Heterologous expression of linalool synthase in Saccharomyces cerevisiae using gap repair technique

Abstract:
Monoterpenoids are a diverse group of secondary metabolites usually found in plant tissue. These compounds are widely used as aromatic additives in cosmetics and food industry. Saccharomyces cerevisiae naturally contains the necessary precursors for the synthesis of terpenoids and therefore can be used as a cell factory for heterologous production of these compounds. In recent years, many genes that encode plant monoterpene synthases (MTS), have been characterized. In this study, in order to produce linalool in yeast Saccharomyces cerevisiae, the sequence of gene encoding linalool synthase from lavender was cloned in the pESC-URA under the control of GAL1 promoter, by using gap repair technique. The resultant yeast strain was named YLIS. The production of linalool in this strain was confirmed by gas choromatography (GC). Using standard curve, the concentration of linalool in YLIS strain was estimated 32 μg/L.
