صفحات 149-157زمینه و هدفاستافیلوکوکوس اورئوس یک باکتری گرم مثبت، فاقد حرکت، بدون اسپور، هوازی و بی هوازی اختیاری است که در ناحیه قدامی بینی افراد کلنیزه می شود. کارکنان ناقل مشغول به کار در صنعت غذایی منبع اصلی برای گسترش بیماری های منتقله از غذا محسوب میشوند. انتروتوکسین استافیلوکوکی مهمترین فاکتور بیماری زایی دخیل در مسمومیت های غذایی استافیلوکوکی است. هدف از این مطالعه، شناسایی ژن های انتروتوکسین sea، seb و sec استافیلوکوکوس اورئوس در ناقلین سالم بود.مواد و روش کاردر این مطالعه توصیفی- مقطعی، تعداد 136 نمونه از سوآپ بینی پرسنل رستوران ها، قصابی ها، لبنیات فروشی ها و فست فودهای شهرستان اردبیل در بهار 1396 جمع آوری شد. نمونه ها پس از کشت اولیه، با آزمون های بیوشیمیایی تایید شدند. نمونه DNA ایزوله های استافیلوکوکوس اورئوس استخراج شده و ژنهای sea، seb و sec به عنوان مارکر برای ردیابی انتروتوکسین های SEA، SEB و SEC با روش PCR بررسی شدند.یافته هادرمجموع، از 136 نمونه سوآپ بینی، 46 نمونه (33/8%) ناقل استافیلوکوکوس اورئوس بودند که این میزان ناقلی با مثبت شدن ژن femA ثابت شد. از مجموع 46 ایزوله استافیلوکوکوس اورئوس، تعداد 11 سویه (23/9%) واجد ژن sea، 6 سویه (13%) واجد ژن seb و 5 سویه (10/8%) نیز واجد ژن sec بودند.نتیجه گیریدرصد قابل توجهی از پرسنل شاغل در زنجیره غذایی، ناقل استافیلوکوکوس اورئوس انتروتوکسیژنیک در بینی خود بودند. از این رو، برنامه غربالگری و کنترل این ناقلین به منظور جلوگیری از شیوع عفونت های استافیلوکوکی پیشنهاد می شود.کلیدواژگان: استافیلوکوکوس اورئوس، انتروتوکسین، PCR

صفحات 158-166زمینه و هدفروش انجماد یکی از بهترین روش های نگهداری طولانی مدت برای قارچ ها است و استفاده از مواد نگهدارنده مناسب می تواند موفقیت این فرآیند را افزایش دهد. هدف از این مطالعه بررسی تاثیر مواد نگهدارنده مختلف در حفاظت از مخمر کاندیدا گلابراتا در شرایط انجماد است.مواد و روش کاردر مطالعه مداخله ای-تجربی تعداد 50 ایزوله بالینی کاندیدا گلابراتا در دمای انجماد و در داخل مواد نگهدارنده متداول شامل: گلیسرول 10% و 40%، گلوکز 4% و دی متیل سولفواکساید 10% نگهداری شد. پس از اتمام دوره انکوباسیون 2 ساله، میزان موفقیت بازیابی نمونه ها در مواد نگهدارنده مختلف مقایسه و با استفاده از آمار توصیفی و آزمون آماری chi-square مورد ارزیابی قرار گرفت.یافته هاگلوکز 4% با توانایی نگهداری 100% نمونه ها به عنوان بهترین ماده محافظت کننده برای کاندیدا گلابراتا تعیین شد. همچنین گلیسرول 40% و دی متیل سولفواکساید 10% با بازیابی 94% نمونه ها قابلیت بالایی در نگهداری مخمر داشتند. درمقابل گلیسرول 10% کمترین اثر محافظتی را نشان داد. نمونه های کشت شده در لوله های شیب دار حاوی SDA و دمای ̊C4 با بازیابی 32 نمونه (64%) اختلاف معنی داری با نگهداری در دمای انجماد داشت. همچنین بازیابی نمونه ها در محیط SDA نتایج بهتری نسبت به محیط SDB به همراه داشت.نتیجه گیریباتوجه به اینکه مواد نگهدارنده مختلف از قابلیت یکسانی در حفظ کاندیدا گلابراتا در شرایط انجماد برخوردار نیستند، انتخاب نوع و غلظت مناسب ماده نگهدارنده می تواند میزان بقای مخمر را در شرایط انجماد افزایش دهد.کلیدواژگان: کاندیدا گلابراتا، انجماد، مواد نگهدارنده، گلیسرول، گلوکز، دی متیل سولفواکساید

