    بیماری پوسیدگی ریشه و زوال خربزه ناشی از قارچ Monosporascus cannonballus یکی از مهم ترین بیماری های قارچی خربزه در ایران می‫باشد. تاکنون اطلاعی از ساختار ژنتیکی جمعیت های قارچ عامل بیماری در ایران در دست نمی‫باشد. در این بررسی، جدایه های M. cannonballus از بوته های آلوده طالبی، خربزه و هندوانه از مناطق مختلف کشور مانند اصفهان، یزد، تهران، فارس، سمنان، مرکزی، خراسان رضوی، قزوین، سیستان و بلوچستان جمع آوری گردیدند. تنوع ژنتیکی50 جدایه با استفاده از نشانگرrep-PCR و آغازگر های B OX و ERIC با تکثیر قطعاتی از DNA تعیین گردید. اندازه قطعات تکثیر شده بین bp100 تا bp1500در نوسان بود. میانگین تنوع ژنتیکی در بین جدایه های این بیمارگر در کشور 32/0 محاسبه شد. تجزیه خوشه‫ای با استفاده از ضریب تشابه دایس و روش UPGMA وجود 100 درصد تشابه را بین برخی از جدایه ها که از مناطق مختلف جمع آوری شده بودند نشان داد. از نظر ویژگی های مورفولوژیکی نظیر شکل و رنگ پرگنه جدایه ها در محیط کشت و تنوع ژنتیکی مولکولی آنها ارتباط وجود داشت به طوری که بر این اساس و در سطح تشابه 80 درصد جدایه ها در سه گروه مختلف قرار گرفتند. در سطح تشابه70 درصد جدایه ها براساس نوع میزبان در دو گروه مختلف قرار گرفتند. قدرت تمایز بالای این نشانگر نشان داد که می توان از rep-PCR به عنوان یک روش مفید و سریع برای بررسی تنوع ژنتیکی جدایه های M. cannonballus استفاده کرد.

    برای دستیابی به روش ساده و حساس جهت ردیابی قارچ Rosellenia necatrix عامل پوسیدگی رزلینیایی ریشه در خاک و ریشه، مقایسه ای بین روش های کشت مستقیم خاک و ریشه آلوده در محیط کشت های، MEAR MEAR+ پنکونازول و MEAS، تکنیک Nested PCR و تله گذاری صورت گرفت. استخراج مستقیم DNA از خاک و ریشه ها انجام شد و ردیابی بیمارگر با استفاده ازآغازگرهای اختصاصیR2، R8 و R7، R10 در Nested PCR انجام شد. به منظور بررسی رابطه بین میزان مایه زنی اولیه و زمان لازم بعد از مایه زنی بیمارگر برای ردیابی و بروز علائم بیماری، آزمایشی در دو تکرار انجام شد، تکرار اول مقدار 2، 4، 8، 16، 32 گرم بذر گندم آلوده به عنوان زاد مایه اولیه به خاک اضافه و سپس گیاه باقلا به عنوان گیاه آزمون کشت شد. نمونه برداری در یک ماه و هر هفت روز یک بار انجام شد. درتکرار دوم مقدار مایه زنی 25/0، 5/0، 1، 2، 4، 8، 16، 32 گرم بذر گندم آلوده و زمان های نمونه برداری 0، 2، 4، 8، 16، 32، 64 و 128 روز تغییر یافت. نتایج نشان داد با افزایش مایه تلقیح، زمان لازم جهت ردیابی و بروز علائم بیماری کاهش می یابد، هم چنین مشخص گردید بیمارگر قبل از این که روی میزبان ایجاد بیماری کند به وسیله Nested PCR قابل ردیابی است. روش Nested PCR از سایر روش های به کار برده شده مناسب تر و سریع تر تشخیص داده شد.

    دی اکسی نیوالنول (DON) یکی از مهم ترین سموم تولیدشده توسط قارچ Fusarium graminearum در دانه های گندم آلوده می باشد. جایگاه عمل DON زیر واحد S60 پروتئین ریبوزومی L3 RPL3)) بوده و تغییرات نقطه ای در اسید آمینه های RPL3 در مخمر باعث ایجاد مقاومت به آن شده است. بررسی توالی های کدکننده RPL3 نشان داده است که هیچ تفاوتی میان اسید آمینه های ارقام حساس و مقاوم گندم وجود ندارد. هدف اصلی از این تحقیق، تشخیص تغییرات احتمالی ایجاد شده در سطوح رونوشت ژن های RPL3 (انواع A و B) تحت تنش بلایت فوزاریومی سنبله در گندم است. بدین منظور، از خوشه های آلوده به قارچ بیماری زا و غیرآلوده سه رقمSumai3، Frontana و فلات در بازه های زمانی یک، سه و هفت روز بعد از آلودگی، استخراج RNA انجام شد. آزمون های RT-PCR نیمه کمی و کمی همراه با آغازگرهای اختصاصی(انواع A و B) برای تشخیص تفاوت در سطح رونوشت این ژن ها بین ارقام حساس و مقاوم انجام گرفت. اطلاعات به دست آمده کاهش در سطح بیان ژن در گیاه آلوده نسبت به غیرآلوده در ارقام Sumai3 و فلات طی 24 ساعت اولیه بعد از آلودگی را نشان می دهد. از طرفی، افزایش در سطح بیان ژن RPL3A در بازه زمانی هفت روز پس از آلودگی در رقم Frontana می تواند بیانگر نقش مشارکت این پروتئین خاص در بروز مقاومت در رقم فوق باشد.

