    ویروس کوتولگی سبزرد هندوانه (Watermelon chlorotic stunt virus، WmCSV) یکی از مخرب ترین ویروس های هندوانه در جنوب و جنوب شرقی ایران است. به منظور شناسائی علف هرزهای میزبان این ویروس، از مزارع بشدت آلوده هندوانه واقع در میناب (استان هرمزگان)، رودبار جنوب (جیرفت) و ارزوئیه (استان کرمان) بازدید گردید و نمونه برداری از داخل و حاشیه مزارع انجام شد. آلودگی و عدم آلودگی نمونه ها با آزمون PCR و با استفاده از آغازگرهای اختصاصی تعیین گردید. نتایج بدست آمده آلودگی تعداد زیادی از علف های هرز شامل چهارده گونه از دوازده جنس و نه تیره گیاهی را به WmCSV به اثبات رساند، بدون آنکه علائم خاصی روی بیشتر آنها دیده شود. تمامی این علف های هرز بعنوان میزبان WmCSV برای اولین بار معرفی می گردند. به منظور مقایسه جدایه های WmCSV در علف های هرز آلوده، قطعه 655 جفت بازی مربوط به ژن پروتئین پوششی 11 جدایه مختلف ویروس، همسانه سازی و تعیین ترادف گردید. مقایسه ترادف های نوکلئوتیدی و آمینواسیدی این جدایه ها با یکدیگر و با جدایه های موجود در بانک جهانی ژن نشان داد که جدایه های مورد مطالعه شباهت زیادی با یکدیگر و با سایر جدایه ها دارند. نزدیکترین جدایه WmCSV به جدایه های علف های هرز مورد مطالعه، جدایه بندرعباس (استان هرمزگان) بود که قبلا از این منطقه گزارش شده بود و از نظر ترادف های نوکلئوتیدی و آمینواسیدی بترتیب 2/ 99٪-9/ 98 و 100٪-8/ 96 با جدایه های فوق شباهت داشت. این نتایج نشان می دهد که این گیاهان بعنوان منابع نگاهدارنده WmCSV عمل کرده و بطور بالقوه می توانند در اپیدمیولوژی ویروس نقش اساسی داشته باشند.

    وجود تنوع ژنتیکی برای بدست آوردن ارقام با عملکرد بالا، کیفیت بهتر، تحمل بیشتر به تنش های زیستی و غیرزیستی و مقاومت بیشتر به آفات و بیماری ها، برای به نژادگران ضروری است. رتروترنسپوزون ها فراوان ترین و رایج ترین عناصر جابه جا شونده در ژنوم یوکاریوت ها به ویژه ژنوم گیاهان می باشند. توزیع گسترده رتروترنسپوزون ها در ژنوم گیاهان استفاده از آن ها را به عنوان نشانگر های مولکولی برای بررسی تنوع ژنتیکی ایده آل ساخته است. در این مطالعه فعالیت رتروترنسپوزون ها و همچنین تنوع ژنتیکی در 10 توده بومی آفتابگردان آجیلی با استفاده از نشانگرهای IRAP (Inter-retrotransposon amplified polymorphism) یررسی شد. از 25 آغازگر و ترکیب آغازگری IRAP، 11 آغازگر قادر به تولید الگوی باندی چند شکل با وضوح بالا بودند. 11 آغازگر IRAP، 116 مکان را تکثیر کردند که از این تعداد 110 مکان چند شکل بود. حداقل تشابه ژنتیکی نی (74/ 0) بین توده های همدان و مشهد، و حداکثر تشابه (88/ 0) بین توده های مرند و اصفهان مشاهده شد. تجزیه کلاستر با استفاده از آلگوریتم UPGMA، 10 توده بومی آفتابگردان آجیلی را در 3 گروه اصلی قرار داد. تجزیه واریانس مولکولی نشان داد تنوع درون توده ها بیشتر از تنوع بین تودها می باشد. بنابراین در برنامه های به نژادی آفتابگردان آجیلی، می توان انتخاب را درون توده ها انجام داد. نتایج حاصل از واکنش زنجیره ای پلی مراز با نشانگرهای IRAP نشان داد که رتروترنسپوزون های مطالعه شده در ژنوم آفتابگردان فعال هستند و به صورت سر به سر، سر به دم و دم به دم در ژنوم ادغام شده اند. نتایج حاصل از مطالعه حاضر نشان می دهد که نشانگر های مبتنی بر رتروترنسپوزون ها ابزاری مفید برای مطالعات ژنومیکس در آفتابگردان می باشند.

    یکی از مهمترین اهداف اصلاحی انگورهای رومیزی تولید دورگ های بیدانه جدید با حبه های درشت، سفت، معطر و مناسب با سلیقه مصرف کنندگان می باشد. سقط جنین در ارقام بیدانه به طور عمده کارآیی اصلاح ارقام بیدانه را محدود می کند. بر اساس اینکه سیتوکینین ها موجب افزایش قدرت مقصد فیزیولوژیکی دانه ها برای اسیمیلات ها می شوند، تحقیق حاضر به منظور مطالعه اثر محلول پاشی بنزیل آدنین قبل از شکوفایی گل در توسعه تخمک و نجات جنین دو رقم انگور بکربار کاذب(عسکری و بیدانه سفید) انجام گرفت. غلظت های بنزیل آدنین صفر،60 و100 میلی گرم در لیتر بود. تخمک های رقم بیدانه سفید،20 روز و رقم عسکری 40 روز بعد از باز شدن گل از درون حبه ها بیرون آورده شد و سپس بر روی محیط کشت نیچ و نیچ حاوی 1 میکرومول جیبرلین،1 میکرومول نفتالین استیک اسید، 2 گرم در لیتر زغال فعال، 20 گرم در لیتر ساکارز و 8 گرم در لیترآگار کشت شدند. صفات مورد بررسی شامل تخمک های قهوه ای شده، کالوس داده و جوانه زده بود. محلول پاشی بنزیل آدنین اثر معنی دار بر قهوه ای شدن تخمک ها نداشت اما درصد تخمک های قهوه ای شده در بین ارقام مورد آزمایش معنی دار بود و بیشترین تخمک قهوه ای شده در رقم بیدانه سفید مشاهده گردید. اثر محلول پاشی بنزیل آدنین با غلظت-های 60 و100 میلی گرم در لیتر بر درصد کالوس دهی و جوانه زنی تخمک ها معنی دار بود. بر اساس نتایج بدست آمده درصد تخمک های جوانه زده، بین دو رقم مورد آزمایش معنی دار بود. تخمک های جوانه زده در رقم عسکری71/ 23 درصد و در رقم بیدانه سفید44/ 14 درصد بود. بطور کلی محلول پاشی با بنزیل آدنین قبل از گل دهی باعث افزایش موفقیت تکنیک نجات جنین در ارقام عسکری و بیدانه سفید می شود.

