    لپتوسپیروز یکی از بیماری های مهم قابل انتقال از حیوان به انسان است. LipL32 یکی از مهمترین پروتئین های غشای خارجی لپتوسپیرا است که تنها در سرووارهای بیماری زا وجود دارد و فاکتور مناسبی جهت شناسایی لپتوسپیراهای بیماری زا می باشد. بنابراین هدف از انجام این پژوهش، تشخیص مولکولی لپتوسپیراهای بیماری زا بر اساس ژن lipL32 می باشد. در این بررسی از سرووارهای بیماری زا و غیر بیماری زای استاندارد لپتوسپیرا استفاده شد. باکتری ها در محیط کشت اختصاصیEMJH  کشت داده شدند و DNA ژنومیک آن ها با روش استاندارد فنول کلروفرم ایزوآمیل الکل تخلیص گردید. پرایمر اختصاصی جهت تکثیر ژن lipL32طراحی شد. دمای اتصال (Annealing) برای این پرایمر 61 درجه سانتی گراد، غلظت مناسب پرایمر 10 پیکومول ، میزان حساسیت   4-10 pg/µl و همچنین تعیین ویژگی آن در PCR مورد بررسی قرار گرفت. با توجه به داده های PCR تایید شد که این ژن فقط در سرووارهای بیماری زای لپتوسپیرا حضور دارد و قطعه bp 272 حاصل که نشان دهنده تکثیر  ژن  lip L32 می باشد، در سرووار غیر بیماری زا مشاهده نگردید.  شناسایی روش های سریع تشخیص لپتوسپیراهای بیماری زا پیش از ورود به فاز حاد بیماری بسیار حایز اهمیت می باشد. بنابراین جهت تشخیص لپتوسپیراهای بیماری زا، روش های مولکولی مبتنی بر PCR با استفاده از ژن lipL32که در کوتاه ترین زمان ممکم پاسخی دقیق و قابل اعتماد ارایه می دهند، پیشنهاد می گردد.

    یکی از توصیه های مهم انجمن های مرتبط با حیوانات آزمایشگاهی، استفاده از حیوانات با فلور میکروبی مشخص است زیرا که نتایج کار بر روی آنها بایستی تکرار پذیر، مطمین و قابل تعمیم باشد. بنابراین انجام پایش های بهداشتی حیوانات جهت صدور گواهی وضعیت سلامت آنها در مراکز تولید و پرورش حیوانات آزمایشگاهی الزامی است. از میان ویروس های توصیه شده توسط FELASA ، ویروس تیلر عامل انسفالومیلیت از مهمترین آنها بوده  که در این تحقیق به بررسی حضور آن در جمعیت موش های آزمایشگاهی نژاد NIH یک مرکز پرورش حیوانات آزمایشگاهی پرداخته شده است. روش تحقیق در این بررسی، RT-PCR ، با استفاده از پرایمرهای اختصاصی ویروس مورد نظر بوده است. طراحی و تهیه نمونه کنترل مثبت، با استفاده از ترانسفورمیشن پلاسمید حاوی مناسب ترین قطعه ژن ویروس در  E.coli  جهت تکثیر ژن بود و سپس استخراج آن صورت گرفت. نمونه ها  از بافت روده (کولون) حاوی مدفوع حیوانات، تهیه و به روش استاندارد آماده سازی و سپس روش RT-PCR انجام گرفت. نتایج نشان می دهد تعداد 13 نمونه از 29 نمونه و با میزان شیوع 45% در بین موش های نژاد NIH  مبتلا به این ویروس بوده اند. محصول  PCR  انجام شده تخلیص و تعیین توالی  گردید. در بررسی همترازی (alignment) جهت تعیین استرین، با توالی های هشت گونه دیگر کسب شده از  بانک ژن NCBI مقایسه شد. در درخت فیلوژنی، سویه KX774639 0.15874  متعلق به این تحقیق بیشترین قرابت را، با گونه M20562 از کشور اتریش و گونه NC_001366  از کشور انگلیس نشان داد. از آنجاکه این عفونت قابل انتقال به انسان نیست، حذف فوری کلنی ضرورت ندارد.