    گونه های P. citrophthora و P. nicotianae از خاک و بافت های آلوده مرکبات جداسازی، خالص سازی و شناسایی شده، نقشه پراکنش آنها در استان مازندران تهیه شد. میانگین تراکم جمعیت دوگونه براساس زادمایه در گرم خاک محاسبه شد. در 8/82 % از باغات نمونه برداری شده گونه P. citrophthora و در 9/85% از آنها گونهP. nicotianae ردیابی شد. 17% از باغات کل استان هر دو گونه، 8/32% باغات گونه P. citrophthoraو 39% از آنها گونه P. nicotianae را با تراکمی بالاتر از 10 زادمایه در هر گرم خاک دارا بودند. میانگین تراکم جمعیتP. citrophthora و P. nicotianae برای کل باغات نمونه برداری شده استان به ترتیب 08/13 و 30/11 زادمایه در هر گرم خاک تعیین شد. بیماری زایی ده جدایه از گونه های P. citrophthora و P. nicotianae روی ریشه گیاهچه های دوازده ماهه سیترنج، سیتروملو، پرتقال شهسوار و لیموشیرین بررسی شد. تجزیه و تحلیل داده های مربوط به وزن تر ساقه و ریشه و شاخص بیماری در سیترنج عمدتا سه گروه را شامل شاهد، جدایه های P. nicotianaeو جدایه های P. citrophthora متمایز کرد که گروه اخیر مهاجم تر از گروه دوم بود. داده های مربوط به وزن تر ساقه و ریشه سیتروملو و پرتقال شهسوار گروه های آماری با تفاوت معنی دار بین جدایه های P. citrophthora و P. nicotianae متمایز نکردند. داده های مربوط به شاخص بیماری سیتروملو و پرتقال شهسوار به طور کامل جدایه های P. citrophthoraو P. nicotianae را از هم جدا کردند؛ جدایه های P. citrophthoraبه طور معنی داری بیماری زاتر از جدایه های P. nicotianae روی گیاهچه های سیتروملو و پرتقال شهسوار بودند. داده های مربوط به وزن تر ساقه و ریشه و شاخص بیماری لیمو شیرین گروه های آماری با تفاوت معنی دار بین جدایه های P. citrophthora و P. nicotianae متمایز نکردند.

    بیماری سفیدک پودری که توسط انگل اجباری Erysiphe necator (مترادف با Uncinula necator) ایجاد می گردد مهم ترین بیماری قارچی در مو به لحاظ اقتصادی است. تقریبا تمامی ارقام تجاری گونه Vitis vinifera به علت اینکه طی روند تکاملی در معرض این بیمارگر نبوده اند به این قارچ بسیار حساس هستند. در این پژوهش روند جوانه زنی قارچ سفیدک پودری مو روی دو ژنوتیپ حساس و مقاوم مو و هم چنین مقاومت انتوژنیک، در بافت های رویشی انگور رقم کابرنت ساویگنون و Arabidopsis thaliana با سنین مختلف مورد بررسی قرار گرفت. 12-2 ساعت پس از آلودگی در هر دو ژنوتیپ حساس و مقاوم، کنیدیوم ها شروع به جوانه زنی کرده و اولین علایم ظهور آپروسوریوم و هوستوریوم در هر دو ژنوتیپ قابل مشاهده بود. نمو ریسه های اولیه 12 ساعت پس از آلودگی در هر دو ژنوتیپ آغاز گردید اما در ژنوتیپ مقاوم پس از 12 ساعت ته نشست های قهوه ای- زرد رنگ در اطراف سلول های اپیدرمی قابل مشاهده بود. در کابرنت ساویگنون حساس نمو ریسه های اولیه به طور طبیعی تا 24 ساعت ادامه داشت اما در ژنوتیپ مقاوم پس از 24 ساعت واکنش فوق حساسیت مشخصا رخ داد و از گسترش ریسه های ثانویه ممانعت نمود در حالی که در کابرنت ساویگنون واکنش فوق حساسیت معنی داری مشاهده نشد. با افزایش سن فیزیولوژیک بافت به دلیل افزایش مقاومت انتوژنیک، میزان کارایی نفوذ قارچ E. necator در رقم کابرنت ساویگنون و قارچ Golovinomyces cichoracearum در آرابیدوپسیس کاهش یافت.

  • کنترل بیولوژیک بیماری کپک خاکستری سیب با استفاده از Candida membranifaciens، Rhodotorula mucilaginosa و Pichia guilliermondii

    جدایه های مخمرهای Candida membranifaciens، Rhodotorula mucilaginosa و Pichia guilliermondii که از میوه های سالم سیب جداسازی شدند برای کنترل بیولوژیک بیماری کپک خاکستری سیب با عامل Botrytis mali و B. cinerea در آزمایشگاه مورد ارزیابی قرار گرفتند. متابولیت های فرار تولید شده توسط تمام جدایه های مخمر به ترتیب 7/82 تا 7/97 درصد و 2/81 تا 3/93 درصد از رشد میسلیومی B. mali و B. cinereaجلوگیری کردند. تمام مخمرها مساحت پوسیده شده سیب توسط عامل بیماری را به طور معنی داری کاهش دادند. C. membranifaciens A2، A4 و A5 و P. guilliermondii A6 بیشترین تاثیر را در کاهش پوسیدگی در دماهای 4، 20 و 3±20 درجه سانتی گراد دارا بودند. جمعیت C. membranifaciens A2 و C. membranifaciens A5 در طول انجام آزمایش در داخل زخم های سیب در دمای 4 درجه سانتی گراد افزایش یافت.

