لوسمی لنفوبلاستی حاد (Acute lymphoblastic leukemia یا ALL) شایعترین لوسمی در کودکان است. تغییر در بیان miRNAها در ALL اتفاق میافتد. هدف از انجام این مطالعه، بررسی اهمیت miR-128، miR-144-3p، miR-181b و miR-451 در روند تشخیص و پاسخ به درمان در کودکان مبتلا به لوسمی حاد لنفوبلاستی B است.
روشهادر این مطالعه، نمونههای سرم از 12 کودک مبتلا به لوسمی حاد لنفوبلاستی B در زمان تشخیص (گروه مورد) و همچنین، 12 فرد سالم به عنوان گروه شاهد، جمعآوری شده است. پس از درمان، بار دیگر از این بیماران نمونهگیری شد و مقدار بیان miR-128، miR-144-3p، miR-181b و miR-451 در بیماران در دو مرحلهی تشخیص و پس از درمان بررسی شد.
یافتههابیان miR-144-3p در گروه مورد نسبت به گروه شاهد کاهش (26/0 برابر و 020/0 = P) و در زمان بعد از درمان نسبت به قبل از درمان افزایش داشت (4 برابر و 001/0 > P). علاوه بر این، بیان miR-128 (11/0 برابر و 006/0 = P) و miR-181b (34/0 برابر و 040/0 = P) در وضعیت پس از درمان در مقایسه با قبل از درمان به طور معنیداری کاهش داشت.
نتیجهگیریکاهش بیان miR-144-3p میتواند به عنوان نشانگر تشخیصی و در مقابل، افزایش بیان این miRNA میتواند به منظور پیگیری پاسخ به درمان در بیماران ALL به کار رود. علاوه بر این، بیان کاهشی miR-128 و miR-181b پس از درمان بیماران، نشان میدهد که این miRNAها میتوانند به عنوان نشانگرهای درمانی در کودکان ALL پیشنهاد شوند.

گونههای فعال اکسیژن (Reactive oxygen species یا ROS) یکی از مهمترین عوامل آسیبرسان بر کیفیت اسپرم در طی فرایند انجماد میباشند که باعث افزایش میزان پراکسیداسیون لیپیدی (Lipid peroxidation یا LPO) غشای سلول میباشد. مطالعهی حاضر، با هدف اندازهگیری غلظت مالون دیآلدیید (Malondialdehyde یا MDA) در مایع سیمن مردان الیگواسپرمی پیش و پس از فرایند انجماد و بررسی تاثیر فولیک اسید و نیکوتینیک اسید بر غلظت MDA این افراد پس از فرایند انجماد انجام شد.
روشهادر این مطالعهی تجربی، نمونهی مایع سیمن از 25 مرد الیگواسپرمی در محدودهی سنی 45-25 سال جمعآوری گردید. هر نمونه به 5 گروه شامل گروه تازه (Fresh) ، گروه انجماد بدون آنتیاکسیدان (شاهد)، گروه انجماد با نیکوتینک اسید (10 میلیمول)، گروه انجماد با فولیک اسید (50 نانومول) و گروه انجماد با ترکیبی از نیکوتینک اسید (10 میلیمول) + فولیک اسید (50 نانومول) تقسیم شدند. میزان غلظت MDA با استفاده از اسپکتروفتومتر در طول موج 535 نانومتر بر حسب نانومول/میلیلیتر در هر گروه ارزیابی گردید.
یافتههامطالعهی حاضر نشان داد که میانگین غلظت MDA در مایع سیمن پس از فرایند انجماد (12/0 ± 73/1 نانومول/میلیلیتر) در مقایسه با قبل از انجماد (02/0 ± 45/0 نانومول/میلیلیتر) افزایش یافت (001/0 > P). همچنین، میانگین غلظت MDA در گروه فولیک اسید + نیکوتینیک اسید (06/0 ± 68/0 نانومول/میلیلیتر) در مقایسه با سایر گروههای بعد از انجماد کمتر بود (001/0 > P).
نتیجهگیریترکیب آنتیاکسیدانهای فولیک اسید و نیکوتینیک اسید همراه محیط انجماد اسپرم منجر به کاهش سطح MDA و LPO غشای اسپرم در طی فرایند انجماد و حفظ پتانسیل باروری مردان الیگواسپرمی میشود.

