    سفیدک داخلی مهم ترین بیماری اندام هوایی خیار در کشت های گلخانه ای است. این بیماری توسط گونه Pseudoperonospora cubensisبه وجود آمده و در صورت عدم کنترل خسارت زیادی وارد می کند. اثر قارچ کش های سیازوفامید (رانمن®% SC 34.5)، پتاسیم فسفیت (فسفیت®WSL 53 % )، فلوپیکولید+ پروپاموکارب (propamocarb hydrochloride 625g/l + fluopicolide 62.5 g/l) (اینفینیتو® (68،75 SC و فاموکسادون + سیموکسانیل (اکوییشن پرو®WG 52.5%) در کنترل بیماری سفیدک داخلی خیار در گلخانه ای در اربطان مرند (آذربایجان شرقی) با سابقه آلودگی به بیماری ارزیابی شد. آزمایش با 11 تیمار و چهار تکرار در قالب طرح کاملا تصادفی در سال 93 انجام گردید. تیمارهای آزمایش، قارچ کش های رانمن به میزان 2/0، 3/0 و 4/0 در هزار، فسفیت به میزان 2، 3 و 4 در هزار، اینفینیتو به میزان 25/1، 5/1 و 2 در هزار، اکوییشن پرو به میزان 3/0 در هزار ، و شاهد بدون سم پاشی بودند. سم پاشی کرت های آزمایشی بعد از مشاهده اولین علایم بیماری در سه نوبت با فاصله زمانی 10 روز انجام و آماربرداری ازکرت های آزمایشی، هفت روز بعد از آخرین سم پاشی انجام شد. شدت آلودگی برای هرکرت براساس میزان لکه روی سطح برگ و تشکیل و یا عدم تشکیل اسپورانژیوم محاسبه شد. تجزیه آماری داده ها با نرم افزار SPSS و مقایسه میانگین ها با آزمون چند دامنه ای دانکن انجام گرفت. نتایج نشان داد که بهترین اثر کنترل بیماری مربوط به تیمارهای رانمن 4/0 در هزار، فسفیت 4 در هزار و اینفینیتو 2 در هزار به ترتیب با شدت بیماری 3/3، 53/7 و 69/8 درصد نسبت به 79 درصد در تیمار شاهد بود (P≤0.01). بدین ترتیب این قارچ کش ها می توانند در گلخانه بصورت متناوب مورد استفاده قرار گیرند.

    قارچ Sclerotinia sclerotiorum (Lib) de Bary یکی از عوامل مهم کاهش عملکرد بسیاری از محصولات مهم اقتصادی محصولات زراعی و باغی می باشد. بسیاری از روش‎های مرسوم در مدیریت بیماری موثر نیست. در این میان کنترل بیولوژیک با قارچ‎های آنتاگونیست، می‎تواند در کاهش بیماری چاره‎ساز باشد. در این پژوهش اثر آنتاگونیستی 14 جدایه بومی قارچ تریکودرما مربوط به چهار گونه مختلف برای کنترل قارچ بیمارگر S. sclerotiorum در شرایط in vitroبا بررسی های کشت دوتایی، ترکیبات ضدقارچی فرار و غیرفرار تولید شده توسط آنتاگونیست‎ها، ارزیابی شد. بازدارندگی رشد میسلیوم‎های S. sclerotiorum به وسیله جدایه های تریکودرما در کشت دوتایی، ترکیبات فرار و غیرفرار بین 4/53-5/35، 33-0 و 100-0 درصد متغیر بود، در این میان Trichoderma harzianum (A10) بیش ترین تاثیر را در مهار رشد میسلیوم‎ها در کشت دوتایی و T. harzianum (A2 و A10) بیش ترین تاثیر را در مهار رشد میسلیوم‎ها با تولید مواد فرار و غیرفرار داشتند. که در نهایت آنتاگونیست T10 (T. harzianum) موثرترین جدایه در کنترل قارچ عامل بیماری درشرایط آزمایشگاه انتخاب شد. در بررسی سه قارچکش تیوفانات متیل، رورال‎تی‎اس و بنومیل در شرایط آزمایشگاه، مشاهده شد که قارچ‎کش تیوفانات متیل در غلظت 10 پی‎پی‎ام و دو قارچ‎کش دیگر در غلظت 100 پی‎پی‎ام توانستند به طور کامل مانع از رشد میسلیوم‎ها شوند. در بررسی مقایسه کنترل شیمیایی و بیولوژیکی در شرایط گلخانه، مشاهده شد که تیمار خاک با آنتاگونیست هشت روز قبل از آلودگی با قارچ بیمارگر، تاثیر بهتری حتی نسبت به کنترل شیمیایی دارد. این مطالعه نشان داده که استفاده از ترکیبات بیولوژیک در زمان مناسب جهت پیشگیری از وقوع این بیماری، نسبت به سموم شیمیایی از کارایی بیشتری برخوردار است.

