فاکتور سلول بنیادی (SCF) یک فاکتور رونویسی است که نقش اساسی در تکثیر، تمایز و بقای سلول های بنیادی دارد.
هدفهدف از این مطالعه بررسی اثر غلظت های مختلف فاکتور سلول بنیادی بر روی تکثیر سلول های CD146+ اندومتر انسانی بود.
مواد و روش هادر این مطالعه، سلول های CD146+ اندومتر از سوسپانسیون سلول های اندومتر انسانی کشت شده در پاساژ چهارم با MACSجداسازی شدند. سلول های CD146+ اندومتر انسانی تعیین کاریوتایپ شدند و سپس به منظور بررسی اثر تکثیر فاکتور سلول بنیادی بر روی سلول های CD146+ اندومتر انسانی با غلظت های مختلف 0، 5/12، 25، 50 و 100 نانوگرم بر میلی لیتر آزمایش شدند و با روش MTT مورد ارزیابی قرار گرفتند.
نتایجآنالیز کروموزومی سلول های کشت شده، گسترش متافازی نرمال XX46 را از خود نشان دادند. میزان تکثیر سلول های CD146+. اندومتر انسانی در حضور 0، 5/12، 25، 50 و 100 نانوگرم بر میلی لیتر فاکتور سلول بنیادی به ترتیب 094/0±945/0، 151/0±962/28، 00/0±988/0، 012/0±679/1، 145/0±129/1 بود. افزایش چشم گیری در تکثیر سلول های بنیادی استرومای اندومتر به دنبال کشت با 50 نانوگرم بر میلی لیتر نسبت به سایر غلظت های فاکتور سلول بنیادی بود (01/0=p).
نتیجه گیریدر مجموع تحقیق حاضر نشان داد که فاکتور سلول بنیادی می تواند بر روی تکثیر و بقای سلول های CD146+ اندومتر انسانی اثر گذارد که خود کاربردهای مهمی در پزشکی و سلول درمانی دارد.

استرس یک علت ناباروری در مردان است. اگر چه وجود هورمون جنسی و کیفیت اسپرم نشان داده شده است که در استرس پایین است، فیزیولوژی اسپرم و پروتئین های عملکردی بیضه، مانند پروتئین فسفوتیروزین، هنوز در این شرایط گزارش نشده است.
هدفهدف از انجام مطالعه، بررسی وضعیت آکروزوم و تغییرات پروتئین بیضه در استرس مزمن در روند اسپرماتوژنز و تولید تستوسترون در موش صحرایی بود.
مواد و روش هاموش های نر به دو گروه شاهد و استرس مزمن (CS) تقسیم شدند (7=n). موش هایCS به مدت 42 روز متوالی بی حرکت شدند (4 ساعت در روز). سطح گلوکز خون (BGL)، کورتیکوسترون، تستوسترون، وضعیت آکروزوم و هیستوپاتولوژی مورد بررسی قرار گرفت. بیان استروییدساز حاد نظارتی بیضوی (StAR)، زنجیره جانبی سیتوکرومP450)CYP11A1(، و پروتئین های فسفریله شده مورد ارزیابی قرار گرفت.
نتایجدر مقایسه بین دو گروه (شاهد در مقابل CS) BGL (71/25±2/22 در مقابل 95/60±36/3 mg/dl)، سطح کورتیکوسترون (4/23±24/33 در مقابل 2/01±36/9 ng/dl)، اسپرم واکنش آکروزومی نشان داده (1/55±3/25 در مقابل 17/71±5/03%)، و اختلال سر اسپرم (0/71±3/29 در مقابل 6/21±1/18%) در گروه CS به طور قابل توجهی بالاتر بود. در مقابل، کیسه منی (0/05±0/41 در مقابل 0/24±0/07 g/100g)، سطح تستوسترون (0/79±3/37 در مقابل 0/61±0/29 ng/dl) و غلظت اسپرم (7/70±115/33 در مقابل 3/65±79/13×106 میلی لیتر/ سلول) در موش های گروه استرس به طور قابل توجهی پایین تر (0/05˂p) نسبت به گروه شاهد بود. برخی از لوله های اسپرم ساز آتروفیک و توده اسپرم کم در موش های گروه استرس آشکار شد. بیان CYP11A1 به جز پروتئین StAR به طور قابل توجهی در موش گروه استرس کاهش یافت. در مقابل، یک پروتئین فسفریله 55 کیلو دالتون در بیضه گروه استرس بالاتر بود.
نتیجه گیریاسترس مزمن بیان CYP11A را کاهش داد که منجر به کاهش تستوسترون و افزایش واکنش آکروزوم اسپرم می شود که به نظر می رسد در نتیجه افزایش پروتئین فسفریله شده 55 کیلو دالتون باشد.

