    مقدمه

    آنزیم ال-آسپاراژیناز به طور موثر برای درمان بیماران مبتلا به سرطان ازجمله لوسمی لنفوبلاستیک حاد، ملانوسارکوما، لنفوما استفاده می شود. منابع باکتریایی جزء منابع مهم و درخور توجه برای تولید انبوه آنزیم ال-آسپاراژیناز است.

    مواد و روش‏‏ها: 

    در این مطالعه، باکتری تولیدکننده ال-آسپاراژیناز از خاک های آذربایجان جداسازی شد. به طور کلی براساس روش اسپکتروفتومتری با معرف نسلر، 4 جدایه با فعالیت آسپاراژینازی بالا مشاهده شد. جدایه دارای فعالیت آسپاراژینازی و فاقد فعالیت گلوتامینازی، برای ادامه مطالعه انتخاب شد. جدایه مدنظر براساس آزمایشات میکروسکوپی و بیوشیمیایی شناسایی شد. بهینه سازی شرایط محیط کشت ازجمله فاکتورهای غذایی (منبع کربن و نیتروژن) و پارامترهای فیزیکی (pH و دما) برای تولید بالای آنزیم آسپاراژیناز سنجش شد.

    نتایج

    مطالعات بهینه سازی نشان می دهند بالاترین میزان آنزیم ال-آسپاراژیناز در 7 pH، دمای 37 درجه سانتی گراد، در حضور 1 درصد آب پنیر به عنوان منبع کربن و 2/0 درصد آسپاراژین به عنوان منبع نیتروژن دیده می شود.

    بحث و نتیجه‏ گیری: 

    از این ارگانیسم می توان برای تولید صنعتی آنزیم ال-آسپاراژیناز و درنهایت، درمان سرطان استفاده کرد. در شرایط بهینه به دست آمده، تولید ال-آسپاراژیناز تا 2 برابر (8/7 واحد بر میلی لیتر) افزایش یافته است.

    مقدمه

    فرسودگی زیستی میراث فرهنگی پدیده ای جهانی است که ارتباط نزدیکی با حضور ماکروارگانیسم و میکروارگانیسم هایی مانند باکتری ها، آرکی ها، قارچ ها و گلسنگ ها و متابولیت های آنها دارد. چنین جوامعی به آسیب فیزیکی و شیمیایی و همچنین تغییر زیبایی در آثار فرهنگی منجر می شوند. برای کنترل فرسودگی زیستی ناشی از رشد میکروارگانیسم های میراث فرهنگی، از مواد ضدعفونی کننده مختلف استفاده شده است. این مطالعه بر استفاده موثرتر از ضدعفونی کننده های شیمیایی متداول در حوزه حفاظت از میراث فرهنگی متمرکز شده و روشی برای کاهش میزان مصرفی میزان ضدعفونی کننده ها در کنترل رشد و حذف میکروارگانیسم ها پیشنهاد کرده است که به کاهش آسیب های زیست محیطی منجر می شود.

    مواد و روش‏‏ها:

     در این مقاله اثرات ضدمیکروبی ضدعفونی کننده های متداول ازجمله کلرید بنزالکونیوم، ایزوتیازولینون، فرمالین، گلوتارآلدیید و پودر ضدعفونی کننده مبتنی بر اکسیژن علیه باکتری ها و قارچ های جداشده از سنگ آرامگاه کوروش بزرگ با روش میکروبراث بررسی شدند. سپس اثر ترکیبی ضدعفونی کننده ها با استفاده از روش چکربورد مطالعه شد.

    نتایج

    نتایج آزمون ها نشان دادند گلوتارآلدیید و پودر ضدعفونی کننده مبتنی بر اکسیژن، MIC بیشتری نسبت به سایر ضدعفونی کننده ها یعنی کلرید بنزالکونیوم، ایزوتیازولینون و فرمالین داشتند.

    بحث و نتیجه ‏گیری:

     نتایج نشان دادند تمام ضدعفونی کننده های انتخاب شده در این مطالعه، اثرات ضدمیکروبی قابل قبولی بر میکروارگانیسم های جداشده از سنگ آرامگاه کوروش داشتند و ترکیب ضدعفونی کننده ها موجب شد مقدار کمتری از این مواد شیمیایی استفاده شود که به لحاظ زیست محیطی بسیار حایز اهمیت است.

    مقدمه

    بیماری برق زدگی نخود با عامل قارچی Ascochyta rabiei در بیشتر مناطق دنیا نقش مهمی در کاهش عملکرد و کیفیت دانه نخود دارد. این تحقیق با هدف بررسی قابلیت برخی قارچ های میکوریزی دارسانه ای (arbuscular mycorrhizal fungi, AMF) در مهار زیستی این بیمارگر وبهبود صفات رشدی در دو رقم نخود انجام شد.

    مواد و روش‏‏ها: 

    پنج گونه AMF شامل Funneliformis mosseae، Gigaspora margarita، Glomus fasciculatum، Glomus versiform و Rhizophagus irregularis تهیه و اثرشان در مقایسه با قارچ کش کلروتالونیل بر مهار برق زدگی و نیز تاثیر آنها بر برخی صفات رشدی در دو رقم نخود سارال و بیونیج در حضور بیمارگر بررسی شد. این آزمایش در گلخانه به صورت فاکتوریل و بر پایه طرح کاملا تصادفی با 16 تیمار و 6 تکرار انجام شد.

    نتایج

    در هر دو رقم نخود، کلروتالونیل بیشترین میزان بازدارندگی از A. rabiei را نشان داد و همه گونه های AMF (به استثنای G. fasciculatum در رقم سارال)، سبب کاهش معنی دار شاخص بیماری نسبت به شاهد آلوده شدند. بیشترین اثر بازدارندگی از بیماری توسط میکوریزهای R. irregularis و G. versiform به ترتیب با 15/46 و 30/42 درصد در رقم سارال و توسط G. fasciculatum با 40 درصد در رقم بیونیج دیده شد. بیشتر گونه های AMF در حضور بیمارگر، سبب بهبود شاخص های رشدی نخود شدند و بهترین اثر در رقم های سارال و بیونیج به ترتیب به R. irregularis و G. fasciculatum تعلق داشت. بیشترین میزان کلنیزاسیون ریشه در هر دو رقم مربوط به G. fasciculatum بود.

    بحث و نتیجه‏ گیری:

     نتایج این پژوهش نشان دادند AMF می تواند سبب افزایش رشد گیاه و کاهش خسارت A. rabiei در دو رقم نخود شود. میزان کاهش بیماری و تحریک رشد به ژنوتیپ گیاه و گونه AMF وابسته بود. رابطه ای بین میزان کلنیزاسیون ریشه توسط AMF و کاهش بیماری برق زدگی مشاهده نشد.

