    استفاده از ژن اولئوزین گیاهی یک روش مناسب برای تولید و تجمع پروتئین نوترکیب در مقیاس زیاد و همچنین استخراج راحت تر و ارزان تر می باشد. اتصال اولئوزین به ژن موردنظر، موجب هدف گیری پروتئین نوترکیب به اجسام روغنی بذر می شود. در سال های اخیر، به منظور افزایش کارایی اولئوزین در تولید پروتئین نوترکیب، چند ژن اولئوزین (پلی اولئوزین) ، به ژن هدف متصل می شود. این امر سبب تولید و تجمع پروتئین نوترکیب در سطوح اقتصادی در بذر می گردد. از این رو برای افزایش میزان تجمع پروتئین پروانسولین در بذر گیاه کلزا، یک سازه ی تلفیقی پلی اولئوزین-پروانسولین طراحی و ساخته شد. در طراحی این توالی به ترتیب از سمت /5 توالی، یک توالی کزاک، یک برچسب هیستیدین، سه ژن اولئوزین، یک جایگاه پروتئولیتیکی برای پروتئاز، ژن پروانسولین و یک کدون پایان تترانوکلئوتید قرار گرفت. این تلفیق ژنی پس از سنتز در ناقل pUC57 قرار داده شد. تلفیق ژنی، پس از هضم آنزیمی به وسیله ی آنزیم های BamHI و SacI از pUC57 جداسازی و در ناقل دوتایی pBI121 همسانه سازی گردید. پس از تائید درج تلفیق ژنی در ناقل pBI121 با استفاده از روش های PCR، هضم آنزیمی و توالی یابی، ناقل نوترکیب به سویه ی LBA4404 اگروباکتریوم انتقال داده شد. این باکتری برای تراریختی ریزنمونه های کوتیلدون و هیپوکوتیل کلزا مورد استفاد قرار گرفت. نوساقه های باززایی شده در محیط انتخابی حاوی کانامایسین گزینش شدند. آنالیز گیاهان تراریخته در سطح DNA با استفاده از روش PCR انجام و یک قطعه ی 120 جفت بازی (مطابق با بخشی از ژن پروانسولین) تکثیر شد. همچنین تکثیر این قطعه ی 120جفت بازی با روش RT-PCR، بیانگر بیان ژن هدف در گیاهان تراریخته بود.

    گون از گیاهان دارویی وصنعتی با ارزش در ایران است و گونهAstragalus verus از گونه های بومی مولد کتیرا می باشد. با توجه به اهمیت این گونه در تولید کتیرا و در معرض انقراض قرار داشتن آن این مطالعه با هدف دست یابی به روشی سریع برای تکثیر آن برمبنای شکستن خواب بذرو جوانه زنی و باززایی مستقیم گیاه در شرایط این ویترو انجام شد. ابتدا تاثیر خراش دادن سطحی در شکستن خواب بذرها بررسی شد. سپس تاثیر نوع محیط کشت و غلظت ساکارز در جوانه زنی بذرها و تولید گیاهچه استریل در قالب طرح کاملا تصادفی به صورت فاکتوریل بررسی گردید. ریزنمونه های محور زیرلپه و بالای لپه جهت شاخه زایی چندگانه در محیط کشت MS غنی شده با ویتامین B5 حاوی ترکیب هورمونی BAP در سه سطح و IAA در دو سطح کشت شدند. ریشه زایی شاخه های چندگانه در محیط کشت MS2/1 با ترکیب هورمونی IAA در دو سطح و IBA در سه سطح در قالب طرح پایه کاملا تصادفی با شش تیمار و پنج تکرار بررسی شد. خراش دهی باعث افزایش میزان جوانه زنی با میانگین 98 درصد شد. غلطت های مختلف محیط کشت و ساکارز تاثیری روی درصد جوانه زنی نداشت. با افزایش ساکارز طول ساقه و ریشه، وزن تر و وزن خشک گیاهچه افزایش یافت. بیشترین درصد شاخه زایی در محیط کشت MS حاوی دو میلی گرم در لیتر BAP بدون IAA با میانگین 6/4 عدد و بیشترین درصد ریشه زایی در محیط کشت MS2/1 با 5/1 میلی گرم در لیتر IAA با میانگین 60 درصد بدست آمد. نتایج این پژوهش می تواند در ریزازدیادی، انتقال ژن و تولید درون شیشه ای کتیرا در آینده استفاده شود.

