    ویروس رگبرگ زرد نکروتیک چغندرقند (Beet necrotic yellow vein virus، BNYVV) در چغندرقند بیماری مهم و خطرناکی بنام ریشه ریشی (root beardiness) یا ریشه گنائی (rhizomania) ایجاد می کند و دارای گسترش زیادی در اکثر نقاط دنیا است. این ویروس دارای چند قطعه آر ان ای ژنومی می باشد و آران ای شماره 3 آن یک پروتئین 25 کیلودالتونی که مسئول بیماری زایی در گیاه چغندرقند است را رمز گذاری می کند. موقعیت های آمینواسیدی 67 و 68 این پروتئین نقش مهمی در شکستن مقاومت ارقام مقاوم دارند. در این مطالعه p25 در سه جمعیت BNYVV از مغان، لرستان و فارس در ارقام حساس و مقاوم چغندرقند مورد بررسی قرار گرفت. دو جمعیت لرستان و فارس در چهار گذرش پیاپی به ارقام مقاوم و حساس مایه زنی شدند و ژن p25 ویروس در آنها بررسی شد. بر اساس نتایج حاصله انتقال پیاپی ویروس در گیاهان مقاوم منجر به تغییراتی در موقعیت های آمینواسیدی 30، 49، 67، 68، 129، 163 و 198 در p25 شد، در حالی که در گیاهان حساس تغییرات آمینواسیدی دیده نشد. بیشتر تغییرات تتراد (آمینواسیدهای 70-67) در موقعیت آمینواسیدی 67 مشاهده گردید. وجود بیوتیپ P در مغان و لرستان تشخیص داده شد. از آنجا که انتخاب تحت فشار ارقام مقاوم می تواند نقش مهمی در ظهور بیوتیپ P ایفا کند، بنابراین حضور بیوتیپ P در دو منطقه در ایران ممکن است به علت کشت ارقام مقاوم بوده باشد. برخی پارامترهای تکاملی و موتیف های p25 مورد بحث قرار گرفته است.

    قارچ های متعلق به جنس Trichodermaاز مشهورترین و موثرترین عوامل کنترل بیولوژیک بیماری های گیاهی هستند. به دلیل اهمیت سویه های تریکودرما در کشاورزی و کمبود اطلاعات در زمینه اکولوژی و دینامیک جمعیت آن ها در زیستگاه های طبیعی کشور، در این تحقیق، تغییرات جمعیت جدایه های تریکودرما در شالیزار و ارتباط آن با عوامل زنده (رقابت میکروبی) و غیرزنده (رطوبت، pH، EC، میزان ماده آلی خاک و نیز میزان بارش و درجه حرارت) مورد مطالعه قرار گرفت. نمونه برداری به صورت ماهانه به مدت یک سال، از عمق 15 سانتی متری خاک سه مزرعه برنج در مناطق مختلف شهرستان آمل انجام شد. جهت جداسازی میکروارگانیسم ها از روش کشت سری رقت روی محیط کشت های انتخابی و عمومی استفاده شد. جمعیت تریکودرما،قارچ و باکتری کل به ترتیب 103 × 1/1- 101 × 1/1، 104 × 1/3-103 × 7/1 و 105 × 5/6 -104 × 2/2 واحد تشکیل دهنده پرگنه در هر گرم خاک شالیزار (cfu/g) محاسبه شد. نقطه اوج اول جمعیت تریکودرما در اواخر فصل بهار و نقطه اوج دوم در فصل پاییز بود و در زمستان این جمعیت کاهش یافت. آنالیز رگرسیونی چند متغیره نشان داد که جمعیت تریکودرما در سطح احتمال 1% با رطوبت و هدایت الکتریکی خاک رابطه معنی دار مثبت و با جمعیت کل قارچ ها رابطه معنی دار منفی دارد. با سایر فاکتورها رابطه معنی داری مشاهده نشد.