    کشت سلول برای تولیدات بیولوژیکی و دارویی علی الخصوص دارو های دامی اهمیت ویژه ای دارد این تحقیق(کشت سلول) در شرایط کشت سوسپانسیون ، انجام شده است. برای بهینه سازی مکمل های غذایی موجود در محیط کشت از روش های متفاوتی مانند تغییر تک پارامترو ثابت نگه داشتن مابقی پارامترها(فاکتوریل) استفاده میشود که بسیار زمان برو دارای خطای بالایی میشود. در این مقاله از روش طراحی آزمایش به روش تاگوچی استفاده شده که نسبت به روش های دیگر دقت بالاتر ،زمان کمتر و تعداد آزمایش کمتری انجام میشود. . با استفاده از این روش طراحی آزمایش سعی بر بهینه سازی محیط کشت مورد استفاده در پژوهش شد.با توجه به تعداد سلول نهایی،  ضریب تزاید و زمان دو برابر شدن و نرخ ویژه رشد هر آزمایش محاسبه گردید و در باره اهمیت هرکدام از مکمل های غذایی و اثر آن ها بر روی سلول کلیه نوزاد همستر(Baby Hamster Kidney) اعمال نظر شده است.هرچه تعداد سلول نهایی بیشتر باشد در نتیجه زمان دو برابر شدن کمتر  و نرخ ویژه رشد بیشتر می شود،که حاصل تمامی این ها باعث می شود کشت سلول بهره وری بیشتری داشته باشد. هدف از  این تحقیق بررسی  بر روی تاثیر مکمل های غذایی مختلف موجود در محیط کشت بر رشد و تکثیر سلول کلیه نوزاد همستر(BHK)متمرکز شده است. در فرآیند کشت سلول نتایج بدست آمده نشان دهنده بهبود رشد و تکثیر سلول ها شد. افزایش تعداد سلول به صورت کلی (در این پژوهش سلول BHK) موجب تولید بیشتر و با کیفیت بالاتر  داروهای بیولوژیک می شود.میزان سرم اضافه شده در محیط کشت  در این روش (10%) بیشترین تاثیر مثبت بر روی ضریب تزاید ، زمان دو برابر شدن و نرخ ویژه رشد را دارد و هم چنین افزایش میزان اسید های آمینه(12 گرم در هر لیتر) تاثیری بر روی ضریب تزاید ، زمان دو برابر شدن و نرخ ویژه رشد ندارد.

    یکی از ضدعفونی کننده های پرکاربرد در صنعت طیور کشور، هیپو کلریت سدیم می باشد. باکتری اشریشیا کلی </em>(E. coli)</em> یکی از مهمترین میکروارگانیسم هایی است که توسط این ضدعفونی کننده کنترل می شود. امروزه، از باکتری E. coli</em>به عنوان باکتری مهمی در نگهداری و انتقال مقاومت های چندگانه ی دارویی و ضدعفونی کننده ها یاد می شود که می تواند این مقاومت را به سایر باکتری ها نیز منتقل کند. اینتگرون ها از جمله قطعات ژنی مهمی هستند که از طریق پذیرش و بیان ژن های مقاومت در انتقال و بروز مقاومت در باکتری های مختلف نقش دارند. در مطالعات گذشته، در کاست ژنی اینتگرون کلاس1 علاوه بر ژن های مقاومت به داروها و مواد ضدعفونی کننده ترکیبات چهارتایی آمونیوم (ژن های qacE</em>) ، توالی هایی با قالب خواندن باز (ORF)، شناسایی شده اند. تاکنون چندین ORF که ژن هایی با عملکرد ناشناخته هستند، در کاست ژنی اینتگرون کلاس1 شناسایی شده اند. در مطالعه حاضر، فراوانی اینتگرون کلاس 1و نیز ژن orfF</em> به روش Multiplex PCR در باکتری های اشریشیا</em> کلی</em> جدا شده از طیور و نقش آن ها در القای مقاومت باکتری به ضدعفونی کننده  هیپوکلریت سدیم با آزمایش کمترین غلظت مهاری بررسی گردید. در این مطالعه فراوانی اینتگرون کلاس1، 61 درصد به دست آمد. فراوانی حضور اینتگرون کلاس1 در میان جدایه های حاصل از نمونه های مدفوع و یا پریکارد تفاوت معناداری نداشتند. همچنین هیچ گونه ارتباط معنی داری بین فراوانی حضور اینتگرون کلاس1 و القای مقاومت به هیپوکلریت سدیم دیده نشد (05/0P>). فراوانی ژن orfF</em> نیز 8 درصد بود که ارتباط آماری معنی داری با حضور اینتگرون کلاس1 و القای مقاومت به هیپوکلریت سدیم دیده نشد  (05/0P>) . علی رغم وجود فراوانی بسیار بالای اینتگرون کلاس1 در میان جدایه های مورد آزمایش، با توجه به عدم وجود ارتباط آماری بین اینتگرون کلاس1 و مقاومت به هیپوکلریت سدیم می توان نتیجه گرفت که ژن های مقاومت به این ضدعفونی کننده از طریق کاست ژنی اینتگرون کلاس1 منتقل نمی شوند و نیاز به مطالعات بیشتری در زمینه درک مقاومت به این ضدعفونی کننده وجود دارد.