    افزایش مقاومت میکروارگانیسم ها به آنتی بیوتیک های رایج منجر به یافتن ترکیبات ضد میکروبی جدید شده است. پپتیدهای ضد میکروبی یکی از گزینه های مناسب برای جایگزینی آنتی بیوتیک های موجود می باشند. لازیوگلوسین ها گروهی از پپتیدهای زیست فعال می باشند که در مطالعه حاضر، عملکرد ضد میکروبی پپتید نوترکیب لازیوگلوسین ɪɪɪ بر پنج عامل بیماری زای باکتریایی غذایی (باکتری های استافیلوکوکوس اورئوس، سالمونلا تیفی موریوم، انتروکوکوس فکالیس، لیستریا مونوسیتوژنز و اشرشیا کلی) و همچنین بررسی دوز سمیت آن بر یک رده سلول انسانی مورد ارزیابی قرار گرفت. نتایج حکایت از عملکرد ضد میکروبی مناسب پپتید نوترکیب دارد به صورتی که حداقل غلظت بازدارندگی (MIC) و کمترین غلظت کشندگی (MBC) پپتید لازیوگلوسین ɪɪɪ بر باکتری های مورد آزمون به ترتیب در محدوده μg/ml 625/8-851/3 و μg/ml 406/15-703/7 بود. نتایج حاکی از آن بود که باکتری استافیلوکوکوس اورئوس بیشترین حساسیت را به این پپتید دارد به طوری که مقادیر MIC و MBC آن به ترتیب 851/3 و 703/7 بود. غلظتی از پپتید که اثر سمی بر روی سلول های کلیه جنین انسان نشان داد (1652 μg/ml) به مراتب بیشتر از مقادیر MIC و MBC بدست آمده بود. نتایج این مطالعه نشان داد خاصیت باکتری کشی این پپتید با آنتی بیوتیک های رایج قابل رقابت می باشد.

    مطالعه حاضر، اولین پژوهش برای شناسایی واریانت های ژنتیکی در لاین آرین با اطلاعات توالی یابی کل ژنوم است. مطالعه خصوصیات لاین آرین در سطح ژنوم، می تواند تفاوت های ژنتیکی آن را در ارتباط صفات مهم اقتصادی از جمله رشد شرح دهد. در این پژوهش، از اطلاعات توالی کل ژنوم سه قطعه مرغ آرین استفاده شد. نتایج نشان داد که 63866 واریانت بین نمونه ها مشترک است که از این تعداد، 17604 واریانت به عنوان واریانت جدید برای اولین بار در این پژوهش گزارش شدند. همچنین، 604 واریانت در مناطق کدکننده ژنوم وجود دارند که از این تعداد، 204 واریانت منجر به تغییر در نوع آمینواسیدی پروتئین ها می گردند. با توجه به مهم بودن کیفیت پروتئین جیره برای رشد و افزایش وزن برای طیور، نتایج ژن آنتولوژی نشان داد که مسیر کاتابولیسم پروتئین ها و مسیرهای تغییرات پس از ترجمه ی پروتئین های مرتبط با رشد دارای اهمیتی دوچندان هستند. همچنین، ژن های کاندیدایSMURF2 و SUMO1 برای این مسیرهای بیولوژیک پیشنهاد شد. یکی دیگر از نتایج قابل توجه ژن آنتولوژی، معنی دار شدن مسیرهای پاسخ به تنش بود. در لاین آرین به دلیل انتخاب شدید برای دستیابی به پیشرفت ژنتیکی بالا در صفات رشد، وجود تنش اجتناب ناپذیر است. بنابراین، معنی دار شدن مسیرهای پاسخ به تنش، می تواند نشان دهنده اهمیت این مسیرها در ارتباط با عملکرد مطلوب باشد.