    در طبیعت گیاهان در معرض طیف وسیعی از میکروب ها که باعث بروز بیماری ها و یا بهبود رشد و کمک به گیاه جهت غلبه بر تنش های زنده و غیر زنده می شوند قرار می گیرند. مهم ترین میکروب های مفید، گونه های تریکودرما و سودوموناس های فلورسنت می باشند. در این تحقیق اثر سویه های Trichoderma harzianum Tr6 و Pseudomonas sp. Ps14 که از فراریشه خیار جداسازی شده و قادر به القاء مقاومت در این گیاه بودند، بر سیستم ایمنی خیار با ارزیابی کمی تغییرات بیان ژن های دفاعی مورد مطالعه قرار گرفت. نتایج نشان داد که بیان ژن های دفاعی بتا-1 و 3-گلوکاناز و کیتیناز به طور گذرا بعد از مایه زنی ریشه ها با این میکروب های مفید افزایش می یابند. افزایش بیان گذرای ژن های PR1 و LOX1 نیز پس از مایه زنی با Tr6 و ترکیب آن با Ps14 دیده شد. در حالی که مایه زنی با Ps14 فقط افزایش بیان LOX1 را در پی داشت. این یافته ها دلالت بر این دارد که در ابتدا سیستم دفاعی گیاه در مقابل این میکروب های مفید فعال شده و سپس این میکروب ها با سرکوب سیستم دفاعی گیاه ارتباط همیاری با گیاه برقرار می کنند.

    جهت بررسی تاکسونومیکی گونه Valsa sordida، عامل بیماری شانکر درختان، تعداد 125 نمونه گیاهی آلوده در طی سال 1387 از استان گلستان جمع آوری گردیدند. بر اساس ویژگی های ریخت شناختی، از بین 49 جدایه قارچی به دست آمده، 35 جدایه به گونه V. sordida تعلق داشتند. تعداد 12 جدایه از لحاظ ویژگی های ریخت شناختی مرحله غیرجنسی تا حدودی به گونه C. chrysosperma شباهت داشتند و Cytospora sp.1 نامیده شدند. یک جدایه (از Populus deltoides) نیز به لحاظ ویژگی های ریخت شناختی تا حدودی به گونه V. sordida شباهت داشته و Valsa sp.1 نامیده شد. یک جدایه دیگر (از Ficus elastica) نیز به لحاظ ویژگی های ریخت شناختی از گونه C. chrysosperma متمایز و به گونه Cytospora ribis نزدیک بود و Cytospora sp.2 به آن اطلاق گردید. به منظور ارزیابی فیلوژنی جدایه های منتخب، نواحی ITS1-5.8S-ITS2 از rDNA هسته ای در 14 جدایه (پنج جدایه متعلق به گونه C. chrysosperma، هفت جدایه متعلق به Cytospora sp.1، یک جدایه متعلق به Valsa sp.1 و یک جدایه متعلق به Cytospora sp.2)، تعیین توالی گردیدند و به همراه 32 توالی مربوطه از بانک ژن مورد ارزیابی قرار گرفتند. نتایج حاصل از بررسی مولکولی و گروه بندی جدایه های قارچی منتخب، با تنوع مشاهده شده در صفات ریخت شناختی منطبق بود.

    نماتود ریشه گرهی از مهم ترین نماتودهای انگل گیاهی با دامنه وسیع میزبانی است. با توجه به خطرات سموم، استفاده از روش های غیرشیمیایی، از جمله مهار زیستی در مبارزه با آفات و بیماری های گیاهی ضروری به نظر می رسد. در این پژوهش، اثر دو عامل کنترل زیستی Metarhizium anisopliae و Pseudomonas fluorescens CHA0 بر نماتود Meloidogyne incognita به طور جداگانه و به طور توام، در چند آزمایش مطالعه گردید. در آزمون غیرفرار تعیین اثر بازدارندگی باکتری بر قارچ، مشخص شد که باکتری بر قارچ اثر بازدارنده نداشت. در بررسی های آزمایشگاهی اثر سوسپانسیون و عصاره کشت این عوامل، بیشترین اثر کشندگی بر لارو سن دوم در تیمار مخلوط عصاره کشت هر دو عامل با قریب 95% و در درجه دوم، در تیمار عصاره کشت باکتری با حدود 80% مرگ و میر، مشاهده گردید. بیشترین کاهش تفریخ تخم (98-93 درصد) نیز در تمامی تیمارهایی که عصاره کشت یک یا هر دو عامل موجود بودند، مشاهده شد. در مقایسه اثر روش های کاربرد این عوامل، در تیمارهایی که خیساندن خاک با سوسپانسیون قارچ صورت گرفت، شاخص های رشدی گیاه به میزان قابل توجهی بهبود یافتند و شاخص های نماتود نیز به مقدار قابل توجه کاهش داشتند، به طوری که در آزمایش اثر تلفیقی عوامل کنترل زیستی در شرایط گلخانه، در تیمار افزودن فرم تجاری قارچ به خاک و خیساندن خاک با سوسپ‍انسیون اسپور، شاخص های رشدی گیاه در بهترین وضعیت قرار داشتند و همین تیمار نیز به میزان قابل توجهی موجب کاهش شاخص های نماتود شد، به طوری که تعداد گال در گیاه تا 30% و فاکتور تولیدمثل تا حدود 75% کاهش یافت. این مقاله اولین گزارش از اثر نماتودکشی قارچ M. anisopliae بر نماتود ریشه گرهی M. incognita می باشد.

