  • اثرات عصاره ی سیر بر بیان ژن های IFNγ، IL-17F و IL-17A در سلول های تک هسته ای خون محیطی در بیماری Multiple Sclerosis

    مقدمه :

    گیاه سیر، دارای ترکیبات استروییدی ساپونینی با قابلیت مهار برخی واکنش های ایمنی مانند التهاب و تکثیر مونوسیت هاست. با توجه به اهمیت لنفوسیت های Th1 و Th17 در پاتوژنز بیماری (Multiple sclerosis) MS، و اثرات ساپونین ها بر تعدیل سیستم ایمنی و بیان برخی سایتوکاین ها، مطالعه ی حاضر با هدف یافتن یک داروی گیاهی جدید برای درمان مکمل MS اجرا گردید.

    روش ها

    این مطالعه ی تجربی در سال 1399 در دانشگاه علوم پزشکی اصفهان انجام شد. ابتدا از پیازهای گیاه سیر عصاره گیری شد. سپس با توجه به این که شایع ترین الگوی مشاهده شده این بیماری، نوع عودکننده- بهبودیابنده (Relapsing Remitting MS) RRMS است از 5 بیمار جدید مبتلا به RRMS خون گیری شد و با استفاده از فایکول، سلول های تک هسته ای خون محیطی جداسازی گردید. سلول ها در محیط کشت RPMI و در مجاورت غلظت های 1، 2/5، 5 و 10 µg/ml از پارتیشن بوتانولی سیر به مدت 18 ساعت کشت داده شدند. سپس، به دنبال استخراج RNA، cDNA سنتز گردید و در نهایت با توجه به نقش سایتوکاین های interleukin-17A، interleukin-17F و interferon-ɣ در پاتوژنز این بیماری، میزان سطح بیان mRNA ژن های فوق ارزیابی گردید.

    یافته ها

    اگرچه تیمار سلول ها با غلظت های مختلف عصاره ی بوتانولی سیر تاثیر معنی داری در بیان ژن IL-17A نداشت، اما در غلظت های 1 و µg/ml 2/5 بیان ژن های IL-17F و IFNγ به صورت معنی داری کاهش یافت.

    نتیجه گیری

    نتایج مطالعه ی حاضر بیانگر اثربخشی بهتر پارتیشن بوتانولی سیر در غلظت های پایین بر روی بیان ژن های IL-17F و IFNγ می باشد و به نظر می رسد با افزایش غلظت افزایش نسبی بیان ژن اتفاق می افتد اگرچه این افزایش از لحاظ آماری، تفاوت معنی داری با نمونه ی کنترل نداشت، با این وجود لازم است این مطالعه بر روی تعداد نمونه و محدوده ی غلظتی بیشتر نیز انجام گردد.

  • اثر درمانی ترکیب اشعه ی ایکس و نانوذرات اکسید آهن متصل به داروی شیمی درمانی داکاربازین بر روی رده ی سلولی ملانوما

    مقدمه:

     پرتو درمانی، یکی از راه های موثر درمان سرطان ها از جمله سرطان پوست می باشد. به علت مقاوم بودن ذاتی سلول های ملانوما به پرتو، برای درمان سرطان پوست نیاز به دز های بالای تابش وجود دارد که این امر باعث افزایش اثرات جانبی بر روی بافت های اطراف می شود. استفاده از نانوذرات اکسید آهن می تواند سلول های ملانوما را به اشعه حساس تر کرده و میزان مرگ و میر سلولی را افزایش دهد. این مطالعه با هدف بررسی اثرات نانوذرات اکسید آهن متصل شده با داروی داکاربازین در حضور تابش اشعه ی ایکس مگاولتاژ صورت گرفت.