    به منظور بررسی اثر باکتری های فراریشه گیاهان بر فعالیت نماتود ریشه گرهی، گونه Meloidogyne javanica، از خیار، سرو و گوجه فرنگی آلوده به نماتود مذکور از مناطق شیراز، مرودشت و گرمسار و هم چنین سرو، چمن و گندم سالم از باجگاه نمونه برداری شد. نماتودهای جداسازی شده در گلخانه روی گوجه فرنگی رقم شفت فلات خالص سازی و تکثیر گردید. ابتدا 40 سویه باکتری جدا شده از فراریشه گیاهان آلوده و یا سالم بر میزان تفریخ تخم پس از سه و هشت روز و هم چنین فعالیت لاروهای سن دوم، پس از 24، 48 و 72 ساعت، در شرایط آزمایشگاه بررسی گردید. سپس از میان سویه های موثر، ده سویه انتخاب و در شرایط گلخانه روی خیار رقم سوپرآملیا در دو آزمایش مستقل مورد مطالعه قرار گرفت. نتایج نشان داد که تمامی سویه ها موجب افزایش رشد گیاه و کاهش شاخص های مربوط به نماتود شدند. در این میان سویه Pseudomonas fluorescens CHAO با کاهش 3/55% و 7/55% تعداد گال و کاهش 1/91% و 8/82% فاکتور تولیدمثل، به ترتیب در آزمایش اول و دوم، به عنوان موثرترین سویه شناخته شد. سویه ای از Pantoea sp. در رتبه بعدی قرار گرفت. هم چنین تاثیر سه سویه از باکتری های fluorescens CHAO. P، Bacillus subtilis و Pantoea sp. به صورت جداگانه و در ترکیب با هم روی دو رقم خیار سوپرآملیا و رویال بررسی شد. براساس نتایج به دست آمده کاربرد توام هر سه باکتری نسبت به استفاده از هریک به صورت جداگانه و یا در ترکیب های دوتایی اثر بیشتری در افزایش شاخص های رویشی گیاه و کاهش شاخص های مربوط به نماتود داشت.

    قارچ Colletotrichum coccodes و باکتری Ralstonia solanacearum از جمله عوامل بیماری زای مهم سیب زمینی در استان همدان هستند. در این پژوهش اثر متقابل این دو بیمارگر با استفاده از دو روش آغشته سازی، روی سه رقم سیب زمینی شامل آگریا، بورن و دیامانت مورد بررسی قرار گرفت. آنالیز نتایج حاصل با نرم افزار SAS نشان داد که حضور هم زمان دو بیمارگر در گیاه روی یکدیگر اثر تشدیدکننده (هم افزایی) داشته و طول، وزن تر و خشک ساقه و ریشه را بیشتر کاهش می دهند. درصد پرگنه سازی ریشه گیاه توسط میکرواسکلروت قارچ C. coccodes در رقم آگریا نسبت به سایر ارقام مورد بررسی بیشتر و بین 50 تا 70 درصد بود. در این رقم وجود قارچ C. coccodes باعث افزایش خسارت باکتری شد. در مجموع رقم آگریا نسبت به سایر ارقام مورد بررسی در برابر قارچ C. coccodes حساس تر بود. در رقم بورن حساسیت گیاه نسبت به دو عامل بیماری زای فوق یکسان و پرگنه سازی ریشه توسط میکرواسکلروت های قارچ 50 درصد بود. در رقم دیامانت در بیشتر موارد تیمار آلوده به باکتری از لحاظ آماری با تیمار شاهد (گیاه غیرآلوده) در یک گروه قرار گرفت که نشان دهنده توان بیماری زایی کم باکتری روی این رقم و وجود مقاومت نسبی است. به علاوه، پرگنه سازی ضعیف ریشه سیب زمینی در رقم دیامانت توسط میکرواسکلروت قارچ C. coccodes گویای مقاومت نسبی این رقم در برابر قارچ عامل بیماری خال سیاه سیب زمینی است.

    بیماری فیلودی کاهو در فارس که ناشی از یک فیتوپلاسما از گروه جاروک نخود کبوتر (pigeon pea witches'' broom) می باشد در طبیعت توسط زنجرک Neoaliturus fenestratus منتقل می شود. خصوصیات انتقال این فیتوپلاسما با زنجرک مزبور شامل زمان گیرش، دوره کمون و زمان دهش مورد بررسی قرار گرفت. حداقل مدت گیرش چهار ساعت بود و با اضافه شدن زمان دسترسی زنجرک سالم به کاهوی آلوده به فیتوپلاسمای عامل بیماری راندمان انتقال بالا رفت. حداقل دوره نهفتگی عامل بیماری در بدن زنجرک ناقل 14 روز بود. راندمان انتقال 32 روز پس از شروع تغذیه به حداکثر رسید و سپس سیر نزولی داشت. حداقل مدت دهش چهار ساعت بود و با اضافه شدن مدت زمان دهش راندمان انتقال بالا رفت. نتایج به دست آمده شبیه نتایجی است که در مورد ویژگی های انتقال فیتوپلاسماهای دیگر و Spiroplasma citri با ناقل های خود گزارش شده است. براساس نتایج این تحقیق فیتوپلاسمای عامل فیلودی کاهو با زنجرک ناقل مانند سایر فیتوپلاسماها رابطه تکثیری دارد.

