    به منظور بررسی تاثیر دگرآسیبی غلظت های مختلف عصاره های برگی گیاه اکالیپتوس (E. globulus) بر جوانه زنی و رشد گیاهچه بادمجان رقم محلی جهرم پژوهشی به صورت فاکتوریل در قالب طرح کاملا تصادفی با 3 تکرار انجام شد. تیمارها شامل 5 نوع عصاره آبی، متانولی، اتیل استاتی، استونی و بنزنی برگ اکالیپتوس و غلظت های مختلف هریک از این عصاره ها در 5 سطح 0، 25/1، 5/2، 5 و 10 گرم بر لیتر بودند. در پایان آزمایش صفات مربوط به جوانه زنی و رشد گیاهچه شامل درصد و سرعت جوانه زنی، طول ریشه چه و ساقه چه، وزن تر و خشک گیاهچه ها اندازه گیری شد. نتایج نشان داد که اثر بازدارندگی با غلظت عصاره ها در ارتباط بود به طوریکه غلظت های بیشتر اثر بازدارندگی قویتری بر جوانه زنی و رشدگیاهچه های بادمجان داشتند. نتایج همچنین نشان داد در تمام موارد عصاره اتیل استاتی بیشترین تاثیر منفی را بر جوانه زنی و رشد گیاهچه های بادمجان داشت در حالیکه در عصاره آبی و پس از آن عصاره استونی بیشترین درصد و سرعت جوانه زنی و بیشترین رشد گیاهچه مشاهده شد.

    در این مطالعه نقش جهش زایی با موتاژن EMS در بهبود تحمل به تنش خشکی در برنج مورد بررسی قرار گرفت. لاین های موتانت نسل M2 در مرحله جوانه زنی و گیاهچه ای در شرایط تنش اسمزی با PEG (8/0- مگاپاسکال) ارزیابی شدند که منجر به جداسازی 9 لاین موتانت متحمل به خشکی گردید. این لاین ها به همراه رقم مادری در مرحله گیاه کامل در شرایط مزرعه با اعمال آبیاری محدود نیز مورد ارزیابی قرار گرفتند که بر اساس شاخص تحمل به تنش (STI) دو لاین متحمل به خشکی به نام های MT90 و MT149 شناسایی گردیدند. برای بررسی سطح تنوع ژنتیکی حاصل از تیمار جهش در لاین های موتانت، از نشانگرهای ISSR استفاده شد که 4/69 درصد آنها چندشکلی بین رقم مادری و لاین های موتانت را نشان دادند. تنوع ژنتیکی حاصل از جهش در ژنوتیپ های مورد مطالعه معادل 5/23 درصد برآورد شد و تجزیه خوشه ایبا روش UPGMA آنها را به سه گروه عمده تفکیک نمود که گروه های حاوی لاین های موتانت نسبت به گروه حاوی رقم مادری در شرایط تنش خشکی هم عملکرد بالاتر و هم شاخص تحمل به تنش بالاتری را نشان دادند. بر اساس نتایج، می توان لاین MT149 را به عنوان لاین متحمل به خشکی در هر دو مرحله گیاهچه ای و گیاه بالغ معرفی نمود. می توان نتیجه گیری کرد که جهش القایی EMS تاثیر مطلوبی در ایجاد لاین های موتانت متحمل به خشکی داشته و از نشانگرهای ISSR می توان برای جداسازی لاین های موتانت متحمل به خشکی در جمعیت های حاصل از جهش در برنج استفاده کرد.