    اعضای جنس Septoria رایج ترین گونه های قارچی عامل بیماری لکه برگی درختان صنوبر و یکی از خسارت زاترین بیمارگرهای اندام های هوایی این درختان می باشند. با وجود شیوع این بیماری در منطقه شمالغرب ایران، هویت گونه های Septoria دخیل در این بیماری ناشناخته باقی مانده اند. هدف از انجام این تحقیق شناسایی گونه های Septoria عامل بیماری لکه برگی درختان تبریزی در استان آذربایجان شرقی و بخش هایی از استان های آذربایجان غربی و اردبیل بر اساس صفات ریخت شناختی و داده های مولکولی بود. برای این منظور، در طول تابستان 1392 از برگ های آلوده گونه های جنس Populus که عمدتا گونه P. nigraیا شالک بودند، نمونه برداری صورت گرفت و بعد از جداسازی و خالص سازی جدایه های متعلق به جنس Septoria، شناسایی جدایه ها بر اساس ویژگی های ریخت شناختی و آغازگرهای اختصاصی معرفی شده برای دو گونه S. populi و S. populicola انجام گرفت. نتایج مطالعات ریخت شناختی نشان داد که گونه S. populi تنها گونه دخیل در بیماری لکه برگی سپتوریایی روی گونه های جنس Populusدر مناطق نمونه برداری شده می باشد. در آزمون PCR به وسیله آغازگرهای اختصاصی گونه S. populi قطعه ای به طول 439 جفت باز از تمامی جدایه های Septoria تکثیر شد، در حالی که قطعه اختصاصی (به طول 329 جفت باز) S. populicola با استفاده از آغازگرهای اختصاصی این گونه در هیچ یک از جدایه ها تکثیرنشد. بنابراین، گونه S. populi به عنوان تنها عامل بیماری لکه برگی سپتوریایی درختان جنسPopulus در مناطق مورد مطالعه شناسایی شد. آزمون بیماری زایی جدایه های منتخب با استفاده از روش برگ بریده روی گونه P.nigraانجام گرفت و نتایج بررسی نشان داد که همه جدایه های بدست آمده بر روی رقم نیگرا بیماری زا بودند.

    ویروس موزائیک معمولی لوبیا (Bean common mosaic virus، BCMV)، ویروس موزائیک معمولی بافت مرده ی لوبیا (Bean common mosaic necrosis virus، BCMNV) و ویروس موزائیک خیار (Cucumber mosaic virus، CMV) از ویروس های بذربرد لوبیا می باشند که به طور جدی باعث افت کمی و کیفی این محصول می شوند. بدلیل وجود گزارش های متعدد از آلودگی مخلوط لوبیا با این سه ویروس، معرفی روشی دقیق، سریع و حساس برای ردیابی همزمان CMV، BCMV و BCMNV در گیاه لوبیا می تواند نقش مهمی را در جلوگیری از انتشار ویروس و گسترش خسارت آنها و نیز غربال گیاهان سالم داشته باشد. در پژوهش حاضر برای ردیابی همزمان BCMV، BCMNV، CMV و نیز ژن 18S rRNA بعنوان کنترل داخلی، روش Multiplex RT-PCR استفاده شد. برای این منظور ابتدا لوبیای آلوده به این سه ویروس توسط DAC-ELISA ردیابی شد. آران آکل استخراج و سپس cDNA با استفاده از آغازگرهای تصادفی شش نوکلئوتیدی ساخته شد. واکنش پی سی آر با استفاده از آغازگرهای اختصاصی برای هر ویروس و کنترل داخلی بصورت جداگانه و یا در ترکیب با یکدیگر بطور موفقیت آمیز قادر به تکثیر قطعات در حدود 470 جفت بازی برای BCMV، حدود 500 جفت بازی برای کنترل داخلی، حدود 704 جفت بازی برای BCMNV و حدود 870 جفت بازی برای CMV گردیدند. این روش قابلیت ردیابی این سه ویروس را در 100 نانوگرم از آر ان ای کل دارد.

