صفحات 1-19ویروس هپاتیت C، یکی از علل اصلی سیروز و سرطان کبد در سراسر جهان است. قابل قبول ترین رژیم دارویی در درمان بیماران مبتلا به عفونت ویروس هپاتیت C استفاده از درمان ترکیبی پلیاتیلن گلیکول- اینترفرون به همراه ریباورین است.
مطالعات متعددی نشان داده است که به دنبال استفاده از رژیم دارویی فوق به عنوان رژیم دارویی استاندارد، درصد نسبتا کمی از مبتلایان به ژنوتیپ های 1 و 4 ویروس به حالت پاسخ های ویروسی پایدار (SVR) (حذف دائم ویروس) می رسند (42-46 درصد) این در حالیست که میزان پاسخ های ویروسی پایدار در مورد ژنوتیپ های 2 و 3 ویروس به مراتب بهتر است (76-82 درصد).
در مجموع، زمان مناسب برای رسیدن به پاسخ های ویروسی پایدار در مورد ژنوتیپ های 1 و 4 یک سال است در حالی که طول این مدت در ژنوتیپ های 2 و 3 حدود 6 ماه است.
از سال 2011 استفاده از داروهای ضد ویروسی با عملکرد مستقیم علیه ویروس هپاتیت C برای درمان بالینی آغاز شد که به طور مستقیم پروتین های کد شده توسط ویروس را مورد هدف قرار می دهند.
نسل اول مهارکننده های «پروتئازHCV NS3/4A » مانند تلاپریویر (Telaprevir) سبب افزایش پاسخ ویروسی پایدار در بیماران مبتلا به ویروس هپاتیت C شد.
با وجود پتانسیل بالقوه این داروها، رژیم درمانی مذکور با عوارض جانبی شدید همراه است. برای بیماران مبتلا به ویروس هپاتیت C که به درمان سخت جواب می دهند، گزینه های درمانی بیشتری مورد نیاز است.
مطالعات بیشتر برای اثر بخشی و کاهش آثارجانبی داروها نگاه ها را به سمت دنیای مولکولی و میکروRNAها سوق داد. میکروRNAها، نقش مهمی در استقرار عفونت ویروس هپاتیت C که در تکثیر ویروس موثر است.
در مطالعه مروری حاضر تلاش شده است که از یک طرف عفونت ویروس هپاتیت C، میکروRNAها، بیوژنز آن ها و سایر موارد بررسی شود و از طرف دیگر نقش میکروRNAها در چرخه تکثیر HCV ونیز ژنوتیپ های مختلف ویروس مورد بحث قرار می گیرد.کلیدواژگان: ویروس هپاتیت C، میکروRNAهای کبدی، داروهای جدید، ژنوتیپ های ویروس هپاتیت C - مقاله پژوهشی

صفحات 21-35هدفمرفین و مشتقات آن به دلیل ویژگی های قوی ضد دردشان در مصارف پزشکی بسیار شناخته شده هستند. اما استفاده پزشکی از آن ها به دلیل ایجاد مقاومت و وابستگی پس از استفاده طولانی به شدت محدود است. گیرنده های افیونی مو در سلول ها بر اثر مرفین دچار اندوسیتوز نمی شوند اما مشابه های انکفالین مانند DAMGO باعث اندوسیتوز سریع گیرنده ها می شوند. استفاده مکرر از داروهای مخدر باعث تغییر بیان ژن ها می شود. مطالعه کنونی به بررسی اثر تیمار طولانی مدت افیون ها بر میزان بیان ژن های Dnm1L و Rab22a پرداخته است.