    انتخاب برای بهبود صفات تولید و تولید مثل در دام ها، امری زمان بر بوده و فقط در سنین بلوغ دام-ها امکان پذیر است. گاو سرابی یکی از نژادهای بومی شیری در ایران است که به طور عمده در شمالغرب کشور پرورش می یابد. شناسایی ژن های کاندیدای موثر بر صفات تولید شیر و عملکرد تولید مثلی در این نژاد دارای اهمیت بوده و می تواند بازده برنامه های اصلاح نژادی را بهبود دهد. هدف از مطالعه حاضر بررسی ارتباط بین چندشکلی موجود در چهار ژن کاندیدا (لپتین، هورمون رشد ((GH)، گیرنده ی هورمون رشد (GHR) و ژن Pit-1 با صفات تولید شیر و برخی صفات تولید مثلی گاو سرابی با استفاده از نشانگر PCR-RFLP بود. نتایج نشان داد که بیشترین فراوانی های آللی برای هر یک از ژن های لپتین، GH، GHR و Pit-1 به ترتیب برای آلل های(A (0/72)، V(0/51)، + (0/65 0) و(B (0/55 بود. ژنوتیپ-هایLL، -/-، AB-BB مربوط به ژن های GH، GHR و Pit-1 نسبت به سایر ژنوتیپ ها به طور معنی داری تولید شیر بیشتری داشتند (p< 0.01). همچنین ژنوتیپ های LL و AB شناسایی شده در ژن های GH و Pit-1 نسبت به سایر ژنوتیپ ها روزهای باز کمتری را نشان دادند (p< 0.01). در مطالعه حاضر هیچ رابطه معنی داری بین ژنوتیپ این ژن ها و سایر صفات بررسی شده مشاهده نشد. در ژنهای کاندید مذکور مقادیر هتروزیگوسیتی مشاهده شده نسبت به مقادیر مورد انتظار کمتر بودند و نتایج تجزیه و تحلیل کای مربع نشان دهنده ی عدم وجود تعادل هاردی- واینبرگ در این جایگاه ها بود. به طور کلی نتایج این تحقیق نشان دهنده وجود ارتباط بین جهش های موجود شناسائی شده در برخی از این ژن ها با صفات تولید شیر و روزهای باز در گاوهای سرابی است و امکان استفاده از این اطلاعات در برنامه ی انتخاب به کمک نشانگرها باید مورد بررسی بیشتر قرار گیرد.

    ویروس پیچیدگی زرد برگ گوجه فرنگی، TYLCV) Tomato yellow leaf curl virus) به همراه ده گونه ویروسی و نژادهایشان که به ویروس های شبه TYLCV معروفند عامل خسارت به گوجه فرنگی (Solanum lycopersicum L.) در مناطق گرمسیر، نیمه گرمسیر و معتدل دنیا می باشند. در این بررسی تعداد سه جدایه TYLCV-IL از گیاهان تاتوره در منطقه بجنورد همسانه سازی و ژنوم کامل آنها تعیین توالی گردید. توالی اسید نوکلئیک ژنوم کامل آنها به همراه تعداد نه جدایه TYLCV-IL گزارش شده ازایران و هشت توالی سایر ویروس های Begomovirus گوجه فرنگی از ایران و سایر مناطق دنیا مورد تجزیه و تحلیل قرار گرفتند. نتایج فیلوژنی جدایه ها نشان داد که تکامل جمعیت نژاد TYLCV-IL در ایران وابسته به جغرافیا بوده و صرفنظر از گونه میزبان واریانت های شمال شرق و جنوب ایران به طور مجزا در دو زیرگروه جداگانه قرار گرفتند. جدایه TYLCV-IL[IR:Sh40:07]، GUO76444 از منطقه شیراز با هیچکدام از جمعیت های شمال شرق و جنوب ایران گروه بندی نشد. این جدایه با نژاد تیپ TYLCV-IL گزارش شده از فلسطین اشغالی قرابت نزدیکی داشته و با یکدیگر در یک زیر گروه مجزا قرار گرفتند. نتایج تجزیه فیلوژنی نشان داد که این جدایه از نظر تکاملی به عنوان پل ژنتیکی بین جدایه های TYLCV-IL حوزه مدیترانه ای و ایران محسوب می شود. این اولین گزارش از گیاه تاتوره به عنوان میزبان نژاد TYLCV-IL در ایران و همچنین حضور این نژاد ویروسی در منطقه بجنورد و استان خراسان شمالی می باشد.

