    بلایت زودرس یا سوختگی سیب زمینی که توسط گونه های مختلف Alternaria ایجاد می شود، یکی از بیماری های مخرب و شایع سیب زمینی در دنیا می باشد. در سال های اخیر بیماری در مناطق تولید سیب زمینی کشور خصوصا استان اصفهان گسترش پیدا کرده است و کشاورزان غالبا برای کنترل بیماری از قارچ کش ها استفاده می کنند. با توجه به مشکل بروز مقاومت در برخی قارچ ها به قارچ کش ها، این مطالعه با هدف ارزیابی حساسیت/مقاومت نه جدایه A. alternata به قارچ کش های رایج (مانکوزب، کلروتالونیل، ایمین اکتادین تریس و کانسنتو) با استفاده از روش بازدارندگی رشد رویشی در شرایط آزمایشگاه و گلخانه انجام شد. نتایج نشان می دهد که جدایه های جمع آوری شده از مزارع سیب زمینی که به طور مرتب با قارچ کش هایی مانند مانکوزب سم پاشی شده اند، دارای کمترین حساسیت به مانکوزب بوده اند. بر عکس، در برابر کلروتالونیل در این جدایه ها کمترین اختلاف در حساسیت مشاهده گردید. در آزمایش های گلخانه ای بیشترین کنترل بیماری توسط کلروتالونیل و مانکوزب صورت گرفت. این دو قارچ کش در مقدار توصیه شده باعث کاهش شدت بیماری به کمتر از 15% در یک دوره 20 روزه پس از مایه زنی شدند. بر اساس نتایج بدست آمده با ملاحظه سطح مقاومت مشاهده شده در شرایط آزمایشگاهی، کاربرد مانکوزب در تناوب با سایر قارچ کش ها مانند کلروتالونیل و ایمین اکتادین تریس می تواند قابل توصیه باشد.

    بیماری آنتراکنوز لوبیا ناشی از قارچ Colletotrichum lindemuthianum یکی از بیماری های مهم لوبیا در ایران است. در این پژوهش وجود یا عدم وجود ژن های مقاومت به بیماری با استفاده از آغازگرهای SCAR در 12 رقم لوبیای ایرانی مورد بررسی قرار گرفت. ارزیابی فنوتیپی بیماری در شرایط گلخانه بررسی شد. ژن Co-42 پیوسته به نشانگر SBB14، ژن Co-4 پیوسته به نشانگر SC08، و ژن مقاومت Co-6 با نشانگر SZ20 در ارقام اختر، درخشان، الماس، تلاش و کوشا ردیابی گردید. در ارقام گلی و ناز، ژن Co-42 ردیابی و در ارقام پاک، صدری و خمین دو ژن Co-4 و Co-42 ردیابی گردید. ژن Co-42 پیوسته با نشانگر SH18، در دو رقم درسا و شکوفا ردیابی گردید. حضور ژن مقاومت Co-2، تنها در رقم الماس مشاهده شد. ژن Co-5 پیوسته با نشانگر SAB3 تنها در رقم افتراقی مربوطه مشاهده شد. ارزیابی فنوتیپی بیماری در شرایط گلخانه براساس شدت شاخص بیماری نشان داد که از بین 12 رقم مورد بررسی چهار رقم (ناز، الماس،درسا و شکوفا) واکنش مقاومت نشان دادند در حالی که دو رقم اختر و گلی در گروه نیمه مقاوم قرار گرفتند. براساس شدت شاخص بیماری ارقام مذکور از یک مقاومت نسبی برخوردار بودند . بررسی همزمان نتایج ملکولی و فنوتیپی نشان داد که تنها دو نشانگر SQ4وSH18 در ارقام مقاوم (الماس، شکوفا و درسا) موفق به تکثیر باند DNA شدند. نتایج بدست آمده از این پژوهش کمک شایانی در تولید ارقام مقاوم لوبیا به بیماری از طریق هرمی سازی ژن های مقاومت خواهد بود.

