صفحات 113-122سابقهطناب نخاعی ظرفیت محدودی برای ترمیم دارد،بدین جهت مداخله پزشکی برای درمان ضایعات نخاعی ضروری می باشد. پیوند سلول های شوان برای درمان ضایعات نخاعی نتایج امیدوارکننده ای داشته است.اما جداسازی واستفاده از سلول های شوان بخاطر ایجاد ضایعه برای دهنده سلول ونیز ظرفیت پایین در تزاید با محدودیت روبرو است.از طرف دیگر،استفاده از منابع سهل الوصولی مانند سلول های بنیادی مغز استخوان توجه های زیادی رو به خود جلب کرده است.بنابراین،در این مطالعه اثر سلول های شوان تمایز یافته از سلول های بنیادی مغز استخوان بر بهبود حرکتی موش های صحرایی پس از ضایعه نخاعی مورد بررسی قرار گرفت.روش هاابتدا در محیط آزمایشگاهی سلول های مزانشیمی مغز استخوان موش های صحرایی کشت و به سلول های شوان تمایز داده شدند.تمایز سلول ها با روش های RT-PCRوایمنوسیتوشیمی تایید گردید.سپس،سلولهای شوان درون داربست های کلاژنی قرار گرفتند ودر ضایعه 3 میلیمتری(lateral hemisection) طناب نخاعی قرار داده شدند.پیشرفت های حسی وحرکتی توسط آزمون های open filed locomotor scale، narrow beamو tail flick برای 8 هفته مورد ارزیابی قرار گرفتند.در انتها، طناب های نخاعی با روش های بافت شناسی و ایمنوهیستوشیمی بررسی شدند.نتایجمشاهدات آزمایشگاهی نشان داد که سلول های تمایز یافته دارای ظاهر و نشان های سلول های شوان می باشند. این مطالعه دارای چهار گروه بود(هر گروه 10موش صحرایی):لامینکتومی، شاهد،داربست و داربست+سلول.نتایج آزمون های حسی و حرکتی گروه دریافت کننده سلول بطور مشخص از گروه های شاهد و داربست بدون سلول بهتر بود.در ارزیابی بافت شناسی، رشد مجدد رشته های عصبی و میلینه شدن در این گروه نیز بهتر از گروه های دیگر بود.نتیجه گیریاین مطالعه نشان داد که سلول های شوان تمایز یافته از مغز استخوان می توانند بعنوان منبعی جهت سلول های شوان برای ترمیم ضایعه نخاعی مورد توجه قرار گیرند.

صفحات 123-128مقدمهمیتوکندری منبع مهم تولید آدنوزین تری فسفات (ATP) در تکوین جنین مراحل قبل از لانه گزینی است. بنابراین هدف از این مطالعه بررسی تاثیر انجماد شیشه ای و تکوین در محیط کشت بر روی پراکندگی میتوکندری و میزان ATP جنین های موش بود.مواد و روش هاجنین ها در مراحل 2-PN، 4 سلولی و بلاستوسیست از لوله فالوپ و شاخ رحمی موشهای حامله تحریک شده جمع آوری شدند، سپس به روش کرایوتاپ با بکارگیری اتیلن گلیکول و دی متیل سولفوکساید منجمد شدند. پس از تعیین درصد حیات جنینهای منجمد شده، تکوین آنها تا مرحله خروج از زونا مورد بررسی قرار گرفت. میزان ATP جنین های حاصل از شرایط in vivo و in vitro در مراحل مختلف تکوینی با استفاده از واکنش لوسیفرین- لوسیفراز اندازه گیری شد. توزیع میتوکندری با استفاده از رنگ آمیزی توسط Mitotracker green و با میکروسکوپ فلوروسنت مورد بررسی قرار گرفت.نتایجمیزان زنده ماندن جنینهای انجمادی در مراحل 2-PN، 4 سلولی و early blastocyst به ترتیب 63/83%، 52/87% و 27/89% بود. میزان خروج از زونا در جنین ها در همان مراحل تکوینی در گروه انجمادی به ترتیب 44/57%، 73/66 و 89/70% و در گروه غیرانجمادی به ترتیب 32/66%، 25/73 و 89/75 بود (P>0.05). بین جنینهای مراحل مختلف تکوینی در گروه کنترل و انجمادی و نیز جنینهای حاصل از شرایط in vivo و in vitro از نظر میزان ATP تفاوت معنی داری وجود نداشت (P>0.05). توزیع میتوکندری در جنینهای 2-PN در گروه های انجمادی و غیر انجمادی مشابه بود ولی تجمعات بزرگ میتوکندری در جنین های 4 سلولی در گروه انجمادی بارز بود.نتیجهروش انجماد شیشه ای بر روی میزان ATP جنین های موش تاثیر نداشت، همچنین در این روش استرسی به سلول وارد نشد و سلامت روش انجماد شیشه ای نشان داده شد.