    Phytophthora drechsleri که قبلا در بسیاری از کشورها از جمله ایران به عنوان عامل بوته میری کدوییان گزارش شده بود، از لحاظ ریخت شناسی و نیاز دمایی با P. melonis، عامل دیگر بوته میری کدوییان، تفاوتی ندارد. هدف اصلی این پژوهش بررسی بیماری زایی دو گونه ی P. drechsleri و P. melonis نسبت به ارقام مختلف جالیز در شرایط گل خانه بود. 87 رقم مختلف کدوییان شامل: خیار، طالبی، خربزه، کدو و هندوانه از مناطق مختلف مورد بررسی قرار گرفت. گیاهان دو ماهه، با مایه چهار هفته ای دو گونه ی فوق که بر روی ورمیکولایت و عصاره دانه شاهدانه رشد یافته، مایه زنی شدند و تا دو ماه پس از مایه زنی مورد بررسی قرار گرفتند. بررسی نتایج نشان داد که هیچ یک از ارقام جالیزی مایه زنی شده با P. drechsleri بیمار نشدند و تفاوتی با شاهد نداشتند. اما اغلب کدوییان مایه زنی شده با P. melonis نشانه های آلودگی را نشان دادند. بنابراین P. melonisبه عنوان یکی از عوامل بوته میری در کدوییان ایران معرفی می گردد. نتایج بررسی واکنش این ارقام به P. melonis نشان داد که، ارقام طالبی حساس تر از سایر ارقام جالیزی هستند. ارقام خربزه و خیار هم حساسیت نسبی به P. melonis دارند و ارقام کدو و هندوانه نیز مقاومت بیشتری نسبت به سایر ارقام به P. melonis نشان می دهند.

    سفیدک پودری از بیماری های مهم گوجه فرنگی در مزارع و گلخانه های دنیا محسوب می شود و در ایران گونه Leveillula tauricaبه عنوان تنها عامل این بیماری گزارش شده است. طی مطالعه قارچ های تیره Erysiphaceae در استان اردبیل، برگ های گوجه فرنگی با علایم سفیدک پودری از گلخانه ای در شهر اردبیل جمع آوری و جدایه ها بر اساس ویژگی های ریخت شناختی مورد شناسایی اولیه قرار گرفتند. سپس DNA ژنومی قارچ به روش Chelex 5% استخراج گردید و ناحیه ITS از rDNA با آغازگرهای ITS1 و ITS4 تکثیر و توالی یابی شد. بر اساس مشخصات ریخت شناسی و توالی یابی ناحیه ITS-rDNA، این قارچ به عنوان گونهPseudoidium neolycopersici (Syn: Oidium neolycopersici) شناسایی شد. بدین ترتیب، این گونه برای اولین بار به عنوان عامل سفیدک پودری گوجه فرنگی (Lycopersicon esculentum) در ایران معرفی می شود. گونه P. neolycopersici یکی از مهم ترین عوامل مولد سفیدک سطحی گوجه فرنگی در سراسر دنیا است و پیدایش آن در ایران تهدید جدیدی برای کشت گوجه فرنگی در کشور می باشد.