    در این تحقیق اثر سالیسیلیک اسید به منظور کاهش بیماری زایی قارچ رایزوکتونیا سولانی بر روی فعالیت آنزیم های پراکسیداز، کاتالاز و پلی فنل اکسیداز در رقم سیب زمینی آگریا در شرایط گلخانه ای بررسی گردید. دو هفته قبل از کاشت ریز غده های سیب زمینی، خاک استریل گلدان ها در قالب یک طرح کاملا تصادفی با قارچ ریزوکتونیا سولانی آلوده شدند. تیمار سالسیلیک اسید در مرحله 8-7 برگی با غلظت های صفر 0 (شاهد یا کنترل)، 2/0 و 5/0 میلی مولار هر هفته (مجموعا چهار هفته) مطابق طرح آزمایش اعمال گردید. بعد از اتمام تیمار اسید سالیسیلیک، میزان فعالیت آنزیم های پراکسیداز، کاتالاز و پلی فنل اکسیداز در برگ تازه گیاهان سالم و آلوده سیب زمینی اندازه گیری گردید. نتایج نشان داد که با افزایش میزان اسید سالیسیلیک، میزان فعالیت آنزیم های پراکسیداز و کاتالاز در گیاهان سیب زمینی سالم و آلوده یک روند کاهشی دارد. در صورتی که فعالیت آنزیم پلی فنل اکسیداز در این شرایط یک روند افزایشی را نشان می دهد. به نظر می رسد به علت کاهش فعالیت آنزیم های کاتالاز و پراکسیداز و تولید انواع اکسیژن فعال (H2O2 ، OH- ، O2) که برای قارچ سمی است، گیاه سیب زمینی با قارچ رایزوکتونیا سولانی مقابله می کند. همچنین ممکن است به علت افزایش فعالیت آنزیم پلی فنل اکسیداز، مکانیسم های دفاعی از جمله لیگنینی شدن و ایجاد پل های عرضی هیدروکسی پرولین در دیواره سلول های ریشه باعث مقاوم سازی بافت های ریشه گیاه سیب زمینی در برابر قارچ رایزوکتونیا سولانی گردیده است.

    TRR14، پروتئین کوچکی است که در کلروپلاست جای دارد. این پروتئین نخستین بار در غربالگری گیاهچه های آرابیدوپسیس مقاوم به ترهالوز (Trehalose Resistant) شناخته شده است. هدف از این تحقیق، بررسی فیزیولوژیکی و مولکولی گیاهان تراریخت شده با ژن TRR14 در مقایسه با گیاهچه های وحشی آرابیدوپسیس در تنش شوری است. به این منظور، بذر های گیاه وحشی آرابیدوپسیس و تراریخت شده با ژن TRR14 در محیط کشت MS حاوی غلظت های مختلف نمک (0، 75، 100 و 150 میلی مولار ) کشت شدند. نتایج نشان داد که در تیمار شوری درصد جوانه زنی و فعالیت آنزیم ترهالاز در گیاهچه های تراریخت بالاتر از گیاهچه وحشی است. همچنین این نتایج نشان داد که با افزایش غلظت نمک میزان آنتوسیانین در گیاهچه وحشی به طور قابل توجهی در مقایسه با گیاهچه تراریخت افزایش یافت. نتایج حاضر نشان داد که در تیمار 75 میلی مولار نمک، غلظت سدیم در گیاهچه وحشی به طور قابل ملاحظه ای افزایش می یابد. سنجش پتاسیم نیز نشان داد که میزان آن در گیاهچه وحشی رشد یافته بر روی محیط MS ، تقریبا سه برابر گیاهچه تراریخت است. اما تیمار 75 میلی مولار نمک، سبب کاهش میزان پتاسیم در هر دو نوع گیاهچه ها شد. همچنین با افزایش میزان نمک شکل و اندازه سلول های اپیدرمی تغییر یافته و تعداد روزنه در واحد سطح در گیاهچه های وحشی افزایش یافت، در حالیکه این فاکتور در گیاهچه تراریخت تغییری نشان نداد. بررسی الگوی بیان ژن نیز نشان داد که تیمار شوری، سبب افزایش بیان ژن های ترهالاز و TRR14 شد.

