    مقدمه
    حاملین سالم شاغل در بیمارستان از دلایل اصلی انتشار عفونت استافیلوکوکی در بیماران محسوب می شوند. این باکتری با ترشح سموم مختلف از جمله انتروتوکسین شرایط تهاجم به میزبان را فراهم می آورد. شناسائی ویژگی ها و تفاوت های استافیلوکوکوس اورئوس های جدا شده از کارکنان بیمارستان ها به عنوان حاملین سالم و انواع جدا شده از بیماران بر مبنای دارا بودن ژن انتروتوکسین Sea، Seb، Sec،s Sed) وSee) از اهداف این مطالعه می باشد.
    روش بررسی
    از 120 مورد از افراد بیمار و80 مورد از حاملین سالم شاغل در مراکز درمانی گرگان استافیلوکوکوس اورئوس جدا گردید. برای مطالعه انتخاب شد. جدایه ها از لحاظ حضور ژن انتروتوکسین تایپ-های A-E (sea-see) با استفاده از پرایمرهای اختصاصی بررسی شدند.
    یافته ها
    استافیلوکوکوس اورئوس های مورد آزمون در این مطالعه(5/87%) دارای حداقل یکی از ژن های انتروتوکسین sea-see بوده و فراوان ترین آن ژن انتروتوکسین sea،(124 مورد) بود. توزیع فراوانی این جدایه ها در نمونه های جدا شده از حاملین به طور معنی داری بیشتر از جدایه های بدست آمده از بیماران است.
    نتیجه گیری
    درصد بالایی از استافیلوکوکوس اورئوس های جدا شده از نمونه های بالینی دارای ژن های انتروتوکسین می باشند که اهمیت توجه به انسان را به عنوان منبع اصلی و حامل استافیلوکوکی به دلیل ارتباط معنی-داری که بین توکسین زا بودن جدایه ها و منبع جداسازی آنها از حاملین و بیماران وجود دارد مشخص می کند.

    زمینه و هدف
    تشابه بسیار میان ژنوم باکتری های مایکوباکتریوم توبرکولوزیس کمپلکس، تشخیص افتراقی آنها با روش های بیوشیمیایی مرسوم آزمایشگاهی را مشکل می نماید. این تحقیق به منظور شناسایی و تعیین هویت جدایه های مایکوباکتریوم توبرکولوزیس کمپلکس عامل بیماری در میان مسلولین استان گلستان صورت پذیرفته است.
    روش بررسی
    جدایه های جمع آوری شده از نمونه های کلینیکی 42 مسلول استان گلستان در محدوده زمانی 1387 تا 1389 به موسسه رازی منتقل و بر روی محیط های کشت لونشتاین- جانسون گلسیرینه و پیرووات دار، تجدید کشت گردیدند. ژنوم همه جدایه ها به روش جوشانیدن استخراج و هویت جدایه ها توسط یک برنامهPCR سه مرحله ای به نام روش Huard-Warren شامل آزمون های 16SrRNA Rv Typing مشتمل بر لوکوس های Rv0577، Rv3877.8، Rv1970، Rv3120، Rv1510، IS1561) و RD typing (شامل لوکوس های (RD1، RD4، RD9، RD12) تعیین گردید.
    یافته ها
    هویت همه جدایه ها مایکوباکتریوم توبرکولوزیس تعیین شد و نشانه ای از آلودگی هیچ یک از بیماران به سایر اعضای مایکوباکتریوم توبرکلوزیس کمپلکس یا آلودگی هم زمان آنها به بیش از یک عضو کمپلکس مشاهده نگردید.
    نتیجه گیری
    با توجه به اهمیت پرورش گاو و گوسفند در اقتصاد منطقه فقدان مایکوباکتریوم بوویس در میان جدایه های بررسی شده تا حدودی غیر منتظره به نظر می رسد. همان گونه که این مطالعه نشان می دهد روش هوارد– وارن که انجام آن ساده و مقرون به صرفه می باشد، می تواند در هر دو گروه از آزمایشگاه های منطقه ای و همچنین مرجع در تشخیص افتراقی میان اعضاء مایکوباکتریوم توبرکلوزیس کمپلکس به کار گرفته شود.

