    سیاهک مهم ترین بیماری نیشکر درایران و بیشتر مناطق کشت این گیاه در جهان است. موثرترین روش مدیریت بیماری، شناسایی و کشت ارقام مقاوم است. هدف این پژوهش بررسی تغییرات سطح ترکیبات فنولی در دو رقم حساس و مقاوم به بیماری و تعیین ترکیبات فنولی شاخص مقاومت، برای شناسایی سریعتر این گونه ارقام بود. دو رقم NCO-310 و CP78-1628 حساس و مقاوم به بیماری انتخاب شدند و با روش کشت مریستم انتهایی ساقه، گیاهچه های چهار برگی از آن ها تولید شدند. گیاهچه ها با سوسپانسیون مخلوطی از اسپوریدیوم های دو تیپ آمیزشی سازگار بیمارگر مایه زنی شدند. ساقه ی گیاهچه های مایه زنی شده هر رقم و شاهد آن ها در زمان های صفرتا 120 ساعت پس ازمایه زنی نمونه برداری و اسیدهای فنولی محلول و موجود در دیواره سلولی آن ها استخراج شدند و با روش کروماتوگرافی مایع با کارآیی بالا (HPLC) ، وجود و غلظت اسیدهای فنولی پاراکوماریک، فرولیک، پاراهیدروکسی بنزوییک و سیرینجیک در آن ها شناسایی وتعیین شدند. نتیجه مقایسه داده های به دست آمده از دستگاه HPLC نشان داد که در بین این اسیدهای فنولی تنها غلظت اسیدهای فرولیک و سیرنجیک متصل به دیواره در رقم مقاوم نسبت به حساس سطح بالاتری دارند، که این حالت نشان دهنده نقش این اسیدهای فنولی در واکنش مقاومت است. بنابراین می توان این اسیدها را به عنوان شاخص مقاومت در نظر گرفت و از این راه ارقام مقاوم را سریعتر شناسایی کرد و بیماری را مدیریت نمود.

    ویروس پیچیدگی برگ شلغم (Turnip curly top virus, TCTV) به عنوان ویروس تیپ جنس Turncurtovirus از خانواده Geminiviridae چند سال قبل از ایران گزارش گردید. این ویروس دارای دامنه میزبانی وسیعی در میان سبزیجات و علف های هرز است. در این پژوهش، آلودگی گیاه تاتوره (Datura stramonium L.) با علائم زردی عمومی، پیچیدگی شدید و فنجانی شدن برگ ها با استفاده از آزمون پی سی آر، به TCTV اثبات گردید. سپس طول کامل ژنوم ویروس با تلفیق پی سی آر و روش تکثیر دایره غلتان (rolling circle amplification, RCA) و استفاده از یک جفت آغازگر همپوشان تکثیر و بعد از همسانه سازی، در ناقل pJET1. 2 تعیین ترادف گردید و ترادف بدست آمده به لحاظ فیلوژنتیکی مورد تجزیه و تحلیل قرار گرفت. نتایج بدست آمده بیانگر آن است که جدایه تاتوره دارای بیشترین و کمترین میزان شباهت (بترتیب 2/99 و 8/85 درصد) با جدایه های همین ویروس می باشد که پیشتر بترتیب از گیاهان شلغم و ترب گزارش شده اند. به رغم مشابهت بالای ترادف نوکلئوتیدی ژنوم جدایه تاتوره با جدایه شلغم، پروتئین rep رمزشده توسط جدایه تاتوره در انتهای N-terminal دارای 14 اسیدآمینه بیشتر است. در میان نژادهای پنج گانه TCTV، جدایه تاتوره متعلق به نژاد C بوده و در کنار جدایه هائی قرار می گیرند که قبلا از گیاه شلغم یا از بدن زنجرک ها گزارش شده است. نتایج این تحقیق، بار دیگر دامنه میزبانی وسیع ترنکرتوویروس ها در علف های هرز و نقش آن ها را در آلودگی سبزیجات تائید می کند.