    خصوصیات آسکوسپورهای هشت جمعیت قارچ های عوامل بیماری ساق سیاه کلزا Leptosphaeria maculans و L. biglobosa در استان های مازندران و گلستان جداسازی شده از بقایای کلزا و خردل وحشی مورد ارزیابی قرار گرفت. نتایج تجزیه واریانس طول و عرض آسکوسپورها نشان داد که اختلاف معنی داری در سطح 1% در جمعیت های هر دو گونه وجود دارد. تغییرات طول و عرض آسکوسپورها به ترتیب 25/6 و 5/2 میکرومتر از بالاترین تغییرات دامنه طول و عرض آسکوسپورهای دیگر مطالعات انجام شده در جهان بیشتر بود. همچنین اختلاف معنی داری از نظر طول لوله تندش پس از 4، 16 و 24 ساعت از آزمایش مورد بررسی مشاهده گردید. هاگ های جنسی جمعیت مناطق دشت ناز ساری، کوهی خیل و فاضل آباد دارای تعداد لوله تندش بیشتر، کوتاهتر، پیچ خورده و با منشاء از سلول های میانی بودند در صورتیکه در جمعیت مناطق لالا، مازاروستاق و خالخیل لوله تندش بلندتر، صاف و از سلولهای طرفین منشاء گرفته بود که با در نظر گرفتن خصوصیات مربوط به ریخت شناسی لوله تندش، به ترتیب هاگ های جنسی بخشی از جمعیت های بررسی شده مکان های اول را تیپ های بیماری زا L. maculansو اکثر جمعیت های مناطق دوم را تیپ غیر بیماری زا L. biglobosaتشکیل می دهد. با توجه به بالا بودن درصد جوانه زنی اغلب جمعیت های قارچ عامل بیماری ساق سیاه کلزا در این بررسی، می توان نتیجه گرفت که بیشتر جمعیت ارزیابی شده از مناطق مختلف، متعلق به گروه غیر بیماری زا است. با توجه به خصوصیات لوله تندش جمعیت های قارچ بر روی خردل وحشی، این جمعیت ها غیربیماری زا تشخیص داده شد. رابطه ایی بین خصوصیات مربوطه به لوله تندش جمعیت های قارچ بر روی خردل و بیماریزا بودن آن مشاهده نشد.

    در این مطالعه تنوع ژنتیکی 34 ژنوتیپ زراعی و وحشی جو به همراه رقم حساس افضل از لحاظ حساسیت و مقاومت به بیماری سفیدک پودری در قالب طرح کاملا تصادفی با سه تکرار در گلخانه دانشگاه ایلام ارزیابی گردید. مایه زنی با قارچ عامل بیماری در مرحله تورم سنبله صورت گرفت. DNA ژنومی نمونه های برگی بر اساس روش دویل و همکاران استخراج گردید. با استفاده از 16 آغازگر ISSR و شش آغازگر RAPD به ترتیب 125 و 32 باند تشکیل شد که در مجموع 27/99 و 100% باندها از چندشکلی برخوردار بودند. میانگین هتروژنی مورد انتظار، مشاهده شده و همچنین میانگین شاخص شانون به ترتیب در نشانگرهای ISSR،32/0، 25/0 و 35/0 و RAPD 28/0، 31/0 و 42/0 محاسبه شد. تجزیه خوشه ایبه روش UPGMA، 34 ژنوتیپ مورد مطالعه را در 5 گروه قرار داد که به ترتیب گروه های 1 و 2 ژنوتیپ های مقاوم ونیمه مقاوم، گروه 3 ژنوتیپ های حساس و گروه 4 و 5 ژنوتیپ های نیمه حساس تا حساس را شامل شدند. به طور کلی، نتایج نشان داد که با استفاده از مارکرهایی همچون RAPD و ISSR می توان ژنوتیپ های حساس را از مقاوم تفکیک نمود که در برنامه های اصلاحی کمک شایانی به محقیقن می نماید.

