صفحات 1-6زمینه مطالعهگزارشات اندکی در مورد فارماکوکینتیک ماربوفلوکساسین در پرندگان وجود دارد و یک داروی مفید در پرورش طیور است.هدفتعیین پارامترهای کینتیک ماربوفلوکساسین پس از تزریق داخل وریدی و تجویز خوراکی در پلاسما و عضله و تخمین دوره قطع دارو می باشد.روش کارجوجه هایی که از نظر کلینیکی سالم بودند با محدوده وزنی kg 0.22±1.08 در دسته های 5 تایی در 2 گروه استفاده گردید. در گروه Aر(450=n)،رmg 2 به ازای هر کیلوگرم ماربوفلوکساسین به صورت داخل وریدی دریافت کردند در حالیکه گروه Bر(65=n) همان دوز از دارو را پس از 12 ساعت محدودیت غذایی بصورت خوراکی دریافت کردند. نمونه های خون و g 5 بافت عضله در زمان های مختلف (در گروه آتا 24 ساعت و در گروه ب تا 120 ساعت پس از تجویز دارو) جمع آوری شدند. ماربوفلوکساسین بوسیله (اچ پی ال سی) اندازه گیری شد.نتایجماربوفلوکساسین در جوجه های تحت محرومیت غذایی تقریبا بطور کامل جذب گردید. (98%=F) متوسط زمان باقی ماندن آن 9/6 ساعت همراه با یک حجم پخش بالا بود. در تجویز خوراکی، میانگین زمان باقی ماندن در پلاسما کمتر بود (9/4 ساعت). زمان های قطع دارو تا 5/2 روز محاسبه شد.
نتیجه گیری نهایی: ویژگی های فارماکوکینتیک ماربوفلوکساسین شبیه سایر فلوروکینولون ها در طیور بود و برای دوره های تولید کوتاه مناسب می باشد.کلیدواژگان: جوجه گوشتی، HPLC، ماربوفلوکساسین، فارماکوکینتیک - کلینیکال پاتولوژی

صفحات 7-17زمینه مطالعهاسترس گرمایی از مهمترین استرس های محیطی است که خسارت تاثیرگذاری در صنعت طیور ایجاد می کند. شناخت این استرس و کنترل آن تاثیر بسیار مهمی در تولید این صنعت خواهد داشت.هدفتاثیر استفاده از ملاتونین خوراکی و رژیم تاریکی بر عوارض منفی ناشی از استرس گرمایی مورد برسی قرار می گیرد.روش کار400 قطعه جوجه گوشتی نژاد آرین تهیه و به چهار گروه تقسیم و تا 42 روزگی نگهداری شدند. جوجه های گروه کنترل در کل دوره از لحاظ دما و نور در شرایط عادی نگهداری شدند. در گروه کنترل مثبت، جوجه های آن تا 35 روزگی مانند گروه اول پرورش یافتند و بین 35 الی 40 روزگی، روزانه 6 ساعت در شرایط استرس حاد گرمایی قرار گرفتند. در گروه تاریکی، جوجه ها ضمن تحمل استرس گرمایی مانند سایر گروه ها، از یک مدل تاریکی استفاده کردند. در گروه ملاتونین، جوجه ها همانند گروه کنترل مثبت پرورش یافته و بین 30 الی 40 روزگی ملاتونین خوراکی به میزان ppm 40 در دان مصرف نمودند. جوجه ها و غذای مصرفی آنها هفتگی توزین و تلفات روزانه کالبدگشایی و گزارش شد. از جوجه ها خونگیری و غلظت هورمون های خون آنها اندازه گیری شد.نتایجاستفاده از برنامه تاریکی و ملاتونین در دان سبب کاهش تعداد تلفات ناشی از استرس گرمایی گردید که این کاهش تلفات به شکل معنی داری با کاهش غلظت T3، نسبت T4/T3 و کورتیکوسترون خون جوجه ها همراه شد. ضمنا میزان TBARS خون جوجه های گروه کنترل مثبت هم به شکل معنی داری نسبت به گروه های دیگر بالاتر بود.