    به منظور تعیین اثر رقم چغندرقند و روش آلوده شدن گیاهان در مقاومت به جدایه ایرانی ویروس پیچیدگی شدید بوته چغندر (BSCTV-Ir) مطالعه ای در قالب طرح آماری کاملا تصادفی روی 18 رقم چغندرقند انجام شد. از هر رقم 18 بوته در 6 گلدان کشت داده شد. برای مایه زنی ارقام از همسانه عفونت زای BSCTV-Ir استفاده و ارقام با دو روش مایه زنی با آگروباکتریوم (Agroinoculation) (آزمایش اول) و انتقال با زنجرک ناقل (Circulifer haematoceps) ویروس دار (آزمایش دوم) مایه زنی شدند. در آزمایش اول پس از 21 روز و 35 روز و در آزمایش دوم پس از 21 روز از زمان مایه زنی میزان همانندسازی ویروس در گیاهان با انجام واکنش زنجیره ای پلی مراز (PCR) مورد بررسی قرار گرفت و شدت علائم بیماری در 4 درجه شامل درجات صفر (فاقد علائم)، 1 (علائم خفیف)، 2 (علائم متوسط) و3 (علائم شدید) یادداشت شد. تجزیه و تحلیل داده ها در آزمایش اول نشان داد تنها رقمی که تمام گیاهان تیمار در آن آلوده به ویروس بوده و علائم شدید نشان دادند رقم Brigita بود. در نتیجه این رقم به عنوان رقم شاهد حساس در نظر گرفته شد و سایر ارقام با آن مقایسه شدند. تجزیه آماری داده ها از نظر شدت بیماری نشان داد که بین ارقام از نظر مقاومت به بیماری با رقم شاهد تفاوت معنی دار وجود دارد. بر این اساس هیچیک از ارقام مقاوم نبودند و ارقام از نظر میزان حساسیت به 3 گروه شامل متحمل (7233، H5505، BR1، HM1990 و FIMMA)، حساس (رسول، افشاری، هیبرید بالک شیراز، زرقان، P.P.8، P.P.22، Dorothea، Rhizofouret، IC، Flores، Hilma و Polyrow) و خیلی حساس (Brigita) طبقه بندی شدند. در نهایت با مقایسه داده های حاصل از مایه زنی گیاهان چغندرقند با دو روش فوق الذکر مشخص گردید میزان آلودگی به ویروس در انتقال با زنجره ناقل ویروس دار در اکثر ارقام کمتر از روش مایه زنی با آگروباکتریوم است.

    زوال درختان چنار سال هاست که به طور گسترده در شیراز و دیگر نقاط کشور وجود دارد. از 50 اصله درخت چنار با علائم زوال و سرخشکیدگی، از 41 درخت قارچ های C ytospora (%85/42)،Alternaria (%9/18)، Aspergillus (%71/12)، Nattrassia mangiferae (%22/3)، Penicillium (%05/3)، Rhizopus (%7/0)، Helminthosporium (%24/0)، Chaetomium (%21/0)، Coniothyrium (%21/0) و Ulocladium (%07/0) جداسازی شد. در این تحقیق از شاخه های بریده چنار برای اثبات بیماری زایی جدایه های قارچی که درصد فراوانی بالایی داشتند استفاده شد. به استثنای Nattrassia mangiferae و چند جدایه Cytospora دیگر جدایه های مایه زنی شده هیچکدام بیماری زا نبودند. مایه زنی 10 جدایه بیماری زای Cytospora به شاخه های بریده سیب، گردو، نارنج، ارغوان، توت سفید، کاج، افرای شبه چناری، زبان گنجشک و چنار صورت گرفت که تنها یک جدایه روی توت و پنج جدایه روی ارغوان علائم بسیار جزیی ایجاد کردند. براساس خصوصیات ریخت شناسی، جدایه های سیتوسپورا از درختان چنار در شیراز گونه Cytospora theryana تشخیص داده شدند. در این گونه دو گروه مورفولوژیکی Cytospora و Cytophoma تشخیص داده شد. نتایج حاصله از آزمایشات صورت گرفته روی قابلیت هدایت الکتریکی (EC) عصاره اشباع، پ هاش و بافت خاک تا عمق 40 سانتی متری خاک اطراف 40 نمونه درختان چنار سالم و دارای علائم زوال نشان داد که بافت خاک درختان سالم و در حال زوال یکسان، EC بین ds/m 4/2-2/0 و پ هاش خاک درختان سالم و درختان دارای علایم زوال نیز قلیایی و بین 7/8-16/8 بود. با توجه به نتایج فوق قارچ های جدا شده نقشی در زوال نداشته و تصور می رود که با توجه به پ هاش بالای خاک، کمبود عناصر غذایی قابل جذب نقش مهمی در زوال درختان چنار در شهرستان شیراز دارد.

    در این تحقیق میزان مقاومت 14 رقم تجارتی سیب درختی شامل: رد دلیشز، گلدن دلیشز، گلاب کهنز، گلاب اصفهان، سلطانی، گلاب کرمانشاه، گلاب خراسان، آزایش، برابورن، فوجی، جاناتان،گالا، اینگرید ماری و دلبار استیوال در قالب طرح بلوک های کامل تصادفی نسبت به سفیدک پودری مورد ارزیابی قرار گرفت. هر تیمار دارای سه تکرار ودر هر کرت سه اصله درخت کشت گردید، فاصله بین تکرارها 5/2 متر و بین نهال ها در هر کرت نیز یک متر در نظر گرفته شد. ارزیابی بیماری براساس میزان آلودگی سطح برگ ها بود و در زمانی که علائم بیماری به حداکثر توسعه خود رسیده بودند انجام شد و تمام ارقام به سه گروه طبقه بندی گردیدند. تمام ارقام مورد آزمایش با شدت های مختلف آلوده شدند. ارقام فوجی، گلدن دلیشز، گالا، آزایش، برابورن و دلبار استیوال به ترتیب با شاخص های 12/30؛ 92/37؛ 71/31؛2/33؛ 91/42 و78/25 نسبت به بیماری حساس(Susceptible) می باشند. در حالی که ارقام جاناتان،گلاب اصفهان، گلاب کرمانشاه، گلاب خراسان وگلاب کهنز در دوسال به ترتیب با شاخص های آلودگی 96/54؛57/55؛71/58؛68/53 و 42/59 بیشترین حساسیت(Highly susceptible) را نسبت به بیمارگر دارند و کمترین میزان آلودگی متعلق به ارقام رد دلیشز و اینگرید ماری با شاخص آلودگی 04/9 و 44/11 می باشد. این ارقام در این طبقه بندی جزو مقاوم ها(Resistance) قرار گرفتند.