    مقدمه

    متیل ترشیاری بوتیل اتر (MTBE) مکمل اکسیژنی محلول و کم تجزیه پذیری برای سوخت است که به سرعت خاک و آب را آلوده می کند. این مطالعه با هدف جداسازی و شناسایی باکتری های تجزیه کننده MTBE از خاک پتروشیمی بندر امام خمینی (ایران) و همچنین یافتن بهترین دما و pH برای تجزیه MTBE انجام شد.

    مواد و روش‏‏ها: 

    نمونه ها از خاک آلوده خط تولید MTBE در شرکت پتروشیمی امام خمینی ماهشهر گرفته شد. غنی سازی و جداسازی باکتری های تجزیه کننده MTBE در محیط پایه نمکی (BSM) حاوی 50 میلی گرم در لیتر MTBE و به دنبال آن، افزایش غلظت MTBE تا 400 میلی گرم در لیتر طی 5 مرحله غنی سازی انجام شد. فرایند تجزیه MTBE توسط باکتری جداشده با روش GC ارزیابی شد. جدایه تجزیه کننده MTBE با تکثیر و تعیین توالی ژن 16S rRNA شناسایی شد؛ درنهایت، پتانسیل باکتری برای تجزیه MTBE در دماهای مختلف (37-25 درجه سانتی گراد) و مقادیر مختلف pH (5/7-5/6) تعیین شد.

    نتایج

    باکتری تجزیه کننده MTBE به عنوان Azotobacter vinelandii شناسایی شد. نتایج تجزیه MTBE نشان دادند این باکتری یک سویه قدرتمند با قابلیت تجزیه 29/91 درصد MTBE در طول 14 روز است. بیشترین تجزیه  MTBE توسط باکتری در دمای 30 درجه سانتی گراد و 7pH= بدون تفاوت معنی دار به دست آمد.

    بحث و نتیجه‏ گیری: 

    سویه Azotobacter vinelandii جداشده در بررسی حاضر، توانایی زیادی برای تجزیه MTBE طی 14 روز انکوباسیون، بدون نیاز به افزودن سوبسترای هم تجزیه داشت و برای حذف این آلاینده از اکوسیستم های آلوده خاکی پیشنهاد می شود.

    مقدمه

    در سال های اخیر تقاضای جهانی برای محصولات کشاورزی افزایش یافته است. یکی از روش های افزایش میزان محصولات همراه با کاهش مصرف کودهای شیمیایی، استفاده از کودهای زیستی است. هدف از مطالعه حاضر تولید یک فرمول کم هزینه از کود زیستی تهیه شده از سودوموناس پوتیدا سویه PT است.

    مواد و روش‏‏ها: 

    ویژگی های مختلف محرک رشد گیاه سودوموناس پوتیدا سویه PT مانند محلول سازی فسفر، تثبیت نیتروژن و تولید ایندول-3-استیک اسید با استفاده از محیط Pikovskaya’s (PVK)، معرف سالکوفسکی و محیط فاقد نیتروژن سنجش شدند. اثر سوبستراهای کم هزینه برای تهیه کود زیستی ارزان قیمت با اندازه گیری وزن خشک باکتری تجزیه و تحلیل شد. کود زیستی به روش غوطه وری بذر تهیه شد و آزمایش های گلدانی با استفاده از کود زیستی تهیه شده از سودوموناس پوتیدا سویه  PTو مکمل های غذایی انجام شدند. اثر تیمارهای مختلف بر رشد و نمو گیاه Ocimum basilicum (ریحان) با اندازه گیری طول ریشه و اندام های هوایی، وزن تر اندام های هوایی و وزن خشک اندام های هوایی ارزیابی شد.

    نتایج

    سودوموناس پوتیدا سویه PT قابلیت محلول سازی فسفات (OD430nm = 1.1)، تولید (OD530nm = 0.86) IAA و تثبیت نیتروژن را نشان داد. بالاترین زیست توده باکتریایی در حضور آب پنیر و به دنبال آن ملاس با درجه بریکس 5/2 همراه با آمونیوم سولفات مشاهده شد. کود زیستی تهیه شده از این باکتری به ویژه در صورت استفاده با فسفات نامحلول، رشد گیاه را به طور چشمگیری افزایش داد. افزودن کود زیستی و فسفات نامحلول به ترتیب باعث افزایش 6/41، 7/83 و 25 درصدی طول اندام های هوایی، وزن تر اندام های هوایی و وزن خشک اندام های هوایی نسبت به گیاهان شاهد تیمارنشده شد.

    بحث و نتیجه ‏گیری: 

    نتایج نشان می دهند سودوموناس پوتیدا سویه PT می تواند برای تهیه کود زیستی مقرون به صرفه به منظور افزایش رشد گیاه استفاده شود.

    مقدمه

    پارک ملی خبر یکی از منابع طبیعی ارزشمند ایران است که جمعیت سیانوباکتری آن تاکنون مطالعه نشده است. سیانوباکتری ها مهم ترین باکتری های تثبیت کننده نیتروژن در محیط های آبی و خشکی اند که نقش حیاتی در اکوسیستم های خشک و نیمه خشک دارند. مطالعه حاضر با هدف تعیین نقش گونه های سیانوباکتری جداشده از مناطق دست خورده و دست نخورده در دو استپ سرد و گرم از پارک ملی خبر در برخی ویژگی های خاک، انجام و تاثیر گونه منتخب بر رشد گیاه مرتعی سکاله مونتانوم بررسی شد.

    مواد و روش‏‏ها:

     در مطالعه حاضر، سیانوباکتری ها از اکوسیستم سخت خشکی، جدا و روی محیط BG11 کشت شد. سپس بیان ژن nifH در تمامی جدایه ها با استفاده از Real Time-PCR سنجش شد. ندولاریا به عنوان بهترین سیانوباکتری تثبیت کننده نیتروژن، انتخاب و فعالیت آن در آزمایش گلدانی به تنهایی و در ترکیب با کود شیمیایی بررسی شد. در این مطالعه از سه خاک با ویژگی های فیزیکی و شیمیایی مختلف استفاده شد و نتایج تجزیه و تحلیل شد.

    نتایج

    نتایج نشان دادند تلقیح خاک با سیانوباکتری منتخب ندولاریا به عنوان یک کود زیستی، میزان نیتروژن را در خاک کشاورزی و خاک مخلوط افزایش داد و بدین ترتیب امکان کاهش مصرف کودهای شیمیایی را فراهم کرد. این باکتری توانایی ارتقای برخی ویژگی های خاک را ازطریق افزایش مقادیر TN، SOC، SOM و تقویت فعالیت آنزیمی دارد.