صفحات 129-133زمینهعفونت توسط سودوموناس آیروژینوزا درتمام جهان گسترش یافته است و گزارشات مکرر انتشار متالوبتالاکتامازها نشان از سرعت رو به رشد آن دارد. هدف از این مطالعه بررسی حساسیت آنتی بیوتیکی و تشخیص ژن های متالوبتالاکتاماز (MBLs) از نوع IMP و VIM در سودوموناس آئروژینوزای ایزوله شده از استان زنجان می باشد.روش بررسیتعداد 70 ایزوله ی سودوموناس آیروژینوزا از بیماران بستری در بخش مراقبت های ویژه، پس از انجام آزمایشات میکروبی و بیوشیمیائی تعیین هویت شدند. الگوی مقاومت آنتی بیوتیکی نمونه های تائید شده، به روش کایربی بوئر تعیین گردید. سپس سویه های تولید کننده ی متالوبتالاکتاماز به روش فتوتیپی (DDST) شناسائی شدند. بعد از استخراج DNA وجود ژن های متالوبتالاکتاماز از نوع VIM و IMP و ژن حمل کننده ی اینتگرون کلاس یک در سویه های فوق باروش PCR بررسی گردیدند.یافته هابیشترین مقاومت آنتی بیوتیکی در ایزوله ها به ترتیب: مروپنم، سفتازیدیم و ایمی پنم بود. در آزمون DDST مشخص گردید که 85/62 درصد ایزوله ها مقاوم به ایمی پنم هستند. 93% از ایزوله ها MIC بیش از 4 میکروگرم برای ایمی پنم نشان دادند. بر اساس نتایج DDST 8/87% ایزوله ها تولید کننده MBL بودند. با انجام آزمون PCR 33 سویه ی تولید کننده ی MBLS فتوتیپ مثبت، تایید شدند که 23 ایزوله حاوی ژن VIM(56%) و10ایزوله حاوی ژن IMP(3/24%) بودند.همچنین مشخص گردیدکه از 44 سویه ی مقاوم به ایمی پنم تنها 31 ایزوله (5/70%) دارای اینتگرون کلاس یک بودند.نتیجه گیرینتایج حاکی از آن است که ژن های متالوبتالاکتاماز از نوع VIM و IMP و ژن اینتگرون کلاس یک در بین ایزوله های سودوموناس آیروژینوزا در منطقه زنجان به صورت غالب وجود داشته و همچنین فراوانی ژن های VIM بیش از IMP می باشد. در این مطالعه برای اولین بار ژن های IMP با فراوانی بیشتری در میان سویه های تولیدکننده MBLS گزارش شد.

