    هدف
    تحقیق ما بیان ژن ITPA را به عنوان یک عامل زمینه ساز ایجاد اختلالات ژنتیکی مشاهده شده در این سلول ها ارزیابی شد.
    مواد و روش ها
    برای ارزیابی بیان ژن مورد نظر از روش RT-PCR نیمه کمی استفاده و برای بررسی صحت عملکرد محصول ژن cDNAهای به دست آمده کلون و تعیین توالی شد. پروتئین های حاصل از cDNAها با استفاده از نرم افزارهای بیوانفورماتیکی از نظر ساختمانی پیشگویی و مقایسه شدند.
    نتایج
    نتایج حاصل از پیش بینی ساختمانی بیانگر آن است که mRNA جدا شده قادر به کد کردن پروتئینی است که فاقد کارایی لازم در اتصال به سوبسترا و انجام واکنش آنزیمی است. با توجه به لزوم تشکیل دایمر برای فعالیت طبیعی پروتئین، عملکرد مونومر طبیعی ITPase نیز در هنگام تشکیل هترودایمر تحت تاثیر قرار می گیرد. بنابراین به نظر می رسد که عمل آنزیمی ITPase در سلول هایK562 طبیعی نبوده و می توان آن را به عنوان یک عامل زمینه ساز ناپایداری ژنتیکی در این سلول ها در نظر گرفت.
    نتیجه گیری
    بررسی بیان ژن در سلول های K562 نشان داد که در مقایسه با بیان ژن کنترل داخلیGAPDH، ITPA بیانی در حد متوسط در این سلول ها داشته و دو نوع رونوشت در این سلول ها تولید می کند. یکی از آنها حاصل پردازش طبیعی hnRNA اولیه است ولی رونوشت دوم دارای یک حذف 51 نوکلئوتیدی در ناحیه کد کننده mRNA است. به نظر می رسد که این رونوشت حاصل یک پیرایش نادر در سلول است.

    هدف
    مطالعات اخیر بیان می کند که هیپرکسی نورموباریک (HO) متناوب و پیوسته باعث ایجاد پدیده تحمل به ایسکمی می شود و از این طریق از آسیب های ناشی از برقراری مجدد جریان جلوگیری می کند. هدف از این مطالعه بررسی تغییرات بیان ناقل اسیدهای آمینه تحریکی شماره 3(EAAT3)، سطح فاکتور نکروز تومور آلفا (TNF-α) سرم و فعالیت فاکتور هسته ای کاپا B به دنبال پیش شرطی سازی با HO پیوسته و متناوب است.
    مواد و روش ها
    رت ها به چهار گروه آزمایشی تقسیم شدند و هر گروه حاوی 21 حیوان بود. دو گروه اول به صورت پیوسته (24 ساعت مداوم) و متناوب (4 ساعت در روز به مدت 6 روز) در معرض اکسیژن 95 درصد به نام HO متناوب و پیوسته قرار گرفتند. دو گروه دوم به عنوان گروه های کنترل همانند دو گروه اول به صورت پیوسته (24 ساعت مداوم) و متناوب (4 ساعت در روز به مدت 6 روز) در معرض اکسیژن 21 درصد به نام نورموکسی نورموباریک (RA؛ هوای اتاق) پیوسته و متناوب قرار گرفتند. هر گروه به سه زیر گروه به نام زیرگروه انسداد شریان راست مرکزی مغز(MCAO)، زیرگروه گروه شم MCAO (جراحی بدون ایسکمیMCAO)، و زیرگروه دست نخورده (بدون هیچ گونه جراحی) تقسیم شدند. بعد از 24 ساعت برقراری جریان خون مجدد بعد از 60 دقیقه ایسکمی، میزان نقص نورولوژیک(NDS) در زیر گروه MCAO بررسی شد. بلافاصله و 48 ساعت بعد از پیش درمان، خونگیری به منظور اندازه گیری سطح TNF-α سرم انجام شد. بعد از 48 ساعت، رت ها برای نمونه برداری مغزی قربانی شدند تا اثر پیوسته و متناوب بر تغییرات فعالیت فاکتور هسته ای کاپا B بیان و EAAT3 و سطح TNF-α سرم بررسی شود.
