وجود کانالهای کلر در غشا داخلی میتوکندری گزارش شده است. این کانالها موجب تنظیم حجم اندامک و سلول، ارتباطات سلولی و اسیدی شدن اندامک ها و حفاظت سلولی می شوند. مطالعات قبلی ما حضور یک کانال کلری با هدایت 301pS را در غشا داخلی میتوکندری مغز نشان داد. در مطالعه حاضر، رفتار بیوفیزیکی یک کانال کلرجدید در غشاء داخلی میتوکندری مغز موش صحرایی مورد بررسی قرار گرفت.
روش هامغز موش صحرایی بالغ جدا و هموژنیزه شد. هموژنا در طی 3 مرحله در MSE-دیژیتونین، آب، و Na2CO3 سانتریفیوژ شد و وزیکول های غشاء داخلی میتوکندری به شکل محلول در MSE جدا گردید. فسفاتیدیل کولین زرده تخم مرغ جهت تشکیل غشاء دو لایه لیپید استخراج گردید.این غشاء در منفذی به قطر µm 200 تشکیل شد. سیگنالهای ثبت تک کانال به میزان 1 kHz فیلترشدند و با سرعت نمونه برداری kHz 10 ذخیره گردیدند. اطلاعات بر اساس Markov's noise free single channel analysis با نرم افزار Pclamp 10 آنالیز گردیدند.
یافته هاکانال الحاق شده در غشاء لیپیدی دولایه در محیط الکترولیتی mM KCl trans 50/mM KCl cis 200 یک کانال آنیونی با کنداکتانس pS158 بود. احتمال باز بودن کانال نشان داد که این کانال یک کانال وابسته به ولتاژ بوده و در محدوده ولتاژی بین mV20± فعالیت بالایی دارد. اضافه کردن ماده مهار کننده کانال کلر DIDS به ناحیه ای که معادل محیط داخل سلولی (سمت سیس) است سبب مهار قوی کانال گردید.
نتیجه گیریاین بررسی حضور یک کانال آنیونی وابسته به ولتاژ در غشای داخلی میتوکندری بافت مغز موش صحرایی را نشان داد. این کانال ممکن است در اعمالی چون تنظیم پتانسیل غشایی، تنظیم pH، سنتز ATP و حفاظت سلولی در مغز نقش کلیدی داشته باشد.

بعضی از بیماریها از قبیل پیسی، پسوریازیس و سرطان پوست در اثر اختلال درتولید رنگدانه های پوست و هموستاز سلولهای تولیدکننده ملانین ایجاد میشود. گیرنده آریل هیدروکربن (AHR) یک گیرنده وابسته به لیگاند میباشد که نقش مهمی در متابولیسم داروها، تکامل سیستم ایمنی، تعیین سرنوشت سلولهای بنیادی، رشد و ازدیاد سلولی، هموستاز سلولی، تنظیم چرخه طبیعی سلول و ساخته شدن ملانین دارد. هدف این بررسی ارتباط متقابل سیستمNADPH oxidase ها (NOXs) و AHR در ایجاد و یا جلوگیری از سنتز ملانین میباشد.
روش هابعد از کشت سلولهای ملانومای موش رده B16/F10 در محیط استاندارد، سلولها با غلظتهای مختلف لیگاند درونزاد گیرنده AHR به نام6-formylindolo[3,2-b] carbazole (FICZ) به تنهایی یا همراه با آنتاگونیست گیرنده AHR 3'-Methoxy-4'-Nitroflavone, MNF,، مهارکنندهی آنزیم سیتوکرومP450 از خانواده1A1 (CYP1A1) و مهارکنندهی آنزیم NADPH oxidase (NOX) (کورکومین 1 میکرومولار) تیمار شدند و سپس فعالیت آنزیم CYP1A1، فعالیت آنزیم تیروزیناز و مقدار ملانین اندازه گیری شد.
یافته هاFICZ به صورت معنادار و با یک الگوی وابسته به دوز و زمان باعث افزایش فعالیت آنزیم CYP1A1 شد. FICZ در غلظت زیاد (100 نانومولار) به تنهایی و یا همراه با مهارکننده CYP1A1 و NOX باعث افزایش فعالیت آنزیم CYP1A1 و افزایش میزان ملانین گردید. FICZ در غلظت کم (1 نانومولار) همراه با مهارکنندهی NOX به صورت معناداری (0/05 < p) باعث افزایش سنتز ملانین گردید.
نتیجه گیریمهار متابولیسم FICZ میتواند باعث تحریک سنتز ملانین در شرایط برون تن شود.

