    اوتیسم، اختلالی نوروژنتیکی است که تا قبل از سه سالگی و با اختلالات عدیده در برقراری ارتباطات اجتماعی و تکلم در کودکان بروز می یابد. با این حال، اخیرا آن را نوعی اختلال چند-ژنی با تاثیرات عمیق اپی ژنتیک بر پاتولوژی آن معرفی کرده اند. بر اساس گزارشات اخیر،، خاموشی اپی ژنتیکی ژن RORa (که نقشی کلیدی در تکامل بافت عصبی دارد)، بر اثر متیلاسیون نواحی پروموتری آن در کودکان اوتیسمی رخ می دهد که قادر است بخش قابل توجهی از پاتوژنز مولکولی اوتیسم را پاسخ دهد. بر این مبنا، تایید صحت و سقم این ادعا در جمعیت اوتیسمی ایران موضوع مطالعه ی پیش رو قرار گرفت. در این مطالعه، وضعیت متیلاسیون یک توالی 136 جفت بازی از یک جزیره CpG در نواحی پروموتری ژن RORa (از 200- تا 64-) که 13 سایت CpG را دربرمی گیرد، در سلول های رده لنفوسیتی 30 کودک مبتلا به اوتیسم (دوقلوهای MZ، DZ و اوتیسم منفرد (individual)) و مقایسه با کودکان نرمال، پس از تیمار بی سولفیت سدیم بوسیله تکنیک آنالیز منحنی ذوب متیلاسیون (MCA-Meth) مورد بررسی قرار گرفت. همچنین آنالیز qRT-PCR نیز جهت بررسی رونوشت های mRNA ژن RORa و تایید نتایج آنالیز MCA-Meth مورد استفاده قرار گرفت. در این مطالعه هیچ میزانی از متیلاسیون در نواحی پروموتری بررسی شده در کودکان بیمار و سالم یافت نشد و منحنی ذوب تمامی نمونه های مورد مطالعه با منحنی ذوب کنترل غیر متیله کاملا تطابق نشان داد. همچنین آنالیز qRT-PCR نیز کاملا نتایج آنالیز MCA-Meth را تایید کرد. متیلاسیون نواحی پروموتری ژن RORa را نمی توان به عنوان یک ریسک فاکتور اپی ژنتیکی عمومی در همه ی مناطق جغرافیایی قلمداد کرد و احتمالا سوال بزرگی را در پاتوژنز مولکولی اوتیسم پاسخ نمی دهد، حداقل در جامعه اوتیسمی ایران.

    شیوع سیاه زخم طبیعی در انسان نادر است اما از آنجایی که این باکتری پتانسیل بالایی جهت استفاده به عنوان سلاح بیولوژیکی دارد تلاش ها برای تولید واکسن علیه این باکتری رو به افزایش است. ترکیب آنتی ژن ها با زیرواحد B کلرا توکسین CTB می تواند پاسخ آنتی بادی موکوزی قوی ایجاد نماید و به عنوان واکسن معرفی شود. در این مطالعه، ابتدا توالی های مربوطه از پایگاه داده NCBI اخذ و پس از بهینه سازی، از لینکری مناسب جهت اتصال توالی ها استفاده شد. نیمه عمر و شاخص ناپایداری پروتئین تعیین و در ادامه پیش بینی ساختارهای ثانویه و نیز ساختار سه بعدی صورت گرفت. در انتها اپیتوپ های خطی سلول های B نیز تعیین شدند.کدون ها به کدون های رایج در باکتری اشریشیاکلی تغییر کرد و محتوی C+G به میزان بهینه تغییر داده شد. ساختارهای فضایی نامناسب در ساختمان دوم mRNA شکسته شد که این تغییرات منجر به طولانی شدن نیمه عمر mRNA و افزایش قابل ملاحظه بیان پروتئین نوترکیب گردید. ساختارهای ثانویه و نیز ساختار سه بعدی نیز تعیین گردید. لینکر EAAAK به خوبی توانست از تداخل دومین های پروتئینی ممانعت نماید. بررسی ها در In silico نشان داد با توجه به تغییراتی که در ژن کد کننده این پروتئین صورت گرفته، توالی نوکلئوتیدی طرحی شده می تواند از بیان بالایی در میزبان E.coli جهت کلون بیان برخوردار باشد. ساختارهای سه بعدی پیش بینی شده با نرم افزارهای آنلاین ارزیابی شده و در ادامه بهترین ساختارها انتخاب گردید. توالی کایمر بهینه شده می تواند کاندیدای مناسبی جهت بررسی های ایمنی زایی به منظور تهیه واکسن موثر در نظر گرفته شود.

    روتئین GRP78 مارکر مهمی برای استرس شبکه آندوپلاسمی به شمار می رود. مرگ سلولها بواسطه استرس شبکه آندوپلاسمی نقش مهمی در پاتوفیزیولوژی تخریب در بیماری های مرتبط با تخریب شبکیه بازی می کند. از سوی دیگر، از جمله مارکرهای التهابی دخیل در فرایند پاتوژنز این بیماری ها می توان به CRP، CFH، C3، CR1، C5، کاسپاز-4، کاسپاز –S 12، MCP، DAF، MIRLاشاره نمود. هدف از این مطالعه بررسی ارتباط احتمالی بین این دو پدیده یعنی استرس شبکه آندوپلاسمی و التهاب از طریق طراحی شبکه ارتباطی مابین GRP78 و فاکتورهای التهابی است. شبکه حاصل از متن کاوی و جستجوی اطلاعات مربوط به میانکنش های پروتئین-پروتئین با داده های بیانی بازیابی شده مربوط به پروتئین های مورد نظر در بافت شبکیه در انسان با استفاده از نرم افزار Cytoscape ادغام گردید. ماژول های ساختمانی و عملکردی احتمالی شبکه با استفاده از دو افزونه jActiveModules و ClusterONE شناسایی شدند. با استفاده از افزونه BiNGO هریک از ماژولهای شناسایی شده از نظر هر گونه فعالیت زیستی مشترک مورد بررسی قرار گرفتند. از میان ماژولهای شناسایی شده با استفاده از داده های بیانی و مکان شناسی، 9 ماژول حاوی محتوای عملکردی مشترک هستند که به عنوان یک فراگره تعریف شدند. در نهایت شبکه ساده شده ای بدست آمد که روابط مابین GRP78 را با فاکتورهای التهابی انتخابی و نیز پروتئین های حدواسط آنها را نمایش می دهد. وجود شبکه پیش بینی شده ارتباطی مابین GRP78 به عنوان مارکر کلیدی فرایند استرس شبکه آندوپلاسمی با 10 فاکتور التهابی مطرح در فرایند بیماریزایی بیماری های مرتبط با تخریب شبکیه کندوکاو شد و بوسیله داده های موجود ترسیم آن امکانپذیر گردید.

