    یکی از مکانیسم های سازگاری سودوموناس های فلورسنت، پدیده تنوع فازی می باشد؛ این پدیده در اثر تغییرات ژنتیکی برگشت پذیر رخ داده که نتیجه آن ظهور زیرجمعیت های متنوع از یک باکتری با رفتارهای متفاوت می باشد که سبب بقا و سازگاری آنها با شرایط مختلف محیطی و تغییرات ناگهانی آنها می شود. در این تحقیق وقوع پدیده تنوع فازی در جدایه B1 باکتری Pseudomonas fluorescens (جدایه واجد ژن تنظیم کننده تنوع فازی) مشاهده شد. جدایه مزبور دارای دو تیپ کلونی بود، کلونی های تیپ وحشی کوچک و کدر بودند، در حالیکه کلونی تیپ ثانویه بزرگ تر بوده و شدت رنگ فلورسنس بیشتری داشتند. این دو تیپ از نظر تولید آنزیم های پروتئاز، لیپاز، فسفاتاز و تولید متابولیت های سیانید هیدروژن، سیدروفور و اکسین و رفتارهای تحرک، تشکیل بیوفیلم و فعالیت ضدقارچی علیه چند قارچ بیماریزای گیاهی مورد مقایسه قرار گرفتند. تفاوت های رفتاری معنی داری بین دو تیپ مشخص گردید؛ هر دو تیپ توانایی تولید پروتئاز، لیپاز و فسفاتاز را داشتند اما تولید سیانید هیدروژن در تیپ ثانویه به شدت کاهش پیدا کرده و برعکس میزان سیدروفور و اکسین تولید شده توسط این تیپ در مقایسه با تیپ وحشی افزایش پیدا کرده بود. میزان تحرک کلونی هایی تیپ ثانویه بسیار بیشتر از تیپ وحشی بوده ولی میزان تشکیل بیوفیلم توسط تیپ اولیه بیشتر از تیپ ثانویه بود. از نظر فعالیت ضدقارچی تیپ وحشی قوی تر از تیپ ثانویه عمل کرد. انگشت نگاری DNA به روش BOX-PCR تشابه کامل واریانت ها را با جدایه اولیه تایید کرد.

    بیماری بلایت فوزاریومی سنبله گندم از لحاظ اقتصادی یکی از مهمترین بیماری های قارچی گندم در سرتاسر جهان می باشد. هدف از انجام این تحقیق بررسی و شناسایی متابولیت های ضدقارچی جدایه های بومی باسیلوس بود. جدایه های باسیلوس از خاک مزارع کشاورزی گندم و جو (واقع شده در جنوب تهران در پاییز 91) جداسازی شد و علیه چندین جدایه فوزاریوم بومی جداسازی شده از دانه های گندم آلوده فعالیت آنتاگونیستی نشان داد. تخلیص و شناسایی متابولیت های ضدقارچی تولید شده توسط جدایه منتخب باسیلوس در محیط نوترینت براث انجام شد. روش کروماتوگرافی مایع با کارایی بالا بر روی عصاره تخلیص شده حضور آنتی بیوتیک های لیپوپپتیدی ایتورین را به عنوان پیک اصلی نشان داد به طوری که با ایتورین آخالص شرکت سیگما قابل مقایسه بود. ساختار متابولیت های ضدقارچی تخلیص شده به وسیله روش های طیف سنجی مادون قرمز و طیف سنجی جرمی بر پایه کروماتوگرافی مایع مورد شناسایی قرار گرفت و ساختار ایتورینی آن تایید گردید. آنالیز طیف سنجی جرمی همچنین توانایی باسیلوس منتخب را در تولید فنجی سین علاوه بر ایتورین آاثبات نمود. توالی ژنومی جدایه باسیلوس منتخب 9/99 درصد با توالی ژنومی باسیلوسآریاب هاتایی و ژنوم جدایه منتخب فوزاریوم 99 درصد با فوزاریوم گرامینئاروم قرابت ژنومی نشان داد. بر اساس نتایج حاصل از این تحقیق، اثرات ضدقارچی جدایه های بومی باسیلوس و نیز اثرات آن ها در کنترل بیولوژیک بیماری بلایت فوزاریومی سنبله گندم مورد تایید قرار گرفت.