    نتایج
    پیش شرطی سازی با HO پیوسته و متناوب باعث کاهش NDS می شود. پیش درمان با HO پیوسته و متناوب باعث افزایش بیان EAAT3 و سطح TNF-α سرم می شود.
    نتیجه گیری
    اگرچه مطالعات بیشتری برای وضوح مکانیسم های تحمل به ایسکمی لازم است، اما HO پیوسته و متناوب ظاهرا تا حدی آثارشان را از طریق افزایش بیان ناقل اسیدهای آمینه تحریکی و سطح TNF-α سرم انجام می دهند.

    هدف
    سلول های دندریتیک در تنظیم وکنترل پاسخ های ایمنی نقش مهمی به عهده دارند. امروزه نظر بر این است که از این سلول ها در درمان بسیاری از بیماری ها می توان استفاده کرد. یکی از راه های ایمونوتراپی ایجاد سلول های دندریتیک تولروژن از طریق ممانعت از بیان مولکول های کمک تحریکی است.CD40 یکی از مولکول های کمک تحریکی بوده که با ممانعت از بیان آن از طریق روش هایی مانند آنتی سنس و یا SiRNA می توان به این مهم (سلول های دندریتیک تولروژن) دست پیدا کرد. با تولید سلول های دندریتیکی تولروژن می توان افقی روشن در درمان بسیاری از بیماری ها ایجاد نمود و با طراحی RT-PCR کمی برای اندازه گیری میزان بیان می توان راه را برای ایجاد این سلول ها هموار نمود. در این تحقیق با طراحی آنتی سنس با استفاده از نرم افزارهای مربوط و منتقل کردن آن به سلول های دندریتیکی با استفاده از ماده”لیپوفکتامین 2000“(Invitrogen) به سلول دندریتیک تولروژن رسیده ایم.
    مواد و روش ها
    در این مطالعه سلول های دندریتیک از طحال موش Balb/c جدا شد و میزان خلوص به وسیله فلوسیتومتری بر اساس نشانگر CD11Cتعیین شد. رده سلولیBCL1 به عنوان گروه کنترل، بیان کننده CD40 در محیط کشت FCS 10% RPMI-1640+ کشت داده شدند. با استفاده نرم افزارهای بیوانفورماتیک Beacon Designer و mfold and Blast آغازگر برای) GADPHبه عنوان کنترل داخلی) و CD 40 طراحی شد، کیت RNeasy Plus Mini Kit (Qiagen) برای استخراج RNA مورد استفاده قرار گرفت و با تعیین O.D280/260 و الکترفورز در آگارز میزان خلوص تعیین شد. در ادامه کار ابتدا PCR کیفی با استفاده از آنزیم Fast Hot Start Taq (Roche) بهینه شد و سپس بعد از ساخت cDNA با استفاده از کیت IQ sybergreen (Biorad)،RT-PCR کمی برای CD40 بهینه و در نهایت با استفاده از همین کیت منحنی استاندارد پس از تهیه رقت های مناسب از RNA برای CD40 و کنترل داخلی محاسبه شد. پس از طراحی آنتی سنس با استفاده از نرم افزار های بیوانفورماتیک با استفاده از ماده”لیپوفکتامین 2000“انتقال، انجام و با استفاده از RT-PCR کمی میزان کاهش بیان ژن مشخص شد.
    نتایج
    دمای بهینه اتصال با استفاده ازشیب Real time PCR و بهترین CT و Rn∆ در 5/95 درجه سانتی گراد برای هر دو ژن CD40 و GADPH تعیین و همچنین دمای ذوب PCR حدود 84 درجه سانتی گراد مشخص شد. شیب و بازدهی واکنش PCR برای ژن های CD40 و GADPH با روش رقت سریال تعیین شد. با استفاده از غلظت نابرابر آغازگرها بازدهی و شیب برابری این دو ژن با دمای اتصال یکسان تعیین شد. (بازده CD40: 5/96، شیب: 408/3-؛ بازده GADPH: 1/94، شیب: 471/3-) میزان کاهش بیان ژن پس از انتقال با آنتی سنس 64/1در سلول های دندریتیکی و در رده سلولی BCL132/1 تعیین شد. ماده منتقل شده تاثیری روی بیان ژن ندارد. بهترین زمان برای اندازه گیری کاهش بیان ژن CD40 48 ساعت پس از انتقال در سلول های دندریتیکی و رده سلولی BCL1 است.
