بررسی اثر سولفورموستارد بر وضعیت متیلاسیون پروموتر ژن FOXP3 در جانبازان شیمیایی با عوارض تاخیری ریویصفحات 1-8مقدمه و هدفسولفورموستارد (SM) یک ماده شیمیایی و سمی است که در بروز عوارض ریوی و فرآیندهای التهابی در افراد مواجهه یافته با این گاز نقش مهمی دارد. FOXP3 یک فاکتور رونویسی مهم در عملکرد سلول های Treg است. تغییر الگوی متیلاسیون FOXP3 تحت تاثیر مواد شیمیایی و سمی باعث بروز بیماری های ریوی مزمن می شود. هدف از این مطالعه بررسی اثر ماده ی شیمیایی و سمی SM بر وضعیت متیلاسیون پروموتر FOXP3 در جانبازان شیمیایی با عوارض تاخیری ریوی است.
مواد و روش هادر این مطالعه، 20 نفر مواجهه یافته با گازخردل بعنوان گروه مورد و 20 نفر سالم بعنوان گروه کنترل می باشند. DNA از نمونه های خون استخراج گردید. وضعیت متیلاسیون پروموتر ژن FOXP3 از طریق تست Methylation specific PCR (MSP) بررسی گردید.
نتایجدر متیلاسیون ژن FOXP3 بین دو گروه اختلاف معناداری وجود نداشت.
نتیجه گیریوضعیت متیلاسیون ژن FOXP3 در گروه مواجهه یافته خفیف و متوسط تغییر معناداری پیدا نکرده است. به منظور یافتن نقش این ژن در پاتولوژی ریه مصدومین شیمیایی نیاز به سنجش سایر فاکتورهای اپی ژنتیکی و بررسی های بیشتر وجود دارد.کلیدواژگان: سولفورموستارد، عوارض ریوی، متیلاسیون -
صفحات 9-20مقدمه و هدفاستفاده از نانوحاملهای حاوی ترکیبات سیتوتوکسیک گیاهی به جای داروهای رایج شیمی درمانی می تواند بسیاری از چالش های پیشروی این روش سرطان درمانی را کاهش دهد. در این مطالعه نانوسامانه های سورفکتانتی حاوی عصاره پوست انار به منظور ارزیابی سمیت آن بر رده ی سلولیMCF-7 سرطان پستان طراحی و ویژگی های فیزیکوشیمیایی آن مورد بررسی قرار گرفته است.
مواد و روش هاوزیکول های نیوزومی با استفاده از Span 60، کلسترول و پلی اتیلن گلیکولبروش فیلم نازک تهیه شده و عصاره پوست انار درون نیوزومها بارگذاری شد. بررسی شاخصه های فیزیکوشیمیایی آن ها با استفاده از دستگاه های زتا سایزر، FT-IR، SEM انجام شد و میزان رهایش عصاره در دمای oC37 و oC42 محاسبه گردید. در پایان میزان سمیت نانوسامانه دارای عصاره بر سلول های سرطانی پستان با استفاده از تست MTT سنجیده شد.
نتایجنتایج پژوهش حاضر نشان می دهد که نیوزومهای دارای عصاره به ترتیب دارای راندمان انکپسولاسیون، اندازه و شارژ سطحی 28/61 درصد، nm 6/143 وmV 9/40-است. است. بررسی رهایش نیز نشان می دهد که نانوحامل نیورومی در دمای oC 37 و oC 42 دارای رهایش کنترل شده است. مطالعه ی FT-IR و SEM نیز عدم برهمکنش عصاره با سامانه و موفولوژی کروی سامانه را تایید می کند. در پایان مشخص می شود که میزان سمیت عصاره پوست انار در شرایط کپسوله نسبت به شرایط کپسوله نشده بر رده ی سلولی MCF-7 سرطان پستان بیشتر است.
نتیجه گیری: با توجه به نتایج حاصل از این پژوهش مشخص می شود که نانو حامل نیوزومی می تواند حاملی مناسب جهت رسانش عصاره پوست انار در شرایط سلول های سرطانی باشد.کلیدواژگان: گیاهان دارویی، پوست انار، نیوزوم، نانوحامل، MCF-7 -
صفحات 21-28مقدمه و هدفاسترس های اجتماعی با تاثیر بر سیستم ایمنی و ویژگی های فیزیولوژیک اندام ها، طیف وسیعی از اختلالات و نیز نارسایی های سایکوپاتولوژیکال از جمله افسردگی و اضطراب را به دنبال می آورند. هورمون اکسی توسین که در سیستم عصبی مرکزی و برخی بافت ها تولید می شود، در کاهش اضطراب و افسردگی ناشی از استرس نقش دارد. همچنین استرس های اجتماعی بر لنفوسیت های کشنده طبیعی سیستم ایمنی ذاتی که نقش مهمی در دفاع در برابر تومورها و عفونت های ویروسی دارند؛ اثر می گذارند. هدف از این مطالعه بررسی اثر استرس بی ثباتی و اکسی توسین برون زاد بر تعداد سلول های کشنده طبیعی در رت های مواجهه یافته با این استرس است.