    در بهار 1388 میوه های گلابی دارای علائم پوسیدگی و توده های اسپور نارنجی فراوان در استان گیلان (رشت) مورد بررسی و مطالعه قرار گرفتند. صفات ریخت شناسی مانند شکل و اندازه طولی و عرضی کنیدیوم و چنگک، امکان تولید خار و سختینه، رنگ و میزان رشد رویشی پرگنه بررسی شد. هم چنین، DNA ژنومی از میسلیوم قارچ به روش Chelex 5% استخراج گردید و ناحیه ITS از rDNA با آغازگرهای ITS1 و ITS4 تکثیر و توالی یابی شد. براساس مشخصات شکل شناسی و ویژگی های مولکولی این قارچ به عنوان گونهColletotrichum acutatum شناسایی شد. بیماری زایی این گونه روی میوه های زخمی و فاقد زخم گلابی با مایه زنی به روش سوسپانسیون کنیدیوم به اثبات رسید. براساس نتایج این بررسی، گلابی به عنوان میزبان جدید گونه مذکور در ایران معرفی می گردد و برای اولین بار توالی ناحیه ITS rDNA این گونه روی میزبان گلابی از ایران منتشر می شود.

    بیماری پیچیدگی بوته چغندرقند یکی از عوامل خسارت زای کشت چغندرقند به حساب می آید و به دلیل ایجاد اپیدمی های شدید، وسیع و غیرقابل پیش بینی تا به حال مورد توجه زیادی قرار گرفته است. مشاهدات گذشته نشان داده است که این بیماری به دلیل کوتاه بودن دوره تولیدمثل، فراوانی بالای زنجرک های ناقل و توانایی تغذیه آنها از گونه های متنوع گیاهی، در مدت کوتاهی به سرعت در یک منطقه پخش می شود. حداقل پنج گونه ویروس متعلق به جنس Curtovirus (تیرهGeminiviridae) به عنوان عوامل ایجادکننده بیماری پیچیدگی بوته چغندرقند معرفی شده اند که از بین آنها دو گونه ویروس ایرانی پیچیدگی بوته چغندرقند (BCTIV) و ویروس پیچیدگی شدید بوته چغندرقند (BSCTV) از ایران گزارش شده اند. پیش از این دو گونه زنجرک Circulifer haematocepsو C. tenellusبه عنوان ناقل ویروس پیچیدگی بوته چغندرقند (BCTV) گزارش شده اند ولی این مطالعات مربوط به زمانی است که BCTIV وBSCTV از هم تفکیک نشده بودند. لذا براساس مطالعات قبلی معلوم نبود که آیا هر دو ویروس یا تنها یکی از آنها با زنجرک های مزبور قابل انتقال هستند. طبق گزارش فتاحی و همکاران (1389) BSCTV با زنجرک C. haematocepsقابل انتقال است. در تحقیق کنونی، انتقال BCTIV مورد بررسی قرار گرفته است. به منظور تعیین ناقل BCTIVاز مزارع چغندرقند آلوده به بیماری پیچیدگی بوته در استان فارس (شهرستان های مرودشت، زرقان، اقلید) با دستگاه D-VAC تعداد زیادی زنجرک جمع آوری شد. براساس شکل شناسی، جمعیت حشره غالب haematoceps C. تشخیص داده شد. در نهایت به زنجرک های جمع آوری شده از یک مزرعه چغندرقند واقع در مرودشت فرصت داده شد طی چند مرحله روی گیاهان چغندرقند سالم در گلخانه زاد و ولد نموده و از پوره های تازه تولد شده آنها برای انتقال BCTIV استفاده شد. از سوی دیگر برای تهیه گیاه منبع آلوده به BCTIV، تعدادی گیاه چغندرقند در مرحله شش برگی با یک همسانه عفونت زای جدایه زرقان BCTIV (ساخته شده در مرکز تحقیقات ویروس شناسی گیاهی) به روش آگرواینوکولیشن مایه زنی شدند. برای گیرش ویروس تعدادی زنجرک روی یک گیاه چغندرقند مایه زنی شده به روش آگرواینوکولیشن انتقال داده شدند و به آنها اجازه داده شد تا به مدت 4 روز از این گیاهان تغذیه کنند و سپس به گلدان حاوی گیاهچه سالم چغندرقند در مرحله 2 برگی (5 زنجرک به ازای 3 گیاهچه با 3 بار تکرار)، انتقال داده شدند و به آنها اجازه داده شد به مدت 6 روز از این گیاهان تغذیه نمایند. علائم بیماری شامل پیچیدگی و راست ایستادن برگ ها و تورم رگبرگ ها در پشت برگ تمام گیاهان مایه زنی شده، پس از 14 روز از زمان مایه زنی، مشاهده شد. وجود این ویروس در این گیاهان از طریق انجام آزمونPCR با استفاده از آغازگرهای اختصاصی BCTIVبه اثبات رسید. بدین ترتیب انتقال BCTIVمانند BSCTV که قبلا مشخص شده بود، توسط زنجرک C. haematoceps به اثبات رسید. این اولین گزارش انتقال BCTIV با زنجرک C. haematoceps در آزمایش های گلخانه ای می باشد.

