    گلوکز اکسیداز (β-D-گلوکز:اکسیژن 1-اکسیدوردوکتاز) با استفاده از اکسیژن مولکولی به عنوان پذیرنده الکترون، اکسیداسیون β-D-گلوکز را به گلوکونیک اسید کاتالیز نموده و منجر به تولید پراکسید هیدروژن می شود. این آنزیم در دامنه وسیعی از قارچ های مختلف، عمدتا جنس های Aspergillus و Penicillium تولید شده و کاربردهای وسیعی در صنایع مختلف از قبیل شیمی، داروسازی، صنایع غذایی، بیوتکنولوژی و غیره دارد. در این مطالعه، ژن کدکننده گلوکز اکسیداز از قارچ Penicillium funiculosum با استفاده از تکنیک واکنش زنجیره ای پلی مراز (PCR) جداسازی و در ناقل pUC19 همسانه سازی شده و ساختار مولکولی، ویژگی های بیوشیمیایی و فیلوژنتیکی آن مورد بررسی قرار گرفت. توالی نوکلئوتیدی این ژن، تحت عنوان PfGOX، به طول bp1818 بوده و یک پروتئین با 605 آمینواسید را رمز می کند. وزن مولکولی محاسبه شده و نقطه ایزوالکتریک پیش بینی شده این پروتئین به ترتیب برابر 65/78 کیلو دالتون و 5/05 می باشد. بررسی ساختارهای دوم و سوم توالی پروتئینی PfGOX نشان داد که این پروتئین از نظر ساختاری مشابه با توالی های گلوکز اکسیداز گزارش شده از قارچ های دیگر می باشد. توالی پروتئینی بدست آمده شباهت زیادی را با توالی های گلوکز اکسیداز سایر قارچ ها از قبیل Penicillium amagasakiense، Talaromyces flavus، Talaromyces stipitatus و Talaromyces variabilis نشان داد. بررسی فیلوژنتیکی نیز مشخص کرد که این ژن به زیرگروه IA گلوکز اکسیدازهای قارچی تعلق دارد.

    باکتری سایکروتروف تجزیه کننده نشاسته از نمونه های خاک جمع آوری شده از حومه جنوب غربی تهران جداسازی شد. با استفاده از روش های بیوشیمیایی و مولکولی براساس تعیین توالی ژن 16S rDNA این باکتری بعنوان گونه ایی از جنس اگزیگوآباکتریوم شناسایی و تحت عنوان اگزیگوآباکتریوم سویه SH3 نام گذاری گردید. این باکتری در دماهای 4، 30، 37، و 48 درجه قادر به رشد و تولید آنزیم های آمیلولیتیک بود. دمای بهینه برای تولید آنزیم های آمیلولیتیک توسط این باکتری °C30 و pH بهینه 7 تعیین شدند. این باکتری قادر بود نشاسته اضافه شده (1%) به محیط کشت را در مدت 9 ساعت بطور کامل تجزیه نماید. سوپرناتانت محیط کشت باکتری بعنوان منبع خام آنزیم برای بررسی فعالیت آمیلازی مورد استفاده قرار گرفت. این سوپرناتانت در دامنه رسیعی از دماها بین 4 تا 45 درجه، و pH بین 5 تا 9 واجد فعالیت آمیلازی بود. دما و pH بهینه برای فعالیت آمیلازی بترتیب 37 درجه و 7pH تعیین شدند. بنظر می رسد که آنزیم های سازگار با سرما بدلیل برخورداری از فعالیت کاتالیتیک زیاد برای کاربرد در دمای محیط مثل شستشو و فراوری غذا نسبت به انواع مزوفیل و گرمادوست از مزیت بیشتری برخوردار هستند.