شکست لانه گزینی یکی از مهم ترین عوامل محدودکننده موفقیت در درمان نازایی می باشد. اکثر مقالات نشانگر تاثیر مطلوب خراش اندومتر در موفقیت لانه گزینی به ویژه در زنان با شکست مکرر لانه گزینی می باشند.
هدفهدف ازاین مطالعه نشان دادن تاثیر خراش اندومتر بر نتایج حاملگی در سیکل های برگشت از فریزمی باشد.
مواد و روش هاما یک مطالعه آینده نگر بر روی 93 بیمار مراجعه کننده به مرکز ناباروری ترتیب دادیم. زنان مورد مطالعه قرار بود درسیکل برگشت از فریز قرار بگیرند. زنان درگروه مداخله تحت خراش اندومتربا کاتترپایپل درسیکل قبل از کاشت فریز قرار گرفتند. پیامدهای اولیه شامل میزان حاملگی بالینی و میزان لانه گزینی بودند.
نتایج45 بیمار در گروه مداخله با 48 بیمار در گروه کنترل مورد مقایسه قرار گرفتند. تفاوت آماری معنی داری در ویژگی های پایه و سیکلیک بین دو گروه وجود نداشت. تفاوت در میزان لانه گزینی از نظر آماری تقریبا معنی دار بود. میزان حاملگی شیمیایی و بالینی و ادامه یافته در گروه مداخله کمتر از گروه کنترل بود. میزان حاملگی دوقلویی و میزان سقط در گروه مداخله کمتر از گروه کنترل بود.
نتیجه گیریبر اساس نتایج این مطالعه خراش اندومتر در سیکل قبل از کاشت جنین فریز تاثیر منفی بر میزان لانه گزینی و میزان حاملگی بالینی داشت.

آنتی ژن G لکوسیت انسانی ((HLA-G جزو خانواده I غیرکلاسیک بوده و توسط سلول هایExtravillous Cytotrophoblast به میزان زیادی بیان می شود. در صورت حذف یک جفت باز در ژن آن، عملکرد آن می تواند توسط ایزوفرم های دیگر مانند HLA-G2 جبران شود. بررسی فراوانی آلل نول در افراد با سابقه سقط مکرر می تواند نتیجه مفید در درک رابطه بین فراوانی این آلل و سقط های مکرر در جمعیت ها ارایه دهد.
هدفدر این مطالعه، هدف ارایه فراوانی آلل نول HLA-G* 0105N و بررسی ارتباط بالقوه این فراوانی با کاهش بیان آن در افراد مبتلا به سقط مکرر بود.
مواد و روش هااز روش وسترن بلاتینگ برای بررسی میزان بیان پروتئین HLA-G استفاده شد. برای بررسی فراوانی آلل نول HLA-G* 0105N در افراد مبتلا به سقط مکرر، از روش PCR-RFLP استفاده گردید. اگزون 3 ژن HLA-G با روش PCR تکثیر گردید. بدنبال آن با استفاده از آنزیم PpuM-1 محصول PCR برش داده شده و بر روی ژل آگارز الکتروفورز انجام و سپس آنالیز گردید.
نتایجبرش با آنزیم محدودکننده وجود 10% هتروزیگوس برای آلل نول HLA-G* 0105N در افراد را نشان داد. همچنین نتایج وسترن بلاتینگ کاهش در بیان ژن HLA-G در بافت جفت افراد مبتلا به سقط مکرر در مقایسه با افراد با تولد نرمال را نشان داد.
نتیجه گیریفراوانی حالت هتروزیگوس آلل نول HLA-G* 0105N در افراد مبتلا به سقط مکرر جنین بالاست. نسبت جهش در این افراد، وجود یک ارتباط بین سقط مکرر و فراوانی جهش در آلل مذکور را پیشنهاد می کند.

