    ویروس موزائیک خیار (Cucumber mosaic virus، CMV) بیش از هزار وسیصد گونه گیاهی را آلوده و سبب افت کمی و کیفی محصولات می شود. در این تحقیق کاربرد خاموشی پس از نسخه برداری با استفاده از سازه سنجاق سری برای بدست آوردن گیاهان مقاوم به CMV بررسی شد. به این منظور ترادف 145 جفت بازی حفاظت شده ویروسی از ژن بازدارنده سیستم خاموشی گیاه میزبان انتخاب شد. این ترادف بصورت سنس و آنتی سنس که توسط اینترون از هم جدا می شوند در حامل دوگانه pFGC5941 همسانه سازی و حامل نوترکیب pFGC-2b-h نامیده شد. حامل ساخته شده به اگروباکتریوم سویه GV3101 منتقل گردید. گیاهان توتون (Nicotiana benthamiana) و لوبیا (Phaseolus vulgaris) بصورت موقت به این سازه تیمار شد و شش روز بعد با ویروس موزائیک خیار مایه زنی شدند. کارائی سازه 15 روز بعد از مایه زنی با ویروس مورد ارزیابی قرار گرفت. نتایج این پژوهش نشان داد این سازه کارائی بالای در کاهش علایم و غلظت ویروس در برگ های بالائی نسبت به گیاه شاهد دارد و از این سازه می توان در تولید گیاهان تراریخت مقاوم به CMV استفاده کرد.

    ویروس موزائیک زرد نواری جو (Barley yellow striate mosaic virus، BYSMV) متعلق به جنس Cytorhabdovirusو خانواده Rhabdoviridaeمی باشد. این ویروس عامل بروز علائم موزائیک ونوارهای کلروتیک در غلات است و توسط زنجرک Laodelphax striatellus با رابطه تکثیری منتقل می شود. در مطالعه حاضر ترادف نوکلئوتیدی ژن های گلیکوپروتئین و فسفوپروتئین جدایه زنجان این ویروس گزارش می شود. پس از خالص سازی نوکلئوکپسید ویروس، ژنوم باروش random RT-PCR تکثیر شد. آغازگر های اختصاصی نیز با نرم افزار version 9.0VECTOR-NTI جهت بدست آوردن ترادف نواحی مابین قطعات تکثیر شده با PCR، طراحی شدند. پس از هم ردیف سازی چندگانه ترادف ترجمه آمینواسیدی دو ژن جدایه زنجان با سایر رابدوویروس های گیاهی موجود در بانک ژن توسط نرم افزار‍CLUSTAL W، این جدایهBYSMV بیشترین میزان شباهت را با NCMV) Northern cereal mosaic virus) نشان داد. در درخت های فیلوژنتیکی ترسیم شده، رابدوویروس های گیاهی در دو گروه اصلی مرتبط با دو جنس Cytorhabdovirus و یا Nucleorhabdovirusقرار گرفتند کهBYSMV و NCMV در یک زیر گروه جداگانه قرار گرفتند. بررسی قابلیت انتقال چهار جدایه BYSMV در این مطالعه نشان داد که جدایه های زنجان، نقده و سرو با کارآیی بالاتری نسبت به جدایه مشهد توسط یک بیوتیپ از زنجره L. striatellusمنتقل می شوند.

    وجود منابع فعال ویروس های موزائیک رگه ای گندم و کوتولگی زرد جو و نیز وقوع آلودگی هم زمان آن ها در مناطق مختلف گندم کاری ایران گزارش شده است. در این تحقیق، اثر انفرادی و ترکیبی متغیر های فوق بر عیار ویروس، شاخص علائم WSMV و برخی متغیر های رشدی و عملکردی سه ژنوتیپ گندم بررسی گردیده است. آزمایش حاضر به صورت یک طرح فاکتوریل در قالب بلوک های کامل تصادفی (RCBD) با 15 تکرار انجام شد. متغیر های آزمایش شامل ترتیب اعمال تیمار ویروسی در شش سطح، ژنوتیپ در سه سطح و زمان پس از آلودگی با WSMV در سه سطح در نظر گرفته شد. اثر انفرادی و ترکیبی متغیر ها بر صفات مورد نظر بررسی گردید. نتایج نشان داد که تیمار های با آلودگی دو گانه بیشترین میزان جذب الیزا و شدت علائم ناشی از WSMV و نیز پایین ترین میزان متغیر های زراعی را دارا بودند. در مقایسه انواع تیمار ویروسی، آلودگی اولیه با WSMV و سپس BYDV و در بین ژنوتیپ ها، رقم حساس مرودشت، بالاترین میزان جذب الیزا و شاخص علائم برای WSMV را به خود اختصاص دادند. بیشترین میزان جذب الیزا و شاخص علائم به ترتیب در 21 و 35 روز پس از آلوده سازی با WSMV مشاهده شد. در برهمکنش ژنوتیپ و تیمار ویروسی، بیشترین میزان جذب الیزا و شاخص علائم به تیمار های آلودگی اولیه به WSMV و سپس BYDV در رقم های مرودشت و رسول اختصاص یافت. در مطالعه برهمکنش سه متغیر، تیمار آلودگی اولیه به WSMV و سپس BYDV در ژنوتیپ های مرودشت و رسول، در 21 روز پس از آلوده سازی بیشترین میزان جذب را دارا بودند. تمامی تیمار های آلودگی دوگانه منجر به کاهش محسوس ارتفاع، عملکرد دانه و وزن اندام هوایی شدند. در این بین پایین ترین میزان شاخص های فوق به تیمار آلودگی هم زمان دو ویروس تعلق گرفت. در ژنوتیپ مقاوم کراس عدل، بیشترین مقدار جذب الیزا در تیمار آلودگی اولیه با BYDV و سپس WSMV مشاهده شد. این درحالی است که کاهش معنی داری در عملکرد محصول در تیمار آلودگی هم زمان دو ویروس دیده شد.