    خانواده Didymellaceae گونه های متنوع ساپروفیت، اندوفیت و بیمارگر را شامل می شود که با انواع مختلف بستره های غذایی مرتبط هستند. برخی گونه های این خانواده از قبیل افراد متعلق به جنس Phoma sensu lato در میان مهمترین و گسترده ترین بیمارگرهای گیاهی قرار دارند. با وجود اهمیت اقتصادی اعضای این خانواده در ایران، مطالعه جامعی بر روی تنوع گونه ای آن ها صورت نگرفته است. بسیاری از اعضای Didymellaceae در ایران بر اساس نمونه های هرباریومی شناسایی شده اند و در مورد اغلب آنها کشت های زنده برای مطالعات مولکولی در دسترس نمی باشد. هدف از مطالعه حاضر بررسی تنوع گونه ای خانواده Didymellaceae در استان های آذربایجان شرقی و غربی بود. پس از جداسازی و ایجاد کشت های تک اسپور از بسترهای غذایی مختلف، جدایه ها بر اساس ترکیب داده های ریخت شناختی و داده های توالی چند ژنی (ITS-rDNA، LSU-rDNA و ژن بتاتوبولین) شناسایی شدند. در مطالعه حاضر Epicoccum nigrum، Boeremia exigua، Boeremia strasseri، Didymella glomerata، Didymella tanaceti، Ascochyta medicaginicola، Ascochyta herbicola، Phoma eupyrena و Heterophoma novae-verbascicola از میزبان های گیاهی مختلف، خاک و همچنین به عنوان میکوپارازیت شناسایی شدند. گونه های B. strasseri، D. tanaceti، P. eupyrena و H. novae-verbascicola گزارش های جدید برای میکوبیوتای ایران هستند. علاوه بر این، برای هر گونه میزبان ها/ بستره های غذایی جدید گزارش شدند. نتایج این بررسی تایید کرد که تنها تکیه بر صفات ریخت شناختی جهت شناسایی این قارچ ها در سطح گونه و جنس ناکافی است.

    ویروس توته ی رگبرگ مرکبات (Citrus vein enation virus, CVEV) گونه احتمالی جنس Enamovirus، با علائم رگبرگ توته ای و گال چوبی در تنه، اخیرا از مرکبات ایران گزارش شده است. در تحقیق حاضر خصوصیات بیولوژیکی و مولکولی تعدادی از جدایه های ویروس مطالعه شد. پس از انتقال پیوندی 29 جدایه مختلف ویروس به سه گونه میزبان شامل نارنج (Citrus aurantium) ، لیموی ولکامریانا ((C. volkameriana و لیموی پوست ناصاف (راف لمون، (C. jambhiri، نتایج نشان داد که زمان بروز، نوع و شدت علائم بیماری وابسته به جدایه و میزبان (پایه پیوند شده) بوده و بر این اساس جدایه های ویروس در سه گروه قرار گرفتند. این ویروس با شته سیاه باقلا (Aphis fabae) و با کارایی بالاتری نسبت به شته سبز پنبه (Aphis gossypii) انتقال یافت. این اولین گزارش از انتقال پذیری CVEV توسط شته سیاه باقلا است. نتایج RT-qPCR نشان داد که بافت های پوست ساقه یک ساله و دمبرگ دانهال نارنج آلوده به ترتیب دارای بیشترین و کمترین غلظت CVEV هستند. پایش ماهیانه ویروس در یک سال با آزمون RT-PCR نشان داد که ویروس فقط در دو بازه زمانی اواخر فروردین تا اوایل تیر ماه و اواسط شهریور تا اوایل مهرماه قابل ردیابی است، با وجود این به کار گیری روش RT-qPCR باعث افزایش شصت درصدی حساسیت ردیابی ویروس در آذرماه شد. نتیجه تبارزایی جدایه های مختلف ویروس بر اساس ترادف نوکلئوتیدی ناحیه همپوشان چارچوب های خوانش 1 و 2 (ORF1 و ORF2) ، آنها را به سه گروه تفکیک کرد که تاحدی با گروه های علائم شناسی همبستگی داشت.

    در این پژوهش اثر آلودگی مخلوط فیتوپلاسماهای جاروک لیموترش (16SrII-B) و فیلودی کاهو (16SrIX-D) بر روی علائم بیماری و تراکم سلول های فیتوپلاسما در بوته های پروانش (Catharanthus roseus (L.) G. Don) مطالعه شد. تعداد چهار بوته پروانش سالم و هم سن از طریق قطعات سس آلوده ابتدا با فیتوپلاسمای عامل فیلودی کاهو و پس از سه هفته با فیتوپلاسمای جاروک لیموترش مایه زنی شدند. علائم بیماری و جمعیت فیتوپلاسما در این بوته ها با علائم بیماری و جمعیت فیتوپلاسما در بوته هایی که تنها با یکی از این فیتوپلاسماها مایه زنی شده بودند مقایسه گردید. در بوته هایی که به صورت مخلوط مایه زنی شده بودند تنها کاهش خفیفی در اندازه برگ و فاصله میانگره ها ایجاد شد که با علائم شدید ناشی از هر یک از دو فیتوپلاسما، کاملا متفاوت بود. بوته هایی که به صورت مخلوط مایه زنی شده بودند در مقایسه با بوته هایی که تنها با یک فیتوپلاسما مایه زنی شده بودند طول عمر بیشتری داشتند. با استفاده از پی سی آر معمولی و آغازگرهای اختصاصی وجود هر دو فیتوپلاسما در بوته هایی که به صورت مخلوط مایه زنی شده بودند و همچنین در بوته های شاهد مثبت به اثبات رسید. آنالیز داده های حاصل از آزمون پی سی آر در زمان واقعی (Real-time PCR) نشان داد که جمعیت هر دو فیتوپلاسما در بوته هایی که به صورت مخلوط مایه زنی شده بودند در مقایسه با بوته های شاهد مثبت به طور معنی داری کاهش داشت. به نظر می رسد برهمکنش این دو فیتوپلاسما در آلودگی مخلوط از نوع آنتاگونیسم متقابل باشد.