    تابع نیروی دانش پایه نوعی از توابع نمره دهی می باشد که از آن در زمینه تشخیص فولد پروتئین ها با موفقیت قابل توجهی استفاده شده است. ما در این مطالعه کارایی نوعی تابع انرژی دانش پایه و تابع نیروی هم ارز آن را در تشخیص کمپلکس های درست پروتئین-پروتئین از کمپلکس های نادرست، با یکدیگر مقایسه کردیم. مقدار نیروی کل که از یک جزء کمپلکس (گیرنده/لیگاند) بر جزء دیگر وارد می شود به عنوان معیار پایداری کمپلکس، مورد استفاده قرار گرفت. چنین انتظار می رود که این نیرو در ساختار طبیعی کمترین مقدار را داشته باشد. جهت ارزیابی کارایی هر روش، دو مجموعه مورد استفاده قرار گرفت که یکی از آنها با الگوریتم داکینگ جسم نرم و دیگری با الگوریتم داکینگ جسم سخت ایجاد شده بودند. نتایج حاصل از این مقایسه نشان می دهد نرخ موفقیت مدل انرژی در انتخاب ساختار های طبیعی و نزدیک به طبیعی بالاتر از روش نیرو می باشد. ظاهرا وابستگی مقدار نیرو به شکل ناحیه اتصال کمپلکس ها باعث ایجاد خطاهایی در مدل نیرو شده که سبب نامناسب گردیدن آن در نمره دهی کمپلکس های داک شده می شود.

    هدف این تحقیق بررسی قدرت نشانگر کلروپلاستی trnL-F در تمایز ژنتیکی و ارتباط فیلوژنتیکی سپیدار (Populus alba) و سفیدپلت (Populus caspica) است. تعداد 15 نمونه برگی از هفت رویشگاه مختلف جنگل های هیرکانی ایران؛ استان های گیلان و مازندران، جمع آوری شدند. DNAی ژنومی از برگ ها با استفاده از روش ترکیبی CTAB و SDS- پتاسیم استات استخراج گردید. واکنش PCRی با استفاده از پرایمرهای جهانی انجام شد و قطعات trnL-F تکثیر شده توالی یابی گردید. نتایج نشان دادند که طول منطقه trnL-F تقریبا برای جمعیت های هر دو گونه در حدود 392-394 نوکلئوتید است. در مقایسه با توالی های موجود در بانک ژنی، تعداد نوکلئوتیدهای آدنین و تیمین در سفیدپلت و سپیدار بیش تر از دیگر نوکلئوتیدها است. ترکیب نوکلئوتیدی این دو گونه بسیار مشابه با حداقل فاصله ژنتیکی از یکدیگر (0/005=) می باشد. آنالیز پارسیمونی از 582 جایگاه نوکلئوتیدی؛ 178 جایگاه محافظت شده، 197 جایگاه متغیر، 151 جایگاه پارسیمون و 46 جایگاه انحصاری را نشان داد. رسم درخت فیلوژنی براساس نشانگرtrnL-F نیز نشان داد که توالی این نشانگر نمی تواند تمایز بین پایه های دو گونه سپیدار و سفیدپلت ایجاد کند چراکه نمونه های بررسی شده در یک گروه مشترک قرار گرفتند. از نتایج این تحقیق می توان استنتاج نمود که نشانگر کلروپلاستی trnL-F نتوانست دو گونه سپیدار و سفیدپلت را از یکدیگر متمایز نماید. بنابراین برای شناخت دقیق این گونه ها، بازسازی فیلوژنی با استفاده از سایر نشانگرهای کلروپلاستی و یا هسته ای پیشنهاد می گردد.

    پروتئین های ترشحی و خلال غشایی در بسیاری از فرایندهای زیستی نقش کلیدی دارند. نرم افزار های بیو انفورماتیکی متعددی جهت یش بینی این پروتئین ها طراحی و ساخته شده است. همچنین چندین روش تجربی نیز جهت به دام انداختن این پروتئین ها معرفی شده است.
    سیستم ترشح مخمر یا به اختصار YST، یکی از روش های تجربی است که در این زمینه مورد استفاده قرار گرفته است. این سیستم شامل حامل های بیانی و یک سویه مخمر جهش یافته است. این سویه فاقد اینورتاز است. اینورتاز آنزیمی است که ترشح آن باعث رشد مخمر بر روی محیط انتخابی سوکرز می شود. سه وکتور بیانی این سیستم، اینورتاز فاقد متیونین آغازی و توالی نشانه را در سه قالب مختلف کد می کنند. عرضه cDNA های حاوی توالی نشانه در ابتدای اینورتاز ترشح آن و در نتیجه رشد مخمر بر روی محیط انتخابی سوکرز را فراهم می آورد.
    هدف از این تحقیق راه اندازی سیستم YST جهت شناسایی پروتئین های ترشحی و خلال غشایی بود. به این منظور عملکرد دو توالی نشانه پیش بینی شده توسط نرم افزار های بیو انفورماتیکی، در انتهای آمین دو پروتئین انسانی Ppic و Trmt1 و توالی نشانه شناخته شدهPi16 به عنوان کنترل مثبت وتوالی نشانه میتوکندری Tfam، به عنوان کنترل منفی با این روش مورد ارزیابی قرار گرفت. نتایج حاصل از کنترل مثبت و منفی حاکی از کارکرد صحیح سیستم بود و در مورد توالی مورد نظر در Trmt1، همانطور که انتظار می رفت در این سیتم فاقد عملکرد بود اما توالی مورد نظر در Ppic، بر خلاف انتظار قادر به عملکرد به عنوان توالی نشانه در این سیستم نبود.

