    بیماری هوانگ لونگ بینگ (Huanglongbing = HLB) یا میوه سبز مرکبات ازنظر اقتصادی خسارت زاترین بیماری مرکبات در دنیا می باشد و بیش از یک دهه است که از جنوب ایران نیز گزارش شده است. تا کنون روش درمان قطعی برای این بیماری شناخته نشده است. باوجود این، یکی از موثرترین راه های مدیریت این بیماری، یافتن ارقامی است که از مقاومت نسبی یا تحمل بالایی در برابر این بیماری برخوردار هستند. در این پژوهش واکنش نهال های سه ساله ده رقم و پایه تجاری مرکبات نسبت به عامل فرم آسیایی بیماری HLB Candidatus Liberibacter asiaticus، ازطریق مایه زنی با پیوند بررسی شد. پس از مایه زنی، ردیابی Ca. L. asiaticus با استفاده از روش های پی سی آر معمولی و آشیان ه ای، با کاربرد آغازگرهای اختصاصی باکتری، در فواصل زمانی دو ماهه انجام و نیز اولین ظهور علایم بیماری، در نهال های مایه زنی شده یادداشت شد. سپس، بر اساس میانگین تعداد روزها تا اولین ردیابی Ca. L. asiaticus با پی سی آر و نیزدوره کمون بیماری، حساسیت و تحمل نسبی نهال ها به HLB مشخص شد. براساس نتایج به دست آمده، لیموترش مکزیکی و لیمولیسبون بیش ترین تحمل و پرتقال والنسیا و اورلاندوتانجلو بیش ترین حساسیت را در برابر این بیماری داشتند. نهال های نارنج و لیموشیرین حساسیت متوسطی به بیماری داشتند و گریپ فروت، پرتقال محلی، نارنگی محلی و نارنگی کینو، بعداز پرتقال والنسیا واورلاندوتاجلو، در درجه دوم حساسیت قرار گرفتند. نتایج این مطالعه برای انتخاب پایه ها و ارقام مناسب تر در مناطق آلوده به بیماری HLB و مدیریت بهتر بیماری می تواند مفید باشد.

    به منظور شناسایی گونه های قارچی عامل بیماری های تنه ی تاک در استان فارس، در ماه های اردیبهشت تا شهریور 1397 تا 1399 از درختچه های بالغ تاک که علایم کاهش رشد، زردی و سرخشکیدگی را در بخش های هوایی وتغییر رنگ بافت چوبی را در برش تنه نشان می دادند نمونه برداری شد. قطعات کوچکی از مرز مناطق تغییر رنگ یافته و بافت نسبتا سالم چوب جدا و پس از سترون سازی به تشتک های حاوی محیط کشت منتقل شدند. جدایه های قارچی پس از رشد به روش نوک ریسه و یا تک اسپور خالص سازی شدند. شناسایی جدایه ها با استفاده از ویژگی های ریخت شناختی و توالی سنجی نواحی مختلف ژنی انجام شد. در مجموع 166 جدایه ی قارچی شامل گونه های Botryosphaeria dothidea، Cytospora chrysosperma، Fomitiporia mediterranea، Kalmusia variispora، Macrophomina phaseolina، Phaeoacremonium minimum و Phaeoacremonium parasiticum شناسایی شد. گونه های C.  chrysosperma و P. minimum با فراوانی 42/5% و 31/28% به ترتیب کم ترین و بیش ترین فراوانی را در میان جدایه ها به خود اختصاص دادند. آزمون بیماری زایی روی نهال های یک ساله ی تاک در گلخانه نشان داد همه گونه ها قادر به ایجاد بیماری در گیاهان مایه زنی شده هستند. به منظور تکمیل اصول کخ، قارچ های عامل بیماری مجددا از گیاهان مایه زنی شده جداسازی و با استفاده از خصوصیات ریخت شناختی و مولکولی شناسایی شدند. تمامی گونه های فوق به استثنای P. minimum و P. parasiticum برای نخستین بار از تاک های آلوده به بیماری های تنه در استان فارس گزارش می شوند.

    زنبورVenturia canescens Gravenhorst (Hymenoptera: Ichneumonidae)  یکی از پارازیتوییدهای مهم کرم گلوگاه انار، Ectomyelois ceratoniae Zeller (Lepidoptera: Pyralidae) می باشد. گیاهان میزبان می توانند به عنوان یک عامل موثر بر خصوصیات زیستی و کارایی پارازیتوییدهای گیاه خوار عمل کنند. هدف تحقیق حاضر، بررسی تاثیر میزبان مختلف گیاهی شامل انار، به، پسته، پرتقال، خرما، سیب و زیتون و همچنین غذای مصنوعی بر فراسنجه های زیستی زنبور V. canescens در اتاقک رشد، شرایط دمایی 1±30 درجه ‏ی سلسیوس، رطوبت نسبی 5±60 درصد و دوره‏ ی نوری 14 ساعت روشنایی و 10 ساعت تاریکی بود. هم چنین، برخی از ترکیبات بیوشیمیایی میزبان ها اندازه گیری شد. بر اساس نتایج، طولانی ترین و کوتاه ترین طول دوره نابالغ پارازیتویید به ترتیب روی به و غذای مصنوعی مشاهده شد. کم ترین نرخ ذاتی افزایش جمعیت (rm) روی به و بیش ترین میزان آن روی انار بدست آمد. بین مقادیر برخی از ترکیبات بیوشیمیایی در میزبان های مورد مطالعه اختلاف معنی داری مشاهده شد. همچنین همبستگی مثبت یا منفی معنی داری بین ترکیبات اولیه و ثانویه گیاهی با فراسنجه های زیستی زنبور پارازیتویید مشاهده شد. فراسنجه های باروری، نرخ خالص تولیدمثل (R0) و نرخ ذاتی افزایش جمعیت (rm) همبستگی منفی معنی داری با مقادیر محتوای ترکیبات فنلی، تانن و فلاونویید گیاهی نشان دادند. نتایج نشان داد که نشوونما و تولیدمثل زنبور V. canescens در تغذیه از کرم گلوگاه روی انار و غذای مصنوعی نسبت به سایر میزبان ها بهتر بود و زنبور کارایی زیستی بالایی روی این دو میزبان نشان داد. یافته به دست آمده در این تحقیق می تواند در پیش بینی نتایج کنترل بیولوژیک کرم گلوگاه در روی میزبان های مختلف مفید واقع شود.