    مشمشه یکی از مهم ترین بیماری های قابل انتقال از حیوانات تک سمی بوده که عامل آن بورخولدریا مالیی است. با توجه به کانون بیماری در ایران و احتمال آلودگی انسان و خطرات ناشی از انتقال این باکتری، شناسایی سریع آن در حیوانات از اهمیت زیادی برخوردار می باشد. لذا هدف از این مطالعه طراحی یک روش تشخیص سریع شامل لکه نمایی نقطه ای برای تعیین حضور حضور آنتی بادی بود. برای این منظور 40 نمونه سرم اسب از  استان های مختلف اخذ و به همراه نمونه آنتی ژن پیکره باکتری بورخولدریا مالیی (کنترل مثبت) و سرم اسب های سالم (کنترل منفی) مورد آزمون قرار گرفتند. غلظت بهینه آنتی ژن-آنتی بادی با استفاده از چکر بورد برای سرم های مثبت و منفی مشخص گردید. پس از طراحی آزمون لکه گذاری نقطه ای، آنتی ژن روی چاهک کاغذ نیتروسلولز قرار داده شد و تثبیت آنتی ژن  با محلول بلاکینگ و سپس افزودن سرم اسب مشکوک به همراه کنترل مثبت و منفی انجام شد. در ادامه به آن آنتی بادی کونژوگه اضافه و نهایتا با افزودن سوبسترا به چاهک ها نتیجه ی واکنش به صورت ایجاد رنگ قرایت گردید. در این بررسی از 40 نمونه، شش مورد مثبت شناسایی گردید. با در نظر گرفتن مالییناسیون به عنوان آزمون استاندارد و مقایسه آن با لکه گذاری نقطه ای به ترتیب حساسیت و ویژگی 100٪ و 9/91٪  شد. نتایج نشان داد روش لکه گذاری نقطه ای به دلیل حساسیت بالا، سرعت، سهولت قرایت و قابلیت کاربردی صحرایی آن، در صورت تایید مراجع مسیول تشخیص در عفونت با بورخولدریا مالیی می تواند استفاده شود.

    واکنش زنجیره‎ای پلیمراز چندگانه (PCR Multiplex) یک روش پیشرفته از تکنیک های تشخیص مولکولی است که امکان شناسایی همزمان چند عامل بیماریزا را در یک نمونه فراهم می کند. مزایای استفاده از روش واکنش زنجیره‎ای پلیمراز چندگانه در مقایسه با نتایج تشخیص به دست آمده با روش های متداول دیگر توسط محققان مختلف بررسی شده است. در این پژوهش از PCR چندگانه طراحی شده در آزمایشگاه به منظور شناسایی هم زمان عوامل مهم سقط جنین گوسفندان شامل: Brucella melitensis ، Salmonella abortusovis ، Chlamydophila abortus وCoxiella burnetii ، استفاده شد. برای این منظور تعداد 98 نمونه محتویات شیردان جنین های سقط شده از سه منطقه جغرافیایی ایران شامل استان های اصفهان، چهارمحال و بختیاری و خراسان رضوی، مورد آزمایش قرار گرفت. میزان آلودگی در نمونه های بررسی شده، 15/3 درصد  Brucella melitensis، 11/2 درصد Salmonella abortusovis، 7/1 درصد Chlamydophila abortus و صفر درصد Coxiella burnetii بود. در کل نتایج این پژوهش نشان می دهد که  Brucella melitensis ، Salmonella abortusovisوChlamydophila abortusاز مهمترین عوامل سقط جنین نشخوارکنندگان کوچک بوده و از آزمون PCR چندگانه، به خوبی می توان برای تشخیص عوامل سقط جنین در گوسفند و بز استفاده کرد.

    آنفلوانزای فوق حاد پرندگان بعنوان بیماری جدی عفونی، توسط یکRNA ویروس سنس منفی متعلق به جنس آنفلوانزا و خانواده اورتومیکسوویریده ایجاد می شود. ویروس های نوع A بر اساس آنتی ژنهای گلیکوپروتئینی سطح آنها شامل هماگلوتینین(HA) و نورامینیداز (NA) به زیر گروه هایی طبقه بندی می شوند. ویروس آنفلوانزای پرندگانH5N1 با حدت بالا (HPAI)، از طریق مرغ های خانگی و پرندگان وحشی در بیش از 60 کشور منتشر شده است. در این مقاله به نحوه بیان هماگلوتنین H5 در سلولهای Sf9 حشره ای پرداخته شده است که متعلق به ویروس A/chicken/Iran/53-3/2008(H5N1) می باشد. توالی نوکلیوتیدی هماگلوتینین، از سویه ایرانی این ویروس که متعل به clade 2.2 است، از بانک ژنی استخراج شده و پس از بهینه سازی کدون ها برای بیان در سیستم سلول حشره، توسط شرکت GenScript سنتز شد. این توالی در پلاسمید pIEx-3 که حاوی توالی پپتید ترشحی AKH و 6xHis-Tag می باشد، داخل شد. پروتئین هماگلوتینین در سلول Sf9 حشره ای، بیان شده و H5 ترشحی با وزن مولکولی 95 کیلودالتون تخلیص گردید. با استفاده از این سیستم بیانی غیر وابسته به باکلوویروس، H5 نوترکیب، در مطالعات بعدی مربوط به واکسن های مبتنی بر پروتئین، انتخاب داروها و ساخت کیت تشخیص آنفلوانزای فوق حاد پرندگان، قابل استفاده می باشد.

