    مقدمه

    اشریشیاکلی انتروتوکسیژنیک عامل اسهال های باکتریایی که منجر به مرگ و میر سالانه صدها هزار کودک می شود. اشیرشیا کلی O157:H7 ، شایعترین سوش اشریشیاکلی انتروهموراژیک است. تولید یک واکسن ترکیبی موثر برای اشریشیاکلی انتروهموراژیک و اشریشیاکلی انتروتوکسیژنیک بسیار حایز اهمیت است.

    مواد و روش ها

    در این مطالعه تکثیر و همسانه سازی ژن sicl ، بعنوان یک DNA واکسن صورت گرفت. ترادف ژنهای کد کننده آنتی ژن از بانک ژنی ارزیابی و اپی توپهای آنها به منظور طراحی پرایمر برای ژن کایمری سنتتیک (تهیه شده از گروه امانی) بررسی شد و تکثیر آن از طریق PCR صورت گرفت ،زیر همسانه سازی ژن کایمریک چند قسمتی در وکتور بیانی یوکاریوتیک به منظور ساخت DNA واکسن انجام شد و در آخر پروتئین با روش کروماتوگرافی نیکل تخلیص و با وسترن بلات ارزیابی شد.

    نتایج

    نتایج ایمونوبلات بیان پروتئین کایمریک SICL به شکل ذرات نامحلول  12 ساعت پس از القا نمایانگر وجود یک باند 76 کیلو دالتونی است. تخلیص پروتئین نوترکیب با استفاده از ترادف هیستیدینی داخل ژن وتایید پروتئین تخلیص شده با آنتی بادی اختصاصی پروتئین نوترکیب ، صحت بیان پروتئین را نشان داد .

    بحث و نتیجه گیری

    مطالعات بیان و تخلیص پروتئین و ارزیابی آن با وسترن بلات ، نشان دهنده بیان پروتئین در میزبان است.

    زمینه تحقیق: 

    صرع یکی از پیچیده ترین بیماری های نورولوژیکی می باشد .علی رغم وجود داروهای صناعی تعدادی از بیماران از اثرات جانبی داروهای موجود و تشنج مقاوم رنج می برند. استفاده از داروهای سنتی گیاهی برای درمان صرع به عنوان یک روش درمانی پیشنهاد شده است. با توجه به ترکیبات زوفا و استفاده آن در طب سنتی، هدف این تحقیق مطالعه اثرات ضد تشنجی زوفا بر صرع است.

    مواد و روش ها

    در این مطالعه تجربی 56 موش سوری نر به 7 گروه (8 تایی) تقسیم شدند که شامل 1- گروه کنترل که فقط پنتیلن تترازول دیافت می کردند، 2- گروه شم که فقط آب مقطر دریافت می کردند، 3- 5 گروه آزمایشی که 30 دقیقه قبل از تزریق PTZ، زوفا را با دوزهای 100، 200، 400 و 800 میلی گرم بر کیلوگرم وزن بدن بوسیله ی گاواژ و 2 میلی گرم بر کیلوگرم وزن بدن دیازپام را با روش تزریق دریافت کردند. کیندلینگ با 12 بار تزریق داخل صفاقی 45 میلی گرم بر کیلوگرم PTZ هر 48 ساعت یکبار شد. آنالیز داده ها نشان داد درمان با زوفا اثر قابل ملاحظه ای بر کیندلینگ شیمیایی داردکه با دیازپام قابل مقایسه است. آن پیشرفت مراحل تشنج رو بطور قابل ملاحظه ای کاهش داد (P˂0.05) و زمان شرع تشنج را یطور معنی داری افزایش داد (P˂0.05)  ولی اثر قابل ملاحظه ای روی مدت تشنج نداشت.  

    بحث: 

    نتایج نشان داد که عصاره ی زوفا اثر ضد تشنجی بر کیندلینگ PTZ دارد بنابراین ممکن است در درمان تشنج مفید باشد.