یادگیری و حافظه نقش اساسی در زندگی روزمره انسان دارد و حافظه پایه و مبنای تمام آموزش ها و یادگیری ها می باشد. با شناسایی ترکیبات تقویت کننده یادگیری و حافظه می توان به بسیاری از افراد که دچار کندی ذهن و ضعف حافظه هستند، کمک بزرگی نمود. هدف مطالعه حاضر بررسی اثر اسانس بابونه و تمرینات بدنی بر میزان حافظه احترازی غیرفعال در موش های صحرایی بود.
روش ها46 موش صحرایی نر (300-250 گرم) به صورت تصادفی به 6 گروه 8 تایی شامل گروه کنترل، گروه دریافت کننده اسانس بابونه با غلظت 50 میلی گرم بر کیلوگرم، گروه دریافت کننده اسانس بابونه با غلظت 75 میلی گرم بر کیلوگرم، گروه دریافت کننده تمرینات بدنی، گروه دریافت کننده تمرینات بدنی+ اسانس بابونه با غلظت 50 میلی گرم بر کیلوگرم و گروه دریافت کننده تمرینات بدنی+ اسانس بابونه با غلظت 75 میلی گرم بر کیلوگرم تقسیم شدند. موش ها به مدت 10 روز با اسانس و تمرینات بدنی تیمار شدند. از روز ششم تجویز اسانس و تمرین بدنی، میزان حافظه احترازی غیرفعال توسط جعبه شاتل به مدت 4 روز سنجیده شد. در انتها خونگیری انجام شده و ظرفیت آنتی اکسیدانی سرم تعیین شد.
یافته هادر آزمون حافظه احترازی غیرفعال تفاوت معنی داری از نظر زمان تاخیر اولیه بین گروه های آزمایشی وجود نداشت. زمان تاخیر ثانویه در گروه های دریافت کننده اسانس 75 میلی گرم، تمرینات بدنی، تمرینات بدنی+ اسانس 50 میلی گرم و تمرینات بدنی+ اسانس 75 میلی گرم به طور معنی داری بیشتر از گروه کنترل بود. ظرفیت آنتی اکسیدانی سرم نیز در گروه های تمرینات بدنی و تمرینات بدنی+ اسانس 75 میلی گرم بیشتر از گروه کنترل بود.
نتیجه گیریاسانس بابونه و تمرینات بدنی به صورت مجزا و توام دارای اثرات تقویت دهندگی حافظه در موش صحرایی هستند.

مورفین پتانسیل اعتیادآوری بالایی دارد و می تواند مورد سوء مصرف قرار گیرد. استفاده طولانی مدت مورفین اثرات جانبی زیادی دارد و از آن جمله می توان به سمیت عصبی، اختلال عصبی، استرس اکسیداتیو و آپوپتوز اشاره نمود. شواهد زیادی نشان داده است که ملاتونین اثرات آنتی اکسیداتیو و آنتی آپوپتوز دارد با این حال، اثرات آن بر سمیت عصبی ناشی از مورفین در سلول های PC12 مطالعه نشده است. این مطالعه به منظور بررسی نقش محافظتی ملاتونین بر سمیت عصبی ناشی از مورفین در سلول های PC12 انجام شد.
روش هاسلول های PC12 به مدت 24 ساعت با غلظت های مختلف ملاتونین (10، 25، 40 و50 میکرومولار) درمان شده و سپس به مدت 24 ساعت در معرض مورفین با غلظت 2/3 میلی مولار قرار گرفتند. زنده مانی سلول ها با استفاده از روش MTT اندازه گیری شد. اثرات آنتی اکسیدانی ملاتونین با ارزیابی میزان تولید گونه های فعال اکسیژن (ROS)، ظرفیت آنتی اکسیدانی تام (TAP)، پراکسیداسیون لیپیدی (LPO)و گروه های تیول مورد بررسی قرار گرفت.
یافته هاملاتونین به طور قابل توجهی زنده مانی سلول های مواجه شده با مورفین را افزایش داد و سلول های PC12 را در برابر تولید بیش از اندازه گونه های فعال اکسیژن محافظت کرد. همچنین ملاتونین میزان TAP و گروه های تیول را افزایش و میزان LPO را کاهش داد.
نتیجه گیریدر سلول های PC12 ملاتونین می تواند با اثرات آنتی اکسیدانی، سمیت عصبی ناشی از مورفین را کاهش دهد.