مواد و روشها: سلول های HEK293 که گیرنده افیونی Mu را بیان می نمایند، با مرفین، DAMGO و متادون به طور جداگانه و طولانی مدت تیمار شد. mRNA از عصاره سلولی استخراج شد و پس از ساخت cDNA، میزان بیان ژن های Dnm1L و Rab22a نسبت به نمونه کنترل به روش Q-RT-PCR مقایسه شد. نتایج با نرم افزار Graph Pad تجزیه و تحلیل آماری شد.نتایجبررسی منحنی های به دست آمده از Q-RT-PCR نشان داد در اثر تیمار طولانی مدت با مرفین در مقایسه با نمونه کنترل بیان ژن Dnm1L 37/2 برابر افزایش یافته و بیان ژن Rab22a 39/0 برابر کاهش یافته است. در حالی که میزان بیان Dnm1L در اثر تیمار با متادون و DAMGO تغییر معنی داری نشان نداده است، بیان ژن Rab22a به ترتیب 41/0 و 45/0 درصد کاهش نشان داده است.
نتیجه گیری: ژن Dnm1L می تواند در مسیرهای مقاومت سلولی ایجاد شده در اثر مرفین که باعث تفاوت آن نسبت به سایر آگونیست ها می شود دخالت داشته باشد.کلیدواژگان: مرفین، Dnm1L، Rab22a، مقاومت، وابستگی -
صفحات 37-51هدفکروسین از ترکیبات عصاره زعفران است که خواص ضد سرطانی آن روی انواعی از سرطان ها به ویژه سرطان پستان در مطالعات اخیر نشان داده شده است. با این حال، اطلاعات بسیار کمی از مکانیسم اثر این ترکیب موجود است. مطالعات قبلی، القای آپوپتوز در سایر سلول های سرطانی تحت درمان با زعفران یا کروسین را نشان داده است. در این پژوهش با هدف بررسی اثر کروسین بر رده سلولی MDA-MB-468 سرطان پستان، در نظر است تا بیان و شکستن کاسپاز 9، پیرایش و بیان پروتئین XBP1 و تجمع پروتئین LC3-II در تیمار شده با این ترکیب، بررسی شود.
مواد و روشها: سنجش MTT برای بررسی اثر سمی کروسین بر رده سلولی نامبرده استفاده شد. سپس پیرایش ژن XBP1 در سطح mRNA و بیان پروتئین پیرایش شده (XBP1s) به ترتیب با RT-PCR و وسترن بلات بررسی شد. بیان کاسپاز 9، کاسپاز شکسته شده و تجمع LC3-II نیز با روش وسترن بلات بررسی شدند. میزان بیان این پارامترها با استفاده از نرم افزار Image J به صورت کمی گزارش شد.نتایجکروسین به صورت وابسته به دوز و زمان، موجب مرگ رده سلولی MDA-MB-468 شد. تیمار با کروسین، تغییر قابل توجهی در فعال شدن کاسپاز 9 و شکستن آن، در نتیجه افزایش نسبت Cleaved-Cas9/Cas9 داشت. پیرایش ژن XBP1 نیز پس از تیمار افزایش یافت. همچنین افزایش معنیدار بیان پروتئین پیرایش شده XBP1 (XBP1s) و تجمع پروتئین LC3-II و افزایش نسبت LC3-II/LC3-I در این سلولها مشاهده شد.نتیجه گیرینتایج نشان داد که این رده از سلولهای سرطان پستان پس از تیمار با کروسین دچار آپوپتوز شد. با توجه به تغییرات نشانگرهای مسیرهای UPR و اتوفاژی، احتمالا این دو مسیر نیز در پیشبرد سلول به سمت مرگ سلولی و تنظیمات درون سلولی، نقش دارند.کلیدواژگان: سرطان پستان، کاسپاز 9 شکسته شده، تنش شبکه اندوپلاسمی، پیرایش XBP1، تجمع LC3-II -