نتیجه گیری نهایی: این آزمایش نشان داد که استفاده از ملاتونین خوراکی و تاریکی در پرورش جوجه سبب کاهش فعالیت متابولیک و تولید گرما در جوجه های گوشتی شده که این مسئله سبب کاهش تلفات و عوارض ناشی از استرس حاد گرمایی خواهد شد.کلیدواژگان: جوجه گوشتی، کورتیکوسترون، استرس گرمایی، ملاتونین، T4، T3 - عوامل عفونی - بیماریها - جراحی

صفحات 19-26زمینه مطالعهیک نگرانی شایع در صنعت مرغداری حضور عوامل بیماریزای قارچی در محیط پرندگان است که خطر قابل ملاحظه ای برای پرندگان، کارگران مرغداری و تمام افرادی است که در نزدیکی مرغداری زندگی می کنند.هدفاهداف این مطالعه تشخیص فلور قارچی در محیط های داخل و خارج سالن های پرورش طیور و مطالعه تاثیر روش های ضدعفونی کردن اسپری و دوددهی روی کاهش غلظت اسپورهای هوا بودند.مواد و روش کارهوای داخلی 12 سالن مرغداری با روش مواجهه پتری دیش های حاوی سابوروگلوکز آگار بعد از برداشت پوشال قدیمی، اسپری کردن با محلول های ضدعفونی کننده و دوددهی با فرمالین بعلاوه پرمنگنات نمونه برداری شدند. پلیت ها در حرارت C°30 بمدت 7 روز گرمخانه گذاری شدند و قارچ ها به صورت میکروسکوپی و ماکروسکوپی مطابق روش های استاندارد قارچ شناسی شمارش و شناسایی شدند.نتایجدر مجموع 182 و 181 کولونی قارچی بترتیب از هوای داخل و خارج سالن های مرغداری بدست آمدند. گونه های کاندیدا (2/30%) و آسپرژیلوس (9/26%) به ترتیب شایع ترین قارچ های مخمری و رشته ای در هوای داخل سالن بودند، در حالیکه گونه های آلترناریا (6/37%) و کاندیدا (3/19%) بعنوان برجسته ترین قارچ ها در هوای خارج سالن های مرغداری شناسایی شدند. ضدعفونی سالن های مرغداری با استفاده از روش های اسپری و دوددهی به ترتیب موجب کاهش 1/38% و 75% در غلظت اسپورهای هوا شدند (0.05>p).
نتیجه گیری نهایی: در مجموع، گونه های کاندیدا و آلترناریا به ترتیب بیشترین غلظت را در هوای داخل و خارج سالن های مرغداری داشتند و بیشترین تاثیر روی کاهش اسپورهای هوا مربوط به روش دوددهی بود.کلیدواژگان: اسپورهای هوا، کاندیدا، دوددهی، فلور قارچی، سالن مرغداری -
صفحات 27-32زمینه مطالعهکلستریدیوم شوای عامل شاربن علامتی است که بعنوان یک عفونت درون زا در گاو شناخته شده است. فلاژلین (liC) بعنوان عامل اصلی در ایجاد ایمنیت حفاظتی در دام ها علیه عفونت های کلستریدیایی نقش دارد. کلستریدیوم شوای دو کپی از ژن فلاژلین ( fliC) که fliA و fliB نامیده می شوند را دارا می باشد.هدفتعیین توالی و آنالیز نوکلئوتیدی هر دو کپی ژن fliC در سویه واکسینال و جدایه های فیلدی کلستریدیوم شوای هدف این مطالعه بوده است.روش کارشش پرایمراختصاصی برای تکثیر ژن فلاژلین و دو کپی آن با استفاده از نرم افزار الیگو طراحی گردید. واکنش زنجیره ای پلیمراز به منظور تکثیر یک قطعه 700 جفت بازی برای هر دو ژن fliA و fliB بکار گرفته شد. درصد تشابه و اختلاف نوکلئوتیدی بین جدایه ها با استفاده از نرم افزارهای BLAST و MegAlign محاسبه گردید.نتایجیافته های آنالیز توالی های نوکلئوتیدی نشان داد که میزان تنوع در ژن fliB بیشتر از fliA می باشد. شش ناحیه کاملا حفاظت شده، سی و یک تغییر تک نوکلئوتیدی، سیزده اختلاف اسید آمینه ای در ژن fliC جدایه های ایرانی کلستریدیوم شوای شناسایی گردید. مقایسه توالی نوکلئوتیدین های fliA و fliB، نشان داد که قرابت سویه واکسینال با جدایه های فیلدی به ترتیب 3/97 و 2/98٪ می باشد.
