Investigating the effect of combination of different levels of 2,4-D and BAP on a medium culture of 1/4 MS on some vegetative characteristics of nodal segment of Dog rose
The present study was carried out in order to obtain a suitable micro propagation method of Rosa canina L. as the most important rootstock for roses. The experiment was a factorial based on a completely randomized design with two concentrations of BAP (0, 0.75, 1.5, 2.5 and 3 mg/l) and 2,4-D concentrations (0, 1.5, 2.5, 4 and 5.5 mg/l) were cultured in MS medium (one quarter) in four replicates (each replicate included two samples). Samples were prepared in the middle of the season in the middle part of the middle shoots of the shrubs located in the fruit stage (red), pieces of 1.5 cm after disinfection With 70% ethanol for one minute, 10% sodium hypochlorite for 15 minutes was cultured horizontally and purification under sterile conditions. The room was then kept under a temperature of 23°C to 25°C and a photoperiod of 16 hours of light and eight hours of darkness. After the three months and without any replanting, the experiment was stopped. Results indicated that both the 2,4-D and BAP factors, as well as their interactions, had a very significant effect on all the test characters (height, weights and shoot length, number of leaves, length of stem and number of new bud). The treatment has 1 mg/l 2,4-D plus 2.25 mg/l BAP, is the best treatment of this combination is to increase seedling height, stem length, weigh weight, and shoot weight. The treatment was 2.5 mg/l 2,4-D with concentrations of 0.75 mg/l BAP, the best treatment for increasing height, weight, and the treatment with 2.5 mg/l 2,4-D with concentrations of 1.5 mg/l BAP is the best treatment for increasing weight, leaf number and new bud.