صفحات 134-139زمینهاحتمال زیادی وجود دارد که ریتالین سو مصرف شود. این دارو انتخاب اصلی جهت کنترل اختلال عدم توجه همراه با بیش فعالی است. دانشجویان کالج جهت بهبود حافظه، کاهش نیاز به خواب (خصوصا در زمان امتحانات) به این دارو روی می آورند، که حداقل تا حدودی اثرات آن می تواند تحت تاثیر سیستم سروتونرژیک باشد. بنابراین، بررسی نقش مصرف ریتالین بر روی مغز بالغ، ارزشمند به نظر می رسد. بررسی های زیادی در مورد اثر ریتالین بر مغز درحال رشد انجام شده است، ولی تعداد بسیار کمی مطالعه بر روی مغز بالغ صورت گرفته است. این مطالعه جهت یافتن اثر ریتالین بر تراکم ترانسپورترریتالین در ناحیه مدیال کورتکس فرونتال در موش بالغ نر انجام شد.روشدر این مطالعه از 30 سر موش ویستار نر استفاده گردید. موش ها به 5 گروه تقسیم شدند: یک گروه کنترل، 2 گروه دریافت کننده ریتالین و دو گروهvehicle. 12 موش به مدت 11 روز متوالی از طریق دهان ریتالین دریافت کردند (20 میلی گرم / کیلوگرم/ 2 بار در روز). سپس یکهفته زمان ترک از ریتالین در نظر گرفته شد. در یک گروه جهت بررسی اثرات کوتاه مدت ریتالین، بعد از دو هفته شش موش قربانی و بررسی شدند. برای بررسی اثرات بلند مدت مصرف ریتالین، گروه بعدی دریافت کننده ریتالین، 12 هفته بعد قربانی شدند. بنابراین این گروه 12 هفته بعد از گروه اول قربانی شدند. جهت بررسی نتایج از روش ایمونوهیستوشیمی استفاده شد.
یافته هامطالعات ایمونوهیستو شیمی تراکم بالاتری از SERT، 2 و 12 هفته بعد از ترک ریتالین را در کورتکس فرونتال میانی نشان داد که البته اختلاف معنی داری بین این دو گروه وجود نداشت.نتایجیافته های ما نشان دهنده اثرات کوتاه مدت و بلند مدت ریتالین بر سیستم سروتونرژیک ناحیه مدیال کورتکس فرونتال مغز بدنبال ترک این ماده بود.

تا کنون، سلولهای استرومایی مغز استخوان به خوبی در بهبود مشکلات حرکتی در موشهای صحرایی مبتلا به ضایعات نخاعی مورد استفاده قرار گرفته اند. یکی از مشکلات عمده در پیوند سلول میزان بالای مرگ سلولها در in vivo است که عمدتا نیز پس از تمایز عصبی در این سلولها اتفاق می افتد. مطالعات قبلی ما نشان داده است که گیرنده p75، با نام گیرنده مرگ، تنها در فاصله زمانی 12-6 ساعت پس از القای تمایز عصبی در سلولهای استرومایی مغز استخوان بیان می شود. همچنین در بررسی جدیدتری ما کاهش در میزان آپوپتوز سلولهای ترانسفکت شده با siRNA p75 را درin vitro نشان داده ایم. از این رو، هدف از تحقیق حاضر بررسی اثرات پیوند سلولهای استرومایی مغز استخوان بیان کننده p75 siRNA در بهبود عملکردی موشهای صحرایی مبتلا به ضایعه نخاعی می باشد. بدین منظور، لامینکتومی در مهره L1 انجام شد تا نخاع برای ایجاد مدل کانتیوژن با استفاده از روش سقوط وزنه در معرض قرار گیرد. موشهای صحرایی ضایعه دیده تیمار شده با PBS (گروه اول) به عنوان کنترل منفی در نظر گرفته شدند که در آنها حفرات در نخاع 10 هفته پس از ایجاد ضایعه به خوبی قابل مشاهده بود. سلولهای استرومایی مغز استخوان ترانسفکت شده با pRNA-U6.1/Hygro (گروه دوم) و pRNA-U6.1/Hygro-p75 shRNA (گروه سوم) با رنگ فلورسانت CM-DiI رنگ آمیزی شده و حدود 7 روز پس از جراحی به داخل محل ضایعه پیوند زده شدند. مقیاس اندازه گیری حرکتی Basso–Beattie–Bresnehan به صورت هفتگی به مدت 10 هفته انجام شد. تفاوت معناداری بین تمام گروه های مورد بررسی از نظر بهبود عملکردی به ویژه بین گروه دوم و سوم وجود داشت (P ≤ 0.05). همچنین میزان بقای سلولهای گروه سوم نیز در in vivo نسبت به گروه دوم بالاتر بود. این نتایج نشان دادند که سلولهای استرومایی مغز استخوان که p75 siRNA را بیان می کنند، کاندید مناسبتری برای درمان ضایعات نخاعی هستند.