    باکتری مولد گال طوقه، Rhizobium vitis و نماتود ریشه گرهی Meloidogyne javanica در خاک های اکثر مناطق ایران، به ویژه باغات میوه، گسترش و شیوع روزافزونی دارند. به منظور بررسی برهمکنش نماتود ریشه گرهی و باکتری عامل گال طوقه در روی پایه های وارداتی GF677 و GN15 بادام و پایه هیبرید هلو × بادام شورابی (تولید شده در استان چهارمحال و بختیاری)، آزمایشی به صورت فاکتوریل در قالب طرح کاملا تصادفی در حضور و غیاب هر یک از دو بیمارگر، در شرایط گلخانه انجام شد. مایه زنی با 3000 تخم و لارو نماتود بر کیلوگرم خاک پس از استقرار پایه های ریشه دار منتقل شده به گلدان های آزمایشی و مایه زنی با باکتری به نسبت یک درصد حجم به وزن با سوسپانسیون cfu/ml 108 سه هفته بعد از مایه زنی نماتود صورت گرفت و سطوح صفر نماتود و باکتری به عنوان شاهدهای آزمایش منظور گردیدند. سه ماه بعد از آخرین مایه زنی، شاخص های رشدی گیاه، نماتود و بیماری زایی باکتری مورد ازریابی قرار گرفت. نتایج نشان داد که به جز پایه ی GN15، سایر پایه های مورد بررسی به نماتود حساس می باشند. در مقابل، پایه ی GN15 نسبت به باکتری مولد گال طوقه بسیار حساس بود. حضور نماتود فقط در پایه ی GN15 تاثیر معنی دار بر بیماری زایی باکتری داشت. میزان این افزایش در مقایسه با شاهد 3/23% بود. همچنین حضور باکتری موجب افزایش شاخص های نماتود در سایر پایه ها به جز پایه ی GN15 نشد.

    طی سال های زراعی 89-1388 و 90-1389 بررسی های مزرعه ای به منظور تعیین اثر تاریخ کشت و تیمار بذر با ایمیداکلوپرید روی عملکرد (بیوماس) ذرت علوفه ای رقم Single cross 704، اجزاء عملکرد و کنترل بیماری ویروسی موزاییک ایرانی ذرت در شهرستان های شهرکرد و لردگان از استان چهارمحال و بختیاری انجام گرفت. آزمایش در قالب طرح بلوک های کامل تصادفی با چهار تکرار به صورت کرت های خردشده بود. کرت های اصلی شامل چهار تاریخ کشت به فاصله 15 روز از 20 اردیبهشت الی پنجم تیرماه و کرت های فرعی تیمار بذر با حشره کش جذبی بود. در هر دو سال عملکرد در کرت های کشت شده در 20 اردیبهشت به طور معنی داری در هر دو شهرستان کمترین و درصد آلودگی به بیماری موزاییک ایرانی ذرت بیشترین میزان بود. وقوع بیماری در کرت های آزمایشی بسته به تاریخ کشت و تیمار یا بدون تیمار بذر از 2% تا 5/44% متغیر بود. آزمون الیزا با استفاده از آنتی سرم ویروس موزاییک ایرانی ذرت این نتایج را تائید نمود. بهترین عملکرد و اجزاء عملکرد ذرت در هر دو سال مربوط به کرت های کشت شده در بیستم خرداد و پنجم تیرماه به ترتیب برای شهرستان های شهرکرد و لردگان بود. عملکرد و اجزاء عملکرد ذرت در پاسخ به تیمار بذر بسته به تاریخ کشت از 3/9 تا 32% افزایش معنی دار نشان داد (P=0.01). در بررسی تغییرات جمعیت زنجرک های Laodelphax striatellus و Unkanodes tanasijevic، ناقلین موزاییک ایرانی ذرت، بیشترین جمعیت زنجرک ها درزمانی شکار شد که بوته های ذرت در تاریخ کشت 20 اردیبهشت در مرحله سه برگی بودند و در سایر تاریخ کشت ها گیاه چه ها هنوز ظاهرنشده بودند. علاوه بر موزاییک ایرانی ذرت علائم آلودگی به کوتولگی زبر ذرت مشاهده شد، همچنین نتایج الیزا آلودگی به موزاییک مخطط گندم و سروتیپ های PAV و MAV ویروس کوتولگی زرد جو را در برخی کرت های آزمایشی نشان داد. تعداد هفت رقم ذرت (ZP677، ZP505، ZP684، Single cross 704، KONSOUR، KSC500، TWC 647) ازنظر وضعیت آلودگی به ویروس های موزاییک ایرانی ذرت در هر دو منطقه در شرایط آلودگی طبیعی در سال زراعی 89-1388 مورد ارزیابی قرار گرفت که بیشترین و کمترین میزان و شدت بیماری به ترتیب مربوط به Single cross 704 و KONSOUR بود.