صفحات 53-66هدفایست قلبی یکی از شایع ترین بیماری های قلبی با نرخ مرگ و میر بالا و هزینه های زیاد محسوب می شود. با وجود مراقبت های ویژه، هنوز هم نرخ بقای یک ساله بیماران دارای ایست قلبی مرحله آخر کمتر از 1 درصد است. در مطالعات مختلفی ارتباط RNA های غیر کدکننده با انواع بیماری های قلبی نشان داده شده است.مواد و روش هادر این مطالعه، مکانیسم عملکردی RNA غیرکدکننده BACE1-AS با استفاده از بررسی بیان ژن، تکثیر سلولی و مرگ سلولی برنامه ریزی شده مطالعه شد که در مطالعات قبلی محققان حاضر در بیماران دارای ایست قلبی افزایش بیان نشان داده بود.نتایجنتایج نشان داد که مهار BACE-1AS باعث کاهش بیان مهارکننده های سایکلین چرخه سلولی یعنی CDKN1A، CDKN1B، CDKN1C، CDKN2A، CDKN2B به غیر از CDKN2C و CDKN2D می شود. همچنین بیش بیان BACE1-AS در سلول های HAOEC باعث افزایش بیان این ژن ها شده است که در نتیجه نشان دهنده تاثیر BACE1-AS روی چرخه سلولی است. مهار BACE1-AS، بیان ژن BCL2 را افزایش و بیان BAX را کاهش می دهد و بالعکس بیش بیان BACE1-AS باعث کاهش بیان BCL2 و افزایش بیان BAX می شود. مهار BACE1-AS تغییری در تکثیر سلول های HAOEC ایجاد نکرد ولی با بیش بیان BACE1-AS تعداد سلول های اندوتلیال به صورت معنی داری کاهش می یابد و باعث افزایش مرگ سلولی برنامه ریزی شده نسبت به نمونه کنترل (سلول های القا شده با ناقل خالی) می شود.نتیجه گیریاین نتایج تاثیر و نقش BACE1-AS در القای مرگ سلولی برنامه ریزی شده و درنتیجه بیماری زایی در ایست قلبی را نشان می دهد که می توان از BACE1-AS به عنوان یک نشانگر زیستی احتمالی و هدف در درمان ایست قلبی استفاده کرد.کلیدواژگان: ایست قلبی، نشانگر زیستی، RNA غیر کد کننده

صفحات 67-76هدفکریپتورکیدیسم شایعترین نقص دستگاه تناسلی مذکر و شایعترین بیماری غدد درون ریز پسران هنگام تولد است. یکی از عوامل آن ترشح ناکافی هورمون آنتی مولرین از سلولهای سرتولی بیضه قبل از تولد است. اخیرا نقش این هورمون و هورمون اینهیبین B در باروری به اثبات رسیده است و به عنوان شاخصی برای ارزیابی عملکرد سلولهای سرتولی شناخته میشود. هدف این مطالعه بررسی رابطه بین هورمون آنتی مولرین، اینهیبینB، تستوسترون و شاخصهای مایع منی در بیماران کریپتوکیدیسم پس از بلوغ است.
مواد و روشها: داده های این مطالعه مقطعی از نمونه خون و مایع منی 20 بیمار دچار سابقه کریپتوکیدیسم یک طرفه در محدوده سنی 20 تا 40 سال مراجعه کننده به بخش اورولوژی بیمارستان دکتر شریعتی تهران بهدست آمد. همچنین غلظت سرمی هورمونهای آنتی مولرین، تستوسترون و اینهیبین B توسط روش الایزا ارزیابی شد. ارزیابی پارامترهای مایع منی توسط نرم افزار CASA (Computer Aided Semen Analysis 6.5.0) صورت گرفت. برای بررسی ارتباط بین متغیرها از آزمون آماری همبستگی پیرسون استفاده شد.نتایجبین تحرک اسپرم با تعداد اسپرم (086/ =r، 01/0>P)، تحرک کلی اسپرم با درصد حرکت سریع (97/0=r، 01/0>P)، تعداد اسپرم با درصد حرکت سریع (9/0=r، 01/0>P)، تعداد گلبولهای سفید مایع منی با تعداد اسپرم (63/0=r، 05/0>P)، تعداد گلبولهای سفید مایع منی با تحرک (66/0=r، 05/0>P)، تعداد گلبولهای سفید مایع منی با حرکت سریع (77/0=r، 01/0>P) و سطح تستوسترون با آنتی مولرین سرمی (6/0=r، 01/0>P) ارتباط مثبت معنیداری وجود داشت.