    تا کنون تلاش های متعددی در جهت جداسازی آنزیم هایی که در شرایط سخت صنعتی پایداراند از میکروارگانیسم های کران دوست از جمله نمک دوست صورت گرفته است. آرکی نمک دوست افراطی سویه Ha25، آنزیم پولولاناز خارج سلولی نمک دوست و گرمادوست نسبی را در شرایط پرتنش در محیط کشت دارای 23% نمک تام تولید می کند. تعیین توالی ژن S rRNA16 و ویژگی های فنوتیپی نشان داد سویه Ha25 متعلق به جنس Halorubrum است. تولید آنزیم با میزان رشد هماهنگ بود و در میانه فاز سکون به بیشترین میزان خود رسید. ترشح این آنزیم در حضور نمک سدیم کلراید صورت گرفته و در حضور غلظت 3 تا 4 مولار نمک NaCl بیشینه تولید آنزیم مشاهده شد (U/ml 85). دما و pH بهینه برای تولید آنزیم به ترتیب 45 درجه سانتی گراد و0/7 تعیین شد. منابع مختلف کربن مانند مالتوز، نشاسته و پولولان به عنوان القا کننده و گلوکوز به عنوان مهار کننده تولید آنزیم شناسایی شدند. غلظت بهینه منیزیوم برای تولید آنزیم 0.1 مولار بوده و این در حالی است که هیچ رشدی و متعاقب آن، هیچ تولیدی در عدم حضور یون های منیزیوم مشاهده نشد. بیشینه فعالیت آنزیم در حضور غلظت 3 مولار نمک NaCl و در دمای 50 درجه سانتی گراد بوده و در دمای 45 و 35 درجه سانتی گراد به ترتیب کاهش 25% و 72% در فعالیت آنزیم مشاهده شد. pH بهینه برای فعالیت آنزیم 8 بوده و در pH 6 و 9، به ترتیب 49.5% و 32.4% از فعالیت آنزیم باقی ماند.

    پیل سوختی میکروبی دستگاهی الکتروشیمیایی است که انرژی شیمیایی موجود در مواد آلی و غیرآلی را با به کارگیری میکروارگانیسم به عنوان بیوکاتالیست (Biocatalyst)، به انرژی الکتریکی تبدیل می کند. در این تحقیق تولید جریان بیوالکتریسیته در پیل سوختی میکروبی بی واسطه مورد ارزیابی قرار گرفته است. جهت ارزیابی عملکرد الکتریکی پیل سوختی ساخته شده از منحنی قطبش استفاده شده است. منبع کربنی به عنوان سوبسترا و پساب بی هوازی فاضلاب شهرک صنعتی بابل به عنوان بیوکاتالیست مورد استفاده قرار گرفت. در انجام آزمایشات از غشای نفیونی (Nafion 117) برای جداسازی محفظه آند و کاتد و انتقال پروتون از آند به سمت کاتد استفاده شد. الکترود های به کار گرفته شده از جنس گرافیت بوده است. بیشینه دانسیته توان و جریان الکتریکی تولیدی در آزمایشات صورت گرفته با غلظت g/l 0/5 گلوکز به عنوان منبع کربنی و 10 ٪ فاضلاب به عنوان ماده تلقیحی به ترتیب به ترتیب mW/m2 186 و mA/m2 1078به دست آمده است