نتیجه گیری نهایی: کپی های ژن fliC بعنوان مارکر های خیلی مناسب برای مطالعات اپیدمیولوزیک و شناسایی تنوع در جدایه های کلستریدیوم شوای شناخته شد. وجود تنوع ژنتیکی در بین دو آلل ژن fliC برای اولین بار از ایران گزارش می شود. علی رغم وجود تفاوت های ژنتیکی مذکور، آزمایش ایمنیت متقاطع قدرت حفاظت واکسن تهیه شده از سویه بومی (توسط موسسه رازی) را در چالنج با سویه های همولوگ و هترولوگ نشان داد.کلیدواژگان: کلستریدیوم شوای، فلاژلین، پلی مورفیسم -
صفحات 33-40مقدمهسویه های مختلف اشریشیا کلی به عنوان عوامل مهم عفونت های داخل و خارج روده ای در انسان و گونه های مختلف دامی شناخته می شوند. روش های مولکولی، برای شناسایی جدایه های باکتریایی، تنوع سکانس نوکلئوتیدی، اطلاعاتی در زمینه ردیابی عوامل باکتریایی دخیل در همه گیری ها، شناسایی تکراردر ساختار ژنتیکی و نیز شناسایی تکامل تدریجی باکتری ها، اهمیت دارند.هدفهدف از این مطالعه ارزیابی میزان کارایی روش RAPD-PCR در تفکیک جدایه های اشریشیا کلی می باشد.روش کاردر این مطالعه 110 جدایه اشریشیا کلی از منابع مختلف به کمک دو الیگونوکلئوتید bp 10 با سکانس های اختیاری به روش RAPD-PCR مورد بررسی قرار گرفتند. این جدایه ها از افراد مبتلا به عفونت ادراری، گوساله های نوزاد مبتلا به اسهال یا سپتی سمی، جدا شده بودند.نتایجتجزیه و تحلیل اطلاعات نشان داد که 5/87٪ ازجدایه های اشریشیا کلی جدا شده از انسان به درستی در گروه میزبان انسان طبقه بندی شدند، درحالی که 3/94٪ از جدایه های اشریشیا کلی به درستی در گروه های مربوط به گوساله قرار گرفتند. همچنین نشان داده شد که 100٪ و 3/93٪ از جدایه ها به درستی به ترتیب درگروه های گوساله اسهالی وسپتی سمی قرار گرفتند.
نتیجه گیری نهایی: آنالیز تنوع ژنتیکی نشان داد که درصد پلی مورفیسم در بین جدایه های اشریشیا کلی از افراد مبتلا به عفونت ادراری، گوساله های مبتلا به اسهال و گوساله های سپتی سمی به ترتیب 71/54٪، 22/61٪ و 5/62 ٪ بودند.کلیدواژگان: E، coli، تنوع ژنتیکی، RAPD، PCR -
صفحات 41-46زمینه مطالعهارزیابی درد در دام های کوچک به دلیل عدم توانایی در بیان درد و فقدان وجود سیستم واحد جهانی سنجش درد همواره با چالش همراه بوده است. مشخص شده است که بین افزایش برخی از پارامترهای بیوشیمیایی خون و میزان درد پس از عمل جراحی رابطه مستقیم وجود دارد.هدفهدف از این مطالعه ارزیابی تغییرات میزان آنزیم کراتین فسفوکیناز سرم و همچنین احتمال وجود رابطه آماری بین تغییرات این آنزیم و درد پس از عمل جراحی در گربه ها بود.روش کاربرداشت رحم و تخمدان به روش معمول جراحی از خط وسط در 24 گربه ماده سالم انجام شد. سطوح سرمی هورمون کورتیزول، آنزیم کراتین فسفوکیناز و گلوکز، در پیش از جراحی و 1، 3 و 24 ساعت پس از جراحی اندازه گیری شد. همچنین میزان درد پس از عمل جراحی با استفاده از مقیاس سنجش درد بصری VAS در 1 ، 3 و 24 ساعت پس از جراحی ثبت شد. داده ها به کمک آزمون آنالیز داده های تکراری و همچنین استفاده از ضریب اسپیرمن برای بررسی احتمال وجود رابطه بین میزان آنزیم کراتین فسفوکیناز و درد پس از عمل جراحی آنالیز شد.نتایجمیزان درد به صورت معنی داری در تمامی گربه ها در 1 و 3 ساعت پس از جراحی افزایش یافت (0.001=p). میزان هورمون کورتیزول و آنزیم کراتین فسفوکیناز پس از جراحی به صورت معنی داری نسبت به مقادیر پیش از جراحی افزایش یافت (0.001>p). نتایج نشان داد که در 24 ساعت پس از جراحی میزان آنزیم کراتین فسفوکیناز همچنان نسبت به میزان پیش از جراحی به صورت معنی داری بالاتر بود. رابطه مستقیم معنی داری بین میزان درد پس از عمل و میزان آنزیم کراتین فسفوکیناز مشخص شد (0.05>p).
