Study of genetic diversity of Dashtiari and Khazak breeds using microsatellite markers
Purpose of this study was determination of genetic diversity and genetic distance within and between two types of native chickens of Sistan and Baluchestan province, using five microsatellite markers. Extracted genomic DNA from feather samples were amplificated by the microsatellite primers and then their product electrophoresed using 8% polyacrylamide gel. In Khazak chickens were detected 26 alleles and in Dashtiari chicken 28 alleles with average of 5.4 and 6 alleles per locus, respectively. The mean of Shannon index were 1.627 and 1.587 for Dashtiari and Khazak, respectively. ADL225 and MCW5 loci had shown the highest Shannon index. The mean of PIC in the two types of native chickens was 0.656. The highest PIC values were observed in ADL225 in Khazak with 0.80 and in Dashtiari with 0.81. Results were indicated that Dashtiari and Khazak chicken might be in one genetic group.