    با توجه به افزایش سطح محصولات دست ورزی شده ژنتیکی، شناسایی و تعیین سطح کمی آنها با روش های مطمئن و حساس ضروری می باشد. در ایران بخش عمده سویای مصرفی، وارداتی و از نوع تراریخت می باشد. در این مطالعه از روش های مولکولی جهت شناسایی کیفی و کمی بذور سویای تراریخت مقاوم به رانداپ در سطوح مختلف شامل تعیین نوع تراژن بکار رفته، سازه های ژنی و رخداد اختصاصی سویای مقاوم به رانداپ استفاده شده است. در ابتدا با استفاده از چندین جفت آغازگر مربوط به عناصر تراژن، آنالیزهای مولکولی غربال گری در سطح کیفی انجام گردید. در ادامه، آنالیزهای نیمه کمی برای سری رقت های مختلفی از DNA بدست آمده از این بذور تراریخت با همان جفت آغازگرها نسبت به قطعه تکثیر یافته با آغازگرهای ژن کنترل اختصاصی سویا (لکتین) نیز انجام شد. بدین ترتیب، میزان حد آستانه مختلفی برای شناسایی این عناصر بدست آمد بطوری که در سنجش مبتنی بر توالی پیش بر 35S نسبت به ژن خانه دار لکتین، میزان 1% و در سنجش مبتنی بر توالی های پایان دهنده nos و cp4 epsps نسبت به ژن خانه دار لکتین میزان 0/5% و در نهایت در سنجش مبتنی بر توالی های سازه ژنی (CTP-35S) و رخداد اختصاصی سوی (pDNA-35S)،میزان 5% را نشان داد. بطور کلی، حساسیت آزمون نیمه کمی برای توالی های پایان دهنده nos و cp4 epsps با استفاده از روش زنجیره ای پلی مراز معمولی بیشتر از سایر عناصر بوده است. بنابراین این روش می تواند جهت شناسایی و غربال گری سریع نمونه ها و محموله های حاوی سویای تراریخت مقاوم به رانداپ به کار رود.

    آلژینات لیازباکتریایی آنزیمی پری پلاسمی است که در تولید و تخریب آلژینات اهمیت دارد. خصوصیات این آنزیم اعم از وزن مولکولی، pI، مقاومت حرارتی، pH و دمای بهینه و تاثیر بر آلژینات باکتریایی در سویه های یک باکتری هم متفاوت می باشد.
    در این مطالعه، به منظور دسترسی به آلژینات لیازی با خصوصیات ویژه و کار آمد در تخریب آلژینات باکتریایی، ازجدایه موکوئیدی Pseudomonas aeruginosa به دست آمده از خلط بیمار مبتلا به عفونت ریوی استفاده شد. از آنجایی که آلژینات لیاز در تولید آلژینات نیز نقش دارد لذا این سویه برای مطالعه انتخاب شد.
    ابتدا آلژینات لیاز با روش شوک حرارتی از فضای پری پلاسمی استخراج و سپس با استفاده از غلظت مناسب سولفات آمونیوم و کروماتوگرافی تعویض آنیونی خالص شد. فعالیت آنزیم با روش تیوباربیتوریک اسید تعیین گردید. وزن مولکولی نسبی این آنزیم حدودkDa 60 تخمین زده شد. اثر زمان واکنش، pH، غلظت سوبسترا و دما بر فعالیت آلژینات لیاز بررسی گردید. نتایج نشان داد که آنزیم بهترین فعالیت را در غلظت mg/ml 0/4 از سوبسترا و بعد از 20 دقیقه واکنش در دمای C 37 و 8/5 pH دارد. فعالیت باقی مانده آنزیم بعد از 4 ساعت قرار گرفتن در دمای C 80، 33/31% تعیین گردید که نشان دهنده پایداری دمایی قابل توجه این آنزیم می باشد.
    این آنزیم توانایی تخریب آلژینات سویهM 8821 P. aeruginosa را دارد که این ویژگی همراه با پایداری حرارتی مناسب، آن را گزینه مناسبی برای درمان کمکی در مبارزه با عفونت های مقاوم به آنتی بیوتیک P. aeruginosa می سازد.

