Vanadium Schiff base compound mediated apoptosis and cytotoxicity effects in K562 cell line
Previous studies revealed that Vanadium complexes exhibit interesting biological properties. The goal of this study was to examine the antiproliferative and apoptosis induction virtue of some newly synthesized Schiff-base of vanadium (VOL Complex: C17H16N2O3V and C19H20N2O5V) on K562 leukemia cells.
In this experimental study، the leukemia cells were cultured in RPMI medium in 25 and 400 µg/L concentration and they were incubated for 48 hours. The vital activity of cells was assessed by MTT method. Apoptosis induction percentage was measured in 12، 24 and 48 hours after encountering with 150، 250 and 350 µg/L concentrations of VOL Complex by Annexin-PI kit and flow cytometry Cell cycle analysis was performed in the 150 µg/L concentration at 24 hours with flow cytometry.
The MTT results showed that the K562 cells viability were sensitive to VOL complex in a concentration. dependent manner With increase in Schiff-base of vanadium concentration the survival percentages of K562 leukemia cells were decreased to 90، 85 and 80 percent. Encountering cells with VLO complex caused to induce apoptosis in relation with time and concentration. The highest apoptosis (34. 11 percent) was happened in 350 µg/ml at 48 hours.
This finding indicated that VOL complex، even in noncytotoxic dose، have just an apoptotic inducing effect and capable of arresting the affected cell in Go/G1 phase of cell cycle.