نتیجه گیری نهایی: رابطه مستقیم معنی داری بین میزان درد پس از عمل جراحی و سطوح سرمی هورمون کورتیزول و آنزیم کراتین فسفوکیناز در ساعات اولیه پس از جراحی مشاهده شد. آنزیم کراتین فسفوکیناز می تواند به عنوان شاخصی در سنجش درد در ساعات اولیه پس از عمل در گربه ها که درد در میزان حداکثر می باشد استفاده شود.کلیدواژگان: کورتیزول، آنزیم کراتین فسفوکیناز، گربه، درد پس از عمل جراحی -
صفحات 47-52زمینه مطالعهرودوکوکوس اکوئی، پاستورلا مولتی سیدا و بروسلا آبورتوس از مهمترین باکتری های پاتوژن دامپزشکی محسوب شده که در سال های اخیر مقاومت این باکتری ها به آنتی بیوتیک های رایج باعث نگرانی هایی در صنعت دامپروری شده است. برای کنترل این باکتری ها، استفاده از ترکیبات ضد باکتریایی جدیدی چون مشتقات تیازول در دامپزشکی ضروری است.هدفدر این مطالعه به بررسی اثر ضد باکتریایی مشتقات جدید تیازول بر روی سه باکتری رودوکوکوس اکوئی، پاستورلا مولتی سیدا و بروسلا آبورتوس پرداختیم.روش کارپس از حل کردن مشتقات در DMSO، برای بررسی مقایسه اثر ضد باکتریایی، از روش انتشار در دیسک برای اندازه گیری قطر هاله مهار رشد و از روش براس میکرودایلوشن برای تعیین حداقل غلظت بازدارندگی رشد (MIC) استفاده شد.نتایجاثر مهاری از هیچ کدام از مشتقات روی بروسلا آبورتوس مشاهده نشد اما قطر هاله مهار رشد mm 4/0±7/12تا 2/0±2/31 و MICرμg/mlر32 تا 256 برای اثر مهاری مشتقات روی رودوکوکوس اکوئی و پاستورلا مولتی سیدا ثبت گردید.
نتیجه گیری نهایی: نتایج این تحقیق قدرت مهاری مشتقات تیازول را بر روی دو باکتری مهم دامپزشکی (رودوکوکوس اکوئی و پاستورلا مولتی سیدا ) اثبات می کند.کلیدواژگان: اثر ضد باکتریایی، بروسلا آبورتوس، پاستورلا مولتی سیدا، رودوکوکوس اکوئی، مشتقات تیازول -
صفحات 53-64زمینه مطالعهویروس نقصان ایمنی گاو یکی از اجرام بیماریزای شناخته شده به عنوان یک عامل سرکوب کننده سیستم ایمنی گاو است اما روند بیماریزایی آن به خوبی مشخص نشده است. در سال های اخیر، این فرضیه مطرح شده است که عفونت با BIV گاو را به آلوده شدن به سایر عوامل بیماریزا مستعد می نماید.هدفهدف از این مطالعه جستجوی عفونت توام BIV و بروسلا است تا در آینده مطالعات بیشتری در مورد مستعد شدن گاو به ابتلا به سایر میکروارگانیسم ها نظیر بروسلا پس از عفونت با BIV انجام شود.روش کارتعداد 2290 نمونه خون گاو از فارم های غیر صنعتی (490=n) و صنعتی (1800=n) پرورش گاو شیری واقع در استان های چهار محال و بختیاری و اصفهان، ایران، اخذ گردید. حیوانات آلوده به BIV با آزمون های Lab- ELISA و nested PCR شناسایی شدند.نتایجمیزان کلی شیوع BIV، 61/1% محاسبه شد (7/5% و 83/0% به ترتیب در فارم های غیرصنعتی و صنعتی).