    کلزا (Brassica napus L.) یکی از مهمترین گیاهان روغنی جهان به شمار می رود. علی رغم تولید گیاهان تراریخته کلزا کارایی انتقال به این گیاه پایین می باشد. با توجه به اهمیت تاثیر ژنوتیپ گیاه و ترکیب محیط کشت بر تراریختی و باززایی و همچنین سمیت کانامایسین برای رشد این گیاه، توسعه یک روش مناسب جهت انتقال ژن بسیار ضروری است. در این پژوهش از ریزنمونه های برگهای لپه ای با استفاده از آگروباکتریوم تومی فاشینس سویه های حاوی ژن انول پیروویل شیکیمات 3-فسفات سنتاز (EPSPS) برای تراریختی رقم7045-91 PF استفاده شد. ریز نمونه ها پس از تلقیح با اگروباکتریوم محیط های مختلف هم کشتی، دمای 25 درجه سانتیگراد و تاریکی قرارداده شد. طی باززایی به منظور جلوگیری از نکروز شدن ریز نمونه ها و گزینش موثر از نیترات نقره به عنوان ممانعت کننده تولید اتیلن و افزایش پلکانی غلظت کانامایسین به عنوان ماده انتخاب کننده استفاده شد. برای این منظور ابتدا ریزنمونه ها در محیط باززایی حاوی نیترات نقره و بدون حضور کانامایسین نگهداری شدند. پس از آن با حفظ غلظت ثابتی از نیترات نقره (5 میلی گرم بر لیتر)، کانامایسین به غلظت 15میلی گرم بر لیتر افزایش داده شد. نوساقه های باززا شده به منظور القا ریشه زایی به محیط MS حاوی 15گرم بر لیتر ساکارز و غلظت 0/2 میلی گرم بر لیتر از IBA منتقل شدند. انتقال موثر ژن به گیاهان با آزمون های مولکولی تایید شد. در این بررسی مشخص شد که با افزایش تدریجی عامل انتخابی، درصد تراریختی نیز افزایش می یابد.

    تجمع مگنتیتFe3O4 در مغز و نقص در تنطیم انتقال و ذخیره آهن، با بسیاری از بیماری های تحلیل سیستم عصبی مانند آلزایمر ارتباط دارد. همچنین ثابت شده است پاتولوژی آلزایمر از طریق مکانیسم های مختلف با ناجور تاخوردگی و ایجاد تجمعات فیبریلی پروتئین تاو همراه است. از اینرو، در این مقاله میانکنش پروتئین تاو انسانی در حضور نانوذرات آهن مغناطیسی مورد بررسی قرار گرفت. نتایج با استفاده از روش های اسپکتروسکوپی فلورسانس و میکروسکوپ الکترونی نشان داد که نانوذرات آهن مغناطیسی قادر به القای تغییرات ساختاری در پروتئین تاو از پیچه نامنظم به ساختار بتا و در نتیجه ایجاد تجمعات فیبریلی پروتئین تاو می باشد. این نتایج، این حدس را تقویت می کند که احتمالا کریستالهای آهن مغناطیسی مغز در القای فیبریلی شدن پروتئین تاو دخالت داشته باشند.

    با هدف دستیابی به کلون های پایدار از سلول های S2 بیان کننده فاکتور 9 انعقادی انسانی (S2-hFIX)، در این مطالعه تاثیر عوامل مختلف بر تراآلایی و بیان پروتئین نوترکیب در سامانه بیانی S2 قابل القاء، با استفاده از یون فلزی به عنوان القاء کننده، مورد ارزیابی قرار گرفت. پس از تراآلایی یک سازه بیان کننده EGFP تحت کنترل پروموتر متالوتیونین(pMT-EGFP) به سلولهای S2 با روش های لیپوفکشن و کلسیم فسفات، بررسی نتایج بیانی نشان داد که انتقال سازه به این سلولها با روش کلسیم فسفات در pH=7.06 و غلظت μg/ml 10 کارآیی بالاتری دارد. الگوی الکتروفورزی محصولات RT-PCR فاکتور 9 ترشح شده از سلولهای S2-hFIX نشان داد که پروموتر متالوتیونین دروزوفیلایی بدنبال القا با سولفات مس از دقت تنظیمی بالایی برخوردار است و قادر به هدایت نسخه برداری در سطح بالا است. جداسازی کلون پایدار در دو محیط واجد میتومیسین (روش غیر مستقیم) و فاقد آن (روش مستقیم) مورد مقایسه قرار گرفت که در هر دو روش زمان تهیه کلون پایدار حدود 3 هفته است که نسبت به زمان تهیه کلون پایدار در سلولهای پستانداران بسیار کوتاهتر است.

