The Effect of Elaidic Acid on TNF-α Gene Expression in Macrophage Cell Line J744

Abstract:
Background and
Purpose
Recent studies reported that not only the quantity, but also the quality of dietary fatty acids can affect lipid parameters. Elaidic acid as one of the most important by-product of partially hydrogenated vegetable oil, participate in the development of cardiovascular diseases, especially atherosclerosis through changing in lipid profile and exacerbation of inflammation and inflammatory factors. On the other hand, the role of TNF-α is evident in inflammation and inflammatory pathways leading to atherosclerosis. Thus we investigated the effect of elaidic acid on TNF-α gene expression.
Materials And Methods
J744 cells were treated with 100 and 200µm concentrations of elaidic acid for 24 hr. The expression of TNF-α gene was evaluated after RNA extraction and cDNA synthesis using Real-Time PCR.
Results
Gene expression of TNF-α in J744 macrophage cell line showed no significant difference compared with that of the control group at both concentrations of elaidic acid.
Conclusion
These results indicated that the elaidic acid does not affect the development of atherosclerosis, through TNF-α gene expression.
