    سابقه و هدف
    سل به عنوان دومین علت مرگ و میر ناشی از عفونت شناخته شده است. روش های مولکولی مانند PCR، تکنیک های مناسبی برای شناسایی مایکوباکتریوم توبرکیلوسیس (MTB) هستند. بروز نتایج متفاوت در آزمایشگاه های مختلف، از معایب این تکنیک مولکولی است. این مطالعه با هدف طراحی، ساخت و کاربرد کنترل داخلی در روش PCR به منظور تشخیص MTB انجام شد.
    مواد و روش ها
    به منظور ساخت کنترل داخلی ویژه MTB، ابتدا پرایمرهای ویژه آزمون PCR برای تشخیص مولکولی بهینه و سپس حساسیت و ویژگی آن مشخص گردید. پرایمرهای مرکب (Composite Primer) برای IC-MTB نیز طراحی، تکثیر و سپس کلون شد. IC-MTB تکثیر شده در پلاسمید pTZ57R الحاق و در باکتری اشریشیا کلی JM107 ترانسفورم و کلون گردید. تعداد حداقل IC در هر واکنش PCR از طریق رقیق سازی و همچنین طیف پاسخ PCR همراه با کنترل داخلی مورد بررسی قرار گرفت.
    یافته ها
    اندازه محصول تشخیصی MTB با پرایمرهای اختصاصی آن برابر با 245 جفت باز و محصول IC-MTB برابر با 660 جفت باز بود. که از نظر اندازه، اختلاف مطلوب را با هم دارند. تعداد حداقل IC در هر واکنش 1000 عدد تعیین شد. حداقل و حداکثر حساسیت روش PCR همراه با IC برای DNA باکتری مایکوباکتریوم توبرکیلوسیس بین 10 میلیون تا 10 باکتری محاسبه گردید. در ارزیابی ویژگی با عوامل مختلف نیز هیچ محصول ناخواسته ای مشاهده نشد.
    نتیجه گیری
    استفاده از یک کنترل داخلی در روش PCR می تواند خطاها را در آزمون تشخیص مولکولی مایکوباکتریوم توبرکیلوسیس شناسایی نماید. در واقع تکثیر IC نشان دهنده انجام صحیح تمامی مراحل تکثیر و تشخیص می باشد.

    سابقه و هدف

    اسینتوباکتر بامانی یکی از علل مهم عفونت های بیمارستانی در سراسر دنیا است. سویه های اسینتوباکتر مقاوم به آنتی بیوتیک ها باعث محدودیت هایی در درمان موثر می شوند. این مطالعه با هدف بررسی جهش در ژن gyrA جدایه های بالینی اسینتوباکتر بامانی مقاوم به کینولون و ارزیابی الگوی حساسیت آنتی بیوتیکی آن ها در اصفهان انجام شد.

    مواد و روش ها

    در این مطالعه مقطعی-توصیفی، 70 جدایه اسینتوباکتر از بیماران بستری در ICU بیمارستان الزهرا اصفهان جمع آوری گردید. به منظور شناسایی بیشتر جدایه ها از تست های بیوشیمیایی استاندارد استفاده شد. حساسیت آنتی بیوتیکی با روش انتشار دیسک برای 8 آنتی بیوتیک انجام گرفت. حداقل غلظت باز دارندگی (MIC) سیپروفلوکساسین و لووفلوکساسین با روش E-test برای جدایه ها مطابق با استاندارد CLSI تعیین شد. همچنین به منظور شناسایی جهش در ژن gyrA در جدایه های مقاوم به سیپروفلوکساسین و لووفلوکساسین از روش PCR-RFLP استفاده گردید.

    یافته ها

    جدایه ها بیشترین میزان مقاومت آنتی بیوتیکی را نسبت به سیپروفلوکساسین (100%)، جنتامایسین (100%) و کمترین میزان را نسبت به ایمی پنم (92.8%) و مروپنم (90%) نشان دادند. در روش MIC میزان مقاومت جدایه ها به سیپروفلوکساسین و لووفلوکساسین 100% و 65.7% بود. میزان مقاومت چند دارویی 66.7% درصد و فراوانی جهش ژن gyrA در جدایه ها 93% بود.