    زمینه و هدف
    شیوه درمانی با گیاهان با عوارض جانبی و مقاومت دارویی کم برای درمان بیماری ها در سراسر دنیا به کار برده می شود. در این تحقیق به مقایسه اثرات مهاری نانو ذرات مس و اسانس بادرنجبویه بر E.coli وS.mutans در شرایط In vitro پرداخت شده است.
    روش بررسی
    در این تحقیق به منظور بررسی قطر هاله عدم رشد از روش انتشار دیسک آگار استفاده شد. سپس اثرات ضدباکتریایی این مواد 24 ساعت بعد از تیمار در غلظت های ppm 100و500 از نانو ذرات مس(nm10) و اسانس روغنی 5/12 تا 100 درصد با استفاده از آزمون ANOVAمورد بررسی قرار گرفت.
    یافته ها
    نانو ذرات مس 24 ساعت بعد از تیمار هیچ تاثیری بر این باکتری ها نداشت. در حالی که میانگین قطر هاله عدم رشد به ترتیب برای اشرشیاکلی و استرپتوکوکوس موتانس در غلظت های مختلف اسانس روغنی بادرنجبویه برابر mm13/0±30/31 و 13/0±13/16 بود. ترکیب غلظت های مختلف اسانس بادرنجبویه و غلظت ppm500 نانو ذرات مس بعد از24 ساعت هیچ اثری بر اشریشیاکلی وجود نداشت در حالی که در استرپتوکوکوس موتانس ترکیب غلظت های 25 و 50 درصد اسانس و ppm500 نانوذرات مس نسبت به غلظت های اسانس به تنهایی دارای تفاوت معنی داری (001/0p=)، (01/0p=) بود.
    نتیجه گیری
    ترکیب نانو ذرات مس و اسانس بر اشرشیاکلی اثر هم افزایی نداشته ولی در بعضی از غلظت های اسانس بر استرپتوکوکوس موتانس اثر هم افزایی داشته است.

    زمینه و هدف
    شناسایی و به کارگیری فیتاز جداشده ازمیکروارگانیسم های موجود در خاک، اهمیت زیادی در تولید این آنزیم به منظور استفاده تجاری در صنایع مختلف دارد.این مطالعه به منظور شناسایی گونه های باسیلوس تولید کننده آنزیم فیتاز و جداسازی ژن تولید کننده این آنزیم صورت گرفت.
    روش بررسی
    نمونه برداری از خاک های مختلف مناطق کوهستانی شهرستان تنکابن انجام شد. جداسازی اولیه باسیلوس در محیط غنی Bacillus Medium Agar صورت پذیرفت. بعد از جداسازی باکتری ها و استخراج ژنوم باکتری، با استفاده از پرایمر اختصاصی ژن تولید کننده آنزیم شناسایی شده و نسخه های زیادی از این ژن با روش PCR تکثیر شد. با استفاده از بررسی های تکمیلی ازجمله SDS-PAGE وسنجش فعالیت آنزیمی اندازه این پروتئین وشرایط بهینه تولید آن ارزیابی گردید.
    یافته ها
    از 40 نمونه خاکی که مورد آزمایش قرار گرفتند یک باکتری ترشح کننده آنزیم فیتاز جداسازی شد و این باکتری مورد توالی یابی قرار گرفت و گونه باسیلوس سوبتلیس سویه اس تی آر تشخیص داده شد. اندازه پروتئین فیتاز تولید شده از این ژن در حدود 45 کیلو دالتون بوده و فعالیت آنزیم در دمای 55 درجه در حدود 1/. در طول موج 415 نانومتر به دست آمد. ژن فیتاز با اندازه حدود bp1200 تکثیر صورت گرفت.
    نتیجه گیری
    با توجه به اینکه میکروارگانیسم های تولید کننده آنزیم فیتازدرشرایط محیطی طبیعی آنزیم را در مقادیر محدود و با کارائی و کیفیتی در سطوح میکروارگانیسم تولید می کنند، بنابرابن شناسایی وجداسازی باسیل های تولید کننده فیتاز و تخلیص این پروتئین از اهمیت ویژه ای برخوردار می باشد.

