    آفتابگردان دانه روغنی (Helianthus annuus L.) به طور گسترده در سراسر جهان کشت می شود. شوری خاک بر بسیاری از صفات مورفولوژیک و فیزیولوژیک آفتابگردان تاثیر منفی می گذارد. ژن های MYB  بخشی از یک خانواده ژنی بزرگ از فاکتورهای رونویسی هستند که پروتیین های هسته ای را رمزگذاری می کنند و در مقاومت به شوری دخالت دارند. آفتابگردان روغنی متحمل به تنش شوری (AS5305)، در محیط کنترل شده در قالب طرح کاملا تصادفی با دو تکرار زیستی در شرایط نرمال و تنش شوری کشت شد. تنش شوری  هشت دسی زیمنس بر متر در مرحله هشت برگی از منبع NaCl اعمال گردید. حدود 24 ساعت پس از اعمال تنش شوری نمونه برداری از برگ ها انجام گرفت. ژن MYB44، به عنوان یکی از ژن های دخیل در مقاومت به تنش شوری، در ناقل های بیانی p35s-N و p35s-G همسانه سازی شده و تجزیه بیوانفورماتیکی بر روی آن انجام گرفت. تجزیه و تحلیل توالی MYB44 کلون شده به ترتیب بیش از 95% و 98% شباهت DNA  و پروتیین را بین ژن کلون شده با توالی MYB44 در NCBI نشان داد که نشان دهنده حفظ شدگی توالی بالای این ژن در بین ژنوتیپ های مختلف آفتابگردان است. به طور کلی، نتایج مطالعه حاضر بالقوه راه را برای به نژادی مولکولی آفتابگردان برای مقاومت به شوری هموار می نماید.

    گوجه فرنگی از نظر غذایی، اقتصادی و علمی از اهمیت زیادی دارد. با توجه به اهمیت زراعی و حساسیت این گیاه نسبت به اغلب تنش های زنده و غیرزنده، استفاده از روش های نوین در اصلاح این گیاه نیاز می باشد. در این تحقیق عوامل موثر در تراریزش یک رقم محلی گوجه فرنگی زراعی بنام بناب از قبیل مدت زمان پیش کشت (1، 2، 3 و4 روز)، غلظت آگروباکتریوم تومفشینس (1 و 8/0، 6/0، 4/0= OD600nm)، مدت زمان تلقیح (5، 10، 15 و 20 دقیقه)، سطوح مختلف استوسیرینگون (0، 100، 150 و 200 میکرومولار)، مدت زمان هم کشتی (1، 2، 3 و 4 روز) و غلظت های مختلف آنتی بیوتیک کانامایسین  (5، 10، 15 و 20 میلی گرم در لیتر) مورد بررسی قرار گرفت. نتایج نشان داد پیش کشت دو روزه ریز نمونه ها در تراریزش گیاهان اثر معنی داری داشت. بیشترین میزان تراریختی در غلظت باکتری 6/0=OD600nm  حاصل شد. همچنین بیشترین میزان تراریختی در مدت زمان تلقیح 10 دقیقه و مدت زمان هم کشتی دو روز حاصل شد. حضور استوسیرینگون اثر افزایشی در کارایی تراریختی گوجه فرنگی داشت، به طوریکه بیشترین میزان تراریختی در غلظت 150 میکرومولار استوسیرینگون بدست آمد. در نهایت حضور ژن کیتیناز در گیاهان تراریخت به وسیله PCR  مورد تایید قرار گرفت. شرایط بهینه بدست آمده در این مطالعه می تواند برای انتقال ژن های هدف به این رقم  گوجه فرنگی استفاده شود.

    تنش خشکی یکی از مهمترین عوامل محدود کننده تولید در بخش کشاورزی است. استفاده از گیاهان جایگزین متحمل به خشکی با کارآیی مصرف آب بالا از اهمیت به سزایی برخوردار است. ارزن دم روباهی (Setaria italicaL.) یکی از غلات مهم غذایی و علوفه ای متحمل به خشکی در مناطق خشک و نیمه خشک است. در این مطالعه، QTLهای شناسایی شده در شرایط آبیاری معمول و تنش خشکی، از مقالات مختلف گردآوری و روی نقشه ژنتیکی تلفیقی کروموزوم های ارزن مکان یابی شدند. از مجموع هشت مطالعه مستقل، تعداد 386 QTL مرتبط با صفات ارتفاع گیاه، کارایی مصرف آب، زیست توده، طول ریشه و طول پانیکول در ارزن دم روباهی جمع آوری شد. در شرایط آبیاری عادی و تنش خشکی به ترتیب تعداد 287 و 96 ناحیه QTL جمع آوری شد و روی نقشه مرجع ژنتیکی قرار گرفتند. متاآنالیز QTLها با استفاده از نرم افزار  BioMercator در نسخه 2/4 صورت گرفت. در مجموع تعداد 32 ناحیه متا QTL (MQTL) روی نه کروموزوم ارزن دم روباهی شناسایی شد که به طور میانگین فاصله اطمینان نواحی MQTL به نسبت 4/3 برابر کم تر از فاصله اطمینان QTLهای اولیه در مجموع صفات مورد بررسی بود. به منظور بررسی الگوی بیان ژن​ها در نواحی MQTL، داده ​های ریزآرایه و RNA-seq موجود در پایگاه داده​ها مورد آنالیز قرار گرفتند. از این بین تعداد 95 و 2172 ژن دارای بیان افتراقی در شرایط خشکی به ترتیب در چهار ناحیه MQTL با CI کمتر از یک میلیون جفت باز و در تمام مناطق MQTLها شناسایی شدند. در آنالیز "هاب" تعداد هشت ژن کلیدی شناسایی شد که جزو گروه عوامل تنظیمی و فاکتورهای رونویسی بودند. امید است نتایج این مطالعه بتواند در راستای گزینش مبتنی بر نشانگر (MAS) و استفاده در برنامه های به نژادی ارزن دم روباهی با هدف معرفی ژنوتیپ های برتر متحمل به خشکی مورد استفاده قرار گیرد.

