Multiple alignment and phylogenetic tree drawn by the neighbor-joinin cp gene of Lettuce mosaic virus in Golestan province

lately the symptoms of this viral diseases Lettuce mosaic virus (LMV) have become prevalent in lettuce farms. This virus destructive and economically important disease which is belonging to the genus potyvirus of potyviridae. In order to detect these viruses plants were visited, and 93 samples with mosaic, mottle, stunting and absence of natural crown symptoms were collected in 2013- 2014 from lettuce fields around Gorgan and transferred to the lab in plastic bags and beside the ice. samples were examined using DAS-ELISA serological test by LMV-IgG monoclonal antibodies. 80 samples with symptoms of mosaic, and mottle showed positive reactions to the ELISA test and yellowing symptoms had negative results. Some of the infected samples were chosen for sequencing. Total RNA of the samples was extracted to confirm the virus infection after cDNA syntheses, to amplify a partial fragment of protein coat gene (CP) by reverse transcription polymerase chain (RT-PCR) using potyvirus universal primers (Oligo1n/Oligo2n). The sequence were compared by different countries in Gen Bank. Multiple alignment and phylogenetic analysis performed by DNASTAR, DNAMAN, ClustalX programs and the neighbor-joinin method was drawn for phylogenetic tree. Sequencing results revealed the samples were infected by LMV. Phylogenetic analysis of iranian isolate (Golestan) showed 99.7% similarity at the nucleotide level with french isolate ) Z78229). this is the first report of sequencing of LMV virus protein coat from lectuce fields in Golestan province of Iran.