    بیماری پوسیدگی سفید ساقه با عامل Sclerotium rolfsii یکی از مهم ترین بیماری های خاک زاد بادام زمینی است که در سطح وسیعی از مزارع کشت بادام زمینی استان گیلان به ویژه در زمان برداشت شایع می شود. به کارگیری میکروارگانیسم های مفید، روش جایگزین برای استفاده از قارچ کش های شیمیایی است. در این پژوهش اثر سه جدایه از Trichoderma spp.، سه جدایه از Penicillium spp.، دو جدایه از Aspergillus spp. و یک جدایه از Cladosporium cladosporioides روی بیمارگر S.rolfsii با استفاده از روش های کشت متقابل، کشت روی اسلاید، متابولیت های فرار و متابولیت های غیرفرار در آزمایشگاه مورد بررسی قرار گرفت. در آزمایش های گلخانه ای، گیاهان بادام زمینی با این جدایه های قارچی همراه با S. rolfsii مایه زنی شدند و صفاتی مانند شدت بیماری، ارتفاع، وزن تر و وزن خشک گیاه مورد مطالعه قرار گرفت. نتایج نشان داد که در روش کشت متقابل، T. harzianum و T. virens به ترتیب با 58/93 درصد و 94/92 درصد بیشترین توانایی را در مهار رشد میسلیومی S. rolfsii داشتند. در روش کشت اسلاید، تمامی جدایه ها به جز T. virens و T. viride در مهار ریسه های S. rolfsii موفق بودند. در آزمون متابولیت فرار، P. glabrum و A. flavus به ترتیب با 25/81 درصد و 75 درصد و در آزمون متابولیت غیرفرار T. harzianum و T. viride به ترتیب با 5/97 و 75/93 درصد بیشترین توانایی را در مهار رشد میسلیومی S. rolfsii نشان دادند. همچنین اثرات این قارچ ها روی میزان مهار بیمارگر S. rolfsii در مطالعات گلخانه ای مورد بررسی قرارگرفت. نتایج نشان داد که P. glabrum با 4/39 درصد و T. viride با 8/38 درصد موثرترین قارچ ها در کاهش وقوع و شدت بیماری بودند. همچنین به کارگیری این قارچ های آنتاگونیست در شرایط گلخانه منجر به افزایش ارتفاع بوته، وزن تر و خشک اندام هوایی و ریشه در حضور قارچ عامل بیماری گردید. تجزیه واریانس و مقایسه میانگین صفات با استفاده از روش حداقل اختلاف معنی دار (LSD)، اختلاف معنی داری بین قارچ های مورد مطالعه در روش های آزمایشگاهی و گلخانه ای را نشان داد. نتایج این تحقیق نشان دهنده این مطلب بود که قارچ های T. viride، T. harzianum،P. glabrum، A. flavus و C. cladosporioides موجود در زیستگان طبیعی گیاه بادام زمینی خواص بالقوه ای برای مهار زیستی بیماری پوسیدگی سفید ساقه بادام زمینی از خود نشان می دهند.

    سویه شدید ویروس پیچیدگی برگ زرد گوجه فرنگی (Tomato yellow leaf curl viru, TYLCV-IL) یک بگوموویروس تک بخشی مخرب با گسترش جهانی است. دو روش غیرتراریخت سازی، واکسیناسیون آران ای دولا و کاربرد سازه سنجاق سری بیان کننده آران ای دولا، به منظور القای تداخل آران ای بر علیه ویروس پیچیدگی برگ زرد گوجه فرنگی مورد استفاده قرار گرفت. بررسی پروفایل آران ای های کوچک مداخله گر مشتق شده از ژنوم ویروس، دو نقطه داغ شامل قسمت هایی از توالی ژن های کدکننده پروتیین پوششی و پروتیین همراه با همانندسازی را در راه اندازی خاموشی ژن پس از ترانویسی مشخص نمود. آران ای های دولای مشتق شده از این توالی ها در شرایط درون شیشه ای تولید شدند و همزمان و یا سه روز قبل از مایه زنی همسانه عفونت زای ویروس، بر روی سطح برگ گیاهان توتون مورد استفاده قرار گرفتند. تمام گیاهان علایم مشخص آلودگی را در زمان 21 روز پس از مایه ز نی نشان دادند و قطعات ژنومی ویروس در گیاهان دارای علایم توسط واکنش زنجیره ای پلی مراز تکثیر شد. علی رغم حرکت سیستمیک آران ای های دولای مورد استفاده در گیاهان توتون، واکنش زنجیره ای پلی مراز در زمان حقیقی، هیچ تفاوت آماری معناداری در سطح تجمع ژنوم ویروس در بین تیمارهای مختلف نشان نداد. در رویکرد دوم، کاربرد سازه سنجاق سری بیان کننده آران ای دولا، بروز علایم آلودگی را تا زمان 21 روز پس از مایه زنی سرکوب کرد. همچنین ژنوم ویروس در گیاهان فاقد علایم ردیابی نشد که نشان دهنده قطع همانندسازی ژنوم ویروس است. این نتایج، کارآیی متمایز دو روش القای تداخل آران ای را علیه یک گونه مهم بگوموویروس نشان می دهد. نقش عوامل مختلف تاثیرگذار بر کارآیی تداخل آران ای علیه ویروس پیچیدگی برگ زرد گوجه فرنگی مورد بحث قرار گرفته است.