بیماری آلزایمر یک اختلال تحلیل برنده نورونی است که موجب اختلال حافظه، تغییرات شخصیتی و رفتارهای غیر معمول می گردد. هدف از انجام این مطالعه بررسی اثر آمپاکین فارامپاتور (Farampator) بر نقص حافظه کاری در موش های صحرایی آلزایمری بود.
روش هادر این مطالعه از 40 سر موش صحرایی نر نژاد ویستار (250 تا 300 گرم) استفاده شد که به 5 گروه آزمایشی تقسیم شدند: (1) گروه شم/کنترل (تزریق درون هیپوکمپ بافر فسفات lµ 1/PBS)، (2) گروه موشهای دارای آلزایمر (تزریق درون هیپوکمپ آمیلویید بتا/PBS l4µg/1µ)، (3 و 4) گروه موشهای دارای آلزایمر + تیمار با فارامپاتور (µg/kg 300 و 30 / خوراکی/ دوبار در روز) و (5) گروه موشهای دارای آلزایمر + تیمار با حلال فارامپاتور (سالین/ خوراکی/ دوبار در روز). جهت بررسی حافظه کاری از آزمون اندازه گیری رفتار تناوبی در ماز Y-شکل استفاده شد.
یافته هاتزریق درون هیپوکمپ آمیلویید بتا 42-1 باعث بروز نقص حافظه کاری در گروه موشهای دارای آلزایمر در مقایسه با گروه شم/کنترل گردید (0/01 >p). تجویز خوراکی آمپاکین فارامپاتور با دوز µg/kg 300 بطور معناداری باعث بهبود عمکرد حافظه کاری در موشهای دارای آلزایمر شد (0/05 > p). تجویز خوراکی µg/kg 30 فارامپاتور تاثیر معناداری در بهبود عملکرد حافظه کاری در موشهای دارای آلزایمر نداشت.
نتیجه گیریآمپاکین فارامپاتور می تواند باعث بهبود نقص حافظه کاری در مدل آزمایشگاهی آلزایمر در موش هایی صحرایی شود.

گزارش شده است که یایسگی با افزایش استرس اکسیداتیو و به دنبال آن اختلال حافظه و اضطراب همراه است. با توجه به خاصیت آنتی اکسیدانی آسکوربیک اسید، اثر این ماده بر استرس اکسیداتیو، اختلال حافظه کاری و رفتار شبه اضطرابی در موش سوری اوارکتومی شده را بررسی کردیم.
روش ها48 سر موش سوری ماده در محدوده وزنی 5±20 گرم بطور تصادفی به شش گروه 8 تایی شامل گروه کنترل آسکوربیک اسید، شم، اوارکتومی تیمار نشده و اوارکتومی تیمار شده با دوزهایmg/kg 500، 300، 100 آسکوربیک اسید تقسیم شدند. نخست حیوانات اوارکتومی شدند و پس از تایید صحت اوارکتومی شدن، به مدت یک ماه با آسکوربیک اسید یا آب گاواژ شدند. سپس حافظه کاری (با آزمون ماز تی شکل)، رفتار شبه اضطرابی (با آزمون ماز مرتفع صلیبی) و سطح فعالیت گلوتاتیون پر اکسیداز در بافت مغز موش ها تعیین گردید.
یافته هادر موشهای اوارکتومی تیمارشده، آسکوربیک اسید با دوز mg/kg100 (0/05<p) و mg/kg500 (0/001< p) حافظه کاری را نسبت به گروه اوارکتومی تیمارنشده بهبود بخشید. آسکوربیک اسید با دوزهای mg/kg 300 و mg/kg 500 در حیوانات اوارکتومی در مقایسه با گروه اوارکتومی تیمار نشده مدت زمان گذرانده شده در بازوی باز را افزایش داد (0/01<p). همچنین در گروه اوارکتومی تیمار شده با دوزهای mg/kg 300 (0/01<p) و mg/kg 500 (0/001p< ) آسکوربیک اسید، درصد دفعات ورود حیوان به بازوی باز افزایش یافت. سطح فعالیت گلوتاتیون پر اکسیداز در گروه اوارکتومی تیمارشده با آسکوربیک اسید با دوز mg/kg 500 نسبت به گروه اوارکتومی تیمارنشده افزایش یافت (0/01<p).
نتیجه گیریآسکوربیک اسید به خصوص با دوز mg/kg 500، نقش موثری در بهبود اضطراب، اختلال حافظه کاری و همچنین افزایش سطح فعالیت آنزیم گلوتاتیون پر اکسیداز در موش های سوری اوارکتومی ایفا کرد.