صفحات 146-151مقدمهسلولهای تشکیل دهنده ی کلونی اندوتلیال (CFU-EC) در ابتدا به عنوان سلولهای پیش ساز اندوتلیوم معرفی شدند. مطالعات بعدی نشان داد که آنها مونوسیتهایی هستند که کنترل کننده رگ زایی می باشند. نشان داده شده است که CFU-EC باعث بهبود جریان خون و تراکم مویرگها در مدلهای حیوانی ایسکمی قلبی و اندامی می شود. نارسایی اصلی این سلولها برای مقاصد بالینی، فراوانی پایین آنها و توان تکثیر پایینشان است. هدف از این مطالعه بررسیفعایت و واریانهای آلترناتیو تلومراز در این سلولها به عنوان دلیلی برای توان تکثیر پایینشان می باشد.
شیوه ها: CFU-EC با استفاده از پروتکلهای استاندارد قبلی از خون محیطی جدا شدند. کلونی ها به شیوه مکانیکی جدا شدند و واریان های آلترناتیو تلومراز با استفاده از real time RT-PCR بررسی شدند. فعالیت آنزیم تلومراز با استفاده از پروتکل تکثیر توالی های تکراری تلومراز اندازه گیری شد. همین موارد در لاین سرطانی Calu6 به عنوان کنترل مثبت اندازه گیری شد.نتایجسلول های تک هسته ای کشت داده شده، کلونی های تیپیکی تشکیل دادند که مونوسیتهای دوکی شکل از مرکز آنها به سمت محیط کشیده شده بودند. این خصوصیت ها نشانگر جداسازی صحیح CFU-EC بوده است. پروتکل تکثیر توالی های تکراری تلومراز و real time RT-PCR هیچ فعایت و واریان آلترناتیوی از تلومراز را در CFU-EC نشان ندادند، در حالی که هر دو این موارد در سطح بالایی در لاین سرطانی Calu6 مشاهده می شد.نتیجه گیریفقدان فعالیت آنزیم تلومراز در CFU-EC نتیجه یک کنترل بیان ژن در سطح قبل از رونویسی است و حاصل مواردی نظیر تغییرات بعد از رونویسی نمی باشد. این یافته می تواند توان تکثیر پایین CFU-EC را تا حدی توضیح دهد. ما احتمال می دهیم که این توان تکثیر پایین به علت ماهیت بالغ مونوسیتی سلولهای تشکیل دهنده CFU-EC باشد که نیاز به مطالعات بیشتر داردکلیدواژگان: تلومراز، سلول آندوتلیال، همانژیوبلاست، آلترناتیو اسپلایسنگ -
صفحات 152-157سابقهبه خوبی مشخص گردیده که بوجود آمدن استرس اکسیداتیو در بافت مغز یکی از عوارض پرفشاری شریانی بوده که منجر به آسیب مغز می شود. بر این اساس مطالعه حاضر در نظر داشت تا اثرات درمانی داروی آتورواستاتین (mg/kg/day20)، به عنوان یک آنتی اکسیدان، جهت جلوگیری از آسیب ناشی از استرس اکسیداتیو در پرفشاری شریانی را مورد بررسی قرار دهد.روش هاآزمایش در چهار گروه از موشهای صحرایی انجام گردید (هر گروه 5 = n): گروه شاهد، شاهد درمان شده، پرفشار و پرفشار درمان شده. جهت القاء پرفشاری از روش تنگی آئورت در بالای شریانهای کلیه استفاده گردید. بعد از 30 روز جهت برداشتن بافت مغز حیوانات تحت بیهوشی عمیق کشته شدند. پس از هموژن کردن بافتها، فعالیت آنزیمهای سوپراکسید دیسموتاز(SOD)، کاتالاز(CAT)، غلظت گلوتاتیون(GSH) و مالون دی آلدئید(MDA) بافت مغز توسط روش های بیوشیمیایی تعیین گردید.