    بیماری باکتریایی نواری یکی از مهم ترین بیماری های گندم است که در صورت مهیا بودن شرایط محیطی خسارت قابل توجهی به میزبان خود در مناطق مختلف دنیا وارد می نماید. این بیماری در سال های اخیر در استان کرمانشاه شیوع پیداکرده و موجب خسارت قابل توجه روی گندم شده است. به منظور بررسی خصوصیات عامل بیماری، طی سال های 1398 و 1399 از بذور و برگ های دارای علایم بیماری در مناطق عمده گندم کاری استان نمونه برداری انجام شد. جداسازی باکتری ها از بافت های گیاهی آلوده به روش معمول تهیه سوسپانسیون از برگ ها و بذور خردشده در آب مقطر سترون و کشت روی محیط کشت آگار غذایی (NA) به صورت خطی انجام شد. پس از رشد، 140 پرگنه زردرنگ شفاف، براق و گرد خالص سازی شدند. برای تعیین خصوصیات فنوتیپی و بیوشیمیایی جدایه ها آزمون های باکتری شناسی مرسوم انجام شد. برای اثبات بیماری زا بودن جدایه ها و نیز تعیین پاتووار عامل بیماری، آزمون بیماری زایی در گلخانه روی چهار میزبان اصلی شامل گندم، جو، چاودار و تریتیکاله انجام شد. برای شناسایی دقیق تر عامل بیماری، ناحیه حفاظت شده 16S rRNA در جدایه های منتخب، با آزمون PCR و استفاده از آغازگرهای 27F و 1492R تکثیر و تعیین توالی گردید. تنوع ژنتیکی بین جدایه ها با استفاده از روش rep-PCR و با آغازگرهای BOX و ERIC بررسی شد. طبق نتایج آزمون های فنوتیپی و بیوشیمیایی، جدایه ها به گونه X. translucens تعلق داشتند. بر اساس نتایج آزمون بیماری زایی، جدایه ها در دو گروه قرار گرفتند. اعضای گروه اول فقط روی گندم بیماری زا بودند و علایم خفیفی در برگ های تریتیکاله ایجاد کردند اما اعضای گروه دوم در هر چهار میزبان ایجاد بیماری کردند، به این ترتیب اعضای گروه اول به پاتووار undulosa و اعضای گروه دوم به پاتووار cerealis تعلق داشتند. نتایج تعیین توالی 16S rRNA، شباهت نوکلیوتیدی 100% جدایه های منتخب را با گونه ی X. translucens (CP043500 - MK356430) موجود در پایگاه ژن بانک نشان داد. بررسی تنوع ژنتیکی نشان داد بین جدایه های مناطق مختلف استان تنوع زیادی وجود دارد، اما ارتباطی بین این تنوع و مناطق نمونه برداری دیده نشد. ضمن اینکه rep-PCR نتوانست پاتووارهای شناسایی شده را از یکدیگر تفکیک نماید.

    حلزون ها و راب ها از آفات مهم گلخانه و مزارع کاهو بوده که با تغذیه از برگ، ساقه، ریشه و بذور گیاه باعث از بین رفتن آن می گردند. کارایی متالدیید لوماکیدین 5 جی® با مقادیر 7/0 (توصیه شرکت سازنده) و 2 (توصیه شده مزرعه ای) گرم در مترمربع، طعمه فریکول® (5 گرم در مترمربع) و رنگ دورکننده سبزآرنگ® در مقایسه با شاهد روی گیاه زینتی اسپاتیفیلوم در سال های 1396 و 1397 در شرایط گلخانه ای مورد مطالعه قرار گرفت. همچنین عملکرد متالدیید لوماکیدین® در دزهای 7/0 و 2 گرم در مترمربع در مزرعه کاهو بررسی شد. تعداد نرم تن زنده، یک روز قبل و 2، 4، 8، 14 و 21 روز پس از تیمارها شمارش شد. طبق نتایج دو ساله گلخانه، در روز بیست و یکم، متالدیید لوماکیدین® 2 و 7/0 گرم در مترمربع، فریکول® و سبزآرنگ® به طور میانگین به ترتیب 95، 5/91، 7/78 و 2/76 درصد کنترل علیه راب Deroceras agreste نشان دادند. بر این اساس، متالدیید لوماکیدین® 2 گرم در مترمربع به طور معنی داری از سه تیمار دیگر موثرتر عمل کرد. مقایسه دو دز متالدیید لوماکیدین® در تجزیه آماری سالیانه، در روز بیست و یکم، درصد کارایی دز کمتر را بدون تفاوت آماری معنی دار با دز دیگر نشان داد. در مزرعه، در سال اول و دوم آزمایش به ترتیب تا 8 و 4 روز، عملکرد بیشتر دز بالاتر و در روزهای آتی عملکرد مشابه هر دو دز مشهود بود. این نشان می دهد که مصرف متالدیید لوماکیدین® 7/0 گرم در متر مربع می تواند ضمن کارایی مطلوب، صرفه اقتصادی نیز داشته باشد.

    شته ها یکی از مهم ترین آفات کشاورزی و جنگلی هستند که به صورت مستقیم با مکیدن شیره اندام های گیاهی و غیرمستقیم با انتقال عوامل بیماری زای میکروبی به میزبان های گیاهی سبب ایجاد خسارت می شوند. همچنین اشکال دیگر خسارت غیرمستقیم می تواند به صورت ایجاد گال، بدشکلی اندام ها، لکه های رنگ پریده بافت مردگی هر سه به دلیل ترشح بزاق سمی و ایجاد منظره کپک دوده ای به علت ترشح عسلک به عنوان منبع غذایی برای قارچ های پوسیده خوار دیده شود. تعداد گونه های شته در حال حاضر بیش از 5200 گونه و در حدود 69 گونه در شش جنس متعلق به خانواده Phylloxeridae است. به استثناء شته فیلوکسرای مو که آفت جهانی تاکستان های صنعتی است، زیستگاه بقیه گونه ها، عرصه های جنگلی و زیست بوم های غیر زراعی می باشد. هدف از مطالعه حاضر شناسایی یک گونه شته تغذیه کننده از بلوط گونه Q. brantii به عنوان مسبب احتمالی بروز لکه های زرد بافت مردگی روی برگ های درخت های آلوده بود. نمونه های شته روی درخت بلوط Q. brantii در اطراف شهرستان خرم آباد جمع آوری شدند. توالی یابی بخشی از قطعه ژنی COI انجام و تبارشناسی مولکولی این گونه با سایر گونه های جنس Phylloxera بررسی و مقایسه شد. به منظور شناسایی ریخت شناسی، نمونه های مونته شده (اسلایدهای میکروسکوپی) شته با روش های مرسوم زیر میکروسکوپ نوری و با کمک کلیدهای تاکسونومیک، تشخیص داده شدند. براساس تشخیص مولکولی و ریخت شناسی، نمونه ها به عنوان Phylloxera quercina (Ferrari, 1872) شناسایی شدند. مطالعه حاضر اولین گزارش وقوع P. quercina به صورت یک آفت در ناحیه رویشی زاگرس می باشد. قطعه ژنی COI بارکد قادر است تا به همراه ویژگی های ریخت شناسی به تشخیص دقیق گونه های جنس Phylloxera در ایران کمک شایانی نماید.