صفحات 167-177زمینه و هدفپونه از جمله گیاهانی است که در صنایع مواد غذایی به عنوان طعم دهنده مصرف فراوانی داشته و به اثرات ضد باکتریایی آن توجه شده است. مطالعه حاضر با هدف بررسی خواص فیتوشیمیایی اسانس پونه (Mentha pulegium L.) به دست آمده از ایران و فعالیت ضد میکروبی آن برعلیه تعدادی از باکتری های بیماری زا انجام شد.مواد و روش کارترکیبات شیمیایی این گیاه دارویی با GC-MS بررسی شد و 16 ترکیب مختلف که در مجموع86/96% کل وزن اسانس گیاه را تشکیل می دهند، شناسایی شدند. فعالیت ضد میکروبی اسانس گیاه پونه در برابر باکتری های استافیلوکوکوس اورئوس، اشریشیا کلی، سودوموناس آئروژینوزا و سودوموناس سیرینگه پاتوار گوجه فرنگی به روش انتشار از دیسک در مقایسه با آنتی بیوتیک سیپروفلوکساسین ارزیابی شد. حداقل غلظت مهارکنندگی اسانس در برابر باکتری های مطالعه شده با استفاده از روش ماکروبراث دایلوشن بررسی شد.یافته هانتایج به دست آمده نشان داد که اسانس پونه از اجزای اصلی پیپریتون (32/1٪)، پیپریتنون (21/71٪) و پولگون (15/85٪) تشکیل شده است. به علاوه، نتایج به دست آمده با استفاده از MICو MBCنشان داد که حداقل غلظت مهارکنندگی اسانس پونه در محدوده mg/mL 40-10 برای باکتری های استافیلوکوکوس اورئوس، اشریشیا کلی، سودوموناس آئروژینوزا و mg/mL 20 برای سودوموناس سیرینگه است. با توجه به نتایج به دست آمده، بیشترین هاله عدم رشد (mm 29/15) در سودوموناس سیرینگه و کمترین هاله عدم رشد (mm 17/08) در سودوموناس آئروژینوزا در غلظت mg/mL 40 اسانس پونه مشاهده شد.نتیجه گیرینتایج به دست آمده حاکی از این است که اسانس پونه دارای اثر مهارکنندگی مناسبی برعلیه باکتری های استافیلوکوکوس اورئوس و اشریشیا کلی می باشد.کلیدواژگان: ضد باکتریایی، پونه، اسانس گیاهی، MIC، MBC

صفحات 178-183زمینه و هدفکلبسیلا پنومونیه که باعث عفونت های دستگاه گردش خون و ادرار می شود حاوی چندین ژن کدکننده عوامل بیمار ی زا است. عواملی مثل توانایی اتصال به سطوح وجود گیرنده های سیدروفور سیستم جذب آهن و تنظیم فنوتیپ مخاطی جزء اساسی ترین فاکتورهای دخیل در ایجاد بیماری از سوی کلبسیلا پنومونیه هستند. این مطالعه با هدف تعیین میزان حضور ژن های کدکننده این فاکتورهای بیماری زا در کلبسیلا پنومونیه جداشده از بیماران بستری شده با روش Multiplex-PCR انجام شد.مواد و روش کاردر این مطالعه توصیفی فراوانی ژن های بیماری زای irp2، wcaG، rmpA، allS و fimH در 100 ایزوله کلبسیلا پنومونیه جمع آوری شده از بیماران بستری در بیمارستان های آموزشی زاهدان با روش Multiplex-PCR در سال 1393بررسی شد.یافته هادرمجموع از 100 ایزوله کلبسیلا پنومونیه مطالعه شده، ژن allS با 90% بیشترین فراوانی و ژن rmpA با 6% کمترین فراوانی ژنی را داشتند. فراوانی ژنی برای ژن های fimH، irp2 و wcaG به ترتیب 88%، 87% و 33% گزارش گردید.نتیجه گیریalS، irp2، fimH و wcaG شایع ترین ژن های کدکننده فاکتورهای حدت در ایزوله های کلبسیلا پنومونیه جدا شده از بیماران تحت مطالعه در زاهدان است. این مسئله می تواند اطلاعاتی در رابطه با اهمیت آن ها در آسیب شناسی عفونت ها که منجر به دستیابی به درمان های موثر می شود در اختیار محققین قرار دهد.کلیدواژگان: کلبسیلا پنومونیه، فاکتورهای بیماری زایی، روش واکنش زنجیره ای پلیمراز

