها

    دراین مطالعه،از نانو کایتوسان، و نانو کپسول به عنوان یک سیستم حامل برای ارائه خوراکیCTXB مورد استفاده شد. ناوکایتوسان حاوی آنتی ژن توسط الکترواسپینینگ محلول کایتوسان / AcOH آماده شد. آنتی ژن در داخل نانوالیاف کایتوسان از طریق تجزیه و تحلیل طیف سنجی مادون قرمز(FTIR)مورد تایید قرار گرفت. خوکچه هندی توسط تلقیح مستقیم نانوالیاف کایتوسان حاوی آنتی ژن به داخل حفره دهانیایمن شدند. پاسخ های ایمونوگلوبولین سرم (G (IgGو ایمونوگلوبولینروده ای (A(IgA آنتی بادی تعیین شد

    نتایج

    خوکچه هندی بوسیله کایتوسان همراه با آنتی ژن و یا آنتی ژن به تنهایی ایمن شدند. نتایج بدست آمده حاکی از پاسخ بسیار زیاد آنتی ژن همراه نانوفیبر و نانوکپسول بود آنتی بادی IgA و IgGپس از تجویز خوراکیCtxBتولید نگردید. نتایج آنتی بادی تولید شده با استفاده از تست ANOVA و LSD مورد تجزیه و تحلیل گردید.

    نتیجه گیری

    نتایج این تحقیق نشان دهنده آن است که در روش خوراکی می توان با نانویی کردن آنتی ژن تیتر آنتی بادی را بالا برد.این سیستم می تواند برای انتقال دیگر آنتی ژن ها مورد استفاده قرار گیرد.

    هدف
    شباهت بیشتر به محیط درون تنی، به افزایش تکثیر و تمایز سلول ها در شرایط برون تنی کمک می کند. در این مطالعه، اثر محیط کشت پویا و وجود نانو ذرات هیدرو کسی آپاتیت nHA)) بر تکثیر و تمایز سلول های بنیادی مزانشیمی (MSCs) به سلول های استخوانی با استفاده از داربستهای الکتروریسی شدهٔ پلی کاپرولاکتون (PCL) بررسی شده است.
    مواد و روش ها
    ابتدا داربست های PCL و PCL-nHA به روش الکتروریسی تهیه شدند. پس از کشت ایستای داربست ها با MSCs، داربست ها در یک دوره 14 روزه در دو گروه کشت ایستا و پویا تقسیم شدند. داربست های کشت پویا بر روی همزن قرار گرفتند. تکثیر و تمایز سلول ها در روزهای 3، 7 و 14، با آزمایش های MTT، کلسیم و آلکالین فسفاتاز بررسی شدند.
    نتایج
    نتایج به دست آمده از بررسی MTT داربست ها در روز 14، افزایش 1/1 برابری میزان تکثیر سلول ها را در کشت پویا نسبت به ایستا نشان داد. همچنین در طی این دوره، میزان کلسیم تولیدی توسط سلول ها برای داربست های PCL و PCL-nHA در حالت پویا نسبت به ایستا در روز 14، به ترتیب 23/1 و 46/1 برابر بود. در بررسی فعالیت آنزیم آلکالین فسفاتاز، میزان فعالیت برای داربست های PCL و PCL-nHA در حالت پویا نسبت به ایستا در روز 14، به ترتیب 1/24 و 1/28 برابر بود.
    نتیجه گیری
    نتایج حاصل از کشت پویا، تکثیر و تمایز بیشتر سلول های بنیادی به استخوانی را برای هر دو نوع داربست PCL و PCL-nHA نسبت به ایستا نشان دادند. همچنین، میزان تکثیر و تمایز سلولی در داربست های حاوی nHA بیشتر از داربست های بدون آن بود.