    بحث و نتیجه‏ گیری: 

    سیانوباکتری منتخب باعث افزایش پایداری خاک ازنظر شکست مکانیکی و مقاومت در برابر خشک شدن شد و از این نظر ساختار خاک را بهبود بخشید؛ این موضوع به ویژه در خاک مناطق خشک و بیابانی بسیار حایز اهمیت است.

    مقدمه

    در صنایع غذایی برای تولید انواع مختلف محصولات تخمیری از تک سویه ها یا مخلوط مزوفیل و ترموفیل باکتری های اسید لاکتیک به عنوان کشت های آغازگر استفاده می شود. نگهداری این کشت های آغازگر مرحله مهمی است که طی آن باید زنده مانی و پایداری ژنتیکی کشت آغازگر در بالاترین حد حفظ شود. روش های سنتی نگهداری کشت های آغازگر، بیشتر براساس روش های انجماد بود. امروزه خشک کردن روش معمول نگهداری کشت های آغازگر شده که عمدتا شامل خشک کردن پاششی و انجمادی است.

    مواد و روش‏‏ها: 

    در این بررسی مروری روایی، انواع روش های نگهداری کشت های آغازگر با کمک جستجو در پایگاه های داده ای مانند Google Scholar، Scopus،PubMed  و سایر پایگاه های اطلاعاتی معتبر مرور شده اند. پس از نگاه کلی به انواع روش های نگهداری، مزایا و معایب آنها مقایسه و راه حل هایی برای غلبه بر مشکل بقا معرفی شده اند.

    بحث و نتیجه ‏گیری: 

    در این مطالعه، مقایسه روش های خشک کردن از نقطه نظرهای مختلف می تواند به طراحی پروژه های تحقیقاتی موثر یا انجام فرایندهای تولید در این حوزه کمک کند.

    مقدمه

    عفونت کریپتوسپوریدیوم به عنوان عامل اصلی شیوع بیماری های منتقله از راه غذا و آب شناخته شده که نگرانی های بهداشت عمومی را در کشورهای توسعه یافته و درحال توسعه ایجاد کرده است. در مطالعه حاضر، شیوع و عوامل خطر ابتلا به کریپتوسپوریدیوم در کودکان مبتلا به اسهال در جوامع شهری و روستایی استان چهارمحال و بختیاری بررسی شدند.

    مواد و روش ها:

     در مجموع، 400 نمونه به صورت تصادفی از مدفوع کودکان مبتلا به اسهال مراجعه کننده به بیمارستان های شهرکرد و آزمایشگاه های تشخیص طبی گرفته شدند. بخشی از نمونه برای تست PCR، فریز و بخشی برای رنگ آمیزی زیل نلسون تغییریافته استفاده شد. نمونه هایی که از لحاظ میکروسکوپی آلودگی به کریپتوسپوریدیوم در آنها مثبت تلقی شدند، برای تعیین سویه انگل نیز با تست PCR ارزیابی شدند.

    نتایج

    میزان شیوع کریپتوسپوریدیوم در نمونه های گرفته شده 5/3 درصد بود. همچنین نتایج نشان دادند 85 درصد کودکان مبتلا به کریپتوسپوریدیوز در سن زیر 6 سال و تنها 15 درصد در سن 6 تا 12 سال بوده اند (P<0.05). براساس نتایج، میزان شیوع کریپتوسپوریدیوم در روستاییان، 5 درصد و در شهر حدود 2 درصد است (P<0.05). همچنین، 95 درصد روستاییان مبتلا و 85 درصد افراد شهری مبتلا، در یک ماه قبل از مراجعه، حداقل یک بار تماس با حیوان داشته اند (P<0.05). نتایج بیان می کنند میزان شیوع در فصل های بهار و تابستان بیشتر از فصول سرد بوده است (P<0.05)؛ اما بین دو جنس مذکر و مونث تفاوت آماری معنی داری مشاهده نشد (P>0.05). نتایج آزمون PCR نشان دادند همه نمونه های مثبت تک یاخته کریپتوسپوریدیوم پارووم بودند.

    بحث و نتیجه‏ گیری: 

    به طور کلی نتایج نشان دادند میزان شیوع و بروز کریپتوسپوریدیوز در کودکان با عواملی مانند سن، سبک زندگی، تماس با حیوان و فصل ارتباط دارد. همچنین گونه غالب در این منطقه کریپتوسپوریدیوم پارووم است.

    مقدمه

    اگزوپلی ساکاریدهای (EPSs) سنتزشده توسط باکتری های اسیدلاکتیک، پلی ساکاریدهایی با فعالیت های درمانی متنوع اند. EPSها به دلیل ایمن بودن می توانند در تهیه غذاهای فراسودمند به کار گرفته شوند. مصرف این محصولات می تواند راهکاری نویدبخش به منظور جلوگیری از بیماری های مهلک و مقاوم به درمان همانند سرطان و عفونت های مرتبط با بیوفیلم های میکربی باشد؛ با این حال، راندمان پایین سنتز، کاربرد صنعتی آنها را بسیار دشوار می کند. مطالعه حاضر با هدف بررسی فعالیت ضدسرطانی و ضدبیوفیلمی اگزو پلی ساکارید به دست آمده از Lacticaseibacillus paracasei AS20(1) و بهینه سازی تولید آن انجام شد.

    مواد و روش‏‏ها: 

    در مطالعه حاضر، سنتز EPS توسط Lacticaseibacillus paracasei AS20(1) باCentral Composite Design  بهینه سازی شد. همچنین تاثیر این EPS بر مهار تشکیل بیوفیلم Staphylococcus aureus (75/0-25/0 میلی گرم بر میلی لیتر)، بقای رده سلولی سرطان کولون انسانی (HT-29) (2-005/0 میلی گرم بر میلی لیتر) و القای آپوپتوز (2 میلی گرم بر میلی لیتر) در آنها بررسی شد؛ درنهایت، سمیت این اگزو پلی ساکارید میکربی با استفاده از آزمون همولیز گلبول های قرمز انسانی (50 و 100 میلی گرم بر میلی لیتر) بررسی شد.

    نتایج

    با توجه به مدل پیش بینی شده، بیشترین تولید EPS در 2/7pH ، غلظت عصاره مخمر 3/2 درصد (وزنی / حجمی) و غلظت سوکروز 3/33 گرم بر لیتر به دست آمد. EPS مطالعه شده فعالیت ضدبیوفیلمی (70-11 درصد) و فعالیت ضدسرطانی نویدبخشی را در برابر HT-29 نشان داد (IC50=1451.42 میکروگرم بر میلی لیتر). تیمار EPS درصد سلول ها را در حالت آپوپتوز اولیه (8/28 درصد) افزایش داد. این EPS میکربی، سمیت ناچیز (<5 درصد) علیه گلبول های قرمز داشت.