    تنش های غیر زیستی بازدهی گیاه پالایی را به عنوان فن آوری سازگار با محیط زیست و مقرون به صرفه در پالایش آلودگی های خاک کاهش میدهد. هدف از این تحقیق بررسی اثر تنش آلودگی نفتی و مسن خاک (10%وزنی: غلظت کل هیدروکربن های نفتی) طی فرایند گیاه پالایی بر دو گونه گراس (جو دوسر وحشی و جو زراعی) بود. به علاوه اثر تلقیح ریزوباکتری های تجزیه کننده نفت بر تحریک رشد و کاهش سمیت آلاینده های نفتی در خاک آلوده مورد بررسی قرار گرفت. بر اساس نتایج آلودگی نفتی خاک در هر دو گیاه باعث کاهش شاخصهای رشد، رنگیزه های فتوسنتزی و افزایش مقدار پراکسید هیدروژن (H2O2)، MDA (مالوندیآلدهید) و آنتیاکسیدانهای آنزیمی (کاتالاز و پراکسیداز) و همچنین افزایش نسخه های ژنهای NCDE و COR2 شد که بیانگر بروز تنش اکسیداتیو در اثر آلودگی نفتی، مشابه با دیگر تنشهای غیر زنده متداول بود. افزایش نسبت کلرفیل a/b و کاهش شدیدتر وزن خشک اندام هوایی در جو دوسر وحشی نسبت به جو زراعی نشان داد جو زراعی در برابر تنش آلودگی نفتی خاک متحمل تر بود. تلقیح باکتریایی باعث کاهش سمیت نفت خام و بهبود رشد گیاه به خصوص در جو دوسر وحشی گردید.

    آنتی بادی های مونوکلونال هترودایمرهای متشکل از چهار زنجیره پپتیدی، دو زنجیره سبک (L) یکسان و دو زنجیره سنگین (H) یکسان هستند. بیان همزمان مقدار برابر ژن های رمز کننده برای تولید پایدار و گردایش به صورت یک آنتی بادی کارکردی ضروری است. به منظور بیان هم زمان چندین ژن، سیستم های مختلفی از جمله وکتورهای مبتنی بر جایگاه میانی ورود ریبوزوم (IRES) توسعه یافته اند. در مطالعه حاضر، یک حامل دی سیسترونیک برای بیان هم زمان ژن های رمز کننده زنجیره های سنگین و سبک آنتی بادی ضد سرطان سینه trastuzumab (Herceptin®) با استفاده از توالی IRES از فاکتور پروتئین شوک حرارتی-1 Nicotiana tabacum (NtHSF1) ساخته شد. کارایی دستواره در Nicotina tabacum به روش آگرواینفیلتراسیون ارزیابی شد. بیان دی سیسترونیک تراژن ها در سطح رونوشت RNA با آنالیز RT-PCR نیمه کمی مشخص شد. آنالیز وسترن بلات بیان کارآمد فرم تترامر trastuzumab را در بافت برگی توتون مورد تایید قرار داد و محتوی آنتی بادی 7 روز پس از اینفیلتراسیون 44/0 درصد پروتئین کل محلول برآورد شد. نتایج مطالعه حاضر نشان دهنده کارایی عناصر IRES گیاهی برای بیان دی سیسترونیک پروتئین های هترومولتی مر در گیاهان است.