    نتیجه گیری

    در این مطالعه مشاهده شد که جدایه های اسینتوباکتر بامانی مقاوم به کینولون دارای جهش در ژن gyrA بودند. این جهش نقش مهمی در افزایش مقاومت در جدایه ها دارد. تشخیص سریع جدایه های اسینتوباکتر بامانی مقاوم به کینولون می تواند کمک مناسبی برای درمان این عفونت ها باشد.

    سابقه و هدف

    سودوموناس آئروژینوزا از مهمترین علل عفونت های بیمارستانی در بیماران سوختگی می باشد. یکی از مشکلات درمان این بیماران بروز مقاومت های آنتی بیوتیکی با مکانیسم های مختلف می باشد. پمپ های افلاکس mex نقش اساسی در بروز مقاومت چندگانه نسبت به داروهای ضد میکروبی دارند. این مطالعه با هدف ارزیابی شیوع ژن های پمپ های افلاکس mexA-B-oprM و نیز بررسی حضور این ژن ها به صورت فنوتیپی انجام شد.

    مواد و روش ها

    در این مطالعه مقطعی 250 نمونه سواب از زخم بیماران دچار سوختگی سطح 2 و 3 مراجعه کننده به بیمارستان سوختگی قطب الدین شیرازی جمع آوری شد. جدایه های سودوموناس آئروژینوزا با آزمون های بیوشیمیایی و روش PCR تایید گردیدند. الگوی حساسیت دارویی با روش انتشار دیسک، بررسی فنوتیپی فعالیت پمپ های افلاکس با روش کارت ویل و بررسی ژن های mex A و mex B با روش PCR انجام گرفت.

    یافته ها

    در مطالعه حاضر 26.40% (66 مورد) نمونه های زخم بیماران سوختگی آلوده به سودوموناس آئروژینوزا بودند. این باکتری به تمامی آنتی بیوتیک های مورد بررسی به جز کلیسیتین مقاوم بود. 66.66% از جدایه ها (44 مورد) دارای پمپ افلاکس بودند. از این میان 92.42% و 87.87% از جدایه ها به ترتیب دارای ژن های mexA و mexB بودند.

    نتیجه گیری

    نتایج نشان داد که روش ژنوتیپی بسیار دقیق و قابل اعتماد تر از روش فنوتیپی در تشخیص پمپ های افلاکس در جدایه های بالینی سودوموناس آئروژینوزا می باشد. با توجه به حضور بیش از 90% ی ژن های پمپ افلاکس در سودوموناس آئروژینوزا، بررسی حضور این ژن ها در پیشنهاد الگوی درمانی مناسب بیماران آلوده به این باکتری حائز اهمیت است.

    سابقه و هدف
    پوسیدگی دندان شایع ترین بیماری مزمن در جهان است و استرپتوکوک های گروه میوتانس اصلی ترین عامل ایجاد پوسیدگی در انسان به شمار می روند. استفاده از پروبیوتیک ها یکی از روش های جدید پیشگیری از پوسیدگی دندان می باشد. از جمله پروبیوتیک های مهم در این فرآیند لاکتوباسیلوس رامنوسوس است. این مطالعه با هدف بررسی تاثیر لاکتوباسیلوس رامنوسوس بر فرآیند اتصال استرپتوکوک های دهانی انجام شد.
    مواد و روش ها
    این پژوهش به صورت مقطعی- توصیفی بر روی 40 سویه استرپتوکوک های میوتانس و غیرمیوتانس از نمونه های پلاک و پوسیدگی دندان افراد داوطلب مراجعه کننده به دانشکده دندانپزشکی دانشگاه آزاد اسلامی واحد خوراسگان اصفهان انجام شد. قدرت تشکیل بیوفیلم آن ها ارزیابی و قوی ترین سویه ها در تشکیل بیوفیلم انتخاب شدند. تاثیر لاکتوباسیلوس رامنوسوس (ATCC4769) بر اتصال استرپتوکوک ها به چند روش بررسی گردید. روش 1، مخلوط هم حجم لاکتوباسیل و استرپتوکوک به طور هم زمان، روش 2، 30 دقیقه قبل از ورود استرپتوکوک به سیستم، روش 3، رسوب سلولی پروبیوتیک و در روش 4 روماند کشت شبانه پروبیوتیک استفاده گردید.
    یافته ها
    لاکتوباسیلوس رامنوسوس در هر 4 روش موجب کاهش اتصال استرپتوکوک ها به سطح گردید. از میان روش های مورد بررسی روش 2 از بقیه موثرتر بود. به طوری که در روش 2 کاهش بیشتری در اتصال استرپتوکوک های میوتانس نسبت به غیر میوتانس ایجاد گردید. بین روش های 3 و 4 نیز تفاوتی از نظر اتصال مشاهده نشد. اما تاثیر هر دو روش بر استرپتوکوک های میوتانس از دیگر استرپتوکوک ها بیشتر بود.
    نتیجه گیری
    استفاده از باکتری لاکتوباسیلوس رامنوسوس به عنوان پروبیوتیک اتصال استرپتوکوک های میوتانس و غیرمیوتانس به سطوح را کاهش داده و می تواند منجر به کاهش شیوع پوسیدگی دندان گردد.