    ویروس کوتولگی سبزرد نخود ایرانی (Chickpea chlorotic dwarf virus, CpCDV) از جنس Mastrevirus و خانواده Geminiviridae در طبیعت دارای دامنه میزبانی بسیار گسترده ای است. علیرغم دارا بودن قابلیت ایجاد خسارت توسط این ویروس، هنوز مطالعه ای روی آن در مناطق شرق و جنوب ایران صورت نگرفته است. در این تحقیق، میزبان های طبیعی این ویروس در گیاهان خانواده Fabaceae در استان های کرمان، خراسان رضوی و فارس مورد بررسی قرار گرفته است و ویژگی های ژنوم کامل یک جدایه از گیاه نخود ایرانی (جدایه شماره 15) از منطقه رابر استان کرمان تعیین گردیده است. بدین منظور، گیاهان زراعی از چند نقطه واقع در این سه استان جمع آوری گردید و آلودگی آن ها با استفاده از آزمون واکنش زنجیره ای پلی مراز بررسی شد. نتایج بدست آمده نشان داد که چندین گیاه لگومینوز شامل انواع حبوباب، سبزیجات، گیاهان علوفه ای و وحشی به CpCDV آلوده هستند. ترادف نوکلیوتیدی مربوط به جدایه نخود ایرانی از منطقه رابر استان کرمان تعیین شد و با ترادف های مشابه در بانک ژن مقایسه گردید. همچنین، سازه عفونت زای ویروس طراحی و ساخته شد و در آزمایش های بیماری زایی قادر به ایجاد بیماری در گیاهان مایه کوبی شده گردید. نتایج این بررسی نشان می دهد که CpCDV در ایران دارای دامنه میزبانی وسیعی در میان گیاهان خانواده Fabaceae است. گیاهان شنبلیله، اسپرس، یونجه و تلخه بیان برای اولین بار به عنوان میزبان های جدید این ویروس در ایران معرفی می گردند.

    پوسیدگی های فوزاریومی ریشه از بیماری های مهم لوبیا (Phaseolus vulgaris) هستنند. به منظور شناسایی فوزاریوم های عامل پوسیدگی ریشه لوبیا در استان مرکزی، در تابستان سال های 1397 و 1398 از مزارع لوبیا بازدید و از بوته های آلوده به پوسیدگی ریشه، نمونه برداری شد و در آزمایشگاه جداسازی و خالص سازی قارچ ها انجام گرفت. براساس خصوصیات ریخت شناختی و توالی یابی دو ناحیه ژنی فاصله ی ترانویسی شده ی داخلی دی ان ای ریبوزومی (ITS) و عامل امتداد ترجمه ی یک آلفا (EF-1α)، 37 جدایه فوزاریوم، شامل گونه های مرکبFusarium oxysporum ، F. solani و F. lateritium و همچنین گونه های F. equiseti ، F. acuminatum و F. redolens شناسایی شدند. همه ی جدایه ها پس از مایه زنی روی لوبیای رقم صدری، پوسیدگی ریشه ایجاد کردند. از آن جا که جدایه هایF. solani بیشترین فراوانی را داشتند، 12 جدایه از این گروه انتخاب و روابط فیلوژنتیکی آنها مطالعه شد. از سه تبار گونه ی مرکبF. solani ، جدایه ها در تبار شماره دو و سه قرار گرفتند. تنها یک جدایه در تبار دو قرار گرفت که خصوصیات آن با مشخصات گونهF. brasiliense مطابقت داشت. سایر جدایه ها در تبار سه و در دو زیرگروه قرار گرفتند. در آزمون بیماریزایی دامنه میزبانی جدایه های F. solani، هیچ کدام از جدایه ها روی عدس و لوبیا چشم بلبلی بیماری زا نبودند، اما تعدادی از جدایه ها موجب پوسیدگی ریشه نخود، سویا، ماش و پوسیدگی غده ی سیب زمینی شدند. گونه های F. lateritium، F. redolensوF. brasiliense برای اولین بار از ایران گزارش می شوند.

