Assessment of dose and time dependant DNA damage induced by ultraviolet -B and C radiation on egg and larvae of rainbow trout (Oncorhynchus mykiss) by alkaline Comet assay

Destructive effects of increased ultraviolet irradiation of Earth’s surface on different animals has particular importance. Nucleic acid content of cells is one of the most important compounds that target of ultraviolet irradiation and deleterious effects of DNA damage cause many physiological disorders such as sperm destruction, decreased fertilization, embryo’s abnormality and finally decreased the fish viability. In this study, destructive dose and time dependant DNA damages of ultraviolet irradiation on early embryonic stages of rainbow trout (Oncorhynchus mykiss) investigated. So, eyed egg and yolk sac larvae of rainbow trout exposed to type B and C of ultraviolet irradiation intensities of 10, 40, 84, 153 and 15, 55, 90, 196 µW/cm2 respectively, in duration of 30 and 60 minutes and amount of DNA damages analyzed by alkaline comet assay. Results indicated, although, increased intensity of ultraviolet irradiation induced DNA damages in both embryo and larvae of rainbow trout, but destructive effects has been more increased in larvae stage, because of protective chorionic layer of embryo (p<0.05). In addition, increasing the duration of exposure from 30 to 60 minutes has not increased the genetic damages, significantly. In conclusion, results of this study while confirmed the applicability of alkaline comet assay for assessment of DNA damages of fish embryonic and larvae stages, indicated the more dose dependant than time dependant destructive effect of ultraviolet irradiations and more ultraviolet sensitivity of larvae than embryo. This finding should be considered in using these irradiations for fish breeding programs.