    زمینه و هدف
    جداسازی نوکاردیا به دلیل رشد سریع تر باکتری های هم جوار در محیط طبیعی بسیار مشکل است. بنابراین انتخاب محیط کشت اختصاصی و ترکیب مناسب که بتواند میکروارگانیسم های سریع الرشد مزاحم را حذف یا کاهش دهد همواره مورد توجه محققین بوده است. در این راستا ما به بررسی و مقایسه سه روش متداول آزمایشگاهی پرداختیم تا بهترین و مطمئن ترین روش جداسازی گونه های نوکاردیا شناخته و معرفی شود.
    روش بررسی
    نمونه های خاک از مناطق مختلف تهران جمع آوری و به آزمایشگاه منتقل شد. نمونه ها پس از آماده سازی، در سه محیط کشت مایع بدون کربن حاوی میله پارافینی، هیومیک اسید ویتامین ب آگار و پارافین آگار تلقیح و سپس در دمای 35 درجه سانتیگراد به مدت 3تا 5 هفته گرماگزاری شدند.
    یافته ها
    از تعداد 110 نمونه خاک جمع آوری شده، 31جدایه نوکاردیا (18/28%)بدست آمد که به ترتیب 19جدایه(27/17%) توسط روش Paraffin Bait، 4 جدایه(63/3%) با روش کشت در محیط هیومیک اسید ویتامین ب آگار و 8 جدایه(27/7%) با روش کشت در محیط پارافین آگار به دست آمد.
    نتیجه گیری
    کارایی روش Paraffin Bait با توجه به جداسازی بیشتر، ارزانی محیط کشت و وضوح کلنی های مشکوک به جنس نوکاردیا بیشتر از روش های دیگر کشت می باشد.

    زمینه و هدف
    کیتین یک پلیمر خطی از واحد های N-استیل گلوکز آمین است که پس از سلولز فراوان ترین بیوپلیمر در طبیعت به حساب می آید. در دهه های اخیر کیتیناز ها به دلیل کاربردهای وسیع، به ویژه به عنوان کنترل زیستی برعلیه قارچ ها مورد توجه زیادی قرار گرفته اند.
    روش بررسی
    جداسازی باسیلوس های مولد کیتیناز با جمع آوری 40 نمونه خاک از چهار منطقه جغرافیایی گرگان انجام گرفت و ویژگی تجزیه کنندگی کیتین آنها با بررسی هاله شفاف در پلیت های حاوی کیتین کلوئیدی مشخص و مورد ارزیابی قرار گرفت. شناسایی سویه های منتخب بر اساس تاکسونومی پلی فازی انجام گرفت. استخراج DNA برای شناسایی دقیق تر و تعیین توالی انجام شد. اثر ضدقارچی با روش چاهک برعلیه قارچ های Candida albicans، Aspergillus niger، Aspergillus flavus (IR 6)، Alternaria alternate و oxyporum Fusarium مورد ارزیابی قرار گرفت.
    یافته ها
    تعداد 9 کلونی باسیلوس کیتیناز مثبت از پلیت های کلوئیدال کیتین آگار جدا شد که 5 مورد از آنها اثر ضدقارچی از خود نشان دادند ودر این بین سویه R6بیشترین اثر، R2 و R3 کم ترین اثر را روی قارچ ها نشان دادند. مقایسه توالی 16S rRNA این جدایه ها با باکتری های شناخته شده، تشابه ژنتیکی حدود 95-97 درصد را نشان دادند.
    نتیجه گیری
    برخی باکتری خاک می توانند با عوامل بیماری زایی انسانی و گیاهی خاک اثرات متضاد داشته باشند.

    زمینه و هدف
    در کشور ما اغلب از آزمایش رایت برای تشخیص بروسلوزاستفاده می شود. به علت حساسیت پایین این آزمایش موارد منفی کاذب بالا گزارش می شود. در این مطالعه نتایج آزمون های رایت و الایزادر افراد مشکوک به بروسلوز مورد مقایسه قرار گرفت.
    روش بررسی
    نتایج آزمون رایت، 2MEرایت، کومبس رایت و تعیین کلاس های IgG، IgM علیه بروسلا به روش الایزا مورد ارزیابی قرار گرفت. از مجموع 1183 افراد با آزمایش رایتارجاع داده شده، 148 مورد آزمون کومبس رایت و 228 مورد آزمون 2ME رایتبرای آنها درخواست شده بود. برای 32 نفر از آنها علاوه بر آزمون رایت، آزمایش های سنجش آنتی بادی بر علیه بروسلا از کلاس های IgG و IgMنیز انجام شد.
    یافته ها
    آزمونرایت در 4/95 درصد موارد منفی بود. 7/8 درصد موارد از لحاظ آزمایش کومبس رایت و 7/4 درصد از آن ها از لحاظ آزمایش 2ME رایت مثبت بودند و در 3/2 درصد موارد رایت منفی، کومبس رایت مثبت بود. 16 نفر از لحاظ آزمون رایت و الایزامنفی بودند. در 8 مورد رایتمنفی، الایزااز کلاس IgM مثبت و IgG منفی و 4 مورد رایت منفی، الایزا از لحاظ IgM منفی و IgG مثبت بود. همچنین 4 مورد از لحاظ رایت مثبت و آنتی بادی از کلاس IgM و IgG مثبت بودند.
    نتیجه گیری
    به علت عدم هم خوانی نتایج آزمایش های رایت و الایزاو با توجه به در دسترس بودن، حساسیت بالا و تعیین نوع کلاس آنتی بادی در روش الایزا این روش برای تشخیص بروسلوز مورد تاکید می باشد.