یافته هادر گروه های پرفشار شریانی میانگین فشار خون به میزان 40 درصد افزایش داشت و فعالیت آنزیمهای مغزی SOD و CAT در مقایسه با گروه شاهد به طور معنی داری کاهش پیدا کرد. همچنین پرفشاری شریانی سبب کاهش معنی دار سطح گلوتاتیون و افزایش سطح MDA بافت مغز گردید. درمان با آتورواستاتین توانست فعالیت آنزیم SOD را به طور معنی دار افزایش دهد و از کاهش معنی دار فعالیت آنزیم GSH جلوگیری نماید.نتیجه گیریبر اساس یافته های این مطالعه درمان با آتورواستاتین ممکن است از آسیب استرس اکسیداتیو ناشی از پرفشاری شریانی در بافت مغز موش صحرایی جلوگیری نماید.

صفحات 158-164سابقهفاکتور محرکه کلونی گرانولوسیتی انسان بعنوان یک محصول دارویی ارزشمند در درمان سرطان خون شناخته شده است. در چند سال گذشته گیاهان ترانسپلاستوم برای تولید انواع داروهای نوترکیب و واکسن ها مورد استفاده قرار گرفته اند و در مقایسه با گیاهان تراریخت بدلیل تولید بالای پروتئین نوترکیب و مسائل ایمنی زیستی برتری هایی را نشان می دهند.روش هاژن hG-CSF بر روی وکتور pCL تحت پروموتر prnn 16S و خاتمه دهنده TspA کلون گردید. وکتور حاوی ژن مورد نظر با نانو ذرات طلا پوشش داده شد و با استفاده از روش تفنگ ژنی به ژنوم کلروپلاست منتقل گردید. باززایی نمونه های بمباران شده در محیط موراشینگ و اسکوگ حاوی هورمون 6-benzylaminopurine and 1-naphthaleneacetic acid انجام گرفت.نتایجبرای تایید صحت وکتور بیانی ابتدا وکتور حاوی ژن نشانگر بتاگلوکرونیداز در کلروپلاست بیان گردید و سپس ژن hG-CSF با همان پروتوکل انتقال یافت. درج هدفمند ژن مورد نظر در ژنوم کلروپلاست لاین های تراریخت با پرایمر های اختصاصی که از روی ژنوم طراحی شده بود مورد تایید قرار گرفت. برای بررسی بیان ژن در سطح پروتئین آزمایش وسترن بلات انجام شد که بیان پروتئین نوترکیب 20 کیلودالتونی را در گیاهان تراریخت و عدم بیان آن در گیاهان شاهد مورد تایید قرار داد.نتیجه گیریاین مطالعه برای اولین بار بیان فاکتور محرکه کلنی گرانولوسیتی انسان را در کلروپلاست گیاه مورد بررسی قرار داده است. امید است که پلت فرم مناسبی برای تحقیقات دارویی و بیان پروتئین های نوترکیب باشد.