    ویروس های کوتولگی زرد جو یکی از مهمترین ویروس های غلات در دنیا می باشند. بررسی های انجام شده بیانگر پراکنش ویروس های کوتولگی زرد جو (Barley yellow dwarf virus، BYDV) و غلات (Cereal yellow dwarf virus، CYDV) در کشور و غالب بودن BYDV-PAV می باشد. به منظور تعیین ترادف کامل ویروس کوتولگی زرد جو (BYDV-PAV) در ایران، جدایه ای از منطقه اسفندقه از استان کرمان که دارای علائم شدید آلودگی به ویروس مذکور بود انتخاب شد. استخراج آر. ان. ای کل از بافت گیاهی آلوده به ویروس با استفاده از کیت TM RNX (-plus) (شرکت سیناژن) انجام شد. کل ژنوم ویروس با استفاده از نه جفت آغازگر اختصاصی PAV با روش RT-PCR تکثیر شد. قطعه های تکثیرشده با استفاده از InsT/AClone PCR Product Cloning Kit (Fermentas) در پلاسمید PTZ57R/T وارد و در سویه DH5αباکتری Escherichia coliهمسانه سازی شد. آنالیز ترادف نوکلئوتیدی کامل نشان داد که جدایه کرمان ویروس کوتولگی زرد جو (KP771878) با 94% در صد تشابه با جدایه های AF235167 (از ایلی نویز آمریکا) و EF043235 (جدایه کانزاس از آمریکا) در یک گروه قرار می گیرند در حالیکه در صد تشابه این جدایه با سایر جدایه های موجود در بانک ژن کمتر از 90% می باشد و کمترین در صد تشابه آن با جدایه AY855920 از چین (78%) است. نتایج حاصل از مقایسه ترادف ها در سطح هر یک از ژن ها نیز، وجود تنوع زیاد در بین جدایه های این ویروس و همچنین احتمال نوترکیبی ژن ها را تایید می نماید. به طوری که ORF4 حفاظت شده ترین و ORF6 متغیرترین ناحیه در ژنوم این ویروس است.

    شش جمعیت از Rhizoctonia solani AG-1 IA از سه مزرعه برنج با آلودگی طبیعی در مناطق رشت، تنکابن و آمل در شمال ایران نمونه برداری شد. تجزیه و تحلیل آللی با استفاده از آغازگر های فلورسنت در نه جایگاه ریز ماهواره ای انجام شد. طیف فراونی آلل ها و ژنوتیپ ها به ترتیب از 002/ 0 تا 962/ 0 و از 40/ 0 تا 32/ 10 درصد در 252 جدایه بودند. فراوانی 13، 11 و 9 آلل به ترتیب در پنج، شش و سه جایگاه در جمعیت های جغرافیایی رشت، آمل و تنکابن صفر بود و از این جایگاه ها دو جایگاه در هر جمعیت در (Hardy–Weinberg Equilibrium (HWE نبودند. در حالیکه آلل bp 145 در جایگاه TC12 در جمعیت های آمل با فراوانی یک، تثبیت شد. همانطور که مشخص شد فراوانی آلل و ژنوتیپ در جمعیت ها دارای پراکنش یکنواختی نبودند. این وضعیت بیانگر تثبیت و حذف تدریجی بعضی آلل ها در جمعیت های این بیمارگر در شالیزارهای شمال ایران می باشد.

    نمونه برداری از غده های سیب زمینی آلوده به پوسیدگی صورتی در انبارهای استان فارس منجر به جداسازی تعدادی جدایه ی فیتوفتورا شد که شباهت کاملی با هیچ یک از عوامل شناخته شده ی ایجاد کننده ی پوسیدگی صورتی نداشتند. واکاوی ژن های سه ناحیه ی هسته ای (فاصله ی ترانویسی شده ی داخلی (آی تی اس)، بتاتوبولین و پروتئین شوک حرارتی 90) و یک ناحیه ی میتوکندریایی (زیر واحد یک سیتوکروم سی اکسیداز) نشان داد این جدایه ها دورگی از Phytophthora erythroseptica و P. cryptogea هستند. واکاوی فیلوژنتیکی بر اساس ره یافت بیسی نشان داد که جدایه های عامل پوسیدگی صورتی سیب زمینی همراه با گونه های P. erythroseptica وGII P. cryptogea در گروهی تک نیا و مستقل از سایر اعضای مهم تبار هشت فیتوفتورا قرار می گیرند. همسانه سازی ژن بتاتوبولین با تولید توالی های والدی، طبیعت دورگ این جدایه ها را تایید کرد. این جدایه ها دارای اسپورانژیوم های بدون پاپیل، بیضی تا تخم مرغی شکل، ااسپورهای گرد و ناپرساز و آنتریدیوم های انتهایی، منفرد و پیراماده بودند. جدایه های به دست آمده با وجود تفاوت های مشاهده شده، از نظر خصوصیات ریخت شناختی شباهت زیادی به والد مادری شان یعنی P. erythroseptica داشتند.