    نتیجه گیری
    در مطالعات مختلف از روش های متفاوتی مانند RT-PCR نیمه کمی RT-PCR کمی مطلق و RT-PCR کمی نسبی برای تعیین میزان بیان ژن های مختلف ازجمله CD40 استفاده شده است. بررسی ها در این تحقیق نشان داد که استفاده از کیت IQ–sybergreen (Biorad) و الیگومر دی اکسی تیمیدین برای سنتز cDNA برای رسم منحنی استاندارد ژن CD40 بسیار مناسب است (در این مورد GADPH کنترل داخلی مناسبی است) که در مقایسه با RT-PCR نیمه کمی دقیق تر و قابل اعتمادتر است.

    هدف
    در این تحقیق روش Real Time RT-PCRبا استفاده از رنگ سایبرگرین به منظور سنجش کمی ویروس نقص سیستم ایمنی انسانی نوع یک (HIV-1) راه اندازی شد.
    مواد و روش ها
    این آزمایش بر اساس تکثیر ناحیه ژن pol ویروس بوده و از دستگاه ترموسایکلرBI 7500 استفاده شد. 26 بیمار مبتلا به HIV-1 با این روش مطالعه شدند و سپس نتایج با آزمون تجارتی استاندارد کوباس آمپلی کور مونیتور تست (COBAS Amplicor HIV-1 Monitor Test)مقایسه شد.
    نتایج
    نتایج حاصل از این تحقیق نشان داد که این روش قادر به اندازه گیری حداقل 500 کپی از HIV-1 RNA در هر میلی لیتر است. علاوه بر این، آزمایش در محدوده 5-109×5 کپی از RNA ویروس در هر میکرولیتر خطی است. از آنجایی که در هیچ کدام از افراد کنترل هیچگونه علامت مثبتی نشان داده نشد، ویژگی این روش 100 درصد محاسبه شد. اندازه گیری کمی HIV-1 در بیماران با استفاده از روش راه اندازی شده و روش استاندارد نشان داد که همبستگی بسیار خوبی بین دو روش وجود دارد (95/0=R2).
    نتیجه گیری
    بر اساس آخرین اطلاعات موجود، شیوع HIV در جمهوری اسلامی ایران در یک وضعیت هشدار دهنده در حال افزایش است. از آنجایی که سطح پلاسمایی ویروس، موثرترین شاخص برای بررسی درمان بیماری است، اندازه گیری سطح پلاسمایی HIV-1 ابزار بسیار مهمی برای پیگیری فعالیت ویروس در افراد آلوده است. در این راستا راه اندازی و توسعه روش های اندازه گیری HIV-1 RNA ابزار منحصر به فردی است که به پزشک برای آگاهی از وضعیت درمان در بیمارانی که تحت درمان ضد رتروویروسی قرار گرفته اند کمک می کند. در این تحقیق روشSYBR-green Real Time RT-PCR را به منظور سنجش کمی HIV-1 در بیماران آلوده راه اندازی شد. از آنجایی که در این روش از RNA استاندارد سنتز شده استفاده شد، محدوده شناسایی بالادست در این روش بسیار بالاتر از روش استاندارد بود (109 ×5 در مقابل 105×5/7). بنابراین اگر نیازی به محدوده دینامیکی گسترده تری باشد- مانند موارد حاد بیماری- این استاندارد مفید خواهد بود. در این تحقیق تکرارپذیری در سطح درون سنجش (Intra assay) و بین سنجش (Inter assay) بررسی شد. ضریب تغییرات Ct به دست آمده در آزمون تکرارپذیری درون سنجش و بین سنجش به ترتیب کمتر از 3 و 5/4 درصد بود که نشان می دهد این روش تکرارپذیری بالایی دارد. با توجه به نتایج به دست آمده روش راه اندازی شده، می تواند جایگزین مناسبی برای روش استاندارد در سنجش کمی HIV-1 باشد.