نتیجه گیری: غلظت سرمی هومونهای آنتی مولرین، تستوسترون و اینهیبین B در افراد کریپتورکیدیسم از محدوده طبیعی پایینتر است. با وجود این که بین هورمون تستوسترون و آنتی مولرین رابطه مثبت معنیداری بهدست آمد اما هیچ یک از هورمونهای مورد مطالعه ارتباط معنیداری با شاخصهای مایع منی نشان نداد.کلیدواژگان: هورمون آنتی مولرین، اینهیبین B، تستوسترون، کریپتورکیدیسم

Pages 1-19Hepatitis C virus (HCV) is a common cause of liver cirrhosis and hepatocellular carcinoma (HCC) worldwide. The combination of ribavirin and peg-interferon, as standard treatment for HCV infection, seems very promising. Many studies have revealed that despite following standard HCV treatment, a high proportion of HCV genotypes 1 and 4 poorly attain (42% to 46%) the SVR condition, whereas it is somehow easier for HCV genotypes 2 and 3 (76%-82%). Overall, genotypes 1 and 4 antiviral therapies must be continued up to one year to achieve SVR, whereas in individuals infected with genotypes 2 and 3 must continue therapy for six months. Since 2011, direct-acting antiviral agents (DAA) have been introduced that target the HCV-encoded proteins which are vital for replication of the virus. The first generation of DAA, telaprevir, in combination with peg-interferon and ribavirin, more efficiently inhibits replication of genotype 1. Although the level of DAA SVR rate is high, the new treatment has some undesirable adverse effects. Micro-RNAs (miRNAs), as the new HCV drug approach, open a new insight into the treatment of non-responder HCV patients. Altered expression of miRNAs is involved in the aspects of HCV infection and HCC. Pages 21-35ObjectiveMorphine and its derivatives are well-known for their strong analgesic effects in medicine. However, their medical long-term administration is severely restricted because of tolerance and dependence. Morphine fails to induce endocytosis of Mu opioid receptors whereas other encephalin analogues such as DAMGO induce rapid endocytosis of these receptors. Repeated use of opioid drugs alters gene expressions. In current study, we analyze the effects of long-term opioid treatment on Dnm1L and Rab22a genes.MethodsHEK293 cell lines that expressed MOP receptors were treated separately with morphine, methadone, and DAMGO. mRNAs were extracted from the cell suspension and we amplified the cDNA. Next, we analyzed Dnm1L and Rab22a gene expressions compared to control samples by qRT-PCR. Results were statistically validated by Graph Pad software.ResultsAmplification graphs from qRT-PCR showed that Dnm1L gene expression was induced by 2.37-fold, whereas Rab22a gene expression decreased by 0.39-fold in response to long-term treatment of cells with morphine compared to control samples. Dnm1L gene expression was not significantly affected by treatment with methadone and DAMGO. Rab22a gene expression decreased by 0.41-fold with methadone treatment and by 0.45-fold in response to DAMGO treatment.ConclusionThe Dnm1L gene could be involved in cellular pathways of morphine-induced tolerance which would indicate the difference between morphine and other similar Mu opioid receptor agonists.Keywords: Morphine, Dnm1L, Rab22a, Tolerance, Dependence

Pages 37-51ObjectiveCrocin, an important saffron ingredient, showed anticancer activity in a variety of cancer types, particularly breast cancer. However, little information is available on the mechanism of its action. Previous studies indicate apoptosis induction by crocin in some cancer cells. This study aims to investigate the effect of crocin on the MDA-MB-468 breast cancer cell line in order to investigate its effect on caspase 9 (Cas9) and cleaved-Cas9, expression and splicing of XBP1, and accumulation of LC3-II.MethodsWe used the MTT assay to investigate the cytotoxic effect of crocin on MDA-MB-468 breast cancer cells. Next, Cas9 and cleaved-Cas9 levels were evaluated by Western blot