    گیاه همیشه بهار Calendula officinalis به علت داشتن ترکیب های دارویی متنوع در صنایع داروسازی حائز اهمیت می باشد. در این پژوهش٬ اثر مایه زنی نماتود ریشه گرهی Meloidogyne javanica در هفت سطح مختلف (صفر، 5/ 0، 1، 5/ 1، 2، 5/ 2 و 5 تخم و لارو /گرم خاک) روی شاخص های رشد این گیاه و تکثیر نماتود ارزیابی گردید. نود روز پس از مایه زنی، مقدار ترکیبات شیمیایی کوئرستین، کامپفرول و اسید یورسولیک در گیاهان آلوده اندازه گیری شد. نتایج نشان داد با افزایش تراکم مایه زنی میانگین فاکتورهای رشد گیاه به طور معنی داری کاهش یافت. فاکتور تولیدمثل نماتود با سطوح مختلف جمعیت رابطه معکوس داشته و بالاترین فاکتور تولیدمثل در کمترین سطح از جمعیت نماتود یعنی سطح 5/0 تخم و لارو مشاهده گردید. سنجش ترکیب های فلاونوییدی و تری ترپنوییدی، کوئرستین و یورسولیک اسید عصاره گل های گیاه همیشه بهار حاکی از افزایش این ترکیب ها در گیاهان آلوده نسبت به شاهد (بدون نماتود) بود.

    پایه GF-677 (هیبرید بین هلو و بادام) یکی از مناسب ترین پایه ها در تولید نهال های بادام و هلو می باشد. طی بازدیدهای سال های 1388 و 1389 از درختان GF-677 در بیدزرد و استهبان (استان فارس) علائم جاروک در آن ها مشاهده گردید. عامل بیماری جاروک GF-677 در بیدزرد و استهبان از طریق پیوند از GF-677، به نهال های بادام تلخ، هلو و GF-677 و با استفاده از سس (Cuscuta campestris Yank.)، از نهال های آلوده بادام تلخ به پروانش و بادنجان منتقل شد و در آنها علائم بیماری های فیتوپلاسمایی تولید کرد. واکنش دی ان ای کل استخراج شده از درختان GF-677 در بیدزرد و استهبان و گیاهان مایه زنی شده با پیوند و سس، در آزمون پی سی آر مستقیم با استفاده از جفت آغازگر P1/P7 و پی سی آر دو مرحله ای با استفاده از جفت آغازگرهای P1/P7 و R16F2n/R16R2 مثبت بود و باند مورد انتظار به ترتیب 1800 و 1200 جفت بازی تکثیر شد. محصول پی سی آر مستقیم جدایه های بیدزرد و استهبان همسانه سازی و تعیین ترادف شد و تحت رس شمارهای JX445141 و JX445142 در بانک جهانی ترادف ها ثبت گردید. جستجو با برنامه بلاست نشان داد که فیتوپلاسماهای عامل جاروک GF-677 در بیدزرد و استهبان، متعلق به گروه ار ان ای ریبوزومی جاروک نخود کبوتر (Pigeon pea witches’- broom، 16SrIX) می باشند. مقایسه چند شکلی طولی قطعات برشی (RFLP) محصول پی سی آر مستقیم فیتوپلاسماهای جاروک GF-677 نشان داد که جدایه های بیدزرد و استهبان، با یکدیگر و همچنین با فیتوپلاسمای عامل جاروک بادام نیریز یکسان می باشند. با استفاده از RFLP مجازی مشخص گردید که جدایه های جاروک GF-677 و فیتوپلاسمای عامل جاروک بادام نیریز، متعلق به زیرگروه 16SrIX-B می باشند و با فیتوپلاسمای عامل جاروک بادام خفر تفاوت دارند. جدایه های بیدزرد و استهبان و فیتوپلاسمای عامل جاروک بادام خفر از نظر علائم بیماری در بادام تلخ و پروانش نیز قابل تشخیص بودند. تعلق جدایه های جاروک GF-677 از بیدزرد و استهبان به گروه 16SrIX و نزدیکی آنها به زیرگروه 16SrIX-B در آنالیزهای فیلوژنتیکی با استفاده از ترادف های کامل ژن 16S rRNA و ناحیه SR نیز به اثبات رسید.

    ترجیح میزبانی نماتد Ivanova، 1967 Helicotylenchus digitiformis، در دو آزمون مجزا، با جمعیت های اولیه 49/ 1 و 33/ 4 نماتد در گرم خاک، در قالب طرح بلوک های کامل تصادفی در شرایط گلخانه انجام شد. نتایج نشان داد که همه گیاهان مورد آزمایش، شامل پنبه (دلتاپاین 16)، چغندرقند (Pb8)، چمن (اسپورت)، ذرت (هیبرید 704)، شبدر (ایرانی)، طالبی (سمسوری)، کلزا (طلایی)، گندم (شیراز)، گوجه فرنگی (راتگرز) و یونجه (همدانی)، میزبان این نماتد هستند، اما بالاترین فاکتور تولیدمثل و جمعیت نهایی نماتد مربوط به پنبه و ذرت بود. همچنین به منظور تعیین سطوح جمعیت خسارت زای گونه H. digitiformis روی هر یک از گیاهان ذرت (هیبرید 704) و پنبه (رقم دلتاپاین 16) دو آزمون جداگانه در قالب طرح بلوک های کامل تصادفی، با جمعیت مزرعه ای، نصف جمعیت مزرعه ای و شاهد، به ترتیب با 33/ 4، 16/ 2 و صفر نماتد در گرم خاک، با شش تکرار در شرایط گلخانه و میکروپلات انجام شد. نتایج نشان داد که تیمارهای جمعیت مزرعه ای و نصف جمعیت مزرعه ای در شرایط میکروپلات و تیمار جمعیت مزرعه ای در شرایط گلخانه باعث کاهش معنی دار اکثر شاخص های رشدی گیاه ذرت نسبت به شاهد گردیدند. با وجود تکثیر نماتد بر روی رقم دلتاپاین 16 پنبه و داشتن فاکتور تولیدمثل بالاتر از یک در تیمارهای دارای نماتد، هیچ یک از سطوح جمعیتی در شرایط گلخانه و میکروپلات کاهش معنی داری بر روی شاخص های رشدی گیاه نسبت به شاهد ایجاد نکرده و به نظر می رسد رقم مذکور در شرایط آزمون های انجام شده نسبت به سطوح جمعیتی نماتد متحمل باشد. همچنین آزمون مشابهی روی دو رقم گندم پیشتاز و شیراز، در قالب طرح بلوک های کامل تصادفی با پنج تکرار در شرایط میکروپلات انجام گردید. از بین چهار سطوح جمعیت 49 /1، 75/ 0، 50/ 0 و صفر نماتد در گرم خاک تنها تیمار دارای 49/1 نماتد در گرم خاک روی اکثر شاخص های رشدی رقم پیشتاز موثر بوده و کاهش معنی داری نسبت به شاهد ایجاد نمود.