    در این تحقیقاثر سالیسیلیک اسید به منظور کاهش بیماری زایی ویروس X سیب زمینی (PVX) بر روی پارامتر های رشد از جمله میزان کلروفیل، سطح برگ، فاصله میانگره، تعداد برگ، طول ساقه، تعداد غده و وزن غده در دو رقم سیب زمینی (آگریا و مارفونا) در شرایط گلخانه ای بررسی گردید. گیاهچه هائی که عاری از عوامل بیماریزا بودند تکثیر شده و گیاهچه های رشد کرده به گلدان ها با خاک مناسب و استریل منتقل گردیدند. گیاهچه ها با غلظت های صفر (شاهد)، 2/0، 5/0، 1 و 2 میلی مولار سالیسیلیک اسید مورد تیمار قرار گرفتند. عصاره فعال برگ توتون رقم تامسون آلوده به PVX برای آلوده سازی گیاهان سیب زمینی به کار رفت. نتایج نشان داد که علائم آلودگی PVX بر روی برگ های گیاهان آلوده نسبت به گیاهان شاهد در دو رقم آگریا و مارفونا خیلی مشهود نبود ولی بر روی برگ گیاهان توتون آلوده با پیچش برگ آن شناخته می شد. میزان سالیسیلیک اسید درگیاهان آلوده تقریبا دو برابر گیاهان سالم بود. همچنین نتایج نشان داد که تیمار سالسیلیک اسید با غلظت 2/0 تا 5/0 میلی مولار باعث کاهش 25/27 درصد خسارات ناشی از PVX می گردد. علاوه براین به نظر می رسد که رقم مارفونا نسبت به رقم آگریا در برابر PVX مقاومت بیشتری دارد.

    چکیده
    قارچ خوراکی دکمه ای (Agaricus bisporus) یکی از محصولات غذایی و دارویی حاوی انواع ویتامین ها و املاح معدنی است که از تامین کنندگان پروتئین و کالری موردنیاز افراد جامعه به حساب می آید. با تشخیص ارزش بالای قارچ های خوراکی و توسعه کشت و پرورش این نوع قارچ ، مدیریت واحدهای پرورش ازنظر کنترل بیماری ها حائز اهمیت است. انواع بیماری ها، خسارت قابل توجهی را به پرورش دهندگان تحمیل می نمایند. لذا تعیین اولویت های مبارزه با عوامل بیماری زای قارچ خوراکی ضروری است. بدین منظور با انتخاب روش تحلیل سلسله مراتبی به ارزش‎گذاری هریک از بیماری های شایع در استان های تهران و البرز با استفاده از شاخص های تعیین شده پرداخته شد. طراحی پرسش‎نامه با استفاده از معیارهایی چون کیفیت و بازار پسندی، مدت نگهداری، هزینه کنترل بیماری، امکان کنترل بی ضرر و میزان شیوع بیماری وغیره انجام پذیرفت. نتایج حاصل از مدل AHP (Analytical Hierarchy Process) نشان داد، با توجه به خروجی پرسش نامه های تکمیل شده توسط تولیدکنندگان قارچ، اساتید و متخصصان و مصرف کنندگان، بیماری باکتریایی لکه قهوه ای در استان های البرز و تهران با وزن 39% نیاز به بالاترین کنترل و مبارزه داشته و بیماری قارچی حباب تر با 33% در رتبه دوم قرار دارد. در انتها نیز بیماری باکتریایی مومی (Pseudomonas spp.) با 19 درصد و بیماری کپک سبز با نزدیک به 8 درصد از کل اوزان قرار دارند. این نتایج نشان دهنده بیشترین سطح خسارت زایی در دو بیماری لکه قهوه ای باکتریایی و حباب تر و لزوم توجه به آن ها نسبت به دیگر بیماری های شایع در این منطقه است. همچنین بیماری کپک سبز با توجه به نتایج تحلیل سلسله مراتبی، کم اهمیت ترین بیماری در منطقه مورد مطالعه محسوب می گردد.