    در سال های اخیر، یک بیماری کمتر شناخته شده که موجب ایجاد لکه های نکروتیک روی اندام های گل و سطح میوه انار می گردد، در برخی مناطق کشت انار در ایران از جمله استان های گلستان و مازندران شیوع پیدا کرده است. تحقیق حاضر با هدف شناسایی عامل بیماری در این دو استان اجرا گردید. به این منظور در بهار 1396 از باغات انار این دو استان بازدید و نمونه برداری از اندام های گل و میوه صورت پذیرفت. جداسازی عامل بیماری با استفاده از روش های رایج در بیماری شناسی گیاهی انجام و کشت های خالص با استفاده از روش نوک ریسه جهت مطالعات ریخت شناختی و مولکولی ایجاد شدند. پرگنه قارچ روی محیط عصاره مالت آگار کند رشد (3-4 میلیمتر در هفته) ، برجسته، مطبق، میسلیوم متراکم در قسمت مرکزی، میسیلیوم در قسمت مرکزی سفید رنگ و در حاشیه متمایل به صورتی بود. ساختارهای اسپورزایی در محیط کشت تشکیل نگردید. در محل لکه ها در سطح میوه های آلوده آسروول هایی مشاهده شدند. آسروول به قطر50 تا200 میکرومتر، کنیدیوفورها متراکم، منفرد و نسبتا کوتاه، سلول های کنیدیوم زا بی رنگ یا قهوه ای روشن و فیالیدیک بودند. همچنین ناحیه ITS-rDNA توالی یابی شد. قارچ عامل بیماری براساس ویژگی های ریخت شناختی و توالی نوکلئوتیدی ناحیه
    ITS-rDNA گونهElsinoë punicae شناسایی گردید. مایه زنی میوه های سالم انار (رقم یزد) با سوسپانسیون مسیلیوم قارچ منجر به ایجاد علایم اسکب روی میوه شد، در حالیکه در نمونه های شاهد علایمی مشاهده نگردید. عامل بیماری از بافت های مایه زنی شده مجدد جداسازی شد.

    گیاه شبدر زینتی یا شبدر بنفش با نام علمی Oxalis triangularisA. St. -Hil. از تیره Oxalidaceae گیاهی چند ساله است که به عنوان گیاه زینتی گلدانی در فضاهای بسته استفاده می شود. در خرداد سال 1397 برخی از گلدان های گیاه مذکور در دانشگاه مراغه واقع در شهر مراغه، آذربایجان شرقی علایم آلودگی به قارچ مولد زنگ را نشان دادند. بر اساس بررسی دقیق نمونه های آلوده ویژگی های زیر روی برگ های گیاه میزبان مشاهد گردید: مراحل اسپرموگونیوم و اسیوم روی گیاه آلوده مشاهده نشدند. اوردینیوم ها گرد به قطر 4/0-2/0 میلیمتر، شکوفا با ظاهری برآمده و پودری به رنگ زرد رنگ پریده تا زرد-نارنجی در حلقه های متحد المرکز به قطر تا 9 میلیمتر روی سطح زیرین برگ تشکیل شده بودند. اوردینیوم ها فاقد پارافیز بودند. اوردینیوسپورها به ابعاد 19-13 × 23-16 میکرومتر اندازه گیری شدند. این اسپورها بیضوی، کروی یا به شکل تخم مرغی وارونه بودند. دیواره اوردینیوسپورها بی رنگ به ضخامت غالبا 1-5/0 و بندرت تا 5/1 میکرومتر و به طور متراکم و ظریف خاردار بود. منافذ تندشی اوردینیوسپورها ناپیدا بودند. تلیوم های مجزا روی نمونه بررسی شده دیده نشد. تلیوسپورها به صورت پراکنده در اوردینیوم ها دیده شدند.

