The Study of Saponin Profile in 3 Cultivars of Alfalfa (Medicago sativa L.) Expressing Different Levels of Resistance to Alfalfa Weevil (Hypera postica Gyll.) Using TLC

Abstract:
Triterpenoid saponins from alfalfa are secondary metabolites which cause plant bittering, decreasing palatability, and increasing the resistance to pests and diseases. Saponins such as glycoside form part of a group of steroid. In present work, the variability of saponins in different cultivars of alfalfa which had presented different levels of resistance towards alfalfa weevil in alfalfa field of germplasm, was studied and analyzed using thin layer chromatography (TLC). Leaf saponins were extracted in methanol solvent from cultivars Tabriz8 as resistant, Deiopoy as susceptible and Kaliverd321588 as semi resistant cultivars before onset of budding stage. The analysis of saponins on TLC plates showed that there are qualitative differences between gel banding patterns. The number of saponins bands in resistant cultivar of alfalfa (Tabriz 8) was 11, in semi resistant (Kaliverd321588) was 8 and in susceptible cultivar (Deiupoy) was observed to be produced 6 bands. In present work, from the results, As a result, it is concluded that, the findings of the present study more numbers of band spots formed at the origin of sample running on the gel, confirms more resistance to the pest in related cultivar.