نتیجه گیری نهایی: با استفاده از آزمون های آماری مربع کای و ضریب همبستگی پیرسون ارتباط معنی داری بین وضعیت آلودگی به BIV و بروسلوز در مورد کل نمونه ها (0001/0=p ،24/0=r) و نمونه های مربوط به چهارمحال و بختیاری (044/0=p ،13/0=p) و در فارم های صنعتی اصفهان (001/0=p ،074/0=p) دیده شد.کلیدواژگان: ویروس نقصان ایمنی گاو، بروسلا، شیوع سرمی - کلینیکال پاتولوژی

صفحات 65-71زمینه مطالعهشتر و گوسفند از مقاومت و گستردگی زیادی در مناطق خشک برخوردارند. اما نتایج متفاوتی از توانایی آنها در تحمل این شرایط گزارش شده است.هدفهدف این آزمایش، بررسی تحمل استرس گرمایی در شتر و گوسفند به وسیله ارزیابی تغییرات متابولیت های سرم خون، گزارش و مقایسه پروفیل بیوشیمیایی سرم خون گوسفند و شتر در طول استرس گرمایی طولانی تابستان است.روش کاردر این آزمایش نمونه های خون گوسفند و شتر در چهار ماه متوالی اخذ و مورد مقایسه قرار گرفت.نتایجتفاوت معنی داری بین مقادیر اوره، گلوکز، کل پروتئین، آلبومین، فسفر، کلسیم،آنزیم های آمینوترانسفراز، اسید اوریک، کلسترول، تری گلیسرید، کل بیلی روبین و لاکتات دهیدروژناز گوسفند و شتر مشاهده شد. بطورکلی اوره، گلوکز، کل پروتئین، آلبومین، فسفر، کلسیم به طور معنی داری در شتر بالاتر از گوسفند بود (0.01>p). برعکس گوسفندان به طور معنی داری مقادیر بالاتری از اسید اوریک، کلسترول، تری گلیسرید، کل بیلی روبین و لاکتات دهیدروژناز نشان دادند (0.01>p). با این وجود آنزیم های آمینوتراسفراز و کراتین دو حیوان تفاوتی با یکدیگر نشان نداد.
نتیجه گیری نهایی: این نتایج نشان داد گوسفند به استرس گرمایی حساسیت بیشتری دارد که با افزایش اسید اوریک، کلسترول، تری گلیسرید، کل بیلی روب ین و لاکتات دهیدروژناز نمایان می شود. بطوریکه شاید بتوان گفت گوسفندان در طول استرس گرمایی الگوی متابولیسمی لیپولیز را دارند. بالابودن لاکتات دهیدروژناز و کل بیلی روبین نشانه تخریب گلبول قرمز یا آسیب کبدی می باشد. همچنین غلظت بالای اسید اوریک خون گوسفندان سبب حساسیت بیشتر آنها به آسیب های کلیوی مانند نقرس می شود.کلیدواژگان: متابولیت، استرس گرمایی، گوسفند، شتر - تغذیه - بهداشت و پرورش

صفحات 73-81زمینه مطالعهعصاره کنگرفرنگی حاوی ترکیبات آنتی اکسیدان طبیعی می تواند بعنوان یک منبع با پتانسیل خوب آنتی اکسیدانی در نظر گرفته شود.هدفهدف از انجام این مطالعه ارزیابی توانایی آنتی اکسیدان عصاره کنگر فرنگی بر کیفیت گوشت جوجه ها بود.روش کار200 جوجه گوشتی نژاد راس به پنج گروه مساوی تقسیم شدند و به ترتیب 100، 200، 300 و 500 عصاره کنگر فرنگی (94%) در آب آشامیدنی (mg/l) و گروه کنترل آب خالص دریافت کردند. پیش از افزودن عصاره به آب آشامیدنی، فعالیت آنتی رادیکالی عصاره توسط روش 1,1-diphenyl-2-picrylhydrazyl (DPPH) و محتوی فنل آن بر اساس میزان گالیک اسید اندازه گیری شد. جوجه های گوشتی با عصاره کنگر فرنگی از روز 35-21 دوره رشد درمان شدند و نمونه های گوشت ران برای آنالیز بیوشیمیایی در روز 42 از دوره پرورش تهیه شد.نتایجفعالیت آنتی رادیکالی عصاره و محتوی فنل به ترتیب 35٪ و/100 g 3/3 عصاره بود. در خصوص آنزیم های آنتی اکسیدانی مانند گلوتاتیون پراکسیداز (GPx) و کاتالاز (CAT)، استفاده ازmg/l 200 عصاره کنگرفرنگی بیشترین خاصیت آنتی اکسیدانی را در مقایسه با گروه های دیگر نشان داد (0.05>p). همچنین، افزودن mg/l 200 عصاره کنگرفرنگی به آب آشامیدنی کمترین فعالیت را برای آنزیم های GPx و CAT در مقایسه با گروه کنترل و mg/l 300 صاره نشان داد (0.05>p). تفاوت های معنی داری بین گروه کنترل و گروه های دریافت کننده عصاره برای شاخص های عملکردی وزن مانند: میزان افزایش وزن، متوسط افزایش وزن روزانه، درصد افزایش وزن در روزهای 35-21 دوره آزمایش نسبت به کل دوره و وزن نهایی وجود نداشت (0.05نتیجه گیری نهایی: این مطالعه نشان داد که تجویز mg/l 200 عصاره کنگرفرنگی در آب آشامیدنی در طول دوره رشد فعالیت آنزیم های آنتی اکسیدانی گلوتاتیون پراکسیداز (GPx) و کاتالاز (CAT) را در گوشت ران جوجه های گوشتی کاهش داد، که احتمالا نتیجه کاهش بیان ژن برای این آنزیم ها می باشد.کلیدواژگان: عصاره کنگرفرنگی، جوجه گوشتی، گلوتاتیون پراکسیداز، پایداری لیپید

Pages 1-6BackgroundReports on the pharmacokinetics of marbofloxacin in birds are scarce, even when it is a useful tool in poultry production.ObjectivesDetermining marbofloxacin kinetic parameters in plasma and muscle arrangement after intravenous and oral administrations and establishing its withdrawal period.MethodsClinical healthy chickens (1.08±0.22 kg) were used as experimental subjects, formed in groups of 5. In Group A (n= 45) birds received 2 mg/kg of marbofloxacin intravenously while group B (n= 65) was given the same oral dose after fasting for 12 hours. Blood samples in the groups and 5 g of muscle were obtained at different times, in A group up to 24 hours and in B group until 120 hours after application. Once the samples were obtained and treated, marbofloxacin was quantified by HPLC.ResultsMarbofloxacin in fasted chickens is almost completely absorbed (F= 97%), mean residence time is moderate (6.9 hours), with a high volume of distribution. Orally, mean residence time is less in plasma (4.9 hours) and the withdrawal time was calculated 2.5 days.ConclusionsThe kinetic results are consistent with those presented by other fluoroquinolones in poultry and are in line with short production cycles in chickens.Keywords: chickens, HPLC, marbofloxacin, pharmacokinetics