    بیماری زنگ قهوه ای (برگی) گندم با عامل Puccinia triticina Eriksson از مخربترین بیماری های قارچی گندم است که هر ساله در بسیاری از مناطق گندم کاری ایران با شدت های مختلف باعث خسارت و کاهش عملکرد محصول گندم می شود. به منظور مطالعه اجزاء مقاومت به این بیماری، 31 رقم تجاری گندم تحت شرایط کنترل شده و در مرحله گیاهچه ای در گلخانه، با یک جدایه زنگ قهوه ای (نژاد CHHLS) از لرستان مایه زنی و در قالب طرح بلوک های کامل تصادفی با 3 تکرار مورد ارزیابی قرار گرفتند. در این تحقیق، اجزاء مقاومت شامل تیپ آلودگی و دوره نهفتگی مورد مطالعه قرار گرفت. تعیین دوره نهفتگی به صورت تعداد روز از مایه زنی تا ظهور اولین جوش روی برگ که 8 روز پس از مایه زنی شروع و تا 14 روز پس از مایه زنی ادامه داشت و تیپ آلودگی به روش رولفز در مقیاس (4-0) محاسبه گردید. نتایج مطالعه نشان داد که صفات تیپ آلودگی و دوره نهفتگی دارای همبستگی منفی (6463/ 0-) در سطح 1% می باشند. ارقام اروم بیشترین دوره نهفتگی و کمترین تیپ آلودگی و بولانی به عنوان رقم حساس کمترین دوره نهفتگی و بیشترین تیپ آلودگی را داشتند. نتایج تجزیه واریانس نشان داد که ارقام برای تیپ آلودگی در سطح 1% دارای اختلاف معنی دار می باشند ولی برای دوره نهفتگی اختلاف آنها معنی دار نگردید. در تجزیه خوشه ایمربوط به صفات مورد مطالعه ارقام گندم در مقاومت به زنگ قهوه ای به سه گروه مجزا مقاوم، نیمه مقاوم و حساس تقسیم شدند. گروه مقاوم شامل (خزر1، گلستان، بک کراس روشن بهاره، بم، نیشابور، بک کراس روشن زمستانه، هیرمند و اروم) گروه نیمه مقاوم شامل (سیستان، کویر، پارسی، نوید، مغان1، مغان2، مهدوی، افلاک، میهن الموت، بهار، داراب2، شیراز، چناب، البرز و فلات) و گروه حساس شامل (زارع، مغان3، اترک، زرین، ارگ، اروند و بولانی) می باشد.

    به منظور شناسایی فون نماتد های انگل گیاهی گندم در شهرستان های بویراحمد و دنا، طی سالهای زراعی 92-1391، تعداد 50 نمونه خاک و ریشه از مزارع گندم جمع آوری گردید. پس از انتقال نمونه ها به آزمایشگاه، عملیات شستشوی خاک و استخراج نماتدها با استفاده از روش سری الک ها و سانتریفیوژ و سپس تثبیت و انتقال آن ها به گلیسیرین خالص با روش دگریسهانجام گرفت و از آن ها اسلاید های میکروسکوپی دائمی تهیه شد. پس از مطالعه میکروسکوپی، اندازه گیری ها و ترسیم های لازم انجام و با استفاده از منابع و کلیدهای معتبر گونه های استخراج شده مورد شناسایی قرار گرفت. در این مطالعه بر اساس مشخصات ریخت سنجی و ریخت شناختی تعداد 28 گونه شناسایی شدند که شرح کامل دو گونهDitylenchus equalis و D. tenuidensدر اینجا ارائه می شود.