    هدف از انجام این پژوهش برررسی اثر منبع و توازن آنیون -کاتیون جیره (DCAD) گاوهای شیری بر جمعیت پروتوزوآها، متانوژن ها، غلظت نیتروژن آمونیاکی، گاز متان و قابلیت هضم ماده خشک در شرایط آزمایشگاهی بود. برای شمارش جمعیت پروتوزوآها و متانوژن ها از تکنیک واکنش زنجیره ای پلیمراز (PCR)رقابتی استفاده شد. این طرح برای گاو شیری در اوایل شیردهی با میانگین580 کیلوگرم وزن بدن، سن 25 ماهگی و تولید روزانه 35 کیلوگرم در قالب آزمایش فاکتوریل 2×2×3 انجام شد. عوامل موثرها شامل DCAD (meq/kgDM 150+، 250+ و 350+)، منابع پتاسیمی (کربنات پتاسیم بدون آب(KC) و کربنات پتاسیم سسکویی هیدرات (KCS)) و منابع منیزیمی (اکسید منیزیم (MO) و کربنات منیزیم (MC)) بود. اثر تیمارهای آزمایشی بر غلظت نیتروژن آمونیاکی معنی دار نبود. در همه تیمارها افزایش DCAD باعث کاهش غلظت نیتروژن آمونیاکی در شکمبه شد. اثر تیمارهای آزمایشی بر تولید گاز متان، قابلیت هضم ماده خشک، جمعیت پروتوزوآها و متانوژن ها معنی دار بود. نتایج پژوهش حاضر نشان داد که تیمار حاوی DCAD 250+ میلی اکی والان تامین شده از دومنبع کربنات پتاسیم سسکویی هیدرات و کربنات منیزیم باعث کاهش جمعیت پروتوزوآ، متانوژن ها، تولید متان و افزایش قابلیت هضم ماده خشک می شود. با توجه به اینکه اختلاف معنی دار بین DCAD 250+ و 350+ مشاهده نشد، بنظر می رسد که منابع کربنات پتاسیم سسکویی هیدرات و کربنات منیزیم به همراه سطح تعادل آنیون - کاتیون جیره 250+ میلی اکی والان از طریق بهبود شرایط بافری شکمبه باعث بهبود پارامترهای تخمیر شکمبه ای می شود. بنابراین این دو منبع احتمالا مکمل های مناسبی در جیره گاوهای شیری هستند.

    بیماری بروسلوز یا تب مالت، همواره از دو بعد اقتصادی و بهداشتی حایز اهمیت بوده به نحوی که هر ساله خسارت های اقتصادی فراوانی را برای سرمایه دامی کشور به بار می آورد. از سوی دیگر از آنجایی که بروسلوز یک بیماری مشترک بین انسان و دام می باشد، اهمیت موضوع از نظر بهداشت عمومی دو چندان می شود. بنابراین لازم است با انجام مطالعات اپیدمیولوژیک به عنوان گام اول، به سمت شناسایی عامل بیماری و سپس کنترل و کاهش موارد ابتلا به آن در کشور قدم برداشت. در این مطالعه، 170 نمونه سقط جنین مربوط به گوسفند و گاو در طی سال های 94 تا 97 از استان های گلستان و خراسان رضوی جمع آوری گردید. پس از کالبد شکافی، از ارگان های مختلف جنین ها در محیط کلمبیا آگار و مک کانکی کشت داده شد و با استفاده از رنگ آمیزی گرم و تست های استاندارد بیوشیمیایی، تمام نمونه ها مورد بررسی قرار گرفتند. در مجموع، 60 ایزوله بروسلا شناسایی گردید. در نهایت با استفاده از تکنیک های مولکولی Bruce-ladder Multiplex PCR و ERIC-PCR، تفکیک گونه و تایپینگ مولکولی ایزوله های جداسازی شده انجام پذیرفت. از میان 60 ایزوله شناسایی شده، 59 گونه مربوط به بروسلا ملی تنسیس و 1 گونه مربوط به بروسلا آبورتوس بود. تمامی ایزوله های شناسایی شده، علیرغم مناطق جغرافیایی متفاوت، دارای الگوی ژنتیکی مشابهی بودند. تکنیکBruce-ladder Multiplex PCR به عنوان یک روش سریع، دقیق و ایمن جهت شناسایی بروسلا در سطح جنس و گونه بسیار موثر است. با وجود اینکه تکنیک ERIC-PCR در بسیاری از مطالعات روش قدرتمندی در تمایز مولکولی باکتری ها بشمار می رود، در صورت امکان بهتر است که برای تایید نتایج این مطالعه از تکنیک های دقیق تر و با قدرت تفکیک بالا همچون MLVA و MLST نیز استفاده گردد.

