The study of d-loop mitochondrial region with the objective of investigating the genetic and phylogenetic diversity in wild sheep and comparing it with Kermani sheep

genetic purity of the sheep has been change due to non-controlled Mating. Mitochondrial geneome sequencing is a suitable method for studying genetic purity. The purpose of this study is to analyse the genetic and phylogenetic diversity in nucleotides sequence in the d-loop region in the mitochondrial genome of rams, ewes, and domestic sheep of Kermani in order to determine their genetic distance and phylogenetic relationship. In order to do this, 15 blood samples of Kermani sheep in the research station of Shahid Bahonar University of Kerman, and 12 blood samples of wild sheep in the regions of Kerman (2 head), Shiraz (5 head), and Tehran (5 head), Alborz mountain is located north of Iran, were collected. After the DNA extraction, the amplification of the 749 component from the d-loop region in mitochondria were done by using polymerase chain reaction with a gene specific primer pair. Sequencing this component was performed by the Korean Bioneer company because it was needed to compare the sequence of this study with the sequences of other researches in order to determine the genetic distance. The results indicate that there is a relatively great haplogroup difference among the nucleotides sequences of the studied samples. The haplogroup investigation showed that the mass of Kermani local sheep in haplogroup B is 75 percent of the wild species and only 25 percent of the wild species in haplogroup A. Moreover, the phylogenetic results, which were generated by using the neighbor-joining method and illustrated that the studied wild rams and ewes with kermani breed sheep are located in the family of ovis urial.