    بحث و نتیجه ‏گیری:

     درمجموع، شرایط بهینه سازی شده ایدیال می تواند برای تولید EPS در مقیاس بزرگ توسط Lacticaseibacillus paracasei AS20(1)  مفید باشد که دارای فعالیت های زیستی ارزشمندی است.

    مقدمه

    انتروویروس ها (EVs) و پاراکوویروس انسانی (HPeVs) نقش حایز اهمیتی در بروز بیماری در کودکان دارند. این ویروس ها قادر به ایجاد طیف گسترده ای از بیماری ها هستند که در مواردی باعث بروز عوارض شدید بالینی و مرگ ومیر می شوند. همچنین روش های تشخیصی مناسب برای جداسازی این ویروس ها به طور معمول در آزمایشگاه بالینی در دسترس نیستند؛ ازاین رو اپیدمیولوژی آنها به صورت دقیق گزارش نشده است. این مطالعه با هدف بررسی فراوانی انتروویروس های انسانی و پاراکوویروس در کودکان بستری در بیمارستان انجام شد.

    مواد و روش‏‏ها:

     سواب های نازوفارنکس از کودکان زیر پنج سال با علایم شدید تنفسی بستری در بیمارستان جمع آوری شدند. برای بررسی وجود EVs و HPeVs، از روش های RT-PCR و Real-time PCR دو مرحله ای استفاده شد.

    نتایج

    از مجموع هشتاد سواب نازوفارنکس جداشده از کودکان با علایم ARTIS، انتروویروس از چهار بیمار جدا شد (5 درصد). نتایج Real-time PCR برای pan-HPeVs هیچ تشخیصی از HPeVs را نشان نداد. هر چهار بیمار، نوزاد بودند. فراوانی انتروویروس و پاراکوویروس در میان جنس ها و گروه های مختلف سنی غیرمعنادار گزارش شد.

    بحث و نتیجه‏ گیری:

     انتروویروس ها و پاراکوویروس های انسانی می توانند باعث بیماری های شبه آنفلوانزا و اختلالات عصبی شوند؛ اما اطلاعات مربوط به ویژگی های اپیدمیولوژیک آنها بسیار محدود است. تحقیقات بیشتر درباره عفونت های دستگاه تنفسی ویروسی برای بهبود سلامت عمومی و جلوگیری از عوارض شدید لازم اند.

    مقدمه

    محصولات گوشتی ارگانیک به دلیل وجود باکتری های اسیدلاکتیک مانند لاکتوباسیلوس ها، لوکونوستوک ها، لاکتوکوس ها و پدیوکوکوس ها بسیار شناخته شده هستند. هدف از این تحقیق جداسازی و شناسایی سویه های جدید باکتری های اسیدلاکتیک از سوسیس های ارگانیک گوشت و ارزیابی فعالیت ضدمیکروبی آنها است.

    مواد و روش‏‏ها: 

    در این تحقیق جداسازی باکتری های اسیدلاکتیک از بسته بندی های تهیه شده در شرایط خلا سوسیس های شرکت سولیکو (تهران، ایران) انجام گرفت. شناسایی براساس ویژگی های مورفولوژی، بیوشیمیایی و تجزیه و تحلیل 16S rRNA انجام شد. ارزیابی اولیه فعالیت ضدمیکروبی سویه ها علیه میکروکوکوس لویتوس PTCC 1408 انجام گرفت. سینتیک رشد سویه MN-B و فعالیت ضدمیکروبی آن علیه لاکتوباسیلوس کازیی ATCC 32392 در محیط  MRSبررسی شد. فعالیت ضدمیکروبی علیه برخی از پاتوژن های منتقل شونده از راه غذا انجام شد. باکتریوسین تولیدشده توسط جدایه  MN-Bتوسط روش جذبی - دفعی و به کار بردن دیالیز به شکل جزیی خالص سازی شد.

    نتایج

    در این تحقیق دو سویه از جنس پدیوکوکوس و سه سویه از جنس لوکونوستوک از سوسیس های بسته بندی شده در شرایط خلا، جداسازی و با علایم اختصاری MN-A3، MN-A2-1، MN-A2، MN-B و MN-A نام گذاری شدند. نتایج شناسایی نشان دادند سویه های MN-B و MN-A متعلق به گونه پدیوکوکس اسیدی لاکتیسی و سه سویه MN-A2-1، MN-A2 و MN-A3 متعلق به گونه لکونوستوک مزانتروییدس هستند. از میان سویه ها، سویه MN-B بالاترین فعالیت ضدمیکروبی را علیه میکروکوکوس لویتوس PTCC 1408 نشان داد. وزن مولکولی باکتریوسین جداسازی شده از این سویه به شکل جزیی 5 کیلودالتون است.

    بحث و نتیجه‏ گیری: 

    سویه MN-B، پدیوکوکس اسیدی لاکتیسی PTCC 1954 فعالیت ضدمیکروبی مناسبی علیه سالمونلا انتریکا زیرگونه انتریکا سروتیپ تیفی موریوم PTCC 1709، استافیلوکوکوس اریوس PTCC 1113، لیستریا منوسیتوژنز PTCC 1302 و لیستریا اینووانی PTCC 1303 نشان داد. با توجه به اثر ضدمیکروبی مناسب، سویه جدید جداسازی شده می تواند کاندید مناسبی به عنوان نگه دارنده در مواد غذایی باشد.

    مقدمه

    میکروب هایی که درون بافت های گیاهی قرار دارند، به نام اندوفیت شناخته می شوند. این میکروب ها مجموعه ای از ترکیبات را تولید می کنند که قابلیت استفاده در پزشکی و کشاورزی مدرن را دارند. هدف از این مطالعه جداسازی، غربالگری و شناسایی باکتری های اندوفیت با فعالیت ضدمیکروبی علیه باکتری های پاتوژن گیاهی بود. گیاهان دارویی مانند Satureja khuzestanica به دلیل داشتن برخی متابولیت های ثانویه در طب سنتی استفاده می شوند؛ اما اطلاعات مربوط به اندوفیت های باکتریایی طبیعی آن محدود است.

    مواد و روش‏‏ها:

     در مطالعه حاضر، 27 سویه اندوفیت از S. khuzestanica جداسازی شد. براساس توالی یابی ژن 16S rRNA، سویه های باکتریایی جداشده با بیشترین فعالیت در برابر باکتری های بیماری زای گیاهی متعلق به جنس های Bacillus، Streptomyces و Pseudomonas بودند. متابولیت های ثانویه زیست فعال این باکتری های اندوفیت با استفاده از اتیل استات استخراج شدند و سپس آنالیز کروماتوگرافی گازی - طیف سنجی جرمی (GC-MS) در شرایط استاندارد انجام شد. حداقل غلظت مهاری (MIC) برای پنج گونه باکتری با روش رقت میکروبراث تعیین شد.