    ژن های سرطان بیضه (CTAs) زیر گروهی از آنتی ژنهای مرتبط با تومور (TAA) می باشند که در سلول های لایه زایا بیضه، تخمدان و تروفوبلاست و در انواع مختلفی از سلول های سرطانی بیان می شوند. عملکرد بیولوژیکی ژنهای CT در لایه زایا و تومورها تاکنون به درستی شناخته نشده است. یکی از مهمترین CTAs، ژن سرطان بیضه 100 (CT100) می باشد که به نامDPPA2 نیز شناخته می شود. هدف ما در این طرح جداسازی، تکثیر و همسانه سازی ژن DPPA2 بوده است. بدین منظور در ابتدا رده های سلولی بیان کننده ژنDPPA2 شناسایی شدند. سپس توسط پرایمرهای طراحی شده، در شرایط بهینه، وا زه ای از رو س (RT-PCR) انجام گرفت. در ادامه ژن DPPA2 از روی ژل آگارز استخراج، در وکتور pTZ57R/T کلون، و سپس به میزبان (TOP10F'') E.coli انتقال داده شد. در نهایت صحت کلنی های نوترکیب، توسط روش کلنی PCR، هضم آنزیمی و تعیین توالی مورد بررسی قرار گرفت. بر اساس نتایج حاصله، ژن DPPA2 از رده سلولی SKOV3 جداسازی و تکثیر شد و پس از همسانه سازی در وکتور pTZ57R/T، کلنی های نوترکیب توسط کلنی PCR، هضم آنزیمی و تعیین توالی مورد تایید قرار گرفتند. با توجه به اهمیت ژن DPPA2 در روند ایجاد سرطان، شناسایی عملکرد این ژن سرطان بیضه دارای اهمیت بسزایی است که جداسازی و همسانه سازی این ژن را حائز اهمیت می سازد.

    عامل نورون زایی مشتق شده از مغز1 (BDNF) زیرمجموعه ای از خانواده ی عوامل نورون زایی است که به مقدار زیاد در سیستم عصبی به خصوص هیپوکامپوس و پوشش مغز وجود دارد. این پروتئین در ابتدا 247 اسیدآمینه دارد که در پردازش های بعدی بخشی از آن بریده شده و تنها 119 اسیدآمینه باقی می ماند. پروتئین BDNF بالغ به بیرون یاخته تراویده شده و به گیرنده های تروپومیوزین کیناز B متصل می شود. اتصال به گیرنده سبب راه اندازی دسته ای از آبشارهای داخل یاخته ای از جمله 2 Ras-MAP کیناز می شود. این آبشارها در رشد و تمایز نورون ها و همچنین ایجاد سیناپس نقش دارند. هدف از این پژوهش همسانه سازی و بیان ناحیه عملکردی پروتئینBDNF در اشریشیاکلای و سویه BL21 بود. توالی ژن مربوط به این بخش دارای 357جفت باز است که به صورت سنتز شده درون ناقل PUC57 تهیه شد. این ناقل به درون باکتری ترانسفورم شد و به میزان زیاد تکثیر شد. ناقل تکثیرشده پس از تیمار با آنزیم های محدودکننده ی Xho1 و BamH1 درون ناقل pET26bقرار گرفت. در مرحله ی بعد ناقل نوترکیب درون باکتری اشریشیا کلای و سویه 10 top ترانسفورم شد و به میزان زیاد تکثیر یافت. سپس ناقل نوترکیب استخراج و جهت بیان ژن مورد نظر به داخل باکتری اشریشیاکلای سویه Bl21 ترانسفورم شد. آنالیز SDSPAGE بیان پروتئین در باکتری را تایید کرد.

    در این مطالعه، پاسخ های القا شده تنش سرما (چهار درجه سانتی گراد) در دو ژنوتیپ متحمل (Sel 11439) و حساس (ILC533) نخود زراعی (Cicer arietinum L.) به کمک روش های بیوشیمیایی و مولکولی بررسی شد. نتایج تجزیه آماری نشان داد که اختلاف معنی داری بین ژنوتیپ ها در تحمل به سرما وجود دارد. در روز ششم تنش، میزان MDA در ژنوتیپ حساس به بالاترین سطح خود رسید اما تجمع کمتری از این ترکیب در ژنوتیپ متحمل مشاهده شد. میزان تجمع آلفا توکوفرول و فنل کل تولید شده در گیاهان در روز ششم تنش سرما افزایش یافت طوری که این افزایش در ژنوتیپ متحمل بسیار بیشتر از میزان آن در ژنوتیپ حساس بود. در ژنوتیپ متحمل و حساس با طولانی شدن دوره تنش میزان بیان ژن لیپوکسیژناز (درگیر در مسیر اکسی لیپین) افزایش یافت طوری که در ژنوتیپ حساس این افزایش در روز سوم و ششم تنش به میزان بالاتری نسبت به ژنوتیپ متحمل بود. نتایج نشان داد که افزایش بیش از حد مسیر اکسی لیپینی که به تولید اسید جاسمونیک و القا ژنهای دفاعی می انجامد به نفع سلول نبوده و در مواردی خسارت بیشتر به غشا سلول وارد می کند که افزایش میزان MDA در روز شش تنش سرما در ژنوتیپ حساس تایید کننده آن بود. به نظر می رسد گیاهان متحمل با مکانیسم های ترجیحی، از عوامل دفاعی متعددی در پاسخ به تنش استفاده می کنند. عدم تغییر معنی دار در میزان پراکسیداسیون لیپیدی تحت تنش سرما در ژنوتیپ متحمل به معنای عدم افزایش خسارت تلقی می شود که در اثر فعالیت گروهی مکانیسم های دفاعی مانند آلفاتوکوفرول، فنل و لیپوکسیژناز ایجاد شده است.