    زمینه و هدف
    عفونت های ادراری و باکتریمی به عنوان یک مشکل عمده در بیماران پیوند کلیه مطرح می باشند. از مهمترین علل آن می توان به رژیم درمانی سرکوب کننده، عمل جراحی و تماس با میکروب های بیمارستانی اشاره نمود. هدف از این مطالعه، تعیین میزان شیوع عوامل باکتریال در کشت خون و ادرار در افراد داوطلب پیوند قبل و بعد ازپیوند بود.
    روش بررسی
    در این مطالعه 33 بیمار داوطلب پیوند کلیه در طی یک سال از نظر کشت ادرار و کشت خون قبل و بعد از پیوند مورد بررسی قرار گرفتند. از هر بیمار یک نمونه ادرار و یک نمونه خون قبل از پیوند و سه نمونه بعد از عمل پیوند تهیه گردید. نمونه ها با استفاده از روش های استاندارد میکروب شناسی کشت و ارگانیسم های عفونت زا شناسایی گردیدند.
    یافته ها
    از 132 نمونه ادرار به ترتیب 5/20 درصد اشریشیاکلی، 3/5 درصد انتروباکتر، 3 درصد کلبسیلا، 5/1درصد استافیلوکوک اپیدرمیدیس جدا گردید. در نمونه های خون تهیه شده از بیماران، 1/9 درصد انتروباکتر، 8/6 درصداشریشیاکلی، 8/3 درصد استافیلوکوک اپیدرمیدیس، 8/0 درصد کلبسیلا جدا شد.
    نتیجه گیری
    بروز عفونت ادراری در بیماران پیوند کلیه بالا است و عفونت با اشریشیا کلی شایع ترین علت آن قبل و بعد از عمل پیوند می باشد.

    زمینه و هدف
    عفونت های باکتریال به ویژه مننژیت، پنومونی و سپتی سمی هنوز از عوامل مهم مرگ میرکودکان به شمار می رود. هدف از این مطالعه، بررسی شایع ترین عوامل باکتریایی عفونت های کودکان زیر14سال و تعیین مقاومت دارویی باکتری های جدا شده در آنها بود.
    روش بررسی
    در طی دوسال تعداد 1897 نمونه به دست آمده از بیماران دارای علائم بالینی عفونت باکتریال مراجعه کننده از نظر کشت میکروبی، توزیع سنی، جنسی و نتایج آنتی بیوگرام مورد مطالعه قرار گرفت. گونه های میکروبی با روش های بیوشیمیایی و سرولوژیک مورد شناسایی قرار گرفتند.
    یافته ها
    از مجموع 1897 نمونه آزمایش شده، تعداد 563 نمونه (6/29%) کشت مثبت باکتریایی به دست آمد که از این تعداد 7/74 درصد باکتری های گرم منفی و 3/25 درصد باکتری های گرم مثبت جدا گردید. شایع ترین باکتری های جدا شده اشریشیا کلی 1/34 درصد، استافیلوکوک اورئوس 1/17 درصد، پسودوموناس آئروژینوزا 4/12 درصد، کلبسیلا 11 درصد و استافیلوکوک اپیدرمیدیس 7/5 درصد بودند. موثرترین آنتی بیوتیک برای هر دو گروه باکتری گرم مثبت و منفی ایزوله شده از بیماران، سفتریاکسون، نیتروفورانتوئین، نالیدیکسیک اسید، آمیکاسین و جنتامایسین بود.
    نتیجه گیری
    باکتری های گرم منفی به ویژه اشریشیا کلی، پسودوموناس آئروژینوزا و کلبسیلا از عوامل اصلی ایجاد کننده عفونت باکتریال کودکان منطقه مورد بررسی به شمار می آیند. اکثر گونه ها نسبت به آنتی بیوتیک های رایج مانند آمپی سیلین، تری متوپریم و کلرامفنیکل مقاومت نسبتا بالایی از خود نشان دادند.