    Root rot and vine decline of cantaloupe and muskmelon plants is an economically important disease in Iran caused by Monosporascus cannonballus. Regarding the recent detection of the disease in Iran, genetic diversity of the causal agent need further research. In this study, isolates of M. cannonballus were collected from Esfahan, Yazd, Tehran, Fars, Semnan, Markazi, Ghazvin, SistanBaluchestan and Khorasan Razavi provinces. Genetic diversity between 50 isolates of the pathogen was studied using Rep-PCR markers; Box and ERIC. Reproducible fragments were between 100 and 1500bp and the genetic diversity amongst the isolates calculated 0.32. There was a correlation between genetic diversity, host and geographic origin of the isolates. M. cannonballus isolates were placed in two groups based on their host plant. Cluster analysis using UPGMA method and Dice's coefficient revealed 100% similarity between some isolates collected from different regions. There was a correlation between colony morphology in the culture media and genetic diversity where at %80 similarity level the isolates were placed in three different groups. The discriminatory power of rep-PCR was high, indicating that rep-PCR is a rapid, effective marker for genotyping M. cannonballus isolates.

    To achieve a simple and sensitive Rosellinia necatrix detection method in soil, three detection methods including soil direct culture method, infected soil in culture media e. g. MEAR, MEAR + penconazole and MEAS, Nested PCR method and bait twig method were compared. Direct DNA Extraction from soil was done. Two specific primer pairs R2، R8 and R7، R10 in Nested PCR were used and the extracted DNA was purified using PVPP column. Among used methods, the direct culture method from soil was not suitable and wasn’t sensitive for detection. However, bait twig method was capable to detect, the pathogen in soil and root but it is time consuming and laborious method. In conventional PCR, using primer pairs R2-R8, 10 pg μl−1 of fungal DNA was detected; however in Nested PCR using R7-R10 primer pairs, 1fg μl−1 of fungal DNA was detected. Among used methods, the Nested PCR was better in detection of the fungus. Nested PCR method of detection in soil and root is more sensitive and reliable.

    Deoxynivalenol (DON) is a major mycotoxin produced by the fungus Fusarium graminearum during infection and is known to be harmful to both animal and human health. DON is believed to act as a virulence factor in fungal pathogenesis by inhibiting eukaryotic protein synthesis. The putative site of action of DON is 60s ribosomal protein L3 (RPL3). Previously, single amino acid changes in RPL3 conferring DON resistance have been described in yeast. Sequence characterization of six wheat genes encoding RPL3 has shown that there is no amino acid alteration between resistant and susceptible cultivars. The main goal of this study, was to determine any induced changes in transcriptional levels of RPL3 (A and B type) genes under stress conditions caused by Fusarium Head Blight (FHB) in wheat. In order to detect expression levels of RPL3 genes, we extracted RNA from both inoculated and non-inoculated spikes of sensitive and tolerant cultivars, during 1, 3 and 7 days post-infection. Quantitative RT-PCRs were performed with A and B type-specific RPL3 primers to detect the expression differences. Primary data indicated a decrease in the genes expression level in Sumai3 and Falat cultivars 24 hours post-infection. On the other hand, increased level of RPL3A transcript in Frontana 7 days post-infection may reveal the role of this specific protein in inducing the resistance in this cultivar.

    A general surveys were conducted in Mazandaran citrus groves for the presence of Phytophthora species causing citrus gummosis. Soil samples and infected citrus tissues were collected and assayed using baiting and selective medium. Phytophthora citrophthora and P. nicotianae were detected in all of the citrus orchards and their distribution was mapped. The soil population of each species was also determined. Eighty two and 86% of the orchards were infested respectively with P. citrophthora and P. nicotianae with average over 10 propagules /g soil.The virulence of five isolates of each species to root of 1-year-old citrange, citromelo, Shahsavar orange, and sweet lime was examined. Data analysis of fresh weight of root and stem, and disease index in citrange revealed three groups including control, P. nicotianae, and P. citrophthora isolates, which the that third group was significantly more virulent than the second group. Data analysis of fresh weight of root and stem in citromelo and Shahsavar orange did not show significant differences between P. nicotianae, and P. citrophthora isolates. On the basis of disease index data of citromelo and Shahsavar orange, P. citrophthora isolates were more virulent than P. nicotianae isolates. Data analysis of fresh weight of root and stem and disease index in sweet lime did not show significant differences between P. nicotianae, and P. citrophthora isolates.

    Powdery mildew caused by an obligate biotrophic fungus Erysiphe necator (synonym Uncinula necator) is the most economically important disease of grapevines. Most widely grown grape cultivars are highly susceptible to E. necator, because they are derived from Vitis vinifera, a species which was not exposed to this pathogen during its evolution. In this study we investigated germinating of grapevine powdery mildew on two susceptible and resistant grapevine genotypes as well as ontogenic resistance in vegetative tissues of Cabernet sauvignon and Arabidopsis thaliana with different age. Conidia germinated and appressorium and haustorium appeared during 2-12 hpi in both genotypes. The primary hypha developed 12 hpi in both genotypes however; yellow deposits could be seen surround epidermal cells 12 hpi just in resistant genotype. Development of primary hypha continued up to 24 hpi in susceptible Cabernet sauvignon, however, 24 hpi the hypersensitive reaction (HR) happened in resistant genotype and blocked secondary hypha development, but not in Cabernet sauvignon. By increased in physiological age related to ontogenic resistance the penetration efficiency of fungi in Cabernet sauvignon and Arabidopsis thaliana reduced.