    فسفر یکی از عناصر غذایی مهم برای گیاهان می باشد که در خاک فراهمی کمی دارد. فسفر در خاک به دو شکل آلی و معدنی یافت می شود. باکتری های تحریک کننده رشد گیاه یا PGPR، باکتری های موجود در خاک و ریزوسفر گیاهان هستند که با سازوکار های مختلف به رشد گیاه کمک می کنند. توانایی برخی ریزسازواره ها به منظور تبدیل فسفر نامحلول به شکل قابل استفاده مانند ارتوفسفات، ویژگی مهمی در PGPR می باشد که باعث افزایش عملکرد گیاهان می شود. گونه هایی از جنس Pseudomonas، Bacillus، Pantoea وRhizobium از قوی ترین حل کنندگان فسفات به شمار می آیند. سازوکار اصلی برای انحلال فسفات معدنی تولید اسیدهای آلی است و در انحلال اشکال فسفر آلی اسید فسفاتازها نقش اصلی را در خاک بازی می کنند. روش مرسوم در جداسازی این دسته از باکتری ها استفاده از منابع فسفاته معدنی و آلی کم محلول یا نامحلول در محیط کشت های مایع یا جامد می باشد، که از طریق پایش تولید فسفات آزاد شده و کاهش pH در محیط مایع یا مشاهده هاله شفاف در اطراف کلنی ها و تولید کلنی های رنگی (سبز، آبی و زرد) در صورت استفاده از سوبستراهای رنگزا در محیط کشت جامد دنبال می شود. گر چه دانش ژنتیک انحلال فسفات هنوز اندک می باشد، چندین ژن رمزکننده فسفاتاز مشخص گردیده و همسانه سازی شده اند و تعدادی ژن درگیر در انحلال فسفات معدنی جداسازی شده است. روش های زیست شناسی مولکولی رویکردی مفید برای به دست آوردن و تشخیص سویه های PGPR کاآمد می باشد. انتقال و بیان ژن های درگیر در انحلال فسفات (فسفات آلی یا معدنی) در باکتری ها یا گیاهان یک راهکار جدید برای بهبود ظرفیت ریزسازواره ها به عنوان مایه تلقیح میکروبی است.

    به منظور شناسایی QTL موثر بر صفات رشد و برآورد میزان اثر آنها، از یک طرح تلاقی F2 حاصل از دو سویه متفاوت بلدرچین ژاپنی (تخمگذار یا سفید و گوشتی یا وحشی) استفاده گردید. با تلاقی 34 پرنده حاصل از تلاقی فوق و به منظور ایجاد جمعیت لازم برای رکورد برداری فنوتیپی، 422 پرنده نسل سوم تولید شد. مشاهدات شامل وزن های هفتگی ثبت شده و نسبت کلیبر مربوط به فاصله زمانی هچ تا 1، 1 تا 2، 2 تا 3، 3 تا 4، 4 تا 5 و هچ تا 5 هفتگی می باشد. تمامی پرندگان هر سه نسل (472 پرنده) برای 8 نشانگر ریزماهواره موجود بر روی کروموزوم شماره 1 و با میانگین فاصله 29 سانتی مورگان از یکدیگر تعیین ژنوتیپ شدند. آنالیز داده های حاصل با استفاده از روش نقشه یابی درون فاصله ای مبتنی بر رگرسیون انجام گرفته و پنج QTL مربوط به نسبت کلیبر محاسبه شده برای 1 تا 2، 3 تا 4، 4 تا 5 و هچ تا 5 هفتگی شناسایی شد. واریانس QTLهای شناسایی شده برای صفات مختلف در محدوده 8/ 0 تا7/ 3 بود. با برازش مدل دارای اثرات افزایشی و غلبه، اثر غلبه برای هیچکدام از صفات معنی دار نشد. همچنین، جایگاه های شناسایی شده، بر هیچکدام از صفات مورد بررسی اثر ایمپرینتینگ نداشتند.

    در دهه های اخیر، پیشرفت در بیوتکنولوژی گیاه، بویژه زراعت مولکولی امکان استفاده از گیاهان را به عنوان یک سیستم تولید جدید و راکتور زیستی برای محدوده ی وسیعی از پروتئین های نوترکیب فراهم کرده است. اینترفرون گامای انسانی یکی از پروتئین های ارزشمند دارویی می باشد که کاربردهای پزشکی وسیعی در زمینه های تشخیصی و درمانی پیدا کرده است. کلونینگ ژن اینترفرون گامای انسانی به همراه ژن مقاومت به آنتی بیوتیک اسپکتینومایسین - استرپتومایسین با استفاده از ناقل کلروپلاستی pKCZ در آزمایشگاه بیوتکنولوژی دانشکده کشاورزی دانشگاه تربیت مدرس صورت گرفته و سپس به روش بیولستیک به کلروپلاست گیاه توتون انتقال یافته است. هدف از انجام این تحقیق بررسی پایداری بیان ژن اینترفرون گامای انسانی در گیاهان ترانسپلاستومیک نسل اول توتون بود. گیاهان رشد کرده در سطح محیط انتخابگر، در سطوح مختلف (DNA، RNA و پروتئین) به منظور تایید درج و بیان ژن مورد بررسی قرار گرفتند. بررسی این گیاهان در سطح DNA با استفاده از آغازگرهای اختصاصی ژن اینترفرون گاما و تکنیک PCR صورت پذیرفت، نتایج حاصل درج این ژن را در ژنوم کلروپلاستی گیاهان تایید کرد، به علاوه با آزمون RT-PCR رونویسی ژن هدف تایید شد. همچنین به منظور بررسی بیان ژن هدف در سطح پروتئین، آزمون های SDS-PAGE، Dot-blot و Western-blot انجام شد و نتایج بدست آمده، بیان این ژن را در سطح پروتئین تایید کرد. در نهایت به منظور تعیین میزان بیان ژن اینترفرون گامای انسانی آزمون ELIZA انجام شد. نتایج نشان داد که بالاترین سطح تجمع اینترفرون گاما در گیاهان ترانسپلاستومیک بالای 2/ 0% پروتئین محلول کل می باشد.