    اامیست گرماپسند Phytophthora parsiana، یکی از عوامل ایجاد کننده بیماری انگومک پسته در ایران، اولین بار از طوقه درخت انجیر در ایران جداسازی شد. hydropathica Phytophthora و Phytophthora irrigata دو گونه جدید گرما پسند مشابه P. parsiana اولین بار از آبهای آبیاری و شاخه های در حال بلایت و پژمردگی گیاهان زینتی در امریکا جداسازی و توصیف شدند اما مطالعات اندکی روی بیماریزائی و دامنه میزبانی آنها صورت گرفته و با توجه به اینکه در مطالعات قبلی بر اساس درختهای فیلوژنی، جدایه های P. hydropathica در بین جدایه های P. parsiana قرار گرفته و جایگاه برخی جدایه های این دو گونه مشابه مشخص نیست بنابراین در این مطالعه تفاوتهای ریخت شناختی و بیماریزایی این جدایه ها جهت تفکیک یا قرار گرفتن این جدایه ها در یک گونه مورد بررسی قرار گرفت. در مطالعه حاضر هر سه بیمارگر در بافت آلوده میزبان کلامیدوسپور های کروی شکل تشکیل دادند، P. hydropathica ااسپور و ااگونیوم، کلامیدوسپور و آماس ریسه بزرگتری نسبت به تمامی جدایه های P. parsiana و P. irrigata تشکیل داد، از میان میزبان های چوبی مایه زنی شده، نهالهای پسته رقم سرخس، بادام و زردآلو بیشترین حساسیت را به هر سه گونه داشتند. بنه، خنجوک، گردو، انجیر و انبه توسط P. hydropathica و P. parsiana و پسته رقم قزوینی توسط P. hydropathica علائم بیماری را از چند روز تا چند ماه پس از مایه زنی نشان دادند. نهالهای ازگیل و کنار به تمامی جدایه های هر سه گونه مقاوم بودند.

    گونه Pyricularia oryzaeعامل بلاست برنج یکی از خسارتزا ترین و مهمترین بیماری های برنج در دنیا می باشد. به منظور شناسایی میزبان های P. oryzaeو جدا کردن این گونه از گونه P. grisea، در بهار، تابستان و پاییز سال های 1391 و 1392 از گیاهان دارای نشانه های بلاست و لکه برگی در مزارع برنج، گندم، جو، ذرت، سورگوم و باغات مرکبات، چای و جنگل های نواحی جنوبی دریای خزر- از آستارا در استان گیلان تا گنبد کاووس در استان گلستان- نمونه برداری به عمل آمد. از 80 جدایه شناسایی شده، 50 جدایه متعلق به گونه P. oryzaeو 30 جدایه متعلق به گونهP. griseaمی باشند که با مطالعه ویژگی های مورفولوژیکیشناسایی گردید. روابط فیلوژنتیکی دو گونهP. oryzaeو P. grisea براساس توالی نوکلئوتیدی نواحی ژنومی rDNA-ITS و MCM7مورد بررسی قرار گرفت و سه گروه اصلی در کلادوگرام حاصل شناسایی شدند. گروه اول شامل گونه های P. oryzae و P. grisea می باشند این دو گونه قارچی به لحاظ مورفولوژی و میزبان نیز متمایز می گردند. دو گروه دیگر گونه های مرتبط با Magnaporthe sensu lato را تشکیل می دهند. آزمون بیماری زایی گونه P. oryzae روی بوته های ذرت، سوروف، چسبک،گندیل، پاسپالوم و برنج انجام گرفت. نتایج بدست آمده نشان داد که گونه P. oryzae روی میزبان هایی ذرت، سوروف، چسبک به شدت بیماری زا می باشد. همچنین در مطالعه حاضر گیاهان ذرت، موز، مرغ، پاسپالوم و گندیل به عنوان میزبان های جدیدی برای عامل بیماری بلاست برنج در ایران معرفی می گردند.

