    مقدمه
    امروزه در سلول درمانی و مهندسی بافت، به سلول های بنیادی مزانشیمی توجه خاصی می شود و جهت رسیدن به تعداد سلول مورد نظر با ویژگی های مطلوب، دستیابی به شرایط مناسب کشت ضرورت دارد. از این رو، پژوهش حاضر با هدف مقایسه ی تاثیر پلاسمای غنی از پلاکت (PRP یا Platelet rich plasma) و سرم جنین گاوی (FBS یا Fetal bovine serum) بر تکثیر و بقای سلول های بنیادی مشتق از چربی (ADSCs یا Adipose derived stem cells) در داربست طبیعی فیبرین انجام شد.
    روش ها
    نمونه های چربی از سه بیمار، طی جراحی لیپوساکشن به دست آمد. سلول های بنیادی از بافت چربی استخراج و کشت شد. سپس سلول های پاساژ سوم در داربست فیبرین و مدیوم حاوی PRP 10 درصد (گروه مورد) یا FBS 10 درصد (گروه شاهد) کشت داده شدند. میزان تکثیر و بقای سلول ها در روزهای چهارم و هشتم با روش MTT (diphenyl tetrazolium bromide assay 5،2 (yl-2- dimethyl thiazol-4،5)3 و رنگ آمیزی با Trypan blue و درصد آپوپتوز سلولی با روش Flow cytometry با استفاده از کیت Annexinᴠ-FITC ارزیابی گردید. اطلاعات جمع آوری شد و با استفاده از آزمون t مستقل مورد تجزیه و تحلیل قرار گرفت.
    یافته ها
    در روزهای چهارم و هشتم میزان بقا و تکثیر و درصد سلول های زنده در گروه مورد نسبت به گروه شاهد به طور معنی داری بیشتر و میزان سلول ها در مرحله ی اولیه ی آپوپتوز در گروه مورد نسبت به گروه شاهد به طور معنی داری کمتر بود (010/0 > P). میزان سلول ها در مرحله ی نهایی آپوپتوز، در روز 8 به طور معنی داری در گروه مورد کمتر از گروه شاهد بود (001/0 > P).
    نتیجه گیری
    تاثیر مثبت PRP بر روند بقا و تکثیر سلول های ADSCs در مقایسه با FBS قابل توجه است.

    مقدمه
    گلیکوپروتئین 3Tim (3T-cell immunoglobulin and mucin domain) یکی از نشانگرهای سطح سلولی سلول های 1Th (1T helper) است که در بسیاری از بیماری های مرتبط با سیستم ایمنی، تغییر بیان دارد. این مطالعه، با هدف بررسی و افزایش بیان پروتئین 3Tim در سطح سلول T293 انجام شد.
    روش ها
    کاست بیانی 3Tim از روی پلاسمید 67M-2682W-EX با استفاده از پرایمرهای دارای جایگاه آنزیمی NheI و MluI تکثیر و در جایگاه های مربوط در پلاسمید pHygro ساب کلون شد. پلاسمید نوترکیب محتوی ژن 3Tim (3pH-Tim) پس از استخراج و خالص سازی، با آنزیم NheI خطی شد و در سلول های T293 با استفاده از روش کلسیم فسفات ترانسفکت شد. سپس سلول های نوترکیب T293 بیان کننده ی 3Tim در محیط حاوی هیگرومایسین انتخاب مثبت شدند. پس از یک ماه بر روی DNA ژنومیک کلون های سلولی باقی مانده، واکنش PCR (Polymerase chain reaction) به منظور بررسی ادغام 3cDNATim (Complementary DNA) در ژنوم سلول T293 انجام شد. همچنین میزان بیان پروتئین 3Tim در سطح سلول به روش فلوسایتومتری ارزیابی گردید.
    یافته ها
    نتایج هضم آنزیمی با آنزیم های NheI و MluI بر روی پلاسمید 3p-H-Tim باند 2312 و 4600 جفت بازی را نشان داد که تایید کننده ی صحت کلونینگ است. در واکنش PCR بر روی DNA ژنومیک، باند 1100 جفت بازی تکثیر شد که نشانه ی ورود ژن 3Tim در ژنوم سلول T293 می باشد. همچنین در فلوسایتومتری، 88 درصد سلول ها از نظر بیان 3Tim مثبت بودند و شدت بیان در قسمت زیادی از سلول ها بالا بود.
    نتیجه گیری
    بیان پروتئین در سیستم های بیانی پروکاریوت ها از نظر زمان و هزینه بهتر از سیستم های یوکاریوتی است، اما در مورد پروتئین های دارای تغییرات پس از ترجمه، این سیستم بیانی جوابگو نیست. در برخی موارد ساختار فضایی طبیعی پروتئین در موقع لنگر انداختن بر سطح غشا، در تولید پروتئین های غشایی مانند 3Tim در سیستم های یوکاریوتی، حفظ نمی شود. در نتیجه، برای استفاده در تحقیقات تهیه ی آنتی بادی هایی که اپی توپ های فضایی را می شناسند و یا انتخاب آپتامر، مناسب نمی باشند. بیان پروتئین 3Tim در سطح سلول، می تواند ارایه دهنده ی پروتئین با ساختار فضایی طبیعی در پروژه های تولید آنتی بادی، نانوبادی و آپتامر باشد.