    به دلیل ضرورت درمان آسیب های سیستم عصبی و همچنین نقش ژن سوروایوین در تکثیر سلولی و مرگ برنامه ریزی شده سلول، تغییرات بیان این ژن در طول دوره ترمیم در اعصاب سیاتیک آسیب دیده و همچنین قطعات نخاعی مرتبط با عصب سیاتیک بررسی شد.
    مواد و روش ها
    موش های نر بالغ نژاد «ان ماری» به عنوان مدل مورد استفاده قرار گرفتند که پس از بیهوش کردن آن ها، عصب سیاتیک پای راست موش ها برش عرضی داده شد و سپس در زمان های مشخص (3، 6، 12، 24، 48، 96 و 144 ساعت) پس از آسیب، موش ها کشته شده و قطعات انتهایی و ابتدایی عصب قطع شده، عصب دست نخورده پای چپ و قطعاتL4- L6 نخاعی که مربوط به عصب سیاتیک هستند، نمونه گیری شدند. RNA کل هر نمونه استخراج و سپس واکنش RT-PCR ((Reverse Transcriptase-Polymerase Chain Reaction نیمه کمی با آغازگرهای اختصاصی برای ژن سوروایوین و ژن 2m، به عنوان کنترل داخلی، گذاشته شد. برای تعیین توزیع سلولی پروتئین سوروایوین، 6 روز (144 ساعت) پس از قطع عصب، بیان این پروتئین با استفاده از آنتی بادی اختصاصی با روش ایمونوهیستوشیمی ارزیابی شد.
    نتایج
    نتایج این تحقیق نشانگر بیان دو واریانت سوروایوین 140 و سوروایوین 40 در قطعه انتهایی و ابتدایی عصب آسیب دیده با شدت های مختلف بود به گونه ای که میزان بیان سوروایوین 140 بیشتر از بیان سوروایوین 40 بود و در قطعات نخاعی، تنها بیان واریانت سوروایوین 140 شناسایی شد. همچنین در بررسی ایمونوهیستوشیمی قطعات نخاعی، هم توزیع هسته ای و هم توزیع سیتوپلاسمی پروتئین سوروایوین مشاهده شد در حالی که بیان این پروتئین در هیچ کدام از قطعات انتهایی یا ابتدایی عصب سیاتیک تشخیص داده نشد.
    نتیجه گیری
    نتایج بیانگر آن است که ژن سوروایوین به طور متمایزی در طول دوره ترمیم در عصب و نخاع آسیب دیده بیان و پردازش می شود. همچنین نتایج نشان دادند که دستکاری در بیان یا پردازش رونوشت اولیه سوروایوین می تواند تاثیر بالقوه ای در فرآیند ترمیم در آسیب های اعصاب یا نخاع داشته باشد.

    هدف
    مهار مرگ برنامه ریزی شده سلول ها، یکی از عوامل آغاز و پیشرفت تومور است و مهارکنندگان مرگ برنامه ریزی شده سلول، در بروز سرطان نقش ویژه ای ایفا می کنند. سوروایوین یکی از مهارکنندگان مرگ برنامه ریزی شده سلول است که اخیرا به عنوان یک نشانگر تومور احتمالی برای تشخیص و پیش آگهی تومورهای مثانه مورد توجه قرار گرفته است. پروتئین سوروایوین عملکرد دوگانه ای هم در تنظیم تقسیم سلولی و هم در کنترل مرگ سلولی دارد و در اغلب سرطان های انسانی افزایش بیان دارد. در این تحقیق شایستگی بیان سوروایوین، به عنوان نشانگرهای ملکولی ذاتی در پیش آگهی مبتلایان به سرطان مثانه، به خصوص بیماران دارای عود مجدد تومور مورد بررسی قرار گرفته است.
    مواد و روش ها
    بلوک های پارافینه از آرشیو بیمارستان لبافی نژاد به دست آمدند و پس از بررسی پرونده پنج ساله بیماران، با تکنیکRT-PCR مورد بررسی قرار گرفتند. در مجموع تعداد 51 نمونه پارافینه متعلق به 30 بیمار مبتلا به سرطان مثانه، از نظر بیان این ژن ارزیابی شدند. همچنین توزیع بافتی و جایگاه درون سلولی پروتئین حاصل از این ژن با استفاده از روش ایمونوهیستوشیمی مورد بررسی قرار گرفت.
