Evaluation of Genetic Variation in some Canola Cultivars Using ISSR Markers

Evaluation of genetic variation and inter/intra relationships among populations, species and different individuals is a basic purpose of plant breeding studies. To this object molecular markers have been widely used. In this study to investigate the genetic diversity of twenty cultivars of Canola, nine ISSR primers were used. Among all, eight ISSR primers produced highest polymorphisms. The average polymorphic band per primer/cultivar was estimated 8.77 and 3.95, respectively. The highest Polymorphic Information Content (PIC) calculated for MM6 primer (0.78), too. The tree diagram from Cluster analysis was drawn using NTSYS software, UPGMA method and Simple similarity matrix. The Cultivars divided into 6 separate clusters, accordingly. Furthermore, a Two-dimensional plot of the Principal Component Analysis (PCA) were drawn and compared with cluster analysis. Cultivars grouping based on two methods had a great similarity. The most genetic similarity observed between Elite and Rinbow cultivars. Genetic variation between Canola cultivars did not relate with their geographic pattern.