انتقال جنین فریز روی اندومتر طبیعی می تواند در بیماران با سابقه عدم لانه گزینی مکرر در سیکل های IVF با فوایدی همراه باشد که می تواند به دلیل تفاوت در سیتوکین ها و فاکتورهای رشد مترشحه در اندومتر طبیعی و اندومتر تحریک شده با هورمون باشد.
هدفهدف از این مطالعه مقایسه میزان حاملگی جنین های فریز منتقل شده روی اندومترطبیعی با اندومتر تحت در مان هورمونی در بیماران با سابقه عدم لانه گزینی مکرر در سیکل های IVF می باشد.
مواد و روش هادر این مطالعه گذشته نگر میزان حاملگی در بیماران با سابقه عدم لانه گزینی مکررو تحت انتقال جنین روز سوم روی اندومتر طبیعی (گروه 1، تعداد= 56) با موارد انتقال جنین روی اندومتر تحت در مان هورمونی(گروه 2، تعداد= 52) مقایسه شدند.
نتایجمیزان حاملگی در گروه 1، %41/07 (23 نفر از 56) در مقایسه با گروه 2، %36/5 بدست امد (0/63=p). میزان سقط در دو گروه تفاوت معنی داری نداشت (0/60=p).
نتیجه گیریدر موارد عدم لا نه گزینی مکرر انتقال جنین فریز روی سیکل طبیعی با انتقال جنین روی سیکل های تحت درمان هورمونی قابل مقایسه است.

رحم بافتی پویا در طول چرخه تولیدمثلی در پاسخ به تغییرات هورمونی است. به این ترتیب، سلول های بنیادی رحم در سال های اخیر مورد مطالعه قرار گرفته است.
هدفتحقیق حاضر، بیان Oct4 و Sox2 در بافت رحم موش اوریکتومی تیمار شده با هورمون های استروییدی را ارزیابی کرد.
مواد و روش هادر این مطالعه تجربی، موش های ماده باکره اوریکتومی شدند و با استرادیولβ 17 )E2(، پروژسترون (P4) و ترکیب E2 و P4 (E2&P4) برای مدت 5 روز تیمار شدند. بافت های رحم جدا شدند و رنگ آمیزی ایمونوهیستوشیمی (IHC) و PCR کمی زمان واقعی مارکرهای Oct4 و Sox2 انجام شد.
نتایجنتایج IHC نشان داد که هر دو Oct4 و Sox2 در آندومتر و میومتر رحم تمام گروه ها بیان شد. بیان Oct4 و Sox2 در گروه تیمار شده با E2 به طور معنی داری در مقایسه با گروه کنترل کاهش یافت. در مقابل، بیان Oct4 و Sox2 در گروه های تیمار شده با P4 و E2&P4 تفاوت معنی داری در مقایسه با گروه کنترل نشان ندادند.
نتیجه گیرینتایج نشان می دهد هورمون های استروییدی تخمدان بیان Oct4 و Sox2 را در بافت رحم موش تغییر می دهد که بیانگر دخالت تنظیم هورمون استروییدی در سلول های بنیادی رحم می باشد.