    نتایج
    بیان سوروایوین در 6/66 درصد نمونه ها مشخص شد. طبق یافته های این تحقیق، بیان سوروایوین با افزایش درجه و بدخیمی تومور، افزایش می یابد. میزان بیان سوروایوین در عودهای دوم و سوم تومور، افزایش داشت. همچنین با افزایش بدخیمی و در نتیجه افزایش بیان سوروایوین شانس زنده ماندن در طی 5 سال پیگیری، به طور معناداری کاهش داشت (036/0(P=. نتایج ایمونوهیستوشیمی نیز مؤید این بودند که پروتئین سوروایوین در سلول های توموری بیان شده و تجمع هسته ای دارد.
    نتیجه گیری
    در مجموع این تحقیق توانست بیان سوروایوین را در نمونه های پارافینه شده از سرطان مثانه، در سطح mRNA و پروتئین، آشکار کرده و نشان دهد که افزایش بیان سوروایوین با افزایش بدخیمی تومورهای مثانه همراه است که این یافته می تواند در پیش آگهی مبتلایان به این تومورها مؤثر باشد و سوروایوین را به عنوان نشانگر پیش آگهی دهنده مناسب برای سرطان مثانه معرفی می کند.

    هدف
    ناحیه 22q11.2یک منطقه مستعد برای وقوع انواع بازآرایی ها است؛ به ویژه حذف هایی که منجر به وقوع انواع مختلفی از اختلالات ژنومی از جمله دی جرج و ولوکاردیوفاسیال می شوند. شاخص ترین ویژگی بیماران دارای این ریزحذف، نقایص قلبی مادرزادی از نوع کونوترانکال است. روش استاندارد برای تشخیص ریزحذف، روش (FISH) است. با این حال این روش دارای اشکالاتی از جمله گران بودن قیمت تمام شده وحساسیت کمتر این روش در شناسایی ریزحذف های غیر شایع و کوچکتر از معمول است.
    مواد و روش ها
    گروه مورد مطالعه در این تحقیق شامل 10 فرد با علائم مشخصه نشانگان دی جرج و دارای ناهنجاری مادرزادی قلبی از نوع کونوترانکال بودند که با استفاده از روش FISH و کاوشگر TUPLE1 بررسی شده بوده اند. علاوه براین سه فرد نرمال به عنوان کنترل منفی ریزحذف فوق در این تحقیق داخل شده اند. در این مطالعه با استفاده از نشانگرهای STSداخل و خارج منطقه مورد مطالعه، واکنش PCR نیمه کمی (SQMPCR) طراحی شد. نتایج با استفاده از نرم افزار TotalLab تجزیه و تحلیل شد.
    نتایج
    4 بیمار دوز ژنی کمتر از 60 درصد نشان دادند که تایید کننده وجود ریزحذف است. روش FISH تنها نشان دهنده یک بیمار مبتلا به ریزحذف بوده است.
    نتیجه گیری
    روش SQMPCR راه اندازی شده در این تحقیق توانست در بیماران مبتلا به ناراحتی مادرزادی قلبی نوع کونوترانکال وجود ریزحذف را در 40 درصد بیماران مشخص کند. به عبارت دیگر در 3 بیمار مبتلا و بدون مشکل شناخته شده ژنتیکی با روش FISH، حذف تشخیص داده شده است. بررسی های قبلی هم نشان دهنده حساسیت بیشتر روش های مولکولی در تشخیص ریزحذف های ناحیه مورد نظر بوده است. این تحقیق نشان دهنده وجود ریزحذف های غیر شایع در جامعه مبتلایان ایرانی و توانایی روش فوق در شناسایی بعضی از موارد منفی با روش FISH است. اگرچه آزمایش ها و بررسی های بیشتری برای تعیین حساسیت این روش لازم است اما به نظر می رسد که این روش می تواند در تشخیص موارد ریزحذف بدون علت شناخته شده ژنتیکی در بررسی FISH ولی دارای فنوتیپ کامل نشانگان 22q11DS استفاده شود.