    مقدمه
    سرطان پستان شایع ترین تومور بدخیم و سرطان منجر به مرگ در زنان می باشد. برخی نشانگرهای زیستی به عنوان عوامل تخمین پاسخ دهی به درمان کاربرد دارند. مثال واضح و رایج در این مورد، گیرنده ی استروژن است و ارتباط معنی داری بین حضور گیرنده (ER+ یا Estrogen receptor) با احتمال ابتلا به سرطان پستان وجود دارد. هدف از این مطالعه، اندازه گیری بیان گیرنده ی استروژن α در زنان مبتلا به سرطان پستان بود.
    روش ها
    در این مطالعه، تعداد 20 نمونه ی بافت پارافینه از افراد مبتلا به سرطان و 10 نمونه ی سالم جمع آوری شد. پس از پارافین زدایی، استخراج RNA با محلول RNX Plus انجام گردید. cDNA (complementary DNA) با روش رونویسی معکوس به وسیله ی آنزیم M-MuLV (Moloney murine leukemia virus) سنتز شد. بیان ژن با روش Real time PCR (Real-time polymerase chain reaction) نسبی سنجیده شد و ژن گلیسر آلدهید فسفات دهیدروژناز (GAPDH یا phosphate dehydrogenase-3 Glyceraldehyde) نیز به عنوان شاهد داخلی استفاده گردید.
    یافته ها
    با مقایسه ی بیان ژن گیرنده ی استروژن α در بافت های سرطانی نسبت به نمونه های سالم، افزایش بیان در تمام نمونه های سرطانی بیشتر دیده شد. نتایج نشان داد که با افزایش مرحله ی بیماری، میزان بیان گیرنده ی استروژن α افزایش می یابد (0001/0 > P).
    نتیجه گیری
    میزان بیان ژن گیرنده ی استروژن α در نمونه های سرطانی به طور قابل توجهی افزایش می یابد. بنابراین، اندازه گیری بیان این ژن، می تواند به عنوان عامل موثر در تشخیص و پی گیری پاسخ دهی به درمان به کار رود.

    مقدمه
    تومورهای مغزی مرگ و میر بالایی را سبب می شوند. علاوه بر این، در درجه های توموری پیشرفته، قدرت تهاجم بالایی داشته، پیش آگهی بدتری دارند. تلومرها، توالی های تکراری و طویل DNA در دو انتهای کروموزوم های خطی هستند که از انتهای کروموزوم در مقابل نوترکیبی، اتصال و تخریب DNA محافظت نموده، از پیری سلولی جلوگیری می نمایند. در مهره داران، تلومرها از تکرار های پشت سر هم شش نوکلوتیدی TTAGGG تشکیل شده اند. تلومرها با وجود ساختار هتروکروماتینی خود، به صورت نوعی RNA غیر کد کننده ی موسوم به TERRA (Telomeric repeat-containing RNA) رونویسی می شوند و این RNA، به عنوان مهارکننده ی طبیعی تلومراز عمل می نماید. با توجه به این امر که آستروسیتوما از شایع ترین تومورهای دستگاه عصبی مرکزی است و پیش آگهی بالینی بدی دارد، هدف از این مطالعه، سنجش میزان بیان TERRA در تومورهای مغزی آستروسیتوما و نمونه های شاهد غیر توموری بود. به علاوه، بیان رونوشت TERRA در درجات توموری مختلف آستروسیتوما سنجیده شد.
    روش ها
    mRNA از 26 نمونه ی تومور آستروسیتوما و 4 نمونه ی شاهد غیرتوموری استخراج شد. DNA مکمل (cDNA) سنتز شد و سطح بیان TERRA با روش Real-time reverse transcription polymerase chain reaction با استفاده از کیت SYBR Green در درجات مختلف توموری به همراه نمونه های طبیعی ارزیابی شد.
    یافته ها
    بر اساس مقایسه ی بیان نسبی ژن TERRA بین درجات توموری مختلف در آستروسایتوما، میانگین ∆Ct در درجات بالای توموری (III و IV) نسبت به درجه ی پایین (II) کاهش نشان داد و این امر حاکی از کاهش بیان TERRA در درجه های بالای آستروسایتوما به میزان 377/4 برابر نسبت به درجه ی پایین بود (036/0 = P). افزون بر این، اندازه گیری بیان TERRA در آستروسایتوما به میزان 969/4 برابر، کاهش بیان در مقایسه با نمونه های شاهد غیرتوموری نشان داد (029/0 = P).
    نتیجه گیری
    مطابق این بررسی و سایر مطالعات انجام شده در این مورد، احتمال می رود، TERRA بتواند به عنوان یک نشانگر پیش آگهی مطرح شود.