مواد و روش هارت های WISTAR به مدت 21 روز در تماس با استرس بی ثباتی بودند به طوری که هم خانه آن ها هر سه روز یک بار عوض می شد و از روز 11 ام یک گروه از رت ها 20 میکرولیتر و گروه دیگر 40 میکرولیتر اکسی توسین با غلظت1mg/ml ویک گروه کنترل نرمال سالین به صورت تزریق اینترانازال دریافت کردند و یک گروه کنترل سالم بدون هیچ گونه تزریق وجود داشت. در پایان مطالعه حیوان ها بی هوش شده و کشته شدند. نمونه خون محیطی و بافت طحال جمع آوری شد و تعداد سلول های NK با روش فلوسایتومتری توسط دو مارکرCD3-CD161+ شمارش شد.
نتایجتعداد سلول های NK در خون محیطی در گروه های استرس بی ثباتی و تیمار شده با دوز های 20 و 40 میکرولیتر با اکسی توسین به ترتیب (0/9±4/4، 1/8±3/6) نسبت به گروه کنترل سالم (2/1±4/5) تفاوت معنی داری نداشت. در طحال تعداد سلول های NK در گروه استرس بی ثباتی تیمار شده با دوز های 20 و 40 میکرولیتر به ترتیب (1/1±4/7،1/6±4/4) بود که نسبت به گروه کنترل سالم (0/6±2/2) افزایش معنی دار داشت. نتیجه گیریاسترس بی ثباتی به همراه تیمار با دوز 20 و 40 میکرولیتر اکسی توسین موجب افزایش تعداد لنفوسیت های NK در طحال رتها می شود که در واقع نشان می دهد که این اثر هم افزایی سیستم ایمنی را در یک حالت آماده باش برای مقابله با عفونت ها و سرطان قرار می دهد.کلیدواژگان: استرس بی ثباتی، سلول کشنده طبیعی، اکسی توسین -
صفحات 29-34مقدمه و هدفسلول های T تنظیمی (Treg) از زیرگروه های لنفوسیت ها هستند که با مکانیسم های وابسته به تماس سلولی و ترشح عوامل محلول باعث کنترل التهاب و برقراری هموستاز می شوند. با توجه به این که در افراد مواجهه یافته با سولفورموستارد عدم تعادل سلول های ایمنی و اختلال در هموستاز وجود دارد، در مطالعه حاضر درصد سلول های Treg با مارکرهای CD4, CD25, CD127, FOXP3 در خون محیطی افراد مواجهه یافته با سولفورموستارد در مقایسه با گروه کنترل و رابطه آن با عوارض ریوی درازمدت بررسی می گردد.
مواد و روش ها از 12 فرد مواجهه یافته با سولفورموستارد و 12 داوطلب سالم دعوت شد. بررسی های بالینی گروه مورد توسط پزشکان فوق تخصص ریه و اسپیرومتری، توسط اپراتور تست ریه انجام شد. پس از جداسازی سلول های تک هسته ای خون محیطی درصد سلول های Treg با روش فلوسایتومتری تعیین شد.
نتایجدرصد سلول های Treg در افراد مواجهه یافته با سولفورموستارد در مقایسه با گروه کنترل تفاوت معناداری نداشت. هیچ ارتباط معناداری نیز بین پارامترهای اسپیرومتری و درصد این سلول ها در افراد مواجهه یافته مشاهده نشد.