    Objective
    The ITPA gene is responsible to remove free deaminated purine nucleotides of ITP, dITP and XTP from nucleotide pool of the cells. It seems that dysfunction in its activity, not only can increas the base substitution mutations frequency but also can works as a contrived factor to creating instability in genetic materials of the cells. There are several reports about the existence of structural and numerical genetic instability in the K562 cell genome. In this research, we examined the expression of ITPA gene as a possible contrived factor in observed genetic instability of this cell line.
    Materials And Methods
    To evaluate the expression of target gene semi-quantitative RT-PCR technique was used. Then to examine the functionality of gene products, its cDNAs were cloned and their sequences were determined. Their proteins products were predicted using available bioinformatics soft wares and the results were compared.
    Results
    The result of structural prediction of second mRNA showed that it has ability to encode a protein which has inability in substrate binding and also in its normal enzymatic activity. With regard to the fact that enzymatic activity of protein is dependent on the dimer formation, the function of hetero-dimer enzyme is changed. Therefore the catalytic activity of ITPase is predicted to be abnormal and it can be considered as a contrived factor for creating genetic instability in K562 cell line.
    Conclusion
    The study of gene expression showed that ITPA gene is expressed in moderate level compared to GAPDH expression as an internal control in K562 cell. Two types of transcripts were detected in this line. One of them was the normal product of splicing process of primary hnRNA, but the second one contained a 51 nucleotides deletion in the mRNA coding region. It seems, this transcript is the product of a rare splicing process in this line.

    Objective
    Recent studies suggest that intermittent and prolonged normobaric hyperoxia (HO) results in ischemic tolerance to preventing ischemia brain injury. In this research attempts were made to see the changes in excitatory amio-acid transporter 3 (EAAT3), TNF-α levels, and NF-κB activity following prolonged and intermittent NBHO preconditioning.
    Materials And Methods
    Rats were divided into four experimental groups, each with 21 animals. The first two groups were exposed to 95% inspired HO for 4h/day for 6 consecutive days (intermittent HO; InHO) or for 24 continuous hours (prolonged HO; PrHO). The second two groups acted as controls, and were exposed to 21% oxygen in the same chamber (normobaric normoxia, RA; room air) continuously for six days (intermittent RA, InRA) or for 24 hours (prolonged RA; PrRA). Each main group was subdivided to MCAO-operated (middle cerebral artery occlusion), sham-operated (without MCAO), and intact (without any surgery) subgroups. After 24h, MCAO-operated subgroups were subjected to 60min of right MCAO. After 24h reperfusion, neurologic deficit score (NDS) were assessed in MCAO-operated subgroups. Immediately and 48h after pretreatment, blood sampling for assessment of serum TNF- levels were purformed. Then, the effect of InHO and PrHO on serum TNF- levels, NF-κB activity and EAAT3 expression were measured.
    Results
    Preconditioning with InHO and PrHO decreased NDS and upregulate EAAT3 and increase serum TNF-α level and NF-κB activity significantly.
    Conclusion
    Although further studies are needed to clarify the mechanisms of ischemic tolerance, InHO and PrHO seems to partly exert their effects via increase in serum TNF-α levels, NF-κB activity and upregulation of glutamate transporters.

    Objective
    Dendritic cells have a critical role in control and regulation of immune responses. It is believed that these cells can be used for the treatment of many diseases. One of the methods used in immunotherapy is based on generating of tolerogenic dendritic cells through inhibition of expression costimulatory molecules. CD40 is one of the costimulatory molecules, and inhibition of expression by antisense or siRNA techniques, can generate tolerogenic dendritic cells. Generation of tolerogenic dendritic cells will be useful in the treatment of many diseases. By developing a quantitive RT-PCR for evaluation of gene expression, generation of these cells could be possible. Using proper software we designed an Antisense and transfection of dendritic cells by lipofectamine 2000 (Invitrogen) could lead us to generate tolerogenic dendritic cells.