    Glucose oxidase (β-D-glucose: oxygen 1-oxidoreductase) catalyzes the oxidation of β-D-glucose to gluconic acid، by utilizing molecular oxygen as an electron acceptor with simultaneous production of hydrogen peroxide. Glucose oxidase has been purified from a range of different fungal sources، mainly from the genus Aspergillus and Penicillium and has wide applications in chemical، pharmaceutical، food، biotechnology and other industries. In this study، a glucose oxidase (GOX) -encoding gene was isolated from Penicillium funiculosum by polymerase chain reaction (PCR) technique and then was cloned into a pUC19 plasmid. Consequently، molecular structure، biochemical and phylogenetic characteristics were analyzed. Nucleotide sequence of the gene، called PfGOX، revealed 1818 bp long، encoding a protein of 605 amino acid residues. The calculated molecular mass and the predicted isoelectric point of the deduced polypeptide were 65. 79 kDa and 4. 50، respectively. Analysis of secondary and three dimensional structures of the deduced polypeptide sequence for PfGOX was shown that PfGOX is similar to those of previously reported glucose oxidase proteins from other fungi. The deduced protein sequence was indicated a high similarity to glucose oxidases isolated from fungi such as Penicillium amagasakiense، Talaromyces flavus، Talaromyces stipitatus، and Talaromyces variabilis. Phylogenetic study was also demonstrated that PfGOX gene belongs to the subgroup IA of fungous glucose oxidases

    A psychrotrophic bacterium with amylolytic activity was isolated from soil samples collected from the southwestern suburb of Tehran. Using biochemical tests and a molecular approach based on 16S rDNA sequencing this bacterial isolate was identified as closely related with Exiguobacterium genus. This bacterium designated as Exiguobacterium sp. SH3 was able to grow and produce amylolytic enzymes at 4, 30, 37, and 48 °C. The best temperature for amylase production by this bacterium was 30 °C at which the starch (1%) added as inducer to the culture medium was totally digested during 9 h of culture. The supernatant of culture medium was used as a source of crude enzyme for amylase assay. The amylolytic activity was observed over a wide range of the tested temperatures from 4 to 45 °C and a wide range of the tested pH conditions from 5 to 9. Cold adapted enzymes with high catalytic activity seem to be more efficient than their mesophilic or thermophilic counterparts for applications at ambient temperature such as laundry and food processing.

    The knowledge-based force function is a new type of the scoring functions that has been used in the field of protein fold recognition with a noticeable success. In this study we compared the performance of a knowledge-based potential function and its corresponding force function in discrimination of the correct protein-protein complexes from the incorrect ones. The total force imposed by one component (receptor/ligand) upon another was used as a measure of complex stability. This force is expected to be the lowest in the native structure. To test the performance of each method, two decoy sets were used; one generated by soft body docking and the other by rigid body docking algorithms. The results of this comparison show that, for both decoy sets, the success rates in native and near native selections of the energy model are higher than that of the force model. It seems, the dependence of amount of the force on shape of the interface region introduces errors in the later model and therefore makes it inappropriate for the scoring of docked complexes.

    The purpose of this study is to evaluate of the choloroplast trnL-F marker’s power in genetic differentiation and phylogenetic relationships between Populus caspica and Populus alba. The 15 leaf samples collected from seven different growthing areas of hyrcanian forests, Iran; Guilan and Mazandaran provinces. Genomic DNA extracted from leaves using the CTAB and SDS-potassium acetatate combined method. PCR performed by universal primers and amplified trnL-F fragments sequenced. Results showed that, the entire length of the trnL-F region was 392-394 nucleotides in both species. In compaire to available sequences of the GenBank, Adenin and Thimine nucleotides in P. caspica and P. alba are more than other nucleotides. These two species have shown high similarity in nucleotide composition of trnL-F spacer region with minimum gegetic distance (=0.005). Parsimony analysis of 582 characters revealed 178 constant, 56 variable, 151 parsimony informative and 46 characters are unique positon. Too, bootstrap consensus trees showed that the trnL-F sequence data cannot distinguish P. caspica from P. alba because, investigated samples have located in common group. Therefore, for accurate recognition of this species, phylogenetic reconstruction using other choloroplastic and nuclear markers suggested.