Pages 7-17BackgroundHeat stress is one of the most important environmental stressors challenging poultry production worldwide. Understanding and controlling environmental stressors is crucial for successful poultry production.ObjectivesThe aim of the present study was to investigate the effects of melatonin supplementation and darkness regime on reducing the negative effects of heat stress in broiler chickens.MethodsA total of 400 broiler chickens (Arian) were obtained and subjected to four different groups. The control chickens were grown in normal conditions without receiving any treatment. The positive control birds were grown similar to the control but exposed to an acute heat stress for 6 h/d from days 35 to 40. The dark group chickens were reared under a dark schedule and exposed to heat stress. The birds of the fourth group were reared similar to the positive control but received 40 ppm melatonin in the diet from days 30 to 40 of age. Dead birds were autopsied. The weekly growth performance of chickens was determined and blood samples were taken for hormonal analysis.ResultsThe number of dead birds due to heat stress was significantly decreased in chickens subjected to the dark program and/or those which received melatonin in the diet compared with those of positive control chickens. Such differences were accompanied with the significantly lower levels of plasma T3, T3/T4 and corticosterone in these groups of chickens. Additionally, plasma thiobarbituric acid reacting substances level was significantly higher in positive control broilers compared to their counterparts.ConclusionsOur findings indicate a reduction of metabolic rate and heat production in dark regime and melatonin supplementation that can reduce the side effects of heat stress and, therefore, reduce the mortality rate.Keywords: broiler chickens, corticosterone, Heat stress, melatonin, T3, T4

Pages 19-26BackgroundA common concern of the poultry industry is the presence of fungal pathogens in the birds environment, which may constitute a considerable health hazard to the birds, farmers, and those living in proximity of the farm.ObjectivesThe aims of this study were to assess the mycoflora in the indoor and outdoor environments of poultry breeding houses and studying the efficacy of disinfection methods, including spraying and fumigation, on reducing airspora concentration.MethodsIndoor air of 12 poultry houses were sampled by exposing Petri dishes containing Sabourauds glucose agar after removal of old litter, spraying with disinfectant solutions, and fumigation with formalin plus permanganate. The plates were incubated at 30 °C for seven days and fungi were counted and identified microscopically and macroscopically according to standard mycological methods.ResultsA total of 182 and 181 fungal colonies were recovered from indoor and outdoor air of poultry houses, respectively. Candida (30.2%) and Aspergillus (26.9%) species were the most common yeast and mold in the indoor, respectively, whereas Alternaria (37.6%) and Candida (19.3%) species were the most predominant fungi in the outdoor air of poultry houses. Disinfection of the poultry houses using spraying and fumigation methods led to a 38.1% and 75% reduction in airspora concentration (pConclusionsBased on the findings of the present study, Candida spp and Alternaria spp had the highest indoor and outdoor concentrations in poultry breeding houses air, respectively, and fumigation was the most efficient method in reducing airspora.Keywords: airspora, Candida, fumigation, mycoflora, poultry house