    یکی از روش های سالم مدیریت بیماری آتشک در گیاه به (Cydonia oblanga) ، فعال سازی سیستم های دفاعی درونزاد گیاه با استفاده از مواد محرک دفاعی از جمله بیون (Benzothiadiazole, Bion) است. هدف از این تحقیق، بررسی مکانیسم دفاع القا شده توسط بیون در گیاه به با تاکید بر تغییر بیان ژن های دفاعی گروه PR1 بود. بدین منظور، ابتدا از برگ گیاهان تیمار که دو نوبت با محلول بیون (400 میلی گرم در لیتر) محلول پاشی شدند و آب (شاهد) RNA استخراج شد و برای ساخت cDNA مورد استفاده قرار گرفت. سپس برای کنترل داخلی، یک قطعه 750 جفت بازی از ژن خانه دار actin همسانه سازی و تعیین توالی شد. نتیجه توالی یابی نشان داد که 99% تشابه بین ژن اکتین گیاه به با گیاه سیب (Malus domesticus) وجود دارد. بر اساس آن توالی، آغازگرهای اختصاصی برای گیاه به طراحی و سپس تغییرات ایجاد شده در بیان ژن PR1 پس از تیمار درختان به با بیون توسط real-time PCR بررسی و با شاهد مقایسه شد. نتایج این واکنش نشان داد که تیمار بیون موجب 10 بار افزایش بیان ژن PR1 می شود. بر این اساس، اثر ماده بیون در کنترل نسبی بیماری آتشک می تواند با فعالیت پروتئین های دفاعی گروه PR1 مرتبط باشد.

    تولید گونه های فعال اکسیژن به خصوص پراکسید هیدروژن امری اجتناب ناپذیر در متابولیسم هوازی است. از آن جا که این ترکیبات در غلظت های متفاوت، بر فرآیندهای گوناگونی در سلول موثر هستند و افزایش بیش از حد آن ها به تنش های اکسیدی منجر می شود، کنترل غلظت آن ها امری ضروری به نظر می رسد. در این پژوهش از اسانس آویشن شیرازی با ترکیبات فنولی (تیمول، کارواکرول) فراوان و پلیمر صمغ زدو به منظور کپسوله سازی و تولید نانوامولسیون استفاده گردید. تاثیر نانوامولسیون صمغ زدو- اسانس آویشن شیرازی در مهار رادیکال های آزاد 2،2-آزینوبیس- (3-اتیل بنزوتیازولین-6-سولفونیک اسید (ABTS: 2, 2-azinobis-3-ethylbenzothiazolin-6-sulfonic acid) و پراکسیدهیدروژن در خارج از سلول و بیان ژن (NOX: NAD(P) H oxidase) با شماره دسترسی AY561153 (بزرگ ترین تولید کننده گونه های فعال اکسیژن) مورد بررسی قرار گرفت. نانوامولسیون با محتوای فنول 88 میلی گرم در گرم، توانایی بالایی در مهار رادیکال های آزاد ABTS و پراکسید هیدروژن از خود نشان داد. از طرفی نانوامولسیون در غلظت های 25 و 50 میکروگرم در میلی لیتر موجب افزایش و در غلظت های 100، 150 و 200 میکروگرم در میلی لیتر موجب کاهش بیان ژن NOX گردید. با توجه به نتایج به نظر می رسد نانوامولسیون در غلظت های متفاوت بر گونه های فعال اکسیژن تاثیر داشته و در نتیجه توانایی کنترل تنش های اکسیدی را دارا است.

    جنس Xanthomonas از باکتری‎های مهم بیمارگر گیاهی است که سبب خسارت اقتصادی در طیف وسیعی از گیاهان میزبان می‎شود. یکی از فاکتورهای اصلی بیماری‎زایی این جنس سیستم ترشحی نوع III است که باکتری‎ها از آن برای انتقال افکتورهای مختلف به گیاه میزبان استفاده‎می‎کنند. افکتورهای شبه‎فعال‎کننده رونویسی (TALE) که در بیشتر زانتوموناس‎ها شناخته‎شده‎اند، پروتئین‎های متصل‎شونده به DNA هستند که با افزایش بیان ژن‎های هدف اغلب به‎تنهایی نتیجه برهم‎کنش میزبان-بیمارگر را مشخص‎می‎کنند. در پرتو ماهیت تعیین‎کننده برهم‎کنش افکتور TAL و ژن هدف آن‎ها، شناخت تالوم (TALome؛ مجموعه افکتورهای شبه‎فعال‎کننده رونویسی یک بیمارگر) ، پیش‎بینی ژن‎های حساسیت و ویرایش ژنوم میزبان با استفاده از ابزارهای موجود راه‎کارهای جدیدی برای مدیریت بیماری‎های ناشی از باکتری‎ها را فراهم‎کرده‎است. این مقاله مروری به ساختار، نقش افکتورهای TAL در بیماریزایی و مقاومت و نیز ایجاد ارقام مقاوم به Xanthomonas با استفاده از اطلاعات حاصل از تالوم می‎پردازد.