    سابقه و هدف
    آنتی بیوتیک های بتالاکتام در حال حاضر رایج ترین درمان برای عفونت های باکتریایی می باشند. از مهم ترین مکانیسم های مقاومت به این آنتی بیوتیک ها تولید آنزیم های بتالاکتامازی توسط باکتری ها می باشد. هدف از این مطالعه بررسی وجود ژن های بتالاکتامازی ampC و esbl در نمونه های اشریشیا کلی جدا شده از موارد انسانی و طیور می باشد.
    مواد و روش ها
    در این مطالعه مقطعی - توصیفی تعداد 400 نمونه ادرار از مراکز درمانی و 200 نمونه سواب کلواک طیور از مرغداری های استان تهران جمع آوری گردید. شناسایی فنوتیپی سویه های مولد بتا لاکتاماز با استفاده از آزمون حساسیت ضد میکروبی به روش انتشار دیسک انجام شد. به منظور تایید حضور ژن های ampC و esbl در باکتری های مولد آن از روش PCR استفاده گردید.
    یافته ها
    در مجموع 120 نمونه انسانی (30%) و 50 نمونه طیوری (25%) آلوده به باکتری اشریشیاکلی بودند. ارزیابی فنوتیپی نشان داد که 54 مورد (45%) از نمونه های انسانی تولید کننده آنزیم بتا لاکتامازی نوع ESBLs و 2 مورد (1.67%) تولید کننده AmpC بودند. در نمونه های طیوری در 3 مورد (21.4%) وجود آنزیم بتا لاکتامازی نوع ESBLs تایید شد. این نمونه ها فاقد ژن ampC بودند. بررسی های ژنوتیپی نشان داد که 2 (1.67%) نمونه انسانی واجد ژن های بتالاکتامازی نوع ampC بودند.
    نتیجه گیری
    نتایج به دست آمده در این مطالعه نشان می دهد که ژن بتالاکتامازی ampC در نمونه های انسانی وجود داشته اما در نمونه های طیوری باید مطالعات دقیق تری صورت پذیرد. به دلیل اهمیت ارگانیسم های تولیدکننده این آنزیم ها در ایجاد عفونت های بیمارستانی و برای جلوگیری از گسترش آن ها توصیه می شود تحقیقات وسیع تری از نظر بررسی شیوع واقعی این آنزیم در ایران انجام شود.

    سابقه و هدف

    آلودگی های نفتی، یکی از مشکلات عمده تهدید کننده محیط زیست به شمار می روند. امروزه روش تجزیه زیستی با استفاده از باکتری های بومی تجزیه کننده هیدروکربن به دلیل مناسب بودن از لحاظ اقتصادی و زیست محیطی مورد توجه می باشد. آلکانی ورراکس به عنوان یک باکتری تجزیه کننده هیدروکربن در محیط های دریایی آلوده به نفت خام شناخته شده است. این مطالعه با هدف بررسی توانایی تجزیه زیستی نفت خام توسط آلکانی ورراکس دیزلولی، جداسازی شده از رسوبات منطقه ساحلی خلیج فارس انجام شد.