    Smut is the most important disease of sugarcane in Iran, and most of the plant growing areas in the world. The most effective method of disease management, is identification and cultivation of resistant varieties. The aim of this study was to investigate changes in the level of phenolic compounds in resistant and susceptible cultivars to disease and detection phenolic compounds resistance index, to accelerate the identification of resistant cultivars. Two cultivars NCO-310 and CP78-1628, susceptible and resistant to the disease were selected, and with apical meristem and lateral buds culture method, their seedlings with four leaves were produced. Seedlings were inoculated with a suspension of a mixture of two compatible mating type of pathogen. Stem of plants were sampled at 0-120 hour post-inoculation. Soluble and cell-wall binding phenolic acids were extracted from samples, and presence and concentration of phenolic acids p-coumaric , ferulic, p-hydroxybenzoic and syringic acids were detected and determined by high performance liquid chromatography (HPLC) method. Comparison the data obtained from HPLC device showed only titer of cell wall-bound phenolic acids ferulic and syringic showed enhancement, in the resistant cultivar, this reflects the role of these phenolic acids in the resistance reaction. Thus, these phenolic acids can be considered as an indicator of resistance, and in this way quickly identify resistant cultivars and management disease.

    Turnip curly top virus (TCTV) is a type member of the genus Turncurtovirus previously reported from Iran. TCTV has a wide host range among vegetables and weeds. In this study, the TCTV infection of the symptomatic jimsonweed (Datura stramonium L.) showing general yellowing, severe curling and cup-shaped leaves was confirmed by PCR. Full-length genome of this isolate was amplified using combination of the PCR and rolling circle amplification (RCA) and cloned into the pJET1.2 vector. Resulted nucleotide sequence indicated that the jimsonweed isolate of TCTV shares maximum and minimum identities (85.8-99.2%) with turnip and radish isolates of TCTV, respectively. In spite of high similarity between full-length nucleotide sequence of jimsonweed and several GenBank isolates, predicted rep protein of the jimsonweed isolate has an additional 14 amino acid residues at the N-terminal. Among five TCTV strains, the jimsonweed isolate was classified in the strain C with turnip and leafhopper isolates of the virus. Results of this study again confirm wide host range of turncurtoviruses in weeds and the role of wild species in infection of vegetables.

    Didymellaceae is a species-rich family including saprophytic, endophytic, and pathogenic species that are associated with different types of substrates. Some species of this family, such as Phoma sensu lato, are among the most important and widespread plant pathogens. Despite the economic significance of species belonging to this family in Iran, no coherent study has been conducted on their biodiversity. Many of presently known Didymellaceae members in Iran have been identified based on herbarium materials and in most of the cases there is no living culture available for molecular studies. The aim of present study was to explore species diversity of the family Didymellaceae in East and West Azarbaijan provinces. After isolation and establishing single spore cultures from different substrates, the isolates were identified based on morphological and molecular data using sequence data of ITS-rDNA, LSU-rDNA, and beta-tubulin gene. In this study Epicoccum nigrum, Boeremia exigua, Boeremia strasseri, Didymella glomerata, Didymella tanaceti, Ascochyta medicaginicola, Ascochyta herbicola, Phoma eupyrena and Heterophoma novae-verbascicola were identified from different host plants, soil and also as mycoparasites. Boeremia strasseri, D. tanaceti, P. eupyrena and H. novae-verbascicola are new records for the mycobiota of Iran. Furthermore, for each species new hosts/substrates are reported. Our results confirm that the sole reliance on morphological features for the identifaction of these fungi, is insufficient both at species and genus level.

    Citrus vein enation virus (CVEV), a tentative species of the genus Enamovirus, family Luteoviridae, which causes vein enation and trunk woody gall symptoms on citrus, has been recently reported from Iran. In the present research, biological and molecular properties of CVEV different isolates were studied. Following graft transmission of 29 CVEV isolates on sour orange (Citrus aurantium), volkamer lemon (C. volkameriana) and rough lemon (C. jambhiri), the results indicated that the incidence, type and severity of symptoms depend on the CVEV isolate and host (rootstock), accordingly virus isolates were classified into three groups. CVEV was transmitted by Aphis fabae with a significantly higher efficiency than that by Aphis gossypii. Thisis the first report of CVEV transmissibility by A. fabae in the world. Based on RT-qPCR results, the maximal and minimal concentrations of CVEV were determined in young bark and petioles tissues of infected sour orange seedlings, respectively. Monthly monitoring of the virus throughout the year by RT-PCR showed that CVEV was only detectable during two periods of late April to early July along with mid-September to early October, however, sensitivity of virus detection raised up to 60 percent in early December by using RT-qPCR. In the phylogenetic analysis of CVEV isolates based on nucleotide sequence of overlapped regions of ORF1 and ORF2, isolates were grouped into three clades which showed some correlation with symptomology groups.