    نماتدهای ریشه گرهی (Meloidogyne spp.)، یکی از عوامل محدودکننده کشت خیار در مزارع و گلخانه ها محسوب می شوند. تقویت خاک با کودهای حیوانی یا شیمیایی در کاهش میزان خسارت ناشی از این نماتدها موثر است. به همین منظور در سال 92-93 طی یک آزمایش اثرات سه سطح 25، 50 و 100 میلی گرم گوگرد/کیلوگرم خاک در زمان کشت و 45 روز قبل از کشت، در خاک سترون و غیرسترون در دو نوبت در گلخانه مورد مطالعه قرار گرفت. ابتدا دو عدد تخم یا لارو سن دوم نماتد/گرم خاک و سطوح گوگرد با خاک گلدان ها مخلوط، سپس در نیمی از گلدان ها بلافاصله و در نیمی دیگر پس از 45 روز بذور خیار (رقم نگین) کشت گردید. به منظور بررسی اثر تبخیر گوگرد بر جمعیت اولیه ی نماتد، نیمی از گلدان های 45 روزه درون کیسه های پلاستیکی قرار داده شد. نتایج آزمایش در دو نوبت کشت نشان داد که استفاده از 50 میلی گرم گوگرد در کیلوگرم در گلدان های حاوی خاک مزرعه، قرار داده شده داخل کیسه پلاستیکی به طور میانگین باعث افزایش 36% وزن خشک شاخساره و 114% وزن محصول نسبت به شاهد گردید. همچنین گوگرد 100 میلی گرم/کیلوگرم خاک جمعیت نهایی و فاکتور تولیدمثل را به ترتیب 70 و 69% نسبت به شاهد کاهش داد.

    ویروس کوتولگی سبزرد هندوانه (Watermelon chlorotic stunt virus، WmCSV) یکی از ویروس های بیماری زای مهم در هندوانه در مناطق جنوب و جنوب شرقی ایران است. این ویروس در سال های اخیر باعث خسارت های سنگینی به محصول هندوانه در مناطق فوق الذکر شده است. در این تحقیق، واکنش 11 رقم تجارتی هندوانه در برابر WmCSV با استفاده از روش مایه کوبی با سازه ی عفونت زای ویروس ارزیابی گردید. نتایج حاصل از این مطالعه نشان داد که ارقام PS (کریمسون سوییت)، Proseed، WLNOOL-201046 و Charlee (چارلستون گری) در برابر WmCSV کمتر حساس بوده و به ترتیب با 4/24، 17/52، 74/67 و 07/74 درصد آلودگی، بهترین واکنش را در برابر آلودگی به این ویروس نشان دادند. در صورتیکه سایر ارقام بعد از مایه کوبی با سازه عفونت زای ویروس 100% آلوده شدند. علاوه بر این، چهار رقم فوق الذکر از نظر شدت آلودگی با هم متفاوت بودند بطوریکه هندوانه کریمسون سوییت رقم PS علائم خفیف تری نسبت به سه رقم دیگر نشان داد. با تولید سازه همسانه ی عفونت زا WmCSV منبع نامحدودی از این ویروس برای ایجاد آلودگی و ارزیابی واکنش ارقام هندوانه و دیگر گیاهان میزبان ویروس فراهم شده است.

    در این تحقیق اثر تلفیقی و جداگانه قارچ Trichoderma harzianum و کود آلی ورمی کمپوست بر فعالیت نماتود ریشه گرهی Meloidogyne javanica در شرایط آزمایشگاهی و گلخانه ای ارزیابی شد. با استفاده از ویژگی های ریخت شناسی، نماتود ریشه گرهی و جدایه ی قارچ بر اساس آغازگرهای اختصاصی نیز مورد شناسایی قرار گرفتند. در شرایط آزمایشگاهی اثر عصاره کشت دو جدایه قارچی روی مرگ و میر لاروهای سن دو و نیز ممانعت از تفریخ تخم ها مورد آزمون قرار گرفت. مطالعات آزمایشگاهی نشان داد، بیشترین میزان مرگ و میر لاروها در غلظت پایه (100%) عصاره کشت بوده و میزان بازدارندگی تفریخ تخم در کاربرد جدایه قارچ اولی بیش از دیگری است. در گلخانه اثر کاربرد تلفیقی و جداگانه ورمی کمپوست و قارچ T. harzianum روی فاکتورهای رشدی گوجه فرنگی و شاخص های تولیدمثل نماتود در دو نوبت و در دو زمان برداشت 90 و 120 روز به صورت مستقل مورد بررسی قرار گرفت. استفاده هم زمان ورمی کمپوست و قارچ تریکودرما، علاوه بر مهار نماتود، عملکرد رشدی گیاه را نیز افزایش داده و این نشان از توانایی مهار نماتود ریشه گرهی با کاربرد تلفیقی دو عامل زیستی است.