    ملخ صحرایی، Schistocerca gregaria Forsskål، به عنوان مخرب ‏ترین آفت مهاجر به مناطق وسیعی از زمین‏ های زراعی و مراتع در نقاط مختلف جهان خسارت وارد می‏ کند. به طور معمول، ملخ‏ های صحرایی با استفاده از حشره‏ کش‏ های شیمیایی کنترل می‏ شوند. با این حال، به دلیل اثرات جانبی حشره‏ کش‏ های رایج بر سلامت انسان، محیط زیست و ظهور حشرات مقاوم به حشره‏ کش‏ ها، توسعه برنامه‏ های جایگزین مدیریت آفت ضروری به نظر می‏ رسد. باتوجه به نقش کلیدی گیرنده اکدایزون (EcR) در رشد و نمو حشرات، این مطالعه با هدف استفاده از برنامه ‏های محاسباتی در جهت کشف ترکیباتی با ویژگی‏ های آنتاگونیستی برای گیرنده اکدایزون ملخ صحرایی صورت پذیرفت. درک ویژگی ‏های بیوشیمیایی و ساختاری گیرنده اکدایزون جهت طراحی آنتاگونیست‏ های اختصاصی مورد نیاز است و بنابراین در این مطالعه ابتدا خواص فیزیکوشیمیایی، ساختارهای ثانویه و توپولوژی گیرنده اکدایزون ملخ صحرایی با استفاده از برنامه‏ های بیوانفورماتیکی مورد بررسی قرار گرفت. مدل‏ های ساختاری سه بعدی گیرنده اکدایزون با استفاده از SWISS-MODEL پیش‏ بینی شده و کیفیت مدل‏ های حاصل با استفاده از برنامه ‏های مختلف مورد ارزیابی قرار گرفت. مطالعات داکینگ مولکولی بین هشت مهارکننده پروتیازی مشتق شده از ملخ ‏ها و مدل پیش ‏بینی شده گیرنده اکدایزون نشان ‏دهنده پتانسیل مطلوب آنتاگونیستی همه مهارکننده ‏های مورد مطالعه در برابر گیرنده اکدایزون بود. با این وجود، مهارکننده 1KJ0 در میان ‏کنش با گیرنده اکدایزون، مطلوب ‏ترین امتیاز داکینگ، انرژی پیوند، ثابت تفکیک، تعداد پیوندهای هیدروژنی و ارتباطات غیرپیوندی را از خود بروز داد که حاکی از پتانسیل آنتاگونیستی بالای 1KJ0 در مقابل گیرنده اکدایزون بود. نتایج حاصل از این پژوهش، اهمیت مطالعات محاسباتی را در شناسایی آنتاگونیست‏ های جدید علیه پروتیین هدف را نشان می‏ دهد. با این حال، تحقیقات in vitro و in vivo جهت اعتبار بخشیدن به ترکیب معرفی شده مورد نیاز است.

    پژوهش حاضر با هدف ارزیابی تاثیر دو ترکیب موثره موجود در اسانس سیر، دی ‏آلیل سولفید و دی ‏ آلیل دی ‏ سولفید، بر سامانه آنتی اکسیدانی و مصرف انرژی سلولی لاروهای شب ‏ پره مینوز گوجه‏ فرنگی انجام شد. پس از انجام زیست ‏سنجی اولیه به طریق غوطه ‏ وری برگ و محاسبه مقادیر LC30 و LC50، لاروها با این غلظت ‏ ها تغذیه شده و نمونه ‏ های آنزیمی از کل بدن آن ها تهیه شد. نتایج نشان داد که فعالیت آنزیم کاتالاز در لاروهای تیمار شده با دی ‏آلیل سولفید و دی ‏ آلیل دی ‏ سولفید نسبت به شاهد افزایش داشته و بیشترین مقدار در تیمار LC50 دی ‏آلیل دی ‏ سولفید مشاهده شد. تفاوت معنی‏‏ داری در فعالیت سوپراکسید دیسموتاز و پراکسیداز لاروهای شاهد و تیمار شده با غلظت LC30 دو ترکیب موثره وجود نداشت. فعالیت گلوکز 6- فسفات دهیدروژناز در همه تیمارها نسبت به شاهد افزایش یافت؛ اما تیمارهای LC50 هر دو ماده موثره بیشترین فعالیت این آنزیم را نشان دادند. لاروهای تیمار شده با غلظت های LC50 آلیل دی سولفید و دی آلیل دی سولفید بیشترین مقدار مالون دی آلدهید را نشان دادند درحالی که کمترین غلظت در لاروهای شاهد ثبت شد. میزان مصرف انرژی در لاروهای تیمار نسبت به شاهد کاهش معنی ‏داری را نشان داد به طوری که کمترین مقدار در لاروهای تیمار با غلظت LC50 دی آلیل دی ‏سولفید مشاهده شد. نتایج حاضر نشان می دهند که علاوه بر کشندگی، دی‏‏ آلیل سولفید و دی ‏ آلیل دی ‏سولفید سبب القای پاسخ های آنتی اکسیدانی و تحمیل انرژی مصرفی در لاروهای تیمار شده در مقایسه با شاهد می شوند. این تغییرات در نهایت کارایی تولیدمثلی و توانایی انتشار آفت را تحت تاثیر قرار داده و در دراز مدت می تواند از طغیان جمعتی آن بکاهد.

    Background and Objectives

    Citrus greening (HLB) is one of the most damaging citrus diseases worldwide. As well, the three most important causal agents of this disease in the world include "Candidatus (Ca.) Liberibacter asiaticus ("CLas"), Ca. Liberibacter africanus ("CLaf"), and Ca. Liberibacter americanus ("CLam"). Accordingly, these were reported from Asian-American, African, and American countries, respectively. Of note, "CLas" species is found as the most important causal agent of this disease in all countries in the world, including Iran. It seems that the most effective way to reduce HLB is finding those cultivars that are resistant or tolerant to HLB. Accordingly, in the present study, an attempt was made to evaluate the reaction of a number of common citrus cultivars to this bacterium.