    توکسوپلاسما گوندی می تواند کبد انسان را تحت تاثیر قرار دهد و ممکن است باعث تغییرات پاتولوژیکی از جمله هپاتومگالی ، گرانولوما ، هپاتیت،  نکروز  و سیروز کبدی و در نتیجه زمینه ساز ابتلا به بیماری مزمن کبدی شود.

    هدف

    تعیین میزان آلودگی توکسوپلاسموز در بیماران مزمن کبدی ناشی از بیماری HCV مثبت و  بیماران کبد چرب درجه 2 و بالاتر بوده است.

    روش کار

    این بررسی از نوع مقطعی بوده و بر روی 150 نمونه خون تهیه شده از سه گروه افراد شامل بیماران هپاتیت C،  بیماران کبد چرب درجه 2 و بالاتر و افراد غیر بیمار فاقد عارضه کبدی به عنوان گروه شاهد طی سالهای 1397 و 1398 انجام شد. جهت انجام تست الایزا  از کیت توکسوپلاسماIgG  Toxo و از دو جفت پرایمر اختصاصی جهت تکثیر ژن   GRA6 توکسوپلاسما گوندی جهت  Nested -PCR استفاده شد.

    نتایج

    از 50 نفر گروه شاهد تنها 3 نفر(6%) و از 50 بیمار HCV+، 21 نفر (42%)  و از 50 بیمار مبتلا به کبد چرب گرید 2 و بالاتر 17 نفر (34%) آلودگی به توکسوپلاسموز مزمن را نشان دادند. این اختلاف نسبت آلودگی، از لحاظ آماری کاملا معنی دار بوده است (P< 0.01). ضمنا آزمایش Nested -PCR برای تشخیص انگل در هیچکدام از نمونه ها مثبت نشد.

    نتیجه گیری

    نتایج این مطالعه  نشان دهنده وجود ارتباط معنی دار بین بیماری توکسوپلاسموز مزمن در بیماران مبتلا به هپاتیت C و کبد چرب گرید 2 و بالاتر است. لذا ممکن است توکسوپلاسموزیس از عوامل زمینه ساز و بقای بیمارهای مزمن کبدی باشد.

    اهداف

    کاهش یا افزایش در بیان RNAهای غیرکدکننده در دستگاه عصبی از طریق تاثیر بر بیان مولکول های خاصی، موجب تغییر در مسیرهای پیام رسانی، مرگ سلول های عصبی و ایجاد بیماری می شود. هدف از این مطالعه، بررسی نقش و مکانیسم مولکولی اثر ncRNAها در بیماری های حرکتی ناشی از تحلیل عصبی است.

    روش ها: 

    مقالات موجود در داده پایگاه PubMed بین سالهای 2000 تا 2020 با کلمات کلیدی شامل ncRNA، miRNA، lncRNA و Neurodegenerative movement disorder جستجو شدند. سپس نقش و مکانیسم عمل ncRNAها در بیماری های پارکینسون، هانتیگتون و اسکلروز جانبی آمیوتروفیک یا ALS بررسی و تحلیل گردید.

    یافته ها:

     بر اساس یافته های موجود، تغییرات در بیان میکروRNAها و RNAهای طویل غیر کدکننده از طریق تاثیر بر بیان مولکول های ضروری برای بقای نورون ها شامل فاکتورهای رشد عصبی و عوامل آنتی اکسیدان، موجب مرگ نورونی و در نتیجه باعث القای بیماری های حرکتی ناشی از تحلیل رفتن اعصاب شامل بیماری های پارکینسون، هانتیگتون و ALS می شوند. همچنین مطالعات پیش بالینی و بالینی نشان داده اند که اندازه گیری تغییرات RNAهای غیرکدکننده در مایعات بدن می تواند به عنوان شناساگر زیستی در تشخیص زودهنگام و نیز در بررسی پیشرفت بیماری های عصبی-حرکتی مورد استفاده قرار گیرند. داروهای جدیدی نیز بر اساس هدف قراردادن RNAهای غیرکدکننده به ویژه میکروRNAها در مدل های حیوانی مورد آزمایش قرار گرفته اند که امید است تا در آینده ای نزدیک، کاندیداهای مناسبی برای استفاده به عنوان داروهای کنترل کننده بیماری های عصبی-حرکتی در بیماران باشند.