Pages 113-122BackgroundSpinal cord has a limited capacity to repair; therefore، medical interventions are necessary for treatment of injuries. Transplantation of Schwann cells has shown a great promising result for spinal cord injury (SCI). However، harvesting Schwann cell has been limited due to donor morbidity and limited expansion capacity. Furthermore، accessible sources such as bone marrow stem cells have drawn attentions to themselves. Therefore، this study was designed to evaluate the effect of bone marrow-derived Schwann cell on functional recovery in adult rats after injury.MethodsMesenchymal stem cells were cultured from adult rats’ bone marrow and induced into Schwann cells in vitro. Differentiation was confirmed by immunocytochemistry and RT-PCR. Next، Schwann cells were seeded into collagen scaffolds and engrafted in 3 mm lateral hemisection defects. For 8 weeks، motor and sensory improvements were assessed by open field locomotor scale، narrow beam، and tail flick tests. Afterwards، lesioned spinal cord was evaluated by conventional histology and immunohistochemistry.ResultsIn vitro observations showed that differentiated cells had Schwann cell morphology and markers. In this study، we had four groups (n = 10 each): laminectomy، control، scaffold and scaffold + Schwann cells. Locomotor and sensory scores of cell grafted group were significantly better than control and scaffold groups. In histology، axonal regeneration and remyelination were better than control and scaffold groups.ConclusionThis study demonstrates that bone marrow-derived Schwann cells can be considered as a cell source for Schwann cells in SCI treatment.Keywords: Rats, Spinal cord injuries (SCI), Bone marrow, Schwann cells, Cell transdifferentiation

Pages 123-128BackgroundThe mitochondria are an important source of adenosine triphosphate (ATP) production in pre-implantation embryo. Therefore، the objective of this study was to investigate the effect of vitrification and in vitro culture of mouse embryos on their mitochondrial distribution and ATP content.MethodsThe embryos at 2-PN، 4-cell and blastocyst stages were collected from the oviduct of stimulated pregnant mice and uterine horns. Then، the embryos were vitrified with the cryotop method using ethylene glycol and dimethylsulphoxide. After evaluating the survival rates of vitrified embryos، their development to hatching stages were assessed. The ATP content of collected in vivo and in vitro embryos at different stages was measured by luciferin-luciferase bioluminescence assay. The distribution of mitochondria was studied using Mito-tracker green staining under a fluorescent microscope.ResultsThe survival rates of vitrified embryos at 2-PN، 4-cell and early blastocyst stages were 84. 3، 87. 87 and 89. 89%، respectively. The hatching rates in previous developmental stages in vitrified group were 57. 44، 66. 73 and 70. 89% and in non-vitrified group were 66. 32، 73. 25 and 75. 89%، respectively (P>0. 05) The ATP content of in vivo or in vitro collected embryos was not significantly different in both vitrified and non-vitrified groups (P>0. 05). Mitochondrial distribution of vitrified and non-vitrified 2-PN embryos was similar، but some clampings or large aggregation of mitochondria within the vitrified 4-cell embryos was prominent.ConclusionsVitrification method did not affect the mouse embryo ATP content. Pages 129-133BackgroundInfectious by Pseudomonas aeruginosa has spread worldwide and metallo-beta-lactamases (MBL) are being reported with increasing frequency. The aim of this study was to investigate the antibiotic susceptibility and distribution of blaVIM and blaIMP genes in P. aeruginosa isolates from Zanjan Province of Iran.MethodsA total of 70 P. aeruginosa isolates were identified from patients admitted at intensive care units. The antimicrobial susceptibility was tested by disk diffusion (Kirby-Bauer) method and for production of MBL using double-disk synergy test (DDST). After DNA extraction، the presence of blaVIM and blaIMP genes and class 1 integron were detected by PCR.ResultsMost of the isolates were resistant to meropenem، cefotaxime and imipenem (IPM). Also، 44/70 (62. 