    کشت انواع صیفی و سبزی از جمله انواع کلم در مزارع سبزی اطراف تبریز، شمال غرب ایران (با مشخصات جغرافیایی38 06 N، 46 ̊15 ́E،، دمای 20-6/6 درجه سانتی گراد و خاک ماسه ای لومی) قدمت طولانی دارد. نماتد سیستی کلم، Heterodera cruciferae Franklin، 1945، به عنوان یکی از عوامل بیمارگر گیاهی روی کلم قمری (Brassica oleracea L. var. gongylodes) و کلم برگ (Brassica oleracea L. var. captita alba) در این منطقه شناسایی و گزارش شده است. علاوه بر دو گیاه فوق brussels sprouts، کلم گل، کلم بروکلی، هویج، Alyssum maritimum، Crambe maritime، Cheiranthus allionii، C. cheiri،Iberis umbellate، Lepidium sativum،Raphanus sativus، Sinapis alba، Lamium album، L. purpureum، Stachysannua، Alyssum montanum، Lunaria annua، Moricandia arvensis، M. sonchifolia، ترب، نخود، سیب زمینی، Annonas quamosa، چغندرقند و چغندرلبویی نیز به عنوان میزبان های این نماتد گزارش شده اند. در طول سال زراعی 1391، طی مراحل بررسی چرخه زندگی نماتد سیستی کلم روی میزبان های اصلی آن در تبریز، از گیاهان زراعی مختلف منطقه و انواع علف های هرز موجود در مزارع نمونه برداری شد و آلودگی آن ها به نماتد سیستی کلم مورد بررسی قرار گرفت. ریشه های گیاهان جهت وجود این نماتد و مراحل مختلف زیستی آن با محلول لاکتو گلیسیرین-اسید فوشین رنگ آمیزی و با استفاده از استرئومیکروسکپ بررسی شدند. بررسی های مرفولوژی و مرفومتری نیز روی سیست های جدا شده از ریشه ها انجام و نتایج نشان داد که گیاهان گشنیز (Coriandrum sativum L.، Umbelliferae)، شیرتیغی (Sonchus asper (L.) Hill.، Asteraceae وSisymbrium loeselii L. Brassicaceae در شرایط طبیعی به Heterodera cruciferae آلوده می شوند، مراحل مختلف رشدی نماتد، ماده بالغ و سیست در روی ریشه ها قابل مشاهده بودند.

    سپتوریوز پسته از بیماری های مهم پسته در برخی از کشورهای دنیا است و توسط گونه های مختلفی ایجاد می شود.گونه Septoria pistacinaاز کشورهای حوزه مدیترانه و اخیرا از استان گلستان گزارش شده است(Aghajani et al. 2009.Austr. Plant Dis Notes 4:29-31).
    نمونه های دریافت شده پسته (Pistacia vera) از باغات پسته قزوین در پائیز 1392 (شکل 1) و برگ های آلوده بنه (P.mutica) جمع آوری شده در سال 1388 از اطراف داراب در استان فارس (شکل 2) دارای لکه های قهوه ای رنگ به اندازه های مختلف بود که سطح آنها دارای تعداد زیادی پیکنیدیوم فرو رفته در برگ دارای فتیله های سیاهرنگ بود مشاهده گردید. پیکنیدیوم ها حاوی تعداد زیادی کنیدیوم های نخی شکل، خمیده، روشن و دو سلولی بودند. کنیدیوم های نمونه های پسته از قزوین 2/5×30mm و در نمونه های بنه داراب 2/5×31/6mm بودند. کلنی های بدست آمده از تک کنیدیوم ها در هر دو نمونه روی PDA رشد مخمری داشتند. بر اساس خصوصیات ریخت شناسی، نحوه رشد و میزبان جدا شده Septoria pistacina تشخیص داده شد (Chitzandis.1956. Anna. Inst. Phyto.Ben. 10: 29-44). این اولین گزارش از حضور این گونه در استان قزوین و روی بنه با اطلاعات موجود در دنیا می باشد. کنیدیوم های نمونه های بنه پس از 4 سال نگهداری برگ ها در شرایط آزمایشگاه هنوز به میزان کم قدرت جوانه زنی خود حفظ کرده بودند. اخیرا گونه Septoria pistacina موجود در موسسه CBS به جنس Pseudocercospora pistacinaتغییر یافته ولی وجود پیکنیدیوم در این مورد ابهاماتی را بوجود آورده است(Crous. 2013. IMA Fungus 4: 187-199). تشکیل پیکنیدیوم روی محیط کشت، تولید سیکل جنسی در شرایط طبیعت و آزمایشگاه و واکنش ارقام تجارتی پسته به گونه مذکور تحت بررسی است.