    نتیجه گیری:

     می توان نتیجه گیری نمود که شناسایی ncRNAهای موثر و مکانیسم های مولکولی دخالت آن ها در بیماری های عصبی-حرکتی می تواند به ارایه روش های جدید تشخیصی و راه کارهای درمانی برای این بیماری ها منجر شود.

    بارداری به عنوان دوره ی سرنوشت ساز در زندگی مادر و جنین، در مقابل آسیب هایی همچون سوء مصرف مواد مخدر به شدت آسیب پذیر است. سالیانه تعداد بسیار زیادی نوزاد معتاد به مواد مخدر در نتیجه اعتیاد مادر متولد می شود. سوء مصرف مواد مخدر در دوره بارداری، منجر به ظهور پیامدهای منفی متعددی برای نوزاد مانند افزایش احتمال تولد زودرس و نقص های مادرزادی می شود. از طرفی با آسیب به دستگاه عصبی مرکزی جنین، تکامل شناختی فرزندان مانند حافظه و یادگیری، توجه، زبان، مهارت حل مساله و فعالیت های اجرایی را تحت تاثیر قرار داده و باعث بروز آسیب رفتاری مانند افسردگی، بیش فعالی و غیره می شود. مواجهه با اوپیوییدها قبل از تولد، رشد و تکامل مغز و ساختارهای عصبی در دوره جنینی یا بعد از تولد را به تاخیر می اندازد.با توجه به تاثیر اعتیاد مادر بر تمام جنبه های سلامتی فرزندان و استعداد ابتلا به اعتیاد آنان در دوره بعدی زندگی، اهتمام بر پیشگیری، درمان و کنترل سوء مصرف مواد در مراقبت های بارداری، اصلی ضروری است. لذا در مقاله حاضر، اثرات مواجهه قبل از تولد با اوپیوییدها بر عملکردهای شناختی فرزندان مورد بررسی قرار می گیرد.

    هدف مطالعه

    هرپس ویروس انسانی تیپ 6 (HHV-6) ویروسی با شیوع بالا در جمعیت جهان است. طی سال های اخیر، فرم جدیدی از ژنوم ویروس کشف شده و تحت عنوان Chromosomally integrated HHV-6 (ciHHV-6) می باشد. ارتباط  ciHHV6-با انواع بیماری ها، نیاز به مطالعات بیشتر دارد. هدف از انجام این مطالعه، بررسی فرم اینتگره شده ژنوم ویروس HHV-6 از جنبه های مختلف بالینی و میزان فراوانی آن در جوامع مختلف می باشد.

    مواد و روش ها

    جهت انجام این مطالعه، کلمات کلیدی HHV-6، Integrated genome، Telomeric regions و Integrated HHV-6 در پایگاه های علمی معتبر مانند Scopus،  PubMedو  Google scholarبدون اعمال محدودیت زمانی مورد بررسی قرار گرفتند.

    نتایج

    تقریبا یک درصد از جمعیت جهان آلوده به ciHHV-6 می باشند که این مقدار در مناطق مختلف جغرافیایی متفاوت است. در صورت ابتلا فرد به ciHHV-6لود ویروس بالاتر از ابتلا به عفونت حاد خواهد بود و جهت تایید فرم اینتگره شده ژنوم  HHV-6لازم است تا در نمونه های فولیکول مو یا ناخن، به جستجوی حداقل یک کپی از ژنوم اینتگره شده پرداخته شود. ciHHV-6 با ایجاد اختلال در عملکرد های تلومریک می تواند سلامتی انسان را تحت تاثیر قرار دهد.