    در این تحقیق، اثر Trichoderma harzianum BI در تحریک مقاومت القایی سیستمیک گیاهچه های گوجه فرنگی رقم ارلی اوربانا، بر اساس میزان فعالیت آنزیم های کیتیناز و گلوکاناز در ریشه گیاه اندازه گیری شد. ریشه های گیاهچه های گوجه فرنگی به وسیله قارچ و لارو سن دوم نماتود ریشه گرهی Meloidogyne javanica و قارچ عامل پژمردگی فوزاریومی Fusarium oxysporum f. sp. lycopersici به روش خیساندن خاک مایه زنی گردید. میزان فعالیت آنزیم های کیتیناز و گلوکاناز در ریشه گیاه از روز اول بعد از مایه زنی به مدت هفت روز با استفاده از روش اسپکتروفتومتری اندازه گیری شد. نتایج تغییرات در میزان فعالیت آنزیم های کیتیناز و گلوکاناز در هر تیمار به تنهایی و تیمار هم زمان نماتود- فوزاریوم در مقایسه با کاربرد قارچ آنتاگونیست قبل از آن تیمار نشان داد که قارچ تریکودرما سبب افزایش میزان فعالیت آنزیم کیتیناز شده اما فعالیت آنزیم گلوکاناز در گیاهچه هایی که هر بیمارگر را به تنهایی و همچنین هر دو بیمارگر را هم زمان دریافت کردند، اختلاف معنی داری نداشت.

    در بازدیدهای سال 1392 از فضاهای سبز و پارک های شهرستان بهشهر (استان مازندران) بیماری فیلودی پروانش مشاهده گردید. عامل بیماری فیلودی از پروانش با پیوند به پروانش و به وسیله سس به بادنجان انتقال داده شد. پروانش های آلوده در طبیعت و گیاهان مایه زنی شده در آزمون واکنش زنجیره ای پلیمراز (PCR) مستقیم با استفاده از جفت آغازگر P1/P7 و واکنش زنجیره ای پلیمراز دو مرحله ای با استفاده از جفت آغازگرهای P1/P7 و R16F2n/R16R2 واکنش مثبت نشان دادند و در آن ها قطعات مورد انتظار تکثیر شد. مقایسه چند شکلی طولی قطعات برشی (RFLP) مجازی نشان داد که فیتوپلاسمای همراه با فیلودی پروانش بهشهر متعلق به زیر گروه A در گروه افژولش شبدر (16SrVI) بود. آنالیز فیلوژنتیکی با استفاده از ترادف کامل ژن آر ان ای ریبوزومی 16Sنیز نتایج آنالیزRFLP مجازی را تایید کرد و نشان داد که فیتوپلاسمای عامل فیلودی پروانش بهشهر با ''Candidatus Phytoplasma trifolii''، فیتوپلاسمایی از زیر گروه 16SrVI-A طبقه بندی می شود. این اولین گزارش از بیماری فیلودی پروانش و تعیین ویژگی های فیتوپلاسمای همراه با آن در استان مازندران می باشد.

    شانکر باکتریایی درختان میوه هسته دار ناشی از Pseudomonas syringae pv. syringae پراکنش وسیعی در ایران پیدا کرده است (Abbasi et al 2013). به منظور ارزیابی تنوع جدایه ها و گونه های احتمالی دیگر در پاره ای از مناطق مرکزی کشور بازدیدهایی از باغ های میوه برخی از استان های مرکزی از جمله اصفهان، چهارمحال و بختیاری، قم و یزد در سال های 1393 و 1394 به عمل آمد.