    نتایج

    تجزیه و تحلیل داده ها نشان دادند تفاوت معنی داری برای فعالیت ضدمیکروبی ثبت شد که حداقل غلظت بازداری آن از 312/0 میلی گرم بر میلی لیتر تا 5/2 میلی گرم بر میلی لیتر بود. حداقل غلظت باکتری کشی 625/0 میلی گرم بر میلی لیتر تا 10 میلی گرم بر میلی لیتر بود.

    بحث و نتیجه‏ گیری: 

    ترکیبات اصلی در تجزیه و تحلیل GC-MS باکتری های اندوفیت در این مطالعه 2،4-di-tert-butylphenol ، beta-d-glucopyranose ، hexadecane، tetradecane، eicosane و dibutyl phthalate بودند. این مطالعه برای نخستین بار اندوفیت های باکتریایی S. khuzestanica با فعالیت ضدمیکروبی علیه فیتوپاتوژن های باکتریایی را گزارش می دهد. یافته های ما بینش جدیدی را درباره فعالیت های ضدمیکروبی باکتری های اندوفیت طبیعی S. khuzestanica ارایه می دهد.

    مقدمه

    اهمیت قارچ ها در اکوسیستم های خشکی بسیار درخور توجه است؛ با وجود این، دانش ما درباره انتشار و تنوع قارچ ها در زیستگاه های طبیعی به ویژه اکوسیستم های نیمه خشک کم است. به منظور داشتن اطلاعات مناسب از ساختار جوامع قارچی و تاثیرعوامل محیطی بر ترکیب جوامع قارچی چنین محیط هایی، تنوع قارچی در کنار ویژگی های شیمیایی خاک بررسی شد.

    مواد و روش‏‏ها:

     نمونه برداری از خاک در بهار و تابستان از دو ناحیه سرد و گرم اکوسیستم نیمه خشک واقع در استان کرمان انجام شد. ویژگی های شیمیایی خاک با استفاده از روش های استاندارد خاک تعیین شدند. قارچ های ساکن در خاک با استفاده از توالی یابی نسل دوم ناحیه ITS (ITS1f-ITS2) تعیین شدند.

    نتایج

    با تجزیه و تحلیل یافته های حاصل از پلت فرم Illumina Miseq، 1542 واحد عملکردی تاکسونومی در نمونه های خاک مطالعه شده شناسایی شدند. تفاوت مشاهده شده در ترکیب جمعیت قارچی دو ناحیه سرد و گرم می تواند ناشی از اختلاف چشمگیر شرایط محیطی این دو ناحیه ازجمله تفاوت در میزان رطوبت و کربن آلی خاک باشد. آسکومیکوتا، شاخه غالب قارچی شناسایی شده بود و در مرتبه بعدی بازیدیومیکوتا فراوانی بیشتری داشت.

    بحث و نتیجه ‏گیری:

     قارچ های متنوعی در این اکوسیستم شناسایی شدند. برای آگاهی از نقش گونه های غالب قارچی در اکوسیستم های نیمه خشک به مطالعات بیشتری نیاز است.

    مقدمه

    فلاونوییدها گروهی از ترکیبات پلی فنولیک اند که در اندام های مختلف گیاهان عالی به طور طبیعی یافت شده اند. بسیاری از فلاونوییدها پایه ساخت داروهای گیاهی هستند و اثرات فارماکولوژیکی موثری دارند. روتین نیز یکی از فلاونوییدهایی است که به عنوان معرف در مطالعات فلاونوییدها کاربرد داشته است و نیز یک آنتی اکسیدان محسوب می شود. این مطالعه به منظور جداسازی و شناسایی مولکولی قارچ های اندوفیت تولیدکننده روتین از گیاه کبر انجام گرفت.

    مواد و روش‏‏ها:

     نمونه برداری از 10 منطقه ایران و از بافت های مختلف (شامل برگ، ساقه، ریشه و میوه) انجام شد. نمونه ها پس از ضدعفونی سطحی روی محیط کشت PDA قرار گرفتند و جدایه های قارچی به دست آمده خالص سازی شدند. شناسایی مولکولی جدایه ها با توالی یابی ناحیه ITS1-5.8S-ITS2 انجام شد. شناسایی مورفولوژیکی قارچ ها نیز با روش تهیه اسلاید میکروسکوپی و کلیدهای شناسایی قارچ ها انجام شد. عصاره متانلی قارچ ها به روش خیساندن، تهیه و وجود روتین با استفاده از تکنیک HPLC بررسی شد.

    نتایج

    در مجموع 24 قارچ شناسایی شدند. تجزیه و تحلیل HPLC نشان داد پیک فلاونوییدهای استاندارد شامل روتین، کویرستین و آپی جنین به ترتیب در 82/3، 60/7 و 40/12 دقیقه ظاهر شد. نتایج نشان دادند تنها دو جدایه قارچی قادر به تولید روتین بودند. مقایسه میانگین تولید روتین تفاوت معنی داری را نشان داد. قارچ Alternaria alternata M28 بیشترین میزان تولید روتین را داشت (94/22 پی پی ام). همچنین نتایج نشان دادند تولید روتین در بافت میسلیوم بیشتر از محیط خارج سلولی است؛ به طوری که در A. alternata M28 و Paecilomyces maximus M7 به ترتیب حدود 5/67 و 56/5 برابر روتین در میسلیوم تولید می شود.

    بحث و نتیجه ‏گیری: 

    روتین قارچی موجود در عصاره های متانولی با HPLC مشخص شد. این نخستین گزارشی است که نشان می دهد قار چ های اندوفیت پتانسیل پذیرفتنی برای تشکیل روتین از گیاه C. spinosa دارند و تحقیقات بیشتر روی این جدایه ها می تواند میزان تولید روتین را در آنها افزایش دهد و احتمالا به تجاری سازی منجر شود.

    Introduction

    L-asparaginase can be effectively used for the treatment of cancers, such as acute lymphoblastic leukemia, melanosarcoma, and lymphosarcoma. Bacterial sources contribute to the mass production of L-asparaginase.

    Materials and methods

    In this study, a novel L-asparaginase-producing bacterium was isolated from the soil of East Azerbaijan Province. Totally, 4 isolates were shown to have high L-asparaginase activities by using Nessler's reagent through the spectrophotometric method. The strain with glutaminase-free L-asparaginase activity was chosen for further studies. The isolated strain was identified via microscopic and biochemical tests. The growth conditions, including nutritional factors (carbon and nitrogen sources) and physical parameters (pH and temperature), were assayed and optimized for maximum L-asparaginase growth rate and production.

    Results

    B. subtilis as a potential microorganism was isolated, screened, and identified for the production of L-asparaginase. The optimization study revealed that maximum L-asparaginase could be produced at pH 7.0 and temperature of 37oC in the presence of 1% whey as the most suitable carbon source and 0.2% asparagine as a nitrogen source.