نتیجه گیریبا توجه به مطالعه حال حاضر می توان گفت در جانبازان شیمیایی با عوارض ریوی خفیف هیچ تفاوتی ازلحاظ درصد سلول های Treg با افراد سالم وجود ندارد و برای درک نقش این سلول ها در عوارض ریوی این بیماران مطالعات بیشتری لازم است.کلیدواژگان: سولفور موستارد، اسپیرومتری، جانبازان شیمیایی، سلول هایTreg -
صفحات 35-42مقدمه و هدفخواب طبیعی باعث تجدید و تقویت قوا می شود. خواب در نتیجه افزایش رطوبت در مغز است که این رطوبت مانع حرکت روح در عصب شده و در نتیجه فرد به خواب می رود. هزینه های بالا، عوارض درمان و همچنین عدم پاسخ مناسب به درمان های رایج علاقه به استفاده از درمان های مکمل را افزایش داده است. مطالعه حاضر به بررسی عوامل مرتبط با بی خوابی و خواب خوب از دیدگاه طب سنتی ایران و طب رایج می پردازد.
مواد و روش هادر این بررسی که یک مطالعه کتابخانه ای است متون قدیمی و پایگاه های اطلاعاتی روز دنیا جستجو شده و خواب و اختلالات مربوطه بر مبنای طب جدید و قدیم مقایسه شده است.
نتایجعلل بی خوابی به دو دسته درونی و بیرونی تقسیم می شوند که عواملی مانند خشکی مغز، از علل درونی و مواردی مانند غم از علل بیرونی می باشند. خواب مناسب با مزاج فصول و میزان رطوبت آن، ورزش ملایم، هضم خوب غذا، دفع مناسب سموم و همچنین کنترل مناسب اعراض نفسانی مرتبط است. ایجاد رطوبت مناسب در مغز، پرهیز از غذاهای نفاخ و خشکی افزای مغز و ماساژ کف پا از درمان های مناسب برای بی خوابی می باشند.
نتیجه گیریهر دو مکتب طبی به زمان بندی مناسب خواب توجه داشته بر پرهیز از خواب روز تاکید دارند. همچنین اصلاح سبک زندگی خط اول درمان در اختلالات مرتبط با خواب از دیدگاه طب جدید و قدیم است.کلیدواژگان: طب سنتی، ابن سینا، بی خوابی

Pages 1-8Background and ObjectiveSulfur mustard (SM) is a toxic and chemical agent. The incidence of pulmonary complications and inflammatory processes play an important role in SM-exposed individuals. FOXP3 is one of the important factors in the development of chronic pulmonary diseases. FOXP3 gene promoter due to exposure to chemical and toxic materials leads to chronic pulmonary disease. In this study, the effect of toxic sulfur mustard was evaluated on the promoter methylation status of FOXP3 in SM-exposed individuals with delayed pulmonary complications.
Materials and MethodsIn this case-control study, 20 SM exposed cases and 20 unexposed as control were studied. DNA was extracted from blood samples. Methylation specific PCR(MSP) was used to evaluate promoter methylation status of FOXP3.
ResultsThere was no significant difference between the promoter methylation status of FOXP3 in the exposed group as compared to control group.
ConclusionThe methylation status of the FOXP3 gene did not significantly change after exposure to sulfur mustard and other mechanisms might be involved in expressing this gene. To determine the effect of SM on methylation and its role in the pathogenesis of chemical victims’ lung complications, it is needed to measure other epigenetic factors and further studies.Keywords: Sulfur mustard, Pulmonary complication, Methylation, FOXP3 -
Pages 9-20Background and ObjectiveUse of nano-carriers that contain plant cytotoxic compounds instead of common chemotherapy drugs can reduce many of the challenges in front of this method of cancer therapy. In this study, surfactant nano-systems containing pomegranate peel extract were designed to evaluate its cytotoxic effects on MCF-7 cell line (breast cancer) and investigated its physicochemical properties.
Materials and MethodsNiosomal vesicles were prepared using span60, cholesterol and polyethylene glycol by thin-film method and the pomegranate peel extract were loaded into the niosomes. Physicochemical characteristics were evaluated using Zeta Sizer, FTIR, SEM, and extract release amount was calculated at 37°C and 42°C. The toxicity of the nano-carrier containing extract was measured on the MCF-7 cell line of breast cancer using MTT assay.
ResultsThe results of this study indicate that the niosomes-containing extract has the encapsulation efficiency, size, and surface charge as 61.28%, 14.36 nm, and 40.9 mV, respectively. The study of its releasing also shows that the niosomal nano-carrier has controlled release at 37°C and 42°C. The study of FTIR confirms that there is no interaction between peel extract and nano-carrier. Also, SEM shows the spherical morphology of the nano-carriers. MTT results demonstrate that the toxic effects of niosomes-contained pomegranate peel extract on MCF-7 cell line of breast cancer is higher than peel extract alone.
