Antioxidant effect of extract of the grape seed in streptozotocin induced diabetic rats
Diabetes mellitus is a metabolic disorder as old as mankind and its incidence is considered to be high all over the world. Oxidative stress is strongly associated with development and the complications of diabetes. Antioxidant agents, especially with the origin of plants, are of more importance in the treatment of diabetic complications. The main objective of the present study was to evaluate the effect of extract of the grape seed on antioxidants status in streptozotocin induced diabetic rats.
In this laboratory experimental study which conducted in Islamic Azad University of Tabriz research center. Forty male Wistar rats were randomly assigned into four equal groups including healthy control group, healthy group treated with grape seed extract (40 mg/kg), diabetic control group and diabetic group treated with grape seed extract (40 mg/kg). The experimental rats were treated in related groups for 12 weeks and at the end of experiment serum level of malonaldehyde (MDA) and anti-oxidant enzymes activity including superoxide dismutase (SOD), glutathione peroxidase (GPx) and catalase (CAT) were measured in red blood cells. Statistically, comparison of the groups was carried out using one-way analysis of variance followed by Bonferroni post-hoc test. Differences were considered statistically significant at p< 0.05.
Diabetic rats showed significant increase in the value of MDA and a decrease in the activities of SOD, GPx and CAT of red blood cells (p<0.001). Oral administration of grape seed extract resulted in significant reduction in the level of MDA and significant increase in the activities of SOD, GPx and CAT of red blood cells (p<0.001).
The results of this study provide confirmatory evidence of oxidative stress in diabetes and show the anti-oxidative effect of grape seed extract. [ZJRMS, 12(1):9-14]