مطالعات نشان داده است که داروهای نیروزا مخصوصا استروییدهای آنابولیک باعث آسیب کبدی می شوند. مطالعه ای تاکنون در رابطه با تاثیر داروهای نیروزا بر روی کبد ورزشکاران پرورش اندام در شهر بوشهر (ایران) انجام نگرفته است. هدف این مطالعه، بررسی عوارض کبدی داروهای نیروزا مخصوصا استروییدهای آنابولیک در ورزشکاران پرورش اندام بوشهر بود.
روش هااطلاعات جمعیت شناختی، نحوه ورزش، مصرف داروهای نیروزا، مصرف الکل و سایر مکمل های مجاز ورزشی ورزشکاران مرد پرورش اندام با استفاده از پرسشنامه، جمع آوری شد. شرکت کنندگان در مطالعه به سه گروه تقسیم شدند: 1) مصرف کننده مواد نیروزا در زمان مطالعه، 2) عدم مصرف در زمان مطالعه، و 3) غیرمصرف کننده. میزان سرمی آنزیم های کبدی آلانین ترانس آمیناز، آسپارتات ترانس آمیناز و بیلی روبین ورزشکاران توسط کیت های اختصاصی اندازه گیری شد. داده ها با استفاده از آنالیز واریانس یک طرفه بررسی شدند.
یافته ها203 ورزشکار پرورش اندام در محدوده سنی 50-18 سال، با شاخص توده بدنی 40-18/6 کیلوگرم/مترمربع و متوسط سابقه ورزشی 6 سال وارد مطالعه شدند. در این مطالعه، 8/9 درصد ورزشکاران در زمان مطالعه مصرف کننده مواد نیروزا بودند، 39/9 درصد ورزشکاران در زمان مطالعه مواد نیروزا مصرف نمی کردند و 51/2 درصد اصلا مواد نیروزا مصرف نکرده بودند. ورزشکاران مصرف کننده داروهای نیروزا از استروییدهای آنابولیک، ترکیبات محرک، هورمون رشد و انسولین استفاده کرده بودند. متوسط مدت مصرف داروهای نیروزا در ورزشکاران استفاده کننده حدود 60 روز در سال بود که این ترکیبات را به صورت خوراکی و تزریقی به صورت هم زمان استفاده کرده بودند. میزان سرمی آنزیم های آلانین ترانس آمیناز (0/003 =p)، آسپارتات ترانس آمیناز (0/00 =p)، و همچنین بیلی روبین (023 =p) به صورت معنی داری در بین سه گروه مورد مطالعه متفاوت بود. میزان سرمی آلانین ترانس آمیناز (0/003 =p) و آسپارتات ترانس آمیناز (0/00 =p) در ورزشکاران "مصرف کننده مواد نیروزا در زمان مطالعه" بیشتر از ورزشکاران غیرمصرف کننده بود، در حالیکه تنها میزان آنزیم آسپارتات ترانس آمیناز در ورزشکاران "غیرمصرف کننده مواد نیروزا در زمان مطالعه" بیشتر از ورزشکارانی بود که اصلا مواد نیروزا مصرف نکرده بودند (0/044 =p).
نتیجه گیریمصرف استروییدهای آنابولیک همراه با سایر داروهای نیروزا باعث افزایش میزان آنزیم های کبدی در ورزشکاران پرورش اندام می شود که ممکن است مقدمه ای بر بروز آسیب کبدی ناشی از این داروها در ورزشکاران پرورش اندام باشد.