    روش ها

    بعد از کشت سلول های A375 به صورت درون آزمایشگاهی و گروه بندی سلول ها، غلظت های بهینه از نانوذرات اکسید آهن و داکاربازین به صورت جداگانه توسط آزمون MTT محاسبه شد. سپس غلظت های 0/4، 0/7 و 1/5 میلی گرم بر میلی لیتر از داکاربازین با غلظتی بدون سمیت از نانوذرات اکسید آهن ترکیب شدند. میزان آپوپتوز القا شده در سلول های تحت تابش با انرژی 6 مگاولت و دز تابشی 4 گری محاسبه و با آپوپتوز صورت گرفته در سلول های بدون پرتوگیری مقایسه شدند.

    یافته ها

    میزان آپوپتوز در سلول های ملانوما که با ترکیب نانوذرات اکسید آهن با داکاربازین تیمار شده اند، افزایش 50 درصدی را نشان داد. این میزان با پرتوگیری سلول ها به بیشترین مقدار افزایش پیدا کرده که 30 درصد بیشتر از میزان مرگ و میر در گروه شاهد است.

    نتیجه گیری

    استفاده از ترکیب همزمان نانوذرات اکسید آهن با داکاربازین می تواند پاسخ سلول های ملانوما به درمان را افزایش دهد. همچنین استفاده از این ترکیب در حضور تابش، کاهش معنی داری در بقای سلولی ایجاد می کند که نشان دهنده ی اثر حساس کنندگی نانوذرات اکسید آهن می باشد.

  • نقش میانجیگرانه ی شاخص توده ی بدنی در رابطه ی بیماری های متابولیک (دیابت و چربی خون) با افسردگی در سالمندان

    مقدمه:

     اختلالات متابولیک و افسردگی، از مشکلات شایع سالمندی است که می توانند در شروع و تشدید یکدیگر موثر باشند. هدف پژوهش حاضر، بررسی نقش میانجیگرانه ی شاخص توده ی بدنی در رابطه ی بیماری های متابولیک (دیابت و چربی خون) با افسردگی در سالمندان بود.

    روش ها

    روش پژوهش، توصیفی و از نوع مدل یابی معادلات ساختاری بود. جامعه ی پژوهش شامل تمامی سالمندان مراجعه کننده به مرکز بهداشت شهرستان فلاورجان در سال 1399-1398 بودند که 360 نفر از آن ها به روش نمونه گیری تصادفی ساده انتخاب شدند. ابزارهای پژوهش شامل پرسش نامه ی افسردگی سالمندان (با آلفای کرونباخ 0/9)، پرسش نامه ی توده ی بدنی و پرونده ی پزشکی آن ها در سامانه ی سیب (سامانه ی یکپارچه ی بهداشت) بود.

    یافته ها

    نتایج نشان داد، بین افسردگی سالمندان و شاخص دیابت، میزان (Body mass index) BMI و کلسترول خون به ترتیب همبستگی 0/81، 0/59 و 0/36 وجود دارد که همگی رابطه ی معنی داری را نشان دادند. همچنین متغیر توده ی بدنی می تواند نقش متغیر میانجی بین افسردگی و بیماری های متابولیک را ایفا کند. مدل تبیین شده نیز از برازش مطلوب برخوردار بود.

    نتیجه گیری

    بر اساس یافته های پژوهش، بیماری های متابولیک به طور مستقیم و همچنین غیرمستقیم از طریق توده ی بدنی با افسردگی سالمندان رابطه دارند بنابراین در برنامه های مربوط به اصلاح سبک زندگی و بهبود سلامت سالمندان باید به مداخله ی همزمان عوامل جسمانی و روان شناختی توجه شود.

  • The Effects of Garlic Extract on IL-17A IL-17F and IFNγ mRNA Expression in Peripheral Blood Mononuclear Cells of Multiple Sclerosis Patients

    Background

     Garlic contains steroidal saponins which modulate many aspects of the immune system including inflammation and monocyte proliferation. Considering the importance of Th1 and Th17 lymphocytes in the pathogenesis of Multiple sclerosis (MS), and the modulatory effects of saponins on the immune system, the present study was performed in search for a novel herbal medicine for treatment of MS.