    نخود (Cicer arietinum) یکی از مهمترین حبوبات است که حساس به سرما بوده و هر ساله کاهش ناگهانی دما منجر به افت عملکرد قابل توجه آن می شود. ریزRNA ها، RNA های کوچک غیررمزکننده بسیار محافظت شده ای هستند که بیان ژن را با تخریب mRNA های هدف پس از رونویسی یا سرکوب ترجمه آنها تنظیم می کنند و نقش آن ها در پاسخ به سرما در گیاهان مختلف به اثبات رسیده است. در این مطالعه، به منظور شناسایی ریزRNA های پاسخ دهنده به سرما، پس از توالی یابی ترانسکریپتوم در یک رقم متحمل (سارال) و یک لاین حساس به تنش سرما (ILC533)، توالی ژن های پاسخ دهنده به تنش سرما در نخود با استفاده ازنرم افزار c-mii مورد تجزیه و تحلیل قرار گرفت. سپس، ژن های هدف تعدادی از ریزRNA های مهم با استفاده از نرم افزار psRNA Target تعیین شدند. نتایج نشان داد که 30 ریزRNA در ژنوتیپ متحمل و 20 ریزRNA در ژنوتیپ حساس نخود به تنش سرما پاسخ دادند. با مقایسه پاسخ ریزRNA ها در دو ژنوتیپ مورد بررسی، ریزRNA هایmiR159 ،miR319 ، miR393 و  miR394 به عنوان ریزRNA های نامزد دخیل در تحمل سرما در نخود تعیین و نقش آنها مورد بررسی دقیق تر قرار گرفت. امید است نتایج به دست آمده در راستای ایجاد ارقام نخود متحمل به سرما مفید واقع شود.

    به این دلیل که بیوپسی مایع نسبت به بیوپسی بافتی ایمن تر و کم تهاجمی تر است، در سال های گذشته بررسی زیست نشانگر های موجود در آن برای پیش آگهی و تشخیص زود هنگام سرطان حایز اهمیت شده است. بررسی تغییر پروفایل بیانی پلاکت های آموزش دیده توموری موجود در بیوپسی مایع می تواند به عنوان یکی از زیست نشانگرها مورد استفاده قرار گیرد. استفاده از مدل های یادگیری ماشین دسته بندی با توجه به فضای ویژگی بر گرفته از داده های بیانی این پلاکت ها، توانایی پیش آگهی و تشخیص زود هنگام سرطان را به ما داده است. در این پژوهش میزان صحت و خطای هفت مدل دسته بندی در دو حالت دو دسته برای تشخیص نمونه های سالم و سرطانی و چند دسته برای تشخیص نمونه های سالم و انواع سرطان ها از یکدیگر مورد ارزیابی قرار گرفتند. این مدل ها روی پروفایل بیانی 2000 ژن پلاکت های آموزش دیده توموری مربوط به بیمارانی با سرطان های سینه، کبد، روده، مغز، پانکراس و ریه و همینطور پروفایل بیانی این ژن ها در 55 فرد سالم بررسی شدند. داده های مورد استفاده سری GSE68086 هستند که از پایگاه داده GEO دانلود شدند. همچنین روی این ژن ها با روش preranked GSEA تجزیه و تحلیل غنی سازی مسیر صورت گرفت. نتایج نشان داد مدل ماشین بردارهای پشتیبان با کرنل خطی و غیر خطی از بین مدل های دو دسته با میانگین خطای 05/0 و مدل ماشین بردارهای پشتیبان خطی از بین مدل های چند دسته با میانگین خطای 33/0 نرخ خطای کمتری دارند. به طور کلی نتایج حاصل دسته بندی پروفایل بیانی پلاکت های آموزش دیده توموری و تجزیه و تحلیل غنی سازی مسیرنشان دهنده این است که پروفایل بیانی پلاکت های آموزش دیده توموری را می توان به عنوان کاندید نشانگر زیستی در نظر گرفت.