    نیوسپورا کانینوم انگل تک یاخته داخل سلولی با گسترش جهانی است. این تک یاخته از عوامل اصلی سقط جنین در گاوهای شیری بوده است. تحقیق حاضر به منظور بررسی میزان شیوع فصلی و جغرافیایی نیوسپورا کانینوم در شیر نشخوارکنندگان به روش مولکولی انجام شد. چهارصد و چهل نمونه شیر خام طی چهار فصل سال از استان های اصفهان، چهارمحال و بختیاری، خوزستان و فارس جمع آوری و به آزمایشگاه منتقل شد. نمونه های DNA استخراج شده با استفاده از کیت بوسیله آزمون nested- PCR به منظور ردیابی ژن NC5 نیوسپورا کانینوم ارزیابی شدند. از مجموع 440 نمونه شیر مورد مطالعه، 54 نمونه (27/12 درصد) آلوده به نیوسپورا کانینوم بودند. شیر گاو بیشترین (26 درصد) و شیر گوسفند کمترین (4 درصد) میزان شیوع نیوسپورا کانینوم را داشتند. نمونه های شیر جمع آوری شده در فصل زمستان بیشترین (85/22 درصد) و نمونه های جمع آوری شده در فصل تابستان کمترین (57/8 درصد) میزان شیوع نیوسپورا کانینوم را داشتند. نمونه های شیر جمع آوری شده از استان چهارمحال و بختیاری بیشترین (94/14 درصد) و نمونه های جمع آوری شده از اصفهان کمترین (38/6 درصد) میزان شیوع نیوسپورا کانینوم را داشتند. با توجه به شیوع نسبی این انگل در شیر خام مورد مطالعه، اتخاذ برنامه های کنترلی بیش از پیش مورد نیاز است. همچنین با توجه به احتمال مصرف فرآورده های لبنی سنتی و انتقال آلودگی به انسان، جوشش کامل شیر قبل از مصرف توصیه می گردد.

    توکسوپلاسما گونده ای یک انگل با انتشار جهانی است که می تواند حیوانات و انسان را آلوده کند. عفونت با این انگل زیونوز عمدتا با مصرف گوشت نیم پز یا خام، که حاوی کیست های بافتی است ایجاد می شود. مطالعه حاضر به منظورپایش میدانی روش MIT درتعیین میزان آلودگی دامهای کشتارگاهی شهرستان رشت به انگل توکسوپلاسماانجام گرفته است. مطالعه حاضر به منظور تعیین شیوع عفونت توکسوپلاسموز در حیوانات اهلی در استان گیلان صورت گرفته است. در این مطالعه تجربی نمونه های سرم از 166 حیوان شامل 48 گاو، 85 گوسفند و 33 بز برای شناسایی آنتی بادی های ضد توکسوپلاسما تهیه شد و با آزمون آگلوتیناسیون اصلاح شده (MAT)بررسی شدند. آنتی بادی های ضد توکسوپلاسما در 30 نمونه سرم به دست آمده از 166حیوان (18/1%) شناسایی گردید. بالاترین میزان عفونت در گوسفند (24/7%) و به دنبال آن آلودگی در گاو (16/7%) و بز (3 %) یافت شد. بیشتر گوسفندان (23%) تیتر آنتی بادی 1:20 داشتند. 8/7 درصد از حیوانات نر و 22/9 از ماده ها سرم مثبت بودند و این تفاوت ازنظر آماری معنی دار بود (5%>p). همچنین برحسب منطقه جغرافیایی اختلاف معنی داری دیده شد (5%>p). شیوع سرولوژیک بالای توکسوپلاسموز که در این منطقه مشاهده شد نشان می دهد که حیوانات اهلی می توانند نقش عمده ای در انتقال عفونت به انسان از طریق مصرف گوشت نیم پز داشته باشند.

    ابیا ها به عنوان یک گروه از هشت گونه ی زنده ی مرغ های پا دراز در جنس اسکولوپکس (Scolopax) در نظر گرفته می شوند که در جنگل های مخلوط یا برگ ریز زندگی و معمولا به صورت منفرد مهاجرت می کنند و همچنین یکی از پرندگانی است که دارای آلودگی های انگلی است. در این پژوهش از مجموع 18 پرنده ابیا اوراسیایی (S. rusticola) (11 نر و 7 ماده) که توسط ماموران محیط زیست مازندران از شکارچیان غیر مجاز ضبط شد، به منظور مطالعه ی انگل های خارجی و داخلی مورد بررسی قرار گرفتند که در نتایج، میزان عفونت کلی انگلی در پرنده های مورد مطالعه 100 % و انگل های خارجی و داخلی پرندگان شامل دو گونه سستود (Hymenolepis spp. و Davinia proglottina)، یک گونه ترماتد (Echinoparyphium)، یک گونه جرب (Megniniacubitalis) و دو گونه شپش (Philopterus و Lipeurus) است. علاوه بر این، شایع ترین آلودگی با 22/72 درصد انگل (M. cubitalis) بوده است. یافته ها و مقایسه ی آن ها با مطالعه های دیگر نشان می دهند که ابیای اوراسیایی با توجه اهمیت مهاجرت، نقش مهمی در انتقال انگل ها داشته و نیاز به تحقیق های بیشتر در مورد این پرنده و سایر پرندگان مهاجر در ایران احساس می شود و همچنین این مطالعه نخستین گزارش انگل های مذکور در پرنده ابیا اوراسایی از ایران است.

