Investigation of effect of different concentrations2,4- D, Kin on callus induction in potato (Solanum tuberosum L.) Agria Variety

Plant tissue culture techniques are a useful tool to increase the genetic diversity and crop production. The purpose of this study to evaluate the different 2,4- D and Kinetin hormones on callus induction in Agria potato variety. The experiment was set up based on factorial experiment and completely randomized design (CRD) with 4 replications. The effects of growth regulators 2,4- D (0, 1, 2, 4 mg/l), Kin (0.25 mg/l), leaf explants two internodals on callus induction were examined. Analysis of variance showed that the two types of leaf and internode explants of traits, diameter and fresh weight of callus induction there is a significant difference. Also of hormone levels in explants showed significant differences in all traits. The results showed that the highest percentage of callus induction (75%) in leaf explants at a concentration of 2 mg/l (2,4- D) and the internodals explants (87/5%) at a concentration of 4 mg/l (2,4- D), respectively. Diameter of callus, fresh weight were, (5/60 mm, 0.55 gr) respectively and the intermodal explants were (9.13 mm, 0.92 gr) respectively at a concentration of 2 mg/l (2,4- D).