    Methods

     This experimental study was performed at Isfahan University of Medical Sciences in 2020. Garlic onions were collected and subjected to extraction. Next, blood samples were collected from five new cases with Relapsing remitting MS (RRMS) as the most prevalent type of MS, and Peripheral Blood Mononuclear Cells were isolated using Ficoll™. Cells were cultured in RPMI medium and treated with 1, 2.5, 5 and 10 µg/ml of garlic butanol partition for 18 hrs. Then, the cells were subjected to RNA extraction, cDNA synthesis and Real-Time PCR for analysis of IL-17A, IL-17F and IFNγ mRNA expression levels.

    Findings

     Although treatment of the cells with the above concentrations of the garlic butanolic extract showed no significant effect on the IL-17A gene expression level, but the 1 and 2.5 µg/ml concentrations significantly reduced mRNA expression levels of IL-17F and IFNγ genes.

    Conclusion

     Since, reduction in the expression level effects was not significant at 5 and 10 µg/ml, it is concluded that the extract might be effective at low concentrations or the effect should not be concentration dependent. Further studies on more samples and with a range of lower concentrations should be performed.

  • The Therapeutic Effect of Combination of X-ray and Iron Oxide Nanoparticles Attached to Dacarbazine Chemotherapy Drug on Melanoma Cell Line

    Background

    Due to the inherent resistance of melanoma cells to radiation, high doses of radiation are required to treat skin cancer, which increases the side effects of radiation. The use of iron oxide nanoparticles can make melanoma cells more sensitive to radiation and increase cell mortality. The aim of this study was to investigate the effects of iron oxide nanoparticles conjugated with dacarbazine in the presence of megavoltage X-ray.

    Methods

    After culturing A375 cells in vitro and grouping the cells, the optimal concentrations of iron oxide nanoparticles and dacarbazine were calculated separately by MTT assay. Concentrations of 0.4, 0.7 and 1.5 mg / ml of dacarbazine were then combined with non-toxic concentrations of iron oxide nanoparticles. The amount of induced apoptosis in irradiated cells with 6 MV energy and 4 Gy radiation dose was calculated and compared with apoptosis in non-irradiated cells.

    Findings

    The rate of apoptosis in melanoma cells treated with a combination of iron oxide nanoparticles with dacarbazine increased by 50%. This rate increased to the maximum with cell irradiation, which is 30% higher than the mortality rate in the control group.

    Conclusion

    Concomitant use of iron oxide nanoparticles with dacarbazine can increase the response of melanoma cells to treatment. The use of this compound in the presence of radiation also causes a significant reduction in cell survival, which indicates the sensitizing effect of iron oxide nanoparticles.

  • Relationship between Metabolic Diseases (Diabetes and Hyperlipidemia) with Depression in the Elderly

    Background

    Metabolic disorders and depression are common problems in old age that can affect the onset and exacerbation of each other. The aim of this study was to investigate the mediating role of body mass index in the relationship between metabolic diseases (diabetes and hyperlipidemia) and depression in the elderly.

    Methods

    The research method was descriptive and structural equation modeling. The study population comprised of all the elderly who were referred to Falavarjan health center in 2019-2020. On total, 360 old aged individuals were selected by simple random sampling using their electronic records. The research tools included body mass questionnaire, elderly depression questionnaire (Cronbach’s alpha) and apple system (integrated health system).

    Findings

    The results showed that there was a correlation between depression in the elderly and diabetes index, Body mass index (BMI), and blood cholesterol 0.81, 0.59, and 0.36 respectively, depicting a significant relationship. Also, body mass variable can play the role of mediating variable between depression and metabolic diseases. The explained model also had a good fit.

    Conclusion

    According to the research findings, metabolic diseases are directly and indirectly related to depression in the elderly through body mass, so in programs related to lifestyle modification and improving the health of the elelderly, interventions for physical and psychological factors should be considered as well.