85%) IPM resistant isolates were confirmed by DDST. Of the 44 clinical isolates، 41 (93%) isolates showed MIC≥4 µg/ml for IPM. Based on the DDST results، 36 (87. 8%) were confirmed to be MBL producers. PCR amplification showed that 23/41 (56%) carried blaVIM and 10/41 (24. 3%) possessed blaIMP gene. Also، 31/44 (70. 5%) isolates contained class 1 integron gene.ConclusionOur results highlight that the genes for Verona integron-encoded metallo-β-lactamase، IPM β-lactamases and class 1 integrons were predominantly present among the IPM-resistant P. aeruginosa tested in our province and also the frequency of blaVIM type is higher than blaIMP. This is the first report of P. aeruginosa strains producing blaIMP with high frequency from Zanjan province of Iran.Keywords: Pseudomonas aeruginosa, Beta, lactamases, PCR

Pages 134-139BackgroundRitalin has high tendency to be abused. It has been the main indication to control attention deficit hyperactivity disorder. The college students may seek for it to improve their memory، decrease the need for sleep (especially during exams)، which at least partially، can be related to serotonergic system. Therefore، it seems worthy to evaluate the effect of Ritalin intake on mature brain. There are many studies on Ritalin effect on developing brain، but only few studies on adults are available. This study was undertaken to find Ritalin effect on serotonin transporter (SERT) density in medial frontal cortex (MFC) of mature rat.MethodsThirty male Wistar rats were used in the study. Rats were assigned into five groups (n = 6 per group): one control، two Ritalin and two vehicle groups. Twelve rats received Ritalin (20 mg/kg/twice a day) orally for eleven continuous days. After one week of withdrawal and another two weeks of rest، in order to evaluate short-term effects of Ritalin، six rats were sacrificed. Another six rats were studied to detect the long-term effects of Ritalin; therefore، they were sacrificed 12 weeks after the previous group. The immunohistochemistry was performed to evaluate the results.ResultsImmunohistochemistry studies showed a higher density of SERT in both 2 and 12 weeks after withdrawal from Ritalin intake in MFC of rat and there was no significant difference between these two groups.ConclusionsOur findings demonstrated both short- and long-term effects of Ritalin on frontal serotonergic system after withdrawal period.Keywords: Ritalin, Serotonin, Rats

Bone marrow stromal cells (BMSC) have been successfully employed for movement deficit recovery in spinal cord injury (SCI) rat models. One of the unsettled problems in cell transplantation is the relative high proportion of cell death، specifically after neural differentiation. According to our previous studies، p75 receptor، known as the death receptor، is only expressed in BMSC in a time window of 6-12 hours following neural induction. Moreover، we have recently reported a decreased level of apoptosis in p75-suppressed BMSC in vitro. Therefore، our objective in this research was to explore the functional effects of transplanting p75: siRNA expressing BMSC in SCI rats.
MethodsLaminectomy was performed at L1 vertebra level to expose spinal cord for contusion using weight-drop method. PBS-treated SCI rats (group one) were used as negative controls، in which cavitations were observed 10 weeks after SCI. pRNA-U6. 1/Hygro- (group two، as a mock) and pRNA-U6. 1/Hygro-p75 shRNA- (group three) transfected BMSC were labeled with a fluorescent dye، CM-DiI، and grafted into the lesion site 7 days after surgery. The Basso-Beattie-Bresnehan locomotor rating scale was performed weekly for 10 weeks.
ResultsThere was a significant difference (P≤0. 05) between all groups of treated rats regarding functional recovery. Specifically، the discrepancy among p75 siRNA and mock-transfected BMSC was statistically significant. P75 siRNA BMSC also revealed a higher level of in vivo survival compared to the mock BMSC.
ConclusionOur data suggest that genetically modified BMSC that express p75: siRNA could be a more suitable source of cells for treatment of SCI.