    مواد و روش ها

    در این مطالعه بنیادی کاربردی، ابتدا عمل جداسازی و غنی سازی باکتری تجزیه کننده نفت از رسوبات منطقه ساحلی خلیج فارس انجام گرفت. باکتری جداسازی شده، با روش شناسایی مولکولی مبتنی بر توالی 16S rRNA شناسایی گردید. توانایی تجزیه زیستی سویه جداسازی شده، در غلظت های مختلف نفت خام مورد بررسی قرار گرفت.

    یافته ها

    با استفاده از روش شناسایی مولکولی باکتری جداسازی شده به عنوان آلکانی ورراکس دیزلولی شناسایی گردید. این باکتری در مدت زمان کمتر از 1 دقیقه به صورت پایدار قادر به متلاشی کردن قطرات نفت بود. نتایج نشان داد که در غلظت 1، 2.5 و 5 درصدی نفت خام، میزان تجزیه زیستی به ترتیب 68.37، 67.97 و 13.2 درصد می باشد.

    نتیجه گیری

    از آنجایی که جنس آلکانی ورراکس، یک باکتری بومی محیط های دریایی آلوده به هیدروکربن می باشد و توانایی آن در تجزیه زیستی نفت خام ثابت شده است، استفاده از این باکتری برای رفع آلودگی های نفتی پیشنهاد می گردد.

    سابقه و هدف
    کرم گلوگاه انار، لارو شب پره اکتومیلویس سراتونیا می باشد که از مهم ترین آفات انار محسوب می گردد. در حال حاضر هر گونه عملیات سم پاشی بر علیه این آفت فاقد نتیجه بود است. یکی از روش های کنترل استفاده از عوامل بیولوژیک مانند ایجاد تغییرات تولید مثلی توسط باکتری ولباکیا است. این مطالعه با هدف ردیابی باکتری همزیست ولباکیا در کرم گلوگاه انار و تعیین جایگاه تاکسونومیکی آن در استان فارس انجام شد.
    مواد و روش ها
    در این پژوهش به صورت تصادفی 100 نمونه از انارهای آلوده مناطق مختلف استان فارس جمع آوری گردید. پس از استخراج DNA از تخمدان حشره، به منظور ردیابی و وجود باکتری همزیست ولباکیا از روش PCR استفاده شد. پس از تعیین توالی قطعه حاصل و مقایسه آن با توالی های موجود در بانک ژن جهانی، جایگاه تاکسونومیکی ولباکیا با استفاده از نرم افزارهای ‍ClustalX وTreeView تعیین گردید.
    یافته ها
    در این مطالعه تنها یک جمعیت از شیراز آلودگی به ولباکیا را نشان داد. پس از تکثیر ژن wsp، محصول یک قطعه 513 جفت بازی بود که به دنبال تعیین توالی با شماره دسترسی KF007903 در بانک جهانی ژن ثبت گردید. با مقایسه توالی به دست آمده و توالی موجود در بانک ژنی مشخص گردید که این باکتری در زیر گروه Fur8 از گروه B متعلق به ولباکیا پیپینتیس قرار دارد.
    نتیجه گیری
    مطالعه حاضر اولین گزارش وجود ولباکیا در کرم گلوگاه انار در ایران و جهان می باشد. از آنجایی که نمونه برداری در فاصله زمانی تیر تا شهریور ماه انجام پذیرفته است، بنابراین شیوع کم باکتری ولباکیا در مناطق مورد بررسی را می توان به احتمال اثر دما به عنوان یک عامل بازدارنده رشد باکتری در حشرات نسبت داد.

