Evaluation of effect of Lactobacillusplantarum subspecies plantarum PTCC 1896as a local probiotic on lifespan by the modelnematodeCaenorhabditis elegansin vivo

According to Metchnikoff observations, the first effect of consumption of probiotics in milk-based fermented products, was long lifespan which needs the intestinal probiotic colonization. The aim of this study was investigation of the effect of Lactobacillusplantarum subspecies plantarumPTCC 1896as a local probiotic compared to Escherichiacoli OP50 as a standard food source on lifespan and their colonization ability in Caenorhabditis elegans intestine in vivo. In this research, lactobacillusrhamnosus GG as a commercial probiotic was used as positive control. To determine the optimum dosage of used probiotics, the concentrations of 0.01, 0.1, 1 and 10 mg bacterial cell/plate were used. Results showed that 0.1 mg bacterial cell/plate of Lactobacillusplantarum subspecies plantarumPTCC 1896 increased the lifespan of C. elegans by 36.95%, compared to Escherichiacoli OP50. Also colonization test of Lactobacillusplantarum subspecies plantarum PTCC 1896 showed a good adhesion ability of the bacterial cells to intestinal epithelial cells of the nematode, in comparison to lactobacillusrhamnosus GG. So it could be concluded that consumption of Lactobacillusplantarumsubspecies plantarum PTCC 1896 as a local probiotic with a good intestinal colonization capability provides the production features of functional foods containing the probiotic species with the purpose of increasing the host’s lifespan.