analysis. Splicing of XBP1 mRNA and expression of the spliced protein (XBP1s) was investigated by RT-PCR and Western blot, respectively. The accumulation of LC3-II was also evaluated by Western blot. The obtained results were analyzed and reported by Image J software.ResultsThe results showed a time and dose-dependent cytotoxic effect of crocin in MDA-MB-468 cells. The expression of Cas9 and its cleavage, therefore, the ratio of cleaved-Cas9/Cas9 significantly increased. Crocin treatment led to a noticeable increase in splicing of XBP1 mRNA, expression of XBP1s, accumulation of LC3-II, and increased the LC3-II/LC3-I ratio in these cells.ConclusionThe data have shown induction of apoptosis in these breast cancer cell lines after crocin treatment. Because of the observed changes in UPR markers and autophagy, it seems that these pathways are possibly involved in this process and in intracellular regulations.Keywords: Breast Cancer, Cleaved-Cas9, UPR, XBP1 splicing, LC3-II accumulation

Pages 53-66ObjectiveHeart failure is one of the most common types of cardiac diseases with high rate of morbidity and mortality, which places a large burden on society. Despite well care system, the survival rate for end-stage heart failure patients is less than 1%. Several studies have recently shown a significant association between long non-coding RNAs with different cardiac diseases.MethodsWe investigated the functional mechanism of BACE1-AS, which was previously reported to be up-regulated in heart failure patients, by assessments of gene expression analysis, cell proliferation, and apoptosis.ResultsBACE1-AS down-regulation significantly reduced expressions of cyclin inhibitors that included CDKN1A, CDKN1B, CDKN1C, CDKN2A, and CDKN2B with the exception of CDKN2C and CDKN2D. BACE1-AS over-expression increased expressions of these cell cycle genes, which implied an effect of BACE1-AS LncRNA on the cell cycle. BACE1-AS down-regulation decreased BCL-2 mRNA expression and increased BAX gene expression. BACE1-AS up-regulation increased BCL-2 and reduced BAX expressions. We did not observe any significant change in cell proliferation with BACE1-AS down-regulation, while BACE1-AS up-regulation decreased cell numbers and induced apoptosis compared with the control group (cells treated with an empty vector).ConclusionThese results showed the important role of BACE1-AS in cell death and, accordingly, pathogenesis of heart failure. It could be considered a potential biomarker for targeting and treatment of heart failure.

Pages 67-76ObjectiveCryptorchidism is the most common deficiency of the male reproductive system and most prevalent endocrine disease of boys at birth. Insufficient prenatal anti-Müllerian hormone (AMH) secretion from Sertoli cells of the testis is one of its causes. The role of this hormone and inhibin B in fertility has recently been proven. AMH is a known indicator of Sertoli cell function. The aim of this study was to assess the relationship between anti-Müllerian hormone, inhibin B, testosterone, and semen parameters in post-pubertal cryptorchidism patients.MethodsWe gathered the data of this cross-sectional study from blood and semen samples of 20 patients who had a history of unilateral cryptorchidism. Patients, 20-40 years of age, referred to the Urology Ward of Shariati Hospital, Tehran, Iran. Serum levels of AMH, inhibin B and testosterone were analyzed by enzyme-linked immunosorbent assay. Semen analysis was performed by means of a computer aided semen analysis system 6.5.0. Pearsons correlation test was used to evaluate the relationships between variables.ResultSignificant positive relationships were existed between total sperm motility and sperm concentration (r=0.086; pConclusionCryptorchidism patients in this study had lower serum concentrations of AMH, inhibin B and testosterone than the normal limits.