    پسته یکی از مهم ترین محصولات باغی است که به دلیل دوپایه بودن و هتروزیگوسیتی بالا، دارای تنوع ژنتیکی زیادی است، از این رو شناسایی تنوع ژنتیکی موجود در بین ژنوتیپ های پسته در جهت اصلاح و تولید ارقام مقاوم به پسیل اهمیت ویژه ای دارد. پسیل پسته یکی از مهم ترین آفات درخت پسته است که پوره و حشرات بالغ این آفت با مصرف شیره گیاه سبب افزایش میزان پوکی، عدم خندانی و همچنین کاهش عملکرد می شود. در این پژوهش چندشکلی های تک نوکلیوتیدی و حذف و اضافه کوچک در چند رقم ایرانی پسته حساس (اکبری، کله قوچی، احمدآقایی، ممتاز) و مقاوم (بادامی زرند، حاج عبداللهی، ایتالیایی، سرخس) به پسیل مورد بررسی قرار گرفت. ابتدا خوانش ها با ژنوم مرجع هم ردیف شدند و سپس چندشکلی های تک نوکلیوتیدی و حذف و اضافه های کوچک مشخص شدند. درصد هم ردیفی خوانش ها با ژنوم مرجع بالای 96 درصد بود که نشان دهنده کیفیت بالای هم ردیفی بود. ارقام حساس به ترتیب دارای 2920688 و 701109 و ارقام مقاوم دارای 2919898 و 701000 چندشکلی تک نوکلیوتیدی و حذف و اضافه کوچک بود. در مجموع، نتایج تجزیه و تحلیل داده ها نشان داد که تنوع ساختاری و چندشکلی تک نوکلیوتیدی و حذف اضافه کوچک ژنوم در ارقام حساس بیشتر از ارقام مقاوم است.

    ویروس پالامپور پیچیدگی برگ گوجه فرنگی (Tomato leaf curl Palampur virus, ToLCPMV) یک  Begomovirus جدید با ژنوم دوبخشی از خانواده Geminiviridaeو تنها ناقل آن Bemisia tabaci می باشد. این ویروس اولین بار در سال 2008 از روی گوجه فرنگی در نواحی نیمه گرمسیری هند و سپس از پاکستان، عمان و عراق گزارش شد. به منظور شناسایی ToLCPMV در سال 1400، حدود 80 نمونه هندوانه دارای علایم ویروسی از مزارع شمال استان خوزستان (دزفول، شوش و اندیمشک) جمع آوری شد. آزمون PCR به کمک آغازگر دژنره Begomovirus (PCRv181/Bc) انجام و قطعه 550 جفت بازی مربوط به ژن Pre-coat protein روی DNA-A در نمونه های آلوده تکثیر شد. مقایسه توالی نوکلیوتیدی این ناحیه در NCBI، حضور ToLCPMV را در مناطق مورد مطالعه تایید کرد. آزمون PCR با استفاده از آغازگرهای اختصاصی گونه، توانست بخشی از DNA-B (ژن پروتیین حرکتی و ناحیه بین ژنی) را در 61 نمونه هندوانه تکثیر کند. ترادف های نوکلیوتیدی تعیین شده به ترتیب و به طور میانگین 67/96-03/90 و 28/97-43/91 درصد تشابه نوکلیوتیدی و اسیدآمینه ای را با دیگر توالی های موجود در NCBI نشان دادند. هر سه جدایه استان خوزستان بیشترین و کمترین شباهت را به جدایه های شهر جیرفت استان کرمان و جدایه های هند و پاکستان نشان دادند. ToLCPMV تاکنون از استان های هرمزگان، کرمان، فارس، یزد و خراسان رضوی از روی گوجه فرنگی، انواع کدو، خیار، خربزه و لوبیا و نیز از روی یک نمونه هندوانه در شهرستان خاش استان سیستان و بلوچستان گزارش شده بود. در پژوهش حاضر، پراکنش و میزان خسارت ToLCPMV در برخی مناطق مانند اندیمشک بسیار زیاد بود. این اولین گزارش از وقوع گسترده ToLCPMV در مزارع هندوانه استان خوزستان می باشد.