    نتیجه گیری

    تا به امروز درمانی جهت حذف ژنوم ویروس از سلول های میزبان کشف نشده است. طی بررسی به عمل آمده در این مطالعه، فرم اینتگره شده ژنوم HHV-6 می تواند با بیماری های مختلف از جمله بیماری های قلبی و خود ایمنی مرتبط بوده و مطالعات بیشتری جهت یافتن ارتباط آن با دیگر بیماری ها در نقاط مختلف جغرافیایی جهان نیاز است.

    هدف

    با وجود پیشرفت های فراوان در زمینه درمان و کنترل سرطان، همچنان کمبود های قابل توجهی در این زمینه وجود دارد و نرخ افزایش سرطان رو به رشد می باشد. با توجه به عوارض جانبی استفاده از مواد شیمیایی و پرتودرمانی، استفاده از گیاهان می تواند عوارض کمتری برای بیمار داشته باشد. هدف از این تحقیق بررسی آثار احتمالی ضد سرطانی عصاره الکلی اسطوخودوس بر مهار و رشد سلولهای کبدی رده سلولی HepG2 است.

    مواد و روش ها

    در این مطالعه تاثیر عصاره الکلی اسطوخودوس با غلظت های  10،100،1000و1میکروگرم/میلی لیتر بر رده ی سلولی سرطان کبد   HepG2مورد مطالعه قرار گرفت. برای بررسی میزان سمیت عصاره الکلی اسطوخودوس و میزان زنده مانی سلولها از روشMTT استفاده شد. تغییرات چرخه سلولی با استفاده از دستگاه فلوسایتومتر مورد بررسی قرار گرفت. تولید گونه های آزاد اکسیژنی و پراکسیداسیون لیپیدهای غشایی نیز سنجیده شد.

    نتایج

    نتایج نشان داد عصاره الکلی گیاه اسطوخودوس بر رده ی سلولی HepG2 اثر ضد سرطانی دارد. در پی تایید تاثیر عصاره اسطوخودوس بر سیکل سلولی، نتایج ناشی از بررسی میزان پراکسیداسیون لیپیدهای غشایی و تولید گونه های آزاد واکنشگر نیز معنادارگزارش شد.

    نتیجه گیری

    با توجه به نتایج حاصل از این مطالعه عصاره ی الکلی گیاه اسطوخودوس به عنوان عامل بالقوه ی ضد سرطانی گزارش شده و از جمله استراتژی های درمانی در سرطان کبد به شمار می رود.

    Introduction

    Enterotoxigenic Escherichia coli (ETEC) and Enterohemorrhagic Escherichia coli O157:H7 , the most common strains,  are causes diarrhea that can kill  hundreds of thousands of children annually. The development of an effective combination vaccine for enterohemorrhagic Escherichia coli and Enterotoxigenic Escherichia coli is very important.

    Materials and Methods

    In this study, the sicl gene was amplified and Subcloned as a DNA vaccine. The sequence of antigen encoding genes was evaluated from the genebank and their epitopes were evaluated to design of primer for the synthetic chimeric gene and was amplified by PCR. Subcloning of  a multipartal chimeric gene in eukaryotic expression vector was performed to make a DNA vaccine and finally the protein was purified by nickel chromatography and evaluated by Western blotting.

    Results

    The immunoblotting results of the expression of SICL chimeric protein indicated the presence of a 76 kDa band in the form of insoluble particles after 12 hours induction. Purification of the recombinant protein using His tag sequencee and confirmation of the purified protein with the recombinant protein specific antibody demonstrated the accuracy of the protein expression.

    Discussion & Conclusion

    Protein expression, purification and verify by western blotting showed that this recombinant chimeric protein (SICL) can be expressed in eukaryotic host.