    لپتین به وسیله بافت چربی سفید تولید می شود و نقش مهمی در تنظیم مصرف غذا، تعادل انرژی، باروری و فعالیت های ایمنی بازی می کند. هدف از این پژوهش، مطالعه بیان ژن لپتین در بافت های چربی، کبد، کلیه، شش و قلب در گوسفند کرمانی بود. از بافت های شش گوسفند کرمانی نمونه برداری و RNA استخراج شد. RNA استخراج شده در دمای منفی80 درجه سانتیگراد نگهداری شد. کیفیت و کمیت RNA بررسی و سنتز cDNAانجام شد. واکنش Real Time PCR برای ژن لپتین و بتااکتین صورت گرفت. محصولات PCR روی ژل آگارز 5/1 درصد نیز الکتروفورز شد و سطوح مختلف بیان در بافت های ذکر شده، مورد بررسی قرار گرفت. نتایج حاصل از این بررسی نشان داد که این ژن در تمامی بافت های بررسی شده بیان شده است و بیشترین سطح بیان در بافت چربی و کبد و کمترین سطح بیان در بافت های قلب مشاهده شد. این نتایج می تواند نشان دهنده این امر باشد که لپتین در متابولیسم چربی نقش ویژه ای دارد. مطالعات بیشتری باید انجام شود تا نقش لپتین در فیزیولوژی ساخت و متابولیسم چربی و مواد دیگر مشخص شود. این امر کمک خواهد کرد تا مکانیسم هایی برای شناخت اثر فاکتورهای تغذیه ای و چربی های بدن در فرآیندهای مختلف درک شوند

    در بازدیدهای تابستان 1396 از باغات درختان میوه در استان های خراسان رضوی و شمالی، علائم زردی و ریز برگی در درختان هلو در سطح باغات دیده شد. نمونه های دارای علائم فوق از باغات جمع آوری و برای شناسایی عامل بیماری به آزمایشگاه منتقل شدند. عامل بیماری به وسیله پیوند به نهال های سالم هلو انتقال یافت و در گیاه مایه زنی شده با پیوند، علائم زردی و ریز برگی مشاهده شد. ردیابی عامل بیماری توسط آزمون های دو مرحله ای PCR با جفت آغازگرهای P1/P7 (دور اول) و R16F2n/R16R2 (دور دوم) صورت گرفت. قطعه های حدود 1250 جفت بازی حاصل از PCR دو مرحله ای توالی یابی شدند. مقایسه توالی های به دست آمده با توالی های موجود در NCBI توسط نرم افزار BLAST و بررسی چندشکلی طولی قطعات برشی مجازی ناحیه تکثیر شده، نشان داد که فیتوپلاسمای همراه با زردی هلو در استان های خراسان رضوی و شمالی شباهت بالایی با Candidatus Phytoplasma phoenicium مربوط به گروه جاروک نخود کبوتر (Pigeon pea witches broom) یا 16SrIX آر ان ای ریبوزومی دارند. این اولین گزارش از بیماری زردی و ریز برگی هلو و تعیین ویژگی های فیتوپلاسمای همراه با آن در این مناطق می باشد.

    The use of plant Oleosin gene is a recommended method for large-scale, easier and cheaper production of recombinant protein. Attachment of oleosin gene to target gene cause to accumulate recombinant protein on seed oil bodies surface. Recent researches indicate that application of tandem Oleosin Genes (polyoleosin) fused to the target gene to facilitate recombinant protein purification is more efficient than single Oleosin . In present research, we designed a synthetic fusion fragment containing 5´- Kozak sequence - His-tags, three Oleosin Genes, a proteolytic site for a peptidase enzyme, Proinsulin protein, and a tetranucleotide stop codon. This synthetic sequence was cloned in binary vector pBI121by enzymatic digestion and ligation method. Then for transformation of cotyledon and hypocotyl explants of Canola, confirmed recombinant construct is transformed to Agrobacterium LBA4404 strain. Probable transgenic shoots were regenerated on selective medium containing kanamycin after 4-6 weeks. The analysis of transgenic shoots at DNA level was performed by PCR and amplification of a 120-bp fragment indicate integration of fusion gene in transgenic plant genome. Finally, the Proinsulin gene transcription was also confirmed by RT- PCR.

