Comparing the efficiency of PCR obtained for the GAPDH, TGF-β and IL-10 genes with the two standard curve and LinRegPCR software

Background And Objective
A basis of the molecular biology techniques is the polymerase chain reaction (PCR), we can amplified specific fragments of DNA by Using this technique. The detection of nucleic acids using PCR techniques with advanced Real-Time CPR is considerably easier, quantitative analysis of mRNA using Real-Time PCR by Relative and Absolute methods widely used in biological studies, the purpose of this study was using Relative and calculated PCR Efficiency by standard curve and LinRegPCR methods and compared them. Methods
After sampling and extraction of RNA and cDNA synthesis the quantitative RT-PCR was performed, then calculate PCR Efficiency by Using the two method including standard curve and LinRegPCR method were performed then the two methods were compared and analyzed.
Results
The efficiency of PCR for the GAPDH, TGF-β and IL-10 genes with the standard curve method were 1.99, 1.81 and 1.87, respectively. The PCR efficiency of these three genes were 1.98, 1.82 and 1.82 by using LinRegPCR software method, respectively,
Discussion
The results showed that analysis of data from the amplified cDNA efficacy of PCR results for the three genes, GAPDH, TGF β and IL-10 with two different standard curve and LineRegPCR method have no statistical difference and the same results can be achieved, so the two methods can be used instead of each other.