    Discussion and Conclusion

    The mentioned organism could be exploited for industrial production of L-asparaginase to be eventually utilized in cancer therapy. Under the obtained optimal conditions, L-asparaginase production was enhanced up to 2-fold (7.8 U/ml).

    Introduction

    The biodeterioration of cultural heritage is a worldwide phenomenon. It is closely linked to the presence of microorganisms such as bacteria, archaea, fungi, lichens, and their metabolites. Microbial colonization leads to physical and chemical damages as well as aesthetic changes of ancient objects. Biocides have been used for all kinds of cultural heritage material to control microbial growth as well as its deterioration. The present study focuses on applying current chemical biocides to improve conservation strategies of the cultural heritage and develops a way to reduce consumption of biocides to prevent the growth of microorganisms and reduce environmental biological damages.

    Materials and Methods

    In this study, the antimicrobial effects of commonly used biocides including benzalkonium chloride, isothiazolinone, formalin, glutaraldehyde, and active oxygen-based disinfectant powder were examined against bacteria and fungi isolated from Cyrus the Great tombstone by microdilution broth methods and the combined effect of biocides was studied by the microdilution chequerboard method.

    Results

    Microdilution tests showed that glutaraldehyde and active oxygen-based disinfectant powder had higher MIC values than other biocides, benzalkonium chloride, isothiazolinone, formalin. On the other hand, other biocides were more effective against the tested bacteria.

    Discussion and Conclusion

    The results of the study showed that all selected biocides had acceptable antimicrobial effects on microorganisms isolated from Cyrus the Great tombstone alone. When biocides were combined, they had a better antimicrobial effect resulting in less use of these materials.

    Introduction

    Ascochyta blight caused by the fungus Ascochyta rabiei plays an important role in reducing chickpea yield and seed quality in many parts of the world. The aim of this study was to investigate the potential of arbuscular mycorrhizal fungi (AMF) in improving plant growth parameters and the biocontrol of this pathogen on two chickpea varieties.

    Materials and Methods

    Five AMF species including Funneliformis mosseae, Gigaspora margarita, Glomus fasciculatum, Glomus versiform, and Rhizophagus irregularis were investigated for their potential in suppressing the Ascochyta blight on two chickpea varieties, Saral and Bivanij in comparison with chlorothalonil fungicide. Their effects were also examined on some plant growth parameters in the presence of a pathogen. This research was performed in greenhouse in a factorial experiment based on a completely randomized design including 16 treatments with six replications.

    Results

    Chlorothalonil had the highest suppression against A. rabiei in both chickpea varieties. All AMF (except G. fasciculatum in variety Saral) significantly decreased the disease index compared to the infected control. The highest disease suppression by AMF was obtained by R. irregularis (46.15%) and G. versiform (42.30%) on variety Saral and by G. fasciculatum (40%) on variety Bivanij. Most of AMF enhanced chickpea growth parameters in the presence of pathogen with R. irregularis and G. fasciculatum showing the best effects on Saral and Bivanij, respectively. G. fasciculatum had the highest root colonization on both chickpea varieties.

    Discussion and Conclusion

    The results of this study showed that AMF can promote plant growth parameters and attenuate the A. rabiei-induced losses in two chickpeas varieties. The rates of disease suppression and growth promotion depended on plant genotype and AMF species. No relationship was observed between the rates of root colonization by AMF and the reduction of Ascochyta blight symptoms

    Introduction

    Methyl Tertiary Butyl Ether (MTBE (is a highly soluble and low degradable oxygenated supplement to fuel that causes rapid contamination of soil and water. The present study aimed to isolate and identify the potent MTBE degrading bacteria from the petrochemical soil of Imam Khomeini Port (Iran), as well as the finding of the best temperature and pH for MTBE degradation.

    Materials and Methods

    Samples were taken from contaminated soils of Mahsahar MTBE production site in Imam Khomeini Petrochemical Company. Enrichment and isolation of MTBE degrading bacteria were done in Basal Salt Medium (BSM) containing 50 mg/l MTBE followed by increasing the MTBE concentration up to 400 mg/l during 5 enrichment steps. The MTBE degradation process by the isolated bacterium was assessed by the GC method. The MTBE degrading isolate was identified by the amplification and sequencing of 16S rRNA gene. Finally, the MTBE degradation potential of the bacterium was detected in different temperatures (25-37 ºC) and pH values (6.5-7.5).

    Results

    The MTBE degrading bacterium was identified as Azotobacter vinelandii. The results of MTBE degradation showed that the bacterium is a potent MTBE degrading strain with the degradation ability of 91.29% during 21 days. The highest MTBE degradation by the bacterium was obtained at 30 ⁰C and pH=7 with no significant differences.

    Discussion and Conclusion

    Azotobacter vinelandii strain isolated in the present study had the high ability to degrade high amounts of MTBE during 14 days of incubation without the need for the addition of co-catalytic substrate and is proposed for the removal of this pollutant from contaminated soil ecosystems.

    Introduction

    In recent years, the global demand for agricultural products has increased. One of the methods to increase crop yield, along with the decreased use of chemical fertilizers, is to apply biofertilizers. The purpose of the present study is to produce a low-cost formulation of biofertilizer synthesized from Pseudomonas putida PT.

    Materials and Methods

    Various plant growth-promoting characteristics of P. putida PT such as P-solubilization, N- fixation, and Indole-3-acetic acid (IAA) production were assayed using Pikovskaya’s (PVK) medium, Salkowski reagent, and N-free medium, respectively. The effects of low-cost substrates to prepare inexpensive biofertilizers were analyzed by measuring the bacterial dry weight. The biofertilizer was prepared by the seed immersion method, and pot experiments were performed by using prepared biofertilizer from P. putida PT and nutritional supplements. The effect of various treatments on the growth and development of Ocimum basilicum (basil) was evaluated by measuring the root and shoot lengths, fresh weight of aerial parts, and dry weight of aerial parts.

    Results

    P. putida PT exhibited capability of P-solubilization (OD430nm=1.1), IAA production (OD530nm=0.86), and N-fixation. The highest bacterial biomass was observed in the presence of cheese whey, followed by 2.5 °Bx molasses plus (NH4)2SO4. The prepared biofertilizer from this bacterium increased significantly plant growth, especially when it was used along with insoluble phosphate. The addition of biofertilizer and insoluble phosphate increased shoot length, fresh weight of aerial parts, and dry weight of aerial parts by 41.6%, 83.7%, and 25% compared to the untreated control plants, respectively.

    Discussion and Conclusion

    The results of the study signify that P. putida PT can be used to prepare a cost-effective biofertilizer for the enhancement of plant growth.