    Astragalus is one of the most valuable medicinal plants in Iran. The species of Astragalus verus is a native species of tragacant producer. Due to the importance of this species in the production of tragacant, exposed to extinction, the role of seed in its reproduction, as well as the possibility of its reproduction using micropropagation, this study was conducted with the aim of breaking seed dormancy, seed germination and direct regeneration of this plant. First, the effect of surface scratching on breaking of seed dormancy was investigated. Then, the effect of culture medium and sucrose concentration on seed germination and sterile seedling production was studied as factorial based on completely randomized design. The explants of hypocotyle and epicotyle were cultured on MS medium supplied by B5 vitamin containing BAP in 3 levels and IAA in 2 levels due to multiple shoot production. Rooting of multiple shoots was carried out on 1/2MS medium containing IAA in 2 levels and IBA in 3 levels based on completely randomized design with 6 treatments and 5 repeat.Scraping increased the germination rate by an average of 98%. Different media and sucrose concentration had no effect on germination percentage. With increasing sucrose, were increased the stem and root length, fresh weight and seedling dry weight. The highest percentage of shoots was obtained in MS medium containing 2 mg /L BAP without IAA with an average of 4.6 pcs. The highest percentage of rooting was obtained with 1.5 mg/l IAA with an average of 60%. The results of this study can be used in micropropagation, gene transformation and in vitro tragacant production.

    The increasing microbial resistance to existing antibiotics has increased the interest in novel antimicrobial compounds. Antimicrobial peptides (AMPs) represent an attractive alternative to classical antibiotics. Lasioglossins are a group of peptides with antimicrobial activity. The inhibitory effects of a recombinant synthetic Lasioglossin, Lasioglossin ɪɪɪ, on five food pathogens (Staphylococcus aureus, Salmonella typhimurium, Enterococcus faecalis, Listeria monocytogenes and Escherichia coli) and its cytotoxicity on the normal cell line was investigated in vitro. The findings showed great antimicrobial function of the peptide. Minimum inhibition concentration and minimum bactericidal concentration of Lasioglossin ɪɪɪ on the pathogens were the range of 3.851-8.625 and 7.703-15.406, respectively. Results indicated Staphylococcus aureus showed the highest sensitivity to Lasioglossin ɪɪɪ. The Lassioglossin demonstrated cytotoxic effect on human embryonic kidney cells in higher concentration (1652 µg/ml) in comparison to MIC and MBC values. The results indicate that this peptide can be competed with common antibiotics in terms of the bactericidal properties.

    This is the first study for discovering genetic variants in Arian line by whole genome sequencing data. The study of Arian line features at genomic level can unravel the genetic background of economic traits such as growth. In this study, the information of three whole genomes of Arian chickens were used. The results of this study indicated that, there were 63866 common variants among samples. 17604 variants Out of 63866 variants were reported as novel variants in this study. Also 604 variants are located in coding regions, which 204 out of them lead to change in the amino acid sequence of the proteins. Considering the importance of dietary protein quality for growth and weight gain for poultry, the results of gene ontology analysis indicated that the pathways of protein catabolism and post-translational modification of proteins, related to growth are considerable. Also candidate genes such as, SMURF2 and SUMO1 was suggested for these biological pathways. Another interesting result of gene ontology was the significance of response to stress pathways. In the Arian line due to intense selection, the existence of stress is inevitable. Therefore, the significance of stress response can indicate the importance of these pathways in relation to the desired performance.