    Isolates of the yeasts Candida membranifaciens, Rhodotorula mucilaginosa and Pichia guilliermondii obtained from healthy apples were evaluated as potential bio. control agents for apple gray mold caused by Botrytis mali and B. cinerea. In-vitro volatile metabolites emitted from all yeast isolates inhibited the mycelial growth of B. mali and B. cinerea by 82.7 to 97.7 and 81.2 to 98.3 percent, respectively. Each of the yeasts significantly reduced the decay area in apples inoculated with the pathogens C. membranifaciens A2, A4, A5 and P. guilliermondii A6 had the greatest effect on decay reduction at 4oC, 20 oC and 20±3 oC. The populations of C. membranifaciens A2 and C. membranifaciens A5 during the experiment increased in apple wounds at 4oC.

    The effect of rhizosphere bacteria on root-knot nematode (Meloidogyne javanica) was investigated on cucumber under greenhouse conditions. The bacteria were isolated from infected cucumber, tomato and cedar from Garmsar, Marvdasht and Shiraz, as well as cedar, grass and wheat from Bajgah. After sampling, purification, rearing on root of tomato and identification of M. javanica species, the effect of 40 isolates were investigated on hatching (in 3 and 8 days after bacteria inoculation) and the activity of second stage juvenile of the nematode (24, 48 and 72 hours after adding bacteria) in vitro condition. Then 10 best isolates were used on cucumber (Super Amelia) in greenhouse. All isolates increased growth of the plants and decreased the indices of the nematode. Pseudomonas fluorescens CHAO was the most effective isolate and caused about 55.31% and 55.72% reduction in the gall number and also respectively 91.13% and 82.84% reduction in reproduction factor (RF) in the first and second experiments. The second one was an isolate of Pantoea sp. Additionally the effects of P. fluorescens, Bacillus subtilis and Pantoea sp. on the infected cucumber (Super Amelia) to M. javanica, as single application and in combination was tested. The results indicated that using combination of three bacteria are more effective than application of single and combination of two bacteria on increasing growth of plant and reducing indices of the nematode.

    The two most important diseases of potato in Hamedan province are black-dot disease caused by Colletotrichum coccodes and bacterial wilt disease caused by Ralstonia solanacearum. In this study interactions between the two pathogens were surveyed on the three potato cultivars, Agria, Boren and Diamant using two inoculation methods. Data analysis revealed that co-inoculation of potato by both pathogens showed pathogenicity synergistic effect, and potato length and stems and roots weight were decreased. In Agria, R. solanacearum increased C. coccodes effectiveness on potato plants. Agria cultivar root colonization by C. coccodes sclerotia was more than other cultivars and was scored as 50 – 70%. In fact, this cultivar was more susceptible to C. coccodes than others. Pathogenicity of C. coccodes and R. solanacearum on Boren potato cultivar were almost equal and potato length and stems and roots weight were decreased in compare to control plants. At the presence of two pathogens plant parameters were more decreased. In this cultivar root colonization by C. coccodes sclerotia was scored as 50%. In most cases, Rs treatment on Diamant has no significant differences with control plants and there was relative resistance to the bacterial wilt in this cultivar. In compar to other cultivars, root colonization in Diamant was low and scored 30 %. Diamant potato cultivar was more resistant than others to C. coccodes.

    Phytoplasmal lettuce phyllody (LP) disease previously was reported from Fars province is vectored by Neoaliturus fenestratus leafhopper. Transmission characteristics of LP phytoplasma with N. fenestratus including acquisition feeding, latency and inoculation feeding period were studied. The minimum acquisition access period (AAP) was four hours. Transmission increased with AAP. With an AAP of 48 hours, 88.88 % of plants were infected. The minimum latent period (LP) in N. fenestratus was 14 days and transmission increased after this minimum LP. At 32 days postacquisition, 92.85% of plants were infected, but the rate of transmission gradually decreased. The minimum inoculation access period (IAP) was 4 hours. Transmission increased with IAP. At an IAP of 48 hours, 96.66 % of the plants were infected. Results of this study indicate that the transmission processes of acquisition, latency and inoculation occur in much the same pattern as in other plant pathogenic mollicutes.

    In order to evaluate the effect of sugar beet genotype and method of plant inoculation on resistance to Iranian isolate of Beet severe curly top virus (BSCTV-Ir), a completely randomized experimental design was performed using 18 sugar beet cultivars. Eighteen plants of each cultivar were grown in 6 pots. Two methods of inoculation, namely, agroinoculation using the infectious clone of BSCTV-Ir and inoculation by viruliferous leafhopper (Circulifer haematoceps) were used. The virus replication in inoculated plants was analyzed by PCR, 21 and 35 days post-inoculation (dpi) in the first experiment and 21 dpi in the second experiment. The severity of disease symptoms of inoculated plants was graded using a 0-3 scale (0, symptomless; 1, mild; 2, moderate and 3 severe symptoms). Data analysis of the first experiment indicated that the cv Brigita was the only cultivar in which all the inoculated plants were infected by the virus and all of them showed severe symptoms. Consequently, this cultivar was considered as a highly sensitive control cultivar and used in comparisons. Statistical analysis of the disease symptom severity index showed highly significant differences among cultivars when compared with the control cultivar. As a result, three distinct groups were identified. The first group including the cultivars BR1, FIMMA, HM1990, 7233, and H5505, and the second group including the cultivars Rasoul, Afshari, hybrid of Balk Shiraz, P.P.8, P.P.22, Dorothea, Zarghan, Rhizofouret, IC, Flores, Hilma and Polyrow were regarded as tolerant and sensitive groups, respectively. The third group included only Brigita cultivar which was regarded as very sensitive. Comparison of the data obtained from two inoculation methods indicated that in most cultivars, the virus infection rate was lower with vector inoculation than with agroinoculation.