Pages 27-32BackgroundClostridium chauvoei is the etiological agent of blackleg as an endogenous infection in cattle. Flagella have been known to play a critical role in the protective immunity of animals to clostridial infections. C.chauvoei has two copies of fliC gene, namely fliA and fliB.ObjectivesThe aim of this study was the determination and nucleotide sequence analysis of both copies of fliC genes in vaccinal strain and Iranian C. chauvoei isolates.MethodsSix specific primers for amplification of fliA, fliB, and flagellin (fliC) genes were designed by Oligo software. Polymerase chain reaction was performed to amplify a fragment of 700 bp for both copies of flagellin (fliA and fliB) genes. The nucleotide percentage identity and divergence among isolates were deduced using BlAST and MegAlign softwares.ResultsIt was found that divergence in fliB was more than fliA by sequence alignment analysis. Six highly conserve regions, thirty-one SNPs and 13 amino acid polymorphisms were found in fliC gene (between fliA and fliB sequences) of Iranian C. chauvoei isolates. In comparative analysis, genomic similarity of the fliA and fliB genes between the vaccinal strain and examined field isolates was proved to be as high as 97.3 % and 98.2%, respectively.ConclusionsThe fliC copies were identified as excellent biomarkers to study the molecular epidemiology and strain diversity among C. chauvoei isolates. The existence of genetic variation between two alleles of fliC gene in C. chauvoei is reported for the first time in Iran. In spite of some genetic variations, the immunologic cross protection test showed a high protection power of the local vaccine (produced by Razi Institute) against homologous and heterologous challenge.Keywords: Clostridium chauvoei, flagellin, polymorphism

Molecular methods are important for the identification of bacterial isolates and nucleotide sequence variations, as well as information on tracking bacterial agents related to the outbreaks, the frequency of the bacterial genetic structure, and the evolution of microbial populations.ObjectivesThe purpose of the present study was to evaluate the efficiency of the RAPD method to differentiate E. coli strains.MethodsIn this study, 110 isolates of E. coli were analyzed by the RAPD PCR method using two 10bp oligonucleotides. These strains were isolated from humans with urinary tract infections and neonatal calves affected by diarrhea or septicemia.ResultsData analysis showed that 87.5% of human E. coli isolates were correctly classified in the human host group, while 94.3% of calf E. coli isolates were correctly placed in calf groups. It also demonstrated that 100% and 93.3% of isolates were accurately assigned to diarrheic and septicemic calf groups, respectively.ConclusionsGenetic variation analysis indicated that the percentage of polymorphism among E. coli isolates from humans with urinary tract infections, diarrheic calves, and septicemic neonatal calves were 54.71%, 61.22%, and 62.5%, respectively.Keywords: E. coli, genetic variation, RAPD, PCR

Increase in biochemical parameters have been shown to have direct correlation with level of postoperative pain.ObjectivesThe purpose of the present study is to evaluate the serum levels of CPK and investigate any correlation in serum levels of CPK and postoperative pain following ovariohysterectomy in feline.MethodsConventional midline ovariohysterectomy was performed in 24 healthy female queens. Serum levels of cortisol, CPK and glucose were measured prior to surgery and at 1 and 3 and 24 hours after the surgery. Also, VAS was used to assess level of post-operative pain. Data was analyzed using repeated measure ANOVA. Spearmans rank correlation coefficient was used to identify any correlation between level of pain and CPK after the surgery.ResultsThe score of pain significantly increased at 1 and 3 hours after the surgery in all of the cats (p=0.001).The concentration of cortisol and CPK significantly increased after the surgery in comparison to the values prior to surgery (pConclusionsIt is concluded that there is a correlation with VAS score of pain and serum levels of Cortisol and CPK at early hours after the surgery. CPK might be used as an indicator of pain in early hours after the surgery in feline. Pages 47-52BackgroundRhodoccocus equi, Brucella abortus, and Pasteurella multocida are important veterinary bacterial pathogens that in recent years have been resisted to current antibiotics, and this problem threats the livestock industry. To control this resistant microorganisms, use of new antibacterial