Molecular detection and phylogenetic position of B strain bean yellow mosaic virus from broad bean fields in Golestan province

In recent years, the viral symptoms (mosaic and yellowing) have been observed in a wide range of broad bean fields in Golestan province in Iran.Viruses of Potyvirus genus are one of the most important viral pathogens in plants. In order to detect these viruses, plants with symptoms of mosaic, leaf deformation and rolling were collected during the growing season in 2013 from legume fields of Golestan Province. Total RNA of the samples was extracted, to confirm the infection. Then, samples were tested by reverse transcription polymerase chain reaction (RT-PCR) using potyvirus universal primers (Oligo1n/Oligo2n). Twelve out of 35 samples showed infection. Some of the infected samples were sequenced. Sequencing results revealed that samples are infected by Bean yellow mosaic virus (BYMV). The results showed that all BWMV sequences can be placed into 4 clades. Golestan isolate was grouped with isolates from Japan and Australia. Determination of the taxonomic position of CP gene in Iranian strain of BYMV and comparison with other strains of worldwide, identified origin, evolution and emergence of the strains in Iran and seems having foreign origin and maximum molecular and host variation occur in Eastern Asia. For result of this study, the broad bean with symptoms of mosaic, leaf deformation were Maximum infection of BYMV in other hosts.