    Phase variation is one of the adaptive mechanisms in fluorescent pseudomonads; this process caused by reversible genetic modification that generate subpopulation diversity with phenotypic differences that eventually lead to bacterial survival and their fitness under certain environmental conditions and is important in niche adaptation. In the present study¡ phase variation was observed in P. fluorescens strainB1 that carrying gene for phase variation. P. fluorescens B1 had two colony types¡ wild-type colony was small and opaque whereas the second type colony was larger in size and more fluorescent pigment production. These two types colony were tested for production of several enzymes such as protease¡ lipase¡ phosphatase and some metabolites production such as hydrogen cyanide¡ siderophore and auxin and for motility behaviors¡ biofilm formation and antagonistic activity against some phytopathogenic fungi. The results showed significant difference between the two types. Both of two types produced protease¡ lipase¡ phosphatase¡ but production of hydrogen cyanide in second type decreased strongly¡ in contrast production of siderophore and auxin in secondry type Increased. The second cell types was more motile than the wild-type and biofilm formation in wild-type was more than the secondry type. Antifungal activity of wild-type was higher than the secondary type. DNA fingerprint using BOX-PCR confirmed the identity of the second colony type with the parental isolate.

    Fusarium head blight (FHB) is economically one of the most important fungal diseases of wheat in the world. The aim of this research was to determine the antifungal metabolites of native Bacillus isolates. Bacillus strains isolated from soil of wheat and barley fields (Rural areas of southern Tehran, autumn 2012), and exhibited in vitro antagonistic activity against some Fusarium species isolated from infected wheat seeds (Wheat fields of Parsabad at Moghan, Ardebil Summer 2012). An attempt was made to partially purify and characterize the diffusible antifungal metabolite/s produced by the selected Bacillus strain in Nutrient broth medium. High Performance Liquid Chromatography (HPLC) of partially purified extract of the strain showed the presence of lipopeptide antibiotic iturin as a major peak that was comparable to that of standard iturin A (11.80 min) from Sigma–Aldrich. The structure was further confirmed by Fourier Transform-Infrared Spectrum (FTIR) and Liquid Chromatographic Mass Spectrometric (LCMS) analysis as iturin A. LCMS analysis also showed the presence of fengycin with iturin A. The genome of the selected isolate of Bacillus had shown 99/9 percent similarity by B. aryabhattai and the genome of the selected isolates of Fusarium had shown 99 percent similarity by F. graminearum. According to the results of this experiment, the antifungal effects of native Bacillus strains and also their effects in the biological control of wheat FHB disease were confirmed.

    Incidence of the root-knot nematode, Meloidogyne javanica and the crown gall bacterium, Rhizobium vitis, are increasing in different regions of Iran. In order to evaluate the response of different almond rootstocks to interaction of the root-knot nematode and the crown gall bacterium, three rootstocks viz. GN15, GF677 and Hybrid of peach´Shoorabi almond, inoculated with M. javanica and R. vitis in a factorial experiment with completely randomized design under greenhouse condition. Almond rootstocks were transferred to the main pots, and then the pots were inoculated by 9000 eggs and second stage juveniles of the nematode per pot. Three weeks after inoculation, the rootstocks were inoculated with a suspension of 108 of bacterial cells at the rate of 1% (V/W). Three month after the last inoculation, the growth indices of plant and nematode as well as pathogenicity indices of the bacterium were measured. The results showed that except GN15, other rootstocks were susceptible to nematode. The rootstock GN15 was susceptible to R. vitis. As a result, nematode infection predisposed GN15 to bacterial infection, but bacterial infection had no effect on nematode indices

    Field studies were conducted in Shahrekord and Lordegan towns of Chahar Mahal va Bakhtiari province, in two growing seasons from 2009 through 2011, to evaluate the effect of planting date, with and without a seed- treatment insecticide (Imidacloprid), on yield (biomass), yield components and control of maize Iranian mosaic disease (MIMD) with natural vectors in maize (Zea mays L.) Single cross 704 cultivar. The experimental design was a split plot arrangement of a randomized complete block with four replications. Main plots consisted of four planting dates, approximately 15 days apart from 10 May through 25 June. Sub plots consisted of a seed-treatment systemic insecticide application. In each of the two years, yields and yield components of plots planted in 10 May were reduced significantly by MIMD infection. Disease incidence in experimental plots varied between 2 and 44.5%. Planting dates of 5 June and 25 June resulted in the highest biomass and yield components in two years. Significant increase (P=0.01) in maize yield and yield components varying from 9.3% to 32 % was observed in plots treated with seed-treatment insecticide and the magnitude of increase depended on planting dates. Disease incidence was determined based on the percentage of plants (total plants per plot) exhibiting symptoms. Also, samples of plants from each of the experimental plots were collected and tested for presence of MIMV with TAS-ELISA method and the results were supported in both years of study. The highest populations of Laodelphax striatellus and Unkanodes tanasijevici were also observed when maize seedlings at the first planting date (10 May) were at three-leaf growth stage. Almost 3% plants in some plots were showed symptoms of infection other viral diseases such as Maize rough dwarf virus (MRDV), Wheat streak mosaic virus (WSMV) and Barley yellow dwarf virus (PAV and MAV) based on symptoms and ELISA test. In 2009, seven cultivars of maize, including: ZP677, ZP505, ZP684, Single cross 704, KONSOUR, KSC500 and TWC 647, were evaluated in view of their resistance level to MIMV, in Shahrekord and Lordegan regions. The highest and the lowest infection rates with MIMV belonged to Single cross 704 (57%) and KONSOUR (3.8%), respectively.