    بیماری آتشک یکی از مخرب ترین بیماری های باکتریایی درختان هسته دار و دانه دار می باشد. عامل بیماری، Erwinia amylovora از اعضای خانواده Enterobacteriaceae است. از میان درختان میوه، گلابی حساس ترین میزبان و درختانی نظیر سیب، به و برخی دیگر از اعضای خانواده روزاسه نیز به این بیماری حساس هستند. در ایران آتشک برای نخستین بار از برغان کرج گزارش گردید (Zakeri & Sharifnabi 1991). این بیماری سبب بروز علائم متعددی نظیر بلایت شکوفه ها، نکروز و تراوش صمغ شیری رنگ باکتری از اندام های آلوده می گردد. در سال 1391 علائم این بیماری شامل سرخشکیدگی شدید و سیاه شدگی میوه های نارس به (Cydonia oblonga)، در مناطقی از شهرستان شهربابک در استان کرمان مشاهده گردید. نمونه هایی در فصل بهار و پائیز از باغات مختلف به تهیه و برای جداسازی بیمارگر به آزمایشگاه منتقل گردید. نمونه هایی از بافت آلوده تهیه شده و روی محیط کشت اگار غذایی حاوی نیم درصد ساکاروز کشت داده شد. پس از 72 ساعت نگهداری در دمای 27 درجه سلسیوس کلنی های سفید و لعاب دار در حال رشد بر سطح محیط کشت جدا و روی محیط EMB مخطط گردید. به منظور شناسایی جدایه ها از روش های مرفولوژیک و بیوشیمیایی استفاده گردید(Schaad et al. 2001). برای بررسی توان بیماری زایی جدایه ها، سرشاخه های جوان به با سوسپانسیونی حاوی 107 کلنی در هر میلی لیتر، مایه زنی شده و در شرایط گلخانه با رطوبت نسبی 80 تا 100 درصد و دمای 27 درجه سلسیوس نگهداری گردید. به عنوان کنترل منفی از آب مقطر استریل برای مایه زنی سرشاخه ها استفاده گردید. پنج روز پس از مایه زنی علائم بیماری بصورت زخم های تیره در سرشاخه ها، ایجاد گردید. در سرشاخه هایی که با آب مایه زنی شده بودند علائمی دیده نشد و از بافت های آلوده باکتری بیمارگر مجددا جداسازی گردید. بیست جدایه باکتری در این تحقیق مورد ارزیابی قرار گرفتند. این جدایه ها روی محیط کشت NAS کلنی های سفیدی ایجاد نموده، گرم منفی، بی هوازی اختیاری بوده و سبب القا واکنش فوق حساسیت در توتون شدند. جدایه ها اکسیداز منفی بوده و توانایی تولید رنگ فلورسنت روی محیط کشت Kings B و رشد در دمای 37 درجه سلسیوس را نداشتند. براساس ویژگی های فنوتیپی و بیوشیمیایی تعیین شده و آزمون بیماری زایی، جدایه ها به عنوان باکتریE. amylovora تشخیص داده شدند. بر اساس اطلاعات ما این نخستین گزارش از وجود بیماری آتشک به، در استان کرمان است.

    در نمونه برداری از مناطق مختلف کشت درختان سیب شهرستان های بویراحمد و دنا در فصول بهار و تابستان 1391 و 1392، گونه ای از جنس Tylenchus به دست آمد که با بررسی های مرفولوژی و مرفومتری T. naranensisMaqbool، Zarina & Ghazala، 1987 تشخیص داده شد که گزارشی جدید برای ایران است و شرح داده می شود. ماده: بدن به سمت شکم خمیده تا C شکل، سر در امتداد بدن با شبکه کوتیکولی ضعیف و دارای چهار شیار، بلندی سر 4/ 3-3/ 2 میکرومتر و عرض آن در پایه شبکه کوتیکولی 6/6-3/ 5 میکرومتر. استایلت ظریف، قسمت مخروطی به طول 7/ 5-3/ 5 میکرومتر و 50-46 درصد طول استایلت. استایلت دارای گره های گرد تا متمایل به عقب. محل ریزش غده پشتی مری در زیر گره استایلت با فاصله 4/ 1-8/ 0 میکرومتر از زیر گره های استایلت. سطح بدن دارای شیار های عرضی ظریف به فاصله یک تا یک و نیم میکرومتر از یکدیگر، سطوح جانبی دارای چهار شیار طولی به عرض 4/5-2/4 میکرومتر و به اندازه 26-19 درصد عرض بدن.. لوله اولیه مری استوانه ای و تقریبا هم اندازه با طول لوله ثانویه. حباب میانی بیضی شکل به عرض 5/ 8-6/6 میکرومتر با دریچه مشخص، واقع در نیمه اول تا اواسط مری. لوله ثانویه مری استوانه ای شکل، حلقه عصبی در نیمه اول لوله ثانویه مری و با فاصله 86-64 میکرومتر از ابتدای بدن واقع شده. همی زونید یک تا دو شیار قبل از روزنه دفعی-ترشحی. دایرید مشخص، در نزدیکی روزنه دفعی-ترشحی و در 106-90 میکرومتری از ابتدای بدن. روزنه دفعی-ترشحی قبل از ابتدای حباب انتهایی. حباب انتهایی رشد یافته و کشیده، به عرض 5/ 9-0/ 8 میکرومتر و طول 25-19 میکرومتر. واژن به طول 12-0/ 6 میکرومتر و 40-30 درصد عرض بدن در ناحیه شکاف تناسلی. شکاف تناسلی با لبه های نسبتا برجسته. لوله تناسلی مستقیم، رو به جلو و یک تخمدانه با کیسه عقبی رحم به طول 13-5/ 7 میکرومتر (نصف عرض بدن در ناحیه شکاف تناسلی). دم مخروطی با خمیدگی شکمی با انتهای گرد و باریک. فاسمید مشاهده نشد.