هورمون های تیروییدی در تنظیم رشد سلولی، متابولیسم و میزان عناصر بدن نقش دارند. این هورمون ها و عناصر مس و روی بر باروری موثر هستند. هدف از این مطالعه بررسی اثر کم کاری تیرویید بر میزان مس و روی در مایع منی موش صحرایی بود.
روش ها24 سر موش صحرایی بالغ نر به سه گروه شامل گروه کنترل، گروه تیمار دوز پایین (20 میلی گرم متی مازول در 100 میلی لیتر آب آشامیدنی) و گروه تیمار دوز بالا (100 میلی گرم متی مازول در 100 میلی لیتر آب آشامیدنی) تقسیم شدند. در پایان یک دوره 42 روزه تیمار، تحت بیهوشی عمیق از حیوانات خونگیری بعمل آمد. نمونه های سرم از خون و مایع منی از وزیکول سمینال جمع آوری شدند. بدن و بیضه حیوانات وزن شدند. هماتوکریت تعیین شد. تیروکسین سرمی با روش رادیو ایمنو اسی و عناصر مس و روی با روش اسپکترومتر انتشاری همراه طیف نوری اندازه گیری شدند.
یافته هامتی مازول، سبب کاهش معنی دار و وابسته به دوز تیروکسین سرم (0/001< p)، روی مایع منی (0/001 < p) و وزن نسبی بیضه (0/01 < p) شد. هماتوکریت در هر دو گروه با کم کارتیروییدی کاهش یافت (0/001 < p). مقدار مس مایع منی و وزن بیضه در گرو ه تیمار با دوز بالای متی مازول نسبت به گروه کنترل کاهش نشان داد (0/01 < p).
نتیجه گیریمتی مازول یا کم کاری تیرویید حاصل از آن باعث کاهش روی مایع منی و کاهش وزن نسبی بیضه و هماتوکریت شد درصورتیکه کاهش مس پلاسمای منی فقط در دوز بالا رخ داد.

Chloride channels are present in mitochondrial membranes where they regulate volume and acidity of cells and organelles, cell signaling, and protective mechanisms of the cells. We have previously reported the presence of a 301 pS chloride channel in mitochondrial inner membrane of the rat brain. In this work we characterized biophysical properties of a new chloride channel in the mitochondrial inner membrane of the rat brain.
MethodsThe rat brain was harvested and homogenized in the MSE-nagarase lysis buffer. The homogenate was centrifuged in 3 steps in MSE-digitonin, H2O, Na2CO3, respectively. Vesicles of mitochondrial inner membrane were then extracted in MSE solution. The membrane lipid L-α-Phosphatidylcholine was extracted from fresh egg yolk. Bilayer lipid membrane was formed in a 200 μm diameter hole. All recorded data were filtered at 1 kHz and stored at a sampling rate of 10 kHz for offline analysis by PClamp10 software. Statistical analysis was performed by Markov's noise-free single channel analysis.
ResultsChannel incorporation into planar lipid bilayer revealed a selective anion channel with a conductance of 158 pS in 200 mM KCl cis/50 mM KCl trans electrolyte. The channel open probability appeared to be voltage dependent as the channel was very active at the voltages between ± 20 mV. Adding the chloride channel inhibitor DIDS in the side corresponding to the cell internal medium (cis) caused a strong inhibition of the channel activity.
ConclusionOur results indicate the presence of a chloride channel in the mitochondrial inner membrane of the rat brain. This channel might play a role in maintaining proper pH level, regulation of membrane potential, ATP synthesis, and cell protection.

Vitiligo, psoriasis and some skin cancers are due to abnormal melanogenesis and disturbance in melanocytes homeostasis. Aryl hydrocarbon receptor (AHR) is a ligand-dependent transcriptional factor involved in many physiological processes including drugs metabolism, development of immune system, cellular homeostasis, regulation of cell cycle, and as an essential factor in melanogenesis. The aim of this study was to investigate whether the NADPH oxidases (NOXs) system can interfere with the normal function of aryl hydrocarbon receptor in melanogenesis.
MethodsB16/F10 mouse melanoma cell line were cultured in DMEM and treated with different concentrations of the endogenous ligand of AHR, 6-formylindolo[3,2-b] carbazole (FICZ) (1-100 nM) alone or in combination with an AHR antagonist 3'-Methoxy-4'-Nitroflavone, MNF, or cytochrome P450 type CYP1A1 inhibitor, or a NADPH oxidase (NOX) inhibitor, curcumin (1µM), for different time periods. The activity of CYP1A1 and tyrosinase enzymes as well as total melanin level were measured at the end of treatments.
ResultsCYP1A1 enzyme activity was induced in B16/F10 cells by FICZ in a dose and time dependent manner. Tyrosinase activity and melanin level also were increased by FICZ 100nM. Importantly, treatment of the cells with combination of FICZ 1nM and curcumin induced melanin synthesis.
ConclusionTaken together, inhibition of metabolic degradation of FICZ might elicit biological/toxicological effects on skin homeostasis.