صفحات 184-191زمینه و هدفمقاومت به درمان آنتی بیوتیکی به ویژه در سویه های استافیلوکوکوس اورئوس مقاوم به متی سیلین یکی از نگرانی های عمده بهداشت عمومی در سراسر جهان است. مطالعه حاضر به سنتز نانوذرات سلنیوم، بررسی اثر ضد میکروبی و امکان استفاده از آن به عنوان نانوحامل آمپی سیلین می پردازد.مواد و روش کارنانوذرات سلنیوم به روش احیای شیمیایی سدیم سلنیت به وسیله اسیدآمینه ال - سیستئین سنتز شد. بارگذاری اتصال آمپی سیلین در سطح نانوذرات سلنیوم با اضافه کردن تدریجی آنتی بیوتیک به محلول نانوذرات و هم زدن مداوم انجام شد. سپس این اتصال با استفاده از طیف سنجی UV-Vis، پراش اشعه x و میکروسکوپ الکترونی روبشی تحت بررسی قرار گرفت. اثر ضد باکتری نانوذره حاصل و نانوذره بارگذاری شده با آنتی بیوتیک در برابر سویه استاندارد استافیلوکوکوس اورئوس و 10 سویه بالینی استافیلوکوکوس اورئوس مقاوم به متی سیلین به روش دیسک دیفیوژن و MIC به روش براث ماکرودایلوشن بررسی شد.یافته هااتصال آنتی بیوتیک در سطح نانوذرات سبب افزایش اثر ضد میکروبی آن ها نسبت به حالت عادی شد. کمترین غلظت مهارکننده رشد سویه های بالینی استافیلوکوکوس اورئوس در نانوذره سلنیوم بین 62/5-7/8، فرم آزاد آمپی سیلین 250-62/5 و آمپی سیلین متصل به نانوذرات 62/5-7/8 میکروگرم بر میلی لیتر بود. این مقادیر در مورد سویه استاندارد استافیلوکوکوس اورئوس به ترتیب 7/8، 31/2 و 3/9 میکروگرم بر میلی لیتر بود. اتصال آمپی سیلین در سطح نانوذرات سلنیوم سبب بهبود کمترین غلظت مهارکننده رشد استافیلوکوکوس اورئوس های مقاوم به متی سیلین شد.نتیجه گیرینتایج به دست آمده، تاییدکننده فعالیت ضد میکروبی نانوذرات سلنیوم است. افزایش فعالیت ضد میکروبی آنتی بیوتیک بارگذاری شده در سطح این ذرات، امکان استفاده از این آنتی بیوتیک ها را در درمان عفونت های سخت درمان افزایش می دهد.کلیدواژگان: نانوذرات سلنیم، اثر ضد میکروبی، استافیلوکوکوس اورئوس مقاوم به متی سیلین، آمپی سیلین

صفحات 192-202زمینه و هدفاستافیلوکوکوس اورئوس مقاوم به متی سیلین شایع ترین عامل باکتریائی عفونت زخم پای بیماران دیابتی (DFI) است. فاکتورهای بیماری زایی این باکتری شدت درجه عفونت زخم را افزایش می دهد و درصورت درمان نشدن به موقع منجر به قطع عضو اندام های تحتانی و مرگ بیماران مبتلا می شود. هدف این پژوهش، جداسازی باکتری استافیلوکوکوس اورئوس مقاوم به متی سیلین حامل ژن mecA و شناسایی 4 ژن ویرولانس hla ، lukED، sei و hlg از جدایه های عفونت پای بیماران دیابتی به منظور ارزیابی نقش آن ها در میزان شدت عفونت است.مواد و روش کاراز ترشحات چرک 30 بیمار مبتلا به عفونت پای دیابتی در استان مازندران نمونه گیری شد و استافیلوکوکوس اورئوس براساس ویژگی های کشت و تست های بیوشیمیائی خالص سازی شد. ارزیابی مولکولی جدایه ها پس از استخراج DNA، با 5 جفت پرایمر اختصاصی ژن های مقصود به روش PCR صورت گرفت.یافته هااز مجموع 30 بیمار 14 ایزوله (46/6%) استافیلوکوکوس اورئوس جداسازی و شناسایی شد. هر 14 نمونه حامل ژن mecA بودند و فراوانی ژن های hla ، lukED ، sei و hlg در جدایه ها به ترتیب 100%، 100%، 71/4% و 64/2% به دست آمد.نتیجه گیریشناسایی این 4 ژن به همراه ژن mecA در استافیلوکوکوس اورئوس جداسازی شده از زخم پای دیابتی می تواند مارکر ژنتیکی موثر و قابل اعتمادی در تشخیص درجه بندی عفونت پای بیماران دیابتی باشد. تشخیص عامل عفونت با روش PCR و درمان آنتی بیوتیکی مناسب و به موقع، در جلوگیری از قطع عضو بیماران دیابتی ضروری است.کلیدواژگان: استافیلوکوکوس اورئوس، مقاومت به متی سیلین، ژن های بیماری زایی، عفونت پای دیابتی