    In this research the effect of mixed infection of witches’ broom disease of lime (WBDL) (16SrII-B) and lettuce phyllody (LP) (16SrIX-D) phytoplasmas on symptom expression and phytoplasma density in periwinkle plants (Catharanthus roseus (L.) G. Don) were investigated. Four healthy periwinkle plants of the same age were first inoculated with LP phytoplasma and after three weeks with WBDL phytoplasma using detached fragments of inoculative dodder (Cuscuta campestris Yank.). Disease symptoms and phytoplasma population in plants inoculated with both phytoplasmas were compared with those in plants inoculated with either WBDL or LP phytoplasma. In doubly inoculated periwinkle plants the disease symptoms mitigated to mild reduction of leaf size and internodes which strongly differed from those in periwinkle plants singly inoculated with either WBDL or LP phytoplasma. Plants with mixed infection lived longer than those inoculated singly with either phytoplasma. Both WBDL and LP phytoplasmas could be detected in singly and doubly infected plants by conventional and real time PCR assays using specific primer pairs. Analysis of the data obtained by real-time PCR showed that the population of both WBDL and LP phytoplasmas in the mixed infection was significantly lower than that in plants singly inoculated with WBDL or LP phytoplasma. It seems that interaction of WBDL and LP phytoplasmas in mixed infection of periwinkle is of the mutual suppression type.

    In recent years a lesser known disease that causes necrotic spots on flower and fruit surface of pomegranate have been observed in orcahrds of Golestan and Mazandaran provinces, Iran. The present study was aimed to characterize and determine the causal agent of this disease. Towards this aim, during spring 2016 pomegranate orchards were inspected in Golestan and Mazandaran provinces and symptomtic flowers and fruits were collected and transported to the laboratory. Isolation was made using routine plant pathology methods on Potato dextrose Agar supplemented with sulphate streptomycin. Pure cultures were established using hyphal technqiue. Colonies on MEA grew very slowly (attained a diameter of 3-4 mm after 7 days at 25℃), raised, floded, dense aerial mycelium in centre, white in centre and pink in margin. Conidiomata did not develop in culture. Acervuli were observed on lesions in fruit surface, subcuticular, subepidermal, 50-200 µm diam; conidiophores dense, individual, short, and hyaline; conidiogenous cells hyaline to subhyalaine, phialidic.Conidia ellipsoidal, aseptate, hyalaine, 4-8 × 2-3.5 µm. The identity of the species was further investigated by PCR amplification and sequence of ITS-rDNA region. A BLAST serach against sequence data in GenBank, showed 100 percent similarity with somesequences avaibale in GenBank. Based on morphological and molecular data the fungus was identified as Elsinoë punicea. Inoculation of healthy pomegranate fruit (Yazd Cultivar) using mycelial suspension of the fungal isolates, resulted in the disease symptoms resembling those observed in field conditions; while, controls did not develop disease symptoms. The causal agent was re-isolated from inoculated tissues. With identification and determination of the causal agent, it is necessary to study the incidence of this disease in other pomegranate producing areas in country, biology of the causal agent, diseases cycle and finally provide proper management strategies for this disease.

    Oxalis triangularis A. St.-Hil. (Oxalidaceae) known as “False Shamrock”, is a perennial ornamental plant native to Brazil. The plant is used as indoor pot plant in Iran. In June 2018, rust-infected leaves were sampled from an indoor pot plant in University of Maragheh, located in Maragheh, East Azarbaijan. Uredinia round, 0.2-0.4 mm in diameter, in orbicular groups forming concentric circles up to 9 mm in diameter, soon naked, erumpent, powdery, yellowish white to orange-yellow, without paraphyses, urediniospores 16-23 × 13-19 µm, ellipsoid, globoid or obovoid, wall 0.5-1(1.5) µm thick, minutely and densely echinulate, colorless, germ pores obscure. Telia not seen, teliospores were sporadically present in uredinia, 21-28 × 15-18 µm, ellipsoid, oblong or broadly oblong, rounded or obtuse at apex, slightly constricted at septum, wall quite colorless, smooth, thin, 0.5-0.8 µm, pedicel colorless, as long as spores or shorter (Fig. 1).