    Beet necrotic yellow vein virus (BNYVV, Benyvirus) is an economically important soil borne pathogen of sugar beet throughout the world. It is a multipartite virus in which RNA-3 encoded p25 protein controls disease symptoms in sugar beet. Under certain conditions, resistance-breaking variants are generated by amino acid changes at positions 67 and 68 in p25. In the present study p25 protein in three populations of the virus from Moghan, Lorestan and Fars were analyzed in susceptible and resistant sugar beet cultivars. Lorestan and Fars populations were serially passaged through resistant or susceptible sugar beet cultivars for 4 generations and p25 was monitored for possible amino acid changes. No significant amino acid changes were observed when the populations were passaged through susceptible cultivars. However, passage of the populations through resistant cultivars resulted in changes of amino acids at positions 30, 49, 67, 68, 129, 163 and 198. Changes were more frequent in position 67 in the tetrad amino acids. Biotype P of BNYVV which was found in Moghan and Lorestan may have appeared as a result of selection under pressure of resistant cultivars. Information on certain evolutionary parameters and motifs of p25 is included.

    Fungi belonging to the genus Trichoderma are one of the most effective biocontrol agents of plant diseases. Because of importance of Trichoderma strains in agriculture and lack of information about their ecology and population dynamics in natural habitats in Iran, we studied seasonal changes of Trichoderma spp. populations and its relation to microbial competition and environmental factors (soil moisture, pH, EC, organic matter as well as precipitation and temperature). Three rice fields in North of Iran (Amol, Mazandaran province) were selected and sampling was performed monthly for one year at 15 cm soil depth. All microorganisms were isolated with dilution plate method, using general and selective media. Trichoderma, total fungi and bacterial populations in paddy soils ranged 1.1×101-11×103, 1.7×103-3.1×104 and 2.2×104-6.5×105 cfu/g soil, respectively. Highest Trichoderma population was recorded in late spring and during autumn for all fields, while, population was decreased in winter. According to the multiple linear regression analysis results, there was positive correlation between Trichoderma populations and soil moisture and electrical conductivity, and negative correlation with total fungal count (P=0.01). There was no significant correlation between Trichoderma populations and other factors.

    In nature, plants are exploited by a plethora of microbes species that can cause severe diseases or establish beneficial relationships with them and improve plant growth or help the plant to overcome biotic or abiotic stresses. Trichoderma and fluorescent pseudomonads are the most useful beneficial microbes. In this study, the effects of Trichoderma harzianum Tr6, Pseudomonas sp. Ps14 and their combination, which isolated from cucumber rhizosphere and were able to induce resistance on cucumber, were evaluated on cucumber immune system by evaluation of defense gene expression. The results showed that expression of β-1,3-glucanase and chitinase transiently increased after inoculation with both microbes. The transient increase of expression of PR1 and LOX1 was observed after inoculation with Tr6 and its combination with Ps14, whereas only LOX1 was increased after inoculation with Ps14. These findings indicate that the plant defense system was activated against beneficial microbes but later these microbes actively suppress the immune system of plants to establish a mutualistic interaction.

    In order to taxonomic investigation on Valsa sordida, the causal agent of canker disease on trees, 125 infected plant specimens were collected from Golestan Province. Based on morphological features, among obtained 49 fungal isolates, 35 isolates were identified as V. sordida. Moreover, asexual morphological characteristics of 12 isolates were nearly similar to C. chrysosperma and named as Cytospora sp.1. Also, morphological characteristics of an isolate (from Populus deltoides) was almost similar to V. sordida and named as Valsa sp.1. Finally, morphological characteristics of an isolate (from Ficus elastica) was different from C. chrysosperma and was near to Cytospora ribis and considered as Cytospora sp.2. To investigate the phylogeny of selected isolates, nucleotide sequence of ITS1-5.8S-ITS2 regions from nuclear ribosomal DNA (rDNA) were determined in 14 isolates (five isolates of C. chrysosperma, seven isolates of Cytospora sp.1, one isolate of Valsa sp.1 and one isolate of Cytospora sp.2), and were used along with 32 related sequences from GenBank (NCBI). The results of molecular grouping confirmed the morphological diversity in the isolates.