    در برنامه تولید بذر هیبرید در سیستم سه لاینی، استفاده از لاین با قابلیت ترکیب پذیری خصوصی مطلوب حامل ژن های بازگرداننده باروری (Rf) ضرورت دارد. در این تحقیق، جایگاه ژنی بازگرداننده باروری Rf3 واقع بر روی کروموزوم شماره یک برنج با روش تلاقی برگشتی به کمک نشانگر، به پس زمینه ژنتیکی لاین نرعقیم ندا-A انتقال داده شد و همزمان اثر آن در هر نسل بر باروری دانه گرده و خوشه لاین گیرنده، مورد بررسی قرار گرفت. جهت انتقال، یک گیاه از جمعیت نسل F2 (حاصل از تلاقی میان ندا-A و IR36) بر اساس نشانگرهای پیوسته به جایگاه ژنی Rf3 (RM1، RM3873 و RM3233) انتخاب و با لاین ندا-A (به عنوان والد تکراری) تلاقی برگشتی داده شد. نتاج نسل اول تلاقی برگشتی (BC1) از نظر وجود نشانگرهای پیوسته به ژن و همچنین از نظر فنوتیپی و به دنبال آن برای 15 نشانگر ریز ماهواره پس زمینه، مورد بررسی قرار گرفتند. تنها دو بوته نسل BC1 که در هر سه جایگاه نشانگری دارای آلل غالب از والد بخشنده بودند باروری بالای خوشه (55% و 65%) را نشان دادند. نتاج نسل دوم تلاقی برگشتی (BC2) از تلاقی دو بوته فوق با والد تکراری حاصل شد. در بین نتاج BC2 یک گیاه با باروری بالاتر خودگشن شده و 170 بوته از نسل BC2F2 از نظر 3 نشانگر RM1، RM3873 و RM3233 و همچنین دانه بندی خوشه مورد بررسی قرار گرفتند. هفت بوته با میزان بالاتری از ژنوم والد تکراری (از 1/91 % تا 5/98%) و در وضعیت هموزیگوتی کامل از نظر سه نشانگر پیوسته به (Rf3 (RM1، RM3873 و RM3233 شناسایی شدند که دارای باروری بالای دانه گرده و همچنین خوشه (75%تا 97%) بودند که نشان می دهد هر چه هموزیگوسیتی جایگاه ژنی Rf3 بیشتر شد، بازگرداندن باروری به نرعقیمی نوع وحشی (WA) در پس زمینه ژنتیکی لاین گیرنده نرعقیم، بیشتر تقویت گردید. بنابراین، می توان نتیجه گیری نمود که جایگاه ژنی Rf3 تاثیر متقابلی با نرعقیمی سیتوپلاسمی نوع WA در جهت افزایش باروری دانه گرده و خوشه برنج دارد و از اینرو، می تواند همراه با سایر ژن های بازگرداننده باروری در برنامه تولید بذر هیبرید برنج به کار گرفته شود.

    پروتئین گیرنده هورمون رشد یک گیرنده سطح سلولی برای هورمون رشد است که جهت انجام نقش هورمون رشد در بافت های هدف ضروری است. هدف تحقیق حاضر، شناسایی و بررسی تنوع ژنتیکی ناحیه پروموتر ژن گیرنده هورمون رشد درگاوهای بومی استان کهکیلویه و بویراحمد می باشد. برای این منظور، ناحیه ای از پروموتر ژن گیرنده هورمون رشد توسط تکنیک تفاوت فرم فضایی رشته های منفرد (SSCP) و تعیین توالی در گاوهای نژاد بومی تعیین ژنوتیپ شد. در کل 50 نمونه بررسی شده 3 الگوی (SSCPs) مجزا مشاهده گردید که با تعیین توالی نوکلوتیدی آنها چند شکلی تک نوکلئوتیدی A/G در جایگاه 154- و دو طول متفاوت 17 و 11 تکراری برای ریزماهواره TG در جایگاه 86- قبل از شروع رونویسی مشاهده گردید. فراوانی ژنوتیپ های (AA (17/17 TG)، GG (11/11 TG و (GA (17/11 TG در جایگاه 154- به ترتیب 34/ 0، 24/ 0 و 52/ 0 بود. بررسی بیوانفورماتیکی چند شکلی های شناسایی شده بر روی جایگاه فاکتورهای رونویسی نشان داد که چند شکلی تک نوکلئوتیدی در جایگاه 154- بسیار نزدیک به جایگاه اتصال برای فاکتور رونویسی C/EBPb قرار دارد که ممکن است فرآیند رونویسی ژن را تحت تاثیر قرار دهد. نقش کاربردی ممکن چندشکلی های شناسایی شده باید توسط تجزیه و تحلیل بیان ژن تایید گردد.

    گیاهان به مکانیسم های سلولی مختلفی در مقابله با اثر سمی فلزات سنگین مجهز شده اند. یکی از مهم ترین این مکانیسم ها، تولید پروتئین ها و پپتیدهای متصل شونده به فلزات، مانند متالوتیونین ها (MT) می باشد. متالوتیونین ها، گروهی از پروتئین ها با وزن مولکولی کم و غنی از آمینواسیدهای سیستئین هستند که دارای ظرفیت بالای اتصال به فلزات می باشند. در این تحقیق توالی کد کننده ی ایزوفرمOsMTI-1b از گیاه برنج، به عنوان گیاه مدل در بین تک لپه ای ها، در ناقل بیانی pET41a همسانه سازی و به میزبان بیانی E. coli سویه ی Rosetta (DE3) منتقل شد. پس از القا محیط کشت توسط IPTG، میزان مناسبی از پروتئین های نوترکیب در فاز محلول تولید و با استفاده از روش کروماتوگرافی جذبی خالص سازی شد. سلول های باکتری بیان کننده ی پروتئین های نوترکیب در محیط LB حاوی نمک NiCl2 رشد داده شدند و منحنی رشد آن ها در مقایسه با شاهد ترسیم شد. مقدار کاهش فلز نیکل در محیط کشت و تجمع آن ها در رسوب باکتریایی توسط دستگاه طیف سنجی پلاسمای جفت شده ی القائی (ICP-AES) به دست آمد. بر اساس نتایج مشخص گردید بیان ایزوفرمOsMTI-1b از طریق افزایش تجمع درون سلولی فلز نیکل موجب افزایش تحمل باکتری E. coli به این فلز می گردد. همچنین، بررسی الگوی جذب نور و واکنش DTNB با پروتئین های انکوبه شده با فلز نیکل در شرایط این ویترو، تشکیل دستجات فلز/تیول در پروتئین نوترکیب GST-OsMTI-1b را اثبات نمود. یافته های پژوهش حاضر می تواند منعکس کننده نقش احتمالی ایزوفرمOsMTI-1b در تحمل گیاه برنج به تنش فلزات سنگین باشد.