    هدف
    دوزهای بالای استامینوفن ایحاد آسیب های کبدی می کند. فاکتور سلول بنیادی (SCF) و گیرنده آن c-kit باعث بهبود آسیب های کبدی ناشی از سمیت استامینوفن می شوند. مطالعه حاضر به بررسی اثر SCF بر فعالیت آنزیم های گلوتاتیون-اس- ترانسفراز (GSTs) در سمیت استامینوفن پرداخته است.
    مواد و روش ها
    45 سر موش نر نژاد بالبسی (Balb/c) به 3 دسته هر یک شامل 3 گروه (هر گروه 5 سر) تقسیم شدند:1) تزریق 300 میلی گرم / کیلوگرم وزن بدن استامینوفن درون صفاقی2) تیمار با 40 میکروگرم/ کیلوگرم وزن بدن SCF، 30 دقیقه پس از تزریق استامینوفن (300 میلی گرم / کیلوگرم وزن بدن) 3) تیمار با نرمال سالین (گروه کنترل).
    دسته ها به ترتیب پس از 1، 12 و 24 ساعت کشته شدند. ارزیابی سمیت با پاتولوژی کبد و سنجش مارکرهای سرمی آسیب کبدی (ALT و AST) در موش های دسته 24 ساعت انجام شد. میزان پروتئین c-kit، و فعالیت آنزیم های GST سنجش شد.
    نتایج
    مشاهدات هیستوپاتولوژی و افزایش معنی دار(p < 0.05) سطحALT و AST، سمیت کبدی ناشی از تزریق دوز 300 میلی گرم / کیلوگرم وزن بدن از استامینوفن را نشان داد. SCF باعث افزایش معنی دار فعالیت GSTs و کاهش معنی دار در میزان پروتئین c-kit پس از 24 ساعت شد.
    نتیجه گیری
    اتصال SCF به گیرنده آن (c-kit) در کبد باعث بهبود آسیب های کبدی ناشی از استامینوفن به واسطه افزایش فعالیت GSTs در کبد موش می شود.

    هدف
    سرطان سینه با شیوع حدودا یک میلیونی یکی از معمول ترین سرطان ها در میان زنان سراسر جهان بوده و هفت درصد مرگ و میرهای ناشی از سرطان را شامل می شود. چندین راهبرد ایمنی زایی و شناسایی زود هنگام با استفاده از آنتی ژن های سطحی سرطان سینه هم اکنون به صورت بالینی توسعه یافته است. در واقع سرطان سینه در اثر افزایش بیان بیش از حد یک سری از ژن ها حادث می شود. HER-2 به عنوان گیرنده تیروزین کینازی در خانواده گیرنده های فاکتور رشد اپیدرمی قرار دارد. نقش HER-2 در سرطان سینه بطور گسترده مورد مطالعه قرار گرفته است. HER-2 تنها در30-25 درصد از بیماران مبتلا به سرطان سینه شایع بوده و هدف درمانی HER-2 با آنتی بادی های انسانی مثل هرسپتین ثابت شده است. هدف از این تحقیق تولید پروتئین نوترکیب HER-2 به منظورتشخیص زود هنگام سرطان سینه می باشد.
    مواد و روش ها
    بخشی از ژن her-2 با طراحی پرایمر مناسب بوسیله ی PCR تکثیر شده و سپس درون ناقل پلاسمیدی pET28a جهت بیان کلون شد، پس از تائید فرایند کلونینگ با روش های هضم آنزیمی و تعیین توالی، بیان این ژن در باکتری E.coli با استفاده از IPTG القا شده و سپس پروتئین مورد نظر توسط ژل SDS-PAGE بررسی شد.
    نتیجه گیری
    بخشی از ژن her-2 انسانی به صورت نوترکیب در باکتری E.coli بیان گردید که می تواند یک عامل تشخیصی مناسب برای سرطان سینه باشد.

    Objective

    Prostate cancer is the fifth most common cancer. In 2012, it was the second leading cause of cancer death for men worldwide. The PI3K/AKT pathway plays an essential role in pathogenesis of prostate cancer; the key role of this pathway in cancer progression makes it an attractive target for prostate cancer therapy. MicroRNAs (miRNAs) that regulate gene expression have a special ability to simultaneously control multiple genes and pathways which make them candidates for therapeutics. This study aims to determine miRNAs which target the PI3K/AKT pathway and evaluate them in prostate cancer cell lines.

    Methods

    In order to determine an effective miRNA for the PI3K/AKT pathway, we assessed six genes from this pathway which have been proposed as drug targets in ten different prediction algorithms. Next, the candidate miRNAs were analyzed in expression profile and pathway analysis databases. Expression of candidate miRNAs in control and prostate cancer cell lines were subsequently evaluated.

    Results

    According to bioinformatics, the miR-29 family could target the most genes from this list. Other bioinformatic estimates confirmed these results. The miR-29 family showed significant downregulation in prostate cancer cell lines LNCAP, PC3 and DU-145 compared to control samples.

    Conclusion

    These results propose the possibility of using the miR-29 family to inhibit the PI3K/AKT pathway in prostate cancer.