    پپلی گالاکتورونازها (GH28)، عمدتا در گیاهان سبب هیدرولیز سوبسترای پکتینی می شوند. پلی گالاکتورونازها به طور گسترده در گیاهان عالی و میکروارگانیسم ها موجود می باشند. استخراج پلی گالاکتوروناز از منابع قارچی، به علت بازدهی بالای تولید، کاربرد فراوان دارد. در این پژوهش، قارچ M. phaseolina، عامل بیماری پوسیدگی ذغالی، برای تولید پلی گالاکتوروناز در محیط تولید آنزیم کشت و عصاره قارچی جدا گردید. بیشترین فعالیت پلی گالاکتورونازی محلول آنزیمی، در دمای 60 درجه سانتی گراد بود. این آنزیم از پایداری دمایی و اسیدیته بالایی برخوردار است به طوریکه، نیمه عمرحرارتی آن در دمای 50 و 70 درجه سانتی گراد به-ترتیب حدود 627 و 345 دقیقه و میزان تغییرات انرژی آزاد غیر فعال سازی حرارتی آنزیم در دمای 70 درجه سانتی گراد تقریبا معادل 114 کیلو ژول بر مول بود که حاکی از پتانسیل بالای این آنزیم جهت کاربرد در صنایع می باشد. این آنزیم از پایداری بسیار بالایی در شرایط بسیار اسیدی و قلیایی برخوردار است. بررسی و مقایسه خصوصیات بیوشیمیایی پلی گالاکتورونازهای منابع مختلف قارچی با استفاده از برنامه ProtParam نیز پلی گالاکتورونازهای M. phaseolina را در گروه پایدارترین پلی گالاکتورونازهای قارچی قرار داد.

    ترانسفرزوم ها وزیکول های بسیار تغییر شکل پذیری هستند که به دلیل خصوصیات الاستومکانیکی فوق العاده خود قادرند از منافذی بسیار کوچک تر از اندازه خودشان عبور کرده و نقش دارورسانی به پوست را ایفا نمایند. در این مطالعه ترانسفرزوم های حاوی هورمون رشد با استفاده از دو فرمول متشکل از فسفولیپید و سدیم داکسی کولات (f1) و سدیم لوریل سولفات (f2) به عنوان سورفاکتانت ساخته شدند. روش های مختلفی برای به دست آوردن ترانسفرزوم با اندازه مشخص وجود دارد. یکی از این روش ها استفاده از تکنیک سونیکاسیون برای تنظیم ابعاد آنهاست. در این تحقیق ترانسفرزوم های حاوی هورمون رشد مورد بررسی قرار گرفته و در راستای تغییر ابعاد، ترانسفرزوم ها تحت تاثیر سونیکاسیون به روش حمام و پروب قرارداده شدند. در هر دو حالت در سه دوره زمانی ابعاد ذرات، پراکندگی ذرات، پتانسیل زتا و فعالیت زیستی هورمون رشد موجود در ترانسفرزوم ها اندازه گیری گردید. نتایج نشان داد که در کلیه زمان های مورد بررسی فعالیت هورمون رشد در سونیکاسیون پروب در هر دو ترانسفرزوم نسبت به سونیکاسیون حمام کاهش بیشتری نشان می دهد لذا، اندازه وزیکول ها و پتانسیل زتا در آزمایش انجام شده توسط سونیکاسیون پروب تعیین شد. میانگین سایز ذرات در وزیکول های f1 از 6/242 تا 6/190 و در وزیکول های f2 از 6/159 تا 37/81 است. پتانسیل زتا در ترانسفرزوم f1 منفی تر از f2 می باشد.

    In order to investigate the allelopathic effects of leaf extracts of E. globulus on germination and seedling growth of eggplant cv. Mahali Jahrom an experiment was carried out using a completely randomized design arranged in a factorial scheme with 3 replicates. Treatments included 5 types of extracts including aqueous, methanolic, ethyl acetate, acetonic and benzene and different concentrations of extract at 5 level (0, 1.25, 2.5, 5 and 10 gram per liter). At the end of experiment germination and seedling growth characters including germination percentage, germination rate, shoot length, root length, fresh and dry weights of seedling were measured. Result indicated that the inhibitory effect was proportional to the concentrations of the extracts and higher concentration had the strongest inhibitory effect on germination and seedling growth. Results also revealed that in all cases ethyl acetate had most negative effect on germination and seedling growth of eggplant whereas aqueous extract followed by acetonic extract caused highest germination percentage, germination rate and seedling growth.