    Background and Objective

    Epilepsy is one of the most complex neurological disease.   Despite existence of synthetics drugs for epilepsy, some patients suffer from side effects of available medicine and unresponsiveness of seizures.  Use of traditional herbal medicine for treatment of disease is proposed as a therapeutic modality.  In attention to components of Hyssopus officinalis L and use of it in folk medicine, the aim of this investigation was to study of the anti-convulsive effects of Hyssopus officinalis L extract on epilepsy

    Material and methods

    In this experimental study 56 adult male mice divided into 7 groups (n=8) including: 1-control group receiving only Pentylenetetrazole (PTZ), 2- sham group receiving distilled water and 5 experimental groups that 30 minutes before PTZ injection received   hyssopus by gavages in doses of 100, 200, 400 and 800 mg/kg of body weight and diazepam by injection in doses of 2 mg/kg.  Kindling was achieved by 12times intraperitoneal injection of 45 mg/kg of body weight of PTZ at every 48 hours.

    Results

    Data analysis indicated that treatment with hyssopus has a significant effect on chemical kindling that comparable with diazepam. It decreases progress of seizure phases considerably (P˂0.05) and increases latency time of seizure significantly (P˂0.05) but don’t has significant effect on seizure duration.

    Conclusion

    The results shows extract of Hyssopus officinalis L has anticonvulsant effect on PTZ kindling. Thus, it may to be beneficial for seizure treatment

    Toxoplasma gondii can affect the human liver and cause pathological changes such as hepatomegaly, granuloma, hepatitis, necrosis and liver cirrhosis may result to chronic liver disease.

    Objective

    The aim in this study was to determine the rate of toxoplasmosis infection in chronic liver disease patients with HCV positive disease and in fatty liver patients.

    Materials and Methods

    This cross-sectional study was performed on 150 samples prepared from three groups, including individuals with hepatitis C, persons with grade 2 and higher fatty liver disease, and patients without liver complications as a control group during 1397 and 1398. Serum and whole blood were taken from each subject for ELISA and PCR study. Toxoplasma IgG kit was used for ELISA test and two pairs of specific primers were used to amplify T.gondii GRA6 gene in Nested-PCR.

    Results

    Out of 50 controls, only 3 (6%) showed positive chronic toxoplasmosis. Of 50 patients with HCV +, 21 (42%) and of 50 patients with fatty liver disease showed 17 (34%) infection with chronic toxoplasmosis. This difference in infection ratio was statistically significant (P <0.01). No positive sample was observed in the Nested-PCR test for detecting T.gondii.

    Conclusion

    The results of this study indicate an association between chronic toxoplasmosis and patients with hepatitis C and fatty liver. Therefore, toxoplasmosis could be assumed as a predisposing factor for the survival of chronic liver disease.

    Aims

    A decrease or an increase in the expression of non-coding RNAs (ncRNAs) in the nervous system, via affecting the expression of certain molecules, induces dysfunctions in signaling pathways leading to neuronal death and neurodegenerative movement disorders. The aim of this study was to evaluate the role and underlying molecular mechanisms of ncRNAs in movement disorders.

    Materials & Methods

    Literature in PubMed between 2000-2020 with keywords of ncRNA, miRNA, lncRNA, and neurodegenerative movement disorder were searched. Then, the related data on the role and molecular mechanisms of the involvement of ncRNAs in Parkinson’s disease, Huntington’s disease, and Amyotrophic Lateral Sclerosis (ALS) were reviewed.

    Findings

    The findings indicate that the alterations in the expression of microRNAs and lncRNAs through affecting the expression of essential molecules for neuronal survival such as neurotrophic factors and anti-oxidants cause neuronal cell death and induce neurodegenerative movement disorders, including Parkinson’s disease, Huntington’s disease, and ALS. Besides, pre-clinical and clinical studies have shown that ncRNAs levels in the body fluids may serve as biomarkers in early diagnostic and monitoring the progression of movement disorders. New therapeutics based on targeting ncRNAs, especially miRNAs, have been developed and examined in animal models, which makes a hope to be appropriate candidates in controlling the progression of movement disorders in human patients in the near future.