    Root knot nematodes are one of the most important plant parasitic nematodes with worldwide distribution and wide host range. Regarding to the hazards of synthetic chemicals, this is necessary to use other measures, such as biological control. In some studies, the separate, as well as the combined effects of two biocontrol agents, Metarhizium anisopliae and Pseudomonas fluorescens CHA0, on Meloidogyne incognita were studied. In a laboratory test, it has been showed that there is no non-volatile inhibitory effect of P. fluorescens CHA0 on M. anisopliae. In case of In vitro assay of culture filtrate and suspension of both biocontrol agents, the mixture of culture filtrates and culture filtrate of the bacterium caused about 95% and 80% larval mortality, respectively. Maximum inhibitory in egg hatch (93-98%) observed in those treatments in which culture filtrate of one or two agents were included. In the experiment that was designed to compare the application methods, the growth factors of tomato plants considerably increased and the nematode-related factors considerably decreased, in those M. anisopliae spore suspension were applied as soil drench. Soil drench of the trade formulation of M. anisopliae to the soil and foliar spray of P. fluorescens CHA0, improved the plant-related factors and decreased up to 30% of gall formation and up to 75% of reproduction factor. This is the first report of nematicidal effect of M. anisopliae on M. incognita.

    Species belonging to the genus Populus are widely used for log production, landscape design, and windbreak plantations. Septoria spp. are the most prevalent fungi causing leaf spot on poplars, which is one of the most damaging foliar diseases of these trees. However, identification of species belonging to this genus through morphological analysis is often problematic. The present work aimed to identify Septoria spp. causing leaf spot disease on poplars in East Azarbaijan, West Azerbaijan and Ardabil Provinces by using species specific primers as well as morphological features. Primers have been designed based on nr ITS rDNA sequences of S. populi and S. populicola. For this purpose, during summer 2013 symptomatic leaves were sampled from poplars in these regions, and pure cultures of isolates belonging to the genus Septoria were established. Genomic DNA of the isolates was extracted using CTAB method, and efficacy of the primers in identification of causative agent(s) of the disease was evaluated. With species-specific primer set of S. populicola we failed to obtain specific amplification products (329 bp). Species-specific primer set of S. populi, however, amplified a 439 bp fragment of all Septoria isolates. Hence, the results revealed S. populi as the only causal agent of Septoria leaf spot disease of poplars in these regions. Finally, on the basis of pathogenicity test all chosen strain caused disease on popular leaves.

    Bean common mosaic virus (BCMV), Bean common mosaic necrosis virus (BCMNV) and Cucumber mosaic virus (CMV) are seed-borne viruses in beans causing severe damage and reduction of yield and quality of this crop. Since there are lots of reports on the occurrence of mixed infection of BCMV, BCMNV and CMV, thus setting up an accurate, reliable and rapid method for detecting of these viruses in bean crops could play an important role in prevention of their spread and damages and also screening of healthy plants. For this purpose, in the present study, we tried to set up a multiplex RT-PCR for simultaneous detection of BCMV, BCMNV and CMV as well as an internal control gene, 18S rRNA. To do this end, the infection of bean plants to above mentioned viruses was confirmed by DAC-ELISA. Following total RNA extraction, cDNA was synthesized using random hexamer primers. cDNA fragments were successfully amplified using specific primers in expected sizes as almost 470 bp for BCMV, 500bp for internal control, 704 bp for BCMNV and 870 bp for CMV, respectively. The results of this study showed that this method is capable of detecting BCMV, BCMNV and CMV in 100 ng of total RNA.

    Phytophthora drechsleri Tucker which earlier was considered to be responsible for cucurbit root rot in various parts of the world including Iran is similar to P. melonis Katsura in terms of morphology and and temperature requirement. The objective of the present study was to compare pathogenicity and reaction of both species on 87 species and vareties of cucurbits including cantaloupe, long melon, cucumber, watermelon, squashes and pumpkin. Two month old cucurbit seedlings were inoculated with 4-6 week old inoculum of both species grown on vermiculate supplemented with hemp seed extract and evaluated after two months under green house conditions. The result showed that none of the cucurbit species infected with P. drechsleri and indicated that P. melonis is one of the major cucurbit root rot in Iran. The reaction of cucurbit species and cultivars showed that cantaloupeis the most susceptible host to P. melonis. Long melon and cucumber were less susceptible cucurbita species and watermelon was more resistant than other cucurbits used.