    سابقه و هدف
    با توجه به سیال پذیری مورفولوژیک سیانوباکتری ها در شرایط مختلف، لازم است برای شناسایی آن ها علاوه بر مطالعات ریخت شناسی مطالعاتی در زمینه فیتوشیمی، فیزیولوژی، زیست شناسی مولکولی و ژنتیک نیز انجام گیرد. این مطالعه برای اولین بار در ایران با هدف شناسایی مولکولی سیانوباکتری لپتولینگبیا اس پی. و بررسی پاسخ های مورفولوژیک آن نسبت به پرتوهای مونوکروماتیک قرمز، سبز و آبی انجام شد.
    مواد و روش ها
    به منظور بررسی پاسخ های مورفولوژیک لپتولینگبیا اس پی. ابتدا نمونه خالص در محیط کشت مایع BG11 در شرایط اتوتروف (نور سفید)، و سپس تحت تاثیر پرتوهای مونوکروماتیک قرمز، سبز و آبی قرار گرفت. از ابتدا تا پایان تیماردهی نوع اجتماعات، سیالیت فیلامنت، بیومتری، وضعیت تریکوم، وضعیت سلول ها و تغییرات ساختاری آن ها با استفاده از میکروسکوپ نوری و الکترونی نگاره (SEM)، در مقاطع زمانی ماهیانه، هفتگی و روزانه مورد بررسی قرار گرفت.
    یافته ها
    مطالعات ساختاری و بیومتری انجام شده توسط میکروسکوپ الکترونی نگاره و نوری، حاکی از این است که سیانوباکتری مورد نظر تحت تاثیر عوامل محیطی مختلف شکل ظاهری و میزان رشد سلول های خود را تغییر داده و در نور سبز و قرمز نسبت به نور آبی سازش بهتری از خود نشان می دهد. لپتولینگبیا اس پی. در نور سبز و قرمز سلول هایی با اشکال متمایز و دارای دیواره قرمز رنگ تولید می نماید که این تاییدی بر تنوع پذیری مورفولوژیک این گونه می باشند.
    نتیجه گیری
    سیانوباکتری لپتولینگبیا اس پی. دارای سیالیت مورفولوژیک بوده و در شرایط نور مونوکروماتیک رفتارهای متفاوتی از خود نشان می دهد.

    Background and Objectives
    Tuberculosis is the second reason of deaths caused by known infectious agents. Although molecular approaches, such as PCR, are suitable techniques for detection of Mycobacterium tuberculosis (MTB), these techniques suffer of variable results in different laboratory conditions. This study aimed to design, manufacture and apply an internal PCR control used for detection of MTB.
    Materials and Methods
    To produce special MTB internal control, first individual PCR test primers was optimized for molecular detection of MTB, and then their sensitivity and specificity were measured. The composite primer for IC-MTB was also designed, replicated and colonized. The amplified IC-MTB was ligated to pTZ57R plasmid and was transformed into E. coli JM107. The minimum IC number in each PCR reaction was studied using dilution and PCR response spectrum with IC.
    Results
    The size of MTB diagnostic products with individual primers and IC-MTB were was 245 bp and 660 bp, respectively, which is a suitable difference in the size aspect. Minimum IC number was identified 1000 for each reaction. Minimum and maximum sensitivity PCR test by IC for MTB DNA was determined 10 and 10 million bacteria, respectively. No unwanted products were observed in characteristic tests by different agents.
    Conclusion
    Using an internal control as an inside control system we could detect the errors in MTB molecular diagnosis test. In fact, IC amplification is representative of correct procedure in amplification and detection steps.

    Background and Objectives

    Acinetobacter spp. is one of the most important ethiology of nosocomial infections worldwide. The presence of multiple antibiotic-resistant strains have limited effective treatments for Acinetobacter spp. The purposes of this study were to screen gyrA gene mutation in quinolone resistance clinical isolates of A. baumannii and their antimicrobial susceptibility pattern in Isfahan.

    Material and Methods

    In this cross-sectional study, 70 isolates of Acinetobacter were collected from patients hospitalized in the ICU of Isfahan Alzahra Hospital. Biochemical tests were used for detection of isolates. Antimicrobial susceptibility tests were performed on all isolates using the Kirby-Bauer Disk diffusion method and employment of eight antibiotics. Minimum inhibitory concentrations (MIC) of ciprofloxacin and levofeloxacin resistant isolates were determined using the E-test method according to the CLSI guideline. Furthermore, a PCR-RFLP test was performed to investigate gyrA gene mutation in ciprofloxacin/ levofloxacin resistance isolates.

    Results

    The most antibiotic resistance was related to ciprofloxacin (100%) and gentamicin (100%) while the lowest antibiotic resistance were observed in case of application of imipenem (8.92%) and meropenem (90%). Based on MIC test the ratio of resistance to ciprofloxacin and levofeloxacin antibiotics were 100% and 65.7%, respectively. Also, overall 66.7% of these isolates showed multidrug resistant and 93% of the isolates carry a mutation at position 83 in gyrA gene.