    Materials and Methods

    In the present research, ten commercial rootstocks and cultivars of the common citrus in the south of Iran, including Mexican lime, sour orange, mandarin orange, Kinnow mandarin, Orlando tangelo mandarin, local orange, Valencia orange ,sweet lemon, grapefruit, and Lisbon lemons were prepared as seedlings. After checking their health, they were inoculated with the citrus greening agent and then kept in an insect-free greenhouse at night at 25°C and during the day at 30°C.By passing two months from the inoculation, the pathogen was detected by performing specialized tests in inoculated seedlings. Thereafter, three two-year-old seedlings from each one of the above-mentioned rootstocks and cultivars were grafted with two infected buds. Two months after the inoculation, normal and nested PCR tests were performed on the vein tissue extract of the infected seedlings, once every two months, for a 12-month duration. Finally, both sensitivity and relative resistance of cultivars were evaluated based on the average number of days until the detection of the first Ca. L. asiaticus by PCR and also the incubation period of the disease, in each seedling.

    Results

    The causal agent of this disease ("CLas") was detected at the earliest time(in Valencia oranges and Orlando tangelo mandarin after 180 and 200 days, respectively) after the inoculation with infected graft, compared to other cultivars and rootstocks. Among the studied species, the "CLas" was detected in both Mexican lime and Lisbon lemon with the longest delay (by passing 330 and 300 days from the inoculation, respectively). According to the results, "CLas" was detected in sour orange and sweet lemon 290 and 280 days after the inoculation with infected graft, respectively. Based on the average number of days until the detection of the first Ca. L. asiaticus by PCR and also the incubation period of the disease, in each seedling, Mexican lime and Lisbon lemon were found as the most tolerant species and Valencia oranges and Orlando tangelo mandarin were found as the most susceptible ones to HLB. Moreover, sour orange and sweet lemon seedlings were moderately susceptible ones; and grapefruit, local orange, local, and Kinnow mandarin were moderately tolerant to HLB.

    Discussion

    Due to the reason that this bacterium does not grow in conventional medium, in this study, it was not possible to multiply it in artificial medium and then contaminate the plant with it. Therefore, performing the inoculation through grafting the seedlings with HLB infected buds was considered as the most appropriate method for seedlings inoculation with the pathogen. The faster the pathogen can settle and multiply and the symptoms appear in the host, the greater the susceptibility of the host to the disease. Early pathogen’s detection and symptoms’ observation in some hosts such as Valencia oranges and Orlando tangelo mandarin, indicate better bacterial proliferation in both cultivars. Notably, no symptoms of the disease were observed in Mexican lime and Lisbon lemons after 15 months, and one year after the inoculation in sour orange and sweet lemon plants. There is evidence that all known citrus cultivars are affected to this pathogen with different degrees of susceptibility. Based on the results of this study and the results of previous researchers in the world, it seems that researchers should look for the sources of resistance factors to HLB in more species and cultivars. Correspondingly, these sources can be used in the following two ways: used as a rootstock or scion, and used in the efforts of breeders to produce a resistant cultivar.

    Background and Objectives

    Grapevine trunk diseases are destructive fungal diseases that have considerably increased in the recent decades worldwide. More than 130 fungal species were associated with grapevine trunk diseases worldwide; however, the pathogenicity tests have not been conducted for some fungal species. From 2018 to 2020, grapevines with stunted growth, yellowing, branch dieback, and discoloration of the wood tissues symptoms were observed during field observation in the Fars province of Iran. This study aims to isolate, identify and characterize the fungal agents that cause these diseases using a combination of the morphological and molecular approaches.

    Materials and Methods

    Grapevines with stunted growth, yellowing, branch dieback, and discoloration of the wood tissues symptoms were used for the isolation of fungal strains. Small pieces of infected wood tissues taken from the margin of symptomatic and approximately healthy parts were surface sterilized and plated onto potato dextrose agar and malt extract agar media amended with ampicillin. Fungal colonies were purified using single spore and hyphal tip methods. Isolates were identified using morphological characteristics as well as sequencing of different gene regions. The pathogenicity of the isolates was tested in greenhouse conditions using one-year-old seedlings of grapevine. To complete Koch's Postulates, fungal agents were re-isolated from inoculated plants and identified.

    Results

    166 fungal isolates were obtained and identified using the morphological characteristics as well as sequencing of different gene regions. The isolates were identified as Botryosphaeria dothidea, Cytospora chrysosperma, Fomitiporia mediterranea, Kalmusia variispora, Macrophomina phaseolina, Phaeoacremonium minimum, and Phaeoacremonium parasiticum. Among the species, P. minimum (28.31%) and C. chrysosperma (5.42%) had the most and least frequency, respectively. Pathogenicity tests using one-year-old seedlings in glasshouse showed all tested isolates can cause grapevine trunk diseases and produced white rot (in F. mediterranea) or wood necrosis (in other species) on inoculated tissue, developing from the point of inoculation. 

    Discussion

    This study's results showed that grapevines in Fars province are infected with fungi causing trunk diseases, which were previously reported from other provinces of Iran and other countries. According to our knowledge, it is the first report of all species, except P. minimum and P. parasiticum, causing grapevine trunk diseases in Fars province. More studies are required to better understand the causal agents of grapevine trunk diseases in other parts of Iran and introduce chemical or natural components that can reduce the economic losses caused by grapevine trunk diseases.

    Background and Objectives

    Venturia canescens Gravenhorst (Hymenoptera: Ichneumonidae) is an important parasitoid of lepidopterous pests, particularly the larval stage of Ectomyelois ceratoniae Zeller (Lepidoptera: Pyralidae), which is a polyphagous and key pest. The host plant of the herbivore can have a significant effect on their parasitoids' biological properties and efficiency. Host plants have a direct impact on the biological parameters of the herbivores. Additionally, they can influence the development and reproductive characteristics of herbivores' natural enemies. The purpose of this study was to determine the effect of seven host plants on the life history of V. canescens, including apple, date, olive, orange, pistachio, pomegranate, and quince, as well as an artificial diet.

    Materials and Method

    scanescens was grown on each host in a growth chamber set to 30±1°C, 60±5% RH, and photoperiods of 14:10 (L:D). Daily observations and records of stage duration, developmental period, and survival rate were made. The raw data were evaluated using TWO SEX-MSChart based on the age-stage, two-sex life table. Moreover, we used a spectrophotometer to determine the phytochemical metabolites of the tested hosts. Correlations between demographic variables and biochemical characteristics of various hosts were then estimated to identify factors affecting tritrophic interaction.

