mRNA extraction from venom glands of Iranian scorpion Odonthobuthus doriae,cDNA synthesis and perfom PCR from OD1 by means of degenerative primers

In most area in Iran، scorpion sting is a serious problem. most of the scorpion stings are due to Buthidae family. Scorpion toxin consists of different biologic active components such as different types of toxins which are encoded by individual gene. There is no genetic information about sequence of genes coding toxins of Buthidae scorpion family in national center for biotechnology information (NCBI). The aim of this study was to amplify of cDNA encoding a neurotoxin from venom gland of Buthidae family scorpions. Samples were provided and identified by Razi reference scorpion laboratory of karaj. Using RNA extraction kit، the total RNA was extracted from the venom glands of specie of Odonthobuthus doriae. In order to amplify cDNA encoding a neurotoxin، the proper primers have been designed. Using RT-PCR technique and after optimizing PCR conditions، a 264 bp cDNA fragment from O. doriae was amplified. Decisive confirmation of the amplified fragment will be possible just by its sequence determination، and after that، determination of the type of the encoded neurotoxin will be easy.