    Introduction

    Khabr National Park is one of the valuable natural resources in Iran, and its cyanobacterial population has not been studied yet. Cyanobacteria are the most important nitrogen-fixing bacteria in all aquatic and terrestrial environments, playing a vital role in arid and semi-arid ecosystems. The present study aimed to determine the role of different cyanobacterial species isolated from the grazed and not grazed areas in two cold and warm steppes of Khabr National Park on some soil properties and the effect of the selected cyanobacteria on Secale montanum growth.

    Materials and Methods

    In the present study, cyanobacteria were isolated from a harsh terrestrial ecosystem and cultured on a BG11 medium. Next, the expression of the nifH gene was assayed in all nitrogen fixing isolates using the Real Time-PCR. Then, the best nitrogen-fixing cyanobacteria (Nodularia. sp.) was selected and its activity was investigated in pot experiments alone and in combination with chemical fertilizer. Three different soils in terms of physical and chemical properties were analyzed.

    Results

    The results indicated that the treated soils with Nodularia sp. as a biofertilizer can significantly increase nitrogen content of both agricultural soil and mixed soil. Consequently, Nodularia sp. has the ability to promote some characteristics of the studied soil through improving TN, SOC, SOM, and enzyme activities.

    Discussion and Conclusion

    It can be concluded that the selected cyanobacterium increase soil stability in terms of mechanical fracture and resistance to desiccation and improve soil structure, which is very important, especially in arid and desert soil.

    Introduction

    Single or multiple strains of mesophilic and thermophilic lactic acid bacteria are used as starter cultures in food industries to produce different types of fermented products. Preservation of these starter cultures is a crucial step in which the highest viability and genetic stability of the starter culture should be retained. Traditional methods for the preservation of starter cultures were mainly based on the freezing method. Nowadays, the usual method for the preservation of starter cultures is drying which mostly included spray drying and freeze-drying.

    Materials and Methods

    In this narrative review of studies, a comprehensive review is conducted on all methods for the preservation of lactic acid bacteria through searches in databases such as Google Scholar, Scopus, Pubmed, and other valid databases. After a glance at the types of preservation methods, advantages and disadvantages are compared and solutions to overcome the problem of survival have been introduced.

    Discussion and Conclusion

    Comparisons of the preservation methods from different points of view in the present overview may help to design more effective research projects or production processes in this area.

    Introduction

    Cryptosporidium infection, which has been identified as a major cause of outbreaks of food and waterborne diseases, has raised public health concerns in developed and developing countries. In the present study, the prevalence and risk factors of cryptosporidium in diarrheal children in urban and rural communities of Chaharmahal and Bakhtiari province were investigated.

    Materials and Methods

    A total of 400 samples were randomly collected from the feces of children with diarrhea referred to Shahrekord hospitals and medical diagnostic laboratories. Stool samples were taken from the patients and part of the sample was frozen for the PCR test and part was used for modified Ziehl Neelsen staining. Samples in which cryptosporidium contamination was considered positive in terms of microscopy were also evaluated by the PCR test to determine the parasites species.

    Results

    The prevalence of Cryptosporidium in the collected samples was 3.5%. The results also showed that 85% of children with cryptosporidiosis were under 6 years old and only 15% of patients were between 6 and 12 years old (P <0.05). According to the results, the prevalence of Cryptosporidium was 5% in rural areas and 2% in urban areas (P <0.05). Also, 95% of infected villagers and 85% of infected urban children had contact with animals (P <0.05). The results showed that the prevalence was higher in spring and summer than in cold seasons (P <0.05). But the results revealed that there was no statistically significant difference between males and females affected (P> 0.05). The PCR test results showed that all positive samples were Cryptosporidium parvum.

    Discussion and Conclusion

    Cryptosporidium parvum is the predominant species in Chaharmahal and Bakhtiari province, which is more prevalent in children under 6 years old, in warm seasons and rural areas, and in people who have more contact with animals.

    Introduction

    Bioactive exopolysaccharides (EPSs) synthesized by lactic acid bacteria are therapeutic polysaccharides. EPSs are safe, so they can be used to make functional foods. Consumption of these foods can be a promising approach to prevent life-threatening and hard-to-treat diseases such as cancer and biofilm-associated infections. However, the low efficiency of synthesis makes their industrial implementation much more difficult. The aim of this study was to evaluate the anticancer and anti-biofilm activities of Lacticaseibacillus paracasei AS20(1) EPS and to optimize its production.

    Materials and Methods

    In the present study, the EPS synthesis by Lacticaseibacillus paracasei AS20(1) was optimized by Central Composite Design. Also, the effects of this EPS on inhibition of biofilm formation of Staphylococcus aureus (0.78-25 mg/ml), the viability of human colon cancer cell line, HT-29 (0.005-2 mg/ml), and the induction of apoptosis (2 mg/ml) in them were evaluated. Finally, the toxicity of this microbial EPS (50 and 100 mg/ml) was evaluated through a hemolysis assay of human red blood cells.

    Results

    According to the predicted model, the maximal EPS production was achieved at pH 7.2, yeast extract, and sucrose concentrations of 2.3%(w/v) and 33.3 g/L, respectively. This EPS showed promising anti-biofilm (11-70%) and anticancer activity against HT-29 (IC50=1451.42 µg/ml). EPS treatment increased the percentage of cells in the early apoptotic state (28.8%). This microbial EPS showed negligible toxicity (<5%) on red blood cells.

    Discussion and Conclusion

    Collectively, the ideal optimized conditions can be useful in large-scale production of EPS by Lacticaseibacillus paracasei AS20(1) which possesses valuable bioactivities.

    Introduction

    Enteroviruses (E.V.s) and human parechovirus (HPeV) play an important role in childhood diseases, from mild infections to severe illnesses which cause morbidity and mortality. Since the diagnostic procedures are not routinely accessible in the clinical laboratory, the epidemiology of enterovirus and parechovirus remains ambiguous. The current study investigates the frequency of human Enteroviruses and Parechovirus in hospitalized children.

    Materials and Methods

    Nasopharyngeal swabs were collected from children younger than five years with severe acute respiratory symptoms admitted to the hospital and screened for E.V.s and HPeV using RT-PCR and a two-step real-time PCR.

    Results

    From 80 nasopharyngeal swabs, viral agents were detected in 4 (5%) hospitalized children with ARTIs. All of these positive cases were infants. The results of RT-PCR of pan-HPeV demonstrate no detection of HPeV. Four samples were positive for enterovirus (5%). The frequency of E.V.s and HPeV between different sexes was not significant. Also, the distribution of E.V.s and HPeV among the other age groups was not significant (P>0.05).