    Plane trees (Platanus orientalis) decline is prevalent in Shiraz and other parts of the country. From 50 plane trees with decline symptoms, forty one yielded several fungi such as Cytospera (42.85%), Alternaria (18.9%), Aspergillus (12.7%), Nattrassia mangiferae (3.22%), Penicillium (3.05%), Rhizopus (0.7%), Helminthosporium (0.24%), Chaetomium (0.2%), Coniothyrium (0.2%) and Ulocladium (0.07%). The fungi were inoculated to detached stem of plane tree. Only N. mangiferae and few isolates of Cytospora were pathogenic. From 41 isolates of Cytospora recovered, only 8 isolates were mildly pathogenic to detached but not to intact plane tree. From ten isolates of Cytospora inoculated to detached stem of Acer pseudoplatanus, Platanus orientalis, Magnolia grandiflora, Morus alba, Citrus aurantium, Malus communis, Juglans regia, Cercis siliquastrum, Fraxinus excelser and Pinus sp. only 1 isolate was pathogenic on M. alba and 5 to C. siliquastrum. None of the 40 Cytospora isolates caused infection of stem the above plants under natural conditions. On the basis of morphological characteristics the fungus was identified as Cytospora theryana. There was no significant differences among soil samples collected around declining and healthy plane tree in Shiraz in respect to soil, EC, pH and soil texture. It is concluded that plane tree decline in Shiraz city is as the result of various environmental and nutritional stresses which predisposed trees to various opportunistic fungi.

    Apple powdery mildew (Podosphaera leucotricha) is one of the most important diseases in this crop causing weakness and yield reduction in Semirom, Isfahan Province. In order to evaluate the reaction of 14 commercial cultivars budded trees (on apple rootstock)were planted in a completely randomised block design, with 3 replicates(3 plants in every plot) 2.5×1m apart, at Kahangan village in Semirom.Sampling were carried out at the time of matimum lesion size. Relative resistance was assessed on the basis of lesion size in natural conditions. The results showed that all of the cultivars infected with different intensity. Fugi, Golden delicious, Gala, Azayesh, Braborn and Delbar Stival cultivars, with Severity Index(SI): 30.12, 37.92, 31.71, 33.2, 42.91 and 25.78 were susceptible. Janatan, Golab Isfahan, Golab Kermanshah, Golab Khorasan and Golab Kohanz cultivars with the SI: 54.96, 55.57, 58.71, 53.68 and 59.42 were highly susceptible while Red Delicious and Ingrid Mary with the SI:9.04 and 11.44 were resistance respectively.

    Pear fruits (Pyrus communis) with rotting symptoms and orange spore masses were collected in Guilan Province (Rasht), Iran in spring of 2009. Morphological characteristics including shape and size of conidia and appressoria, presence or absence of setae and sclerotia and color and growth rate of colony were studied. Moreover, total DNA was extracted with Chelex 5% method. The ITS1- 5.8S-ITS2 regions of rDNA were amplified and sequenced using ITS1 and ITS4 primers. According to morphological and molecular characteristics the fungus was identified as Colletotrichum acutatum. In pathogenicity test, the species caused fruit rot on wounded and unwounded pear fruits. Based on this study, pear is reported as a new host of C. acutatum for Iran and this is the first report of ITS regions of rDNA of this fungus from Iran on Pear.

    Beet curly top is a disease of sugar beet which has been in the center of attention due to its unpredictable epidemics and its economic importance. It has been shown that due to short life cycle and high populations of the leafhopper vector, the disease often spreads very rapidly in a region. At least five virus species of the genus Curtovirus (Geminiviridae) are recognized as the cause of beet curly top disease. Two of these species, namely, Beet severe curly top virus (BSCTV) and Beet curly top Iran virus (BCTIV) have been reported from Iran. Two leafhopper species, Circulifer haematoceps and C. tenellus, have been reported as the vector of Beet curly top virus before recognition of different species (Fattahi et al., 2010). Therefore, it was not certain whether one or both viruses are transmitted by the leafhoppers. Recently, it was shown that C. haematoceps could transmit BSCTV from sugar beet plants infected via an infectious clone of the virus. In the present study, the possibility of transmission of BCTIV by C. haematoceps was examined. Leafhoppers were collected in sugar beet fields of northern Fars province using D-Vac. The dominant species was C. haematoceps. The leafhoppers were allowed to breed on healthy sugar beet in the greenhouse. The newly born nymphs were placed on sugar beet plants previously agroinoculated with an infectious clone of BCTIV (Zarghan isolate) for an acquisition access period of four days, and then transferred to healthy seedlings at 2-leaf stage (5 leafhoppers / 3 seedlings). Typical symptoms of curly top appeared on all inoculated seedlings after two weeks. Presence of BCTIV in symptomatic test plants was verified by PCR using specific BCTIV primers. The experiment was repeated two more times with similar results. It is concluded that like BSCTV, BCTIV is transmitted by C. haematoceps ‌under greenhouse condition.