    Results

    The results indicated that various hosts had a significant effect on the demographic characteristics of the V. canescens. The immature development period was the longest and shortest on quince and artificial diets, respectively. Additionally, V. canescens' intrinsic rate of increase (rm) was lowest on quince and highest on an artificial diet. Furthermore, significant differences in biochemical metabolites were observed in this study between the various hosts. Moreover, significant positive or negative associations were discovered between life history variables and the biochemical characteristics of various hosts. The time required for parasitoid development was positively correlated with the phenol, tannin, and flavonoid content of E. ceratoniae hosts tested. There was a significant negative correlation between the total phenolic content, tannins, and flavonoids of various hosts and the fecundity, net reproductive rate (R0), and intrinsic rate of increase of V. canescens.

    Discussion

    Our findings may provide basic information for a comprehensive understanding of the tritrophic interactions between plants, herbivores, and parasitoids. Combining host plants and biological control agents may be an effective integrated pest management strategy for E. ceratoniae and other lepidopterous pests. Most importantly, our findings indicated that host plant quality variations could impact the performance and multitrophic communication behavior of V. canescens.

    Background and Objectives

    White stem rot caused by Sclerotium rolfsii is a soilborne disease which is extensively common across the groundnut farms of Guilan province, especially during the harvest period. Using the beneficial microbial agents is an alternative method for applying the chemical fungicides.

    Materials and Methods

    To find the appropriate fungal antagonistic isolates for the biological control of peanut white stem rot, the effect of three isolates of Trichoderma spp., three isolates of Penicillium spp., two isolates of Aspergillus spp. and one isolate of Cladosporium cladosporioides were studied on S. rolfsii in vitro using dual culture, slide culture, volatile metabolites and non-volatile metabolites. Peanut plants were inoculated with these fungal isolates as well as S. rolfsii in greenhouse experiments, and parameters including disease severity, height, fresh weight, and dry weight of the plant were measured.

    Results

    The results showed that in the dual culture method, T. harzianum and T. virens were most capable of suppressing the mycelial growth of S. rolfsii with a suppression capability of 93.58% and 92.94%, respectively. In the slide culture method, all isolates turned out to be effective in suppressing the mycelia growth of S. rolfsii, except for T. virens and T. viride. In the volatile metabolite assay, P. glabrum and A. flavus with 81.25% and 75%, respectively and in the non-volatile metabolite method, T. harzianum and T. viride with 93.75% and 97.5% respectively showed the greatest ability to inhibit S. rolfsii mycelial growth. Moreover, the effects of these fungi on the control of S. rolfsii pathogen were explored in greenhouse trials. The results revealed that P. glabrum was 39.4% effective and T. viride was 38.8% effective in reducing the extent and intensity of the disease. Under the presence of the pathogen, the treatment of these antagonistic fungi in greenhouse conditions enhanced plant height, shoot and root fresh and dry weight. In the in vitro and greenhouse approaches, an analysis of variance and a comparison of means of the attributes using the least significant difference (LSD) indicated significant differences (P ≤0.01 and P≤ 0.05) among the examined fungi.

    Discussion

    The results of the in vitro and greenhouse trials showed that T. viride, T. harzianum, P. glabrum, A. flavus, and C. cladosporioides, which are present in the natural flora of groundnuts, have the potential capability of the biological control of S. rolfsii as the pathogen of groundnut stem white rot.

    Tomato yellow leaf curl virus-Israel (TYLCV-IL) is a worldwide destructive monopartite begomovirus. Two nontransformative methods, namely the dsRNA (double-stranded RNA) vaccination and the hairpin construct expressing dsRNA were applied to induce RNA interference (RNAi) against TYLCV-IL infection. Analysis of TYLCV-IL-derived small interfering RNAs profile revealed two hot spots triggering post-transcriptional gene silencing, including a part of the coat protein (CP) and the replication-associated protein (Rep) genes. CP and Rep derived dsRNAs were produced in vitro and applied simultaneously or three days before agroinoculation of TYLCV-IL by rubbing on the leaf surface of Nicotiana benthamiana. All tested plants showed symptoms of typical infection at 21 days' post-inoculation (dpi), and viral genomic fragments were amplified in symptomatic plants by PCR as expected. Despite the systemic movement of applied dsRNAs in N. benthamiana plants, quantitative PCR showed no statistically significant difference between TYLCV-IL genome accumulation in virus infection alone or at the presence of dsRNA molecules. In contrast, the hairpin RNA construct (hpRep) triggered a high resistance against TYLCV-IL infection in tomato plants by targeting the same Rep sequence. HpRep construct suppressed expression of symptoms up to 21 dpi. Moreover, no virus genome was detected in symptomless plants indicating disruption of TYLCV-IL replication. These results indicate the distinct efficiency of two methods of RNAi induction against an important begomovirus species, TYLCV-IL. The role of different factors on the RNAi efficiency against TYLCV-IL has been discussed.

    Background and Objectives

    Bacterial stripe caused by Xanthomonas translucens is one of the most important diseases of wheat worldwide. The disease was first reported on barley in America in 1917. In Iran, it was reported for the first time on wheat and barley in Kerman province in 1987. Based on the standards for naming pathovars, the currently accepted pathovars as pathogens infecting cereals include pv. cerealis, pv. translucens, pv. undulosa, and pv. secalis. Following climate changes, especially heavy rainfalls in recent years, and probably the transmission of infected seeds, the disease has shown significant outbreaks and damages on wheat in Kermanshah province, Iran. The present study was performed to (I) characterize the pathogen in Kermanshah province, (II) determine pathogen distribution, and (III) assess the genetic diversity of the strains.