    جنس Botrytis P. Micheli ex Pers. یکی از بیمارگر های کارآمد و همه جازی است که سبب ایجاد بیماری های مخرب و خسارت های مهم اقتصادی در تعداد زیادی از گیاهان در سراسر دنیا می شود. به منظور شناسایی گونه های این جنس از میزبان های مختلف گیاهی، گیاهان دارای علایم آلودگی به این قارچ از باغات و گلخانه ها در شهرستان های ارومیه، میاندوآب، سردشت و پیرانشهر (استان آذربایجان غربی) و سنندج (استان کردستان) جمع آوری شدند. در مجموع تعداد 271 جدایه از گیاهان انگور (168 جدایه)، توت فرنگی (44 جدایه)، سیب (18 جدایه)، گیاهان زینتی شمعدانی، آلویه ورا و کاکتوس (39 جدایه) و کلم سفید (دو جدایه) به دست آمد. بر اساس مقایسه ویژگی های ریخت شناختی و تجزیه و تحلیل تبارشناختی نواحی ژنومی G3PDH، HSP60 و RPB2، سه گونه شامل B. cinerea، B. prunorum و B. sinoviticola شناسایی گردید. گونه ی B. prunorum با 125 جدایه، فراوانترین گونه و گونه های B. cinerea با 116 جدایه و B. sinoviticola با 30 جدایه در رتبه های بعدی از نظر فراوانی قرار گرفتند. گونه B. prunorum گونه جدید برای بیوتای قارچی ایران است و گیاهان انگور، توت فرنگی، سیب و شمعدانی میزبان های جدید برای این گونه معرفی می شوند. همچنین، گیاهان انگور و سیب میزبان های جدیدی برای گونه B. sinoviticola گزارش می شوند. نتایج آزمون بیماریزایی جدایه های منتخب از گونه های شناسایی شده روی برگ ها و میوه های انگور رقم بی دانه سفید نشان داد که جدایه های گونه های B. cinerea و B. prunorum به ترتیب بیشترین و کمترین شدت بیماریزایی را روی برگ ها و میوه ها نشان دادند و جدایه های گونه B. sinoviticola شدت بیماریزایی متوسطی داشتند.

    ویروس موزاییک خفیف فلفل (Pepper mild mottle virus, PMMoV)، یک بیمارگر مهم فلفل و بعضی گونه های تیره سولاناسه است که خسارات عمده ای را به این محصولات در سرتاسر نواحی معتدله تا گرمسیری وارد نموده است (Peng et al. 2015). عامل بیماری، ویروس موزاییک خفیف فلفل از جنس Tobamovirus و خانواده Virgaviridae است (Colson et al. 2010). آلودگی مخلوط این ویروس با توباموویروس های دیگر از جمله ویروس چروکیدگی قهوه ای گوجه فرنگی (Tomato brown rugose fruit virus, ToBRFV) ممکن است منجربه افزایش علایم بیماری و شدت بیماری روی گوجه فرنگی -شود(Lu et al. 2012). این ویروس اغلب علایم خفیفی را در برگ های فلفل آلوده ایجاد می کند، این علایم معمولا به صورت موزاییک و نکروز خفیف ایجاد می شود. اما در برخی از واریته ها علایم در برگ ممکن است وجود نداشته باشد. به منظور بررسی سبب شناسی بیماری موزاییک فلفل ، گلخانه هایی در مناطق مختلف استان اصفهان در تابستان و پاییز 1400 مورد بازدید قرار گرفتند. از بوته های فلفل دارای علایم موزاییک و بدشکلی میوه، موزاییک و نکروز برگ هاو همچنین از تدادی بوته های با علایم شدید بیماری شامل نکروز میوه، نمونه برداری انجام گرفت. آر ان ای کل از بافت برگ ها و میوه های جمع آوری شده با ترکیب ترایزول استخراج و در واکنش زنجیره ای پلی مراز رونویسی معکوس (RT-PCR) بکار رفت.