    Objective
    Dendrimers are three-dimensional nanostructures that have numerous applications in medicine, including drug delivery and imaging. Although anionic dendrimer polyethylene glycol–citrate has a high potential to increase solubility of water-insoluble drugs and drug delivery, its multi-step synthesis procedure is time consuming. In addition, toxic substances such as dichloromethane are used in its synthesis procedure. In this study, we have developed a simple one-step synthesis method using green chemistry.
    Methods
    We examined four different methods to improve the synthesis method of this dendrimer. Products were characterized by FTIR, LC-MS and DLS. Cytotoxicity was assessed by the XTT method.
    Results
    We synthesized a G2 polyethylene glycol–citrate dendrimer in one-step without purifying G1. This process was chosen as a beneficial method for synthesis of the G2 dendrimer. When compared with previous methods, this procedure had higher efficiency and greatly reduced response. This procedure used nontoxic materials. XTT assay results showed that this dendrimer created by green chemistry had no cytotoxicity in Hela and Vero cells up to a concentration of 800 µM.
    Conclusion
    One-step synthesis of anionic polyethylene glycol-citrate G2 dendrimer is a simple, beneficial production method. The dendrimer is biocompatible and can be used as a suitable carrier for drug delivery purposes.

    Objective

    In order to improve the water solubility and bioavailability of curcumin in cancer therapy, we prepared and tested a novel waterborne cationic polyurethane (PU) as a nano-carrier for curcumin loading (CU-PU). We studied the effect of this prepared nano-drug on melanoma (F10B16) and fibroblasts cells (L929).

    Methods

    Morphology, size and cell internalization ability of the prepared nanoparticles were analyzed by zetasizer, SEM, AFM and fluorescent microscopy, respectively. We anticipated that curcumin was loaded in the hydrophobic core of the PU carrier. Next, the suitable dose and therapeutic effects of CU-PU for both skin cancer and normal cell lines were evaluated by the MTT assay and real-time PCR.

    Results

    The average diameters and polydispersity of the nanoparticles were 62.37 ± 1.7 nm and 0.080 ± 2.1 at 25 ̊C, respectively. The drug encapsulation efficiency was 87 ± 0.2%. The morphological analysis confirmed both a spherical shape and good dispersion without remarkable aggregation. The MTT assay results showed that the IC50 at 24 hours was 36.2 µM, whereas it was 25.4 µM at 48 hours. Real-time PCR results indicated that the CU-PU significantly decreased mRNA expressions of VEGF, Bcl-2, MMP-9 and COX-2 genes. An increase in mRNA expression of the BAX gene was also observed.

    Conclusion

    Our result provided acceptable evidence for cell proliferation inhibition and the apoptotic effect of CU-PU on skin cancer cells. There were no adverse effects detected for normal cells.

    Objectve: Endometriosis is a common disease in which the endometrial stroma and glands grow abnormally outside the uterine cavity. The establishment of animal models can be effective for determining the etiology of endometriosis. In this study we compare two endometriosis models using endometrial fragments and isolated cultured endometrial cells.

    Methods

    We obtained endometrial tissues that were in the proliferative or secretory phases from women who underwent hysteroscopies for benign reasons at Imam Khomeini Hospital (Tehran). Following confirmation of the tissue's normality, we cut the tissues into 2 mm cube pieces. The remainder of endometrial tissues were used for isolation and cultured to the fourth passage. In the first model of endometriosis, the endometrial tissue fragments and in the second model, 2×106 isolated endometrial cells were subcutaneously transplanted into gamma irradiated mice. The mice were kept under controlled, sterile conditions for 20 days. The mice sera were collected before and after transplantation for assessment of 17β estradiol. The ectopic tissues in both models were assessed for morphological staining using hematoxylin and eosin, as well as periodic acid Schiff (PAS) for gland secretion. The gland sections per mm2 were analyzed.

    Results

    At 20 days after tissue transplantation, we observed endometrial-like glands in the subcutaneous tissue of both endometriosis models. The number of gland sections was 57.55 ± 17.18/mm2 for the first model and 271.57 ± 77.98/mm2 for the second model. This result was significantly higher in the second model when compared to the first model. Gland secretion was positive for PAS. The level of 17β estradiol was higher in both models compared to the control group. This level was significantly higher in the second model compared to the first (P≤0.05).

    Conclusion

    The endometriosis model that used cultured endometrial cells showed more efficiency in morphology, gland formation and level of17β estradiol.