    Materials and Methods

    Wheat seeds and leaves showing symptoms of the disease were collected from major wheat-growing areas in Kermanshah province during 2019 and 2020. In the laboratory, samples were rinsed with tap water and were macerated into a few drops of sterile distilled water. Two loops full of the resulting suspension were streaked on the nutrient agar medium. Plates were incubated at 25°C and pale yellowish colonies were purified. Physiological and biochemical tests were performed to identify and characterize 140 isolates. To determine the pathovar(s) of the pathogen, the pathogenicity of the strains was tested on wheat, barley, rye, and triticale in the greenhouse. Nearly complete 16S rRNA sequences were determined for two strains representatives of two pathovars distinguished by pathogenicity test using 27F and 1492R primers. Genetic diversity among the isolates was evaluated in rep-PCR utilizing BOX and ERIC primers.ResultsBased on physiological and biochemical tests, Xanthomonas strains from wheat in Kermanshah province were identified as X. translucens. Pathogenicity tests revealed two groups among the strains. Members of the first group induced disease symptoms on wheat and triticale exclusively and were identified as pv. undulosa, while the strains belonging to the second group infected wheat, barley, rye, and triticale, and were identified as pv. cerealis. The rep-PCR using ERIC and BOX primers demonstrated genetic variation among the strains. However, there was no significant difference between the results obtained by ERIC- and BOX-PCR. Nearly complete 16S rRNA sequences were determined as the representative strains of two pathovars distinguished by pathogenicity test and based on the comparison of sequences with the GenBank database. The two strains matched X. translucens with 100% similarity. The nucleotide sequences of the pathogen strains were deposited in the GenBank database under the accession numbers MW173461 and MW193070.

    Discussion

    According to the findings of this study, X. translucens strains collected from wheat in Kermanshah province belonged to pathovars undulosa and cerealis. Considering the wide host range of these pathovars, the disease could be found on other cereals and grasses belonging to the Poaceae in the province and this subject must be taken into consideration in future studies. Although rep-PCR showed significant diversity among the strains, there was no correlation between the rep-PCR groups and the pathovars identified in the pathogenicity test or the geographic origin of the strains. Biochemical and physiological tests, as well as rep-PCR and 16S rRNA sequencing, could not distinguish the two pathovars. This confirms that pathogenicity tests and host range are still the main methods for determining pathogen pathovar(s).

    Background and Objectives

    Snails and slugs as terrestrial molluscs are the important pests of greenhouses and lettuce fields, which cause destruction by feeding on the leaves, stems, roots, and seeds of plants. Using molluscicides is the most important control method against their damages. Metaldehyde is considered as one the most important chemical compounds which is widely used to control slugs and snails. Lumakidin 5G® (Basir Shimi Company), as a product containing metaldehyde, has been at field recommended dose, 2g/m2, while its 0.7g/m2 has the potential to control slugs and snails according to the manufacturer. Ferricol® is bait including 1% iron phosphate, and Sabzarang® is snail repellent paint containing insoluble copper salts, special sticking, and binding agents. So far, no research has been conducted on the effectiveness of Lumakidin 5G®, Ferricol® and Sabzarang® in the ornamental flower greenhouse against snail/slug. Therefore, this study aims to evaluate the efficiency of Lumakidin 5G®, Ferricol®, and Sabzarang® in the ornamental greenhouse of Spathiphyllum wallisii in Guilan province, along with the effect of Lumakidin 5G® at 0.7 and 2 g/m2 in lettuce in Lactuca sativa field in Mazandaran province.

    Materials and Method

    Treatments included 1, 2: Lumakidin 5G® at two doses of 0.7 and 2 g/m2, 3: Ferricol® (5g/m2), 4: Sabzarang®, and 5: control (untreated) against slug/snail in the ornamental greenhouse of S. wallisii during 2018 and 2019. In greenhouse, Lumakidin 5G® and Ferricol® were applied between the pots, and Lumakidin 5G® in lettuce field was used between the rows. The pots were painted with Sabzarang®, as a 10 cm wide band, to prevent the snails from climbing the plants. In addition, the effectiveness of Lumakidin 5G® at 0.7 and 2g/m2 was evaluated in the lettuce, L. sativa field. The number of live slugs/snails was counted one day before and 2, 4, 8, 14, and 21 days after the treatments. During the 21st day after the treatment, the percentage of leaves damaged by snails/slugs in the greenhouse and the percentage of damaged plants in the lettuce field in Lumakidin 5G® treatment were calculated.

    Results

    Based on the two-year results of the greenhouse, Lumakidin 5G®, doses of 2 and 0.7 g/m2, Ferricol® and Sabzarang® caused 80.9, 69.9, 68.1, and 65.7% mean efficiency against slug, Deroceras agreste, respectively. Mean comparison indicated that Lumakidin 5G® (0.7 g/m2), Ferricol® and Sabzarang® failed to have a statistically significant difference, while Lumakidin 5G® (2g/m2) was significantly more effective than the other three treatments. Under these conditions, the mean comparison of two doses of metaldehyde in the annual statistical analysis on the twenty-first day showed no statistically significant difference. In the lettuce field, the application of doses of 0.7 and 2g/m2 Lumakidin 5G® against slug, D. agreste in two years of experiment had a similar trend. Thus, more efficiency of higher doses was observed in the first and second year of the experiment up to 8 and 4 days, respectively, while similar efficiency of both doses was evident in the following days, which low-dose of Lumakidin 5G® (0.7 g/m2) can be as effective as the recommended dose (2g/m2) against the slug. The percentage of leaves damaged by snails/slugs in the greenhouse and the percentage of damaged plants in lettuce field in Lumakidin 5G® treatment showed the effective role of both doses in reducing the damage. The use of this compound in both greenhouse and field conditions reduced plant damage about 3 to 4 times compared to the control.

    Discussion

    Due to the effectiveness of Lumakidin 5G® (at 0.7 g/m2), it is recommended to be used in the greenhouse and field to reduce snails/slugs damage. The application of this product decreases the cost of control by reducing the amount of consumption per m2/hectare. Further, it can have less environmental pollution than other doses. In organic agriculture, the use of Ferricol® is recommended due to the environmental safety of the compound. However, it is more expensive than metaldehyde. Thus, it can be considered as a good choice for a greenhouse due to its smaller scale than the field. Sabzarang®, as a repellent color, can be another alternative to chemicals, but the compound does not kill snails/slugs and only keep them away from that area. Finally, the paint is more expensive than the other two compounds.

    Background and Objectives

    Aphids are one of the most important agricultural and forest pests that may directly injure by sucking the sap of plant organs and indirectly by the transmission of microbial pathogen agents into the plant hosts. Moreover, other indirect forms of damage from feeding may be seen as gall formation, deformation of plant organs, chlorosis / necrosis spotting of the leaves all three due to the injection of poisonous saliva, and the presence of sooty mold view resulting in the honeydew excretion as a food source for the saprophytic fungi. The number of aphids is currently more than 5200 species, of which about 69 species within six genera belong to the family Phylloxeridae. Apart from the grape Phylloxera, a commercial pest of grapevines worldwide, the habitats of remaining species are forest trees and non-agroecosystems. The oak leaf phylloxeran aphids have a complex life cycle as exclusively egg-laying with parthenogenic forms. These aphids overwinter as egg inside seams and shell gaps of small branches or sometimes as first nymphal instar in branches. This study aimed to identify an aphid species feeding of Brant's oak, Quercus brantii, as a probable cause of yellow chlorosis spotting on the leaves of infected trees. Various methods have been proposed to assign unknown specimens to known species using their DNA barcodes.