    High temperature oomycete Phytophthora parsiana is one of the causal agent of pistachio gummosis in Iran which originally was isolated from fig in Iran. Phytophthora hydropathica and P. irrigata two newly described high temperature species similar to P. parsiana were recently isolated from irrigation streams and some ornamental plants from USA .In the present study all three species produced chlamydospores in infected crown and root tissues of apricot.. No morphological differences were observed among species. Among woody plants inoculated Pistacia vera cv Sarakhs, almond and apricot were highly susceptible to all species. Pistacia mutica, P. khinjuk, walnut, fig and mango were susceptible to P. parsiana and P. hydropatica but P.vera cv Qazvini was infected only by P. hydropathical. Loquat ( Eriobotryae japonica) and Christ,thorn jupube (Zizyphus spina-christi) were not infected by all species. All annual herbaceous plant species used none were infectd with all species.

    Pyricularia oryzae is the causal agent of rice blast disease, the most destructive, and an important disease of rice worldwide. To identify of host plants for P. oryzae in the north of Iran, and separation of this species from P. grisea, the infected tissues were sampled from rice, corn, sorghum, barley and wheat cultivation regions, tea, citrus orchards, and forest in southern of Caspian Sea from Astara in Guilan province to Gonbad-e Qabus in Golestan province during spring, summers and fall of 2012 and 2013. Out of 80 isolated pathogen, 50 and 30 isolates were identified as P. oryzae and P. grisea on the basis of morphological characteristics respectively. In order to investigation of phylogenetic relationships of studied isolates, ITS region of ribosomal DNA and MCM7 were amplified. Three major clades in cladogram of ITS and MCM7 were identified. Species of the P. oryzae and P. grisea put in clade one. Species that stand into this group have phylogenetic, morphology and host differences. Two other group associated with Magnaporthe sensu lato. Pathogenicity test of P. oryzae species were applied on corn (susceptible line B73), barnyard grass, foxtail, rice, eleusine and paspalum in greenhouse. The results showed that this species cause blast and leaf spot symptoms on corn, barnyard-grass, and foxtail. In this study, corn, banana, bermuda grass, paspalum and eleusine are reported as new hosts for the fungus in Iran.

    Root-knot nematodes (Meloidogyne spp.) are one of the main pests of cucumber production in greenhouses and farms. Applications of chemical and organic fertilizers are effective in reducing the damages caused by these nematodes. For this purpose, during 2013-2014, in an experiment, effects of 25, 50 and 100 mg/kg of elemental sulfur at planting time and 45 days before planting, in sterilized and non-sterilized soil were studied in two turns in greenhouse. First, two eggs or second stage juveniles of the nematode/g of soil and sulfur levels were mixed with the soil. Then cucumber seeds cv. Negin were sown immediately in the half of the pots and in the other half after 45 days. In order to evaluate the effect of sulfur evaporates on the initial nematode population, one half of 45 days pots were placed in plastic bags. The results of two trails showed that sulfur 50 mg/kg of soil in 45 days pots, filled with non-sterilized soil and placed inside plastic bags caused 36% increase in the shoot dry weight and 114% in the yield, on average. In addition, sulfur 100 mg/kg of soil caused 70% and 69% decreases in the final population and reproduction factor of the nematode, respectively.