    نروتروفین ها، فاکتورهای رشد دستگاه عصبی هستند، هر چند مکانیسم های زیربنایی این اثرات نامشخص است. عامل عصبی تغذیه ای مشتق از مغز (BDNF)، یک نروتروفین حیاتی برای یادگیری و حافظه، انعطاف و سلامت نرونی است و نقش مهمی در کنترل اختلالات نرونی و بیماری های ژنی مانند آلزایمر، پارکینسون، افسردگی شدید، صرع و... دارد هرچند اطلاعات در این زمینه متناقض است. با اینحال گزارش شده است که مغز به طور قابل توجهی به ورزش پاسخ می دهد و باعث ایجاد تغییراتی در سطوح آناتومیکی، سلولی و مولکولی بدن می شود. یافته ها، نشان می دهد که ورزش و فعالیت، به حفظ عملکرد شناختی و ساختار کلی مغز به خصوص بر اثر افزایش سن کمک می کند. انعطاف مغز که به وسیله BDNF تعدیل می شود، می تواند مبنای تاثیرات مثبت ورزش روی سلامت مغز باشد. همچنین، گزارش شده که ورزش بقای نرونی را بهبود می بخشد و مقاومت در برابر بیماری های پاتوژنی را افزایش می دهد. این مقاله به نقش اثرات متفاوت ملکولی BDNF و تاثیر ورزش بر روی بیان این فاکتور و کنترل بیماری های مرتبط می پردازد.

    در دهه های اخیر همزمان با پیشرفت ژنتیک ملکولی تحقیقات زیادی برای شناسایی ژنها و یا قطعات کروموزومی موثر بر صفات مهم اقتصادی در حیوانات اهلی انجام شده است. علیرغم پژوهش های متعددی که در ارتباط با انتخاب به کمک نشانگر (MAS) انجام شد، به دلیل محدودیت های این روش کاربرد های آن محدود و افزایش پیشرفت ژنتیکی کم بوده است. با تکمیل موفقیت آمیز توالی یابی ژنوم انسان و پیشرفت های عظیم در تکنولوژی ژنوتیپ کردن چندشکلی های تک نوکلئوتیدی (SNP) سطح ژنوم با هزینه کم و سرعت و دقت بالا، این امکان فراهم شده است که بتوان انتخاب بر پایه نشانگر را در یک مقیاس ژنومی استفاده نمود که به این روش، اصطلاحا انتخاب ژنومی گفته می شود. انتخاب ژنومی با استفاده از نقشه های نشانگری با تراکم زیاد، بخصوص SNPهایی که کل ژنوم را شامل می شوند و اغلب در عدم تعادل پیوستگی با QTL های مجاور خود هستند، ارزش ژنتیکی کل را پیش بینی می کند. محاسبه ارزشهای اصلاحی ژنومی نیازمند برآورد اثرات نشانگرها در شماری از حیوانات یک جمعیت بوده که هم دارای اطلاعات فنوتیپی و هم دارای ژنوتیپ نشانگرها هستند و به آنها جمعیت مرجع گفته می شود. سپس اثرات برآوردی نشانگرها در جمعیت مرجع برای محاسبه ارزش اصلاحی ژنومی حیوانات کاندید انتخاب در جمعیت تایید که فقط دارای اطلاعات نشانگری هستند، استفاده می شوند. بر اساس مطالعات انجام شده عوامل موثر بر صحت ارزیابی های ژنومی توارث پذیری صفت، اندازه جمعیت مرجع، درجه خویشاوندی کاندیداهای انتخاب با جمعیت مرجع، عدم تعادل بین نشانگرها و QTL ها، روش مورد استفاده در برآورد اثرات نشانگرها و توزیع اثرات QTL ها هستند. به طور خلاصه از مزایای انتخاب ژنومی نسبت به انتخاب سنتی می توان به کاهش فاصله نسل و افزایش صحت ارزیابی ها اشاره کرد که علاوه بر کاهش هزینه ها سبب افزایش پیشرفت ژنتیکی می شوند. از طرفی روش انتخاب ژنومی از نشانگرها برای برآورد دقیق واریانس نمونه گیری مندلی استفاده کرده و از این طریق باعث کاهش نرخ همخونی نسبت به روش های سنتی انتخاب می شود.

  • مروری بر فعالیت و ساختار مولکولی نوروتروفین ها

    نوروتروفین ها، مهم ترین عوامل تروفیکی شناخته شده در سیستم عصبی بوده که خانواده برجسته ای از عوامل رشدی پلی-پپتیدی می باشند و از فاکتورهای اصلی تعدیل رشد و توسعه سیناپسی هستند. همه نوروتروفین ها یک ساختار اصلی رایج دارند اما در نواحی خاص عمل می کنند و به گیرنده های ویژه متصل می شوند، در نتیجه عملکردهای متفاوتی نشان می دهند و سرانجام در مسیر بقا یا آپوپتوز قرار می گیرند. یک راه برای افزایش فعالیت نوروتروفین ها در سیناپس ها و نورونهای مختلف، افزایش تعداد گیرنده های آن ها در سطح سلول است. اصلی ترین عضو این خانواده- عامل عصبی تغذیه ای مشتق از مغز- توسط نورونها به ویژه نورونهای هیپوکمپ و کورتکس که در یادگیری و حافظه درگیر هستند، ساخته می شود و بیان آن توسط نوروترانسمیترهای مرکزی و هورمون های محیطی تنظیم می شود. بسیاری از توانایی های مغز برای سازگاری یا تغییر، در پاسخ به تجربه یا محیط، به انعطاف نرونی و ارتباط های سیناپسی مرتبط است. هرچند، مکانیسم های مولکولی که الگوهای فعالیت نورونی را برای ایجاد تغییرات ویژه سیناپسی ترجمه می کنند ناشاخته است.