    Downy mildew caused by Pseudoperonosprora cubensis is the most important foliar disease of greenhouse cultivated cucumber which causes significant economic damages in Iran annually. The effect of five fungicides including Cyazofamid (Ranman® SC 34.5), potassium phosphate (Fosphite® WSL 53%), fluopicolide (5.53%w/w) (55.3%w/w) propamocarb hydrochloride (Infinito® SC 687.5) and famoxadone cymoxanil (Equation pro® WDG 52.5%) on the control of the disease was assessed in a greenhouse in Arbatan (Marand, East Azerbaijan, Iran) with precedent infection. The experiment was performed in a completely randomized design with 11 treatments and four replications during 2014. Treatments included Ranman at the rates of 0.2, 0.3 and 0.4 ml/l, Fosphite at the rates of 2, 3 and 4 ml/l, Infinito at the rates of 1.25, 1.5 and 2 ml/l, Equation pro at the rates of 0.3 g/l and control without spraying. Spraying was carried out immediately after the appearance of disease symptoms and was repeated for 3 times at intervals of 10 days. Data on disease incidence was recorded one week after the third spraying of fungicides. For this purpose 100 leaves were collected from 10 plants randomly from each plot and disease severity was assessed by measurement of diseased spots on the surface of leaves and sporangium formation density. Collected data were analyzed by SPSS software and means comparison were done by Duncan's multiple range tests. Results indicate that fungicide application caused significant decrease in the disease severity compared with the control treatment. Minimum disease severity occurred in Ranman 0.4 ml/l, Fosphite 4 ml/l and Infinito 2 ml/l with the disease severity of 3.3, 7.53 and 8.69% respectively compared to the control treatment 79%. These fungicides can therefore be applied as alternatives to other chemicals in the control of cucumber downy mildew.

    Sclerotinia sclerotiorum (Lib.) de Bary is a major plant pathogen that plays a crucial role in yield reduction of economically important crops. Most of the conventional methods are not effective in management of S. sclerotiorum. The Biological control by fungal antagonists could be useful in reducing disease. In the present research, antagonistic effects of indigenous Trichoderma species on S. sclerotiorum were determined in vitro. Dual culture technique, production of volatile and non-volatile compounds were used to determine the percentage of mycelial growth inhibition. The mycelial growth inhibition of S. sclerotiorum by Trichoderma strains ranged from 35.5-53.4, 0-33 and 0-100 percent in dual culture, volatile compounds and non volatile metabolites tests, respectively. Trichoderma harzianum (A10) showed the highest impact on mycelial growth inhibition in dual culture and T. harzianum (A2 & A10) had the highest effect on the inhibition of mycelial growth by the production of volatile compounds and non-volatile compounds. On the whole, antagonist T10 (T. harzianum) was most effective in controlling pathogen isolates under laboratory conditions. Also effects of three fungicides including thiophanate- methyl, Rovral TS and Benomyl were studied in laboratory conditions. Results showed that thiophanate-methyl (10 ppm) and the other two fungicides at concentration of 100 ppm were able to completely inhibit the mycelial growth. Comparison of effectiveness of chemical and biological control methods in greenhouse conditions indicated that inoculation of soil with antagonist prior to infection with the pathogen had better effect than chemical control did. This study has shown that the use of biocontrol agents at the right time is more efficient in prevention of the disease than chemical fungicides.