    In this research, EMS mutagen was applied to create genetic variability in rice (cv. Neda) and its mutagenesis effect was investigated in drought tolerance improvement. M2 mutants were evaluated at germinating and seedling stages under osmotic stress of PEG (-0.8 MPa) which resulted to isolating 9 drought tolerant lines. The lines along with their original cultivar also were evaluated in field under restricted irrigation regime which two mutants viz. MT90 and MT149 based on stress tolerance index (STI) were identified as drought tolerant lines. ISSR markers were used to assess genetic variability induced by mutation in mutant lines, 69.4 percent of which detected polymorphism between original cultivar and mutant lines. Mutation-induced genetic variation was estimated equal to 23.5 percent in the studied genotypes and cluster analysis using UPGMA method differentiated them into three main classes which mutants-containing groups produced higher yield and STI relative to original cultivar group under drought stress. Based on the results, MT149 mutant can be introduced as a drought tolerant line at both seedling and whole plant stages. It can be concluded that EMS-induced mutation had a desire effect in developing drought tolerant mutants and ISSR markers can be applied for differentiating drought tolerant mutants in rice mutation derived populations.

    This study was carried out to evaluate the effects of salicylic acid on the peroxidase, catalase and polyphenoloxidase activities in potato Agria cultivar for reducing the pathogenesis of Rhizoctonia solani under greenhouse conditions. The potato minitubers (without any infection) were transferred to pots containing soil steril for growth. The sterilized soil of pots had infected by R. solani two weeks before planting. The 4 weeks-old plants (7-8 leaves) have been treated by 0.1, 0.2, 0.5 and 1 mM Salicylic acid every week for 4 times. Results showed that as salicylic acid increased, polyphenoloxidase activity were increased in potato plants (healthy and infected) whereas were decreased peroxidase and catalase activities under the same conditions. On the other hand, it seems that due to decreased activites of catalase and peroxidase and production of reactive oxygen species (H2O2, OH-, O2-) which are toxic for fungi, so potato plants can be resistance to R. solani. Also, it seems that to increase polyphenoloxidase activity, defensive mechanisms such as lignification and build bridges transverse hydroxyproline in root cell walls cause strengthen in the root tissues, so potato plants can be resistance against R. solani.

    TRR14 is a small protein which is located in chloroplast. This protein was first identified in screening of trehalose resistant (TRR14) Arabidopsis seedlings. The aim of present study was physiological and molecular investigation of TRR14 re-transformed (T17) seedlings under salt stress, compared to wild type Arabidopsis seedlings (WT). For this purpose, WT and T17 seeds were grown on MS medium complemented with 0, 75, 100 and 150 mM NaCl. The results showed that under salt stress, the germination rate and trehalase activity in T17 seedlings were higher than WT. Meanwhile the results showed that anthocyanin level in WT seedlings was higher than T17 under salt stress. The Na level was highly increased in WT seedlings under 75 mM NaCl treatment, compared to T17 seedlings. The K measurements revealed that the K level in T17 seedling was 3 times more than WT seedling when grown on MS medium. But K level was decreased under 75 mM NaCl treatment in both WT and T17 seedlings. On the other hand, by adding NaCl to medium culture, the shape and size of epidermal cells were changed in both WT and T17 seedlings. While stomata numbers increased in WT leaves under NaCl treatment, it remained unchanged in T17 seedlings. The gene expression pattern showed that trehalase and TRR14 expression were increased under salt stress.