یکی از دلایل عفونتهای بیمارستانی، پراکندگی اسپورهای Aspergillus در محیط میباشد. ترشح آنزیمهای هیدرولیتیک به عنوان یک عامل ویرولانس در گونههای Aspergillus در نظر گرفته میشود. این مطالعه، با هدف شناسایی گونههای Aspergillus محیطی با تعیین توالی ژن بتا توبولین و ارزیابی قدرت تولید آنزیم پروتییناز و فسفولیپاز در شرایط آزمایشگاهی انجام شد.
روشها93 کلنی Aspergillus از فضای بخش اورژانس، جراحی، مراقبتهای ویژه و اتاق عمل دو بیمارستان آموزشی استان قزوین جمعآوری شد. ناحیهی ژنی بتا توبولین به روش Polymerase chain reaction (PCR) تکثیر شد و 40 ایزوله تعیین توالی شدند. جهت ارزیابی قدرت تولید آنزیم پروتییناز و فسفولیپاز به ترتیب از محیط Yeast carbon base (YCB) و آلبومین سرم گاوی و محیط Egg yolk agar استفاده شد.
یافتههابر اساس توالی بتا توبولین، Aspergillus flavus (3 درصد)، Aspergillus tubingensis (25 درصد)، Aspergillus fumigatus (20 درصد) Aspergillus niger (10 درصد)، Aspergillus sydowii (5/7 درصد)، Aspergillus terreus (5 درصد) و Aspergillus nidulans (5/2 درصد) شناسایی گردید. ارزیابی آنزیمهای خارج سلولی نشان داد که 5/82 درصد از ایزولهها توانایی پروتییناز با میانگین پروتییناز 13/0 ± 73/0 و 5/52 درصد از ایزولههای Aspergillus مورد بررسی دارای فعالیت فسفولیپازی با میانگین 17/0 ± 81/0 میباشند.
نتیجهگیریمطالعهی حاضر نشان داد که سویههای محیطی دارای تولید بالای پروتییناز میباشند. بنابراین، به نظر میرسد که درک بهتر ارتباط عوامل ویرولانس با عفونت Aspergillus در مطالعات آینده، ضروری میباشد.

Acute lymphoblastic leukemia (ALL) is the most common leukemia in children. Changes in the expression of microRNAs occur in ALL. The purpose of this study was to investigate the importance of miR-128, miR-144-3p, miR-181b, and miR-451 in diagnosis and response to treatment in children with B-ALL.
MethodsIn this study, serum samples were collected from 12 children with B-ALL at diagnosis, as well as 12 healthy individuals as a control group. All patients received chemotherapy and after treatment entered in this study, and the expression of miR-128, miR-144-3p, miR-181b, and miR-451 was evaluated after and before treatment of patients.
FindingmiR-144 was significantly downregulated in newly diagnosed patients with ALL compared to control groups (0.26 fold, P = 0.020), and also the expression of miR-144-3P significantly increased after treatment in comparison to before it (4 fold, P < 0.001). In addition, miR-128 (0.11, P = 0.006), and mir-181b (0.34 fold, P = 0.040) expression significantly decreased after treatment compared to newly diagnosed patients.
ConclusionThe reduced expression of miR-144-3p can be served as diagnostic biomarker, and upregulation of miR-144-3p can predict treatment response in patients with ALL. Moreover, the abnormally reduced expression of miR-128 and miR-181b after the treatment of patients, indicate that these microRNAs can be candidate as therapeutic biomarker in pediatric ALL.

Reactive oxygen species (ROS) are of the most important detrimental factors on sperm quality during freezing process, which increases the lipid peroxidation (LPO) of cell membranes. In this study, the concentration of malondialdehyde (MDA) levels in semen of men with oligospermia before and after cryopreservation process was measured to evaluate the effect of folic acid and nicotinic acid on the MDA concentration after freezing.
MethodsIn this experimental study, semen fluid sample was collected from 25 men with oligospermia in age range of 25-45 years. Each sample was divided into 5 groups: Fresh group, Freeze group without antioxidants (control), Freeze group with nicotinic acid (10 mM), Freeze group with folic acid (50 nM), and Freeze group with a combination of nicotinic acid (10 mm) + folic acid (50 nM). The concentration of MDA in nmol/ml was measured in each group with spectrophotometer at 535 nm.
FindingsThe mean concentration of MDA in semen increased after freezing (1.73 ± 0.12) compared to before freezing (0.45 ± 0.02) (P < 0.001). Mean concentration of MDA in the group of folic acid + nicotinic acid (0.68 ± 0.06) was lower compared to other groups after freezing (P ± 0.001).
ConclusionThe combination of folic acid and nicotinic acid antioxidants with sperm freezing medium reduces the level of MDA and LPO of sperm membrane during freezing process, and thereby maintains the fertility potential in men with oligospermia.

One of the causes of nosocomial infections is the dispersion of Aspergillus spores in the environment. The secretion of hydrolytic enzymes is considered as a virulence factor in Aspergillus species. The aim of this study was to identify environmental Aspergillus isolates via sequencing the beta-tubulin gene and evaluating the ability to produce phospholipase and proteinase in vitro.
Methods93 Aspergillus colonies were collected from the emergency, surgical wards, intensive care unit, and operation theatres of two teaching hospitals in Qazvin Province, Iran. The β-tubulin gene region was amplified using polymerase chain reaction (PCR) method, and 40 isolates were sequenced. Evaluation of proteinase and phospholipase production was performed using yeast carbon base (YCB) with bovine serum albumin and egg yolk agar medium, respectively.
FindingsBased on β-tubulin sequence, Aspergillus (A.) flavus (30%), A. tuberculosis (25%), A. fumigatus (20%), A. niger (10%), A. sydowii (7.5%), A. terreus (5%), and A. nidulans (2.5%) were identified. Evaluation of extracellular enzymes showed that 82.5% of the isolates had proteinase ability with a mean proteinase of 0.73 ± 0.13, and 52.5% of the studied Aspergillus isolates had phospholipase activity with a mean of 0.81 ± 0.17.
ConclusionOur study showed that environmental strains have high proteinase production. Therefore, it seems necessary to better understand the association of virulence factors with aspergillosis infection in future studies.