    آنالیز های ترانسکریپتوم در ایجاد اطلاعات و درک کافی از سیستم های واکنشی گیاهان در برابر عوامل زیستی و غیر زیستی بسیار کارآمد هستند. cDNA-AFLP به عنوان روشی مناسب علاوه بر تکرار پذیری بالا، نیاز به اطلاعات اولیه در مورد توالی ژن ها ندارد. البته عوامل متعددی در حساسیت، دقت و تکرار پذیری آن موثر هستند که توجه به این عوامل می تواند باعث افزایش کیفیت ارزیابی ها و دستیابی به اطلاعات صحیح گردد. در تحقیق حاضر cDNA-AFLP برای آنالیز واکنش گیاه سیب زمینی در برابر استرس ناشی از باکتری بیماریزای Ralstonia solanacearum در شرایط in vitro بهینه سازی گردید. در این راستا پارامترهای مختلف و اثر تعامل آنها با یکدیگر در افزایش حساسیت و تکرار پذیری این روش بررسی شد. نتایج نشان داد روش استخراج RNA و استفاده از mRNA از فاکتور های تاثیر گذار در موفقیت آزمون cDNA-AFLP است به طوری که RNA های استخراج شده با استفاده از روش مبتنی بر ستون از یکنواختی و کیفیت بالاتری برخوردار هستند و استخراج mRNA علاوه بر حذف RNA های ریبوزومی، در کاهش ممانعت کننده ها و اجرای صحیح سایر مراحل آزمون تاثیر گذار است. همچنین بهینه سازی فاکتورهایی از قبیل رقت مناسب محصول پیش تکثیر که به عنوان الگو در مرحله تکثیر انتخابی استفاده می شود، تعداد سیکل PCR در مرحله پیش تکثیر، غلظت مناسب MgCl2 و Taq DNA Polymerase در مراحل پیش تکثیر و تکثیر انتخابی، نقش تعیین کننده ای بر روی تعداد و غلظت قطعات cDNA تکثیر شده و وضوح آن بر روی ژل دارد.

    تنوع ساختار کروموزومی در مکانیزم های بیولوژیکی از اهمیت خاصی برخوردار است. به منظور تعیین تعداد و توزیع این تنوع در ژنوم سه کروموزم 6، 14 و 20 که حضور جایگاه صفات کمی موثر بر تولید در آن تایید شده است٬ مورد بررسی قرار گرفت. به این منظور از 580 راس گاو هلشتاین نمونه خون گرفته شد و پس از استخراج DNA٬ تعیین ژنوتیپ با استفاده از بسته نشانگری 50K گاوی شرکت ایلومینا صورت گرفت. پس از تصحیح داده ها برای شدت سیگنال و نواحی غنی از GC، تعداد 383 نمونه برای آنالیز نهایی باقی ماند. داده ها بر اساس اسمبلی UMD3.1 ژنوم گاو برای کروموزم های 6، 14 و 20 آنالیز شد. پس از اعمال کلیه فیلترها تعداد 199 تنوع ساختار کروموزومی (132 حذف و 67 اضافه) با متوسط 5/ 0 برای هر فرد و میانگین طول kb 147/3 و میانه kb 139/4 شناسایی شد. نسبت حذف به اضافه برابر با 97/ 1 و کروموزم های 6 و 20 به ترتیب دارای بیشترین و کمترین تنوع ساختار کروموزومی بودند. آنالیز های بیوانفورماتیکی مشخص نمود که 77 ژن با این نواحی تنوع ساختار کروموزومی در ارتباط هستند. نتایج این پژوهش نشان داد تنوع ساختار کروموزومی بخش قابل توجهی از این سه کروموزم که حاوی تعداد قابل توجهی جایگاه صفات کمی هستند را پوشش می دهد. به دلیل اثر این تنوع بر ایجاد تغییر در ساختار و دز ژن ها، به نظر می رسد تنوع ساختار کروموزومی می تواند بر واریانس فنوتیپی صفات کمی موثر باشد لذا احتمالا این تنوع قابلیت استفاده در برنامه های اصلاح نژادی را دارد.

    وجود تنوع ژنتیکی شرط ضروری برای موفقیت در برنامه های به نژادی است. به منظور بررسی تنوع ژنتیکی از طریق برخی از صفات مورفوفیزیولوژیکی و نشانگرهای SSR، 40 رقم گندم نان در یک طرح بلوک کامل تصادفی با 3 تکرار در مزرعه پژوهشی دانشگاه شهید باهنر کرمان در سال 1390-1389 مطالعه شدند. ژنوتیپ های گندم نان برای تمامی صفات مورد بررسی تنوع ژنتیکی قابل ملاحظه ای نشان دادند. تعداد پنجه های بارور بیشترین همبستگی را با عملکرد دانه نشان داد. در آزمایش SSR از مجموع 10 آغازگر انتخابی، 9 آغازگر چند شکلی قابل توجهی نشان دادند. برای مجموع ژنوتیپ ها 31 نوار چند شکل با میانگین 4/ 3 نوار به ازای هر آغازگر مشاهده شد. هتروزیگوسیتی مورد انتظار کل جمعیت در تمام جایگاه های ژنی به طور متوسط 36/ 0 ارزیابی شد و جایگاه ژنی wmc 420 دارای بیشترین تنوع بر روی جمعیت مورد مطالعه بود. تجزیه کلاستر با استفاده از روش وارد انجام شد و ارقام بر اساس میزان شباهت ها گروه بندی شدند. اطلاعات این گروه بندی می تواند در پروژه های به نژادی برای افزایش عملکرد در شرایط تنش مورد استفاده قرارگیرد.

