The Use of Nodal Segments Explants and Different Combinations of Plant Growth Regulators for Cannabis Sativa L. Regeneration

Cannabis sativa L. is a dicotyledon plant belonging to cannabinaceae family which produces a diverse array of secondary metabolites. In this study, nodal segments explants were cultured on MS media containing different concentrations of hormones including: BA (0.1, 0.2, 0.5, 1, 2, & 3 mg/l; alone or combined with 0.5 mg/l IBA), TDZ (0.1, 0.2, 0.5, 1, 2, & 3 mg/l; alone or combined with 0.5 mg/l of IBA), NAA (0.5, 1, 2, & 3 mg/l; alone or combined with 0.5 mg/l of BA), and 2,4-D (0.1, 0.2, 0.5 & 1 mg/l; alone or combined with 0.5 mg/l of BA). The results showed that the concentrations of 0.2 mg/l TDZ and 0.5 mg/l IBA were the best treatments for callus induction. The maximum length of produced branch belonged to the concentration of 0.1 mg/l TDZ and 0.5 mg/l IBA. For root induction of regenerated shoots, two growth regulators (IBA and NAA) have been used separately in 0.1, 0.2, 0.5, and 1 mg/l concentrations. The greatest root induction belonged to the concentration of 0.1 mg/l IBA and the maximum length of root was observed in the concentration of 0.5 mg/l NAA.