    Leptospirosis is one of the most important diseases Transferable from animal to human. LipL32 is one of the most important outer membrane proteins that exist only in pathogenic Leptospira spp., which is a suitable factor for the detection of pathogenic Leptospira. Therefore, the aim of this study is the molecular detection of pathogenic Leptospira based on lipL32 gene. Saprophytic and pathogenic Leptospira serovars were used in this study. The bacteria were inoculated into the selective culture medium and extraction of the genomic DNA was performed by standard Phenol-Chlorophorm method. The specific primers for proliferation of lipL32 gene were designed. In terms of annealing temperature was 61°C, suitable concentration 10 pmol, sensitivity was -2 11 pg/ μl and its characterization in PCR were investigated. According to PCR data, it was confirmed this gene was present only in pathogenic Leptospira serovars and 272 bp fragment indicating the replication of the lipL32 gene and was not observed in non-pathogenic species. Identify methods for rapid detection of pathogenic leptospires before entering the acute phase of the disease is very important. The results showed that PCR using lipL32 gene was high specificity and sensitivity. So for the detection of pathogenic Leptospires, molecular methods based on PCR using lipL32 gene in the shortest possible time offer accurate and reliable response is recommended.

    One of the important recommendations of laboratory animals related associations is the use of animals with defined microbial flora because the results of the work on them should be repeatable, reliable and generalizable. Therefore, animal health monitoring is mandatory for certification of their health status at animal breeding and production centers. Mouse hepatitis virus and theiler's encephalomyelitis virus are the most important recommended by FELASA that to examine them in the NIH Laboratory mice colony in this project. The research methods in this study was RT-PCR, using specific primers of the virus. Designing and preparing a positive control sample was carried out using plasmid transformation containing the most appropriate viral gene fragment in E.coli for amplification and extraction. The samples are collected from intestine (colon) containing feces and prepared by standard methods and RT-PCR was performed. The virus theiler's encephalomyelitis, number 13 out of 29 samples with prevalence of 45% among NIH mice were infected with the virus. PCR product was purified and sequenced. In a strain alignment study, it was compared with the eight other species obtained from the NCBI gene. In the phylogenic tree, the KX774639 0.15874 strain belonged to this research most closely related to M20562 from Austria and NC_001366 from England. Because the infection can not be transmitted to humans, immediate removal of the colony is unnecessary.

    Cell culture is one of the most commonly used techniques in the production of biological products. Many physical and chemical parameters factors may affect cell growth and proliferation. This research (cell culture) has been conducted under conditions of suspension culture. To optimize the nutritional supplements available in the culture medium, different methods are used, such as factor, which takes a long time and has a high error. This study Using the experimental design and Taguchi method, which is more accurate, shorter, and has fewer tests than other methods. . Using this method of experimental design, an attempt was made to optimize the culture medium used in the research. According to the number of final cells, multiplication factor, doubling time and the specific growth rate of each experiment were calculated, and the importance of each of the supplements Food and its effect on the baby hamsters kidney's (BHK) has been considered. The higher the number of ultimate cells, the more time it doubles and the special rate of growth increases, which results in all of these causes cell cultures More productive. The amount of serum added in the culture medium in this method (10%) has the most positive effect on the growth rate, doubling time and specific growth rate. Also, increasing the amount of amino acids (12 gr/li) has no effect on the growth rate, doubling time and specific growth rate.

    The regular use of disinfectants in broiler farms is one of the main bases of the biosecurity. Disinfectants destroy the different disease agents that remained from the last breeding season. Hypochlorite is one of the most suitable disinfectants used in the poultry industry. Nowadays, resistant pathogens to disinfectants and antibiotics are growing worldwide, becoming a serious concern in the public health. Resistance factors are coded by different genes that are commonly present in the different gene cassettes like Integron1 that distribute resistance genes among bacteria. In the previous studies, the relationship between Integron1 and multidrug resistance genes also quaternary ammonium compounds (QACs) resistance genes was clarified. Still, there is no study on the relationship of Integron1 and other disinfectants like Sodium hypochlorite resistance genes. Escherichia coli (E. coli) is one of the most important pathogens of poultry industry. In the present study, 100 isolates of E. coli from the cloak of healthy broilers and from pericarditis cases were studied. Presence of Integron1 gene cassette and orfF gene by Multiplex PCR was detected. Also, the amount of resistance to the Sodium hypochlorite by MIC method was measured. The results showed that there are no significant relationships between the Presence of Integron1 gene cassette and the resistance of the Sodium hypochlorite disinfectant (P<0.05). This result indicates that the genetic reserve and translocation of the resistance gene of Sodium hypochlorite are not related to the Integron1 gene cassette, which shows the need for more studies.