  • ژن درمانی در بیماری مالتیپل اسکلروزیس (MS)

    بیماری مالتیپل اسکلروزیس (MS) یک بیماری خودایمن مربوط به دستگاه عصبی مرکزی است و سوئ عملکرد دستگاه سیستم ایمنی در آن سبب دمیلینه شدن عصب و ناتوانی نورولوژیکی پیشرونده می شود. برای درمان بیماری از روش های ضد التهابی وحفاظتی استفاده می شود. با این وجود استراتژی های حفاظتی دربیماری به خوبی شناخته نشده اند و استفاده از روش های ضد التهابی به صورت سیستمیک هم به دلیل وجود سد خونی مغزی اثربخشی مناسبی ندارد. مطالعات اخیر ژن درمانی، هدفگیری در جایگاه التهاب را پیشنهاد می دهد که در حال حاضر به صورت پیش بالینی در آنسفالومیلیت اتوایمیون آزمایشگاهی (EAE)، یک مدل حیوانی از MS شروع شده است. با این وجود آزمایشات بالینی برروی انسان در زمینه ژن درمانی بیماری های خودایمن هنوز در مراحل اولیه می باشد. برای مثال در مطالعات پیش بالینی، یک وکتور پلاسمیدی به نام BHT-3009 بوسیله ایجاد تغییراتی در پلاسمید pVAX1 ساخته شد. با استفاده از این پلاسمید، اولین تلاش های موفقیت آمیز واکسن های DNA در تعدیل پاسخ ایمنی در افراد انسانی گزارش شد. بعد از آن این روش در نوع عودکننده بیماری ام اس (RRMS) و یا بیماران با ام اس پیشرونده ثانویه مورد بررسی قرار گرفت. افرادی که BHT-3009 را دریافت کرده بودند کاهش مشخصی در IFN- γ و سلول های T CD4+ واکنشگردر خون محیطی نشان دادند. همچنین در فاز II اقدامات بالینی، بیماران MS تحت درمان با دارونما و دو دوز مختلف از BHT-3009 قرار گرفتند. در بیماران دریافت کننده ی دوز، در مقایسه با دارونما، به میزان 61% آسیب ها در افرادی که با 5/. میلی گرم از BHT-3009 تیمار شده بودند کاهش یافته بود. و میزان آسیب های پیشرونده در 48 هفته با استفاده BHT-3009 در مقایسه با دارونما به میزان 51 درصد کمتر بود و همچنین در این بیماران کاهش قابل توجهی در سطح اتوآنتی بادی های مشاهده شد. بنابراین حدس زده می شود که BTH-3009 می تواند به عنوان یک درمان امیدوارکنند در MS مورد توجه قرار بگیرد اما با این همه چنین مواردی نیازمند بررسی دقیق تری می باشد.

    بیماری های عفونی ناشی از میکروارگانیسم ها همچنان به عنوان علل عمده مرگ در سراسر جهان مطرح بوده و افزایش تعداد سویه های مقاوم آنها باعث نگرانی زیادی شده است. از زمان کشف خاموشی ژن توسط RNA مداخله گر (RNAi)، انقلابی در زمینه کاربرد درمانی آن به راه افتاده است. با به کارگیری این روش از تکثیر و پیشرفت عفونت های ویروسی، قارچی، باکتریایی و انگلی جلوگیری به عمل آمده است. برای مثال مهار عفونت ویروس نقص ایمنی انسانی (HIV)، ویروس هپاتیت C (HCV)، ویروس سارس، هپاتیت (B (HBV و همینطور ویروس کشنده ابولا و نیز مهار باکتری های استافیلوکوک آرئوس مقاوم به متی سیلین (MRSA)، مایکوباکتریوم توبرکلوزیس، فرانسیسلا تولارنزیس و همچنین قارچ های بیماریزای کریپتوکوکوس نئوفرمنس، آسپرجیلوس فلاووس، کاندیدا آلبیکنس و نیز انگل های پروتوزوئری و کرمی را می توان نام برد. درمان ژنی بر اساس RNAi روز به روز جالب تر و امیدبخش تر شده است. این مطالعه مروری به جنبه های درمانی پدیده RNAi و عمدتا جنبه درمانی آن در درمان بیماری های عفونی انسان پرداخته است.

  • الگوریتم های خوشه بندی دوبعدی در داده های بیان ژنی: یک مطالعه مروری

    در سال های اخیر تکنولوژی ریز آرایه امکان مشاهده و بررسی بیان هم زمان هزاران ژن را وقتی سلول تحت شرایط مختلف یا پروسه های متفاوت قرار می گیرد را فراهم کرده است و نقشی اساسی در سرعت بخشیدن و بالا رفتن کارایی مطالعات بیان ژنی ایفا می کند. یکی از مهم ترین تکنیک های تشخیص الگو در داده های بیان ژنی تحلیل خوشه بندی دوبعدی است که می تواند گروه هایی از ژن ها با الگو های بیان شده مشابه را بیابد. مطالعه حاضر که یک مطالعه مروری است با هدف ارزیابی پژوهش های موجود در زمینه خوشه بندی دوبعدی بیان ژن و بررسی الگوریتم های مختلف، انجام شده است. در این مطالعه الگوریتم های معرفی شده در سال های 2000 تا 2014 میلادی مورد ارزیابی قرار گرفته است. نتایج جستجو در سایت های مختلف شامل 16 الگوریتم اصلی بود که همه آن ها به زبان انگلیسی چاپ شده بودند. با استفاده از نتایج به دست آمده از این مطالعه، بهترین الگوریتم های موجود مربوط به الگوی داده کاوی رابطه بود. اما این تکنیک به این دلیل که از روش جستجوی فراگیر استفاده می کند و همه فضاهای مربوط به خوشه های دوبعدی را پیدا می کند لذا بسیار وقت گیر و پرهزینه است. به نظر می رسد نیاز است بر روی سایر الگوریتم های ارائه شده راه کارهایی جهت رفع مشکلات موجود پیاده کرد تا بتوان نتایج قابل قبول تری به دست آورد.