    پپتیدهای ضد میکروبی در برابر پاتوژن های گیاهی فعالیت های ضد میکروبی قوی نشان می دهند و جایگزین های خوبی برای سموم متداول هستند. در این مطالعه برای انتقال ژن پپتید ضد میکروبی rT1 گیاه چای ترش به گندم از سویه LBA4404 اگروباکتریوم حامل پلاسمید نوترکیب pBI121-rT1 استفاده شد. ریزنمونه های جنین بالغ چهار رقم گندم (سایسون، بهار، چمران و آرتا) پس از هم کشتی با سوسپانسیون باکتری به محیط گزینشی کالوس دهی حاوی دو میلی گرم در لیتر 2,4-D و دو میلی گرم در لیتر Picloram به همراه 50 میلی گرم در لیتر آنتی بیوتیک کانامایسین و 500 میلی گرم در لیتر سفوتاکسیم منتقل شدند. کالوس های جنین زا پس از رشد در محیط گزینشی، انتخاب و بعد به محیط باززایی حاوی 25 میلی گرم در لیتر کانامایسین و 250 میلی گرم در لیتر سفوتاکسیم انتقال یافتند. حضور و بیان ژن rT1 در سطح رونویسی در گیاهان تراریخت به کمک آغازگرهای اختصاصی به ترتیب با استفاده از روش های PCR و RT-PCR تایید شد. براساس نتایج از بین 15 گیاه تراریخت احتمالی فقط 11 گیاه تراریخت بودند. از بین 11 گیاه تراریخت، پنج گیاه مربوط به رقم سایسون با درصد تراریختی 6/1، چهار گیاه مربوط به رقم بهار با درصد تراریختی 3/1 و دو گیاه مربوط به رقم چمران با درصد تراریختی 6/0 بودند. از رقم آرتا هیچ گیاه تراریختی بدست نیامد.

    به منظور مقابله با Fusarium oxysporum عامل بیماری پژمردگی فوزاریومی در گوجه فرنگی، از اسید سالیسیلیک به عنوان القا کننده مقاومت استفاده شد. ریشه گوجه فرنگی با سوسپانسیون اسپور قارچ مایه زنی و با اسید سالیسیلیک 1/0 میلی مولار محلول پاشی شد. نمونه های برگی در فواصل صفر، 1، 3، 5، 7 و 14 روز پس از محلول پاشی تهیه و برای بررسی تغییر بیان ژن های کد کننده آنزیم های کاتالاز (CAT)، پراکسیداز (POX)، سوپراکسید دیسموتاز (SOD) و پروتیین های مرتبط با بیماری زایی (PR1 و PR10) به روش RT-qPCR مورد استفاده قرار گرفت. نتایج نشان داد که بیان ژن های CAT و SOD به عنوان آنزیم های آنتی اکسیدانت و موثر در مقابله با آثار منفی بیماری در بوته های آلوده در مقایسه با شاهد به شدت افزایش داشت و روند صعودی بیان این ژن ها تا روز چهاردهم پس از محلول پاشی ادامه داشت. ژن POX تا روز پنجم بیان افزایشی و سپس، کاهشی داشت. این امر نشان می دهد که ژن کاتالاز نقش موثرتری در ایجاد مقاومت به بیماری نسبت به پراکسیداز دارد. ژن های PR در بوته های سالم و بیمار در واکنش به تیمار اسید سالیسیلیک طی 14 روز روند افزایش داشتند که این افزایش برای بوته های بیمار بیشتر از بوته های سالم بود و بیانگر نقش القایی اسید سالیسیلیک در مقاومت به پژمردگی فوزاریومی است. بنابراین، تیمار اسید سالیسیلیک می تواند به عنوان یک گزینه مناسب برای ایجاد مقاومت و جلوگیری از خسارت به محصول گوجه فرنگی استفاده شود.

    نخود (Cicer arietinum) در رقابت با علف های هرز بسیار حساس بوده و دچار کاهش عملکرد و کیفیت محصول می شود. لذا تولید گیاهان مقاوم به علف کش های عمومی نظیر گلایفوسیت مفید می باشد. در این تحقیق، ازAgrobacterium tumefaciens  سویه LBA4404 حاوی پلاسمید pBI121 و ژن موتاسیون یافته epsps برای ایجاد مقاومت به علف کش گلایفوسیت استفاده شد که در آن ژن nptII به عنوان گزینشگر تحت پیش بر 35S قرار گرفته است. به منظور انتقال ژن از محورهای جنینی نخود رقم ثمین و بیونیج استفاده شد. ریزنمونه ها پس از هم کشتی با سوسپانسیون باکتری به مدت سه روز در محیط هم کشتی B5 قرار گرفت و پس از آن به منظور باززایی به محیط کشت MS به همراه غلظت مناسبی از هورمون های BAP، Kin و NAA منتقل گردید. درنهایت پس از گزینش های متوالی با استفاده از آنتی بیوتیک کانامایسین، ریشه زایی و استقرار گیاهان T0 در محیط هیدروپونیک انجام شد و با استفاده از آزمون PCR، در 15گیاه ژن اختصاصی epsps  و ژن  nptII تکثیر و تراریختگی آنها تایید شد. عدم آلودگی گیاهان مشکوک به تراریخته با استفاده از آغازگرهای نواحی ITS باکتری و همچنین با بررسی بیان ژن مورد نظر با استفاده از آزمون RT-PCR تایید شد و درصد تراریختگی در این بررسی 75/0 درصد بود.