    Cucumber mosaic virus (CMV) infects more than 1300 plant species that causes reduction in yield and quality of crops. In this research, the application of the hairpin-mediated RNA silencing technology was investigated for obtaining resistance to CMV. Therefore, a fragment of 145 nt from the silencing suppressor gene of CMV was selected. The CMV sequences from NCBI were aligned and the consensus sequence was selected and synthesized. This gene sequence was cloned as sense and antisense in pFGC5941 binary vector and the recombinant vector named pFGC-2b-h. The obtained plasmid was transferred to GV3101 agrobacterium strain. Transient assay on bean (Phaseolus vulgaris) and Nicotiana benthamiana was performed by the agrobacterium containing pFGC-2b-h and six days later plants were mechanically inoculated by CMV. Fifteen days postinoculation, plants were assessed using symptoms appearance and ELISA test. Results showed that the construct can effectively reduce the symptoms and virus accumulation level. Furthermore, the results suggested that the pFGC-2b-h construct can be used to obtain transgenic plants resistance to CMV.

    Barley yellow striate mosaic virus (BYSMV) belongs to the genus Cytorhabdovirusin the family Rhabdoviridae. It causes mosaic and chlorotic striation in gramineous plants and is transmitted by Laodelphax striatellus in a propagative manner. In the present study, nucleotide sequences ofglycoprotein and phosphoprotein genes of BYSMV Zanjan isolate are reported. Following thepurification of viral nucleocapsid, the nucleotide sequence of G and P genes was determined using random-PCR method (rPCR) followed by PCR with specific primers. Putative amino acid sequences of the two proteins were aligned with other members of Rhabdoviridae using CLUSTAL W software and the phylogenetic trees were depicted. BYSMV G and P proteins showed the highest similarity to the Northern cereal mosaic virus (NCMV). In phylogenetic analysis, the viruses were grouped into two main clusters reflecting the Cytorhabdovirus and Nucleorhabdovirus genera. These results support our previous phylogenetic analysis based on polymerase gene.Analysis of transmission efficiency showed that Zanjan, Naghadeh and Sero isolates were transmitted with a higher efficiency than Mashhad isolate by the same biotype of L. striatellus.

    Prevalence of effective sources of Wheat streak mosaic virus (WSMV) and Barley yellow dwarf virus (BYDV-PAV) and their mixed infections have been reported in several wheat growing regions in Iran. This research, investigates the effect of above mixed infections on viral titer and severity of one of the challenging viruses, WSMV, along with some yield components in three different wheat genotypes. The experiment was conducted in factorial design based on RCBD with 15 replications. Variables were viral treatments (six levels), genotypes (three levels) and the days post infection (DPI, three levels) of WSMV. The main and interactional effects of variables on above features were studied. Results showed that all doubly infections caused higher WSMV symptom scores and virus titer, those were accompanying with lower agronomic features. Among viral treatments, WSMV→BYDV-PAV inoculation and between genotypes, WSMV susceptible cultivar, Marvdasht, showed the maximum ELISA values and symptom scores. The highest viral content and symptom score were evaluated at 21 and 35 days post WSMV inoculation, respectively. Mutual interaction analysis between genotypes and viral treatments revealed the highest ELISA value and symptom score for WSMV→BYDV-PAV in Marvdasht and Rasul cultivars. Three ways interaction analysis of the above factors showed the highest ELISA value and symptom score for WSMV→BYDV-PAV treatment for Marvdasht and Rasul cultivars at 21 DPI. All doubly infections, particularly co-inoculation, resulted in the lower plant height, grain yield and shoot weight, comparing to single infections. For WSMV resistant genotype, Cross Adl, BYDV-PAV→WSMV inoculation was resulted in the higher ELISA value comparing to WSMV single infection, while, significant decrease in grain yield was observed in co-inoculation of two viruses.

    BYDV-PAV is one of the major agents causing yellowing and dwarfing of cereals worldwide. Previous study has been report the wide distribution of BYDVs in Iran and the prevalence of BYDV-PAV in cereal fields of the country. Due to the importance of this virus, and to investigate the taxonomic position of the Iranian isolate, the full length sequence of an Iranian severe isolate of BYDV-PAV from Kerman province (KP771878) was determined. Total RNA was extracted usingRNX (-plus)TM Kit. Nucleic acid extracts were reverse transcribed and amplified using PAV specific primers. The amplified fragments were inserted into PTZ57R/T vector using Ins T/A clone PCR Product Kit (MBI Fermentas) and cloned in Escherichia coli (DH5α) and sequenced. Complete nucleotide sequence of this isolate comprises 5668 nucleotides. Sequence analysis at nucleotide level showed that the Kerman isolate has maximum homology of about 94% with two American isolates (AF235167 and EF043235) of BYDV-PAV and grouped in the same cluster, while it showed the least similarity with a Chinese isolate of BYDV-PAV (AY855920) with only 78% homology. In addition, nucleotide sequence comparison of each ORF showed that the ORF4 was the most conserved while the ORF6 was the most divergent regions.

    Six populations of Rhizoctonia solani AG-1 IA were sampled from three naturally infested rice fields in Rasht, Tonekabon and Amol counties, in north of Iran. Allele fragment analyses were performed using fluorescent-labeled primers from nine microsatellite (SSR) loci. The ranges of allele and genotype frequencies were from 0.002 to 0.962 and from 0.40 to10.32 percent respectively among 252 isolates. The frequency of 13, 11 and 9 alleles was zero on five, six and three loci across geographical populations of Rasht, Amol and Tonekabon respectively, so from them two loci werent in Hardy–Weinberg Equilibrium (HWE) in each population.While the allele 145 bp at a locus TC12 was fixed in the Amol populations at a frequency of 1.0. As there was populations didnt show an even distribution of allele and genotype frequencies. This situation represents the gradual fixation and removal of some alleles in populations of the pathogen in the paddies of northern Iran.