    همچنانکه جمعیت دنیا و تقاضا برای محصولات کشاورزی رو به افزایش است، نیاز به فراهم سازی و کشف راه های جدید در افزایش و ارتقا کمیت و کیفیت محصولات کشاورزی وجود دارد. بکارگیری روش های شیمیایی برای افزایش کمیت محصولات، سبب تغییرات منفی بر روی ویژگی های خاک، آب و اتمسفر و سمیت آنها شده است. به طوریکه شرایط نامناسبی برای رشد و نمو گیاهان فراهم آمده است. بنابراین محققان تمرکز ویژه ایی بر بکارگیری روش های بیوفیزیک داشته اند. اگرچه مکررا از لیزر برای اهدف های صنعتی و پزشکی استفاده شده، اما استفاده از لیزر در بیولوژی از دهه1960آغاز شد. لیزر تقویت نشر نور برانگیخته بوده که دارای ویژگی های منحصر به فرد و اثرات چشمگیری است. نتیجه ی اثرات لیزر افزایش، پتانسیل و انرژی بذر، مقاومت گیاه، فعالیت آنزیم ها، نرخ جوانه زنی، بنیه بذر، فعالیت رنگدانه ای، فتوسنتز، سنتز DNA و RNA، متابولیسم و تسریع رسیدگی است. مکانیسم لیزر هنوز به خوبی شناخته نشده اما چند فرضیه در نظر گرفته شده از جمله اثر بر زنجیره انتقال الکترون در میتوکندری و سیستم رنگدانه ای فیتوکروم. اغلب منابع در دسترس در مورد تاثیر لیزر بر افزایش محتوای ATP سلول هم نظر هستند. اختلاف اثرات لیزر ناشی از تفاوت جذب نور توسط گیاه است. عوامل گوناگونی وجود دارد که بر نتایج اثرات لیزر تاثیر می گذارد. اول شرایط سلول و گیاه است و سپس ویژگی های فیزیکی لیزر. هدف این مقاله بررسی و ارزیابی مکانیسم ها، اثرات و کاربرد های لیزر کم توان بر روی سلول ها است. هم چنین ما لیزر کم توان را به عنوان ابزار جانبی برای باروری گیاهان در کشاورزی و بیوتکنولوژی معرفی می کنیم تا به اهداف عنوان شده دست پیدا کنیم.

  • راهبردها و چالش های بیوتکنولوژی در تولیدات دامی کشورهای در حال توسعه

    بیوتکنولوژی در تولید حیوانی به فنآوری های بیولوژیکی، شیمیایی و فیزیکی تقسیم می شود که می تواند از جنبه های مختلف نظیر تولیدمثل، اصلاح نژاد، تعذیه و بهداشت بر کیفیت و کمیت تولید موثر باشد. این تکنیکها در کشورهای در حال توسعه به دلیل زیرساختهای ضعیف، ظرفیت های آموزشی و تکنیکی ابتدایی و رشد اقتصادی کم، کمتر استفاده شده است. افزایش رشد جمعیت و نیاز به مواد غذایی بیشتر در کشورهای در حال توسعه سبب ضرورت استفاده از بیوتکنولوژی در این کشورها شده است، زیرا سبب افزایش سرعت تولید و کاهش هزینه های محصولات دامی می شود. این امر سبب افزایش تمایل به سرمایه گذاری در بخش دامپروری می شود که به نوبه خود سبب افزایش تقاضا برای زمین و در نتیجه افزایش قیمت آن، افزایش دستمزد و خدمات غیرکشاورزی، افزایش قدرت خرید فقرا و رشد اقتصادی می شود. همچنین بیوتکنولوژی روش هایی مهیا می کند که سبب تعادل بین بهره برداری اقتصادی از منابع زیستی و حفظ آنها برای آینده می شود. به نظر می رسد در برخی کشورها بیوتکنولوژی بیش از ظرفیت آنها برای ایمنی استفاده شده است. این مهم نیازمند فرآیندهای سیاسی و تنظیمی موثر بر بیوتکنولوژی و محصولات آن است که در یک رقابت حرفه ای، دارای بالاترین استاندارد باشند. در این مرور اثر بیوتکنولوژی بر بهبود تولیدات حیوانی در کشورهای در حال توسعه و همچنین فرصت ها و چالش های آن بررسی شده است.