    Discussion and Conclusion

    Enteroviruses and human Parechoviruses can cause severe consequences like neurological impairments, and more importantly, the epidemiological characteristics information in this regard is limited. Therefore, further research on viral respiratory tract infections is needed to improve public health and prevent severe complications.

    Introduction

    Organic meat products are well known for their probiotic values due to the presence of probiotic lactic acid bacteria such as Lactobacillus, Leuconostoc, Lactococcus, and Pediococcus strains. The present study aimed to isolate and characterize new strains of lactic acid bacteria (LABs) from organic meat sausages and evaluate their antimicrobial activities.

    Materials and Methods

    In this study, LAB strains were isolated from vacuum-packed organic meat sausages bought from Solico Group Co Ltd. (Tehran, Iran). All isolates were characterized by morphological, biochemical, and 16S rRNA analysis. The primary antibacterial activity of the isolates was evaluated against Micrococcus luteus PTCC 1408. The growth kinetics and antimicrobial activity of the strain MN-B against Lactobacillus casei ATCC 39392 were determined in an MRS broth medium. In addition, a bacteriocin produced by the strains MN-B revealed antimicrobial activity against selected foodborne pathogens. Bacteriocin produced by the MN-B strain was partially purified using the adsorption-desorption method followed by dialyzes.

    Results

    In this study, two Pediococcus spp. and three Leuconostoc spp. with antimicrobial activity were isolated from vacuum-packed organic meat sausages and designated as MN-A, MN-B, MN-A2, MN-A2-1, and MN-A3. The results showed that the isolate MN-A and MN-B belonged to the strain Pediococcus acidilactici and the MN-A2, MN-A2-1, and MN-A3 isolates were identified as Leuconostoc mesenteroides strains. Amongst the isolates, the strain MN-B showed the highest antimicrobial activity against the Micrococcus luteus PTCC 1408. The molecular weight of bacteriocin was about 5.0 KDa.

    Discussion and Conclusion

    Pediococcus acidilactici (strain MN-B) showed significant antimicrobial activity against Salmonella enterica subsp. enterica serotype Typhimurium PTCC 1709, Staphylococcus aureus PTCC 1113, Listeria monocytogenes PTCC1302, and Listeria inovani PTCC 1303. Therefore, this strain can be considered a potential candidate in different sectors such as the food industry as a preservative.

    Introduction

    Microbes that reside inside the plant tissues are known as endophytes. These microbes produce an array of compounds that have the potential to be used in modern medicine and agriculture. The aim of this study was to isolate, screen, and identify endophytic bacteria that have antimicrobial activity toward phytopathogenic bacteria. Aromatic plants such as Satureja khuzestanica are used in traditional medicine due to their secondary metabolites but information regarding their naturally occurring bacterial endophytes is limited.

    Materials and Methods

    In the present study, 27 strains of endophytes were isolated from S. khuzestanica. Based on the sequencing of the 16S rRNA gene, isolated bacterial strains with the highest activity against phytopathogenic bacteria belong to Bacillus, Streptomyces, and Pseudomonas genera. The bioactive secondary metabolites of these endophyte bacteria were extracted using ethyl acetate, followed by Gas Chromatography-Mass Spectrometry (GC-MS) analysis under standard conditions. The minimum inhibitory concentration (MIC) values were determined for five species of bacteria via the microbroth dilution technique. The crude extracts of the five selected bacterial endophytes indicated an antimicrobial activity against five phytopathogenic species.

    Results

    Data analysis showed significant differences in antimicrobial activity that ranged from 0.312 mg/ml to 2. 5 mg/ml. The minimum bactericidal concentrations ranged from 0.625 mg/ml to 10 mg/ml.

    Discussion and Conclusion

    The major compounds in GC-MS analysis of endophyte bacteria in this study were 2,4-di-tert-butylphenol, beta-d-glucopyranose, hexadecane, tetradecane,  eicosane, and dibutyl phthalate. This study reports for the first-time bacterial endophytes of S. khuzestanica, with antimicrobial activity against bacterial phypathogens. Our findings provide new insights into the antimicrobial activities of natural bacteria endophytes from S. khuzestanica.

    Introduction

    The fugal importance in terrestrial ecosystems is significant. However, our knowledge based on fungal distribution and its diversity in natural communities, especially in semi-arid ecosystems, seems to low. To have proper information on fungal community structure and the impact of environmental factors on the community composition, we assessed fungal diversity accompanied by soil chemical properties.

    Materials and Methods

    Soil sampling was done in two seasons, spring and autumn, from two different sites, which were both cold and warm in a semiarid ecosystem located in Kerman province, Iran. Soil chemical properties were measured according to standard methods. The fungal community was estimated by the next-generation sequencing of the ITS region (ITS1f-ITS2).

    Results

    By Illumina Miseq data analysis, 1542 Operational Taxonomic Units in soil samples were identified. Variation in the fungal community between the two sampling sites may be because of remarkable differences in environmental conditions, such as moisture and soil organic carbon. The dominant phylum was Ascomycota, followed by Basidiomycota.

    Discussion and Conclusion

    Various fungi were identified in this ecosystem. Further studies are necessary to reveal the possible roles of the dominant fungal species inhabiting semiarid soils.

    Introduction

    Flavonoids are a group of polyphenolic compounds  naturally found in plants. Many flavonoids are the basis of herbal medicines and have effective pharmacological effects. Rutin is one of the flavonoids  used as a reagent in the study of flavonoids and is also considered an antioxidant. This study was conducted to isolate and identify rutin-producing endophytic fungi from C. spinosa.

    Materials and Methods

    Sampling was performed in 10 regions of Iran and was taken from different tissues (including leaves, stems, roots, and fruits). After surface disinfection, explants were placed on a PDA medium and the fungal isolates were purified. Molecular identification of isolates was performed by sequencing ITS1-5.8S-ITS2. Morphological identification of fungi was performed by microscopic slide preparation and fungus identification keys. Methanolic extract of fungi was prepared by the maceration method and the presence of rutin was investigated using the HPLC technique.

    Results

    A total of 24 fungi were identified. HPLC analysis showed that the peak of standard flavonoids including rutin, quercetin, and apigenin appeared at 3.82, 7.60, and 12.40 min, respectively. Results showed that only two isolates were able to produce rutin. A comparison of the mean production of rutin showed a significant difference between the two fungi. Alternaria alternata M28 (22.94 ppm) had the highest amount of rutin production. Results also showed that rutin production in the mycelial tissue was more than in the extracellular medium, so A. alternata M28 and Paecilomyces maximus M7 had rutin content of about 67.5 and 5.56 times, respectively.

    Discussion and Conclusion

    Fungal rutin in methanolic extracts was characterized by HPLC. This is the first report showing that endophytic fungi have a potential for rutin production from C. spinosa and more research on these isolates can increase the amount of rutin production in them and lead to the possible commercialization.