  • تعیین الگوی مقاومت و اثر هم افزایی تلفیق چند آنتی بیوتیک بر روی هلیکوباکتر پیلوری جدا شده از نمونه های بیوپسی اولسر معده در شرایط آزمایشگاهی

    زمینه و هدف
    یکی ازمهمترین دلایل عود و شکست درمان عفونت های هلیکوباکتریایی، ایجاد مقاومت در هلیکوباکتر پیلوری به آنتی بیوتیک ها است. با مصرف همزمان چند دارو که با هم اثر هم افزایی دارند می توان تا اندازه ای این مقاومت را مهار نمود. هدف ازاین مطالعه تعیین الگوی مقاومت و بررسی اثر هم افزایی تلفیق چند آنتی بیوتیک بر روی هلیکوباکتر پیلوری بود.
    روش بررسی
    از 100 بیمار دارای اولسر معده مراجعه کننده به بیمارستان امام رضا (ع) شهرستان آمل نمونه بیوپسی تهیه شد. بعد از جداسازی و شناسائی هلیکوباکترپیلوری تست آنتی بیوگرام با دیسک های متفاوت حاوی آنتی بیوتیک، ترکیب دو آنتی بیوتیک (مترونیدازول + کلاریترومایسین) و سه آنتی بیوتیک (مترونیدازول + کلاریترو مایسین + سیپرو فلوکسازین)انجام گرفت.
    یافته ها
    در این مطالعه از 53(53٪) نمونه بیوپسی، هلیکوباکترپیلوری جدا و شناسائی شد که 49 مورد آن(5/92 ٪) مقاوم به مترونیدازول، 14مورد آن(26٪ (مقاوم به آموکسی سیلین،10 مورد آن (19٪ (مقاوم به کلاریترومایسین، 7مورد (13٪) مقاوم به تتراسایکلین، 13مورد (5/24٪) مقاوم به فورازولیدون و7 مورد (13٪) مقاوم به سیپروفلوکساسین بودند. در بررسی اثر هم افزایی جلوگیری از رشد دیسک های تلفیقی دو و سه تای نسبت به موثرترین آنتی بیوتیک قطر هاله تا 5 میلی متر افزایش دیده شد.
    نتیجه گیری
    قطر هاله دیسک های تلفیقی دو آنتی بیوتیک و سه آنتی بیوتیک نسبت به قطر هاله عدم رشد هر آنتی بیوتیک به تنهایی بیشتر بود.

    زمینه و هدف
    مصرف بی رویه آنتی بیوتیک های وسیع الطیف باعث مقاومت دارویی میکروب ها به ویژه ارگانیسم های بیمارستانی می شود. با توجه به بروز بالای مقاومت آنتی بیوتیکی در بیمارستان ها، در این مطالعه مقاومت آنتی بیوتیکی باکتری های گرم منفی در یک مرکز آموزشی درمانی بررسی شد.
    روش بررسی
    دراین مطالعه مقطعی اطلاعات مربوط به نمونه های بیولوژیک سال 1385-1387 که کشت آنها مثبت شده بود وارد مطالعه شدند. جهت تعیین مقاومت میکروبی ارگانیسم جدا شده در هرمورد با آنتی بیوگرام نسبت به آنتی بیوتیک ها مورد ارزیابی قرار گرفت. بعدازکشت نمونه ها نوع باکتری با استفاده از محیط های کشت افتراقی و اختصاصی وآنتی سرم های خاص، مشخص شد. سپس مقاومت آنتی بیوتیکی با، روش disk diffusion در هر مورد انجام شد.
    یافته ها
    شایع ترین میکروارگانیسم های بدست آمده از کل کشت های گرم منفی باکتری Ecoli بود که کمترین مقاومت دارویی این باکتری به نیتروفوزانتوئین بوده است.
    نتیجه گیری
    نتایج این مطالعه نشان داد که مقاومت آنتی بیوتیکی باکتری ها دراین منطقه قابل توجه است. بنابراین پیشنهاد می شود جهت تعیین الگوی دقیق مقاومت آنتی بیوتیکی و انتخاب رژیم های درمانی مناسب و موثرتر مطالعات گسترده تری دراین منطقه انجام گیرد.