    Conclusion

    The present study revealed that A. baumannii isolates carry a mutation in gyrA gene. This mutation causes increases in the microbial resistance to quinolone. Rapid detection of quinolone resistant A. baumannii isolates can help physicians to determine effective treatment for these infections.

    Background and Objectives

    Pseudomonas aeruginosa is an important cause of nosocomial infections in burnt patients. Antibiotic resistance through various mechanisms is one of challenges for treatment of these patients. The mex efflux pumps play a vital role in the development of multiple resistances to antimicrobial drugs. This study aimed to analyze the prevalence of genes responsible for efflux pumps mexA-B-oprM and to investigate their phenotypes in the isolates.

    Materials and Methods

    In this cross-sectional study, 250 swabs were obtained from wounds of the patients suffering of burns levels 2 and 3 who admitted in Ghotbodin Shirazi hospital, Shiraz. Presence of P. aeruginosa isolates were confirmed by biochemical tests and PCR. Drug susceptibility, the phenotypic activity of efflux pumps and the presence of mex A, mex B genes were determined using disk diffusion method, cartwheel method and PCR methods, respectively.

    Results

    In this study, 26.40% (66 cases) of patients with burn wounds were infected with P. aeruginosa. These bacteria were resistant to all antibiotics tested except for colistin. Totally, 66.66% of the isolates (44 cases) had an efflux pump, among them 42.92% and 87.87% of the isolates carried mexA and mexB genes, respectively.

    Conclusion

    Our finding showed that genotypic method is very accurate and reliable than phenotypic methods for detection of efflux pumps in clinical isolates of P. aeruginosa. Due to presence of the efflux pump genes in more than 90% of the P. aeruginosa isolates, analysis of the presence of these genes is very important for suggestion of an effective treatment model for the patients with bacterial infection.

    Background and Objectives
    Dental caries, caused mainly by mutans Streptococci, is the most common chronic disease in the world. Employment of probiotics is a new emerged technique to prevent dental caries production, and Lactobacillus rhamnosus is one of the important common probiotics in this strategy. This study aimed to evaluate the effects of L. rhamnosus on the adhesion of oral Streptococci.
    Materials and Methods
    This cross-sectional study was carried out on 40 isolates of mutans and non-mutans Streptococci isolated from dental plaque and caries of the volunteers who admitted in dental school of Islamic Azad University, Khorasgan. The biofilm formation ability of these strains was investigated and the strongest isolates in this term were selected. The effect of L. rhamnosus (ATCC4769) on the adhesion of Streptococci was investigated by several methods; 1: an equal volum of Lactobacillus and Streptococcus, 2: application of Lactobacilli 30 minutes before induction of Streptococci into the system, 3: probiotic pellet 4: a supernatant of the overnight culture of probiotic.
    Results
    Overall, L. rhamnousus led to reduction of Streptococci adhesion to surface using all four strategies. The second method was the most effective, its effect was more efficient on S. mutans adhesion than non-mutans. There was no significant difference between third and fourth methods, but effect of both methods on S. mutans was more than other Streptococci.
    Conclusion
    Application of L. rhamnousus as probiotic could reduce the adhesion of mutans and non-mutans Streptococci to dental surfaces and therefore can reduce dental decay.

    Background and Objectives
    Beta-lactam antibiotics are currently the most common treatment for bacterial infections. The production of beta-lactamase enzymes are the most important reason of bacterial resistance to these antibiotics. The aim of this study was to investigate the presence of ampC and esbls genes in E. coli isolated from human and poultry.
    Materials and Methods
    In this cross-sectional study, 400 urine samples were collected from medical centers and also 200 swab poultry cloaca samples were collected from poultry farms located in Tehran province. Phenotypic identification of the beta-lactamase producing strains was performed using disk diffusion method. The presence of ampC and esbls genes in bacteria was studied using PCR approach.
    Results
    A total of 120 (30%) human sample and 50 (25%) poultry samples were infected to E. coli. Phenotyping evaluation showed that 54 cases (45%) of the human samples carried esbls beta-lactamase gene while 2 cases (1.67%) carried ampC beta-lactamase gene. In poultry samples, 3 cases (21.4%) were confirmed for ESBLs enzymes and none of them carried ampC gene. Based on genotyping analysis 2 (1.67%) of the strains isolated from human samples carried ampC gene.
    Conclusion
    Based on the results of this study, the ampC beta-lactamase gene was found in human samples, but more accurate studies are required for poultry. Due to high risk factor of the beta-lactamase producing organisms in nosocomial infections further studies is suggested to prevent their spread in community.

