Comparison of DNA Extraction Methods and Development of Simplex PCR to detect residual genomic DNA in Tomato paste

Abstract:
Based on today biotechnology and molecular biology studies for investigation of organism presence in food products through evaluation of DNA markers by PCR method, the quality of extracted DNA for analysis of DNA markers is and important criteria and development of a DNA extraction method with high quality is ideal. In variant DNA extraction methods, three parameters are in high demand, quality, speed and quantity of isolated DNA. In current study, six variant DNA extraction methods were developed to analyze food product samples to investigate presence of GMO (Genetic Modified Organism) and food adulteration. The isolated DNA with high quality was analyzed by PCR technique through using specific primers of mitochondrial D-loop marker as a reaction template.The results showed that between six developed methods, the Wizard method which was based on the specific binding of DNA molecule to silica membrane (Column) was selected and amplification of D-loop DNA marker with band size of 280bp was successfully developed. Analysis of variant genomic DNA extraction methods and development of simplex PCR for specific amplification of tomato-loop DNA marker revealed that through using molecular biology method we can extract and detect specifically presence of low or residual amount of wild , GMO or adulteration tomato food with high sensitivity and quality.