    Watermelon chlorotic stunt virus (WmCSV) is one of the destructive viruses of watermelon in south and southeastern Iran. In order to identify the wild hosts of the virus, watermelon growing areas of Minab (Hormozgan province), Roodbar-e-Jonob and Arzuiyeh (Kerman province) were surveyed and weed samples collected within and around the severely infected farms. The infection of the samples was tested by PCR using specific primer pair. Results showed that 14 weed species from 12 genera and nine plant families are infected with WmCSV. Most of the infected weeds were symptomless and did not show any specific symptoms. All the infected weed species in this study are reported for the first time as the WmCSV host in the world. In order to compare WmCSV isolates from weed species, the amplified 655 bp segment of coat protein (CP) gene from 11 infected weeds were cloned and sequenced. Comparison of nucleotide and amino acid sequences of 11 WmCSV isolates showed close similarities with each other and with the GenBank isolates. The closest GenBank isolate was the previously reported Iranian isolate from Bandar-Abbas (Hormozgan province) with 98/9-99/2 and 96/8-100% homologies for nucleotide and amino acid sequences, respectively. According to the results of this study, weed species are secondary hosts and serve as the reservoirs of the WmCSV. Thus, these weeds could be potentially important in epidemiology of the virus.

    Genetic diversity is necessary for plant breeders to obtain new cultivars either with high yield, better quality, more adapted to abiotic stress or more resistant to pest and pathogens. Retrotransposons are current component of plant genomes. Ubiquitous, activity and abundant of retrotransposons thorough the plant genome, make them useful molecular marker. We used inter-retrotransposon amplified polymorphism (IRAP) markers to assess genomic diversity levels and survey the activity of LTR retrotransposon elements in 10 confectionary sunflower populations. Out of 25 single and combined IRAP primers, 11 primers produced scorable and polymorphic banding patterns. A total of 116 loci were amplified by using 11 IRAP primers on confectionary populations. 110 loci out of 116 were polymorphic. The lowest (0.74) and highest (0.88) Nei genetic similarity was observed between Hamedan and Mashhad and Marand and Esfahan populations, respectively. Cluster analysis based on UPGMA algorithm grouped the studied populations in 3 main classes. Analysis of molecular variance revealed that the high part of total variation was due to within populations. So it will be better to do selection within populations in breeding programs. The results showed that retrotransposons are active in sunflower genome and they are inserted in sunflower genome as head-to-head, head-to-tail and tail-to-tail orientations. Our results showed that retrotransposon-based molecular markers can be used as a valuable tool for genomic assessment in sunflower.

    One of the main objectives of table grape breeding programs is producing new seedless hybrids with large, firm, high flavor berries and consistent with consumers taste. Abortion of zygotic embryo in seedless grapes largely limits the efficiency of seedless cultivars breeding. Since cytokinins are known to increase the sink strength of seeds for assimilates, the present investigation was conducted to study the influence of pre-bloom sprays of benzyladenine (BA) on ovule development and embryo rescue of two stenospermic grape cultivars (Askari and White Seedless).Concentrations of BA were 0, 60 and 100 ppm. Ovules of White Seedless, 20 days and Askari 40 days after flower opening were dissected out of berries and then cultured in Nitsch & Nitsch medium containing 1μM GA3, 1μM NAA, 2 g/l activated charcoal, 20 g/l sucrose and 8 g/l agar. Evaluated characteristics were ovule browning, callusing and germination. BA Spraying had no significant effect on ovules blacking but percentage of browned ovules were significant between examined cultivars and the highest ovule browning were observed in White Seedless cultivar. Effect of BA sprays whit 60 and 100 ppm concentrations were significant on callusing and ovules germination. Results showed the significant effect of cultivars on the percentage of germination ovules. Ovules germination was 23.71% in Askari cultivar and 14.44% in White Seedless. Finally pre-bloom sprays of benzyladenine (BA) increase successfully of embryo rescue technique in Askari and Bidaneh sefid Cultivars.

    Selection for improving of production and reproductive performance traits in livestock is a time-consuming process and can only be applied to adult animals. Sarabi cow is one of the local Iranian dairy breeds with dominate geographical distribution on NorthWest of Iran. Characterization of candidate genes associated with milk yield and reproductive performance traits is important and can help to improve the efficiency of breeding programs in thisbreed. Therefore, the objective of the present study was to evaluate the association among four candidate genes (Leptin, Growth Hormone (GH), Growth Hormone Receptor (GHR) and Pit- 1) with milk yield and some reproductive performance traits in Sarabi cattle breed using PCRRFLP marker. The obtained results revealed that the highest allele frequencies were observed for A (0.72), V (0.51), + (0.65) and B (0.55) for Leptin, GH, GHR and Pit-1 genes respectively. The respective LL, -/- and AB-BB genotypes of GH, GHR and Pit-1 genes showed higher milk yield production compare to other genotypes of these genes (P< 0.01). Also, LL and AB genotypes of GH and Pit-1 genes were related to shorter open days(P< 0.01). No evidence showing the association between other genotypes of these genes and the other investigated traits were observed in the present study. Overall, candidate genes showed deficiency of observed heterozygosity value compare to the expectedheterozygosity value. In addition, the results of chi-square analysis showed that all candidate genes were in Hardy-Weinberg disequilibrium. As a conclusion, the finding of the present study revealed the association of investigated candidate genes with milk yield and open days in Sarabi cattle breed and the application of these polymorphisms for marker assisted selection (MAS) should be further evaluated.