    Early blight of potato caused by Alternaria species is one of the most destructive and prevalent diseases of potato in the world. In recent years, the disease has become important in Iranian potato-producing regions, especially in Isfahan province, and farmers often use fungicides to control the disease. Considering the problem of resistance to some common fungi, this study aimed to evaluate the susceptibility/resistance of nine isolates of A. alternata to conventional fungicides (mancozeb, chlorothalonil, iminoctadine tris, Concento) by using vegetative growth inhibition method under in vitro and greenhouse conditions. The results revealed that most isolates collected from potato fields that were regularly treated with fungicides such as mancozeb exhibited less sensitivity to mancozeb. In contrast, against chlorothalonil, the least difference was observed in the susceptibility of different isolates. The greenhouse experiment showed that the highest control of the disease could be achieved by chlorothalonil and mancozeb treatment. These two fungicides at their recommended concentrations reduced the severity of the disease to less than 15 percent for 20 days after inoculation. Based on the results, concerning the resistance levels observed in the laboratory conditions, mancozeb application might be recommended in alternation with other fungicides such as chlorothalonil and iminoctadine tris.

    Bean anthracnose disease caused by Colletotrichum lindemuthianum is one of the most important bean diseases in Iran. In this study, the presence of disease resistance genes was investigated using SCAR primers in 12 Iranian bean cultivars. Also, disease symptoms were evaluated based on the disease severity index in the greenhouse condition. Amplification of SCAR markers SAS13, SY20, SZ04 and SB12 indicated the presence of Co-42, Co-4, Co-6 and Co-9 resistant genes respectively. Cultivars Akhtar, Derakhshan, Almas, Talash and Koosha contained Co-42, Co-4 and Co-6 resistance gene revealed by amplification of SBB14, SC08 and SZ20 markers accordingly. Co-42 gene was detected in Goli and Naz cultivars and Co-4 and Co-42 genes were present in Pak, Sadri and Khomein cultivars. Co-42 gene that is linked to SH18 marker was observed in Dorsa and Shokofa cultivars. The presence of Co-2 resistance gene was observed only in Almas cultivar. Co-5 gene linked to SAB3 marker was only observed in the related differential cultivars. Phenotypic evaluation for disease symptoms showed that out of 12 cultivars studied, four (Naz, Almas, Dorsa and Shokofa) showed resistance reaction and two (Akhtar and Goli) were moderately resistant. Based on the disease severity index, these cultivars were considered relatively resistant. Simultaneous analysis of molecular and phenotypic data showed that only the two markers of SQ4 and SH18 succeeded in band amplification in resistant cultivars Diamond, Shokofa and Dorsa. The results of this study will be of help in producing resistant bean cultivars to anthracnose disease by resistance gene pyramiding.

    Chickpea chlorotic dwarf virus (CpCDV) (Mastrevirus, Geminiviridae) has a wide natural host range. Despite of possessing the potential loss, until now the virus has not been studied in eastern and southern Iran. In this research, natural leguminous hosts of CpCDV in Kerman, Razavi Khorasan and Fars provinces were studied and the full-length genome of a chickpea isolate from Rabor (Kerman province) was characterized. Samples were collected from leguminous plants in different regions of the three aforementioned provinces and the CpCDV infection was tested in PCR. Results showed that leguminous plants including pulse, vegetables, foliage and wild species have been infected with the virus. The full-length genome of the Rabor isolate of CpCDV was sequenced and the resulted sequence was compared with the sequences of similar isolates available in the GenBank. Furthermore, the infectious clone of CpCDV was constructed and agroinoculated into some plants. Thus, the pathogenesis of the virus was demonstrated. Collectively, the results of this study indicated that CpCDV has a wide natural host range in the family Fabaceae. Fenugreek, common sainfoin, alfalfa and sophora are reported as the new hosts of the virus in Iran.