    Background and Objectives

    Oil pollutions are one of the major problems threatening environment. Biodegradation using hydrocarbon-degrading indigenous bacteria is considered as a convenient method both economically and environmentally. The genus Alcanivorax is known as a petroleum hydrocarbon degrader in petroleum-contaminated marine environments. The purpose of this study was to evaluate the capacity of Alcanivorax dieselolei, isolated from the coastal sediments of Persian Gulf for crude oil biodegradation.

    Material and Methods

    In this applied-fundamental study, first oil degrading bacteria were isolated from the coastal sediments of Persian Gulf and were enriched in media. The isolated bacterium was identified using molecular identification based on 16S rRNA sequence. The oil degrading ability of the isolates was assessed using various concentrations of crude oil.

    Results

    Based on molecular approaches the isolated bacterium was identified as Alcanivorax dieselolei. This bacterium was able to disintegrate oil droplets stably in less than one minute. Results showed that the biodegradation rates at 1, 2.5 and 5% concentrations of crude oil were 68.37, 67.97 and 13.2% respectively.

    Conclusion

    Since Alcanivorax genus is an indigenous bacterium in hydrocarbon polluted marine environments and its capability in biodegradation of crude oil has been proved, using this bacterium to remove oil pollutants is certainly possible.

    Background and Objectives
    Ectomyelois ceratoniae is one of the main pests of pomegranate. By now, no pesticides showed efficient effects on this pest. Biological control, such as changes in the reproduction ability, is a new emerged strategy for control of the population of this pest. This study was aimed to identify the symbiont bacteria in Ectomyelois ceratoniae and to determine their taxonomic classification.
    Materials and Methods
    In this study, totally 100 infected pomegranate were randomly collected from different parts of Fars providence. After DNA extraction from ovum of the insects, a PCR approach was used to identify any symbiont bacteria in this samples. Following sequencing of the products and their analysis on gene banks, the taxonomic classification of Wolbachia was determined using ClusalX and TreeView.
    Results
    In this study only one sample collected from shiraz was contaminated to Wolbachia. After amplification of wsp gene, OCR produced a 513 bp fragment which is available in gene bank under accession number KF007903. Analysis of the fragment in gene bank showed that this bacterium belonged to subclass Fur8 from group B, referred to as Wolbachia pipientis.
    Conclusion
    According to our knowledge, the present study is the first report of the presence of Wolbachia in E. ceratoniae. Since the sampling were performed through June to August, the low prevalence of the bacterium Wolbachia sp. in these area can be due to effects of temperature, which is an inhibitory factor for growth of the bacterium in the insect.

    Background and Objectives
    Due to pleomorphic feature of Cyanobacteria in different conditions, confirmation of their identification required several phytochemical, physiologic, molecular and genetic analysis after preliminary morphological analysis. This study is the first report in Iran in which was aimed to identify Cyanobacteria Leptolyngbya sp. Isc25 based on molecular approaches and to analyze their morphologic response to monochromatic red, green and blue light rays.
    Material and Methods
    In order to investigate the morphologic responses of Leptolyngbya sp. Isc25, the purified isolate of this Cyanobacterium in BG11 media were exposed first to autotrophic conditions (white light) and then to to monochromatic red, green and blue light rays. The morphological responses, flock production, structural changes, biometry and situation of trichome of Leptolyngbya sp. were evaluated using light microscopy and scanning electron microscopy (SEM) at different time periods, daily, weekly and monthly.
    Results
    Based on the morphologic and biometric analysis of the samples using light and scanning electron microscopy, the morphology and growth factor of this Cyanobacteria was affected by different environmental factors, and the best adaptation is obtained in green and red lights in comparison to blue light. Leptolyngbya sp. produced special cells with red pigments in their wall in case of exposure to green and red lights, which confirmed the variability of this species.
    Conclusion
    The results show that Leptolyngbya sp. has morphological diversity and response differently in each light condition.