ConclusionAccording to the results of this investigation, it is indicated that the carrier niosomal nano vesicles can be a suitable carrier for delivery of pomegranate peel extract in conditions of cancerous cells.Keywords: Medicinal plants, Pomegranate peel, Niosome, Nano-carrier, MCF-7 cell line -
Pages 21-28Background and ObjectiveSocial stresses which affect the immune system and the physiological features of the organs, cause a range of disorders, as well as psychopathological deficits such as depression and anxiety. The oxytocin hormone which is produced in the central nervous system and some tissues plays an important role in reducing anxiety and depression caused by stress. Also, social stress affects the natural killer lymphocytes of the innate immune system that play an important role in defense against tumor and viral infections. The purpose of this study was to investigate the effect of unstable stress and extracellular oxytocin on the number of natural killer cells in rats exposed to this stress.
Materials and MethodsWistar rats were subjected to instability stress for 21 days, hence their cage-mate were changed every three days. From the 11th day, a group of rats received 20 microliter and the other group received 40 microliter of 1 mg/ml oxytocin and the control group received normal saline by intranasal rout and there was a healthy control group without any injections. At the end of study, the animals were anesthetized and then were killed. Blood sample and spleen tissue were obtained and the number of NK cells was counted by flow cytometry with CD3-CD161+ markers.
ResultsThe number of NK cells in peripheral blood in the instability stress groups treated with 20 and 40 μL doses of oxytocin (3.6 ± 1.8, 4.4 ± 0.9) compared with healthy controls (4.5 ± 2.1) did not have a significant difference. In the spleen, the number of NK cells in the instability stress group treated with doses of 20 and 40 μL was (4.4 ± 1.6, 4.7 ± 1.1), which was significantly higher than the control group (2.2 ± 0.6) (P value = 0.025 and P value = 0/008, respectively).
ConclusionInstability stress combined with treatment with 20 and 40 μL of oxytocin increases the number of NK lymphocytes in the spleen of the rats, which in fact indicates that this synergistic effect puts the immune system in a precautionary state to confront infections and cancerKeywords: Instability stress, Natural killer cell, Oxytocin -
Pages 29-34Background and ObjectiveRegulatory T cells (Treg) are the subgroups of lymphocytes that control inflammation response and regulates homeostasis by mechanisms of cellular contact and secretion of soluble agents. Here, we investigated percentage of Treg in peripheral blood of sulfur mustard-exposed patients with CD4, CD25, CD127, FOXP3 markers in comparison with healthy individuals and the correlation between these cells and long-term pulmonary complications.
Materials and MethodsIn this study, 12 sulfur mustard-exposed patients and 12 healthy volunteers were invited. Clinical inspections of both groups were done by pulmonary specialists as well as spirometric evaluation that was conducted by pulmonary function test operators. After isolation of peripheral blood mononuclear cells, the percentage of Treg cells was determined by flow cytometry.
ResultsPercentage of Treg cells was not significantly different in those exposed to sulfur mustard. Also, there was no significant correlation between spirometric parameters and percentage of these cells.
ConclusionAccording to the current study, there is no difference in the percentage of Treg cells between exposed patients with mild pulmonary complications and healthy volunteers and more studies are needed to understand the role of these cells in the pulmonary complications of these patients.Keywords: Regulatory T cells_Sulfur mustard_Spirometry_Chemical victims -
Pages 35-42Background and ObjectiveNatural sleep increases the strength of the body. Sleep is caused by increased moisture in the brain and the moisture in the nerves prevents the movement of the soul and thus the person goes to sleep. High costs, complications of treatment, and lack of appropriate response to common treatments have increased the interest in using complementary therapies. The present study examined the factors associated with insomnia and sleep of high quality from the perspective of Persian medicine and modern medicine.Materials and MethodsThis study was a library research based on searching ancient medical books and databases. Finally, sleep and related disorders in Persian medicine were compared with allopathic medicine.ResultsThe causes of insomnia are divided into internal and external categories in which factors such as dryness of the brain are internal causes and factors like sadness are external causes. The proper sleep is related to the temperament of the seasons and its moisture content, mild exercise, good digestion of food, the proper disposal of pesticides and waste materials, as well as proper control of mental movement and response. Proper moisture in the brain, avoiding unhealthy foods and increasing dryness of the brain and feet massage are good treatments for insomnia.ConclusionBoth modern and old medical schools emphasize on proper timing of sleep to avoid daytime sleep. Also, lifestyle modification is the first line of treatment for sleep-related disorders in both medical disciplines.Keywords: Traditional medicine, Avicenna, Insomnia