صفحات 203-209زمینه و هدفاکتینومایستال به گروهی از باکتری های گرم مثبت گفته می شود که ازنظر مورفولوژی دارای ساختمانی رشته ای با قطری حدود یک میکرون هستند. در این گروه، باکتری هایی از خانواده اکتینومایسس، نوکاردیا و استرپتومایسس جای می گیرند. در این پژوهش، عوامل اتیولوژیک عفونت های ناشی از اکتینومایستال ها در بیماران مراجعه کننده به دانشکده بهداشت دانشگاه علوم پزشکی تهران تعیین شده است.مواد و روش کاراین پژوهش که از سال1390 تا 1394 به منظور تعیین وضعیت بیماری های ناشی از اکتینو مایستال ها، تعداد 465 نمونه از بیماران گوناگون، شامل ترشحات ریوی (BAL)، مایع مغزی- نخاعی، بیوپسی پوست، آبسه های مختلف و مواد کلینیکی دیگر ارسالی از بیمارستان های مختلف شهر تهران، آزمایش مستقیم میکروسکوپی و کشت شدند.
یافته ها و بحث: از تعداد 465 نمونه ارسالی، تعداد 20 ایزوله (4/3%) از بیماران، شامل 11 مورد (55%) ایزوله اکتینومایسس و 9 مورد (45%) ایزوله نوکاردیا به دست آمد. بیشترین موارد مثبت مربوط به عفونت ریوی (بال) بوده است. در بین مبتلایان به این بیماری ها، 12 مورد (60%) بیماری در مردان و 8 مورد (40%) در زنان تشخیص داده شد.نتایج این مطالعه نشان داد که شیوع اکتینومایسس و نوکاردیا همانند است و در مردان بیشتر از زنان وجود دارد. برای به دست آوردن نتیجه مناسب بهتر است هنگام مراجعه به پزشک و پیش از آغاز درمان، نمونه گیری انجام شود. همچنین از روش های مولکولی مانند PCR و ریل تایم PCR برای شناسایی دقیق تر و مقایسه با روش های فنوتیپی توصیه می شود.کلیدواژگان: اکتینو مایکوزیس، نوکاردیوزیس، اکتینومایستال، BAL -
صفحات 210-216زمینه و هدفسودوموناس آئروژینوزا از مهمترین عوامل عفونت های بیمارستانی محسوب می شود که سویه های استاندارد و مقاوم آنتی بیوتیکی آن می توانند اندام های حیاتی بدن از قبیل ریه ها و سیستم ادراری را آلوده سازند. در این تحقیق، اثرات مهارکنندگی نایسین، گلایسین، پلی ال لیزین، نانوذرات منیزیم اکسید و عصاره هیدروالکلی اسپند بر سویه های استاندارد و مقاوم چند دارویی سودوموناس آئروژینوزا بررسی شده است.مواد و روش کارنانوذرات منیزیم اکسید به روش شیمیایی مرطوب در اندازه nm 50-30 تهیه گردیدند. ساختار نانوذرات توسط آنالیز پراش اشعه X (XRD) و میکروسکوپ الکترونی روبشی (SEM) تائید شدند. عصاره گیری از دانه های اسپند به روش خیساندن در محلول آب-اتانول 1:1 صورت پذیرفت. محلول هایی از گلایسین، پلی ال لیزین و نایسین تهیه شدند و توسط فیلتر μm 0/22 استریل گردیدند. اثرات بازدارندگی کلیه ترکیبات در قالب مقادیر قطر هاله مهاری رشد (IZD)، حداقل غلظت بازدارندگی رشد (MIC) و حداقل غلظت کشندگی (MBC) با استفاده از روش های انتشار در دیسک و میکرو رقیق سازی براث طبق استانداردهای CLSI تعیین گردیدند.