Learning and memory play a key role in daily human life. Memory is the basis of all teaching and training programs. Identification of compounds that improve learning and memory would be helpful for people with mental and memory impairment. This study aimed to evaluate the effects of physical training and essential oil (EO) of Matricaria chamomilla on passive avoidance memory in rats.
MethodsForty-six rats (250-300g) were randomly divided into 6 groups of 8 animals, including control group, group that received 50 mg/kg M. chamomilla EO, group that received 75mg/kg M. chamomilla EO, group that received physical training +50 mg/kg M. chamomilla EO, groups that received physical training +75 mg/kg M. chamomilla EO. Animals were treated with physical training and M. chamomilla EO for 10 days. Passive avoidance learning was evaluated after the 6th EO administration for 4 days using shuttle box apparatus. Blood samples were then collected and serum antioxidant capacity was determined.
ResultsIn passive avoidance test, there was no significant difference between experimental groups in initial latency. The secondary latency in groups receiving 75 mg/kg EO, physical training, physical training+50 mg/kg EO and physical training +75 mg/kg EO was significantly higher than the control group. Serum antioxidant capacity in physical training and physical training +75 mg/kg EO groups were higher than the control group.
ConclusionPhysical training and chamomile essential oil alone or in combination improve learning and memory in rats.

Morphine has a high addictive potential and can be abused. Long-term use of morphine is associated with some pathological consequences including neurotoxicity, neuronal dysfunction, oxidative stress and apoptosis. Increasing evidence has shown that melatonin has antioxidant and antiapoptosis properties. However, its effects on morphine-induced neurotoxicity in PC12 cells have not been studied. This study was carried out to evaluate protective effect of melatonin against morphine-induced neurotoxicity in PC12 cells.
MethodsPC12 cells were treated for 24 h with several doses of melatonin (10, 25, 40, and 50 μM). The cells were then treated with 3.2 mM morphine for 24 h. The viability of PC12 cells was measured using MTT assay. The antioxidant activity of melatonin was evaluated by measuring the levels of reactive oxygen species (ROS), lipid peroxidation (LPO) and total thiol groups, and total antioxidant power (TAP).
ResultsMelatonin significantly enhanced the viability of PC12 cells and protected the cells against morphine-induced overproduction of ROS. Melatonin has increased TAP and total thiol groups and decreased LPO.
ConclusionMelatonin attenuates neurotoxicity of morphine in PC12 cells via antioxidant activity.

Alzheimer’s disease (AD) is a neurodegenerative disorder that can cause memory loss, personality changes and unusual behavior. The purpose of this study was to investigate the effect of ampakine Farampator on impairment of working memory in rat model of Alzheimer's disease.
MethodsForty male Wistar rat (250-300g) were randomly divided into five experimental groups including: Group 1: Sham/control (intra-hippocampal microinjection of phosphate buffer solution (PBS)/1μl); Group 2: Rat model of Alzheimer's disease (intra-hippocampal microinjection of 4μg/1μl amyloid beta 1-42), Groups 3 & 4: Rat model of Alzheimer's disease + Farampator (30 and 300 µg/kg/by oral gavage/ twice per day); Group 5: Rat model of Alzheimer's disease + Farampator vehicle (saline/ by oral gavage / twice per day). Y-maze spontaneous alternation test was used to evaluate the working memory.
ResultsIntra-hippocampal injection of amyloid beta 1-42 caused significant defects in working memory in rat model of Alzheimer's disease (p < 0.01). Farampator 300 µg/kg significantly improved working memory task in rat model of Alzheimer's disease (p < 0.05). Farampator 30 µg/kg had no significant effect on working memory impairment in rat model of Alzheimer's disease.
ConclusionAmpakine farampator can improve the working memory defect in rat model of Alzheimer's disease.