    Glanders is one of the most important zoonotic diseases that caused by Burkholderia mallei. Considering that Iran as the major center for glanders and risks of transmission of it, quick detection is very important. Therefore, the aim of this study is designing of Dot-Elisa system. So, 40 serum samples were collected from different provinces, along with antigen sample of Burkholderia mallei as positive control and healthy horses’ serum (negative control) was used. The optimum concentrations of antigen-antibody were determined by checkerboard titration using glanders positive and negative horse sera. After designing the dot-blot test, an antigen was coated into the nitrocellulose membrane wells and stopped by blocking solution and the suspicious equine serum was poured into the wells along with positive and negative control. In the following conjugated antibody was added on it, finally substrate was added to the wells and reaction was read as color signal. In this study, out of 40 samples, 6 positive cases were positive. Considering mallein test as a standard test and comparing with DotElisa, the sensitivity and specificity were 100% and 91.9% respectively. The results showed that dot-blot test was successful for detection of Glanders due to its high sensitivity, speed, readability and field applicability.

    Multiplex polymerase chain reaction (mPCR) is an advanced method of molecular diagnostic techniques which allows for simultaneous detection of multiple pathogens in the same sample. The advantages of the multiplex PCR method were assessed by various researchers by comparing the diagnosis results obtained with different other conventional methods. An in-house multiplex PCR method was designed and evaluated for detection of Brucella melitensis, Salmonella abortusovis, Chlamydophila abortus and Coxiella burnetii, which are the most frequent agents in ewe’s abortion. For this purpose, 98 samples of abomasal liquid's contents of the aborted fetuses were collected from three geographic areas of Iran include Isfahan, Khorasan Razavi and Chaharmahal and Bakhtiari provinces and tested by multiplex PCR. Brucella melitensis Salmonella abortusovis and Chlamydophila abortus were found in 15.3%, 11.2% of 7.14% of samples respectively. Coxiella burnetii was not found in any sample. In general, the results of this study were showed that Brucella melitensis, Salmonella abortusovis, Chlamydophila abortus are the most important pathogens of ewe’s abortion and this in-house multiplex PCR test can be used to identify the causative agent of abortion in ewe in one tube.

    Highly Pathogenic Avian influenza (AI) is a serious infectious disease caused by negative sense single stranded RNA viruses that belong to the genus Alphainfluenzavirus in the family Orthomyxoviridae. The type A viruses are classified into subtypes based on their surface glycoprotein antigens including hemagglutinin (HA) and neuraminidase (NA). Highly pathogenic avian influenza (HPAI) virus of the H5N1 subtype has spread through domestic poultry or wild birds in more than 60 countries. This article reports heterologous expression in Sf9 insect cells of H5-Hemagglutinin derived from avian influenza virus A/chicken/Iran/53-3/2008(H5N1). The HA nucleotide sequence of an Iranian strain of H5N1 virus from clade 2.2, (A/chicken/Iran/53- 3/2008), was obtained from Genbank. The nucleotide sequence data was codon optimized for insect cells. Using pIEx-3 vector a molecular construct, encoding a secretion signal peptide of adipokinetic hormone (AKH) and 6xHis-Tag coding sequence, upstream of the cloning site for expressing fusion recombinant HA protein with N-terminal tags, were designed and ordered for synthesis by GenScript, Recombinant H5-Hemagglutinin was expressed in Sf9 insect cells as a 95kDa secreted protein. This insect-based Baculovirus-Free heterologous expression system provided functionally recombinant H5-Hemagglutinin that should be useful in anti-influenza drug screening, detection, and diagnostic tests and also as a potential protein-based vaccine.

    The aim of this study was to evaluate the effect of source and balance of dietary anion-cation (DCAD) of dairy cows on protozoa and methanogens populations, ammonia nitrogen concentration, methane gas and digestibility of dry matter in vitro. Competitive polymerase chain reaction (cPCR) techniques were used to count the protozoan and methanogens population. This design for dairy cow in early lactation with an average body weight of 580 kg, age 25 months and daily production of 35 kg was carried out in the form of a 2 × 2 × 3 factorial experiment. Effective factors included DCAD (+150 meq / kgDM, +250 and +350 meq / kgDM), potassium sources (potassium carbonate (KC) and Potassium carbonate sesquihydrate (KCS)) and magnesium sources (magnesium oxide (MO) and magnesium carbonate (MC)). The effect of experimental treatments on ammonia nitrogen concentration was not significant. In all treatments, DCAD increased cause reduced ammonia nitrogen concentration in the rumen. The effect of experimental treatments on gas production, dry matter digestibility, protozoan and methanogens population was significant. The results of this study showed that the treatment containing DCAD +250 meq / kgDM provided by Potassium carbonate sesquihydrate and magnesium carbonate is reduced the protozoa and methanogens population, production of methane and increasing the digestibility of dry matter. Considering that there was no significant difference between DCAD +250 and +350, it seems that the sources of Potassium carbonate sesquihydrate and magnesium carbonate plus the anion-cation balance level of +250 meq / kgDM through the rumen buffering conditions improve, improves the parameters of rumen fermentation. Therefore, these two sources are probably appropriate supplements in the dietary of dairy cows.