    Tomato yellow leaf curl virus (TYLCV) and a complex of ten virus species and their strains referred to as TYLCV-like viruses cause damage on tomato (Solanum lycopersicum L.) crops in tropical, subtropical and temperate regions of the world. In this study, full-length genome of three TYLCV-IL isolates originating from Datura stramonium L. in Bojnurd region, Iran were cloned, sequenced and subsequently entered the phylogenetic analysis with nine TYLCV-IL isolates from Iran and eight selected begomoviruses of tomato from Iran and other part of the world. The phylogenetic analysis suggested the geographical-dependent evolution of Iranian TYLCV-IL populations so that regardless of host species the monophyletic group of the northeastern and southern isolates was clustered separately into two subgroups. TYLCV-IL [IR: Sh40:07], GU076444 isolated in Shiraz region was not clustered with none of the Iranian TYLCV-IL isolates but rather was grouped with the type strain of TYLCV-IL from Israel in a separate group. Evolutionally, this isolate could be a “genetic bridge” between Mediterranean and Iranian TYLCV-IL isolates. This is the first report of Datura stramonium L. as the host species of TYLCV-IL in Iran and also the first-time occurrence of the viral stain in North Khorasan province, Bojnurd, Iran.

    Phosphorus is one of the major plant nutrients that is least available in the soil. There are two components of P in soil, organic and inorganic phosphate. Plant growth promoting bacteria (PGPR) are soil and rhizosphere bacteria that can benefit plant growth by different mechanisms. The ability of some microorganisms to convert insoluble P to an accessible form, like orthophosphate, is an important trait in a PGPR for increasing plant yields. The use of phosphate solubilizing bacteria as inoculants simultaneously increases P uptake by the plant and crop yield. Strains from the genera Pseudomonas, Bacillus, Pantoea and Rhizobium are among the most powerful phosphate solubilizers. The principal mechanism for mineral phosphate solubilization is the production of organic acids, and acid phosphatases play a major role in the mineralization of organic phosphorous in soil. The main method for isolation PSB is carrying out by using insoluble organic and inorganic phosphate source in solid or liquid media with monitoring of production of free orthophosphate and decreasing pH in liquid media or production halo zone around colonies or production green, blue and yellow colonies in presence of chromogenic substrates in solid media. Although knowledge of the genetics of phosphate solubilization is still scanty, several phosphatase-encoding genes have been cloned and characterized and a few genes involved in mineral phosphate solubilization have been isolated. Molecular biology methods are a benefit approach to access and characterization of improved PGPR. Transfer and expression of phosphate (organic and inorganic phosphate) solvent encoding genes in bacteria or plants, is a new way for improving of microorganism capacitance as an inoculant.

    A three-generation resource population was developed by using two distinct Japanese quail strains, wild and white to map quantitative trait loci underlying Kleiber ratio, an indirect criterion of feed efficiency. Eight pairs of white (S) and wild (W) birds were crossed reciprocally and 34 F1 birds were produced. The F1 birds were intercrossed to generate 422 F2 offspring. All of the birds from three generations (472 birds) were genotyped for eight microsatellite markers on chromosome 1. Liveweight data from hatch to 5 weeks of age were collected on the F2 birds. QTL analysis was conducted applying the line-cross model and the least-squares interval mapping approach. The results indicated QTL affecting Kleiber ratio for 1 to 2, 3 to 4, 4 to 5 and hatch to 5 weeks of age on chromosome 1. The F2 phenotypic variance explained by the detected QTL effects ranged from 0.8 to 3.7 for different traits. Modeling both additive and dominance QTL effects revealed QTL with significant additive mode of action. However, there was no evidence for imprinting (parent-of-origin) effects.

    In the recent decades the progress in plant biotechnology, especially in the field of molecular farming, has made it possible to produce a wide range of recombinant protein using plants as a novel production system and bioreactor. human gamma interferon is one of the valuable pharmaceutical proteins which has found wide medical applications in diagnosis and therapeutic. Cloning of human gamma interferon gene associated with spectinomycinstreptomycin antibiotic resistance gene using chloroplast pKCZ vector has been performed at agricultural biotechnology laboratory of Tarbiat Modares University and has been transferred into the tabacco chloroplast genome with biolistic method. The purpose of this study is to determine gamma interferon gene expression stability in T1 tobacco transplastomic plants. plants, which are grown on selective media, were analysed at different levels of (DNA, RNA and protein) to confirm integration and expression of the gene into the tobacco chloroplast. Analysis of plants was performed at DNA level by using of PCR and specific primers for human gamma interferon gene, results confirmed integration of this gene into the chloroplast genome. In addition, transcription of target gene was confirmed by RT-PCR. In order to study expression of target gene at protein level SDS-PAGE, Dot blotting and Western-blot were also done and the obtained results show target gene expression at protein level. Finally, ELISA test was done to determine quantity of gamma interferon gene expression. Results indicated that the highest gamma interferon level in transplastomic plant is up to 0.2% of total soluble protein.

    In hybrid seed production program based on three-line system, the use of fertility restorer line with desirable specific combining ability carrying fertility restoration (Rf) genes is indispensible. In this research, fertility restoration locus Rf3 was transferred into CMS line ‘Neda-A’ using marker-assisted backcrossing (MAB) method and simultaneously its effect on pollen and panicle fertility of recipient line was evaluated in each generation. For transferring the locus, a single F2 plant (derived from ‘Neda-A’ x ‘IR36’ cross) was selected based on Rf3- linked three markers (RM1, RM3233 and RM3873) and backcrossed to ‘Neda-A’ (as the recurrent parent). The BC1 progenies were screened for Rf3-linked markers and also phenotyped and subsequently screened for 15 background SSR markers. Only two BC1 plants with donor dominant allele at all three loci showed high panicle fertility (65% and 50%). BC2 progenies were developed after backcrossing these two plants to recurrent parent. Among BC2 progenies, a single plant having a higher fertility was self-pollinated and 170 resultant BC2 2 plants were screened with 3 foreground Rf3-linked markers (RM1, RM3233 and RM3873) and also evaluated in terms of seed setting. Seven plants were identified with a higher rate of recurrent parent genome (91.1% to 98.5%) and in complete homozygote state at three Rf3- linked markers. These plants had a high pollen and panicle fertility (75% to 97%), indicating that with increasing homozygosity of Rf3 locus, fertility restoration to WA cytoplasmic male sterility in the genetic background of CMS recipient line was further enhanced. Therefore, it can be concluded that Rf3 locus has an interaction to WA cytoplasmic male sterility in favor of increasing the rice pollen and panicle fertility, and hence it can be utilized along with other restoring fertility genes in hybrid seed production program of rice.