یافته ها و بحث: اثرات مهاری شاخصی از گلایسین، پلی ال لیزین، نانوذرات منیزیم اکسید و عصاره گیاهی در بالاترین غلظت به کاررفته بر سودوموناس آئروژینوزا مشاهده نشد. اثرات بازدارندگی فقط در مورد نایسین با قطر هاله عدم رشد mm 08/13 و 36/10 و MIC معادل μg/mL 128 و 64، و MBC معادل μg/mL 512 و 256 علیه دو سویه استاندارد و مقاوم ثبت گردید. نایسین به عنوان یک نگهدارنده طبیعی مواد غذایی می تواند به تنهایی یا در ترکیب با آنتی بیوتیک های رایج، در درمان بیماری های ناشی از سودوموناس آئروژینوزا بکار برده شود.کلیدواژگان: سودوموناس آئروژینوزا، اثرات ضدمیکروبی، نایسین، پلی ال لیزین، گلایسین، نانوذرات منیزیم اکسید، عصاره اسپند

Pages 149-157Background And AimsStaphylococcus aureus is a gram-positive, non-motile, non-spore-forming, aerobic, and facultative anaerobic bacterium, which is colonized in anterior part of the human nasal cavity. The carrier's individuals working in the food industry are the main source of spreading food-borne diseases. Staphylococcal enterotoxins (SEs) are the key virulence factors in the staphylococcal food poisoning (SFP). The aim of this study was to identify the S. aureus enterotoxin genes of sea, seb and sec among healthy carriers.Materials And MethodsIn the current descriptive cross-sectional study, 136 nasal swab samples were collected from the personnel working at butchers, dairy stores, and fast food restaurants through Ardabil city. The samples were cultured and then confirmed using the biochemical tests. The DNA of S. aureus isolates was extracted to detect sea, seb and sec genes as markers for SEA, SEB and SEC enterotoxins using the PCR method.ResultsAmong the 136 nasal swab samples, 46 (33.8%) were positive for S. aureus, that were confirmed by the presence of femA gene. Out of 46 isolates, the sea, seb and sec genes were found in 11 (23.9%), 6 (13%) and 5 (10.8%) isolates, respectively.ConclusionsAccording to the findings, a significant percentage of the food-chain personnel were nasal carriers of enterotoxigenic S. aureus. Therefore, for prevention and distribution of Staphylococcal infections, screening program and control of such carriers are recommended.Keywords: Staphylococcus aureus, Enterotoxin, PCR

Pages 158-166Background And AimsCryopreservation is one of the best long time preservation methods for fungi and using suitable cryoprotectants can increase the success of cryopreservation process. The aim of this study was to examine the effect of various cryoprotectants to protect of Candida glabrata in freezing conditions.Materials And MethodsIn this study, 50 clinical isolates of C. glabrata were stored at low temperature within three common cryoprotectants including; glycerol 10% and 40%, glucose 4% and DMSO 10%. After completing the preservation period, samples recovery rate was evaluated after two years using descriptive statistics and chi-square test.ResultsIn this study, 4% glucose with the ability to keep 100% of the samples, was determined as the best cryoprotectant for C. glabrata. Also, 40% glycerol and 10% DMSO with 94% success in retrieving samples, were comparable to each other and had a fairly good protective characteristic. While the 10% glycerol showed the lowest protective effect on C. glabrata. Recovery of samples cultured in SDA slant at 4ºC with a total of 32 (64%) positive cultures, showed a significant difference compared to preservation in freezing conditions. Also, the sample recovery in SDA had priority over SDB and yielded better results.ConclusionsThere was a significant difference between the cryoprotectants and by selecting a suitable cryoprotectant and appropriate concentration, survival of microorganisms can be increased in freezing conditions. Generally, assessment of a valid and reliable method for long-term preservation of a fungal strain should be associated with further studies on assessing changes in infectivity and pathogenicity, sporulation power and other features of microorganisms.Keywords: Cryopreservation, Cryoprotectant, Candida Glabrata