    کیفیت میکروبی آب آشامیدنی اهمیت اساسی و جدی برای سلامت عمومی دارد. با توجه به کمبود اطلاعات جامع در خصوص میزان آلودگی آب آشامیدنی در سطح استان تهران این پژوهش به منظور ارزیابی میزان آلودگی آب آشامیدنی به باکتری سودوموناس آیروژینوزا مورد بررسی قرار گرفت. جداسازی ژن TEM-1 در سودوموناس آیروژینوزاهای مولد بتالاکتامازهای با طیف اثر گسترده (ESBL) و بررسی الگوی مقاومت آنتی بیوتیک این سویه ها اطلاعات مفیدی در مورد این باکتری بیماری زای فرصت طلب فراهم می آورد. در این پژوهش 300 نمونه آب آشامیدنی مورد بررسی قرار گرفت. تعیین حساسیت آنتی بیوتیکی نسبت به چهار آنتی بیوتیک به روش دیسک دیفیوژن انجام شد. از نظر ژنوتیپی وجود ژن بتالاکتاماز TEM-1 با روش PCR بررسی گردید. در این بررسی 17 نمونه (67/5 درصد) از آب های آشامیدنی به سودوموناس آیروژینوزا آلوده بودند. بیشترین میزان مقاومت نسبت به آنتی بیوتیک های ایمی پنم/رلباکتام (76/11درصد) و سفتازیدیم/آویباکتام (100%) در آب های آشامیدنی مشاهده شد. در بررسی ژنوتیپی با انجام آزمون PCR از 17 سویه فنوتیپ مثبت، 16 نمونه (1/ 94درصد) شامل ژن TEM-1 بودند. نتایج مشخص کرد که آب های آشامیدنی در برخی موارد دارای آلودگی با باکتری سودوموناس آیروژینوزا هستند. اغلب سویه های جداشده مقاوم به آنتی بیوتیک هستند و در میان سویه های تولید کننده ESBL، ژن TEM-1 دارای فراوانی بیشتری می باشد. نتایج بیانگر این است که ژن TEM-1 در ایجاد مقاومت به آنتی بیوتیک در سویه های جدا شده نقش بسزایی دارد. داده های حاصل از این پژوهش به توسعه اقدامات پیشگیرانه در برابر شیوع سویه های مقاوم به آنتی بیوتیک کمک می کند.

    حفظ ژرم پلاسم سیب زمینی (Solanum tuberosum) با توجه به نقش این گیاه در تامین امنیت غذایی از اهمیت بسیار زیادی برخوردار است. با توجه به هتروزیگوسیتی بالای سیب زمینی و تفرق شدید در نتاج حاصل از تولید مثل جنسی، ضروری است که از تکثیر غیرجنسی برای حفظ ریخته ارثی ژنوتیپ های منتخب استفاده شود. گرچه کشت درون شیشه ای، روشی کارآمد برای نگهداری منابع ژنتیکی سیب زمینی است ولی تحت شرایط بهینه نگهداری، به دلیل رشد سریع گیاهان، واکشت های متعدد نیاز است. از آنجا که این امر منجربه افزایش هزینه ها و افزایش احتمال وقوع تنوع سوماکلونال می شود، بنابراین ایجاد شرایط رشد حداقل در گیاهان تحت نگهداری در شرایط درون شیشه ای ضروری به نظر می رسد. در پژوهش حاضر به منظور یافتن بهترین تیمار موثر بر کاهش سرعت رشد ریز نمونه ها با حداقل اثر منفی بر آنها، اثر دو نوع آگار شامل آگار-آگار و آگار شیرینی پزی، اثر دو عامل اسموتیک سوربیتول و مانیتول در دمای 6 درجه سانتی گراد بر سه ژنوتیپ سیب زمینی مورد بررسی قرار گرفت. ریزنمونه ها به مدت 9 ماه بدون واکشت تحت تیمارهای رشدکند نگهداری شده و صفات طول اندام هوایی، تعداد جوانه بالقوه و وضعیت گیاهچه ها ثبت شد. نتایج حاصل از تجزیه و تحلیل داده ها نشان داد که در هر سه ژنوتیپ تحت بررسی، تیمار سوربیتول (در ترکیب با آگار-آگار یا آگار شیرینی پزی) در دمای 6 درجه به دلیل داشتن طول گیاهچه کمتر و وضعیت گیاهچه مطلوب و همچنین داشتن تعداد جوانه کافی جهت واکشت، تیمار برتر جهت نگهداری میان مدت ژرم پلاسم سیب زمینی است.