HLA-E یک آنتی ژن غیرکلاسیک MHC کلاس یک است که بر روی سلول های سیتوتروفوبلاست بیان می شود که با اتصال به رسپتورهای مهاری سلول های کشنده طبیعی (NKG2A)، منجر به کاهش پاسخ های ایمنی در سطح ارتباطی مادر-جنین می شود وتحمل ایمنی مادر به جنین را فراهم می کنند.
هدفاین مطالعه به منظور بررسی فراوانی واریانت های عملکردی E0101، E0103 در ژن HLA-E در زنان یزدی مبتلا به سقط مکرر صورت گرفته است.
مواد و روش هاپلی مورفیسم E0101، E0103 در ژن HLA-E در بیماران وکنترل با استفاده از روش ARMS-PCR انجام شد.
نتایجداده ها به ما نشان می دهد که تفاوت معناداری در فراوانی آلل ها از ژن HLA-E بین بیماران مبتلابه سقط مکرر وزنان کنترل بارور وجود ندارد. ژنوتیپ هتروزیگوت HLA-E 0101/0103 در افراد مبتلا به سقط مکرر به طور فابل توجهی بالاتر بود (73/1=OR، 006/0=p). بنابراین این ژنوتیپ ممکن است استعداد ابتلا به سقط مکرر خود به خود را افزایش دهد.
نتیجه گیریبر خلاف ژنوتیپ هتروزیگوت HLA-E 0101/0103، ژنوتیپ هموزیگوت HLA-E 0103/0103 باعث کاهش خطر سقط مکرر می شود. به هر حال نتایج ما نشان می دهد که HLA-E 0103/0103 نقش حفاظتی بر علیه سقط مکرر از خود نشان می دهد و بین این ژنوتیپ و بهبود سقط مکرر ارتباط وجود دارد (0/56=p=0/013,OR).

مقدمه:
پارگی رحم در بارداری عارضهای نادر است و اغلب تهدید کننده حیات بوده و با بروز بالای عوارض و مرگ و میر مادری و جنینی همراه است. تقریبا تمام موارد پارگی رحم به دنبال میومکتومی در سه ماهه سوم بارداری یا در طول زایمان رخ میدهد.
مورد:
مادر بارداری است که در هفته 20 بارداری با درد شکم مراجعه نمود که سابقه قبلی میومکتومی شکمی را در 6 سال پیش داشته است. رحم دچار پارگی شده بود و جنین همراه با کیسه آب در داخل حفره شکم قرار داشت.
نتیجه گیریتشخیص زودهنگام پارگی رحم در مواردی از میومکتومی که منجر به باز شدن رحم شده باشد، می تواند جان بیمار را از مرگ حتمی نجات دهد.

Stem cell factor (SCF) is a transcriptional factor which plays crucial roles in normal proliferation, differentiation and survival in a range of stem cells.
ObjectiveThe aim of the present study was to examine the proliferation effect of different concentrations of SCF on expansion of human endometrial CD146 cells.
Materials And MethodsIn this experimental study, total populations of isolated human endometrial suspensions after fourth passage were isolated by magnetic activated cell sorting (MACS) into CD146 cells. Human endometrial CD146 cells were karyotyped and tested for the effect of SCF on proliferation of CD146 cells, then different concentrations of 0, 12.5, 25, 50 and 100 ng/ml was carried out and mitogens-stimulated endometrial CD146 cells proliferation was assessed by MTT assay.
ResultsChromosomal analysis showed a normal metaphase spread and 46XX karyotype. The proliferation rate of endometrial CD146P P cells in the presence of 0, 12.5, 25, 50 and 100 ng/ml SCF were 0.945±0.094, 0.962±0.151, 0.988±0.028, 1.679±0.012 and 1.129±0.145 respectively. There was a significant increase in stem/ stromal cell proliferation following in vitro treatment by 50 ng/ml than other concentrations of SCF (p=0.01).
ConclusionThe present study suggests that SCF could have effect on the proliferation and cell survival of human endometrial CD146P cells and it has important implications for medical sciences and cell therapies.