Pages 167-177Background And AimsMentha pulegium L. is one of the medicinal plants used in the food production as a flavoring substance and its antibacterial effect has also been considered. The present study was designed to evaluate the phytochemical composition of Mentha pulegium L. originated from Iran and its antimicrobial activity against several pathogenic bacteria.Materials And MethodsThe chemical compounds of this medicinal plant were analyzed by GC-MS and 14 components were identified, representing 86.98% of its total essential mass. The antibacterial activity of the essential oil (EO) of Mentha pulegium L., has been evaluated against Staphylococcus aureus, Escherichia coli, Pseudomonas aeroginosa, and Pseudomonas syringae pv. tomato using disc diffusion method compared to synthetic antibiotic ciprofloxacin. The minimum inhibitory concentration (MIC) was evaluated against the above tested bacteria using macrobroth dilution method.ResultsResults showed that the EO of Mentha pulegium L., was formed by the main components of piperitone (32.1%), piperitenone (21.71%) and pulegone (15.85%). In addition, the results obtained using MIC and MBC showed that the minimum inhibitory concentration of the EO of M. pulegium was in the range of 10-40 mg/mL for S. aureus, E. coli, P. aeroginosa and 20 mg/mL for Ps. syringae pv. tomato, respectively. According to the results, the maximum zones of inhibition (29.15 mm) was observed on P. syringae P. tomato in 40 mg/mL of essential oil, and the minimum zone of inhibition (17.08 mm) was related to P. aeroginosa in the same concentration.ConclusionsThe results revealed that EO of Mentha pulegium L. had an appropriate inhibitory effect on Staphylococcus aureus and Escherichia coli.Keywords: Antibacterial activity, Mentha pulegium, Essential oil, MIC, MBC

Pages 178-183Background And AimsKlebsiella pneumoniae that causes bloodstream and urinary tract infections has several virulence factors encoding genes. Factors like the ability of adherence to the host cell surface, presence of sidrophore receptors and regulation of mucoid phenotype are the common virulence ones involved in the pathogenesis of K. pneumoniae. The aim of this study was to determine the frequency of virulence factor encoding genes in K. pneumoniae isolates, collected from hospitalized patients by Multiplex PCR.Materials And MethodsIn this descriptive study, the frequency of genes encoding virulence factor irp2, wcaG, rmpA, rmpA¡ allS and fimH were evaluated in 100 K. pneumoniae isolates, collected from patients referred to teaching hospitals of Zahedan city by Multiplex PCR.ResultsAmong 100 clinical isolates of K. pneumoniae, allS gene had the highest frequency, 90%, and rmpA had the lowest frequency, 6%. The frequency of genes fimH, irp2, wcaG and rmpA were 88%, 87%, 33% and 6% respectively.ConclusionsThe results of this study showed that the allS, irp2 and fimH genes were more frequent among K. pneumoniae isolated from studied patients in Zahedan city. These findings provide important information on the importance of these virulence factors on pathology of urogenital tract infection, management of Klebsiella infection and success of treatment strategies.Keywords: Klebsiella pneumoniae, Virulence Factors, PCR

Pages 184-191Background And AimsDrug resistance, particularly methicillin resistant in Staphylococcus aureus strain is a major worldwide public health concern. The present study aimed to synthetize selenium nanoparticles, investigate its antibacterial effect and its ability to be used as ampicillin nanocarrier.Materials And MethodsSelenium nanoparticles were synthetized via chemical regeneration of sodium selenite by L-systein amino acid. Loading of ampicillin on the surface of selenium nanoparticles was done by gradual addition of antibiotic to nanoparticle solution and continuous shaking. Then this attachment was investigated by using UV-Vis spectroscopy, XRD and scanning electron microscope. Antibacterial properties of produced selenium nanoparticles and antibiotic loaded selenium nanoparticles against standard strain of S. aureus and 10 methicillin resistant S. aureus strains were tested by disc diffusion method. Minimum inhibitory concentration (MIC) of bacterial growth was determined by broth macrodilution method.ResultsThe produced selenium nanoparticles showed antibacterial effect against all strains of S. aureus. Loading of ampicillin on the surface of selenium nanoparticles enhanced the antimicrobial activity of this drug. The mean MIC of selenium nanoparticles against S. aureus strains ranged between 7.8-62.5 µg/mL free form of ampicillin between 62.5- 250 and ampicillin bound to nanoparticles was 7.8-7.8 μg / mL. These values for standard strain of Staphylococcus aureus strains were 7.8, 31.2 and 3.9 μg / mL, respectively. Loading of ampicillin on the surface of selenium nanoparticles enhanced the MIC against methicillin resistant