    Materials And Methods
    In this study dendritic cells were extracted from of Balb/c mice Spleen and the purity of this extraction was determined by flow cytometry. BCL1 cell line as a CD40 expressing control group and Wehi-164 cell line were cultured in RPMI-1640+10%FCS. Primer design for CD40 gene and house keeping gene (GADPH) was done by bioinformatic soft wares such as Beacon designer, mfold and Blast. RNasy plus mini kit (Qiagen) was used for RNA extraction and the Purity and integrity were determined by O.D at 260/280 and agarose gel electrophoresis. In the next step cDNA synthesized and quantitative RT-PCR for CD40 using IQ sybergreen (Biorad) were setup. Finally, standard curve for CD40 and internal control in different RNA concentrations were performed. After transfection with lipofectamin 2000 the amount of gene suppression were quantified by qualitative RT-PCR.
    Results

    Objective
    Hepatitis C virus is the major cause of viral hepatitis and its diagnosis in suspected specimens is of great importance. The risk of transfusion- transmitted virus infection is primarily the result of failure in serological screening tests to detect recently infected donors in the pre-seroconversion window period of infection. Therefore, sensitive and accurate diagnosis of HCV prior to antibody production to reduce window period is necessary.
    Materials And Methods
    In the present study, a sensitive and specific RT-Nested PCR method for detection of a conserved HCV 5'UTR sequence was developed. Two pairs of primers for amplification of the target sequence in two rounds of PCR were selected. The developed RT-Nested PCR assay was performed on HCV-antibody confirmed positive samples as well as negative controls and standard samples. In order to compare the results, One Step RT-PCR kit was used in this study.
    Results
    25 HCV-positive plasma samples whose positivity were confirmed by ELISA and Western Blot tests, also as well as 10 fold dilutions of a high viral load plasma sample obtained from a HCV-positive patient as standard samples and 25 negative control plasmas from healthy blood donors were collected and tested by this assay. In all of positive samples a 175bp band was observed on agarose gel electrophoresis, but no band could be detected in negative control plasma. Results from developed RT-PCR assay and One Step RT-PCR kit showed a good correlation.
    Conclusion
    According to the results of this study, the developed RT-Nested PCR assay has a good sensitivity and specificity for diagnosis of HCV infection. It has the advantage of viral genome detection prior to seroconversion and can be used to detect HCV infection during window period

    Objective
    In this study, a SYBR Green real-time RT-PCR assay for quantification of HIV-1 viral RNA was developed.
    Materials And Methods
    This assay was performed based on amplification of the pol region of HIV-1 and product analysis by an ABI 7500 system. We quantified HIV-1 viral load in 26 seropositive patients by this system and the data were subsequently compared with results obtained with a reference technique represented by COBAS AMPLICOR HIV-1 Monitor test.
    Results
    The results demonstrated that this technique could detecte up to 500 HIV-1 RNA copies/ml of plasma. The linearity of this approach was conserved over a wide range of HIV-1 copy numbers (5×102-5×109). Since no positive signal was observed in seronegative volunteers, the specificity of the test was calculated as 100%. Comparison of the results with those obtained by the reference quantification method, revealed a significant correlation between the results (R2= 0.95).
    Conclusion
    On the basis of the most recent recorded cases for HIV-1 infection and AIDS in Iran, the prevalence of this disease is rising rapidly and the situation has been called to be alarming by national health representatives. Determination of HIV-1 viral load in plasma has been considered as the most effective single prediction tool of clinical outcome. Indeed, the development and stabilization of HIV-1 RNA assays have given physicians a unique tool for monitoring HIV-1 patients treated with antiviral drugs. In this study, we have developed a SYBR-Green Real Time RT-PCR assay for quantitative analysis of HIV-1 in infected patients.     Objective
    Because of the necessity of more effective treatments for the nervous system injuries and considering the role of survivin in cellular proliferation and apoptotic cell death, we have monitored survivin gene expression changes during the course of regeneration in injured sciatic nerves and also L4-L6 segments of spinal cord.
    Materials And Methods
    We used adult male NMRI mice as a model. After anesthetizing the animals, the right sciatic nerve was transected and at the indicated times (3, 6, 12, 24, 48, 96 and 144 hours) the animals were sacrificed and both distal and proximal segments of the transected sciatic nerve, intact left sciatic nerve and L4-L6 segments of spinal cord were dissected. The total RNA was extracted from each sample and semi-quantitative RT-PCR with specific primers for survivin and also 2-microglobulin genes, as an internal control, was performed. To determine cellular distribution of survivin protein, 6 days (144 hours) after the axotomy, survivin protein expression was evaluated using immunohistochemistry technique.