    Watermelon chlorotic stunt virus (WmCSV) is a bipartite begomovirus and one of the destructive watermelon viruses in the south and south-eastern Iran. WmCSV is responsible for huge losses of watermelon production in these regions. In this study, 11 commercial watermelon cultivars from the most current available cultivars were selected and used to evaluate their reaction against WmCSV by agroinoculation method with an infectious clone of the virus. Results showed that cultivar PS of Crimson Sweet watermelon, and Proseed, WLNOOL-201046 and Charlee cultivars of Charleston Gray watermelon are relatively less susceptible to WmCSV with infection rate of 24.4, 52.17, 67.74 and 74.07%, respectively, whereas watermelon plants of the rest of the cultivars were completely susceptible. In addition, four relatively resistant cultivars were differently reacted to WmCSV infection and among them, Proseed cultivar showed the mildest symptoms. The infectious construct is a rapid tool to evaluate the reaction of cultivars and can facilitate the identification of the resistant genetic sources against viral diseases.

    Integrated and separate application of Trichoderma harzianum and vermicompost on activity of the root-knot nematode, Meloidogyne javanica was examined under in vitro and green house conditions. The root-knot nematode identified using morphological characters and fungal isolate with the help of species-specific primers as well. Ability of two fungal isolates on egg hatching and mortality of M. javanica juveniles were tested under laboratory conditions. The results of the laboratory test indicated that the mortality of M. javanica juveniles was highest in the base concentration (100%), and inhibition of egg hatching of the first isolate was more than the other one. In the green house experiment, effects of integrated and separate application of vermicompost and T. harzianum on tomato plants growth factors and nematode reproduction indices, in two independent of 90 and 120 days harvesting time, were tested. Results showed that integrated using of vermicompost and the fungus, in addition of controlling nematode, increased plant growth and indicated a promising potential of nematode control by using combination of these two biological agents.

    Effect of the fungus Trichoderma harzianum BI on stimulation of induced systemic resistance in tomato seedling, cv. Early Urbana, based on activity of chitinase and glucanase enzymes in root was measured. Roots of seedling were inoculated by Trichoderma harzianum and Fusarium oxysporum f. sp. lycopersici fungi and second stage juveniles of the root-knot nematode, Meloidogyne javanica, by method of soil drenching. Changes of chitinase and glucanase enzymes in roots were daily measured by spectrophotometer from the first day of inoculation up to seven days. The results of this enzymes level in plants inoculated with nematode alone, plants inoculated with Fusarium alone and in simultaneous inoculation of nematode and Fusarium, as compared to the application T. harzianum BI before inoculation with nematode and Fusarium, indicate that T. harzianum BI significantly increased chitinase enzyme activity. While glucanase enzyme activity did not increase as a result of antagonist fungus.

    In 2013 surveys conducted in the green area and parks of Behshahr city (Mazandaran province of Iran), phyllody disease of periwinkle was observed. Agent of Behshahr periwinkle phyllody (BPP) was transmitted from naturally phyllody affected periwinkle to periwinkle by grafting and to eggplant via dodder inoculation. Naturally affected periwinkle plants and all inoculated plants reacted positively in direct PCR using P1/P7 primer pair and nested PCR using P1/P7 and R16F2n/R16R2 primer pairs and expected fragments were amplified. Virtual restriction fragment length polymorphism (RFLP) of nested PCR product sequence (1.2 kbp), percent homology and phylogenetic analysis of full length 16S rDNA sequence showed that BPP phytoplasma belongs to phytoplasma in the clover proliferation (16SrVI) phytoplasma group. The same analyses classified BPP phytoplasma with 'Candidatus Phytoplasma trifolli', a member of A subgroup in the 16SrVI group. This is the first report of periwinkle phytoplasma disease and characterization of associated phytoplasma in Mazandran province.

    Bacterial canker of stone fruit trees caused by Pseudomonas syringae pv. syringae has become widespread throughout most fruit growing areas of Iran (Abbasi et al 2013). To evaluate the diversity of the strains and possible species inciting the disease in the central areas of the country, surveys were made in 2014 and 2015 in some central provinces including Charmahal-and-Bakhtiary, Isfahan, Qum and Yazd. The twigs and shoots of apricot (Prunus armeniaca), peach (P. persica), plum (P. domestica) and sweet cherry (P. avium) with typical cankers which almost invariably were associated with exudation of gum were collected and brought to the laboratory. The affected shoots were thoroughly washed under tap water. The bark tissues of a segment encompassing a canker were excised and chopped with a sterile razor blade in a few drops of sterile distilled water.