  • اپیژنتیک: چالشی جدید پیشروی اصلاح نژاد دام

    با وجود پیشرفت های صورت گرفته در زمینه ارزیابی های ژنتیکی به دلیل استفاده از اطلاعات ژنومی، ورود اطلاعات اپیژنتیک در این ارزیابی ها ضروری به نظر می رسد. با پیدایش علم اپیژنتیک و پیشرفت های صورت گرفته در آن، مشخص گردیده که بخش بزرگی از عوامل محیطی (شاید حتی تمام آن) از طریق اپیژنتیک بر فنوتیپ حیوان تاثیر گذار است. اپیژنتیک مجموعه تغییرات در ساختارهای کروماتینی است که به طور تجمعی الگوهای متفاوتی از بیان ژنی و خاموشی را در ژنوم با توالی یکسان ایجاد نموده، که طی تقسیمات میوزی و میتوزی منتقل می گردد. هدف این مقاله، توضیح مکانیسم های ایجاد کننده تنظیمات اپیژنتیکی، بحث پیرامون کاربرد اپیژنتیک در اصلاح دام و فرصت های جدید ایجاد شده به وسیله آن می باشد. همچنین در این مقاله سه حالت اپی آللی در مدل های اپیژنتیکی که شامل اپی آلل های دائمی، موقتی و بین نسلی می باشند، پیشنهاد می شوند. به علاوه، در این مطالعه با بررسی تحقیقات صورت گرفته در منابع مختلف مدلی ارائه گردیده که در آن چگونگی ارتباط بین اپیژنتیک، ژنتیک و محیط در سطح ملکولی را توضیح می دهد. با توجه به این مدل ارائه شده، استفاده از اثرات متقابل بین محیط، ژنتیک و اپیژنتیک می تواند باعث بهبود مدل-های پیش بینی ارزش های اصلاحی حیوانات شده، و صحت پیش بینی ها افزایش می یابد.

    Autism which occurs before age 3 is a neurogenetic disorder with a variety of impairments in social communication and speech development of children. However، recently it has been introduced as a multi-gene disorder with profound epigenetic impact on its pathology. According to the recent reports، epigenetic silencing of RORa gene (which has essential role in neural tissue development) that happen due to the methylation of its promoter region in autistic children، can explain a significant portion of the molecular pathogenesis of autism. Therefore، in current study the accuracy of this opinion was evaluated in Iranian autistic population. Methylation status of a 136 bp sequence of a CpG island in the promoter regions of RORa gene (from -64 to -200) which encompasses 13 CpG sites was examined in lymphoma cells of 30 children with autism (autistic individuals، monozygotic and dizygotic twins) and after sodium bisulfite treatment، through the Melting Curve Analysis-methylation assay (MCA-Meth) compared with normal children. Also، qRT-PCR analysis was used for evaluation of mRNA transcripts of RORa gene and confirmation of MCA-Meth analysis. In this study، no amount of methylation was found in evaluated promoter regions of healthy and unhealthy children and melting curves of all studied cases matched perfectly with unmethylated melting curves of controls. Also، qRT-PCR analysis completely confirmed the results of MCA-Meth analysis. Methylation of RORa promoter regions cannot be considered as a general epigenetic risk factor in all geographic areas and probably don’t respond any significant question about molecular pathogenesis of autism، at least in the autistic population of Iran.

    Natural anthrax is rare among humans، but since this bacterium has the potential to be used as a biological weapon، efforts to produce a vaccine against the bacteria are increasing. Combination of the Cholera toxin B subunit (CTB) with other antigens can produce a strong antibody response and be used as a vaccine. This study aimed to link the gene sequence ctB and paD4 and predicate three-dimensional structure of the Chimeric protein. In this study، first، Nucleotide sequences were obtained from NCBI database and optimized. Sequences were fused together by appropriate amino acid linker in order to find the best epitope exposing chimeric antigen. After Protein half-life and instability index were determined، and then the prediction of the secondary structure and the three-dimensional structure was analyzed. Finally immunodominant linear B cells were also settled. Codons were changed with respect to codon bias of E. coli and the content of C + G was converted to an optimal level. Inappropriate structures of RNA were removed. Howerver، These changes led to a prolonged half-life and increased mRNA expression of the recombinant protein. Secondary structures and three-dimensional structure was specified. EAAAK linker could prevent the protein domain interactions well. Due to the changes over gene encoding of this protein، Silico analysis represented that Nucleotide sequence can have a high expression in E. coli. The predicted Three-dimensional structures of proteins were evaluated by online software and the best one was choosen. Optimized chimeric sequence can be a good candidate in immunogenicity study to produce an effective vaccine.

    GRP78 is an important endoplasmic reticulum stress marker. Apoptosis through endoplasmic reticulum stress plays an important role in the pathogenesis of retinal degenerative diseases. On the other hand، inflammatory factors، CRP، CFH، C3، CR1، C5، Caspase-4، Caspase-12S، MCP، DAF and MIRL are implicated in the pathogenesis of these diseases. The aim of this study was to identify a possible association between these two phenomena; endoplasmic reticulum stress and inflammation through an investigation into a possible relationships between GRP78 & inflammatory factors. The resulted network from text-mining and protein interaction databases querying was integrated with retrieved expression data of desired proteins in human retinal tissue using Cytoscape. Putative complexes were identified using ClusterONE and jActiveModules plugins. The common biological function of each identified modules were assessed using BiNGO plugin. Among the identified modules، using expression and topological data، 9 modules which contained common biological themes were defined as a meta node. Thus، the simplified network illustrated the relationship between GRP78، inflammatory factors and other intermediate proteins. A hypothesized communication network between GRP78 as a marker for endoplasmic reticulum stress and ten key inflammatory factors associated with retinal degenerative diseases was investigated.

    Cancer/testis antigens (CTAs) are a subgroup of tumor-associated antigens which are expressed in trophoblast، ovary and testis germ line cells. CTAs are also expressed in a variety of cancer cells and their biological function in germ line and tumors cells is unclear yet. One of the most important CTAs is CT100 which is also known as DPPA2. The aim of this study was isolation، amplification and cloning of DPPA2 coding sequence. Having examined the gene expression pattern of different cell lines، we amplified coding sequence of DPPA2 gene and cloned it in pTZ57R/T vector. Recombinant colonies were analyzed and confirmed by colony PCR، double digestion and sequencing. Considering the importance of DPPA2 roles in the development، its role analysis in carcinogenesis seems valuable. Our results prepare a suitable platform to analyze its biological function.