    Fusarium rot is one of the most important diseases of beans (Phaseolus vulgaris). In order to identify Fusaria that cause bean root rot in Markazi Province (Iran), in the summer of 2018 and 2019, several bean fields were visited and infected plants with root rot were sampled and the fungi were isolated in the laboratory. Based on morphological and sequencing characteristics of internal transcribed spacers of rDNA (ITS) and translation elongation factor 1-α (EF-1α) gene regions, 37 Fusarium isolates, including Fusarium oxysporum species complex, F. solani species complex, and F. lateritium species complex, as well as F. equiseti, F. acuminatum and F. redolens were identified. All isolates caused root rot on Sadry bean cultivar. Since F. solani isolates were the most common, twelve isolates of this group were selected and their phylogenetic relationships were studied. Of the three Clades of F. solani species complex, the isolates were placed in Clade 2 and 3. Only one isolate was found in Clade 2 whose properties matched those of F. brasiliense. The other isolates were classified in Clade 3 divided into two subgroups. In host range tests of F. solani isolates, none of the isolates was pathogenic on lentils, vetch and cowpea. A number of isolates caused root rot in chickpeas, soybeans, and potato tubers. F. lateritium, F. redolens and F. brasiliense are reported for the first time from Iran.

    The genus Botrytis P. Micheli ex Pers., is an efficient and cosmopolitan plant pathogenic fungus, causing destructive plant disesaes and significant crop losses in a wide variety of plant species all over the world. In order to identification of Botrytis species from different crop plants, symptomatic plants were collected from different orchards and greenhouses in Urmia, Miyandoab, Sardasht and Piranshahr (West Azarbaijan province) and Sanandaj (Kurdistan province). Totally, 271 Botrytis isolates were recovered from grapevine (168 isolates), strawberry (44 isolates), apple (18 isolates), ornamental plants (Geranium, Aloe vera and Cacti) (39 isolates) and white-headed cabbage (2 isolates). Based on comparision of morphological characteristics and phylogenetic analyses using sequences of three protein coding genes G3PDH, HSP60 and RPB2, three species including Botrytis cinerea, B. prunorum and B. sinoviticola were identified. Botrytis prunorum with 125 isolates were the most prevalent species followed by B. cinerea (116 isolates), and B. sinovticola (30 isolates). Botrytis prunorum is a new species to Iran mycobiota and apple, grapevine, geranium and strawberry are introduced as new hosts (matrix nova) for this species. Also, apple and grapevine are reported here as new hosts for B. sinovticola. Results of pathgenicity tests of the selected isolates from the identified species on leaves and berries of Thompson seedlees grape showed that isolates of B. cinerea and B. prunorum had the highest and lowest aggressiveness on both leaves and fruits, respectively and B. sinoviticola isolates had moderate aggressiveness.

    Pepper mild mottle virus (PMMoV) is a serious disease in pepper and some other plant species in the Solanaceae family, with significant damage to these crops throughout the temperate to tropical regions (Colson et al. 2010). The causative agent of the Pepper mild mottle disease single-stranded RNA virus belong to the Tobamovirus genous, family Virgaviridae, that induce (Peng et al. 2015). Mixed infection of plant viruses is a common feature in nature which may lead to more severe symptoms than their single infection. During a survey conducted in the fall and summer of 1400, capcicum plant samples showing symptoms of mild mosaic symptoms on leaves and fruit blistering and necrosis, were collected from a number of greenhouses in Isfahan. PMMoV with an another tobamovirus, Tomato brown rugose fruit virus (ToBRFV), may cause more severe symptoms on pepper or tomato plants (Lu et al. 2012). Total RNA was extracted from collected leaves and fruits tissues by using a TRIzol reagent (Sinaclone) and subjected to reverse transcription-polymerase chain reaction (RT-PCR) assay using a PMMoV specific primer pair PMMoVCP / a and PMMoVCP / s (Peng et al. 2015) a 500 bp DNA fragment was amplified from affected pepeer plants. Furthermore, To assess the presence of other viruses in mixed-infection with PPMoV, in plant samples showing severe symptoms, the extracted total RNA was used in an RT-PCR using four primer pairs including: a degenerate primer pair of potiviruses Nib1 / Nib3R (Riechmann et al. 2015);