    Ascospore dimensions of eight populations of Leptosphaeria maculans and L. biglobosa, causal agents of rapeseed blackleg disease, isolated from debris of rapeseed (Brassica napus) and wild mustard (Sinapis arvensis) were evaluated in Mazandaran and Golestan provinces. Results of ANOVA indicated significant differences in the dimensions of ascospores of the different populations. The greatest length and width of sexual spores were 87.5 and 12.5 micrometer, respectively. Variations in the range of Length and width of ascospores in the studied populations was measured, 46.25 and 7.5 micrometer, respectively also these variations were 6.25 and 2.5 micrometers greater than the dimensions of ascospores reported by other studies in the world. The length of sexual spores isolated from stem debris of mustard was much bigger than those of canola. Germination percentage of most studied populations was more than reported in other studies, so likely the most of studied populations of different regions belong to non aggressive group. Sexual spores of aggressive types have greater number of germ tubes, shorter in length, twisted and originate from the middle cells. While in the nonaggressive type the germ tubes grow out of end cells and are not twisted. With regard to morphological characteristics of sexual spore germ tube and also cotyledon test characteristics, field symptoms, pigment production and colony growth rate it was determined that ascospores of the studied populations from Dasht-e-Naz of Sari, Kohikheil and Fazel Abad were of the aggressive type (L. maculans) and most of population in Lala, Mazarustaq and Talookola areas belonged to nonaggressive type (L. biglobosa). There was no correlation between germ tube characteristics of fungus populations on mustard and their pathogenicity.

    Genetic diversity of 34 wild and cultivated barley cultivars along with susceptible cultivar namely Afzal was evaluated regarding their sensitivity and resistance to powdery mildew in a randomly complete design with three replications in greenhouse of Ilam University. Plants were inoculated at anthesis stage with Blumeria graminis f.sp. hordei. When 16 ISSR and 6 RAPD primers were used, 125 and 32 alleles were amplified where 99.27% and 100% of the alleles had polymorphism, respectively. Average expected and observed heterogeneity and average Shanon index in ISSR were calculated to be 0.32, 0.25 and 0.35 and for RAPD were 0.28, 0.31 and 0.42. Cluster analysis by UPGMA method categorized genotypes in five clusters of which groups 1 and 2 were resistant to semi-resistant, group 3 as susceptible, and also group 4 and 5 were moderately­ susceptible to susceptible ones, respectively.In general, our results indicated that using molecular markers such as RAPD and ISSR can differentiate genotypes into resistant or susceptible groups that would help researchers in breeding programs.

    In this study, in order to reduce the pathogenesis of potato virus X (PVX) in two potato (Solanum tuberosum L.) cultivars (Marfona and Agria), the effects of salicylic acid on growth parameters (chlorophyll content, leaf area, distance between nodes, number of leaves, stem length, number and weight of tubers) were investigated under greenhouse conditions. Healthy potato plantlets were propagated and transferred to pots containing suitable and sterilized soil. The plantlets were sprayed with different concentrations (0, 0.2, 0.5, 1 and 2 mM) of salicylic acid weekly at the rate of 10 ml per plant and for 4 weeks. The leaves of experimental plants were inoculated with PVX, extracted from infected tobacco leaves. Although viral infection symptoms on tobacco were manifested as leaf rippling, no such symptoms were visible on infected potato leaves. Results showed that salicylic acid content in infected plants was about twice that in healthy control plants. Also, treatments with 0.2 and 0.5 mM of salicylic acid resulted in 27.25% reduction in potato damage. Moreover, our study revealed that Marfona cultivar was more resistant to PVX than Agria cultivar.

    Button mushroom (Agaricus bisporus) is one the food and drug products containing different kinds of vitamins and minerals. It is a good supplier of protein and calorie that are required for human nutrition. The high value of mushroom and its cultivation indicate the impact of management of mushroom farms in terms of disease control. A number of diseases cause considerable damage to this product. Therefore, we found it necessary to determine their priorities in combating mushroom pathogens. For this and other reasons, AHP (Analytical Hierarchy Process) was used to evaluate common disease indices for the two provinces under study (Tehran and Alborz). A questionnaire using criteria such as quality and marketing, storage duration, cost of disease control, possibility of disease control and disease outbreaks and so on was designed. The questionnaires were filled out by mushroom producers, experts and consumers in both Tehran and Alborz provinces. The results showed that, bacterial brown blotch disease with 39% weight and fungal wet bubble disease, with 33% weight receive priority for control, respectively. The bacterial disease mummy with nearly 19 percent and green mold disease with 8 percent of the total weight ranked in descending order. These results indicate that the highest level of damage in the two areas are caused by brown blotch and wet bubble diseases and greater attention should be paid to their control than to the other common diseases. The green mold disease, according to results of AHP, is considered the least important one in the study area.