    مقدمه
    میلیوم کلوئید یک وضعیت نادر پوستی با حداقل سه نوع متمایز است. از نظر بالینی، بیماری با ظهور پاپول های گسترده، نیمه شفاف و مایل به رنگ زرد و یا پلاک های تشکیل شده بر روی پوست آفتاب دیده، و از نظر بافت شناسی، با شناسایی کلوئید در ناحیه ی پاپیلای درم قابل تشخیص است.
    معرفی بیمار: بیمار مردی بود با پاپول های کوچک و متعدد بر روی پشت دست ها که بیماری میلیوم کلوئید توسط پاتولوژیست برای وی تایید شده بود. میلیوم کلوئید اغلب در بیماران با پوست روشن مشاهده می شود و بدون تغییر باقی می ماند. اما این بیمار، پوست به نسبت تیره از نوع Fitzpatrick skin type III داشت و ضایعات او در تابستان افزایش و در زمستان کاهش پیدا می کرد.
    نتیجه گیری
    این مورد، تاکیدی بر رابطه ی نزدیک بین قرار گرفتن طولانی مدت در معرض آفتاب و ابتلا به میلیوم کلوئید در بزرگ سالان است.

    Background
    Today, stem cells are the best candidate for cell therapy and tissue engineering. To achieve intended number of cells with optimal characteristics, it is necessary to have appropriate condition of culture. Therefore, in this study, we compared the effects of platelet rich plasma (PRP) and fetal bovine serum (FBS) on proliferation and survival of adipose-derived stem cells (ADSCs) in a natural fibrin scaffold.
    Methods
    Adipose tissue specimens were obtained from 3 patients undergoing liposuction surgery. adipose-derived stem cells were isolated from adipose tissue and cultured and encapsulated in fibrin scaffolds in third passage with culture media containing PRP-10% (experimental group) or FBS-10% (control group). Then, the rate of viability and survival were evaluated using MTT [3-(4, dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide] assay and Trypan blue staining. Percentage of apoptotic cells was evaluated via flow cytometry using Annexinᴠ-FITC kit on fourth and eighth days. Data were collected and statistical analyzed using t-test.
    Findings
    The rates of viability, survival and percentage of living cells in the experimental group were significantly higher than control group on fourth and eight days (P < 0.010 for all). Percentages of early apoptotic cells in the experimental group were significantly lower than control group on fourth and eight days (P < 0.001 for both). On the eighth day, percentage of late apoptotic cells in experimental group was significantly lower than control group.
    Conclusion
    This study showed that the positive effect of platelet rich plasma on the survival and proliferation of adipose-derived stem cells in compared with fetal bovine serum was significant.