Stress is a cause of male infertility. Although sex hormones and sperm quality have been shown to be low in stress, sperm physiology and testicular functional proteins, such as phosphotyrosine proteins, have not been documented.
ObjectiveTo investigate the acrosome status and alterations of testicular proteins involved in spermatogenesis and testosterone synthesis in chronic stress in rats.
Materials And MethodsIn this experimental study, male rats were divided into 2 groups (control and chronic stress (CS), n=7). CS rats were immobilized (4 hr/day) for 42 consecutive days. The blood glucose level (BGL), corticosterone, testosterone, acrosome status, and histopathology were examined. The expressions of testicular steroidogenic acute regulatory (StAR), cytochrome P450 side chain cleavage (CYP11A1), and phosphorylated proteins were analyzed.
ResultsResults showed that BGL (71.25±2.22 vs. 95.60±3.36 mg/dl), corticosterone level (24.33±4.23 vs. 36.9±2.01 ng/ml), acrosome reacted sperm (3.25±1.55 vs. 17.71±5.03%), and sperm head abnormality (3.29±0.71 vs. 6.21±1.18%) were significantly higher in CS group in comparison with control. In contrast, seminal vesicle (0.41±0.05 vs. 0.24±0.07 g/100g), testosterone level (3.37±0.79 vs. 0.61±0.29 ng/ml), and sperm concentration (115.33±7.70 vs. 79.13±3.65×106 cells/ml) of CS were significantly lower (p
ConclusionCS decreased the expression of CYP11A, resulting in decreased testosterone, and increased acrosome-reacted sperm, assumed to be the result of an increase of 55 kDa phosphorylated protein.

Implantation failure is one of the most important factors limiting success in IVF treatment. The majority of trials have demonstrated favorable effect of endometrial injury on implantation success rate especially in women with recurrent implantation failure, while some studies failed to detect any benefit.
ObjectiveThe purpose of our trial was to explore whether endometrial injury in luteal phase prior to frozen-thawed embryo transfer cycles would improve pregnancy outcomes?
Materials And MethodsWe conducted a prospective controlled trial of 93 consecutive subjects at a research and clinical center for infertility. All women were undergone frozen-thawed embryo transfer (FTE) cycles. Women in the experimental group underwent endometrial biopsy with a Pipelle catheter in luteal phase proceeding FET cycle. Primary outcomes were implantation and clinical pregnancy rates and secondary outcomes were chemical, ongoing and multiple pregnancy and miscarriage rates.
Results45 subjects who underwent endometrial injury (EI) were compared with 48 control group which did not include any uterine manipulation. There were no significant differences in baseline and cycle characteristics between two groups. The difference in implantation rate was trend to statistically significance, 11.8% in EI group vs. 20.5% in control group (p=0.091). The chemical, clinical and ongoing pregnancy rates were lower in EI group compared with control group but not statistically significant. The multiple pregnancy rate and miscarriage rate also were lower in EI group compared with control group.
ConclusionBased on results of this study, local injury to endometrium in luteal phase prior to FET cycle had a negative impact on implantation and clinical pregnancy rates.

Human leukocyte antigen-G (HLA-G) is a non-classical class I molecule highly expressed by extravillous cytotrophoblast cells. Due to a single base pair deletion, its function can be compensated by other isoforms. Investigating the frequency of null allele in Recurrent Miscarriage (RM) subjects could be useful in understanding the relationship between frequency of this allele and RM in a given population.
ObjectiveThis study aimed to determine the frequency of HLA-G*0105N null allele and its potential association with down-regulation of HLA-G in subjects with RM.
Materials And MethodsWestern blotting was used to assess the level of HLA-G protein expression. For investigating the frequency of HLA-G*0105N null allele in RM subjects, PCR-RFLP method was used. Exon 3 of HLA-G gene was amplified by polymerase chain reaction (PCR). Subsequently, PpuM-1 enzyme was employed to digest the PCR products and fragments were analyzed using gel electrophoresis.
ResultsDigestion using restriction enzyme showed the presence of heterozygous HLA-G*0105N null allele in 10% of the test population. Western blotting results confirmed the decrease in expression of HLA-G in the placental tissue of subjects with RM compared to subjects who could give normal birth.
ConclusionThe frequency of heterozygous HLA-G*0105N null allele was high to some extent in subjects with RM. The mutation rate in subjects suggested that there is a significant association between RM and frequency of mutations in this allele.