    Phytoremediation efficiency -a cost effective and environmentally sound technology to clean up contaminated soil - is reduced considering abiotic stresses. The research objective was to determine the effects of aged petroleum contaminated soil (10% W/W: Total petroleum Hydrocarbons) on two grasses specious (wild oat and barley) during the phytoremediation process. In addition the effects of petroleum degrading rhizobacteria inoculation were evaluated on plant growth promotion and decreasing toxicity of petroleum hydrocarbons. Results demonstrated that petroleum contaminated soil reduced growth, photosynthetic pigments and increased hydrogen peroxide (H2O2), MDA (malondialdehyde), enzymatic antioxidants (catalase and peroxidase) and NCDE and COR2 gene transcripts that indicate oxidative stress happened because of petroleum pollution similar to other common abiotic stress. a/b chlorophyll ratio enhancement and more decrease in shoot dry weight in wild oat showed barley was more tolerate to petroleum pollution. Bacterial inoculation reduced petroleum contamination toxicity and improved plant growth especially in wild oat.

    Monoclonal antibodies are heterotetramers consisting of four peptide chains—two identical light (L) chains and two identical heavy (H) chains and simultaneous equimolar expression of the corresponding genes is required to stable produce and assembly into a functional antibody. In order to co-express of multiple genes, different systems have been developed such as internal ribosome entry site (IRES) based vectors. IRES sequences are genetic elements that can drive cap-independent translation initiation of cistrons located downstream of the element. In this study, we constructed a bicsitronic vector for co-expression of genes encoding the H and L chains of the anti-breast cancer antibody trastuzumab (Herceptin®) using an IRES sequence derived from the Nicotiana tabacum heat-shock factor 1 (NtHSF1). The effectiveness of construction was evaluated in Nicotina tabacum using agroinfiltration method. Bisictronic expression of transgenes at the RNA transcription level was detected by semi-quantitative RT-PCR analysis. Western blot analyses revealed the efficient accumulation of tetrameric form of trastuzumab in tobacco leaf tissues and antibody content were ca. 0.44% of total soluble protein within 7 days after infiltration. The results of this study demonstrate the efficacy of a plant derived IRES element for bicistronic expression of heteromultimeric proteins.

    Polygalacturonases (GH28) hydrolyze the pectic substances, present mostly in plants. Polygalacturonases are widely distributed among some bacteria, fungi, and higher plants. Microbial polygalacturonases, mainly produced by fungi, are utilized in many large-scale industrial processes. Production of a polygalacturonase from a fungal phytopathogen, M. phaseolina was evaluated in this study. M. phaseolina was cultured in the production medium. The crude extract was separated from fungal cells. The highest enzyme activity was observed at 60 °C. This enzyme had a high stability at wide ranges of pH and temperature and exhibited a t1/2 of 400 min at 70 °C. The value for free energy of the heat inactivation at the temperature of 70 °C was about 108 kJ, suggesting its potential to be used as an industrial enzyme. Analysis of biochemical characteristics of polygalacturonase from different fungi using ProtParam demonstrated that the M. phaseolina polygalacturonase was amongst the most stable fungal polygalacturonases.

    The transfersomes are ultraflexible vesicles¡ which due to their elastomechanic properties are able to penetrate into pores smaller than their own size¡ and thereby have the potential to deliver drugs to the skin. In this study growth hormone transfersomes were constructed from two formula made of phospholipids¡ sodium deoxycholate (f1) and sodium lauryl sulphate (f2) as surfactants. There are different ways of obtaining transfersomes with a specified size range. One of the methods is the use of sonication to regulate their size and dimensions. Accordingly¡ in order to study transfersomes carrying the growth hormone¡ the vesicles were sonicated using both the probe and bath methods. In each case¡ dimensions¡ dispersity¡ the zeta potential of transfersomes and biological activity of the growth hormone were measured during three different time cycles of sonication. The results showed that during investigation of all the different time periods¡ growth hormone activity was found to decrease further following probe sonication when compared to that of bath sonication. Therefore¡ vesicle size and zeta potential were measured in the probe sonication tests. The mean size was found to vary between 242.6 to 190.6 and 81.37 to 159.6¡ for the f1 and f2 vesicles¡ respectively. The zeta potential of the f1 transfersomes was more negative than that of f2.