    Conclusion

    It can be concluded that knowing effective ncRNAs and the related molecular mechanisms involved in movement disorders will result in the development of novel diagnostic and therapeutic approaches for the movement disorders.

    Pregnancy, as one of the crucial periods in the mother and fetus life, is extremely vulnerable to impairments such as drugs abuse. Every year, a large number of drug-addicted newborns are born due to motherchr(chr('39')39chr('39'))s addiction. Drug abuse during pregnancy leads to appearance of several negative consequences for baby, such as an increased risk of preterm birth and congenital deficits. On the other hand, it affects the cognitive development of children like learning and memory, attention, language, problem solving skills and execuative functions by causing impairments in the central nervous system learning, memory and attention of the fetus and it also leads to cognitive and behavioral disorders such as depression, hyperactivity and anxiety. Opioids are lipophilic drug, whereas the placental membrane is a lipoprotein, diffused from maternal to fetal compartments easily. Prenatal exposure to opioids delays the growth and development of the brain and neural structures in the embryonic or postnatal period.
    Considering the effect of maternal addiction on all aspects of health and susceptibility to addiction in later life of children, efforts to prevent, treatment and control drug abuse in pregnancy are essential. Therefore, in the present article, the effects of prenatal exposure to opioids on cognitive functions are reviewed.

    Aims

    Human herpes virus 6 (HHV-6) is a ubiquitous virus with a high rate of prevalence worldwide. In recent years, a new form of the virus genome has been identified called Chromosomally integrated HHV-6 (ciHHV-6). Further studies are required to elucidate the relation between ciHHV-6 and other clinical complications. The purpose of this study was to summarize the literature on different clinical aspects of the ciHHV-6 integration and its prevalevce in different communities.

    Materials and methods

    Search keywords include HHV-6, integrated genome, telomeric regions and integrated HHV-6. Scientific databases such as scopus, PubMed and google scholar with no specified start date were used for information collection.

    Findings

    About 1% of global populations are infected with ciHHV-6. Real-time PCR can be used to differentiate between ciHHV-6 and HHV-6 acute infection in which the viral load will be higher in ciHHV-6 compared to the acute infection. However, ciHHV-6 can be confirmed by evidence of one copy of viral DNA in hair or nail follicles. It has been shown that ciHHV-6 may disrupt the telomer stability.

    Conclutions:

     To date, no treatment has been discovered to remove the viral genome from the host cells. Taken together, the integrated form of the HHV-6 genome could be linked to the various diseases, including heart and autoimmune diseases. But further studies are needed to find its association with other diseases in different geographic area.

    Objective

    Despite many advances in cancer treatment and control, significant deficiencies remain and the rate of cancer growth is increasing. Due to the side effects of using chemicals and radiation, the use of herbs can have fewer side effects for the patient. The aim of this study was to investigate the potential anticancer effects of alcoholic extract of Lavender on inhibition and growth of HepG2 cell lines.

    Materials and Methods

    In this study, the effect of alcoholic extract of lavender (10,100,1000 and 1 μg / ml) on HepG2 liver cancer cell line was studied. MTT assay was used to evaluate the toxicity of lavender extract and cell viability. Cell cycle changes were assessed using a flow cytometer.Oxygen-free species production and membrane lipid peroxidation were also measured

    Results

    Results showed that alcoholic extract of Lavender has anti-cancer effect on HepG2 cell line. After confirming the effect of lavender extract on cell cycle, the results obtained from the evaluation of membrane lipid peroxidation and production of free reactive species were also significant.

    Conclusion

    According to the results of this study, the alcoholic extract of Lavender was reported as a potential anticancer agent and is one of the therapeutic strategies in liver cancer.