    Sunflower oilseed (Helianthus annuus L.) is widely planted around the world. Soil salinity negatively affects many morphological and physiological traits of sunflowers. MYB genes are a large gene family of transcription factors that encode nuclear proteins and involved in salinity resistance. Oil seed sunflower line tolerant to salinity stress (AS5305) was planted in normal and salinity stress conditions in a completely randomized design with two biological replications in a controlled environment. Salinity stress of 8 ds.m-1 was applied in 8-leaf stage from NaCl source. About 24 hours after salinity stress, leaves were sampled. The MYB44 gene, as one of the genes involved in salinity stress resistance, was cloned in expression vectors p35s-N and p35s-G, and bioinformatics analysis was performed about it. Analysis of cloned MYB44 sequence revealed more than 95% and 98% DNA and protein similarity respectively, between cloned gene with the MYB44 sequence in the NCBI, indicating high sequence conservation of this gene among different sunflower genotypes. Overall, the results of present study potentially pave the way for molecular breeding of sunflower for salinity resistance.

    Tomato is an important economic crop with nutritional and scientific value for study. Due to its agronomic importance and the sensitivity to biotic and abiotic stresses, using new methods in breeding this crop is necessary. In this study, the effects of some factors, such as pre-cultivation time (1, 2, 3 and 4 days), Agrobacterium concentration (OD600nm = 0.4, 0.6, 0.8, and 1), inoculation time (5, 10, 15 and 20 minutes), Acetosyringone concentration (0, 100, 150, and 200 μM), co-cultivation period (1, 2, 3 and 4 days) and different concentrations of the kanamycin (5, 10, 15 and 20 mg/l) on the transformation of a local cultivar of tomato called Bonab, were evaluated. The maximum transformation efficiency was obtained at the OD600= 0.6. Also, the highest transgenic plants were obtained in the 10 minutes of inoculation and 2 days of co-cultivation. The presence of Acetosyringone had an increasing effect on the tomato transformation, and the highest percentage of transformation was obtained at 150 M of Acetosyringone. Finally, the presence of a Chitinase gene in the probable transgenic plants was confirmed by PCR. The optimal conditions that were obtained in this study can be used to transfer target genes to this tomato cultivar.

    Drought stress is one of the most important factors limiting production in the agricultural sector. The use of drought-tolerant alternative plants with high water use efficiency is of great importance. Foxtail millet (Setaria italica L.) is one of the important drought tolerant fodder and food grains in semi-arid regions. In this study, drought stress associated QTLs were collected and located on the integrated genetic map of millet chromosomes. From a total of eight studies, 386 QTLs associated with yield controlling traits including plant height, water use efficiency, plant biomass, root length and panicle length were collected in foxtail millet. Under drought stress and normal irrigation conditions, 96 and 287 QTLs were collected and projected on the reference consensus genetic map, respectively. The meta-analysis of QTLs was conducted using BioMercator software V4.2. A total of 32 meta-QTLs (MQTLs) were detected on 9 foxtail millet chromosomes with a confidence interval of 3.4 times lower than the average of the original QTLs. In order to investigate the pattern of gene expression in the MQTL regions, available microarray and RNA-seq data sets in the databases were analyzed. Based on the achieved results, 95 and 2172 differentially expressed genes at drought conditions were identified in 4 MQTLs with a CI of less than 1 Mbp and in all MQTL regions, respectively. Also, eight key genes were identified by the hub analysis which belong to transcription factors and regulatory elements. We hope that the results of this study would be useful for marker assisted selection (MAS) and foxtail millet breeding programs with the aim of development of new high yielding drought-tolerant genotypes.

    Chickpea (Cicer arietinum) is one of the most important legumes, classified as a cold-sensitive species, and each year, the sudden drop in temperature leads to a significant loss of chickpea yield. miRNAs are highly conserved small noncoding RNAs that regulate expression of their target genes by degradation of their mRNAs or suppressing their translation, and their roles have been proved in cold stress response in various plants. In the current study, in order to identify cold stress responsive miRNAs in chickpeas, following transcriptome sequencing of a cold stress tolerant cultivar (Saral) and a cold stress sensitive line (ILC533) in response to cold stress, the sequences of the cold responsive genes were analyzed by c-mii software to identify possible miRNAs. Then, the target genes of a number of important miRNAs were identified using psRNA target software. The results showed that 30 and 20 miRNAs responded to cold stress in the tolerant and sensitive genotypes, respectively. Through comparative analysis of cold responsive miRNAs in the contrasting genotypes, miR319, miR393, miR394 and miR159 were demonstrated as candidate miRNAs involved in cold tolerance and their roles was further inspected. It is hoped that the obtained results will be useful for development of cold tolerant chickpea cultivars.