    Objectives

    CtxB contributes to a vaccine's efficacy by stimulating production of the anti-CtxB antibody. Various attempts have been made to increase production of this antibody. Chitosan is a mucoadhesive polysaccharide that has tremendous potential for oral vaccine delivery in terms of its exclusive features that include biocompatibility, biodegradability, high charge density and non-toxicity. We investigated the potential for chitosan nano­fibers and nanocapsules as novel carrier systems for the oral delivery of CtxB.

    Methods

    Antigen-containing chitosan nanofibers were prepared by electrospinning a chitosan/AcOH solution. Encapsulation of the antigen inside the chitosan nanofibers was confirmed through infrared spectroscopy analysis (FTIR). Guinea pigs were immunized with chitosan and antigen or antigen alone by direct administration of antigen-containing chitosan nanofibers into the buccal cavity. Serum immunoglobulin G (IgG) and intestinal immunoglobulin A (IgA) antibody responses were determined

    Results

    The results indicated that antigen in the chitosan nanofibers or nanocapsules elicited very high IgA and IgG responses. No detectable IgA and IgG responses were obtained after oral immuniza­tion with CtxB. The results of the antibody titer were analyzed using the ANOVA and LSD tests.

    Conclusion

    CtxB inside the nanofiber increased antibody production when administered orally. This system might be used for delivery of other antigens.

    Objective
    More similarity to in vivo medium may help to increase the proliferation and differentiation of cells at in vitro condition. The present study has investigated the effect of a dynamic mediumand nano hydroxyapatite (nHA) presence on proliferation and differentiation of mesenchymal stem cells (MSCs) to bone cells using electrospun polycaprolactone (PCL) scaffolds.
    Methods
    We prepared PCL and PCL-nHA scaffolds by electrospinning. After static culturing of the scaffolds with MSCs, the scaffolds in a 14-day period, they were divided into two groups of static and dynamic cultures. The dynamic culture scaffolds were placed on a shaker. Cell proliferation and differentiation at days 3, 7 and 14 were investigated by MTT, and the calcium and alkaline phosphatase assays.
    Results
    The obtained results from the MTT assay on day 14 showed an increase of 1.1 times in cell proliferation in the dynamic culture compared to the static culture. During this period, the calcium content produced by cells in the dynamic culture at day 14 were 1.23 times higher for the PCL scaffold and 1.46 times higher for the PCL-nHA scaffold compared to the static culture. Alkaline phosphatase levels for the dynamic state PCL scaffold were 1.24 times more and for the PCL-nHA scaffold they were 1.28 times more compared to the static culture at day 14.
    Conclusion
    The obtained results from dynamic culture, showed higher proliferation and differentiation of stem cells to bone for both PCL and PCL-nHA scaffolds compared to the static culture. The amount of cell proliferation and differentiation in the scaffolds that contained nHA was more than scaffolds that did not have nHA.

    Objective
    Acetaminophen (APAP) overdose causes acute liver injuries. Studies show that stem cell factor (SCF) and its receptor, c-Kit, enhance liver recovery from APAP-induced injuries in mice. In this study we explore the effect of SCF on activity of glutathione S-transferase (GSTs) enzymes which are considered to be important in APAP metabolism.
    Methods
    We divided 45 Balb/c mice into three groups. Within each group there were three sub-groups of five mice per subgroup. The groups included: 1. APAP (300 mg/kg B.W., i.p.); 2. SCF (40 µg/kg B.W., i.p.) given.30 minutes after APAP (300 mg/kg B.W., i.p.), and 3.control mice treated with normal saline. The mice were sacrificed at 1, 12 and 24 hours, respectively. Hepatotoxicity was evaluated in the 24 hour group by histopathology and assessment of biochemical serum markers (ALT and AST). We assessed the levels of SCF receptor (c-Kit) protein and GST enzyme activities in the liver tissues.
    Results
    Hepatotoxicity was induced by APAP (300 mg/kg, B.W) as evident by both histopathological observations and a significant (p<0.05) increase in serum ALT and AST levels, which were reversed by SCF administered post-APAP. SCF administration after APAP administration significantly increased GSTs enzyme activity levels by 24 hours, however it led to a significant decrease in c-Kit protein level compared to the control and APAP groups.
    Conclusion
    Our data suggest that SCF binding to its receptor (c-Kit) on liver cells may attenuate APAP-induced liver injuries by increasing GST activities in the livers of mice.