It has been reported that menopause is associated with increased oxidative stress and subsequent memory deficits and anxiety. According to the antioxidant property of Ascorbic Acid (AA), we tested the effect of AA on oxidative stress, impairment of working memory and anxiety like-behavior in ovariectomized mice.
MethodsForty-eight female mice weighting 20 ± 5 g were randomly divided into six groups of eight including AA control group, sham, untreated ovariectomized, and ovariectomized treated with the doses of (100, 300, 500 mg/kg, by oral gavage for 1 month) of AA. Then, working memory (by T-maze), anxiety like-behavior (by Elevated Plus-maze) and the activity of Glutation peroxidase (GPx) in mice brain tissues were determined.
ResultsAA by the doses of 100 (p < 0.05) and 500 mg/kg (p < 0.001) improved working memory of ovariectomized mice compared to untreated ovariectomized group. AA by the doses of 300 and 500 mg/kg increased Open Arm Time (%OAT) of ovariectomized animals compared to untreated ovariectomized group (p < 0.01). Open Arm Entries (%OAE) were also increased in ovariectomized treated group by the dose of 300 (p < 0.01) and 500 mg/kg (p < 0.001) of AA. The activity level of GPx increased in ovariectomized mice treated by AA 500 mg/kg compared to untreated ovariectomized group (p < 0.01).
ConclusionAA especially at the dose of 500 mg/kg showed a significant improvement in anxiety, working memory as well as the level of GPx activity in ovariectomized mice.

Studies have shown that performance-enhancing drugs (PEDs) especially anabolic steroids cause liver damage. There is no study on the PEDs effects on the liver of bodybuilding athletes in Bushehr city (Iran). Aim of this study was to evaluate the hepatic effects of PEDs, particularly anabolic steroids in male bodybuilders in Bushehr.
MethodsDemographic information, exercise program, use of PEDs, alcohol and dietary supplements were recorded in male bodybuilders using a questionnaire. Participants were divided into three groups: 1) current users (CU), 2) non-current users (NCU), and 3) non-users (NU). The serum levels of alanine transaminase (ALT), aspartate transaminase (AST) and bilirubin were measured in the athletes by using the commercially available kits. The data were analyzed by using one-way analysis of variance.
ResultsTwo-hundred and three bodybuilders with the age range of 18-50 years, body mass index of 18.6-40 kg/m2, and average of 6 years exercise were included in the study. In the present study, 8.9% of athletes were current users, 39.9% were non-current users, and 51.2% were non-users. Athletes have been used anabolic steroids, stimulants, growth hormone, and insulin. The average period of PEDs use in athletes was 60 days/year by simultaneous oral and injection routes. The serum levels of ALT (p = 0.003), AST (p = 0.000) and bilirubin (p = 0.023) were significantly different between the three groups. The serum levels of ALT (p = 0.000) and AST in current users were higher than non-user athletes, while only the AST level in non-current users was higher than non-users (p = 0.044).
ConclusionThe use of anabolic steroids with other PEDs may raise the serum level of hepatic enzymes in male bodybuilders. This effect may be the sign of PEDs-induced hepatic injury in these athletes.

Thyroid hormones play a role in cell growth regulation, metabolism, and body trace elements. These hormones, copper and zinc are effective on fertility. The aim of this study was to evaluate the effect of thyroid function on the amount of copper and zinc in rat seminal plasma.
MethodsTwenty four male Wistar rats divided in 3 groups (n = 8) including: control group, hypothyroid groups which treated by methimazole low dose (20 mg in 100 ml of drinking water) and which treated by methimazole high dose (100 mg in 100 ml of drinking water). At the end of a 42-day treatment period, blood samples were taken from animals under deep anesthesia. Serum samples were collected from blood, and semen plasma samples were collected from seminal vesicle. The body and testis of animal were weighted. Hematocrit (HCT) was determined. Serum levels of thyroxin hormone (T4) were measured by Radioimmunoassay. The content of the Copper (Cu) and Zinc (Zn) in the seminal plasma was determined with inductively coupled plasma optical emission spectrometry.
ResultsMethimazole decreased the serum T4 level (p < 0.001), Zn in the seminal plasma (p < 0.001) and relative weight of the testes (p < 0.01) in a significant and dose-dependent manner. Hematocrit level decreased in two hypothyroid groups (p < 0.001). The testis weight and Cu in the seminal plasma reduced in the high dose treatment group (p < 0.01).
ConclusionMethimazole or hypothyroidism induced by methimazole attenuated zinc concentration in seminal plasma, testes relative weight and Hematocrit level. However, copper reduction in the seminal plasma occurred only by high dose of methimazole.