    Background And Objective
    The main cause of spreading staphylococcal infections among patients is the healthy carriers working in hospitals. With the secretion of different sorts of toxins such as entrotoxin, this bacteria can provide the conditions for attacking on the host. The main objective of this study is identification of the characteristics and differences in the Staphylococcus aureus isolated from healthy carriers and from the patients on the basis of enterotoxin genes (sea-see).
    Material And Methods
    One hundred and twenty of the patients and 80 of healthy carriers worked in health centers of Gorgan, north of Iran, were investigated for S. aureus isolate. The isolates were evaluated by PCR for Enterotoxin Genes A-E (SEA to SEE).
    Results
    Enterotoxin genes (SEA to SEE) was found in 87.5% of the total isolates and the most frequent one was enterotoxin gene sea (N= 124). The prevalence of these isolates in healthy carriers was significantly higher than those of the patients.
    Conclusion
    Based on the results, the high percentage of S. aureus isolated from clinical samples contains enterotoxin genes. Therefore, Human as the source and carrier of S. aureus is paramount importance, which is due to significant relationship between being toxigenic strains and the source of isolation.

    Background And Objective
    A high level of homogeneity observed within all bacteria in the Mycobacterium tuberculosis complex makes a property that seriously challenges traditional biochemical-based identification methods of these pathogens in the laboratory. The work presented here was conducted to characterize Mycobacterium tuberculosis complex isolates in Golestan, Northern Iran.
    Material And Methods
    Between 2008 and 2010, 42 mycobacterial isolates were collected from clinical tuberculosis-suspected patients in Golestan province. The isolates were sub-cultured on fresh Mycobacterium-specific culture media including glycerinated and pyruvated Lowenstein-Jensen slopes. The isolates were subsequently subjected to a PCR-based identification scheme coined Huard-Warren method. This strategy consisted of three individual algorithms namely, 16SrRNA; RV typing (Rv0577, Rv3877.8, Rv1970, Rv3120, Rv1510 and IS1561) and RD typing (RD1, RD 4, RD9 and RD12).
    Results
    All isolates were proved to be M. tuberculosis. Furthermore, none of the patients were being infected with any other member of the M. tuberculosis complex or simultaneously co-infected with two mycobacteria. This fundamental observation was independently obtained by specific culture media, RV typing and also RD typing.
    Conclusion
    Considering the fact that cattle and sheep farming play an important role in the economy of the region, absence of Mycobacterium bovis in the studied isolates can be unexpected to some extent. Huard-Warren which is a simple and cost-effective identification method can be used in both reference and regional laboratory for differential diagnosis of tuberculosis.

    Background And Objective
    the plants with the less side effects and drug resistance have been used worldwide. In this study, we compared the inhibitory effects of nanocopper and essential oil of Melissa officinalisL.a on E.coli and S.mutans in In Vitro condition.
    Material And Methods
    for studying diameter of inhibitory zone, disk agar diffusion method was used. Then, antibacterial effects of these substances were evaluated by treating them for 24 hours in Nanocopper particles (concentration of 100 and 500 ppm) and essential oil (12.5% to 100%), and analyzed by ANOVA.
    Results
    twenty-four hours after treatment, nanocopper had no inhibitory effect on these bacteria. However, the diameter of inhibitory zone for E.coli and S.mutans was 31.30±0.13mm and 16.30±0.13mm, respectively. There was not any synergistic effect between different concentrations of this plant and 500ppm of nanocopper after 24 hours of treatment on E.coli. But for S. Mutans, the diameter of inhibitory zone for mixture of 50% & 25% essential oil and 500ppm concentration of nanocupper was increased significantly compared to only essential oil (p=0.001,p=0.01).
    Conclusion
    based on the findings, nanocopper particles and essential oil of Melissa officinalis L.a have not any synergic effects on E.coli, but with some concentrations of this plant the reverse is true on S.mutans.