    Secreted and transmembrane proteins are key players in various biological processes. they are accessible to various drug delivery mechanisms and they have a critical role as diagnostic and prognosis factors in clinical states. There are several bioinformatic softwares applied in the prediction of signal peptides and there are several experimental methods to trap signal peptides and identify secreted and transmembrane proteins. YST abbreviated form of yeast secretion trap is one of experimental methods based on pYST vectors and a host Saccharomyces cerevisiae strain 066-2. The host yeast is mutant and lacks invertase. The three pYST expression vectors encoding a mutant invertase in one of three frames and lacking the start codon and signal sequence. In this work we intended to set up YST system and for this purpose we examined two electronically predicted signal sequences in the amino terminal of cyclophilin c (ppic) and tRNA methyl transferase (Trmt1) human proteins. In addition we used putative signal sequence of pi16 as positive control and putative mitochondrial signal sequence of Tfam as the negative control to confirm functionality of the system. these sequences were amplified by PCR and then cloned to in frame in pYST expression vectors. Subsequently, The host yeast strain 066-2 recombinant were transformed with recombinant vectors and cultured on the selective agar media. The results of expriments confirmed the functionally of the system. Neither trmt1 nor ppic amino terminal predicted signal sequence did not function as signal sequence in this system.

    The industrial application of enzymes which can withstand in harsh condition such as high salinity and temperature has greatly increased. An extracellular halophilic and moderately thermophilic pullulanase was produced under stress conditions by a newly isolated extreme haloarchaeal strain designated as Ha25 in culture medium with 23% of total salt. 16S rRNA gene sequence analysis with phenotypic characterization places strain Ha25 in the genus Halorubrum. The enzyme production corresponded with growth and reached a maximum level during the mid-stationary phase. The isolate was capable of producing pullulanase in the presence of sodium chloride and the maximum enzyme production was at 3-4 M NaCl (85 U/ml). Optimum pH and temperature for enzyme production were 7.0 and 45 °C, respectively. Various carbon sources including maltose, starch and pullulan was inducing enzyme production, while glucose repressed production of enzyme. No cell growth and enzyme production was observed in the lack of Magnesium ions and the optimum concentration was 0.1 M. The maximum pullulanase activity was obtained in the medium containing 3 M NaCl at 50 °C, with 25% and 72% activity reductions at temperature 45 and 35°C, respectively. The optimum pH for activity of enzyme was 8.0, with 49.5% and 32.4% residual activities at pH 6 and 9, respectively.

    Microbial fuel cell (MFC) is an electrochemical device that can directly convert chemical energy of organic and inorganic materials to bioelectricity. The reduction of substances in anode compartment was catalyzed by the living cells in such as microorganisms or enzymes. In this study power generation was studied in mediator-less MFC. The polarity technique was used to characterize the electrical performance of the fabricated MFC. Glucose was used as electron donors and anaerobic effluent of wastewater treatment plant was selected as inoculums. The produced electrons are transported to anode surfaces and protons are moved through proton exchange membrane toward cathode. Nafion 117 was implemented to transfer of produced protons from anode to cathode chamber. The substrate consumption and cell growth was monitored for the incubation period. Maximum generated power and current densities were 186 mW.m-2 and 1078 mA.m-2, respectively. The generated power in the microbial fuel cell was monitored and power production was quite stable for the duration of 100 hours.

    As genetically modified organisms (GMOs) development is now increasing, detection and determination of their quantitative threshold using reliable and sensitive methods would be necessary. The aim of this study was to introduce reliable methods for qualitative and quantitative detection of Roundup-tolerant soybean particularly at event level via molecular approaches. For primary screening, we used polymerase chain reaction with several element–specific primers for detection of different percent of transgenic and non-transgenic Roundup ready soybean seed samples. Furthermore, semi-quantitative assays were performed using the same primers in combinations with soybean lectin-specific primers. The limit of detection (LOD) obtained for CTP-35S construct and event-specific (pDNA-35S) was 5%. This index for cp4 epsps and nos terminator sequences was 0.5% and for CaMV 35S promoter was 1%. Results showed that the sensitivity of analysis for nos terminator and cp4 epsps sequences was more than other elements and therefore, this method could be applied for rapid screening of Roundup Ready transgenic soybean samples.