S. aureus.ConclusionsThe obtained results confirmed antibacterial activity of selenium nanoparticles. Pages 192-202Background And AimsMethicillin-resistant Staphylococcus aureus is the most common bacterial agent of diabetic foot Infection (DFI).The Virulence factors of this bacterium increases the severity of the degree of wound infection, and if not treated promptly, results in lower limb amputation and death of the affected patients. The aim of this study was to isolate methicillin resistant Staphylococcus aureus carries the mecA gene and to detect 4 Virulence genes hla, lukED, sei and hlg from patients with diabetic foot infection in order to evaluate their role in severity of infection.Materials And Methods30 cases of diabetic foot infections in Mazandaran province were collected from pus drainage and Staphylococcus aureus was purified according to culture characteristics and biochemical tests. The molecular evaluation of the isolates after DNA extraction was performed with five pairs of specific primers of intended genes, by PCR method.ResultsOf the 30 patients, 14 isolates (46.6%) of S. aureus were isolated and identified. Each 14 samples contained mecA gene and the frequency of hla, lukED, sei and hlg genes in isolates was 100%, 100%, 71.4% and 64.2%, respectively.ConclusionsIdentification of these 4 genes with the mecA gene in S. aureus isolated from diabetic foot Infection can be an effective and reliable genetic marker for diagnosis of foot infection in diabetic patients. Pages 203-209Background And AimsActinomycetes are a group of Gram-positive bacteria that have filamentous morphology with about one micron diameter. Actinomyces, Nocardia and Streptomyces are classified in this group. In this study, the etiological agents of actinomycetal infections were determined among patients referred to the Health faculty of Tehran University of Medical Sciences (TUMS).Materials And MethodsTo determine the status of actinomycetes diseases, 465 specimens including the broncho-alveolar lavage (BAL), cerebrospinal fluid (CSF), tissue biopsy, abscess discharge and other clinical materiasl were examined via microscopic observation and culture methods. (2011-2015)
Results andConclusionsOut of the 465 specimens, 20 (4.3%) were diagnosed as infected which included 11(55% ) Actinomyces and 9 (45% ) Nocardia. The most positive results were associated with pulmonary infections. Among the infected patients, 60 % (12) were male and 40% (8) were female. The results of this study showed that the prevalence of Actinomyces and Nocardia was similar, being more predominant among men compared to women. In order to obtain the appropriate results, it is recommended that samples are taken during doctor's visit and before starting a treatment. It is also recommended to use molecular methods such as PCR and RT-PCR for better identification and comparison with phenotypic methods.Keywords: Actinomycosis, Nocardiosis, Actinomycetal, BAL -
Pages 210-216Background And AimsPseudomonas aeruginosa is one of the most important common causes of nosocomial infections that its standard and antibiotic-resistant strains can infect critical body organs such as the lungs and the urinary tract. In this study, inhibitory effects of nisin, glycine, poly-L-lysine, magnesium oxide nanoparticles and hydroalcoholic extract of Peganum harmala were assessed against standard and multidrug-resistant strains of P. aeruginosa.Materials And MethodsMgO nanoparticles with sizes of around 30-50 nm were synthesized via wet chemical method. Their structure were characterized using X-ray diffraction (XRD) analysis and scanning electron microscope (SEM). P. harmala seed extract was prepared by soaking in water-ethanol 1:1. Solutions of glycine, poly-L-lysine and nisin were prepared, and sterilized using 0.22 μm filter. Inhibitory effects of all compounds as inhibition zone diameter (IZD), the minimum inhibitory concentration (MIC) and the minimum bactericidal concentration (MBC) values were determined according to CLSI guidelines using disk diffusion and broth microdilution methods.
Results andConclusionsNo significant inhibitory effects against P. aeruginosa were observed at the highest used concentrations of glycine, poly-L-lysine, MgO nanoparticles and plant extract. Inhibitory effects against both standard and resistant strains were only recorded for nisin with inhibition zone diameter of 10.36, 13.08 mm, MIC of 64, 128 μg/mL and MBC of 256, 512 μg/mL.