    Background
    T-cell immunoglobulin and mucin domain-3 (Tim3) is known as a marker of cell surface of T-helper-1 (Th1) cell and has a key role in many diseases with cell-mediated immunity. Over expression of Tim3 has reported in autoimmune and atopic diseases. Though, many studies done about this inhibitory help protein molecules, but yet need more research. In research project on Tim3, native protein with proper post translation modification should be provided. In this study, Tim3 was expressed on the surface of HEK293 T-cell line.
    Methods
    Tim3 expression cassette was amplified from cDNA clone EX-W2682-M67 via polymerase chain reaction (PCR). The PCR product and pHygro plasmid were digested by NheI and MluI restriction enzymes. The linearized pHygro and digested PCR product were ligated together with T4DNA ligase and transformed into Escherichia coli TOP 10 F'. The resulted pH-Tim3 plasmid was linearized with NheI and transfected into 293T cell line. The transfected cells were positive selected with hygromycin and their genomic DNA was extracted and PCR was done on them to amplify complementary DNA (cDNA) of Tim3. Also, protein expression levels were assessed via flow cytometry.
    Findings
    The result of PCR on selected cells confirmed integratioin of cDNA clone of Tim3 in genomic DNA. Based on flow cytometry, about 88% of cells were expressed Tim3 sharply.
    Conclusion
    In order to understand the role and mechanism of Tim3 protein, we need a large amount of purified native Tim3. Prokaryotic expression systems are simpler and cheaper than eukaryotic expression system, but they are not proper system for expression of proteins that modified after translation. Expression of membrane proteins like Tim3 on the surface of cell has even more native conformation in comparison with recombinant Tim3. The cells display Tim3 could be used in antibody, nanobody or aptamer production projects. In this project, we constructed a 293T cell which overexpressed Tim3 on its surface compared to untransfected ones.

    Background
    Breast cancer is the most common malignant tumor and cause of cancer death in women. Some biomarkers are used to estimate the response to therapy. One of the most common indicators is estrogen receptors (ER). It seems that there is a significant correlation between receptor existence (ER +) and risk of breast cancer. This study aimed to measurement the estrogen receptor alpha expression in women with breast cancer.
    Methods
    In this study, 20 cancerous paraffin-embedded tissue blocks from patients with breast cancer and 10 non-cancerous were collected. After deparaffinization, RNA was extracted with RNX Plus solution. Complementary DNA (cDNA) was synthesized via reverse transcription reaction using Moloney murine leukemia virus (MMULV) enzyme. Estrogen receptors gene expression was measured using relative real-time polymerase chain reaction (real-time PCR) reaction. In this study, glyceraldehyde-3-phosphate-dehydrogenase (GAPDH) was used as internal control gene.
    Findings
    Estrogen receptor alpha expression in cancerous tissues had significant over expression compared to normal tissues. Also, increasing the stage of disease, expression of estrogen receptor alpha was increased (P < 0.0001).
    Conclusion
    According to the results, it can be concluded that the estrogen receptor alpha gene expression strongly increases in tumor samples. So, determination of estrogen receptor alpha expression can be used as indicator agent for response to treatment and follow-up.

    Background
    Brain tumors include burden of mortality from cancers. High grades of brain tumors are more aggressive and have poor prognosis. Introduction telomeres are the ends of linear chromosomes that serve as a protective cap to avoid permanent proliferation arrest, termed replicative senescence. In vertebrates, telomeres consist of tandem repeats of TTAGGG hexanucleotide sequences. In spite of heterochromatin structure of telomeres, they are transcribed into a non-coding RNA called telomeric repeat-containing RNA or TERRA which acts as a natural inhibitor of telomerase activity. Considering astrocytoma, as one of the most common tumors of the central nervous system (CNS), and a very poor prognosis tumor, the aim of this study was to evaluate the TERRA expression level in astrocytoma tumors and nontumoral controls. Furthermore, expression levels of TERRA were compared between different grades of astrocytoma.
    Methods
    The mRNA of 26 brain tumor samples and 4 samples as nontumoral controls were extracted and cDNA was synthesized. Then, real-time reverse transcription polymerase chain reaction (SYBR Green kit) for quantitation of total TERRA levels was developed.
    Findings
    We demonstrated the correlation between total TERRA levels of expression with different grades of astrocytoma. High grades (III and IV) of astrocytoma tumors had lower mean of ΔCt than low grades (II) and down-regulation for TERRA mRNA was 4.377-fold (P = 0.036). Additionally, measurement of TERRA expression in astrocytoma showed 4.969-fold less compared to levels of TERRA expression in nontumoral controls (P = 0.029).
    Conclusion
    According to our study, TERRA may be prognostic marker in astrocytoma tumors.

    Background
    Colloid milium is a rare cutaneous condition with at least, three distinct subtypes, characterized clinically by the development of yellowish translucent papules or plaques on sunexposed skin, and histologically by the presence of colloid in the dermal papillae.Case Report: We present a man with multiple small papules on dorsum of his hands pathologically confirmed to be colloid milium. Colloid milium is more commonly observed in fair-skin patients and remain unchanged; however, our patient had dark skin type (Fitzpatrick skin type III) and lesions were increasing in summer and decreasing in winter.
    Conclusion
    This case emphasizes the close relationship between long-term sun exposure and adult colloid milium.