    While sampling from potato tubers infected with potato pink rot in Fars province's potato stores, several Phytophthora isolates were recovered which differed from those known to cause pink rot. Various gene analyses including three nuclear (internal transcribed spacers (ITS), ß-tubulin and heat shock protein 90) and one mitochondrial (cytochrome c oxidase subunit I) gene regions showed these isolates were hybrids between Phytophthora cryptogea and P. erythroseptica. Phylogenetic analysis using Bayesian approach showed the causal agent of potato pink rot, P. erythroseptica and P. cryptogea GII form a monophyletic group which is independent of major members of Phytophthora clade 8. Cloning of the ß-tubulin region produced both parental ß-tubulin sequences and confirmed the hybrid nature of these isolates. Isolates produced non-papillate and ellipsoid to ovoid sporangia, globose and aplerotic oogonia, and amphigynous terminal antheridia. While there were some differences, based on morphological characteristics, these isolates were most similar to their maternal parent, P. erythroseptica.

    Marigold, Calendula officinalis, due to having various medicinal compounds is of high importance in pharmaceutical industry. In the present research, the influence of different initial inoculum levels of Meloidogyne javanica (0.0, 0.5, 1.0, 1.5, 2.0, 2.5 and 5.0 eggs and second stage juveniles/g. soil) on growth parameters of pot marigold and nematode reproduction was investigated. 90 days after nematode inoculation, the chemical compounds quercetin, kaempferol and ursolic acid were measured in infected plants. The results showed that with increasing the initial nematode population, the means of plant growth factors significantly decreased. In addition, nematode reproduction factor was inversely related to the levels of population and the highest reproduction factor was observed with the lowest nematode population. Results of analysis and comparison of chemical compounds of infected and healthy plants, showed that flavonoid and triterpenoid compounds namely quercetin and ursolic acid were increased in the nematode infected plants.

    GF-677 (prunus amygdalus × prunus persica) is one of the most suitable rootstocks for almond and peach trees. GF-677 cuttings were imported from Europe and planted in Bidzard and Estahban areas (Fars province) for further propagation and distribution to other Iranian stone fruit growing areas. During 2009 and 2010 surveys of GF-677 trees, witches’-broom disease (GFWB) was observed in both Bidzard and Estahban regions. The disease agents of GFWB from Bidzard (BGFWB) and Estahban (EGFWB) were transmitted from GF-677 to GF-677, bitter almond and peach seedlings by grafting and from experimentally inoculated bitter almond to seed-grown eggplant and periwinkle plants via dodder inoculation, causing phytoplasma-type symptoms. Total DNA extracted from naturally affected GF-677 trees in Bidzard and Estahban and experimentally inoculated plants reacted positively in direct PCR using P1/P7 and nested-PCR using P1/P7 (first round) and R16F2n/R16R2 (second round) primer pairs. On the basis of disease symptoms, positive reaction in PCR, transmission by dodder and grafting, BGFWB and EGFWB have phytoplasmal etiology. Blast searching showed that BGFWB and EGFWB phytoplasmas belong to the pigeon pea witches` broom (16SrIX) group. Using AluI, HpaII and HaeIII enzymes, RFLP patterns of BGFWB and EGFWB phytoplasmas were indistinguishable from each other and those of Neyriz (Fars, Iran) almond Witches’- broom (NAWB) phytoplasma. P1/P7 PCR ampliconsequences of BGFWB and EGFWB isolates were submitted to the GenBank database under the accession numbers JX445141 and JX445142, respectively. Virtual RFLP using 17 enzymes showed that BGFWB and EGFWB isolates are indistinguishable from NAWB phytoplasma (representative of 16SrIX-B subgroup) but different from Khafr (Fars, Iran) almond witches` broom (KAWB) phytoplasma which is classified with 16SrIX-C subgroup phytoplasmas. Phylogenetic analysis and homology percent, confirmed that among 16SrIX subgroups, BGFWB and EGFWB phytoplasmas were more similar to subgroup B of 16SrIX group than other subgroups.

    Host preference of Helicotylenchus digitiformis Ivanova, 1967 on 10 common field crops including cantaloupe (Samsuri), canola (Golden), clover, corn (704-Hybrid), cotton (Deltapine 16), lucerne (Hamedani), sugar beet (Pb8), tomato (Rutgers), turf grass (Sport) and wheat (Shiraz) was studied in two separate trials. Tests were performed in randomized complete block designs, with initial field populations of 1.49 and 4.33 nematodes/g of soil, under greenhouse conditions. The results showed that all plants are hosts for the nematode but the highest final population and reproduction factor were happened on the cotton (Deltapine 16) and corn (hybrid 704). In addition, to determine the damaging levels of the nematode on corn (hybrid 704) and cotton (Deltapine 16), two tests were conducted in randomized complete block designs with initial field population, ½ field population and control (4.33, 2.16 and Zero nematodes/g of soil, respectively) and six replicates, under greenhouse and microplot conditions. The results showed that all of the field populations under greenhouse conditions and treatments with nematode under microplot conditions caused significantly decreases in most of growth indices of corn plants. Nevertheless, the nematode population has not any significant effect on the growth indices of cotton, although the nematode multiplied on it and reproduction factor was more than one. Also, similar tests were carried out on two local wheat cultivars (Shiraz and Pishtaz), with field population, ½ field population, 1/3 field population and control (1.49, 0.75, 0.50 and Zero nematodes/g of soil, respectively) in a randomized complete block design with five replicates, under microplot conditions. Results showed that only the field population significantly caused decreases in most of growth indices of wheat (Pishtaz).