    Brucellosis has always been among the most important economic and public health issues, somehow, each year it causes huge economic losses for the livestock industries. On the other hand, since brucellosis is a zoonotic disease, the importance of the issue in terms of public health is doubled. Therefore, it is necessary that by conducting epidemiological studies as a first step, identify the agent and then control and reduce the incidence of the disease. In this study, 170 samples were collected from aborted livestock in sheep, goats and cattle in the Golestan and Razavi Khorasan provinces, between 2015-2018. After the autopsy, different organs of the fetuses were cultured on the Columbia agar and MacConkey agar and then all of the samples were examined by using gram staining and standard biochemical tests. Overall, 60 Brucella spp. isolates were recovered and eventually, molecular typing and identification of Brucella spp. were conducted by using molecular techniques including Bruce-ladder Multiplex PCR and ERIC-PCR. Out of 60 Brucella spp. isolates, 59 strains were identified as Brucella melitensis and 1 strain as Brucella abortus, also all isolates despite their different geographic locations, had the same genetic patterns. The Bruce-ladder Multiplex PCR seemed to be a quick, safe and cost-effective method for molecular identification of Brucella isolates at the genus and species level. Although the ERIC-PCR technique in many studies is a powerful method for molecular differentiation of bacteria, it is better to use more precise and high-resolution techniques such as MLVA and MLST for confirmation the results of this study.

    Neospora caninum is an intracellular protozoan parasite with global dissemination. This protozoa has been the main cause of abortion in dairy cows. The present research was done to study the seasonal and geographical prevalence of Neospora caninum in ruminants milk by molecular method. Four-hundred and forty raw milk samples were collected from Isfahan, Chaharmahal Va Bakhtiari, Khuzestan and Fars provinces through 4 seasons of the year and transferred to the laboratory. Extracted DNA samples by kit were analyzed in order to detect the Neospora caninum NC5 gene using the nested-PCR assay. From a total of 440 studied milk samples, 54 samples (12.27%) were infected with Neospora caninum. Cow milk had the highest (26%) and sheep milk had the lowest (4%) prevalence of Neospora caninum. Milk samples collected in winter had the highest (22.85%) and samples collected in the summer had the lowest (8.57%) prevalence of Neospora caninum. Milk samples collected from Chaharmahal Va Bakhtiari province had the highest (14.94%) and samples collected from Isfahan had the lowest (6.38%) prevalence of Neospora caninum. Regarding the relative prevalence of this parasite in raw milk, adoption of monitoring plans are required more than before. Additionally, considering the possibility of consumption of traditional dairy products and transmission of infection to human, complete boiling of milk is recommended before consumption.

    Toxoplasma gondii is a worldwide parasite which infects animals and human. Infections with this zoonotic parasite are acquired mostly by consumption of undercooked or raw meat, which contains tissue cysts. The current study was conducted to field evaluation of the modified agglutination test (MAT) for determine the rate of Toxoplasma antibody positive in slaughtered livestock.Sera were obtained from 346 farm animals including 48 cows, 85 sheep and 33 goats evaluated by Modified Agglutination Test to detect anti-Toxoplasma antibodies. In this study, Anti-Toxoplasma antibodies were detected in sera of 30 out of 166 (18.1%) animals. The highest rate of infection (24.7%) was found in sheep followed by cow (16.7%) and goat (3%). Most of sheep's (23%) had antibody titer of 1:20. Male consisted 7.8 % and females 22.9% of seropositive animals and was statistically significant (P>0.05). The difference in terms of geographical area was statistically significant (P>0.05). High Seroprevalence of toxoplasmosis observed in this region indicates that farm animals may play a major role in transmitting the infection to human through consumption of undercooked meats.

    Woodcocks are considered as a group of eight living species of Wading birds in the Scolopax genus which often migrate individually, choosing mixed or deciduous forests for their Habitation and are also One of the birds with parasitic infections. In this research, a total of 18 Eurasian woodcocks (S. rusticola) (11 males and 7 females), recorded from unauthorized hunters by Mazandaran environmental agents, were examined in order to study their external and internal parasites which showed 100% of total parasitic infection in the results. Many species of external and internal parasites were found including two species of cestoda (Hymenolepis spp. and Davinia proglotina), one species of Termatoda (Echinoparyphium), one species of mite (Megninia cubitalis) and two species of lice (Philopterus and Lipeurus). In addition, M. cubitalis with the infection rate of 72.22%, was the most prevalent parasite. Comparison of The findings in this research with other studies show that Eurasian woodcock have an important role in the transmission of parasites due to the importance of their migration and further researches seem required on this bird and other migratory birds in iran. This study is also the first report of the parasites in Eurasian woodcock in Iran.