    The bacterial alginate lyase is a periplasmic enzyme that is important in the production and disruption of alginate. Characterization of this enzyme such as molecular weight, pI, thermal stability, optimum pH and temperature, and effectiveness on bacterial alginate can differ in the strains of a bacterium. In this study, to obtain an alginate lyase containing specific and efficient properties in destroying of bacterial alginate, a mucoid strain of Pseudomonas aeruginosa isolated from patient‘s sputum was used. Since alginate lyase also has a role in alginate production, this strain was selected for this assay. Alginate lyase was extracted from periplasmic space by heat shock method and was purified by using suitable concentration of ammonium sulfate as well anionic exchange chromatography. Subsequently, enzyme activity was assayed by using thiobarbitoric acid method. The relative molecular weight of the enzyme was estimated about 60 kDa. The effect of reaction time and pH, substrate concentration and impact of temperature on alginate lyase activity was surveyed. Results showed that the optimum activity of the enzyme was achieved at 0.4 mg/ml substrate concentration after 20 min in 37˚C and pH 8.5. The enzymatic residual activity after 4 hours incubation in 80˚C was determined 31.33% that indicated to the spectacular thermal stability of the enzyme.This enzyme has the ability to disrupt the alginate of P. aeruginosa M 8821, and this ability associated with the thermal stability make it an excellent option for adjunctive therapy in the combat against antibiotic-resistant of P. aeruginosa infections.

    Canola (Brassica napus L.) is one of the most important oil producing plants in the world. In recent years, in spite of transformation in canola via Agrobacterium, the efficiency of this method is still low. To consider the effect of plant genotype and cultivation medium composition on transformation and regeneration in canola and the high toxicity of kanamycin for growth of this plant, it is necessary to develop more appropriate and efficient method for transformation of this oily plant. Here we used cotyledon explants and Agrobacterium tumefaciens strains LBA4404, GV3850, GV3101 harboring mutated gene EPSPS (pBI121+EPSPS) for transformation of canola PF7045-91 cultivar. After induction with agrobacterium, explants were placed in different cocultivation medium under dark condition at 25 degree centigrade. In regeneration process, to prevent necrosis of explants, the fixed amount of silver nitrate (5mg/L) was used for blocking ethylene production. The regeneration and selection procedure were started in medium containing silver nitrate without kanamycin and this carried on for one week. Subsequently, the kanamycin was added (5 mg/L) and its concentration was increased up to 15 mg/L in a ten days period. For root induction, the regenerated shoots were transferred to medium containing the MS salt with 15 g/L sucrose and 0.2 mg/L IBA. Transformation was confirmed by molecular analysis. Our data showed that gradually increasing kanamycin concentration and blocking ethylene production could improve the transformation of B. napus.

    Accumulations of biogenic magnetite (Fe3O4) in the brain and dysregulation of iron transport and storage have been found to be associated with many neurodegenerative disorders, such as Alzheimer disease. AD is a progressive neurodegenerative disorder that causes disruptions in memory and cognition. Several studies have shown that pathogenesis of AD is also related to protein misfolding and aggregation of tau protein in the brain. Microtubule-associated tau protein belongs to a family of intrinsically disordered proteins that promotes microtubule assembly and stability. In the present study, we investigated the interaction between recombinant human tau and magnetic iron nanoparticles by using ThT fuorescence and transmission electron microscopy. Our results showed that magnetic iron nanoparticles induced tau conformational changes from random-coil to β-sheet and consequently resulted in the formation of tau fibrils. In conclusion, our results proposes a probable mechanism for the correlation between aggregation of tau protein and biogenic magnetite crystals in AD.

    Our data point to optimized experimental conditions for high expression of heterologous proteins by Drososphila melanogaster S2 cells. Following construction of pMT-EGFP and transfection of S2 cells with calcium phosphate or lipofection methods, the EGFP expression analysis showed that the transfection efficiency with calcium phosphate method with pH=7.06 and 10 μg/ml of DNA is more than that of lipofection method. The electrophoresis pattern of the RT-PCR products of S2 driven hFIX indicated that the Drosophila metallothionein promoter is tightly regulated and capable of driving high-level heterologous transcription upon induction by CuSO4. Nevertheless, the two methods were designed for construction of a recombinant stable S2 cell. In both methods, with and without Mitomycin, stable clones were recovered after only 3 weeks of hygromycin selection that is shorter than selection time in the mammalian cells. Therefore, our results show that Drosophila S2 cells do not have the limitations mentioned for mammalian cells.