    Brain-derived neurotrophic factor، also known as BDNF، is a member of the neurotrophic family of growth factors. The BDNF protein is present in high concentration in hippocampus and cerebral cortex of brian. The precursor protein contains 247 amino acids and in the later process the protein is cleaved to yield the 119 amino acid residue BDNF. This mature BDNF is secreted to extracellular sites and bind to TrkB receptor. Interaction of BDNF to TrkB can lead to a variety of intracellular signaling cascades such as Ras-MAP kinase cascade. These cascades help to keep the survival of existing nerve cells، and stimulate the growth and differentiation of new neurons and their synapses. The aim of this research was to clone the functional BDNF coding sequence in a prokaryotic vector to produce BDNF in E. coli (BL21). Functional part of protein، containing 357bp، was synthesised into PUC57 plasmid. This plasmid transformed in E. coli (DH5) for amplifying purposes. Then the plasmids were extracted and treated by BamH1 and Xho1 restriction endonucleases and the 372bp fragments were inserted into the BamH1-Xho1 site in pET26b vector. At the next step، the recombinant vector was transformed into TOP10 E. coli cells for amplifying purposes. The recombinant plasmid was extracted and transformed into BL21 E. coli cells for gene expression. The expression of the recombinant protein was confirmed by SDS-PAGE technique.

    In this study، we analyzed cold-induced responses (4°C) in tolerant (Sel11439) and susceptible (ILC533) chickpea (Cicer arietinum L.) genotypes using biochemical and molecular methods. The data analysis revealed that there were significant differences between genotypes in their cold tolerance. On the sixth day of cold stress، MDA content in susceptible plants was maximum whereas less accumulation of MDA was observed in tolerant plants. Total phenol and α-tocopherol also increased on the sixth day of cold stress effectively in tolerant genotype compared to susceptible one. However، the longer the cold period، the level of lipoxygenase gene expression (involved in oxylipin pathway) in both genotypes increased so that this increase on the third and sixth day of cold stress in susceptible genotype was more compared to tolerant genotype. The results showed that the high activity of oxylipin pathway which produce jasmonic acid and induce defense genes may cause cell membrane damage under cold stress so that the increase of MDA content on the sixth day of cold stress in susceptible genotype confirmed these results. It seems that using preferred mechanisms in tolerant plants، triggered several defense factors in response to cold stress. Non-significant change of MDA content in tolerant genotype during cold stress which showed no cell damage، was induced by several defense mechanisms like total phenol، α-tocopherol and lipoxygenase activity.

    Neurotrophies، are the major growth factors in the nervous system، although the exact mechanisms underlying these effects have been remained obscure. Brain Derived Neurotrophic Factor (BDNF) is a critical neurotrophine for learning، memory، and neuronal plasticity. Moreover، it exerts an important role in controlling neuronal disorders and genetic diseases such as Alzheimer، Parkinson، depression، epilepsy etc. Although there are several conflicting data in this respect، it has been reported that brain considerably responds to exercise and results in significant changes in the anatomical، molecular and cellular level of body. The findings suggest that exercise and activity helps to maintain cognitive function and brain structure، especially due to aging. Brain plasticity that is controlled by BDNF can be the basis of the beneficial effects of exercise on brain health. The present study discusses different molecular effects of BDNF and the effect of exercise on the expression of this factor and controlling related diseases.

    In recent decades، along with the developments of the molecular approaches، many researches were done to detect genes or chromosome segments that affect the economical traits of domestic animals. Notwithstanding extensive researches towards marker assisted selection (MAS) method، there has been limited implementation of MAS and minor increases in genetic gain due to its several limitations. The successful completion of the human genome sequence and huge developments regarding high-throughput assays for genotyping single nucleotide polymorphisms (SNP) across the genome has made the possibility to apply MAS in genomic scale which is called genomic selection. Genomic selection predicts the total genetic values using genome-wide dense marker maps so that all QTLs are in linkage disequilibrium with at least one marker. Calculation of genomic breeding values requires estimating SNP effects from a subset of animals in the population with information on both phenotypes and markers genotype called the reference population. Estimated markers effects in the reference population is then used to calculate genomic breeding values of selection candidate individuals in evaluation population which has only markers information. According to the literature، factors affecting accuracy of genomic evaluations was the heritability of the trait، the size of the reference population، extent of relationships between selection candidates and the reference population، the method used to estimate marker effects، linkage disequilibrium among markers and QTLs and the distribution of QTLs effects. In a nutshell، genomic selection have many advantages over traditional methods including an increase in accuracies of breeding value prediction، shortening the generation interval for many traits and thus improving the rate of genetic gain and decrease evaluation costs. Furthermore، genomic selection by using genetic markers better predict Mendelian sampling term of breeding values and decrease inbreeding rate compared to traditional selection.

  • A Review on The Function and Molecular Structure of Neurotrophins

    Neurotrophins have been recognized as the main trophic factors in the nervous system. They are a broad family of polypeptide growth factors and the main factors responsible for modulating of synaptic development. They share a common basic structure but in certain areas which connect to their specific receptors. Thus، they exert different performances triggering survival or apoptosis pathway. One way to increase the activity of neurotrophins in different synapses and neurons is increasing the number of respective receptors on the cell surface. The main member of this family is Brain-Derived Neurotrophic Factor which is constructed by neurons especially those located in hippocampus and cortex which are involved in learning and memory. Thereby، its expression is regulated by central neurotransmitter and peripheral hormones. Many of the brain''s abilities to adapt or change in response to experience or environment depended on the synaptic neuronal plasticity and connections. However، the molecular mechanisms that translate neuronal activity patterns for specific changes in synapses are not clearly understood.