    The growth hormone receptor (GHR) is a cell surface receptor for growth hormone (GH) that is required for GH to carry out its effects on target tissues. Therefore, the aim of this study was to identify and analysis of SNPs in the promoter region of GHR gene in Iranian local cattle breed (Bos taurus). The part of promoter region of GHR gene was screened by single strand conformation polymorphism (SSCP) method and DNA sequencing. A total of 3 distinct SSCP patterns were observed which further revealed an A/G transition at position - 154 and two length of TG microsatellite (11 and 17 TG repeats) upon sequence analysis in amplified fragment. The genotype frequencies of AA (TG17/17), G (TG11/11) and GA (TG17/11) were 0.34, 0.24 and 0.52 respectively. In silico analysis has been shown that the single nucleotide polymorphism (SNP) at position -154 is very close to the putative binding site for C/EBP transcription factor. The possible functional activity of identified genetic variation should be approved using gene expression analysis.

    Plants process several potential cellular mechanisms for detoxification of heavy metals. One of the most important mechanisms is synthesis of metal binding peptides and proteins such as metallothioneins (MTs). MTs are low molecular weight and cystein-rich proteins that can bind metal ions through their thiol groups. In this study the coding sequence of gene encoding OsMTI-1b isoforms from rice (), was cloned in pET41a and transferred into expression host, Escherichia coli strain Rosetta¬ (DE3). After induction with IPTG, considerable amount of recombinant proteins was produced in the soluble fraction of the E.coli transforman. Recombinant proteins were purified using affinity chromatography. The tolerance of cells expressing recombinant proteins toward Ni, were compared to control by plotting their growth curve in addition to determination of the amount of accumulated Ni ions in bacterial cells and culture medium using inductively coupled plasma atomic emission spectroscopy (ICP-AES). According to the results, over-expression of OsMTI-1b isoform increased the tolerance of E. coli cells to Ni through accumulation of more metal ions inside cells. Furthermore, the UV absorption spectra and competitive reactions of in vitro Ni-incubated proteins with 5-5' dithiobis (2-nitrobenzoic) acid (DTNB) revealed that GST-OsMTI-1b protein is able to form Ni-thiolate clusters. Taken together, these data indicate that OsMTI-1b isoform may be involved in protection of rice cells against heavy metal toxicity.

    Transcriptome analysis reveals novel insights in the plant responses against biotic and abiotic stresses. cDNA-AFLP is appropriate and repeatable method for genome wide expression analysis when genomic information is limited. Although several factors may affect sensitivity, accuracy and repeatability of cDNA-AFLP technique so attention to these factors increases quality of results. In this study cDNA-AFLP technique was optimized for analysis of potato against Ralstonia solanacearum under in vitro conditions through examining various factors and their interactions, that affecting the results of such analysis. Results showed that RNA extraction method and the use of mRNA has large effects on the reliability of results. RNAs extracted based on using a purification column were monotonous. Ribosomal RNA and inhibitors alleviated by mRNA extraction, Also optimization of factors such as concentrations of template RNA, Mgcl2 and Taq DNA polymerase, in pre and selective amplification has significant effects on number, clarity and concentration of amplificated cDNA fragments.

    Copy number variation (CNV) is important on biological mechanism. For CNV detection and distribution, three bovine autosomal chromosomes, BTA6, BTA14 and BTA6 that improved for quantitative trait loci (QTL) previously, were investigated. Blood sample we obtained from 580 animals and after DNA extraction the samples were genotyped by Illumina BovineSNP50v2 BeadChip. Three hundred eighty three samples were remained for further analysis, after correction for signal intensity and GC content. Data were analyzed based on UMD3.1 bovine genome assembly for BTA6, BTA14 and BTA20. After filtration 199 CNV were detected (132 losses and 67 gains) with 0.5 CNV per animal and mean and medium of 147.3 kb and 139.4 kb respectively. A proportion of loss to gain was 1.97 fold. The Max and Min Number of CNV detected on BTA6 and BTA20 respectively. The bioinformatics analysis showed CNV region's coverage some part of 77 reference gene in bovine genome. These results showed that CNV coverage remarkable part of the three chromosomes that contain several QTL. Because of the influence of CNV on gene dosage and gene structure, it can cause phonotypic variation in quantitative traits, so probably it can be useful on livestock breeding programs.

    Genetic diversity is a requirement for success in the breeding programs. In order to evaluate genetic diversity of 40 bread wheat genotypes through morpho-physiological traits and SSR markers, wheat genotypes were cultivated in Shahid Bahonar university research field in a randomized complete block design with 3 replications during 2011-2012. Wheat genotypes showed a considerable genetic diversity for all traits. Fertile tillers had the highest correlation with grain yield. In SSR experiment, 9 primers showed remarkable polymorphism. Considering all genotypes, 31 polymorphic bands with the mean of 3.4 per each primer were detected. The average expected heterozygosis was 0.36 for all loci. Wmc 420 had the highest diversity in evaluated population. Cluster analysis was performed based on Ward’s method and cultivars were classified based on the similarity. The cluster Information can be used in the breeding programs for yield improvement in drought prone environments.