    Since liquid biopsy is less invasive than tissue biopsy, studies on liquid biopsy biomarkers for the early detection of cancer and diagnosis are taken into consideration. Expression profiles of tumor-educated platelets (TEP) in liquid biopsy can be used as one of the biomarkers. The use of classification machine learning models, according to the features space derived from the expression data of TEPs, has given us the ability to predict cancer. In this study, we evaluate different types of classification models namely SVM, LDA, logistic regression, boosting, classification tree, and random forest, on the expression profile of TEPs in 230 patients with breast, liver, colorectal, brain, pancreatic, and lung cancers, as well as the expression profile of these genes in 55 healthy individuals. These models were examined on the expression profile of 2000 high variance selected genes. Also, pathway enrichment analysis was performed on these genes by the GSEA preranked method. The results showed that linear SVM and polynomial SVM models have lower error rates than two-class models and linear SVM models have lower error rates than multi-class models. In general, the results of TEP expression profile classification and pathway enrichment analysis indicate that the expression profile of TEPs can be considered as candidate biomarkers.

    Pistachio is one of the most important crops that have a large genetic diversity due to its dicotyledonous and high heterozygosity, so identifying these genetic differences is of particular importance for breeding and production of psyllid-resistant pistachio cultivars. Pistachio psyllid is one of the most important pests of the pistachio tree. The nymphs and adult insects of this pest increase the amount of porosity, and reduce yield and lack of laughter, by consuming plant sap. In this study, single nucleotide polymorphisms (SNP) and small indels were investigated in Iranian pistachio sensitive cultivars (Akbari, Kalehghoochi, Ahmad Aghaei, Momtaz) and resistant cultivars (Badami Zarand, Haj Abdollahi, Italian, Sarakhs) to psyllids. The readings were first aligned with the reference genome, and then single-nucleotide polymorphisms and deletions, and small indels were identified by the GATK toolkit. The percentage of alignment of the readings with the reference genome was above 96%, which indicated the high quality of alignment. Sensitive cultivars had 2920688 and 701109 and resistant cultivars had 2919898 and 701000 SNP and small indels, respectively. In general, the results of data analysis showed that the structural and polymorphic diversity of mononucleotides and the small indels are greater in susceptible cultivars than in resistant cultivars.

    Tomato leaf curl Palampur virus (ToLCPMV) is a novel Begomovirus from Geminiviridae family with a bipartite genome and Bemisia tabaci is its uniqe vector. This virus was reported on tomato in the subtropical regions of India for the first time in 2008 and then from Pakistan, Oman and Iraq. In order to identify this virus, about 80 samples of watermelon with viral symptoms were collected from watermelon fields in the north regions of Khouzestan province (Dezfoul, Shoush and Andimeshk) in 2021. PCR was performed with Begomovirus degenerate primer (PCRv181/Bc) and 550 bp fragment of pre-coat protein gene on DNA-A was amplified in the infected samples. Comparison of the nucleotide sequence of this region in NCBI confirmed the presence of ToLCPMV in the studied cities. PCR was able to amplify a part of DNA-B (a part of Movement protein gene and intergenic region) using species-specific primers in 61 watermelon samples. The determined nucleotide sequences showed an average of 90.03-96.67 and 91.43-97.28% nucleotide and amino acid identity with the other NCBI sequences, respectively. The isolates of Khouzestan province showed the most and the least similarity to the isolates of Jiroft city (Kerman province) and the isolates of India and Pakistan. ToLCPMV has been reported from Hormozgan, Kerman, Fars, Yazd and Khorasan Razavi provinces in terms of tomato, types of squash, cucumbers, melons and beans, as well as a watermelon sample in Khash city of Sistan and Balouchestan province. In the present study, the distribution and the amount of damages of ToLCPMV was too much in some areas such as Andimeshk. This is the first report of the widespread occurrence of ToLCPMV in the watermelon fields in Khouzestan province.

    Antimicrobial peptides display powerful antimicrobial activities against a large set of plant pathogens and are a good alternative to conventional toxins. To transfer rT1 gene of Hibiscus sabdariffa to wheat, Agrobacterium tumefaciens strain LBA4404 harboring the pBI121-rT1 plasmid was used. Mature embryo explants of four wheat cultivars including the Saison, Bahar, Chamran and Arta after co-cultivated with A. tumefaciens suspension were transferred to a selective callus medium containing 2 mg/l 2,4-D, 2 mg/l Picloram, 50 mg/l Kanamycin and 500 mg/l Cefotaxime. Embryogenic calli were selected after growth in a selective medium and then transferred to a regenerative medium containing 25 mg/l Kanamycin and 250 mg/l Cefotaxime. The presence and expression of rT1 gene at the transcriptional level in transgenic plants were confirmed by PCR and RT-PCR methods respectively using specific primers. Based on the RT-PCR results, the rT1 gene was expressed in transgenic wheat plants. In this study, out of 15 possible transgenic plants, only 11 were transgenic. Among transgenic plants, five plants belonged to Saison cultivar with 1.6 transgenic percentage, four plants represented to the Bahar cultivar with 1.3 transgenic percentage and two plants belonged to the Chamran cultivar with 0.6 transgenic percentage. No transgenic plant was obtained for the Arta cultivar.