    Background And Objective
    Recognizing and using of isolated phytase in the soil microorganisms are paramount importance to produce the Phytase enzyme utilized commercially in different industries. This study was conducted to recognize different bacillus species which are Phytase producers and detection of the gene that can produce this enzyme.
    Material And Methods
    Soil samples were gathered through different parts of mountainous areas. The early isolation of bacillus was carried out in Bacillus Medium Agar. After isolating the bacteria and genome extraction, the responsible gene of enzyme producer recognized and amplified by PCR method. The size of this protein and the optimal production situation in supplemental exploitation such as SDS-PAGE and the enzymatic activity of its size were evaluated.
    Results
    Of 40 samples, one bacterium secreting Phytase enzyme was isolated. This bacterium was sequenced and recognized Bacillus Sobtlis species that is classified in STR Genus. The size of protein phytase produced by this gene was about 45 KD and the enzyme activity at 55 degrees was measured about 5.65 in wavelength of 415 NM. The phytase gene with the size of 1200 bp was propagated.
    Conclusion
    the microorganisms, in natural conditions, produce Phytase enzyme in limited amount and with the quality appropriate to microorganisms. Thus, isolating the bacilli producing Phytase enzyme and purifying this protein are highly significant.

    Background And Objective
    The Isolation of Nocardia species is complex and time-consuming, which is due to rapid growth of adjacent bacteria. Because of the importance of a specific medium with the ability of controlling intrusive microorganisms, this study aimed at comparing three laboratory methods to introduce the reliable isolation technique for Nocardia species.
    Material And Methods
    The soil samples were collected from different regions of Tehran province, Iran, and carefully transferred to the laboratory. The samples were cultured in three different media including Paraffin Baiting,Humic acid vitamin B agar and Paraffin agar, and incubated for 3-4 weeks at 35 °C.
    Results
    Of 110 soil samples, 31 Nocardia isolates (28.18%) were obtained from the media including Paraffin Baiting, (19; 17.27%), Humic acid and vitamin B agar (4; 3.63%), and Paraffin agar, (8; 7.27%).
    Conclusion
    because of high rate of isolation, low cost and the clearance of colonies suspected nocardia, Paraffin Bait technique is more reliable and efficient compared to the other methods.

    Background And Objective
    Chitin, which is a linear polymer of N-acetyl glucosamine residues, has been the most abundant polymer in nature after cellulose. In recent decades, Chitinases have received increased attention because of their wide range of applications, especially in biological control against fungi.
    Material And Methods
    the isolation of bacilli producing chitinolytic enzymes was performed by collecting 40 soil samples from various regions of Gorgan, northern of Iran. The chitinolytic potential of the isolates was indicated by observation of clear zone in colloidal chitin agar medium. Identification of selected strains was performed by polyphasic taxonomy, and subtler identification and sequensing were carried out by extraction DNA. Antifungal effect was evaluated by well method against Candida albicans (ATCC 10231) Aspergillusniger (ATCC 2029)، Aspergillusflavus (IR6) Fusariumoxyporum (PTCC 5115) and Alternariaalternata (PTCC 5224).
    Results
    Nine colonies of chitinase positive bacillus were isolated on choloidal Chitin Agar (CCA) and five of them had antifungal effect. R6 strain had the highest, and R2 and R3 had the lowest effect on fungi. The 16S rRNA sequence of these isolations in comparison with the known bacteria has 95-97% similarity.
    Conclusion
    Some of the soil bacteria can have antagonestic effects on human and phytopathogenic agents existed in soil.

    Background And Objective
    In our country, the Wright test routinely is used for diagnosing brucellosis. Because of its low sensitivity, the range of false-negative results is high. Therefore, we aimed at comparing Wright and ELISA in the people suspected brucellosis.
    Material And Methods
    The results of Wright, 2ME, Coombs Wright tests were compared with Anti-Brucella IgG, Anti-Brucella IgM. Of 1183 subjects referred for Wright test, 148 of them were investigated for Coombs Wright and 228 for 2ME Wright. In addition to Wright test for 32 cases, Brucella IgG and IgM classes were also experimented.
    Results
    Wright test was negative in 95.4% of cases. Of these negative results, 2.3% were positive for Coombs Wright. Eight-point-five percent of the cases were positive for Coombs Wright test and 4.7% for 2ME Wright test. Sixteen cases were negative for both Wright and ELISA. In 8 cases of Wright-negative, ELISA IgM class was positive and IgG class was negative, and in 4 cases of Wright-negative, ELISA IgM was negative and IgG was positive. About 4 cases of Wright-positive, IgM and IgG antibody classes were positive.
    Conclusion
    Due to the mismatch between the results of Wright agglutination test and ELISA method and with regard to availability, high sensitivity and determining the type of antibody classes in ELISA, it is focused on ELISA method for brucellosis diagnosis.