    Leaf rust caused by Puccinia triticina Eriksson is a major foliar disease of wheat, resulting in yield loss all over the world. In order to study the resistance components to leaf rust, a collection of 31 wheat cultivars was screened under controlled conditions at seedling stage inoculated with an isolate (race CHHLS) which had been collected from Lorestan province in complete randomized block design with 3 replications. Latent period was determined by the number of days after inoculation to the appearance of first pustule on the leaf 8 days after inoculation started and continued until 12 days after inoculation. Infection types on primary leaves were rated 12 days after inoculation using the Roelfs method in a scale of 0-4. The cultivar Orom, showed the lowest infection type, the longest latent period however, the cultivar Bolani had the shortest latent period and the highest infection type. Analysis of variance showed that there was a significant difference for infection type, but not for latent period among cultivars. There was a strong negative correlation (r= - 0.6463) between infection type and latent period. Cluster analysis based on ward’s method, categorized the cultivars into three groups as Resistant (Khazar1, Golestan, BKRB, Bam, Neyshabour, BKRZ, Hirmand and Oroum), Moderately Resistant (Sistan, Kavir, Parsi, Navid, Moghan1, Moghan2, Mahdavi, Aflak, Mihan, Alamout, Bahar, Darab2, Shiraz, Chanab, Alborz and Falat) and Susceptible (Zare, moghan3, Atrak, Zarin, Arg, Arvand and Bolani).

    In order to identify the plant parasitic nematodes of wheat fields of Boyer-Ahmad and Dena regions, Kohgiluyeh and Boyer-Ahmad province, Iran, 50 soil and root samples were collected during 2012-2013. By using sieve series and centrifugal flotation technique, the nematodes were extracted. They were fixed and transferred to anhydrous glycerin and the permanent slides were prepared from the extracted nematodes. Morphological and morphometrical studies were done, using a drawing tube attached to a light microscope. Among 28 identified species, Ditylenchus equalis as a new report for nematode fauna of Iran and D. tenuidens are described here.

    Fire blight is one of the most destructive diseases of stone and pome fruits. The causal pathogen is Erwiniaamylovora, species in the family Enterobacteriaceae. Pears are the most susceptible, but apples, quinces and some other rosaceous plants are also vulnerable. In Iran, the disease has been reported for the first time in Baraghan in Alborz province (Zakeri & Sharifnabi 1991). Fire blight produces several different types of symptoms like blossom blight, necrosis of fruits and leaf tissues, and dark sunken cankers on stems and exudation of droplets of bacterial ooze from the affected plant tissues. Typical symptoms of fire blight were observed on quince (Cydonia oblonga) trees, in Shahrbabak in Kerman province in 2013. Dieback of terminal shoots and dark necrotic lesions on immature fruits were among the major symptoms observed. Samples containing regions of dead and healthy tissues were washed, macerated in drops of sterile distilled water and bacteria were isolated on sucrose nutrient agar (NAS) medium. After 72 h of incubation at 27°C, single whitish and round colonies were purified by sub-culturing on eosine methylene blue agar (EMB agar). Results of morphological and biochemical tests, demonstrated that the bacterium belongs to Enterobacteriaceae (Schaad et al. 2001). Pathogenicity was tested by injecting healthy young quince shoots with a 10-μl bacterial suspension of 107CFU/ml. Controls were injected with sterile distilled water. Shoots were kept at 27°C and 80 to 100% relative humidity. Dark brown-to-black lesions were observed five days after inoculation on the inoculated shoots. The bacterium was reisolated from the lesions formed on the inoculated shoots. No lesions were observed on the controls. All of the twenty strains used in tests for identification, produced white colonies on NAS, were gram-negative, induced a hypersensitive reaction in tobacco (Nicotiana tabacum L. ‘White Burley’) leaves, did not produce a fluorescent pigment on medium B of King and did not grow at 37°C. On the basis of the symptoms, colony morphology and biochemical features, the pathogen was identified as E. amylovora. To our knowledge, this is the first report on the incidence of fire blight on quince in Kerman province.

    During a survey conducted in apple orchards of Boyer-Ahmad and Dena regions, in summer of 2013 and 2014, a species of Tylenchus was recovered which was identified as T. naranensisMaqbool, Zarina & Ghazala, 1987 based on morphological and morphometric studies. This is a new report for Iran and describe here. Female: Ventrally curved to C-form, head continuous with body contour, cephalic framework weakly sclerotized with 4 annules, 2.3–3.4 µm high and 5.3–6.6 µm wide. Stylet delicate with conus in 5.3-5.7 µm, about 46–50% of total length, with small rounded to posteriorly directed knobs. Dorsal esophageal gland orifice at 0.8–1.4 µm of stylet base. Body cuticle finely striated, striations 1.0–1.5 µm apart in mid-body; lateral field 4.2–5.4 µm, about 19–26% of body width; four lines in lateral field. Procorpus cylindrical, as long as isthmus; median bulb oval and muscular, located at first half of the esophagus, 6.6–8.5 µm in width, with distinct cuticular thickenings; isthmus slender, nerve ring located at first half of isthmus, 64-86 µm from the anterior extremity. Hemizonid 1–2 annules anterior to excretory pore. Deirids near to excretory pore, 90-106 µm from the head apex.Excretory pore near anterior end of basal bulb; terminal bulb elongated, 8.0–9.5 µm across and 19–25 µm long; Vagina 6–12 µm and 30–40% of vulval body diameter; vulva with projected lips.