    Materials and Methods

    Aphid specimens were collected on Q. brantii oak trees in eight selected forest sites around Khorramabad County, Lorestan Province. They were identified based on molecular and morphological evidences and was subjected to molecular characterization. DNA sequencing of partial COI gene fragment was performed, and the phylogeny of this species was compared with other species within the genus Phylloxera. In order to identify, the mounted aphid samples (microscopic slides) were morphologically identified by the conventional method under a light microscope with the help of taxonomical keys. 

    Results

    Samples were recognized as Phylloxera quercina (Ferrari, 1872) based on molecular and morphological identification. The phylogenetic tree was constructed as a fully resolved tree with Phylloxera genus as monophyly, dichotomous branching, and near-full bootstrap values.

    Discussion

    The present study is the first report showing the occurrence of P. quercina as a pest in the Zagros vegetation zone. Here we suggest the annual monitor of seedling oak and investigation of the geographical distribution of pest species. The utility of COI barcode gene fragment, along with the morphological characters, can help accurately identify Phylloxera species in Iran.

    The desert locust, Schistocerca gregaria Forsskål, is the most destructive migratory pest, which continually damages large areas of cropland and pastures in various parts of the world. Chemical insecticides are currently being used to control desert locusts. However, due to the harmful effects of conventional insecticides on human health and the environment, as well as the emergence of insecticides-resistant insects, alternative pest management programs must be developed. Given the critical role of the ecdysone receptor (EcR) in insect development, this study aimed to use computational tools to identify compounds with antagonistic properties against the desert locust EcR. Understanding the biochemical and structural properties of EcR is required for designing target-specific inhibitors, so we first used several bioinformatics tools to investigate the physicochemical properties, secondary structures, and topology of EcR from S. gregaria. SWISS-MODEL was used to predict the three-dimensional structural models of EcR, and the reliability of the predicted model was validated by various programs. Molecular docking studies between eight locust-derived protease inhibitors and the predicted model of EcR revealed the antagonistic capacity of all the studied inhibitors against EcR. However, the inhibitor 1KJ0 had the best docking score, the lowest binding energy and dissociation constant, and the greatest number of hydrogen bonds and non-bonded contacts with EcR, indicating its strong antagonistic potency against EcR. Our findings highlight the importance of computational studies in identifying novel antagonists to a target protein. However, in vitro and in vivo investigations are further required to validate the potency of the introduced compound.

    Background and Objectives

    Tuta absoluta Meyrick (Lep.: Gelechidae) has recently caused significant damages to tomato production in greenhouses and fields, necessitating sustainable and efficient management. Garlic has antimicrobial and antioxidant properties due to the presence of various compounds such as mono, di, and trisulfides. Meanwhile, diallyl sulfide and diallyl disulfide have demonstrated effective insecticidal activity against various insects. Due to their rapid decomposition and almost specific lethality, natural compounds may be a viable alternative to chemical compounds in the control of tomato leafminer. The purpose of this study was to determine the effect of diallyl sulfide and diallyl disulfide LC30 and LC50 concentrations on the antioxidant system and energy allocation of tomato leafminer.

    Materials and Methods

    The leaf disc method was used to conduct the larval bioassay. Initially, preliminary testing was performed to determine the final concentrations of 0.0625, 0.125, 0.5, 1, and 2%. Each leaf was submerged in the toxic solution for ten seconds before being transferred to 8 cm Petri dishes. Then, second instar larvae (seven to ten days old) were placed on the leaves and allowed to feed on both control and treated leaves. After 24 hours, larval mortality was counted, and POLO-Plus software was used to calculate lethal concentrations. The second instar larvae were initially immersed separately at LC30 and LC50 concentrations of each component to evaluate the antioxidant system and energy allocation. The control larvae were fed only solvent and each treatment used 60-second instar larva separately. After 24 hours, the treated and control larvae were divided into three groups of 10, and each group was homogenized in distilled water. Samples were centrifuged at 20,000 ×g at 4 ° C for 20 min, and supernatants were used to determine the activity of catalase, superoxide dismutase, peroxidase, glucose-6-phosphate dehydrogenase, and malondialdehyde concentration, as well as energy consumption.

    Results

    Catalase activity was significantly higher in treated larvae than in control larvae, with the highest activity observed following LC50 treatment with diallyl disulfide. Except for the concentration of diallyl sulfide LC30, the treated larvae had increased peroxidase activity compared to the control. The diallyl disulfide LC50 concentration demonstrated the highest activity. Although larvae treated with diallyl disulfide at the LC50 concentration had the highest superoxide dismutase activity, there was no significant difference in the LC30 concentrations of the two compounds compared to control larvae. The highest glucose-6-phosphate dehydrogenase activity was observed in larvae treated with both compounds at LC50 concentrations; however, the lowest was observed in larvae treated with both components at LC30 concentrations. Finally, the malondialdehyde concentration was significantly higher in the treated larvae than in the control larvae. The amount of energy allocated to tomato leafminer treated with diallyl sulfide and diallyl disulfide was significantly lower than the control. Finally, the lowest value was observed in larvae treated with diallyl disulfide at its LC50 concentration, but there was no significant difference between the other treatments.

    Discussion

    The two compounds, diallyl disulfide and diallyl sulfide, increased the antioxidant enzyme activities in tomato leafminer larvae through fat peroxidation and oxidative stress. In treated larvae, the amount of energy required for metabolic activity increased while the amount of energy available to the body decreased significantly. As a result, it is concluded that these two ingredients, garlic essential oil and tomato leafminer mortality, would effectively cause physiological disturbances, most notably in the antioxidant system and energy allocation. Based on the findings, field experiments are necessary to determine the environmental toxicity of these compounds and their importance in reducing crop damage compared to control. Additionally, the efficacy and combination of these compounds can be compared to other field control methods that aim to maintain a healthy crop with minimal pest damage.
