Genetic diversity of zagros zebrafish (Aphanius vladykovi) in Madar-o-dokhtar spring and Chehelgazi spring in the Chaharmahall-o-Bakhtiari Province by using microsatellite markers

This study was aimed to investigate the genetic diversity of zagros Mosquito fish (Aphanius vladykovi) which is a small plagic fish from cyprinodontidae family and an endemic fish of Chaharmahall-o-Bakhtiari Province, by using microsatellite markers. For this purpose 46 fishes were collected in fall 2011 from Chehelgazi and Madar-o-dokhtar spring. Six polymorphic microsatellite loci were used in this study. Number of observed alleles in populations with an average was 11.41 and the observed heterozygosity in the Chehelgazi and Madar-o-dokhtar spring was 0.993 and 0.957, respectively that indicated Aphanius long-term reproductive is a proof on reproductive success and increasing heterozygosity. The result of Fst (0.023) and Rst (0.092) indicated low genetic differentiation among populations. Analysis of Molecular Variance indicated only 2 percent genetic diversity between populations and genetic distance between populations was 0.322. Gene flow between two regions was equal to 19.097 that this high amount of gene flow may be affected by the statistical calculations. In the study of Hardy-Weinberg equilibrium, after Bonferroni correction factor, among the 12 tests of population-loci, 3 tests were in equilibrium. The present study indicated preliminary reasons and results for the existence of distinct populations of zagros Mosquito fish so attention and strict measures to protect of its genetic resources is essential.