Pages 146-151IntroductionEndothelial progenitor colony forming unit-endothelial cells (CFU-EC) were first believed to be the progenitors of endothelial cells، named endothelial progenitor cells. Further studies revealed that they are monocytes regulating vasculogenesis. The main hindrance of these cells for therapeutic purposes is their low frequency and limited replicative potentials. This study was undertaken to determine telomerase activity and alternative splicing variants in CFU-EC as a potential cause of limited replicative capacity in these cells.MethodsCFU-EC were isolated from peripheral blood using a standard cell culture assay. Colonies were detached mechanically and alternative splicing variant mRNA were evaluated using real-time PCR. Telomerase enzyme activity was assessed using telomerase repeat amplification protocol. The same procedures were done on the cancer cell line Calu6 as the positive control.ResultsThe cultured peripheral blood mononuclear cells formed colonies with spindle-shaped monocytic cells sprouted from the clusters. These morphological characteristics fulfill the definition of CFU-EC. Telomere length amplification protocol assay revealed no telomerase activity and real-time PCR showed no expression of telomerase enzyme mRNA in CFU-EC. Both parameters were significantly higher in the cancer cell line Calu6 taken as the positive control.ConclusionThe absence of telomerase activity in the CFU-EC is a result of pre-transcriptional regulation of gene expression rather than other mechanisms for controlling telomerase activity such as post-transcriptional modifications. This finding can explain the limited proliferative activity of CFU-EC cells. Pages 152-157BackgroundIt is well known that the development of brain oxidative stress is one of the most serious complications of arterial hypertension that evokes brain tissue damage. The aim of this study was to examine the effects of atorvastatin treatment (20 mg/kg/day)، as an antioxidant، to prevent the brain tissue oxidative stress in the hypertensive (HTN) rats.MethodsExperiments were performed in four groups of rats (n = 5 each group): sham، sham-treated، HTN and HTN treated. Rats were made HTN by aortic constriction above the renal arteries. After 30 days، rats were slaughtered under deep anesthesia to remove brain hemispheres. After tissue homogenization، enzyme activities of superoxide dismutase (SOD) and catalase (CAT)، as well as glutathione (GSH) content and malondialdehyde (MDA) level were determined by biochemical methods.ResultsIn HTN rats، arterial blood pressure was increased about 40% and brain enzyme activities of SOD and CAT were significantly decreased compared with sham group. Induction of hypertension significantly decreased GSH content and increased MDA level of brain tissue. Treatment with atorvastatin enhanced the activity of SOD and prevented from GSH decrement during hypertension.ConclusionBased on the findings of this study، treatment with atorvastatin might have saved the brain tissue of HTN rats from hypertension-induced oxidative stress.Keywords: Atorvastatin, Aortic coarctation, Oxidative stress, Hypertension

Transplastomic plants have been emerged as a new and high potential candidate for production of recombinant biopharmaceutical proteins in comparison with transgenic plants due to extremely high level expression، biosafety and many other advantages.MethodshG-CSF gene was cloned into pCL vector between prrn16S promoter and TpsbA terminator. The recombinant vector was coated on nanogold particles and transformed to lettuce chloroplasts through biolistic method. Callogenesis and regeneration of cotyledonary explants were obtained by Murashige and Skoog media containing 6-benzylaminopurine and 1-naphthaleneacetic acid hormones. The presence of hG-CSF gene in plastome was studied with four specific PCR primers and expression by Western immunoblotting.ResultshG-CSF gene cloning was confirmed by digestion and sequencing. Transplastomic lettuce lines were regenerated and subjected to molecular analysis. The presence of hG-CSF in plastome was confirmed by PCR using specific primers designed from the plastid genome. Western immunoblotting of extracted protein from transplastomic plants showed a 20-kDa band، which verified the expression of recombinant protein in lettuce chloroplasts.ConclusionsThis study is the first report that successfully express hG-CSF gene in lettuce chloroplast.