    Background And Objective
    Urinary tract infections and bacteremia are the major problems in renal transplant patients, which are mostly due to immunesuppressive regimens, surgery, and exposure to the germs in hospital. The aim of this study was to determine the prevalence of bacterial agents in the blood and urine samples of kidney transplant candidates.
    Material And Methods
    In this one-year-long study, thirty-three renal transplant candidates were assessed for urine and blood cultures. One urine and blood samples from each patient before transplantation and three samples after transplantation were collected. The Samples, using standard microbiological methods, were investigated and infectious organisms identified.
    Results
    In 133 urine samples, Escherichia coli (20.5%), Enterobacter spp. (5.3%), Klebsiella spp. (3 %) and Staphylococcus epidermidis (1.5%) were isolated. In the blood samples, Enterobacter spp. (9.1%), Escherichia coli (6.8%), Staphylococcus epidermidis (3.8%) and Klebsiella spp. (0.8%) were isolated.
    Conclusion
    The results indicate that urinary tract infection was high in patients with transplanted kidney, and E. coli is the most common cause of this infection.

    Background And Objective
    Bacterial infectioins in particular meningitis, pneumonia and septicemia are still some of the most causes of mortalities in children.The aim of present study was to identify the most common bacterial agents causing infectionis in children under 14 and detection of antibiotic resistance paterns.
    Material And Methods
    During two years,1897samples were obtained from the patients suspected bacterial infectioins. They were investigated for bacterial cultures, age, sex and antibiogram patterns. The species were identified by biochemical and serological methods.
    Results
    Of 1897 samples, 563 (29.6%) had positve bacterial culture. Of these 74.7% were gram negative and 25.3% gram positive. The most common species were Escherichia coli(34.1%), Staphylococcus aureus (17.1%), Psuedomonas aeroginosa (12.4%), Kelebsiella (11%) and Staphylococcus epidermidis (5.7%). The most effective antibiotics against both gram positive and gram negative bacteria were ceftriaoxne, nitrofurantoin, nalidixic acid, amikacin and gentamycin.
    Conclusion
    The gram negative bacteria in particular Escherichia coli, Psuedomonas aeroginosa and Kelebsiella are the predominant causes of bacterial infections in children under 14 in these regions. Most species showed a high relative resisitance to routine antibiotics such as ampicillin, trimethoprim and chloramphenicol.

    Background And Objective
    Resistance of Helicobacter Pylori (H. pylori) to antibiotics is the main cause of relapse into Helcobacterial infections. With the use of several antibiotics that have synergistic effect، we can inhibit this antibiotic resistance. Thus، we aimed at determining resistance patterns and assessing the synergy of combining multiple antibiotics on H. pylori.
    Material And Methods
    Biopsy specimens were taken from 100 patients with gastric ulcer referred to Imam Reza hospital in Amol، north of Iran. After isolation and identification of H. Pylori، antibiogram was performed with different antibiotic disks containing one antibiotic، a combination of two antibiotics (metronidazole + clarithromycin) and three antibiotics (metronidazole + Claritromycin + Ciprofloxacin).
    Results
    In this study، H. pylori were isolated from 53 (53%) biopsy specimens. Of these، 49 (5. 92%) were resistant to metronidazole، 14 (26%) to amoxicillin، 10 (19%) to clarithromycin، 7 (13%) to tetracycline، 13 (5/24%) to furazolidone and 7 (13%) to ciprofloxacin. In survey of synergistic effect، an increase in inhibition zone diameter around of combined disks was seen up to 5mm compared to the most effective antibiotic.
    Conclusion
    The inhibition zone diameter of discs containing two and three antibiotics was large، in comparison with one antibiotic.

    Background And Objective
    The excessive use of broad-spectrum antibiotics will lead to drug resistance of microorganism and specially nosocomial organisms. Because of high incidence of antibiotic resistance in hospitals, we aimed to study antibiotic resistance to gram negative bacteria.
    Material And Methods
    This cross-sectional study was conducted on the data of biological samples (2006-2008), with positive culture result. Using antibiogram, microbial resistance to isolated microorganism was determined, and after culturing the samples, bacteria were identified by using differential media and antiserum. Then, antibiotic resistance was performed by disk diffusion.
    Results
    The most common gram-negative microorganism obtained from all cultures was Ecoli with the lowest drug resistance to Nitrofurantoin.
    Conclusion
    Based on the results, antimicrobial resistance pattern is not the same in different places and furthermore it is ever changing. Therefore, further research is needed to be done to have an accurate pattern of antibiotic resistance to provide effective treatment regimens.