Natural endometrium in Frozen-thawed Embryo Transfer (FET) may have some benefits upon implantation in patients with Repeated Implantation Failure (RIF). It might be due to possible differences between natural and stimulated endometrial growth factors and cytokins secretions.
ObjectiveThe objective of this study was to compare the pregnancy rate of FET on modified natural cycle versus hormone replacement therapy (HRT) cycle endometrium in patients with RIF.
Materials And MethodsIn this observational study the pregnancy rate of patients with RIF undergoing day 3 FET in natural cycle endometrium (group 1, n=56), were compared with another group of patients with RIF in whom frozen-thawed day 3 embryos were transferred on HRT cycle (group 2, n=52).
ResultsThe pregnancy rate in group 1 was 41.07%, compared with the pregnancy rate of group 2; 36.5% (p=0.63). The abortion rate was not significantly different among the groups.
ConclusionIt can be concluded that FET in a modified natural cycle is comparable with HRT cycle in patients with RIF.

The uterus is a dynamic tissue responding to hormonal changes during reproductive cycles. As such, uterine stem cells have been studied in recent years. Transcription factors oct4 and sox2 are critical for effective maintenance of pluripotent cell identity.
ObjectiveThe present research evaluated the mRNA expression of oct4 and sox2 in the uterine tissues of ovariectomized mice treated with steroid hormones.
Materials And MethodsIn this experimental study, adult virgin female mice were ovariectomized and treated with estradiol 17β (E2), progesterone (P4), and a combination of E2 and P4 (E2 & P4) for 5 days. Uterine tissues were removed, and immunofluorescent (IF) staining and quantitative real-time PCR of oct4 and sox2 markers were performed.
ResultsIF showed oct4 and sox2 expression in the uterine endometrium and myometrium among all groups. The mRNA expression of oct4 (p=0.022) and sox2 (p=0.042) in the E2-treated group significantly were decreased compared to that in the control group. By contrast, the mRNA expression of oct4 and sox2 in the P4 (p=0.641 and 0.489 respectively) and E2 & P4-treated groups (p=0.267 and 0.264 respectively) did not show significant differences compared to the control group.
ConclusionThe results indicate ovarian steroid hormones change the expression of oct4 and sox2 in the mice uterine tissues, which suggest the involvement of steroid hormonal regulation in uterine stem cells.

Human leukocyte antigen-E (HLA-E)is a non-classical major histocompatibility complex (MHC) class I antigens which expressed on extra villous cytotrophoblast, which interacts with NKG2A, is an inhibitory receptor on natural killer (NK) cells and leading to down regulation of immune response in the maternal-fetal interface and provides maternal immune tolerance of the fetus.
ObjectiveThis study was designated to investigate the gene frequencies of E0101 and E0103 in HLA-E gene in Iranian women with recurrent spontaneous abortion (RSA).
Materials And MethodsAmplification Refractory Mutation System (ARMS-PCR) technique was carried out to detect polymorphism in exon 3 of the HLA-E gene in women with RSA and controls (n=200). Differences between groups were analyzed by SPSS19 software using χ2 test.
ResultsThere was no significant difference in the allele frequencies of the HLA-E polymorphism between RSA and fertile controls but HLA-E 0101/0103 heterozygous genotype was found to be significantly higher in RSA group (p=0.006, OR=1.73), so this genotype might confer susceptibility to RSA.
ConclusionOur results suggest that HLA-E 0101/0103 heterozygous genotype leads to increase of RSA risk. It seems that by genotyping of HLA-E polymorphism, we can predict the risk of RSA in infertile women.

Uterine rupture in pregnancy is rare and often could be life threatening and catastrophic. Myomectomy is one of very common surgeries in gynecology, performed as the vaginal, abdominal and laparoscopic surgeries. Pregnancies occured after abdominal and laparoscopic myomectomy are high risk for uterine rapture.
Case:
Patient was a 28 Years old female, pregnant woman at the 20 wks of gestational age with abdominal pain and a history of abdominal myomectomy 6 yrs ago. Uterus was ruptured and fetus in amniotic sac was found in abdominal cavity.
ConclusionEarly diagnosis of uterine rupture after myomectomy can save patients from death.