    Results
    Our results demonstrated the expression of both survivin140 and survivin40 in distal and proximal segments of sciatic nerve with different intensity, where the expression of survivin140 was higher than survivin40. In spinal cord segments, only survivin140 expression was detected. In Immunohistochemistry analysis of spinal cord segments, both the nuclear and cytoplasmic distribution of survivin protein was observed. In contrast, survivin protein has not been detected in either distal or proximal segments of sciatic nerve.
    Conclusion
    Our data suggest that survivin is differentially expressed and spliced during the course of regeneration in damaged nerve and spinal cord. It seems that manipulation of expression and/or splicing of survivin could potentially affect the process of regeneration in nerve and/or spinal cord injuries.

    Objective
    Inhibition of apoptosis may favor the onset and progression of cancer. Survivin is an inhibitor of apoptosis that has been considered as a potential marker for diagnostic and/or prognostic of bladder cancer. The survivin protein regulates both cell division and cell death and is overexpressed in the vast majority of human cancers. In this study, the expression pattern and potential prognostic value of survivin was assessed in Formalin-Fixed Paraffin-Embedded (FFPE) samples of bladder tumor.
    Materials And Methods
    FFPE samples, from patients with a well-known five-year survival record, were assessed by semi-quantitative RT-PCR technique. 51 samples from 30 patients were analyzed on the basis of Survivin expression. Tissue distribution and subcellular localization of survivin protein in tumor tissues was also examined by immunohistochemistry (IHC).
    Results
    The expression of survivin was detected in 66.6% of the samples, with an increase of expression in higher grades of tumor. Furthermore, survivin was overexpressed in 2nd and 3rd recurrences of the same patients. Also, with the increased malignancy and accordingly increased expression of surviving, the overall 5-year survival rate of patients was significantly declined (P=0.036). IHC results also localized a nuclear localization for Survivin protein in tumor tissues.
    Conclusion
    In conclusion, we were able to detect the expression of survivin in FFPE samples of bladder tissues, at the level of mRNA and protein and find a correlation between the level of Survivin expression and the degree of malignancy of the tumors. Our findings introduce Survivin as a suitable prognostic marker for predicting the bladder tumors.

    Objective
    22q11.2 chromosomal region is a hot spot for many cytogenetic rearrangements especially microdeletions which are responsible for DiGeorge and VeloCardioFacial syndromes. The most characteristic sign in these patients is congenital cardiac conotruncal anomalies. The gold standard diagnostic test for these microdeletions is FISH (Fluorescent In Situ Hybridization). However this diagnostic technique has some drawbacks such as high final cost and low sensitivity in smaller and uncommon microdeletions found in this region. The aim of this study was to introduce a less expensive and a priori more sensitive molecular method to help small and peripheral laboratories to find genetic causes of congenital heart diseases and DiGeorge syndrome.
    Materials And Methods
    10 patients with congenital conotruncal anomalies and symptoms of DiGeorge syndrome were included in this study. These patients had been analyzed by FISH probe TUPLE1 before the inclusion. 3 normal persons were included as normal controls for microdeletion region. Semi Quantitative Multiplex PCRs were designed based on known markers in and out of the region of intrest. Results were analyzed by TotalLab software.
    Results
    4 patients showed a decrease in gene dosage more than 60% compared to normal persons. FISH analysis found only one patient with microdeletion.
    Conclusion
    The designed method based on semi quantitative PCR was able to find 4 patients (40%) with microdeletion in a population of 10 patients with congenital cardiac anomalies. This technique was also able to find microdeletions in three FISH negative patients. Molecular diagnosis of microdeletions is supposed to be more sensitive than FISH in small microdeletions. This study confirms the presence of atypical deletions in Iranian patients and shows that the applied technique can detect some FISH negative patients. However further studies are needed to determine the sensitivity and specificity of the mentioned molecular diagnosis. It seems that this can be used at least for the patients with typical phenotypic features of 22q11DS and negative FISH results.