    In order to control Fusarium oxysporum, responsible for fusarium wilt disease, in tomatoes, salicylic acid was used as a resistance inducer. Tomato root was inoculated with pathogen suspension and sprayed with 0.1 mM salicylic acid. Leaf samples were prepared at 0, 1, 3, 5, 7 and 14 days after foliar application to evaluate the expression of genes encoding catalase (CAT), peroxidase (POX), superoxide dismutase (SOD) and pathogenesis related proteins (PR1 and PR10) using RT-qPCR. The results showed that the expression of CAT and SOD genes as antioxidant enzymes and effective against negative consequences of disease in infected plants increased sharply compared to the control and the upward trend of expression of these genes continued until the fourteenth day after foliar application, but the POX gene increased until the fifth day and then decreased. This indicates that the catalase gene has a more effective role in disease resistance than peroxidase. PR genes in both healthy and diseased plants increased over 14 days in response to salicylic acid treatment, which was greater for diseased plants than for healthy plants and indicates the induced role of salicylic acid in Fusarium wilt resistance. Therefore, salicylic acid treatment can be used as an ideal candidate to create resistance and prevent damage to the tomato crop.

    Chickpea (Cicer arietinum) is very sensitive in competition with weeds and suffers from a decrease in yield and product quality. Therefore, it is useful to produce plants resistant to general herbicides such as glyphosate. In this research, Agrobacterium tumefaciens strain LBA4404 was used. This bacterium contains plasmid pBI121 and mutated epsps gene of Escherichia coli to create resistance to glyphosate herbicide. In this plasmid nptII gene is placed under 35S promoter as a selective gene. In order to transfer the gene, embryonic axes of chickpea varieties (Samin and Bivanij) were used. Chickpea embryonic axes were placed on B5 co-culture medium after co-cultivation with bacterial suspension for 3 days and then transferred to MS culture medium with appropriate concentration of BAP, Kin and NAA hormones for regeneration. Finally, after successive selections using kanamycin antibiotic, rooting and establishment of T0 plants were done in hydroponic environment. Then, PCR amplified epsps and nptII genes in 15 plants and their transgenecity was confirmed. Non-contamination of DNA in putative transgenic plants was confirmed by using the primers of the bacterial ITS regions. Also the expression of the target gene verified using RT-PCR test and the transgenic percentage was 0.75% in this study.

    The microbial quality of drinking water is fundamental and has serious importance for public health. Regarding to the clinical significance, Pseudomonas aeruginosa was surveyed to evaluate the degree of the contamination of drinking water and the bacterial resistance to antibiotics. Isolation of the TEM-1 gene in ESBL-producing Pseudomonas aeruginosa and the pattern of antibiotic resistance provide useful information about this opportunistic pathogen. In this study, the prevalence and antibiotic resistance of Pseudomonas species distributed in drinking water were investigated. 300 samples of drinking water were examined. Determination of antibiotic susceptibility of strains to four antibiotics was performed by disk diffusion method. The ESBL-producing P. aeruginosa containing TEM-1 gene were detected by PCR using TEM-1 specific primers, 17 samples (5.67%) in drinking water showed P. aeruginosa contamination. The highest resistance to imipenem/relabactam (11.76%) and ceftazidime/avibactam antibiotics (100%) was observed in drinking water. According to PCR results of 17 positive phenotype strains, 16 isolates (94.11%) carried TEM-1 gene. Results indicated the contamination of drinking water with P. aeruginosa. Most isolated strains are resistant to antibiotics and TEM-1 gene is more abundant among ESBL-producing strains. The results indicate that TEM-1 gene plays an important role in antibiotic resistance in isolated strains.

    Conservation of potato germplasm (Solanum tuberosum L.) is of great importance due to its role in providing food security. Considering high heterozygosis of potato and severe segregation in the progeny obtained from sexual reproduction, it is necessary to use asexual propagation to maintain selected genotypes. Although in vitro culture is an efficient method for maintaining potato genotypes, rapid growth of plants under optimum growth conditions, makes multiple subcultures necessary to be done. The later leads to increased costs and an increase in the probability of somaclonal variation. Therefore, it seems to be necessary to induce slow growth in plants maintained under in vitro conditions. In the current research the aim was to find out the best in vitro treatment for reducing the growth of potato shoots with the least negative effect on them. To approach the aim, two types of agar (including agar-agar and confectionery agar) and two osmotic agents (including sorbitol and mannitol) were evaluated on three potato genotypes in 6℃. The traits of shoot length, number of possible buds and microplant condition were recorded after 9 months. The results of data analysis showed that in all the three investigated genotypes, sorbitol osmotic agent (in combination with agar-agar or confectionery agar) at the temperature of 6 ℃ was the best treatment for medium-term storage of potato germplasm. It was mainly due to having the least shoot length, good microplant condition and also having enough buds for subsequent subcultures.