  • Gene Therapy In Multiple Sclerosis (MS)

    Multiple sclerosis (MS) is an autoimmune disease of the central nervous system (CNS) in which immune system attacks the myelin of the nerves and demyelination occurs in the white matter of the brain and progressive neurological disability. For disease treatment، protective aspects of MS are unclear and due to the blood– brain barrier، systemic anti-inflammatory drugs are not effective. The latest research on gene therapy proposes the possibility of site-directed، long-term expression، and what is presently being under a particular examination in experimental autoimmune encephalomyelitis (EAE). However، human clinical trials on gene therapy in autoimmunity disease are in their initial stage. In the practical study، a plasmid vector، BHT-3009، was made in modified pVAX1 plasmid. Using this plasmid was observed as the first effective try by a DNA vaccine in modulating the autoimmune response in human. Then this method directed in a cohort of relapsing–remitting MS or secondary progressive MS patients. Cases who received BHT-3009 had discrete reduction in both IFN- γ producing reactive CD4+ T cells in the peripheral blood. Also، in secondary Phase of clinical trial، MS patients were managed with placebo، and two different dose of BHT-3009. Compared with placebo، in patients who carry out vectors، there were 61% lower lesions with 0. 5 mg BHT-3009. The Mean volume of increasing lesions at 48 weeks was 51% lower than 0. 5 mg BHT-3009 compared with placebo and a significant decrease in autoantibody titers was also observed. Therefore، it is suggested that BHT-3009 may be hopeful therapy for MS.

    Infectious diseases caused by microorganisms are still major causes of death all over the world and the increasing number of their resistant strains results in great concerns. Since the discovery of gene silencing using RNA interference (RNAi), there has been a revolution in its medical applications. By using RNAi, proliferation and progression of viral, bacterial, and parasitic infections such as the HIV virus, hepatitis C virus (HCV), SARS virus, hepatitis B virus (HBV), Ebola virus, methicillin resistant Staphylococcus aureus (MRSA), Mycobacterium tuberculosis, Francisella tularensis, and protozoa and helminthes parasites are prevented. RNAi-based gene therapies have become more and more interesting and promising. This study has reviewed various therapeutic characteristics of RNAi, especially its therapeutic characteristics on human infectious diseases.

  • Biclustering Algorithm in Gene Expression Data: A Review Article

    In recent years، DNA microarray technology has provided monitoring of thousands of gene expressions simultaneously when cells are under different conditions and various processes. This technology has a key role in accelerating and increasing the efficiency of gene expression studies. Biclustering has been one of the most important techniques used for detecting pattern recognition and finding groups with similar expression patterns. The main goal of this study was to provide a review of biclustering methods in gene expression data and the evaluation of different algorithms. In order to serve this purpose، we investigated published articles from 2000 to 2014. Search results in multiple sites yielded 16 English articles on main algorithms. The results showed that the best algorithm was the data mining pattern. However، this algorithm makesuse of exhaustive search for discovering biclusters and assesses all of combinations، which make it time consuming and costly. We believe it is prudent to find solutions to issues in others algorithms to access more proper results.

  • The Manner in Which Low Level Laser Makes Biochemical, Physiological, Morphological, and Genetic Changes in Cells and the Consequences of These Changes in Living Creatures

    As the world population and the demand for agricultural products grow, it becomes more necessity to provide and explore new ways to enhance and improve the quantity and quality of agricultural products. Applying chemical methods for increasing the quantity of products leads to negative changes in the characteristics of the soil, water, and atmosphere and causes toxicity. Hence, researchers have focused specially on applying biophysical techniques. Although lasers have been used frequently for industrial and medical purposes, the use of lasers in biology began in the 1960s. Laser is amplification of stimulated emission light which has unique characteristics and causes significant effects. The resulting effects of laser is to enhance the potential and energy of the seed, plant resistance, enzyme activity, rate of germination, seed vigor, pigments activity, photosynthesis, DNA and RNA synthesis, plant metabolism, and early ripening.The laser mechanism is not well understood; however, several hypotheses are considered such as the effect on the mitochondrial electron transport and photochromic system. The most of the available resources are unanimous about the resulting effect of laser on increasing cell ATP content. Differences in laser effects are due to the amount of light absorption by the plant. There are several factors which can affect the results of laser; first, cell and plant conditions and then, physical characteristics of the laser. This article is supposed to investigate and evaluate the mechanism, effects, and applications of low level laser on cells. Also, we introduced low level laser as an alternative tool in plant breeding in agriculture and biotechnology to achieve the mentioned goals.

  • Challenges and Opportunities of Biotechnology on Animal Production of Developing Countries

    In animal production، biotechnology can be divided into biological، chemical and physical techniques which affect the quality and quantity of animal products through different approaches including reproduction، breeding، nutrition and hygiene. These techniques have been less used in the developing countries due to lack of appropriate infrastructures، primitive technical and educational skills and low economic growth. Therefore، using biotechnology is necessary in developing countries because of high rate of population growth and increasing demand for foods. Using biotechnological approaches accelerate animal production and lower the costs of animal production which motivates poepole to invest more on animal production enterprises. This in turn will lead to increase the demand for land and its price، increase of labour cost and and the cost of non-agricultural services، increase of purchasing power of poor people and increase of economic growth rate. Besides، biotechnological approaches provide a balance between economic exploitation of natural resources and their conversation for the future. It seems that in some countries biotechnology passed the borders of safety and put the publich health endeger. In such cases، regulatory and political rules has to set to avoid abusing biotechnological techniques and to produce high quality and safe animal products. In this review opportunity، challenges and the role of biotechnology on the improvement of animal production in developing countries has been reviewed.

  • Epigenetics: A new Challenge In Animal Breeding

    In spite of advances in genetic evaluations through genomic information، using epigenetic information is still required. Recent progress in epigenetic indicates that the most environmental factors (maybe all of them) affect animal’s phenotype via epigenetic. Epigenetic includes alterations in chromatin structure and collectively establishes different patterns of gene expression and silencing in genome and is transferred during mitotic and meiotic divisions. In the present study، the mechanisms causing the epigenetic regulations and application of epigenetic in animal breeding are discussed، and new opportunities created by this interesting field have been explained. Three types of epialleles including permanent، temporary and intergenerational، which can be used in epigenetic models have also been proposed. Furthermore، using previous results of other researches we firstly present a model at the molecular level explaining the relationship between epigenetic، genetics and environment in quantitative traits. According to this model، using interaction between epigenetic، genetics and environment can improve estimation of breeding values and reduce their biases.