    Powdery mildew is an important disease in the fields and greenhouse-grown tomatoes in the world. Leveillula taurica has been reported as only tomato powdery mildew causal agent in Iran. During study on Erysiphaceae in Ardabil province, tomato leaves with powdery mildew symptoms in one greenhouse in Ardabil city were collected and isolates were identified based on morphological characteristics. Moreover, total DNA was extracted with Chelex 5% method and the ITS region of rDNA were amplified and sequenced using ITS1 and ITS4 primers. According to morphological and molecular characteristics, the fungus identified as Pseudoidium neolycopersici. Thus, this species reported for the first time as powdery mildew on Lycopersicon esculentum in Iran. P. neolycopersici is one of important causal agents of tomato powdery mildew in the world and its occurrence is a new treat on tomato culture in Iran.

    Traditionally, in suburbs of Tabriz city, northwest Iran (GPS 38˚06΄N, 46˚15΄E, mean temperature of 6.6-20.0°C, sandy loam soil type), different types of vegetables including cabbages have been cultivated for many years. The cabbage cyst nematode, Heterodera cruciferae was reported as one of the plant parasitic nematodes infecting kohlrabi (Brassica oleracea L. var. gongylodes) and white cabbage (Brassica oleracea L. var. captita alba) in the region. Besides these plants, brussels sprouts, cauliflower, broccoli, radish and Alyssum maritimum, Crambe maritime, Cheiranthus allionii, C. cheiri, Iberis umbellate, Lepidium sativum, Raphanus sativus, Sinapis alba, Lamium album, L. purpureum, Stachysannua, Alyssum montanum, Lunaria annua, Moricandia arvensis and M. sonchifolia, carrot, pea, potato, sweet sop (Annonas quamosa), sugar beet and garden beet, are known as hosts of the H. cruciferae worldwide. During a study on the life cycle of cabbage cyst nematode on its main hosts in the region, 2012-2013, several vegetable crops and different weeds were sampled and checked for the possibility of infection with the cyst nematode. The roots of the plants were stained using lactoglycerin-acid fuchsin solution, examined under stereomicroscope and different developmental stages of the nematode monitored. The morphological and morphometric characters of cysts were determined. As a result, the plants Coriander (Coriandrum sativum L., Umbelliferae), Sonchus (Sonchus asper (L.) Hill., Asteraceae) and Sisymbrium loeselii L. Brassicaceae were found as naturally infected hosts of the cabbage cyst nematode (Fig 1) and herein are reported as new hosts of H. cruciferae. According to available literature, there is no previous documentation of these plants as hosts of the nematode. Other plants such as Chenopodium album, Raphanus sativus and Lepidium sativum found that are infected by the cabbage cyst nematode in the area, too.

    Septoriosis is one of the most important disease in pistachio tree worldwide and is incited by several species of Septoria among which S. pistacina islimited to Mediterranea regions and recently reported on Pistacia vera in Golestan province of Iran (Aghajani et al. 2009. Austr. Plant Dis. Notes 4: 29-31). Leaf samples of P. vera from Ghazvin (Fig. 1) collected in October 2013, and samples from P. mutica (Baneh) (Fig. 2) collected also in October 2009 from Darab in Fars province showed various sizes of discolored leaf spots with many pycnidia embedded in leaf tissue extruding dark cirri. Pycnidiospores were filiform, curved, hyaline with one septum measuring 2.5 x 30 µm in Ghazvin and 2.5 x 31.6 µm from Darab samples. Single spore conidia from both locations exhibited yeast like growth on PDA. Based on morphological and growth pattern and host the fungus was identified as Septoria pistacina (Chitzandis 1956. Anna. Inst.Phyto. Ben. 10: 29-44). The species has recently been proposed to be transferred to Pseudocercospora pistacina (Crous et al. 2013. IMA Fungus 4: 187-199) which needs further support. Pycnidiospores from P.mutica samples maintained low germination after 4 years in dry leaf under laboratory conditions. This is the first report on the presence of the fungus in Ghazvin on P. vera and based on available information for the first time on P. mutica in world.