compounds, such as thiazole derivatives, in veterinary is necessary.ObjectivesIn this study, antibacterial effects of thiazole derivatives on Rhodoccocus equi, Brucella abortus, and Pasteurella multocida were evaluated.MethodsSynthesized thiazole derivatives were prepared in DMSO, then the disk diffusion method was used to measure growth inhibition zone diameter and the broth microdilution method was applied to determine the minimum inhibitory concentration (MIC).ResultsResults showed that thiazole derivatives had no significant inhibitory effects on B. abrotus, while they had inhibitory effects on R. equi and P. multocida with inhibition zone 12.7±0.4 -30.1±0.2 mm and MICs 32- 256 μg/ml.ConclusionsResults of this study indicate that thiazole derivatives have considerable inhibitory effects on R. equi and P. multocida as veterinary bacterial pathogens.Keywords: antibacterial activity, B. abrotus, P. multocida, R. equi, thiazole derivatives

Pages 53-64BackgroundBIV is a well-known bovine immunosuppressive cause, but its pathogenesis has not been well characterized. In recent years, it has been hypothesized that infection with BIV might predispose cattle to be infected by other agents.ObjectiveThis study was performed to investigate of BIV and Brucella co-infection so that in the future more studies will be done on the issue of predisposing cattle to other microorganisms like Brucella after BIV infection.MethodsBlood samples were collected from a total of 2290 cattle in Iran (490 and 1800 cattle in non-industrial and industrial dairy farms, respectively from Isfahan and Chaharmahal and Bakhtiari provinces). The BIV-positive animals were detected by Lab-ELISA and nested PCR tests.ResultsIn this study, the overall prevalence of BIV in Iran was 1.61% (4.5% and 0.83% in non-industrial and industrial dairy farms, respectively).ConclusionsThere was a statistically significant relationship between BIV status and Brucella infection using Chi square and Pearsons correlation coefficient test for all of the samples (p=0.0001, r=0.24), samples from Chaharmahal and Bakhtiari (p=0.044, r=0.13) and from industrial farms in Isfahan (p=0.001, r=0.074).Keywords: Bovine immunodeficiency virus, Brucella, seroprevalence

But different results of harsh condition tolerance ability of them have been reported.ObjectivesThe objective of this study was to determine the heat stress tolerance in camel and sheep by evaluating changes in blood serum metabolites and to report and compare the serum biochemical profile of sheep and camel during long heat stress of warm summer.MethodsIn this experiment, blood metabolites of camel and sheep were taken and compared with each other in four consecutive months during warm months (high summer).ResultsThere was a significant difference between the values of urea, glucose, total protein, albumin, phosphors, calcium, ALT, ALP, uric acid, cholesterol, triglyceride, total bilirubin, and LDH of sheep and camels. Pages 73-81BackgroundArtichoke extract (AE), containing natural antioxidant compounds, can be considered as a good source of antioxidant potential.ObjectivesThe aim of this study was to evaluate antioxidant abilities of AE on broiler meat quality.Methods200 Ross chicken broilers were divided into five equal groups and received 100, 200, 300, and 500 mg/liter of AE in drinking water and pure water in the control group, respectively. Antiradical activity and phenolic content of AE were determined by 1,1-diphenyl-2-picrylhydrazyl (DPPH) method and gallic acid measurement before adding extract into drinking water. The broilers received AE extract form 21-35 day of growing phase and the samples from thigh muscles were taken for biochemical analysis in the 42 day of the growing phase.ResultsAntiradical activity of AE was 35% and phenolic content was 3.3 g/100g of dry extract. Regarding antioxidant enzymes, such as glutathione peroxidase (GPx) and catalase (CAT), the AE with dosage of 200 mg/l indicated maximum antioxidant ability compared to the other groups (pConclusionsThis study showed that administration of 200 mg/l AE in drinking water during growing phase decreased GPx and CAT activities in chicken meat presumably due to down-regulation of gene expression for antioxidant enzymes.Keywords: Artichoke extract, broiler chicken, glutathione peroxidase, lipid stability