    One of the safe methods for fire blight disease management in quince plant (Cydonia oblanga) is through activating plant indigenous defense systems using defense elicitors such as Bion (Benzothiadiazole). Investigating the effect of PR1 defense genes group in induced defense mechanism by Bion in quince was the purpose of this study. For this, RNA was extracted from leaf tissues of bion-sprayed (400 mg/L) and water sprayed (control) plants was used for cDNA synthesis. First, a 750 bp fragment of the actin gene from quince, as a house keeping gene, was cloned and sequenced. The result of sequencing showed it has 99% homology with appleactin gene. Specific primers for quince actin gene were designed and used to study changes in PR1 gene expression in bion-treated comparing control plants using real-time PCR method. Based on the results, Bion treatment caused 10 times increase in the PR1 gene expression. Based of result, the effect of Bion in relative control of fire blight in quince can partly be related to PR1 defense proteins activities.

    The production of reactive oxygen species, such as hydrogen peroxide is inevitable in aerobic metabolism. Since, these compounds are effective at different concentration in a variety of processes in the cell and their excessive production lead to oxidative stresses, Therefore, their concentration must be controlled. In this study, Zataria multiflora essential oil with abundant phenolic compounds (thymol, carvacrol), and zedo gum polymers were used to encapsulate nanoemulsion production. The effect of zedo gum - Zataria multiflora essential oil nanoemulsion were investigated in the inhibition of ABTS (2,2-azinobis-3-ethylbenzothiazolin-6-sulfonic acid), hydrogen peroxide free radicals intercellular and expression of the (NOX:NAD(P)H oxidase) gene with accession number AY561153 (the largest producer of reactive oxygen species). Nanoemulsion with 88 mg/gr phenol content showed a high ability to inhibit the free radical ABTS and hydrogen peroxide. On the other hand, at concentration of 25 and 50 µg/ml increased the expression of NOX and at concentration of 100, 150 and 200 µg/ml decreased it. Our results showed that, different concentration of nanoemulsion have different effect on reactive oxygen species and, therefore, they have the ability to control oxidative stresses.

    Xanthomonas is an important plant pathogenic bacteria and cause economic losses in a wide range of crop plants. The type III secretion system as a crucial pathogenicity factor is used to inject effector proteins into the host cell. Transcription activator like effectors (TALEs), known in the most Xanthomonas species, are DNA-binding proteins which determine solely the outcome of plant-pathogen interaction. Because of determinative nature of TALE-target gene interaction, the knowledge of pathogen TALome diversity, forecasting susceptibility genes, genome editing using new developed methods have provided novel strategies for management of plant diseases. This article reviews TALEs structure, their role in pathogenicity and resistance, as well development of resistant plants to Xanthomonas using TALome derived results.

    Leptin is produced by white adipose tissue and plays an important role in regulation of feed intake, energy balance, fertility and immune functions. The aim of this research was to study leptin gene expression in adipose tissue, liver, kidney, lung and heart of Kermani sheep. Tissue samples obtained from 6 animals and RNA was extracted. Extracted RNA were immediately stored at -80°C. Quality and quantity of RNA were evaluated and cDNA was synthesized and Real Time PCR was performed. PCR Products were electrophoresed on 1.5% agarose gel and were evaluated different levels of expression in studied different tissues. Results showed that the leptin gene was expressed in all the tested tissues and the highest level of expression was observed in adipose tissue and liver and the lowest level was detected in heart. These results may show that leptin plays a particular role in fat metabolism. Further studies are needed to clarify role of leptin in the physiology of fat metabolism and other materials. This would help us to better understand the mechanisms for the known effect of nutritional factors and body fatness on various functions.

    In 2017, leaf stunting and yellowing were observed on twigs and branches of stone fruit trees in production regions in Razavi and North Khorasan provinces of Iran. For diagnosis of disease and identification of the causal agent, symptomatic leaf samples were collected in these orchards and were carried to the laboratory. The disease agent was successfully transmitted by grafting from naturally symptomatic samples to peach (Prunus persica) seedlings. Total DNA was extracted from plant samples and indexed by nested PCR using phytoplasma generic primers, P1/P7 and R16F2n/R2. PCR products were sequenced and the nucleotide sequences were compared with those of other phytoplasmas in GenBank and analyzed by virtual restriction fragment length polymorphism (RFLP). The phytoplasmas associated with leaf stunting and yellowing disease in Razavi and North Khorasan provinces were identified as members of the 16SrIX group or Pigeon pea witches broom (Candidatus Phytoplasma phoenicium). This is the first report of leaf stunting and yellowing disease and characterization of associated phytoplasma in these regions.
